Fluorescence study of domain structure and lipid interaction of human apolipoproteins E3 and E4

Human apolipoprotein E (apoE) isoforms exhibit different conformational stabilities and lipid-binding properties that give rise to altered cholesterol metabolism among the isoforms. Using Trp-substituted mutations and site-directed fluorescence labeling, we made a comprehensive comparison of the conformational organization of the N- and C-terminal domains and lipid interactions between the apoE3 and apoE4 isoforms. Trp fluorescence measurements for selectively Trp-substituted variants of apoE isoforms demonstrated that apoE4 adopts less stable conformations in both the N- and C-terminal domains compared to apoE3. Consistent with this, the conformational reorganization of the N-terminal helix bundle occurs at lower guanidine hydrochloride concentration in apoE4 than in apoE3 as monitored by fluorescence resonance energy transfer (FRET) from Trp residues to acrylodan attached at the N-terminal helix. Upon binding of apoE3 and apoE4 variants to egg phosphatidylcholine small unilamellar vesicles, similar changes in Trp fluorescence or FRET efficiency were observed for the isoforms, indicating that the opening of the N-terminal helix bundle occurs similarly in apoE3 and apoE4. Introduction of mutations into the C-terminal domain of the apoE isoforms to prevent self-association and maintain the monomeric state resulted in great increase in the rate of binding of the C-terminal helices to a lipid surface. Overall, our results demonstrate that the different conformational organizations of the N- and C-terminal domains have a minor effect on the steady-state lipid-binding behavior of apoE3 and apoE4: rather, self-association property is a critical determinant in the kinetics of lipid binding through the C-terminal helices of apoE isoforms.

Surfactants, Salt, and pH Alter Nanoparticle-Model Cell Membrane Interactions

Due to their small size, nanoparticles have the ability to penetrate pulmonary and vascular tissue, and as a result, are classified as potential human carcinogens. On the other hand, nanoparticle insertion into targeted cells can play a key role in drug delivery and gene therapy applications, prompting a need to more thoroughly characterize nanoparticle/membrane interactions. Polystyrene nanoparticles with modifications in surface functionalization and detergent conditions remain monodisperse in a variety of aqueous solutions as measured by dynamic light scattering, but tend to aggregate in phosphate buffered saline at neutral pH potentially due to electrostatic screening effects. Calcein leakage assays with small unilamellar egg phosphatidylcholine vesicles were run to measure their interactions with nanoparticles and determine applicability of previous experiments performed with lipid monolayers at the air-water interface that modeled the outer leaflet of a cell membrane. In pure water, adding surfactant to detergent-free nanoparticle solutions increased the magnitude of membrane permeabilization compared to nanoparticles alone. In a buffered solution, the opposite was true; addition of surfactant decreased the nanoparticle induced membrane permeabilization. To better understand nanoparticle-detergent-membrane interactions, either nanoparticles or detergents were individually introduced to the vesicles, and following a time lapse, the other component was introduced. A model of detergent sequestration by the polystyrene nanoparticles explains these results and provides insight to the mechanism of vesicle leakage caused by surfactant-nanoparticle mixtures.

Fluorescence Analysis of the Lipid Binding-Induced Conformational Change of Apolipoprotein E4

Apolipoprotein (apo) E is thought to undergo conformational changes in the N-terminal helix bundle domain upon lipid binding, modulating its receptor binding activity. In this study, site-specific fluorescence labeling of the N-terminal (S94) and C-terminal (W264 or S290) helices in apoE4 by pyrene maleimide or acrylodan was employed to probe the conformational organization and lipid binding behavior of the N- and C-terminal domains. Guanidine denaturation experiments monitored by acrylodan fluorescence demonstrated the less organized, more solvent-exposed structure of the C-terminal helices compared to the N-terminal helix bundle. Pyrene excimer fluorescence together with gel filtration chromatography indicated that there are extensive intermolecular helix-helix contacts through the C-terminal helices of apoE4. Comparison of increases in pyrene fluorescence upon binding of pyrene-labeled apoE4 to egg phosphatidylcholine small unilamellar vesicles suggests a two-step lipid-binding process; apoE4 initially binds to a lipid surface through the C-terminal helices followed by the slower conformational reorganization of the N-terminal helix bundle domain. Consistent with this, fluorescence resonance energy transfer measurements from Trp residues to acrylodan attached at position 94 demonstrated that upon binding to the lipid surface, opening of the N-terminal helix bundle occurs at the same rate as the increase in pyrene fluorescence of the N-terminal domain. Such a two-step mechanism of lipid binding of apoE4 is likely to apply to mostly phospholipid-covered lipoproteins such as VLDL. However, monitoring pyrene fluorescence upon binding to HDL(3) suggests that not only apoE-lipid interactions but also protein-protein interactions are important for apoE4 binding to HDL(3).

