Rosa roxburghii trattresidue: A novel feed resource for cattle indicated by the non-deleterious performance and blood metabolites.

Rosa roxburghii tratt residue (RRTR) is a regional and uncommon byproduct in Guizhou. Little information is available on whether RRTR can be used as feed for ruminants. In this study, the feasibility of using RRTR as a new feed resource for cattle was investigated by chemical composition analysis, in vitro gas production (Trial 1) and animal feeding experiments (Trial 2). In trial 1, compared to the commonly used fruit residues, RRTR had a proximate nutrient level; the in vitro gas production curve and dynamics fell within the normal range. In trial 2, 16 cattle were allocated to the control and treatment groups, with 8 replicates of 1 cattle each. Cattle in the control group were fed a basal diet without RRTR, while those in the treatment group were fed a diet containing 30% RRTR to replace 30% whole corn silage in basal diet. Feeding RRTR had little effect on the growth performance of the control and treatment cattle (P > 0.05). The feed-to-gain ratio was greater in the treatment group than in the control group throughout the trial period (P < 0.05). The plasma urea levels in the treatment group were lower (P < 0.05) than that in control group, and the levels of other plasma biochemical metabolites were not different between the two groups of cattle (P > 0.05). The in vivo rumen fermentation parameters did not differ between the control and treatment groups (P > 0.05). Our findings indicate that RRTR has a nutritional profile (crude protein, neutral detergent fiber, acid detergent fiber, and crude fiber) similar to that of commonly used fruit residues (such as apple, pineapple, and citrus residue et al.); improves plasma protein utilization efficiency; and has no negative impact on growth performance, albeit with limited effects on feed conversion; blood metabolites, and rumen fermentation parameters in cattle. Accordingly, we conclude, based on the above-mentioned result, that RRTR can serve as a novel feed alternative resource when considering the affordability and as a practical choice for low-cost diets for cattle.

Effects of different forms of amino acid supplementation on the performance and intestinal barrier function of laying hens fed a low-protein diet

The aim of this study was to investigate the effects of low-protein diets and the sustained release of synthetic amino acids (AA) on the performance, intestinal barrier function and nitrogen excretion of laying hens. Two hundred eighty-eight 39-week-old Hyline brown laying hens of were randomly divided into 3 groups with 8 replicates per group. The crude protein level in the control group (CON) was 16%, the crude protein levels in the crystal AA supplement group (LCP-CAA) and microencapsulated AA group (LCP-MAA) were both 13%, and the AA levels in the LCP-CAA and LCP-MAA groups were consistent with that in the CON group. The experiment lasted 12 wk, and production performance was assessed weekly. The FCR and ADFI values were significantly greater for the LCP-CAA group than for the CON and LCP-MAA groups (P < 0.05). Two hours after feeding, His levels were significantly greater in the LCP-CAA group than in the LCP-MAA group (P < 0.05); 4 h after feeding, the contents of Met, Thr, Leu and Val were significantly greater in blood from the LCP-MAA group (P < 0.05); 6 h after feeding, Trp, Ile and Arg levels were significantly greater in the LCP-MAA group (P < 0.05). The chylase content significantly decreased in the duodenum of the LCP-CAA group (P < 0.05), and the chylase and trypsin were contents increased in the ileum of the LCP-MAA group (P < 0.05). In the LCP-MAA group, significantly increased mRNA expression levels of Occludin, ZO-1 in duodenum; Occludin, ZO-1, y+LAT1 in jejunum; and ZO-1 in ileum were detected at 8 and 12 weeks (P < 0.05). The fecal nitrogen content significantly decreased in the low protein diet group (P < 0.01). In conclusion, reducing dietary crude protein levels and supplementing with microencapsulated AAs can improve intestinal barrier function, promote digestive enzyme secretion, increase the expression of AA transporters, improve dietary protein utilization efficiency, and reduce nitrogen emission in laying hens.

Decreased Amino Acid Transporter LAT2 Is the Main Determinant of Impaired Protein Utilization During Aging

As the global demographic shifts toward an aging population, understanding the efficiency of protein utilization in older adults becomes crucial. Our study explores the intricate relationship between protein intake and aging, with a focus on precision nutrition for older people. Through a meta-analysis, we confirm a decline in protein-utilization capacity in older individuals and examine the different contributions of plant and animal protein. In experiments involving mice of different ages, older mice exhibited decreases in the biological utilization of four proteins (casein, beef protein, soy protein, and gluten), particularly casein. In subsequent research, casein was studied as a key protein. A decline in gastric digestion function was observed through peptidomics and the examination of pepsin levels using casein. Nevertheless, this decline did not significantly affect the overall protein digestion during the aging process. The combined application of targeted amino acid metabolomics identified abnormal absorption of amino acids as the underlying cause of decreased protein utilization during aging, particularly emphasizing a reduction in branched-chain amino acids (BCAAs) in older mice. Delving deeper into the proteomics of the intestinal protein digestion and absorption pathway, a reduction of over 60% in large neutral amino acid transporter 2 (LAT2) protein expression was observed in both older humans and aged mice. The reduction in LAT2 protein was found to be a key factor influencing the diminished BCAA availability. Overall, our study establishes the significance of amino acid absorption through LAT2 in protein utilization during aging and offers a new theoretical foundation for improving protein utilization in the older adults.

