Highly sensitive and selective nanostructured microbiosensors for glucose and lactate simultaneous measurements in blood serum and in vivo in brain tissue

Highly sensitive and selective nanostructured lactate and glucose microbiosensors for their in vivo simultaneous determination in rat brain were developed based on carbon fiber microelectrodes (CFM) modified with nanoporous gold (NPG) using the Dynamic Hydrogen Bubble Template (DHBT) method. Electrodeposition of platinum nanoparticles (PtNP) onto the NPG film enhances the sensitivity and the electrocatalytic properties towards H2O2 detection. The nanostructured microelectrode platform was modified by glucose oxidase (GOx) and lactate oxidase (LOx) enzyme immobilization. High selective measurements were achieved by covering with a perm-selective layer of electropolymerized m-phenylenediamine, deposition of a Nafion® film and by using a null sensor. The morphological characteristics and electroanalytical performance of the microbiosensors were assessed, by scanning electron microscopy and electrochemical techniques, respectively.The PtNP/NPG/CFM shows a high sensitivity to H2O2 (5.96 A M-1 cm-2) at 0.36 V vs. Ag/AgCl, with a linear range from 0.2 to 200 μM, and an LOD of 10 nM. The microbiosensors were applied to the simultaneous determination of lactate and glucose in blood serum samples. Moreover, the basal extracellular concentrations of lactate and glucose were measured in vivo in four different rat brain structures. These results support the potential of the microbiosensor to be used as a valuable tool to investigate brain neurochemicals in vivo.

Single-Entity Electrocatalysis at Electrode Ensembles Prepared by Template Synthesis

Nanoelectrode ensembles (NEEs), prepared by Au template synthesis, are presented as a proof-of-concept sample platform to study individual electrodeposited materials by scanning electrochemical cell microscopy (SECCM). With this platform, the non-conductive membrane support does not contribute to the electrocatalytic activity recorded at each electrode. Use of low-density template membranes results in electrodes that are isolated because initial membrane pores are typically separated by significant (microscale) distances. Electrodeposition of catalytic nanoparticles onto the electrodes of the array and observation of electrocatalytic activity are demonstrated to be suitable for correlative SECCM voltammetric mapping and electron microscopy. Suitability of NEEs for studies of surface Au oxidation, hydrazine oxidation, and hydrogen evolution (hydrogen evolution reaction, HER), and at Pt particles on NEEs (Pt-NEEs) for HER is demonstrated.

MoS2@Ti3C2 nanohybrid-based photoelectrochemical biosensor: A platform for ultrasensitive detection of cancer biomarker exosomal miRNA

As a class of newly identified biomarkers, miRNAs show enormous potential in cancer diagnosis. The sensitive detection of abnormal miRNAs concentration to realize early diagnosis of malignant tumors is a frontier in the field of biosensing. In this work, a photoelectrochemical (PEC) biosensor based on MoS2@Ti3C2 nanohybrid was fabricated for the ultrasensitive detection of miRNAs. The hybridization of Ti3C2 with excellent electron transfer capability significantly enhances the photocurrent response of the PEC biosensor. Moreover, the electrodeposition of Au nanoparticles on the surface of MoS2@Ti3C2 nanohybrid further enhances the photocurrent. The detection performance of the PEC biosensor has been tested using colorectal cancer-related exosomal miRNA (miR-92a-3p) as the target. The PEC biosensor shows a broad linear detection ranged from 1 fM to 100 nM and a calculated detection limit of 0.27 fM. In terms of selectivity, the PEC biosensor can distinguish miR-92a-3p from mismatched sequences. The 16 continuous radiation source on-off cycles test indicates the high stability of the PEC biosensor. Furthermore, the accurate detection of exosomal miR-92a-3p concentrations of patients and healthy controls demonstrates the clinical feasibility of the PEC biosensor. Based on these outcomes, the PEC biosensor exhibits the prospect of realizing the ultrasensitive point-of-care detection of miRNAs.

Inherent Surface Activation of Laser-Scribed Graphene Decorated with Au and Ag Nanoparticles: Simultaneous Electrochemical Behavior toward Uric Acid and Dopamine.