Effects of the Core Lipid on the Energetics of Binding of ApoA-I to Model Lipoprotein Particles of Different Sizes

Interaction of apolipoproteins (apo) with lipid surfaces plays crucial roles in lipoprotein metabolism and cholesterol homeostasis. To elucidate the thermodynamics of binding of apoA-I to lipid, we used lipid emulsions composed of triolein (TO) and egg phosphatidylcholine (PC) as lipoprotein models. Determination of the level of binding of wild-type (WT) apoA-I and some deletion mutants to large (120 nm diameter; LEM) and small (35 nm diameter; SEM) emulsions indicated that N-terminal (residues 44-65) and C-terminal (residues 190-243 and 223-243) deletions have large effects on lipid interaction, whereas deletion of the central region (residues 123-166) has little effect. Substitution of amino acids at either L230 or L230, L233, and Y236 with proline residues also decreases the level of binding, indicating that an alpha-helix conformation in this C-terminal region is required for efficient lipid binding. Calorimetry showed that binding of WT apoA-I to SEM generates endothermic heat (DeltaH approximately 30 kcal/mol) in contrast to the exothermic heat (ca. -85 kcal/mol) generated upon binding to LEM and egg PC small unilamellar vesicles (SUV). This exothermic heat arises from an approximately 25% increase in alpha-helix content, and it drives the binding of apoA-I to LEM and SUV. There is a similar increase in alpha-helix content of apoA-I upon binding to either SEM or SUV, but the binding of apoA-I to SEM is an entropy-driven process. These results suggest that the presence of a core triglyceride modifies the highly curved SEM surface packing and thereby the thermodynamics of apoA-I binding in a manner that compensates for the exothermic heat generated by alpha-helix formation.

The structure and organization of synthetic putative membranous segments of ROMK1 channel in phospholipid membranes

The hydropathy plot of ROMK1, an inwardly rectifying K+ channel, suggests that the channel contains two transmembrane domains (M1 and M2) and a linker between them with significant homology to the H5 pore region of voltage-gated K+ channels. To gain structural information on the pore region of the ROMK1 channel, we used a spectrofluorimetric approach and characterized the structure, the organization state, and the ability of the putative membranous domains of the ROMK1 channel to self-assemble and coassemble within lipid membranes. Circular dichroism (CD) spectroscopy revealed that M1 and M2 adopt high alpha-helical structures in egg phosphatidylcholine small unilamellar vesicles and 40% trifluoroethanol (TFE)/water, whereas H5 is not alpha-helical in either egg phosphatidylcholine small unilamellar vesicles or 40% TFE/water. Binding experiments with 4-fluoro-7-nitrobenz-2-oxa-1,3-diazole (NBD)-labeled peptide demonstrated that all of the peptides bind to zwitterionic phospholipid membranes with partition coefficients on the order of 10(5) M-1. Tryptophan quenching experiments using brominated phospholipids revealed that M1 is dipped into the hydrophobic core of the membrane. Resonance energy transfer (RET) measurements between fluorescently labeled pairs of donor (NBD)/acceptor (rhodamine) peptides revealed that H5 and M2 can self-associate in their membrane-bound state, but M1 cannot. Moreover, the membrane-associated nonhelical H5 serving as a donor can coassemble with the alpha-helical M2 but not with M1, and M1 can coassemble with M2. No coassembly was observed between any of the segments and a membrane-embedded alpha-helical control peptide, pardaxin. The results are discussed in terms of their relevance to the proposed topology of the ROMK1 channel, and to general aspects of molecular recognition between membrane-bound polypeptides.

