Identification of three types of O-glycosylated flavonoids in Dendrobium loddigesii, Dendrobium primulinum, Dendrobium crepidatum, Dendrobium porphyrochilum, and Dendrobium hancockii using mass spectrometry.

Flavonoids, representing the pharmacologically active ingredients, are found widely in Dendrobium species. The biodiversity of Dendrobium makes the identification of its varieties all the more complicated. Previous studies showed that C-glycosylated flavones and a few O-glycosylated flavonols could be used in the identification of various Dendrobium species. Accordingly, this study further explores the significance of the identification of various types of O-glycosylated flavonoids in Dendrobium species. High-performance liquid chromatography coupled with electrospray ionization multistage tandem mass spectrometry (HPLC-ESI-MSn ) was used to identify the chemical constituents in five types of Dendrobium: Dendrobium loddigesii, Dendrobium primulinum, Dendrobium crepidatum, Dendrobium porphyrochilum, and Dendrobium hancockii. A total of 41 O-glycosylated flavonoids and 3 C-glycosylated flavones were identified, among which O-glycosylated dihydroflavones were the main flavonoids in D. loddigesii and D. primulinum, O-Glycosylated flavonols were rich in both D. crepidatum and D. porphyrochilum characterized by the main aglycone, substituted sugars, and their structural characteristics, and O-glycosylated flavones were the main constituents in D. hancockii. In this study, three types of O-glycosylated flavonoids in the five Dendrobium species were determined to have certain significance. This also provides a reference for the identification of other O-glycosylated flavonoids in Chinese herbs.

Identification of C-glycosyl flavones and O-glycosyl flavones in five Dendrobium species by high-performance liquid chromatography coupled with electrospray ionization multi-stage tandem MS.

Flavones are widely used in traditional Chinese medicine (TCM) and are the pharmacologically active ingredients of many medicinal plants, such as Dendrobium. With the increasing demand for medicinal Dendrobium, the identification of characteristic flavones that can serve as chemical markers for quality control is critical step for quality assurance and safety in the TCM industry. High-performance liquid chromatography coupled with electrospray ionization multi-stage tandem mass spectrometry (HPLC/ESI-MSn ) was used to identify the chemical constituents in five types of Dendrobium: D. crystallinum, D. falconeri, D. strongylanthum, D. moniliforme, and D. gratiosissimum. A total of seventy-six C-glycosyl flavones and three O-glycosyl flavones were identified, of which fifteen C-glycosyl flavones were found in D. crystallinum, twenty four were found in D. falconeri, thirty were found in D. strongylanthum, seven were found in D. moniliforme (also called "Huangtongpi", from Anhui, China), fifteen were found in D. moniliforme (also called "Zitongpi", from Yunnan, China) and seventeen were found in D. gratiosissimum. Additionally, three flavone O-glycosides were all found in D. strongylanthum. The results of this study may be useful for the quality assessment and for the application of D. crystallinum, D. falconeri, D. strongylanthum, D. moniliforme, and D. gratiosissimum. This study provides comprehensive information for identification of flavones from other Chinese herbs.

HPLC coupled with electrospray ionization multistage MS/MS and TLC analysis of flavones-C-glycosides and bibenzyl of Dendrobium hercoglossum.

Dendrobium hercoglossum Rchb. f. (D. hercoglossum), as one of the origins of medicinal Dendrobium, has been widely used as a health food and nutrient source promoting fluid production. Due to a lack of quality control, it is often counterfeited or mixed with other Dendrobium. In this study, a high-performance liquid chromatography characteristic chromatogram method is established for the quality evaluation of D. hercoglossum. Based on the high-performance liquid chromatography characteristic chromatogram, D. hercoglossum is divided into two classes, each with different flavone peaks. These flavone peaks were identified using high-performance liquid chromatography coupled with electrospray ionization multistage tandem mass spectrometry. Among them, the acylated (3-hydroxy-3-methylglutaryl, p-coumaroyl, feruloyl, or sinapoyl) flavones-C-glycosides are first found in D. hercoglossum in this study. In addition, one unique band was found in D. hercoglossum by thin-layer chromatography, which can be used to distinguish it from other Dendrobium species as a characteristic marker of this plant. Combining the high-performance liquid chromatography characteristic chromatogram and high-performance liquid chromatography coupled with electrospray ionization multistage tandem mass spectrometry, the unique band was identified as 4,5-dihydroxy-3,3'-dimethoxybibenzyl. These analysis methods can be applied for the quality control and identification of D. hercoglossum as well as providing reference for the identification of similar constituents in other Dendrobium species.