Dietary CP and digestion kinetics influence BW loss, litter weight gain, and reproduction by affecting postprandial amino acid metabolism in lactating sows

To avoid a high body protein mobilization in modern lean sows during lactation, an adequate dietary amino acid (AA) supply and an efficient AA utilization are crucial. This study evaluated the effects of dietary CP and in vitro protein digestion kinetics on changes in sow body condition, litter weight gain, milk composition, blood metabolites, protein utilization efficiency and subsequent reproductive performance. We hypothesized that a slower digestion of dietary protein would improve AA availability and utilization. In total, 110 multiparous sows were fed one of four lactation diets in a 2 × 2 factorial design, with two CP concentrations: 140 g/kg vs 180 g/kg, and two protein digestion kinetics, expressed as a percentage of slow protein (in vitro degradation between 30 and 240 min): 8 vs 16% of total protein. Feeding sows the high CP diets reduced sow weight loss (Δ = 7.6 kg, P < 0.01), estimated body fat loss (Δ = 2.6 kg, P = 0.02), and estimated body protein loss (Δ = 1.0 kg, P = 0.08), but only at a high percentage of slow protein. A higher percentage of slow protein increased litter weight gain throughout lactation (Δ = 2.6 kg, P = 0.04) regardless of CP concentrations, whereas a higher CP only increased litter weight gain during week 3 of lactation (Δ = 1.2 kg, P = 0.01). On Day 15 postfarrowing, serial blood samples were taken from a subsample of sows fed with the high CP diets. In these sows, a high percentage of slow protein resulted in higher plasma AA concentrations at 150 and 180 min after feeding (Δ = 0.89, P = 0.02, Δ = 0.78, P = 0.03, mmol/L, respectively) and lower increases in urea at 90 and 120 min after feeding (Δ = 0.67, P = 0.04, Δ = 0.70, P = 0.03, mmol/L, respectively). The higher dietary CP concentration increased total nitrogen loss to the environment (Δ = 604 g, P < 0.01) with a reduction of protein efficiency (Δ = 14.8%, P < 0.01). In the next farrowing, a higher percentage of slow protein increased subsequent liveborn litter size (Δ = 0.7, P < 0.05). In conclusion, feeding sows with a high dietary CP concentration alleviated maternal weight loss during lactation when the dietary protein digestion rate was slower, but lowered protein efficiency. A slower protein digestion improved litter weight gain, possibly by reducing AA oxidation and improving plasma AA availability, thus, improving protein efficiency.

145 Performance, postweaning diarrhea, and apparent total tract nutrient digestibility responses of weanling pigs to nucleotide supplementation of low-protein diets

Abstract Feeding low protein (LP) diets to nursery pigs has been reported to improve intestinal health, reduce incidence of post-weaning diarrhea (PWD) and nitrogen excretion. However, these benefits are often undermined by significant reduction in growth performance. This study evaluated the effects of low protein diets supplemented with dietary nucleotides on growth performance, post weaning diarrhea and nitrogen utilization. A total of 210 piglets (Duroc × Landrace × Yorkshire) were weaned at 21 d of age and allowed a 3-d adaptation to a common solid post weaning diet. At 24 d old, pigs were reweighed (6.02 ± 0.05 kg) and allocated to 5 dietary treatments in a completely randomized block design to give 7 replicates per treatment (n = 6 piglets per replicate). The 5 dietary treatments included i) a high protein positive control diet (PC) with 24% crude protein (CP); ii) a low protein negative control (NC) with 16% CP; iii) NC with 0.1% dietary nucleotide inclusion (NC01); iv) NC with 0.3% dietary nucleotide inclusion (NC03); and v) NC with 0.9% dietary nucleotide inclusion (NC09). All diets were corn-soybean meal based, supplemented with lysine, methionine, tryptophan, and threonine, with all low protein diets (LP) additionally fortified with crystalline isoleucine, and valine to meet the ideal amino acid requirements. Diets were provided ad libitum for 35 d and weekly feed intake (FI) and body weight (BW) were measured. On d 35, blood and fecal samples were collected to determine serum metabolites and nutrient digestibility respectively. Data were analyzed by PROC GLM and orthogonal polynomial contrast of SAS. Comparison between positive and negative control diets and nucleotide levels were performed with orthogonal contrast analysis. Relative to PC, NC diet had decreased overall average daily gain (343.5 vs. 305.5 g/d), incidence of PWD (2.5 vs. 1.2 fecal consistency score), blood urea nitrogen (BUN; 11.3 vs. 3.4 mg/dL) and total tract digestibility of energy (81.3 vs. 80.1 %; P &amp;lt; 0.05, P &amp;lt; 0.05, P &amp;lt; 0.0001 and P &amp;lt; 0.1, respectively). Relative to PC, the NC01, NC03 and NC09 treatments had comparable average daily gain (P &amp;gt; 0.05) while maintaining reduced (P &amp;lt; 0.05) BUN and incidence of PWD. Total tract digestibility of dry matter (83.1 vs. 82.1%) and energy (81.3 vs. 80.1%) were similar between PC and NC09 treatments (P &amp;gt; 0.05). Crude protein reduction decreased serum glutathione (P &amp;lt; 0.05), but without an effect of nucleotide supplementation. Serum glucose and insulin concentrations were not different across treatments (P &amp;gt; 0.05). Results suggest that feeding LP diets supplemented with dietary nucleotide after weaning can increase protein utilization efficiency, reduce incidence of PWD while partially ameliorating the negative effects of LP diets on growth performance.