Laser-scribed graphene electrodes (LSGEs) have attracted great attention for the development of electrochemical (bio)sensors due to their excellent electronic properties, large surface area, and high porosity, which enhances the electrons' transfer rate. An increasing active surface area and defect sites are the quickest way to amplify the electrochemical sensing attributes of the electrodes. Here, we have found that the activation procedure coupled to the electrodeposition of metal nanoparticles resulted in a significant amplification of the active area and the analytical performance. This preliminary study is supported by the demonstration of the simultaneous electrochemical sensing of dopamine (DA) and uric acid (UA) by the electrochemically activated LSGEs (LSGE*s). Furthermore, the electrodeposition of two different metal nanoparticles, gold (Au) and silver (Ag), was performed in multiple combinations on working and reference electrodes to investigate the enhancement in the electrochemical response of LSGE*s. Current enhancements of 32, 27, and 35% were observed from LSGE* with WE:Au/RE:LSG/CE:LSGE, WE:Au/RE:Au/CE:LSGE, and WE:Au/RE:Ag/CE:LSGE, compared to the same combinations of LSGEs without any surface activation. A homemade and practical potentiostat, KAUSTat, was used in these electrochemical depositions in this study. Among all of the combinations, the surface area was increased 1.6-, 2.0-, and 1.2-fold for WE:Au/RE:LSG/CE:LSGE, WE:Au/RE:Au/CE:LSGE, and WE:Au/RE:Ag/CE:LSGE prepared from LSGE*s, respectively. To evaluate the analytical performance, DA and UA were detected simultaneously in the presence of ascorbic acid. The LODs of DA and UA are calculated to be ∼0.8 and ∼0.6 μM, respectively. Hence, this study has the potential to open new insights into new surface activation strategies with a combination of one-step nanostructured metal depositions by a custom-made potentiostat. This novel strategy could be an excellent and straightforward method to enhance the electrochemical transducer sensitivity for various electrochemical sensing applications.

Semi-sacrificial template growth-assisted self-supporting MOF chip: A versatile and high-performance SERS sensor for food contaminants monitoring

Surface-enhanced Raman scattering (SERS) with its outstanding performance in ultra-sensitivity has been applied in contaminants monitoring. Herein, a facile strategy of integrating oriented layered MOF nanoarray with controlled electrodeposition of gold nanoparticles via semi-sacrificial template method was proposed to fabricate a versatile self-supporting MOF chip (S-MOF@Au) for efficient SERS detection of food contaminants. The S-MOF was ingeniously formed by directly using flexible Ni sheet as both metal source and scaffold, which in favour of greatly improving intrinsic electrical conductivity during electrodeposition process as well as endowing prepared chip with uniform distribution of gold nanoparticles and high stability. Furthermore, amounts of SERS “hot spots” in the oriented layered S-MOF@Au can yield strong electromagnetic field for Raman amplification with superior reproducibility. By virtue of good flexibility and pre-concentration effect of S-MOF@Au, tartrazine (TA), chloramphenicol (CP), imidacloprid (IDP) and crystal violet (CV) in food samples were sensitively detected using three collected approaches (drip coating, soaking and wiping) with low limits of detection (1.0 × 10−9 M for TA, 8.0 × 10−11 M for CP, 6.4 × 10−11 M for IDP and 5.0 × 10−12 M for CV), wide linear detection ranges (1.0 × 10−8 – 1.0 × 10−3 M for TA, 1.0 × 10−10 – 1.0 × 10−6 M for CP, 1.0 × 10−10 – 1.0 × 10−7 M for IDP and 1.0 × 10−11 – 1.0 × 10−7 M for CV) and satisfactory recoveries (96.2 – 125.1%). This method provides a novel strategy to develop an advanced SERS platform for efficient food contaminants monitoring.