Characterization of supramolecular peptide-polymer bioconjugates using multistage tandem mass spectrometry

Electrospray ionization multistage tandem mass spectrometry (ESI-MSn) was employed to examine the non-covalent complexes between poly(styrene sulfonate) (PSS) and poly-l-lysine (PLL). During single-stage ion activation, the PLL peptide chain mainly underwent backbone cleavages without disruption of the non-covalent interaction which could only be broken via sequential application of electron transfer dissociation (ETD) and collisionally activated dissociation (CAD), indicating strong binding interactions between the two polyelectrolyte chains. Such binding properties make PSS a potential “non-covalent (supramolecular) label” for determining the surface accessibility of basic residues on a peptide or protein. To probe this premise, non-covalent complexes of substance P and PSS were characterized by ESI-MSn using different ion activation methods. Both MS2 and MS3 experiments on the substance P + PSS complex resulted in the formation of bn (on CAD) or cn (on ETD) fragments attached non-covalently to the intact PSS chain. All peptide fragments containing the intact PSS chain included Arg1, Lys3, and Gln5, pointing out that these residues, which are located near the N-terminus, are most likely involved in the non-covalent interaction with PSS. In contrast, Gln6 was excluded from this fragment series, attesting a much weaker interaction with PSS due to lesser accessibility. The strong tendency of PSS to bind peptides non-covalently at sites that can be elucidated by MSn demonstrates a proof-of-concept for the capacity of this approach to unveil higher order structure in proteins.

A novel bacteriocin from Enterococcus faecalis 478 exhibits a potent activity against vancomycin-resistant enterococci.

The emergence of multidrug-resistant enterococci (MDRE) and particularly vancomycin-resistant enterococci (VRE) is considered a serious health problem worldwide, causing the need for new antimicrobials. The aim of this study was to discover and characterize bacteriocin against clinical isolates of MDRE and VRE. Over 10,000 bacterial isolates from water, environment and clinical samples were screened. E. faecalis strain 478 isolated from human feces produced the highest antibacterial activity against several MDRE and VRE strains. The optimum condition for bacteriocin production was cultivation in MRS broth at 37°C, pH 5–6 for 16 hours. The bacteriocin-like substance produced from E. faecalis strain EF478 was stable at 60°C for at least 1 hour and retained its antimicrobial activity after storage at -20°C for 1 year, at 4°C for 6 months, and at 25°C for 2 months. A nano-HPLC electrospray ionization multi-stage tandem mass spectrometry (nLC-ESI-MS/MS) analysis showed that the amino acid sequences of the bacteriocin-like substance was similar to serine protease of E. faecalis, gi|488296663 (NCBI database), which has never been reported as a bacteriocin. This study reported a novel bacteriocin with high antibacterial activity against VRE and MDRE.

Improvement of shoot proliferation and comparison of secondary metabolites in shoot and callus cultures of Phlomis armeniaca by LC-ESI-MS/MS analysis

Phlomis armeniaca Willd. is a medicinal plant in the Lamiaceae family endemic to Turkey. The present study describes efficient plant regeneration and callus induction protocols for P. armeniaca and compares phenolic profiles, total phenol and flavonoid contents, and free radical scavenging activity of in vitro-derived tissues. Stem node explants from germinated seedlings were cultured on Murashige and Skoog medium (MS) supplemented with 75 plant growth regulator (PGR) combinations. The highest shoot number per explant, frequency of shoot proliferation, and frequency of highly proliferated, green, compact callus were obtained on MS medium containing 0.25 mg L−1 thidiazuron (TDZ) and 0.25 mg L−1 indole-3-acetic acid (IAA). The best root formation was on MS basal medium (control). Methanol extract of leaves obtained from regenerants contained higher total phenol and flavonoid contents than the callus extract. The callus extract showed stronger free radical scavenging activity than leaves with IC50 [concentration inhibiting 50% of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical] values of 4.30 ± 0.08 and 2.21 ± 0.04 mg g−1 dry weight in leaves and callus, respectively. Apigenin, caffeic acid, p-coumaric acid, luteolin, rutin hydrate, vanillic acid, ferulic acid, salicylic acid, sinapic acid, and chlorogenic acid were detected by liquid chromatography–electrospray ionization multistage tandem mass spectrometry (LC-ESI-MS/MS) analysis in in vitro-grown leaves and callus tissue. Rutin hydrate, p-coumaric acid, and vanillic acid were found at approximately tenfold higher levels in callus than in leaves. This new micropropagation protocol, the first for P. armeniaca, could be used in industrial production for new herbal tea and germplasm conservation.