Maintenance requirement and growth utilization efficiency of digestible methionine and cysteine determined for striped catfish (Pangasianodon hypophthalmus) using ration level technique

Knowledge of the amino acid (AA) requirements for striped catfish (Pangasianodon hypophthalmus) is very limited. The main objective of the study was to determine maintenance requirements and utilization of digestible methonine (Met) and other essential amino acids (EAA) for growth in juvenile striped catfish. To evaluate the objective, a series of three different trials were carried out. Initially, a 28-d feed deprivation study showed an allometric relationship between energy, protein, or AA losses (y) and geometric body weight (BW), where the relationship was expressed as a power function (y = aBWb) with the exponent values (b) being 0.84 for energy, 0.83 for protein, and 0.81–0.90 for various EAAs. Next, a digestibility trial was carried out to determine apparent digestibility coefficients of energy, protein and AA from the ingredients, including: fish meal, soybean meal, wheat bran, regular and defatted rice bran. These values were then used in the final trial to formulate two diets on a digestible AA basis with similar crude protein (31%), energy (17.1 kJ g−1) and AA composition - except for digestible Met and Met+Cys, which were 0.47% and 0.90% for Diet A, and 0.61% and 1.05% for Diet B. The diets were fed to quadruplicate groups of fish (∼10 g initial weight; 20 fish per tank) at increasing ration levels (∼ 35%, 55%, 77%, 93% and 100% apparent satiation) for 64 days. One group was deprived of feed for 28 days. The results showed that feeding catfish at increasing ration levels significantly improves the growth performance in both dietary treatments. Additionally, the group fed Diet B showed significantly better weight gain than the corresponding group fed Diet A at 93% and 100% satiation. The maintenance requirement was determined to be 21.77 mg digestible (dig) Met kg−0.83 d−1 and 35.3 mg dig Met+Cys kg−0.83 d−1 based on a linear model and to be 18.8 mg digestible (dig) Met kg−0.83 d−1 and 30.7 mg dig Met+Cys kg−0.83 d−1 based on an exponential fit to the data. The efficiency of digestible Met utilization (kDMetassessed based on Diet B) for growth above maintenance was found to be 63% (including data of all ration levels) or 71% (excluding data of 100% satiation), while that for the Met+Cys (kDM+C) was found to be 46% (including data of all ration levels) or 51% (excluding data of 100% satiation). The efficiency of digestible protein utilization for weight gain with Diet B was found to be higher (66%) than that with Diet A (57%), indicating the importance of balancing striped catfish diets for dietary Met. The study also determined maintenance requirements and utilization efficiency of other EAAs which need to be further improved. These data can be used to derive AA requirements for striped catfish based on a factorial modelling approach which relies on maintenance requirements, growth utilization of AA, and targeted growth or protein deposition.

Genomic analyses of nitrogen utilization efficiency, its indicator trait blood urea nitrogen and the relationship to classical growth performance and feed efficiency traits in a Landrace × Piétrain crossbred population.

Improving the nutrient efficiency in pork production is required to reduce the resource competition between human food and animal feed regarding diet components edible for humans and to minimize emissions relevant to climate or the environment. Thereby, protein utilization efficiency and its equivalent nitrogen utilization efficiency (NUE) play a major role. Breeding for more nitrogen (N) efficient pigs bears a promising strategy to improve such traits, however, directly phenotyping NUE based on N balance data is neither cost-efficient nor straightforward and not applicable for routine evaluations. Blood urea nitrogen (BUN) levels in the pig are suitable to predict the NUE and, therefore, might be an indicator trait for NUE because BUN is a relatively easy-to-measure trait. This study investigated the suitability of NUE as a selection trait in future breeding programs. The relationships to classical growth performance and feed efficiency traits were analysed as well as the relationship to BUN to infer the role of BUN as an indicator trait to improve NUE via breeding. The analyzes were based on a Landrace F1 cross population consisting of 502 individuals who descended from 20 Piétrain sires. All animals were genotyped for 48,525 SNPs. They were phenotyped in two different fattening phases, i.e., FP1 and FP2, during the experiment. Uni- and bivariate analyses were run to estimate variance components and to determine the genetic correlation between different traits or between the same trait measured at different time points. Moderate heritabilities were estimated for all traits, whereby the heritability for NUE was h2 = 0.293 in FP1 and h2 = 0.163 in FP2 and BUN had the by far highest heritability (h2 = 0.415 in FP1 and h2 = 0.460 in FP2). The significant genetic correlation between NUE and BUN showed the potential of BUN to be considered an indicator trait for NUE. This was particularly pronounced when NUE was measured in FP1 (genetic correlations and between NUE and BUN measured in FP1 and FP2, respectively). The genetic correlations of NUE and BUN with important production traits suggest selecting pigs with high growth rates and low BUN levels to breed more efficient pigs in future breeding programs.

Effects of Individual Essential Amino Acids on Growth Rates of Young Rats Fed a Low-Protein Diet.