Dexamethasone Electroanalysis at Copper Nanoparticle Modified Electrodes Formed Using a Potentiostatic Triple Pulse Sequence

Our goal is the electrochemical evaluation of anti-inflammatory/antioxidant therapeutic agents with quantification realised via electrode modifiers which facilitate detection at the appropriate level and selectivity for operation in complex clinical matrices. The analyte dexamethasone, a glucocorticosteroid and potent anti-inflammatory agent found to play a role in the treatment of severe COVID-19 cases [1], was examined in alkaline conditions at bare and copper nanoparticle (CuNP) modified glassy carbon electrodes (GCE). A three-step template free potentiostatic pulse sequence [2] was applied for the first time, realising electrodeposition of copper nanoparticles (CuNPs). This method comprised pre-treatment (E1) to remove adsorbed Cu2+ ions from the electrode surface, a nucleation pulse (E2) to seed Cu nuclei and finally a growth pulse (E3) to allow the formation of the CuNPs from the seeded nuclei, via diffusion. Optimisation was carried whereby E2 and E3 were varied along with their corresponding pulse times, where at the optimised CuNPs, the dexamethasone reduction response increased 4.5-fold, relative to the bare GCE.

Ultrasensitive early detection of insulin antibody employing novel electrochemical nano-biosensor based on controllable electro-fabrication process

An ultrasensitive novel electrochemical nano-biosensor for rapid detection of insulin antibodies against diabetes antigens was developed in this research. The presence of insulin antibodies has been demonstrated to be a strong predictor for the development of type 1 diabetes in individuals who do not have diabetes but are genetically predisposed. The proposed nano-biosensor fabrication process was based on the optimized sequential electropolymerization of polyaniline and electrodeposition of gold nanoparticles on the surface of the functionalized gold electrode. The morphological and chemical characterization of the modified electrode was studied by field emission scanning electron microscopy (FE-SEM), energy dispersive X-ray spectroscopy (EDX), atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), and micro Raman spectroscopy. Moreover, the role of each component in the modification of the electrode was studied by electrochemical methods systematically. After immobilizing insulin antigen and blocking with bovine serum albumin, the nano-biosensor was used for determining different concentrations of insulin antibody under the optimal conditions. This nano-biosensor could respond to insulin antibody with a linear calibration range from 0.001 ng ml−1 to 1000 ng ml−1 with the detection limit of 0.017 pg ml−1 and 0.034 pg ml−1 and selectivity of 18.544 μA ng−1 ml.cm−2 and 31.808 μA ng−1 ml.cm−2 via differential pulse voltammetry and square wave voltammetry, respectively. This novel nano-biosensor exhibited a short response time, high sensitivity, and good reproducibility. It was successfully used in determining the insulin antibody in human samples with a standard error of less than 0.178. Therefore, the nano-biosensor has the potential for the application of early detection of type 1 diabetes. To our best knowledge, label-free electrochemical detection of insulin antibody based on immunosensor is developed for the first time.

Two-Dimensional Restructuring of Cu2O Can Improve the Performance of Nanosized n-TiO2/p-Cu2O Photoelectrodes under UV-Visible Light.

p-Cu2O/n-TiO2 photoanodes were produced by electrodeposition of octahedral p-type Cu2O nanoparticles over n-type TiO2 nanotubes. The photoresponse of the composite p–n photoanodes was evaluated in photoelectrochemical cells operating at “zero-bias” conditions under either visible or UV–vis irradiation. In both operating conditions, the produced electrodes invariably followed the p–n-based photoanode operations but exhibited lower photoelectrochemical performance as compared to the bare n-TiO2 photoanode under UV–vis light. The reported experimental analysis evidenced that such decreased photoactivity is mainly induced by the scarce efficiency of the nanosized p–n interfaces upon irradiation. To overcome such limitation, a restructuring of the originally electrodeposited p-Cu2O was promoted, following a photoelectrochemical post-treatment strategy. p-Cu2O, restructured in a 2D leaf-like morphology, allowed reaching an improved photoelectrochemical performance for the p–n-based photoanode under UV–vis light. As compared to the bare n-TiO2 behavior, such improvement consisted of photoanodic currents up to three times larger. An analysis of the mechanisms driving the transition from compact (∼100 nm) octahedral p-Cu2O to wider (∼1 μm) 2D leaf-like structures was performed, which highlighted the pivotal role played by the irradiated n-TiO2 NTs.