Lateral Organization of Pyrene-labeled Lipids in Bilayers as Determined from the Deviation from Equilibrium between Pyrene Monomers and Excimers

In lipid bilayers, pyrene and pyrene-labeled lipids form excimers in a concentration-dependent manner. The aromatic amine N, N-diethylaniline (DEA), which has a high membrane-to-medium partition coefficient, quenches the monomers only, and therefore it is expected that under conditions in which the monomers are in equilibrium with the excimers due to the mass law, the Stern-Volmer coefficient (Ksv) for monomers (M), defined as KM, should be identical to that of the excimer (E), defined as KE, and KE/KM = 1. 0. This is indeed the case for pyrene and pyrene valerate in egg phosphatidylcholine small unilamellar vesicles. However, for pyrene decanoate and pyrene dodecanoate in these vesicles, and for N-[12-(1-pyrenyl)dodecanoyl]sphingosylphosphocholine in a matrix of either N-stearoyl sphingosylphosphocholine or 1-palmitoyl-2-oleoyl phosphatidylcholine, KE < KM. This can be explained either by the existence of (a) two subpopulations of excimers, one in fast equilibrium with the monomers and the other, related to ground-state protoaggregates of pyrene lipids; (b) two monomer subpopulations where part of M cannot be quenched by DEA; or (c) two monomer subpopulations, both quenched by DEA, but only one of which produces excimers. The good agreement between the photophysical processes determined by steady state and time-resolved measurements supports the third explanation for the bilayers containing pyrene phospholipids. It also suggests that the main factors determining the immiscibility of pyrene lipids in phospholipid bilayers are the temperature, the difference in the gel-to-liquid-crystalline phase transition temperature (deltaTm) between the matrix and the pyrene lipid, and the structural differences between the matrix lipid and the pyrene-labeled lipid. These results indicate that the KE/KM ratio can serve as a very sensitive tool to quantify isothermal microscopic immiscibility in membranes. This novel approach has the following advantages: applicability to fluid phase immiscibility, requirement of a relatively low mol fraction of pyrene lipids, and conceivably, applicability to biological membranes.

Cell-free transfer of cholesterol from lysosomes to phospholipid vesicles.

The objective of this work was to develop a cell-free system for studying the transfer of cholesterol from lysosomes to membrane acceptor particles. The methods involved: 1) loading of CHO cells at 15 degrees C with [3H]cholesteryl oleate-reconstituted LDL, such that it accumulated undegraded in endosomes; 2) homogenization of cells, followed by preparation of an endosome-lysosome donor fraction; 3) incubation of the donor fraction at 37 degrees C in a defined cytosol-like medium containing acceptor particles of egg phosphatidylcholine small unilamellar vesicles (PC-SUV); and 4) measurement of cholesteryl oleate (CO) hydrolysis and transfer of the resulting free cholesterol (FC) to vesicles. During cell-free incubation, LDL-loaded endosomes fused with lysosomes leading to the lysosomal hydrolysis of LDL cholesteryl ester. Maximal hydrolysis of approximately 50% was achieved in 4-8 h. This hydrolysis was inhibited by lysosomotropic agents, proton ionophores, or removal of ATP and GTP from the medium, indicating that it took place in sealed lysosomes. In the absence of PC-SUV, the release of LDL-derived FC from lysosomes was "< or =" 10%/8 h. This was increased to a maximum of 25-30%/8 h at 3 mg/ml of PC-SUV. In contrast, the release of undegraded CO was 5-15%/8 h and not stimulated by PC-SUV, suggesting that the transfer of FC to PC-SUV was selective and not due to the uncontrolled release of lysosomal contents. In comparisons between CHO-K1 cells and sterol transport-defective CHO(2-2) cells, lysosomes from the latter cell were 35% less efficient as donors of cholesterol for transfer to egg phosphatidylcholine small unilamellar vesicles, indicating that these methods reproduce an important aspect of sterol trafficking in cells. In addition, this result suggests that the mutation in CHO(2-2) has a direct effect on the lysosomes of these cells.