To investigate the effects of individual essential amino acids (EAA) on growth and the underlying mechanisms, EAA individually supplemented a low-protein (LP) diet fed to young rats in the present study. Treatments were an LP diet that contained 6% crude protein (CP), a high-protein (HP) diet that contained 18% CP, and 10 LP diets supplemented with individual EAA to achieve an EAA supply equal to that of the HP diet. The CP concentration of the LP diet was ascertained from the results of the first experiment, which examined the effects of dietary CP concentrations on growth rates, with CP ranging from 2% to 26%. Weight gain was increased with the supplementation of His, Ile, Lys, Thr, or Trp as compared to the LP diet (p < 0.05). Feed intake was greater for the His-, Lys-, and Thr-supplemented treatments as compared to the LP group (p < 0.05). Protein utilization efficiency was lower for the HP group than other groups (p < 0.01). The supplementation of Leu, Lys, and Val led to reduced protein utilization efficiency (p < 0.05), but the supplementation of Thr and Trp led to greater efficiency than the LP group (p < 0.05). Compared to the LP group, plasma urea concentrations were elevated with individual EAA supplementation, with the exception of the Thr addition. The added EAA resulted in increased concentrations of the corresponding EAA in plasma, except for Arg and Phe supplementation. The supplementation of Arg, His, Leu, Lys, and Met individually stimulated mTORC1 pathway activity (p < 0.05), and all EAA resulted in the decreased expression of ATF4 (p < 0.05). In summary, the supplementation of His, Ile, Lys, Thr, or Trp to an LP diet improved the growth performance of young rats. Responses to His and Lys additions were related to the activated mTORC1 pathway and feed intake increases. The improved growth performance resulting from the addition of a single EAA is not solely attributed to the increased plasma availability of EAA. Rather, it may be the consequence of a confluence of factors encompassing signaling pathways, the availability of amino acids, and other associated elements. The additivity of these factors results in independent responses to several EAA with no order of limitation, as is universally encoded in growth models for all production animal species.

Effect of milk fat globules on growth and metabolism in rats fed an unbalanced diet.

We assessed the effects of supplementing milk fat globules (MFG) on the growth and development of the skeleton in rats fed a Western unbalanced diet (UBD). The UBD is high in sugar and fat, low in protein, fiber, and micronutrients, and negatively impacts health. The MFG-a complex lipid-protein assembly secreted into milk-has a unique structure and composition, which differs significantly from isolated and processed dietary ingredients. Rats consuming the UBD exhibited growth retardation and disrupted bone structural and mechanical parameters; these were improved by supplementation with small MFG. The addition of small MFG increased the efficiency of protein utilization for growth, and improved trabecular and cortical bone parameters. Furthermore, consumption of UBD led to a decreased concentration of saturated fatty acids and increased levels of polyunsaturated fatty acids (PUFA), particularly omega-6 PUFA, in the serum, liver, and adipose tissue. The addition of small MFG restored PUFA concentration and the ratio of omega-6 to omega-3 PUFA in bone marrow and adipose tissue. Finally, large but not small MFG supplementation affected the cecal microbiome in rats. Overall, our results suggest that natural structure MFG supplementation can improve metabolism and bone development in rats fed an UBD, with the effects depending on MFG size. Moreover, the benefits of small MFG to bone development and metabolism were not mediated by the microbiome, as the detrimental effects of an UBD on the microbiome were not mitigated by MFG supplementation.

Effects of dietary methionine supplementation on the growth performance, immune responses, antioxidant capacity, and subsequent development of layer chicks

Deficiencies or excesses of dietary amino acids, and especially of methionine (Met), in laying hens can lead to abnormal protein anabolism and oxidative stress, which affect methylation and cause cellular dysfunction. This study investigated the effects of dietary methionine (Met) levels on growth performance, metabolism, immune response, antioxidant capacity, and the subsequent development of laying hens. A total of 384 healthy 1-day-old Hyline Grey chicks of similar body weight were randomly allocated to be fed diets containing 0.31%, 0.38%, 0.43% (control group), or 0.54% Met for 6 wk, with 6 replicates of 16 chicks in each. The growth performance of the chicks was then followed until 20 wk old. The results showed dietary supplementation with 0.43% or 0.54% Met significantly increased their mean daily body weight gain, final weight, and Met intake. However, the feed:gain (F/G) decreased linearly with increasing Met supplementation, from 0.31 to 0.54% Met. Met supplementation increased the serum albumin, IgM, and total glutathione concentrations of 14-day-old chicks. In contrast, the serum alkaline phosphatase activity and hydroxyl radical concentration tended to decrease with increasing Met supplementation. In addition, the highest serum concentrations of IL-10, T-SOD, and GSH-PX were in the 0.54% Met-fed group. At 42 d of age, the serum ALB, IL-10, T-SOD, GSH-PX, T-AOC, and T-GSH were correlated with dietary Met levels. Finally, Met supplementation reduced the serum concentrations of ALP, IL-1β, IgA, IgG, hydrogen peroxide, and hydroxyl radicals. Thus, the inclusion of 0.43% or 0.54% Met in the diet helps chicks achieve superior performance during the brooding period and subsequently. In conclusion, Met doses of 0.43 to 0.54% could enhance the growth performance, protein utilization efficiency, antioxidant capacity, and immune responses of layer chicks, and to promote more desirable subsequent development during the brooding period.