Characteristic fragmentation behavior of tobacco‐specific N‐nitrosamines using electrospray ionization multistage tandem mass spectrometry incorporating deuterium labeling

Tobacco-specific N-nitrosamines (TSNAs) mainly include 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), N-nitrosonornicotine (NNN), N-nitrosoanabasine (NAB) and N-nitrosoanatabine (NAT) that are formed from tobacco alkaloids during the curing process and contained in tobacco and tobacco smoke. They are linked with carcinogenesis. Analytical methods for quality control of products and determination of their metabolites are therefore of great importance. The characteristic fragmentation behaviors of tobacco-specific TSNAs have been studied by electrospray ionization multistage tandem mass spectrometry. The deutero-labeled TSNA compounds have also been employed to clarify the fragmentation mechanism, which further confirms the proposed fragmentation patterns. Detailed analysis of the resultant fragments shows there are two different kinds of fragmentation patterns with the general fragment backbone of pyrrolidine or piperidine rings. In one route, pyrrolidine or piperidine rings undergo direct fragmentation and form some stable intermediates without affecting the parent rings. The other, however, involves ring opening and then ring closure at the pyridine-2 carbon atom to form multi-membered rings. This characteristic fragmentation behavior therefore provides useful information on identification of TSNAs that may be used to monitor such kinds of compound in the biological metabolism.

Structural characterization of arabinoxylans from two African plant species Eragrostis nindensis and Eragrostis tef using various mass spectrometric methods

The arabinoxylans are one of the main components of plant cell walls and are known to play major roles in plant tissues properties depending in particular on their structural features. It has been recently shown that one of the strategies developed by resurrection plants to overcome dehydration is based on cell wall composition. For this purpose, the structural characterization of arabinoxylans from desiccation-tolerant grass Eragrostis nindensis (E. nindensis) was compared with its close relative, the desiccation-sensitive Eragrostis tef (E. tef) in order to further understand mechansism of desiccation tolerance in resurrection plants. Ion mobility spectrometry coupled to mass spectrometry (IM-MS) in combination with the conventional mass spectrometric approaches, including matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS), electrospray ionization multistage tandem mass spectrometry (ESI-MS(n)) and gas chromatography/mass spectrometry (GC/MS), were used to characterize arabinoxylan fragments obtained after endo-xylanase digestion of leave extracts from E. nindensis and E. tef. Whole fingerprinting by MALDI-MS analysis showed the presence of various arabinoxylan fragments within leaves of E. nindensis and E. tef. The monosaccharide composition and some linkage information were determined by GC/MS experiments. Information regarding the branching and sequence details was obtained by ESI-MS(n) experiments after sample permethylation. The presence of structural isomeric ions with different collision cross sections was evidenced by IM-MS which could be differentiated using ESI-MS(n). We have shown that an orthogonal approach, and especially IM-MS associated to ESI-MS(n) (n = 2 to 4) and GC/MS allowed characterization of arabinoxylan fragments of E. nindensis and E. tef and revealed the presence of isomeric structures. The same arabinoxylan structures were identified for both species but in different relative abundance. Moreover, this work illustrated that IM-MS can efficiently separate isomeric structures and advantageously complements the conventional mass spectrometric methodologies used for arabinoxylan structural characterization.

THE STRUCTURAL ELUCIDATION OF THE STRYCHNOS ALKALOIDS BY HPLC-ESI-MSn

The characteristic fragmentation of four strychnos alkaloid compounds has been studied by using electrospray ionization multi-stage tandem mass spectrometry (ESI-MSn). In the obtained product ions spectra, some diagnostic neutral losses were observed, which are very helpful for the identification of strychnos alkaloids. According to the structural information collected from spectrum, four strychnos alkaloids have been identified on-line without time-consuming isolation, and two unknown alkaloids have been detected in the extract of crude alkaloids of Strychnos nux-vomica L. An effective and rapid HPLC-ESI-MSn method for analyzing constituents in Strychnos nux-vomica L. has been established and proved.