Kinetics of incorporation of porphyrins into small unilamellar vesicles

The kinetics of hematoporphyrin or deuteroporphyrin incorporation in egg phosphatidylcholine small unilamellar vesicles have been investigated by fluorescence stopped-flow measurements. The processes can be described by a fast equilibrium. The on-rate constant is nearly diffusion controlled regardless of the compound used and the pH. The affinity of these porphyrins for the vesicles is merely governed by the exit rate which depends on the structure of the porphyrin and on its charge determined by pH.

The interaction of Saccharomyces cerevisiae trehalase with membranes

Plasma membranes isolated from cells of Saccharomyces cerevisiae previously submitted to a heat-shock showed a 10-fold increase in membrane-bound trehalase activity. Trehalase was purified to a high specific activity and was shown to be inhibited by glucose 6-phosphate and by the addition of a neutral phospholipid-like surfactant. Purified trehalase binds spontaneously to egg phosphatidylcholine small unilamellar vesicles, when in its active, phosphorylated form. When the enzyme was treated with alkaline phosphatase no binding was observed. The significance of this reversible binding for the control of trehalose metabolism in yeast cells is still unknown.

Differential susceptibility of phosphatidylcholine small unilamellar vesicles to phospholipases A 2, C and D in the presence of membrane active peptides

Activities of phospholipases C and D along with A 2 were followed on egg phosphatidylcholine small unilamellar vesicles in the presence of membrane active peptides melittin, gramicidin S and alamethicin. Decrease in the activity of phospholiapse C and D and enhancement of phospholipses A 2 activity suggest that these enzymes are sensitive to alterations in the lipid packing in the membranes in the presence of these peptides. Phospholipase C and D, which have not been used to study peptide - membrane interactons, have potential use in studying membrane perturbations, since their activities are very sensitive to lipid packing.

Membrane proteins exposed on the external side of the intestinal brush‐border membrane have fusogenic properties

The intestinal brush-border membrane contains one or several membrane proteins that mediate fusion and/or aggregation of small unilamellar egg phosphatidylcholine vesicles. The fusion is accompanied by a partial loss of vesicle contents. Proteolytic treatment of the brush-border membrane with proteinase K abolishes the fusogenic property. This finding suggests that the fusogenic activity is associated with a membrane protein exposed on the external or luminal side of the brush-border membrane. Activation of intrinsic proteinases of the brush-border membrane liberates water-soluble proteins (supernate proteins). These proteins behave in an analogous way to intact brush-border membrane vesicles; they induce fusion of egg phosphatidylcholine vesicles and render the egg phosphatidylcholine bilayer permeable to ions and small molecules (Mr less than or equal to 5000). Furthermore, supernate proteins mediate phosphatidylcholine and cholesterol exchange between two populations of small, unilamellar phospholipid vesicles. Supernate proteins are fractionated on Sephadex G-75 SF yielding three protein peaks of apparent Mr greater than or equal to 70,000, Mr = 22,000 and Mr = 11,500. All three protein fractions show similar phosphatidylcholine-exchange activity, but they differ in their effects on the stability of egg phosphatidylcholine vesicles. The protein fraction with an apparent Mr greater than or equal to 70,000 has the highest fusogenic activity while the protein fraction of apparent Mr = 11,500 appears to be most effective in rendering the egg phosphatidylcholine bilayer permeable.

Possible eye-irritant test using polysaccharide-coated liposomes as a corneal epithelium model.

An alternative nonanimal test to the Draize test to evaluate the eye irritancy of surfactants has been newly developed. The physicochemical lysis of liposomal membranes by various surfactants, such as sodium dodecyl sulfate (SDS), dodecyldimethylbenzylammonium chloride (BTC), tetradecyldimethylbenzylammonium chloride (Sanisol M-100), benzyldimethy1-2- (2-p-1, 1, 3, 3-tetramethylbutylphenoxy) ethoxyethylammonium chloride (Hyamine 1622), dodecanoyldiethanolamide (LDE), and polyoxyethylene (n=21) -sorbitan monostearate (Tween 80), was correlated with the results of the Draize test with the same surfactants. Egg phosphatidylcholine small unilamellar vesicles (SUVs) coated with O-palmitoylamylopectinsulfonic acid were employed as a model of the human corneal epithelium. The lysis of liposomes with the surfactants was monitored quantitatively by following the release of carboxyfluorescein (CF) from the interior water phase of the liposomes suspended in an aqueous medium containing almost the same salt composition as that in the human tear film. The order of efficiencies of these surfactants in perturbing the liposomal membrane was Sanisol M-100 > Hyamine 1622 >BTC> (Tween 80>) LDE>SDS. With the exception of Tween 80, whose critical micelle concentration (cmc) is relatively low (9.6 × 10-6M) compared with those of the other surfactants (approximately 5-7× 10-4M), a reliable correlation was found between the results of animal and nonanimal tests.