Protein requirements for pregnant dairy cows

We aimed to estimate the protein requirements of pregnant Holstein × Gyr cows. Sixty-one Holstein × Gyr (HG) cows were used, with an average initial BW of 480 ± 10.1 kg and 5 ± 0.5 yrs of age. Cows were divided into 3 groups: pregnant (n = 43), non-pregnant (n = 12), and baseline (n = 6). Baseline animals were slaughtered before starting the experiment to estimate the initial body composition of the remaining animals. Pregnant and non-pregnant cows received 2 diets: maintenance and ad libitum. Pregnant cows were slaughtered at 139, 199, 241, and 268 gestation days (GD). First, we used data only from non-pregnant cows to determine the requirements for maintenance and growth in adult cows. The requirements of metabolizable protein for maintenance (MPm; g/EBW0.75/d) were estimated using a linear regression between the metabolizable protein intake (MPI, g/d) and average daily gain (g/d), and the MPm was defined as the intercept divided by the average BW0.75. Net protein requirements for gain (NPg; g/d) were estimated by the first derivative of the allometric equation between final crude protein (CP) in the body (kg) and the final EBW (kg). The efficiency of use of metabolizable protein for gain (k) was calculated from the regression between the retained protein (g/EBW0.75/d) and the MPI (g/EBW0.75/d), and k was the slope of this regression. The MPI was estimated by combining microbial protein synthesis (purine derivatives) with the digestible rumen undegradable protein [(total protein intake minus RDP) × intestinal digestibility]. Second, an exponential model was used to fit the protein accumulation in the gestational components in the function of GD. The first derivative of that model was considered the net requirement for gestation (NPgest). The efficiency of protein utilization for gestation (kgest) was calculated by the iterative method using the equation: Δ = MPI - (MPm + NPg/kg + NPgest/kgest). The iteration was performed aiming at a zero deviation between observed MPI and MP estimated by the requirements determined herein. We obtained a value of 3.88 g/EBW0.75/d for MPm. The estimation of NPg can be calculated according to the following equation: NPg = 0.716 × (EBWopen-0.308) × EBGcorrected, where EBWopen is the empty BW (kg) for non-pregnant animals and EBGcorrected is the empty body gain (kg/d) corrected for the gestational component. The k was determined as 0.347. The net protein requirements for gestation (NPgest) were determined as NPgest (g/d) = 0.0008722 × exp(0.01784 ×GD) × (calfweight/35). The efficiency of using metabolizable protein for gestation (kgest) was 0.625. It is important to highlight that different methods of MP estimates should not be mixed and that the proposed method requires the estimate of microbial protein (estimate it via urinary estimates) and might be a limitation for practical application. In conclusion, new studies should be conducted to validate our results and the methodology adopted to determine protein requirements for pregnancy in dairy cows. Due to the pattern of protein accumulation in the gestational components, we suggest that exponential model to describe protein requirements for pregnancy for dairy cows.

Methionine Supplementation Affects Fecal Bacterial Community and Production Performance in Sika Deer (Cervus nippon).

Amino acid balance is central to improving the efficiency of feed protein utilization and for reducing environmental pollution caused by intensive farming. In previous studies, supplementation with limiting amino acids has been shown to be an effective means of improving animal nutrient utilization and performance. In this experiment, the effects of methionine on the apparent digestibility of nutrients, antler nutrient composition, rumen fluid amino acid composition, fecal volatile fatty acids and intestinal bacteria in antler-growing sika deer were investigated by randomly adding different levels of methionine to the diets of three groups of four deer at 0 g/day (CON), 4 g/day (LMet) and 6 g/day (HMet). Methionine supplementation significantly increased the apparent digestibility of organic matter, neutral detergent fiber (NDF) and acid detergent fiber (ADF) in the LMet group (p < 0.05). The crude protein and collagen protein of antlers were significantly higher in the LMet and HMet groups compared to the CON group and also significantly higher in the HMet group compared to the LMet group, while the calcium content of antlers was significantly lower in the HMet group (p < 0.05). Ruminal fluid free amino acid composition was altered in the three groups of sika deer, with significant changes in aspartic acid, citrulline, valine, cysteine, methionine, histidine and proline. At the phylum level, Firmicutes and Bacteroidetes were highest in the rectal microflora. Unidentified bacterial abundance was significantly decreased in the HMet group compared to the CON group. Based on the results of principal coordinate analysis (PCoA) and Adonis analysis, there was a significant difference in the composition of the intestinal flora between the CON and HMet groups (p < 0.05). At the genus level, compared with the CON group, the abundance of Rikenellaceae_RC9_gut_group and Lachnospiraceae_UCG-010 in the LMet group increased significantly (p < 0.05), the abundance of dgA-11_gut_group in the HMet group decreased significantly (p < 0.05) and the abundance of Lachnospiraceae_UCG-010, Saccharofermentans and Lachnospiraceae_NK3A20_group increased significantly. Taken together, the results showed that methionine supplementation was beneficial in increasing the feed utilization efficiency and improving antler quality in sika deer, while affecting the composition of fecal bacteria.

Dietary supplementation of pyridoxine can enhance the growth performance and improve the protein, lipid utilization efficiency of mandarin fish (Siniperca chuatsi).