NaBH4-electrooxidation mediated electrodeposition of catalytic Pt nanoparticles on a honeycomb-gold electrode for hydrogen evolution reaction

We report the NaBH4-electrooxidation mediated electrodeposition of platinum nanoparticles (PtNPs) on a pulse-plated gold (Aupla) and three-dimensional honeycomb-like porous nano-golds (3DHPNG) modified glassy carbon electrode (GCE), for electrocatalytic hydrogen evolution reaction (HER). The 3DHPNG of large surface area was deposited on Aupla/GCE at −4.76 V versus RHE for 200 s in 5 M aqueous H2SO4 containing 15 mM HAuCl4, by dynamic hydrogen bubble template. The uniform electrodeposition of PtNPs on 3DHPNG/Aupla/GCE at 1.24 V versus RHE was achieved in 25 mM aqueous NaClO4 containing 5.37 μM PtNPs dispersion that was obtained by mixing aqueous NaBH4, trisodium citrate and H2PtCl6. The electrodeposition mechanism is explained by the electrogeneration of H+ through the electrooxidation of residual NaBH4 and its oxidation intermediates, which can neutralize the negative surface-charges of some PtNPs and make the PtNPs coagulate on the electrode surface, and some new insights into the oxidation of so widely used NaBH4 on the anode and by H2PtCl6 and water are also provided. The optimized PtNPs/3DHPNG/Aupla/GCE exhibited good electrocatalytic performance for HER in 0.5 M aqueous H2SO4, giving an overpotential of 10.0 mV at −10 mA cm−2 and a Tafel slope of 25.2 mV dec−1, being better than those of many reported electrodes.

A flexible bioelectrode based on IrO2–coated metallized polypropylene micromembrane

We demonstrate the fabrication of a flexible and conductive bioelectrode via the electrodeposition of IrO2 nanoparticles on a metalized polypropylene micromembrane (PPMM). The metalized PPMM is prepared by carrying out conformal electroless deposition of Au on individual polypropylene (PP) fibers of PPMM using both polydopamine and polyvinyl alcohol as the surface–modifying agents to improve the hydrophilicity of PPMM. Images from scanning and transmission electron microscopes, as well as analysis from energy dispersive X–ray spectrometer and X–ray photoelectron spectroscopy confirm the successful formation of IrO2 nanoparticles on individual PP fibers of metalized PPMM. For bio–stimulating purpose, the IrO2–coated PPMM reveals an impressive charge–storage–capacity of 54.6 mC/cm2 in cyclic voltammetry, In addition, under a symmetric bi–phasic current pulse, its charge–injection–capacity reaches 466 μC/cm2. Since the entire constituents for the IrO2–coated PPMM are bio−compatible, we envision its potential use as a bioelectrode in implantable biomedical devices.

Band-gap shrinked NiO@Co3O4 nanotubes as high-performance supercapacitor electrodes

Novel band-gap shrinked NiO@Co3O4 nanotube arrays on Ni foam have been fabricated via a two-step synthesis route. Hexagonal NiO nanotube arrays are obtained by electrodeposition of Ni nanoparticles onto ZnO nanoprisms followed by selective dissolution of ZnO. The inside and outside surfaces of the NiO nanotubes are homogeneously covered with Co3O4 nanosheets via hydrothermal treatment, which enlarges the surface area and accelerates electron transferring. Benefiting from the hierarchically hollow structure and the synergistic effects, the obtained NiO@Co3O4 nanotube arrays deliver a superior specific capacitance of 1769.2 F/g at 1 A/g and outstanding cycling stability (87.5% capacitance retention after 10,000 cycles at 20 A/g). Density functional theory calculations reveal that the disappearance of the band-gap of the NiO@Co3O4 composite is one intrinsic reason that the conductivity and the capacitive charge storage performance are enhanced significantly. The as-prepared NiO@Co3O4 nanotube arrays are highly promising electrodes for high-performance supercapacitors.

Electronic Metal-Support Interactions for Electrochemiluminescence Signal Amplification.