Spectrofluorimetric study of porphyrin incorporation into membrane models - evidence for pH effects

The effects of hydrophobicity and charges of dicarboxylic porphyrins upon their interactions with membrane model systems are investigated. Four protonation steps are evidenced from fluorescence emission studies of hematoporphyrin IX and its more hydrophobic parent compound lacking of alcoholic chain, deuteroporphyrin IX. They are attributed to the successive protonations of the inner nitrogens of the porphyrin cycle (p K = 4.7 and 2.9 for hematoporphyrin and 4.4 and 2.7 for deuteroporphyrin) and successive deprotonations of propionic groups (p K ≈ 5.0 and 5.5 for hematoporphyrin and 5.4 and 6.0 for deuteroporphyrin). These porphyrins, as well as their dimethyl ester forms, are shown to incorporate as monomers into the hydrophobic bilayer of egg phosphatidylcholine small unilamellar vesicles, although the esterified forms are highly aggregated in aqueous solutions. In the case of the non-esterified forms, the incorporation of the porphyrins into the lipidic bilayer is reversible and strongly pH-dependent. A theoretical model is presented which takes into account the various protonation steps and the partition equilibria of the porphyrin between the vesicle lipidic phase and the water medium. The neutral form of the porphyrin (i.e., carboxylic groups protonated) presents the higher affinity, with constants of K ≈ 2 · 10 5 and K ≈ 6 · 10 6 M −1 (relative to lipid concentration) for hematoporphyrin and deuteroporphyrin, respectively. Protonation of one inner nitrogen leading to the monocationic form is sufficient to prevent incorporation into the hydrophobic bilayer. On the other hand, deprotonation of the peripheral propionic chains leading to anionic forms is less effective. These interactions between vesicles and porphyrins lead to shifts of the apparent p K of nitrogens and carboxylic groups, the latter one being now in the range of physiological pH. These results are discussed with regards to the hypothesis of a possible role of pH in the preferential uptake of porphyrins by tumors.

Role of apolipoproteins in cellular cholesterol efflux

The effects of serum apolipoproteins, particle size and concentration on the effectiveness of phosphatidylcholine (PC)-containing acceptor particles in causing release of cholesterol from cells growing in culture have been investigated. The acceptor particles were prepared by detergent-dialysis procedures and were either egg PC small unilamellar vesicles (SUV) or discoidal complexes of egg PC with apoproteins from human high-density lipoprotein (HDL). Gel filtration chromatography was employed to isolate particles of defined composition and size. The half-times (t 1 2 ) for the unidirectional efflux of cholesterol from cells prelabeled with [ 3H]cholesterol were measured as a function of acceptor PC concentration in the extracellular medium. HDL apolipoprotein-egg PC discoidal complexes at 100 μg PC/ml gave the following (t 1 2 ) values when incubated with rat Fu5AH hepatoma, human HepG2 hepatoma, human GM3468 skin fibroblast, L-cell and mouse J774 macrophage-tumor cells: 11 ± 2, 22 ± 5, 84 ± 18, 17 ± 2 and 32± 6 h, respectively. Equivalent experiments using purified apolipoprotein A-I or the total apolipoprotein C fraction to form the egg PC complexes showed that the (t 1 2 ) values for the hepatoma cells were unaltered. However, with the fibroblasts, L-cells and J774 macrophages, the apolipoprotein C complexes gave significantly longer (t 1 2 ) than complexes of egg PC with either apolipoprotein A-I or HDL apolipoprotein which gave the same (t 1 2 ) . An analysis based on the theory of fast coagulation of colloid particles to describe collisions between desorbed cholesterol molecules and acceptor particles predicts that the dependence of (t 1 2 ) for cholesterol efflux from a given cell to different acceptors should be normalized when the extracellular level of acceptors is expressed in terms of the product of the radius of the particle times the number concentration of acceptor particles. The decrease in (t 1 2 ) for cholesterol efflux from fibroblasts when the egg PC acceptor was changed from an SUV to an apolipoprotein HDL discoidal complex is consistent with the above concepts. The primary effect of the apolipoproteins in promoting cellular cholesterol efflux seems to be the solubilization of PC so that the PC is present in the extracellular medium as many small particles.