This study aimed to assess the effect of pyridoxine supplementation in the mandarin fish diet on growth performance, protein and lipid metabolism, and liver and intestinal histology. Mandarin fish were fed six diets with different levels of pyridoxine (2.67 mg/kg (control), 4.41 mg/kg, 6.57 mg/kg, 10.25 mg/kg, 17.93 mg/kg, 33.12 mg/kg diet) for 8 weeks, and samples were collected for analysis. The findings demonstrated that feeding mandarin fish a diet with 6.57 mg/kg pyridoxine led to a significant increase in weight gain rate (WGR), protein efficiency ratio (PER), whole-body crude protein, whole-body crude lipid, serum protein, cholesterol (CHO), triacylglycerol (TG), high-density lipoprotein-cholesterol (HDL-C), low-density lipoprotein-cholesterol (LDL-C), and alkaline phosphatase (ALP), as well as significantly lower serum glucose (GLU) and feed conversion ratio (FCR), compared to the control group (P < 0.05). Furthermore, we found a significant upregulation of the relative expression of genes associated with hepatic lipid oxidation and synthesis (hl, lpl, pparα, cpt1, cs, srebp1, and fas) and proteolysis (ast, alt, and gdh) in fish fed a diet containing 6.57 mg/kg pyridoxine (P < 0.05). Regarding the histological analysis, we observed a notable decrease in the quantity of intestinal mucus-secreting cells when the fish fed a diet containing 10.25 mg/kg pyridoxine (P < 0.05). These findings suggest that dietary pyridoxine supplementation promotes mandarin fish growth by improving the efficiency of protein and lipid utilization. Additionally, we used a broken-line regression analysis to estimate the optimal dietary pyridoxine requirement for mandarin fish in the range of 6.17-6.41 mg/kg based on WGR, FCR, and PER.

Insights into the mechanism of growth and fat deposition by feeding different levels of lipid provided by transcriptome analysis of swamp eel (Monopterus albus, Zuiew 1793) liver.

Lipid is an important source of energy in fish feeds, and the appropriate fat content can improve the efficiency of protein utilization. However, excessive lipid content in the feed can lead to abnormal fat deposition in fish, which has a negative effect on the growth of fish. Therefore, the effects of feed lipid levels on swamp eel were studied. Essential functional genes were screened using transcriptomics. We divided 840 fish into seven groups (four replicates). A mixture of fish and soybean oils (1:4), 0%, 2%, 4%, 6%, 8%, 10%, and 12% was added to the basic feed were named groups one to seven (L1-L7), respectively. Isonitrogenous diets were fed swamp eel for 10 weeks. Growth performance, visceral index, nutritional components, and biochemical indexes were measured and analyzed. Livers of the 0%, 6%, and 12% groups were subjected to transcriptome sequencing analysis. The results of our study showed that: the suitable lipid level for the growth of swamp eel was 7.03%; the crude fat content of whole fish, liver, intestine, muscle, and skin increased with the increase of lipid level, with some significant difference, and excess fat was deposited in skin tissue; triglyceride, total cholesterol, and free fatty acid contents increased with the increase of feed lipid level. High-density lipoprotein levels in the L3 and L4 groups were higher than in the other groups. Blood glucose concentrations in the L5, L6, and L7 groups increased; the liver tissue structure was damaged when the lipid level was too high. two-hundred-and-twenty-eight differentially expressed genes were found. Several critical pathways regulating glucose metabolism and energy balance (e.g., glycerolipid metabolism, glycolysis synthesis, degradation of ketone bodies, and Janus Kinase/Signal Transducer and Activator of Transcription signaling pathway) were enriched in swamp eel compared with the Kyoto Encyclopedia of Genes and Genomes (KEGG) database. Suitable lipid levels (7.03%) can promote the growth of swamp eel, and excessive lipid levels can cause elevated blood lipids and lead to liver cell damage. Regulatory mechanisms may involve multiple metabolic pathways for glucose and lipid metabolism in eels. This study provides new insights to explain the mechanism of fat deposition due to high levels of lipid and provides a basis for the production of efficient and environmentally friendly feed for swamp eel.

Fecal metabonomics combined with 16S rDNA sequencing to analyze the changes of gut microbiota in rats fed with different protein source diets.

When blended, animal and plant proteins can complement each other in terms of amino acid composition and release time. In this study, we investigated whether the blended protein diet has a better feeding effect than the single protein diet, and to reveal the differences in growth and intestinal microbiota composition caused by the blended protein diet. Forty Sprague Dawley (SD) rats received diets with different protein sources, including casein (C), whey protein (WP), black soybean protein (BSP), and black soybean-whey blended protein (BS-WP), for eight weeks. To investigate the effects of blended protein supplement on gut microbiota and metabolites, we performed a high throughput 16S rDNA sequencing and fecal metabolomics profiling. In addition, we determined growth and serum biochemical indices, and conducted intestinal morphology analyses. Compared to those in the BSP and WP groups, the daily body weight gain and feed conversion efficiency increased in the BS-WP group. Serum biochemical indices indicated that the protein utilization efficiency of the WP and BS-WP groups was relatively high, and the BS-WP blended protein diet improved the protein adoption rate. The BS-WP blended protein diet also improved intestinal tissue morphology and promoted intestinal villi development compared to the single protein diets. Furthermore, dietary protein altered the composition of gut microbiota, the gut microbial diversity of rats fed with the BS-WP diet was significantly (P < 0.05) higher than that of the other groups. The difference in dietary protein corresponded with an alteration of fecal amino acids and their metabolites, and tryptophan and tyrosine metabolism were the key mechanisms leading to the changes in fecal microbial composition. Dietary protein sources played an important role in the growth and development of rats by influencing intestinal metabolism and microbial composition. The BS-WP blended protein diet was more conducive to nutrient absorption than the single protein diet. Furthermore, blended protein increased the diversity of intestinal microbes and aided the establishment of intestinal barrier function.