Au nanoparticle-amplified electrochemiluminescence (ECL) signals are generally realized by nanoparticle morphology modification, functionalization, and nanoalloys formation. It remains a great challenge to utilize the intrinsic catalytic activity of spherical Au nanoparticles for ECL performance improvement. In this work, we prepared the oxygen vacancy-rich CoAl-layered double hydroxide (LDH-Ov)-supported spherical Au nanoparticles via alkali etching of LDH and electrodeposition of Au nanoparticles on the surface of LDH. It was found that the luminol ECL signals of the as-prepared system were significantly enhanced by forming the strong electronic metal-support interaction (EMSI) between Au nanoparticles and LDH-Ov. The further mechanism study demonstrated that EMSI can increase the electron density of interfacial Au atom (Auδ-) due to a redistribution of charge and promote electron transfer between Au species and LDH-Ov. This study not only introduces EMSI to the ECL field but also paves a new way to the applications of the intrinsic activity of spherical Au nanoparticles in ECL signal amplification. We anticipate that EMSI would be applied to other metal nanocatalysts for the development of highly efficient ECL systems.

Electrochemical detection of hepcidin based on spiegelmer and MoS2NF-GNR@AuNPs as sensing platform

Spiegelmers, mirror image L- RNA oligonucleotides, possesses high plasma stability and non-immunogenicity. Herein, a novel spiegelmer based impedimetric biosensor grafted with Au nanoparticles and molybdenum disulfide nanoflowers/graphene nanoribbons nanocomposite has been designed to detect hepcidin in spiked-in human serum sample. Firstly, molybdenum disulfide nanoflowers/graphene nanoribbons (MoS2NF-GNR) hybrid was drop-casted onto the FTO electrode followed by electro deposition of Au nanoparticles (AuNPs). Hepcidin specific thiolated spiegelmer was then immobilized on the MoS2NF-GNR@AuNPs for hepcidin detection. Electrochemical impedance spectroscopy was used to assess the performance of the sensing platform based on the variation of charge transfer resistance (ΔRct) relative to the Fe(CN)64−/3- electrochemical probe in the presence of hepcidin. The impedance signals were recorded at the frequency range of 10−1 to 105 Hz and potential was set as 0.18 V. Under optimized conditions, the limit of detection of spiegelmer based sensor for hepcidin was 0.173 pgmL−1 within a wide linear range of 0.005–10 ngmL−1. The biosensor possesses selectivity, acceptable reproducibility with RSD as 4.76% and stability for up to 20 days. The satisfactory recovery result (89.8–103.1 %) in human serum indicates that the sensor has applicability in clinical monitoring of hepcidin.

A Cu(II)-MOF Based on a Propargyl Carbamate-Functionalized Isophthalate Ligand as Nitrite Electrochemical Sensor.

This paper investigates the electrochemical properties of a new Cu(II)-based metal-organic framework (MOF). Noted as Cu-YBDC, it is built upon a linker containing the propargyl carbamate functionality and immobilized on a glassy carbon electrode by drop-casting (GC/Cu-YBDC). Afterward, GC/Cu-YBDC was treated with HAuCl4 and the direct electro-deposition of Au nanoparticles was carried at 0.05 V for 600 s (GC/Au/Cu-YBDC). The performance of both electrodes towards nitrite oxidation was tested and it was found that GC/Au/Cu-YBDC exhibited a better electrocatalytic behavior toward the oxidation of nitrite than GC/Cu-YBDC with enhanced catalytic currents and a reduced nitrite overpotential from 1.20 to 0.90 V. Additionally GC/Au/Cu-YBDC showed a low limit of detection (5.0 μM), an ultrafast response time (<2 s), and a wide linear range of up to 8 mM in neutral pH.

A novel label-free electrochemical immunesensor for ultrasensitive detection of LT toxin using prussian blue@gold nanoparticles composite as a signal amplification