Identification of the Major Components of Resina Draconis Extract and Their Metabolites in Rat Urine by LC-ESI-MS n

The aim of our study was to employ a liquid chromatography coupled with electrospray ionization multistage tandem mass spectrometry (LC-ESI-MSn) method for the identification of the major components of Resina Draconis extract (RDE) and their metabolites in rat urine. Based on the above, 18 compounds were tentatively identified from the RDE. Among them, 4 compounds were unambiguously characterized by the comparison of the retention time and mass spectra with those of reference compounds and 14 compounds were tentatively identified by comparing the mass spectra with those of literature. In vivo, 21 compounds, including 13 parent compounds and 8 metabolites, were detected in rat urine after oral administration of RDE. The results may be helpful for future research on traditional Chinese medicine.

Identification of isomeric disaccharides in mixture by the 1-phenyl-3-methyl-5-pyrazolone labeling technique in conjunction with electrospray ionization tandem mass spectrometry

1-Phenyl-3-methyl-5-pyrazolone (PMP) labeling technique has hitherto proved to be a convenient and sensitive method for separating and detecting oligosaccharides. However, the detailed fragmentation of the derivatives by tandem mass spectrometry has been reported limitedly and no characteristic fragment ions for isomers have been detected. In this study, eight disaccharide isomers were labeled with PMP and analyzed by positive ion electrospray ionization multi-stage tandem mass spectrometry (ESI-MSn). In comparison with the native disaccharides, PMP labeled disaccharides gave rise to more fragment ions in the tandem mass spectra. The distinctive diagnostic fragment ions formed from cleavage of CC bonds have been detected in the fragmentation of PMP-labeled disaccharide linkage isomers, allowing unambiguous assignment of the position of the glycosidic linkages. This feature is particularly useful for the structural determination of unknown isomeric disaccharides mixed together. In addition, the anomeric configurations can also be easily assigned based on the relative abundance ratios of the selected ion pairs. To verify the feasibility of the method used in the analysis of natural product, water soluble Panax Ginseng extract has been further investigated to identify its unknown disaccharides. The results confirmed that the PMP labeling technique in conjunction with ESI-MSn could offer a powerful and convenient tool for differentiation of structurally closely related isomers, even the unknown mixtures of isomeric disaccharides with different linkage types.

Characterisation of “caramel-type” thermal decomposition products of selected monosaccharides including fructose, mannose, galactose, arabinose and ribose by advanced electrospray ionization mass spectrometry methods

The chemical analysis of caramel, formed upon heating of carbohydrates, remains a significant challenge due to the complexity of the resulting product mixture. Identification of the products formed upon heating of monosaccharides including fructose, mannose, galactose, arabinose and ribose is essential to understand the composition and properties of carbohydrate-rich processed foods. In this work, we report on the use of combined mass spectrometry techniques, including high performance liquid chromatography and electrospray ionization multi-stage tandem mass spectrometry (ESI-MS(n)). The composition of the obtained caramel was examined by high resolution mass spectrometry along with van Krevelen and Kendrick analysis. We found that caramel is composed of oligomers with up to six carbohydrate units formed through unselective glycosidic bond formation, their dehydrated products by losing up to eight water molecules, hydrated products and disproportionation products. An accurate mass measurement and subsequent fragment ion studies of all caramel samples (around 40 compounds) can thus be identified. Glycosidic bond and ring cleavages of sugar moieties were the major observable fragmentation pathways during this experiment. The innovative analytical strategies for the complex mixture analysis used provide a comprehensive account of the chemical composition of caramel, one of the most popular dietary materials over the world.

Structural characterization and identification of five triterpenoid saponins isolated from Momordica cochinchinensis extracts by liquid chromatography/tandem mass spectrometry

In this work, we reported the use of high-performance liquid chromatography (HPLC) with electrospray ionization multi-stage tandem mass spectrometry (ESI-MSn) to study the characterization of five triterpenoid saponins from Momordica cochinchinensis extracts in positive ion mode and negative ion mode, revealed the fragmentation behavior of these five M. cochinchinensis saponins in different ion modes. Results showed that in negative ion mode, fragmentation on the M. cochinchinensis triterpenoid saponins at positions C28 and O bond were the primary pathways. In positive ion mode, the predominant diagnostic ions were a series of ions resulting from the rupture of the saccharic chain on C28. Comparison of different fragmentation patterns showed that the results from positive and negative ion ESI MSn were complementary. Both modes can yield structurally different information for the characterization of triterpenoid saponins. This study provides a powerful method for the online structural identification of five triterpenoid saponins from M. cochinchinensis, and some of which have not been reported.