Utilization of functional feed additives to produce cost‐effective, ecofriendly aquafeeds high in plant‐based ingredients

AbstractFishmeals (FM) are the most superior protein sources in aquafeed. World‐wide growing demand and limited supply of FM limit its use as sole protein sources in aquafeed and force fish nutritionists to develop alternate protein‐based aquafeeds. However, increasing alternate proteins over a certain percentage reduces fish performance. Many attempts have been made (viz., manipulation of processing technology of feed ingredients and feed formulations, supplementation strategy) to overcome the negative effects of increasing alternative protein inclusion in aquafeeds. Among them, supplementation of functional feed additives (FuFAs) has received promising attention. Recently, several FuFA (nucleotides, organic acid, probiotics, prebiotics, and certain amino acids) have been administrated in alternative protein‐based aquafeeds and demonstrate increased efficiency of alternative protein utilization along with improved fish growth and health performances. However, numerous gaps remain in existing knowledge about the administration of exogenous FuFA in alternative protein‐based aquafeeds, including the effects on digestion, age/size‐related responses, species variations, absorption, metabolism, and influences on physiological responses. Application of biochemical and molecular technologies is important to better understand the underlying mechanisms of FuFA functions in alternate protein‐based aquafeed to improve fish and shrimp performance. Efforts are also needed by researchers and industrialists regarding production technologies to reduce functional feed supplement costs. Moreover, in‐depth research knowledge is needed considering the use of functional feeds to combat fish diseases and reduce dependence on antibiotics and chemical therapeutants, thereby making aquaculture more eco‐friendly.

Protein-Sparing Effect of α-Lipoic Acid in Diets with Different Protein/Carbohydrate Ratios for the Oriental River Prawn, Macrobrachium nipponense

Carbohydrates are commonly used in aquaculture feed because they are the cheapest energy source. Promoting carbohydrate catabolism for energy production can increase the dietary protein utilization efficiency (i.e., a protein-sparing effect). Alpha-lipoic acid (α-LA) is a cofactor of some rate-limiting key enzymes (pyruvate dehydrogenase complex and α-ketoglutarate dehydrogenase) in carbohydrate metabolism. To determine whether α-LA could promote carbohydrate catabolism to have a protein-sparing effect, we investigated the growth, activities of key enzymes involved in carbohydrate metabolism, transcript levels of genes encoding enzymes involved in energy metabolism, and the hepatopancreas structure of Macrobrachium nipponense fed with diets containing different protein/carbohydrate ratios (P/C) with and without α-LA. Six experimental diets for M. nipponense were formulated with casein and fish meal as the protein sources, fish oil and soybean oil as the lipid sources, and corn starch as the carbohydrate source. The six diets consisted of three different P/Cs ( P 41 / C 18 , P 37 / C 24 , and P 33 / C 30 ) without or with α-LA (at 1300 mg/kg). Each diet had six replicates and was fed to prawns (initial weight 0.110 ± 0.010 g) twice daily to apparent satiation. Dietary supplementation with α-LA significantly increased the survival rate of prawns, regardless of the P/C ratio. In the low-P/C group ( P 33 / C 30 ), the weight gain did not vary between those that consumed α-LA and those that did not. The P/C ratio and α-LA significantly affected the activities of key enzymes involved in glycolysis and the citric acid cycle. In the P 41 / C 18 group, the hexokinase (HK) and pyruvate dehydrogenase (PDH) activities were significantly higher in prawns that consumed α-LA than in those that did not. In both the P 37 / C 24 and P 33 / C 30 groups, the pyruvate kinase (PK) activity was significantly higher in prawns that consumed α-LA than in those that did not. Significant interactions between P/C and α-LA were found for the transcript levels of genes encoding adenine ribonucleotide-dependent protein kinase subunits (AMPK αand AMPK β), HK, glucose-6-phosphate dehydrogenase (G6PDH), acetyl-CoA carboxylase (ACC), and fatty acid binding protein 10 (FABP 10), all of which are involved in energy metabolism. In the low-P/C group, α-LA increased the transcript levels of genes encoding AMPK α, AMPK β, HK, phosphoenolpyruvate carboxykinase (PEPCK), G6PDH, and MDH, decreased the transcript level of the gene encoding ACC, and did not affect the transcript levels of genes encoding FABP10 and palmitoyl transferase 1 (CPT1). The P/C ratio and α-LA did not affect the overall morphology of the hepatopancreas, but at a low P/C, dietary supplementation of α-LA increased the number of hepatopancreatic B cells. In conclusion, supplementation with α-LA at 1300 mg/kg in a low-P/C diet of M. nipponense increased its carbohydrate utilization efficiency, energy metabolism, and the number of hepatopancreas B cells, thereby having a protein-sparing effect.