In the current study, a novel electrochemical label-free immunosensor is proposed for sensitive detection of heat-labile enterotoxin (LT) from Escherichia coli. Firstly, a glassy carbon electrode (GCE) was modified by a mixture containing reduced graphene oxide/room temperature ionic liquid (rGO/RTIL) composite. Then, simultaneous electrodeposition of prussian blue and gold nanoparticles led to formation of prussian blue@gold nanoparticles (PB@GNPs) composite on the electrode surface. The modified electrode was characterized by field emission scanning electron microscopy (FE-SEM), energy dispersive spectroscopy (EDS), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) techniques. After immobilization of anti-LT and blocking the unreacted sites with BSA (bovine serum albumin), the analytical performance of the proposed immunosensor was evaluated under optimal conditions (i.e. optimal pH, incubation time and temperature of incubation). Square wave voltammetry (SWV) was used to determine different concentrations of the LT antigen. The linear dynamic range of the proposed immunosensor was from 0.01 to 50 µg/mL and the detection limit of the immunosensor was found to be 0.0023 µg/mL. An acceptable selectivity in the real sample, long-term stability and goodreproducibility made the fabricated immunosensor a good candidate for detecting LT.

MnO2@Reduced Graphene Oxide Nanocomposite-Based Electrochemical Sensor for the Simultaneous Determination of Trace Cd(II), Zn(II) and Cu(II) in Water Samples.

The rapid detection of trace metals is one of the most important aspect in achieving environmental monitoring and protection. Electrochemical sensors remain a key solution for rapid detection of heavy metals in environmental water matrices. This paper reports the fabrication of an electrochemical sensor obtained by the simultaneous electrodeposition of MnO2 nanoparticles and RGO nanosheets on the surface of a glassy carbon electrode. The successful electrodeposition was confirmed by the enhanced current response on the cyclic voltammograms. The XRD, HR-SEM/EDX, TEM, FTIR, and BET characterization confirmed the successful synthesis of MnO2 nanoparticles, RGO nanosheets, and MnO2@RGO nanocomposite. The electrochemical studies results revealed that MnO2@RGO@GCE nanocomposite considerably improved the current response on the detection of Zn(II), Cd(II) and Cu(II) ions in surface water. These remarkable improvements were due to the interaction between MnO2 nanomaterials and RGO nanosheets. Moreover, the modified sensor electrode portrayed high sensitivity, reproducibility, and stability on the simultaneous determination of Zn(II), Cd(II), and Cu(II) ions. The detection limits of (S/N = 3) ranged from 0.002–0.015 μg L−1 for the simultaneous detection of Zn(II), Cd(II), and Cu(II) ions. The results show that MnO2@RGO nanocomposite can be successfully used for the early detection of heavy metals with higher sensitivity in water sample analysis.

This makes the mechanistic interpretation sometimes contradictory, preventing unambiguous predictions about the nature and structure of the electrodeposited material. Herein, the complexity of Ni nanoparticles (NPs) electrodeposition on indium tin oxide (ITO) is unraveled operando and at a single entity NP level by optical microscopy correlated to ex situ SEM imaging. Our correlative approach allows differentiating the dynamics of formation of two different NP populations, metallic Ni and Ni(OH)2 with a &lt;25 nm limit of detection, their formation being ruled by the competition between Ni2+ and water reduction. At the single NP level this results in a self‐terminated growth, an information which is most often hidden in ensemble averaged measurements.

Facile electrodeposition of Mn3O4 nanoparticles on wood-derived porous carbon for high-performance asymmetric supercapacitor

In this study, we report a facile method of anchoring Mn3O4 nanoparticles on the surface of wood-derived porous carbon via electrodeposition to enhance the specific capacitance and cycling performance of electrodes for supercapacitors. Owing to the synergistic effect of the natural tubular porous structures of wood-derived porous carbon and the high theoretical specific capacitance of Mn3O4, wood-derived porous carbon-Mn3O4 composites (WPC-Mn3O4) deliver a high specific capacitance (315 F g−1 at the current density of 1 A g−1) and an outstanding cycling stability (91% of maximum capacitance after 10,000 cycles at 10 A g−1) in 1 M Na2SO4 electrolyte. Furthermore, the WPC-Mn3O4 and activated carbon (AC) assembled asymmetric supercapacitor (ASC) WPC-Mn3O4//AC delivers a high energy density of 34.85 Wh kg−1 at 700.0 W kg−1, demonstrating a high applicability in practical energy storage devices. The resultant WPC-Mn3O4 composites with excellent electrochemical properties are expected to be used as a potential cathode material for the development of high-performance supercapacitors.