Mulberrofuran G and Isomulberrofuran G from Morus alba L.: Anti-hepatitis B Virus Activity and Mass Spectrometric Fragmentation

Mulberrofuran G (1) and isomulberrofuran G (2), a pair of isomeric Diels-Alder-type adducts, were isolated from the root bark of Morus alba L. Isomulberrofuran G (2) as a new IIB-type Diels-Alder-type adduct, was elucidated by extensive (1)H, (13)C, and two-dimensional (2D) nuclear magnetic resonance (NMR) and mass spectrometry (MS) spectroscopic analyses. A fragmentation study on compounds 1 and 2 was performed by high-resolution electrospray ionization (ESI) multistage tandem mass spectrometry linked with ion-trap (IT) and time-of-flight (TOF) mass analyzers (ESI-MS(n)/IT-TOF) in negative mode, which resulted in obviously different fragmentations. In the MS(2) experiments, the characteristic ions at m/z 451 and 439 could be revealed as their respective diagnostic ions. Mulberrofuran G (1) showed moderate activity, inhibiting hepatitis B virus (HBV) DNA replication with the IC(50) value of 3.99 μM, according to the anti-HBV assay on the HepG 2.2.15 cell line in vitro.

Systematic screening and characterization of the major bioactive components of Poria cocos and their metabolites in rats by LC‐ESI‐MSn

Poria cocos is a well-known medicinal plant widely used in China and other East Asian countries owing to its various therapeutic effects. However, the bioactive constituents responsible for the pharmacological effects of Poria cocos and their metabolites in vivo are still unclear to date. The aim of the present study was to develop a practical method based on the combined use of the liquid chromatography coupled with electrospray ionization multistage tandem mass spectrometry (LC-ESI-MS(n) ) for the comprehensive and systematic separation and characterization of the bioactive constituents of Poria cocos extract and their metabolites in rats. Based on the proposed strategy, a total of 34 compounds were characterized from the extract of Poria cocos. Among them, eight were unambiguously identified by comparing their retention times and mass spectra with those of reference standards, and 26 were tentatively identified on the basis of their MS(n) fragmentation behaviors and molecular weight information from literatures. In vivo, seven compounds were successfully detected in rat urine whereas one was found in rat plasma. This study proposed a series of potential bioactive components and provided helpful chemical information for further research on the action mechanism of traditional Chinese medicine.

Structural characterization and identification of biflavones in Selaginella tamariscina by liquid chromatography‐diode‐array detection/electrospray ionization tandem mass spectrometry

Biflavonoids, a special class of flavonoids, are widely distributed in gymnosperm plants and have various biological activities. They are also major bioactive ingredients in Selaginella tamariscina. In this work, we report the use of high-performance liquid chromatography with a diode-array detector (HPLC-DAD) and electrospray ionization multi-stage tandem mass spectrometry (ESI-MS(n)) to study the fragmentation behavior of three main types of biflavonoids using seven biflavonoid reference compounds and analyze the biflavonoids in Selaginella tamariscina. The most useful fragmentations in terms of structural identification are those involving the C-ring cleavage of biflavonoids. For amentoflavone-type biflavonoids (containing flavonoid parts I and II), fragmentation on the flavonoid part II at positions 1/3 and 0/4 are the primary pathways, whereas the chances are greater for C-ring cleavage fragmentation occurring on flavonoid part I at positions 1/3 and 1/4 for robustaflavone-type biflavonoids. However, the predominant diagnostic ions of the specific C-O-C-connected hinokiflavone-type biflavonoids are a series of ions resulting from the rupture of the connective C-O bond. Based on the fragmentation patterns of these reference compounds, 17 biflavonoids were identified in an extract of Selaginella tamariscina, three of which have not been previously reported as constituents of this plant. This study provides a powerful approach for the online structural elucidation and identification of different types of biflavonoids and positional isomers from Selaginella tamariscina and other biflavonoids distributed in related plants and prescriptions.

In vivo metabolism study of polygalic acid in rat using HPLC‐ESI‐MSn

A very simple and direct method has been established for the determination of polygalic acid and its metabolites in rat urine based on HPLC coupled with electrospray ionization multi-stage tandem mass spectrometry (HPLC-ESI-MS(n)). The rats were administered a single dose (100 mg/kg) of polygalic acid by oral gavage. The urine samples were collected and purified through a C(18) solid-phase extraction cartridge, and then these pretreated samples were injected into a reversed-phase C(18) column with a gradient elution program, whereas acetonitrile-0.5% aqueous formic acid was used as mobile phase and detected by an on-line MS/MS system. As a result, the parent drug and its four metabolites were identified and characterized in rat urine for the first time by comparing their changes in molecular mass (ΔM), retention times and full-scan MS(n) spectra with those of the parent drug. A possible metabolic pathway of polygalic acid was investigated and proposed. More importantly, the results demonstrated that the newly developed method (HPLC-ESI-MS(n)) was sensitive, simple and suitable for the determination of polygalic acid and its metabolites in biological samples.