Effect of the concentration and the molecular structure of phosphonic acid dialkyl esters on the kinetics of Pr 3+ permeation through egg phosphatidylcholine small unilamellar vesicles

The dependence of the Pr 3+ permeation kinetics in unilamellar vesicles of egg phosphatidylcholine on the incorporation of phosphonic acid dialkyl esters in the membrane was studied by 31P- and 1H-NMR. The phosphonic acid dialkyl esters cause drastic changes of the permeation half-time t 1 2 of the Pr 3+ ions into the inner vesicle compartment. The ion permeation induced by the esters was interpreted as due to the formation of metastable pores. The permeation half-time t 1 2 depends on the ester concentration in the membrane and on temperature. The threshold concentration of the ester necessary for a drastic decrease in permeation half-time t 1 2 reflects certain structural differences of the esters. The results suggest a correlation between the effective molecular shape of the phosphonic acid esters and the ion permeation. The esters are mainly incorporated in the outer vesicle monolayer. This could be understood because of the positive molecular asymmetry of the phosphonic acid dialkyl esters. The permeation half-time t 1 2 of Pr 3+ ions out of the vesicles is much larger than the half-time t 1 2 for the opposite process.

Partition of amphiphilic molecules into phospholipid vesicles and human erythrocyte ghosts: measurements by ultraviolet difference spectroscopy.

Molar partition coefficients for chlorpromazine and methochlorpromazine between phospholipid vesicles or human erythrocyte ghosts and buffer are determined by ultraviolet difference spectroscopy. The partition coefficients between small unilamellar egg phosphatidylcholine vesicles and buffer at pH 7.4 are 4.4 X 10(5) for chlorpromazine and 0.8 X 10(5) for methochlorpromazine, determined with 10 microM amphiphile. An increase in the partition of chlorpromazine into vesicles is seen as the pH is increased to the pKa of chlorpromazine at 9.2. Chlorpromazine also partitions preferentially into fluid-phase phospholipid compared to solid-phase phospholipid. Molar partition coefficients between unsealed human erythrocyte ghosts and buffer at pH 8.0 with 10 microM amphiphile are determined to be 6.5 X 10(5) for chlorpromazine and 2.5 X 10(5) for methochlorpromazine. Difference spectroscopy is an equilibrium technique that does not require separation of bound from free amphiphile, as do many other methods of determining membrane-buffer partition coefficients. This method is useful for any amphiphile that has an appreciable absorbance below its critical micelle concentration and whose absorbance is sensitive to environment.

On the size-dependent disintegration of small unilamellar phosphatidylcholine vesicles in rat plasma. Evidence of complete loss of vesicle structure.

The destruction of small unilamellar egg phosphatidylcholine vesicles in rat plasma was monitored by measuring release of encapsulated 125I-poly(vinylpyrrolidone) or carboxyfluorescein and by determining transfer of radiolabelled phosphatidylcholine to plasma lipoproteins by means of gel filtration. The susceptibility of the vesicles to the destructive action of plasma increased with decreasing vesicle size, as observed by incubating plasma with individual fractions constituting the small-vesicle peak on Sepharose CL-2B. This results in selective destruction of small vesicles when heterogeneous vesicle populations are incubated with plasma. Samples of homogeneous vesicle populations were incubated with a wide range of plasma concentrations, which resulted in extents of solute and phospholipid release ranging from 10 to 90%. When the extents of solute release were plotted against the extents of lipid release a linear, virtually 1:1, relationship was found, for both carboxyfluorescein and poly(vinylpyrrolidone) as the solute. This suggests that the release of solutes from small unilamellar phosphatidylcholine vesicles as a result of their interaction with plasma (lipo)proteins involves the total destruction of a fraction of the vesicles, the magnitude of which is determined by the vesicle: plasma ratio. Our results argue against a previously presented view suggesting that the interaction between such vesicles and plasma results in the formation of pores through which encapsulated solutes diffuse at Mr-dependent rates [Kirby & Gregoriadis (1981) Biochem. J. 199, 251-254]. The discrepancies between the two studies in observations as well as in interpretation are discussed.