Eimeria maxima infection impacts the protein utilisation of broiler chicks from 14 to 28 days of age

In floor-raised broilers, coccidiosis is responsible for reducing the use of nutrients, mainly by impairing intestinal tissue function and activating the immune system. Understanding and quantifying how balanced dietary protein (BP) is used when birds are challenged will allow nutritionists to make decisions regarding challenged flocks. This study aimed to determine the effects of Eimeria maxima on broiler performance and body composition, and to calculate changes in the maintenance and efficiency of protein utilisation (Ep). A total of 2 400 male 14-day-old Cobb500 broiler chickens were randomly allotted to ten groups with six replications of 40 birds each, with a 5 × 2 factorial arrangement of treatments. Five levels of BP in reference to digestible lysine (3.6, 7.2, 10.8, 14.4, and 18.0 g/kg) were fed to unchallenged (NCH) and challenged (CH) broilers with 7 × 103E. maxima sporulated oocysts from 14 to 28 days of age. Performance and body deposition were measured using a comparative slaughter technique to compare BP maintenance requirements and Ep. ANOVA followed by a posthoc test was performed to compare the effects of BP levels, challenge, and their interactions. A monomolecular model describing the responses of NCH and CH broilers to BP intake, maintenance, and maximum protein deposition was compared. There were significant interactions between body weight gain and digestible lysine intake among the factors studied. Infection had a negative impact on all variables analysed, proving the efficacy of the challenge. The maintenance did not differ between the CH and NCH groups. Increased levels of dietary BP did not recover the maximum protein deposition in CH broilers. Eimeria maxima significantly reduced Ep by a factor of 0.09 times on Ep compared to the control group. The Eimeria maxima challenge was responsible to modify the use of BP altering the body composition and impairing broilers performance.

Growth performance and quality indicators of European seabass (Dicentrarchus labrax) fed diets including refined glycerol

Glycerol is a by-product of the biodiesel industry with growing availability and applicability in animal feeds. However, its use as a source of energy in aquafeeds has only been investigated in a small number of fish species. The effects of dietary refined glycerol inclusion rate on growth performance, nitrogen and energy balances, and fish quality were investigated in juvenile European seabass. Twelve fish were assigned to each of the nine tanks and used to test the experimental diets in triplicate. The diets comprised 0, 25, or 50 g kg−1 of refined glycerol (G0, G25, and G50, respectively), being not isoenergetic, although having similar digestible protein-to-energy ratios (22 mg kJ−1). Fish were fed twice a day to apparent satiation for 90 days. The G25 group displayed a higher absolute feed intake and a higher weight gain than the G0 or G50 groups, although feed efficiency and the use of protein as an energy source were similar between the three groups. Seabass fed the G25 diet also displayed a higher specific growth rate than fish fed the G50 diet. Higher hepato-somatic and visceral lipid-somatic indices of seabass were observed with increased levels of dietary glycerol, although proximal body composition remained similar across groups. The apparent digestibility coefficients for dry matter, crude protein, and energy increased with increasing dietary glycerol inclusion rate, whereas the apparent lipid digestibility coefficient was highest for the G25 group. No differences in the energy retained as protein, nitrogen efficiency, and total ammonia excretion rate were detected between seabass fed the different diets. At the end of the growth trial, physical and microbiological quality parameters of the skin and gill surfaces were assessed after 1 and 10 days of storage on ice. Fish quality decreased with storage time but was not affected by the dietary glycerol inclusion rate. In summary, the inclusion of refined glycerol in seabass diets may have economic benefits for feed manufacture and a positive effect on growth performance but does not change the efficiency of protein utilization in this carnivorous species.

Effects of increasing levels of whole Black Soldier Fly (Hermetia illucens) larvae in broiler rations on acceptance, nutrient and energy intakes and utilization, and growth performance of broilers

Meal of black soldier fly larvae (BSFL), which requires extraction of protein and fat, is a novel protein source for poultry, while unprocessed whole BSFL could even directly be fed to chickens. Newly hatched Ross-308 chicks (n = 252) received whole BSFL at 10% (L10), 20% (L20), or 30% (L30) of voluntary feed intake (FI) of control chickens (CON) that received no BSFL but only age-specific diets (n = 63 birds / group) for 42 days (d). Acceptance and nutrient and energy intake of birds by BSFL and FI were calculated. Plasma metabolites were measured using an automatic enzymatic analyzer and immunoglobulins with ELISA. Depending on the variable, data were analyzed using ANOVA or repeated measures ANOVA to address treatment, time and interaction effects. Birds consumed all offered larvae. With the exception of d1, time spent by birds eating their daily portion of larvae (TSL, min/pen) did not differ among the larvae supply groups (P = 0.982). The L10 had a higher larvae eating rate (LER) that is, speed of larvae intake than did L20 and L30 (P < 0.05), implying increased competition for less available BSFL. The ratio of LER to feed eating rate (FER) was greater than 50 fold change difference (FCD), indicating a strong interest of chickens in BSFL over regular feed. Whole BSFL intake up to 30% of voluntary FI did not adversely affect broiler growth (P > 0.05). The L30 had lower total dry matter and metabolizable energy intakes (P < 0.05), although total fat intake was higher in L30 than in CON (P < 0.05). Compared with CON, 30% whole BSFL increased dietary protein-to-energy ratios, plasma uric acid and serum alkaline phosphatase concentrations (P < 0.05). We conclude that whole BSFL can be included in broiler rations up to 20% without negatively affecting growth performance and nutrient conversion efficiency, whereas a higher proportion is associated with lower protein utilization efficiency, possibly due to lower total energy intake.