Identification of Amino Acid Phosphorodiamidates of Antiviral Nucleosides Using Electrospray Ionisation Tandem Mass Spectrometry

Several amino acid phosphorodiamidate derivatives of d4T as anti-HIV prodrugs were synthesized and investigated using electrospray ionization multistage tandem mass spectrometry (ESI-MS(n)). A novel methyl group migration in gas phase was observed in ESI-MS(2) of the sodium adducts of amino acid methyl ester of phosphorodiamidates of 2',3'-didehydro-2',3'-dideoxythymidine (d4T). The proposed structures of the rearrangement ions were confirmed by high resolution tandem mass spectrometry. A possible mechanism involving the pentacoordinate phosphoric-carboxylic phosphate anhydride was proposed, in which a seven-membered ring intermediate was formed by coordination with the metal ion between the phosphoryl group and carbonyl oxygen atom. Thus, the intrinsic properties of phosphoryl group might be the key factors responsible for this migration.

Saponins of Glycine max Merill.: Isolation and further characterization by NMR and ESI-MS/MS

Legumes play a pivotal role in the traditional diets of many regions throughout the world. Soybeans are unique among the legumes because they are a concentrated source of soysaponins. Soysaponins are basically triterpene glycosides and comprise a hydrophobic aglycone linked to one or more hydrophilic mono- or oligosaccharide moieties [1]. Chemical and health effects of soysaponins are numerous and suggest a number of biological properties [2]. As a continuation of our work on the chemical elucidation of bioactive markers from plants, ten soysaponins were isolated from Glycine max. All saponins were isolated from saponin concentrate of said plant by using CHCl3-MeOH-H2O lower layer in an almost isocratic way through a gravity column. All isolated compounds were characterized by NMR. A number of constituents and different possibilities of sugar chain composition and attachment cause greater diversity of soysaponin structures. In order to determine the structures of isolated compounds, ten soysaponins were analyzed under positive and negative mode respectively, by electrospray ionization multi-stage tandem mass spectrometry (ESI-MSn) coupled with an ion-trap analyzer. In positive mode, the sodium adduct molecular ions [M+Na]+ and [M+2Na-H]+ were easily detected. However, β-D-glucopyranosiduronic acid required negative mode to detect [M-H]-, [M-H-sugar(s)]- and [aglycone]-. The proposed fragmentation pathway is useful for structural characterization of soysaponins.

An Isotope (18O, 15N, and 2H) Technique to Investigate the Metal Ion Interactions Between the Phosphoryl Group and Amino Acid Side Chains by Electrospray Ionization Mass Spectrometry

Cationic metal ion-coordinated N-diisopropyloxyphosphoryl dipeptides (DIPP-dipeptides) were analyzed by electrospray ionization multistage tandem mass spectrometry (ESI-MS(n)). Two novel rearrangement reactions with hydroxyl oxygen or carbonyl oxygen migrations were observed in ESI-MS/MS of the metallic adducts of DIPP-dipeptides, but not for the corresponding protonated DIPP-dipeptides. The possible oxygen migration mechanisms were elucidated through a combination of MS/MS experiments, isotope ((18)O, (15)N, and (2)H) labeling, accurate mass measurements, and density functional theory (DFT) calculations at the B3LYP/6-31 G(d) level. It was found that lithium and sodium cations catalyze the carbonyl oxygen migration more efficiently than does potassium and participation through a cyclic phosphoryl intermediate. In addition, dipeptides having a C-terminal hydroxyl or aromatic amino acid residue show a more favorable rearrangement through carbonyl oxygen migration, which may be due to metal cation stabilization by the donation of lone pair of the hydroxyl oxygen or aromatic π-electrons of the C-terminal amino acid residue, respectively. It was further shown that the metal ions, namely lithium, sodium, and potassium cations, could play a novel directing role for the migration of hydroxyl or carbonyl oxygen in the gas phase. This discovery suggests that interactions between phosphorylated biomolecules and proteins might involve the assistance of metal ions to coordinate the phosphoryl oxygen and protein side chains to achieve molecular recognition.