Incorporation of the transmembrane hydrophobic domain of glycophorin into small unilamellar phospholipid vesicles Ion Flux studies

Human erythrocyte glycophorin is one of the best characterized integral membrane proteins. Reconstitution of the membrane-spanning hydrophobic segment of glycophorin (the tryptic insoluble peptide released when glycophorin is treated with trypsin) with liposomes results in the production of freeze-fracture intrabilayer particles of 80 Å diameter (Segrest, J.P., Gulik-Krzywicki, T. and Sardet, C. (1974) Proc. Natl. Acad. Sci. U.S.A. 71, 3294–3298), with particles appearing at or above a tryptic insoluble peptide concentration of 4 mmol per mol phosphatidylcholine. In the present study, increasing concentrations of tryptic insoluble peptide were added to sonicated small unilamellar egg phosphatidylcholine vesicles and the rate of efflux of 22Na + was examined by rapid (30 s) gel filtration on Sephadex G-50. Below a concentation of 3–5 mmol tryptic insoluble peptide/mol phosphatidylcholine, 22Na + efflux occurs at a constant slow rate at given tryptic insoluble peptide concentrations. Above a concentration of 3–5 mM, the rate of efflux is biphasic at given tryptic insoluble peptide concentrations, exhibiting both an initial fast and a subsequent slow component. On the basis of graphic and computer curve-fitting analysis, with increasing tryptic insoluble peptide concentration, the rate of the slow component reaches a plateau at a tryptic insoluble peptide concentration of 3–5 mM and remains essentially constant until much higher concentrations are reached; the fast component increases linearly with increasing tryptic insoluble peptide concentration well beyond 5 mM. The most consistent interpretation of this data is as follows. The slow 22Na + efflux component is due to perturbations of small unilamellar vesicle integrity by tryptic insoluble peptide monomers. At a tryptic insoluble peptide concentration of 3–5 mmol/mol, a critical concentration is reached following which there is intrabilayer tryptic insoluble peptide self-association. The fast 22Na + efflux component is due to the increasing presence of tryptic insoluble peptide self-associated multimers the 80-Å particles seen by freeze-fracture electron microscopy) which results in a significantly larger bilayer defect than do tryptic insoluble peptide monomers. The failure of complete saturation of efflux by the fast component is ascribed to the presence of two populations of small unilamellar vesicles, some of which contain tryptic insoluble peptide multimers and some of which do not. Addition of cholesterol to the tryptic insoluble peptide/phosphatidylcholine vesicles decreases the rate of 22Na + efflux by inhibiting primarily the fast component. Freeze-fracture electron microscopy indicates that the presence of cholesterol has no effect on the size, number or distribution of 80-Å intra-bilayer particles in the tryptic insoluble peptide/phosphatidylcholine vesicles. These results are consistent with a mechanism to explain the fast Na + efflux component involving protein-lipid boundary perturbations. Efflux of 45Ca 2+ from phosphatidylcholine vesicles is also enhanced by incorporation of tryptic insoluble peptide, but only if divalent cations (Ca 2+ or Mg 2+) are present in the external bathing media as well as inside the sonicated vesicles. If monovalent Na + only is present in the bathing media no 45Ca 2+ efflux is seen. Under conditions where 45Ca 2+ efflux is seen, both a fast and a slow component are present, although both appear lower than corresponding rate constants for 22Na + efflux. These results suggest a coordinated mechanism for ion efflux induced by tryptic insoluble peptide and, together with the 22Na + efflux studies, may have mechanistic implications for the transbilayer phospholipid exchange (flip-flop) suggesed to be induced at glycophorin/phospholipid interfaces (de Kruiff, B., van Zoelen, E.J.J. and van Deenen, L.L.M. (1978) Biochim. Biophys. Acta 509, 537–542).
