Leptin fragment modulates the stimulatory effects of chorionic gonadotropin on testicular steroidogenesis in a model of diet-induced obesity in rats

Leptin, secreted by adipose tissue, indirectly stimulates the activity of GnRH-producing neurons of the hypothalamus and thus regulates the functional activity of the hypothalamic-pituitary-testicular (HPT) axis. As is known, the obesity is accompanied by systemic hyperleptinemia and impaired leptin transport in the central nervous system, which limits the use of full-length leptin as a drug. It was previously shown that intranasally administered leptin fragment MA-[D-Leu4]-OB3 (LF) enhances the steroidogenic effect of human chorionic gonadotropin (hCG) in rats fed a standard diet. An even more urgent task is to assess its effect on testicular steroidogenesis in conditions of obesity, which reduces reproductive functions in men. The aim of the work was to study the ability of LF (200 μg/kg, intranasally, 3 days) to modulate the effect of hCG (10 IU/rat, subcutaneously, once) on testicular steroidogenesis in rats with obesity induced by a high-fat/high-carbohydrate diet (HFHCD), and also to evaluate influence of the GnRH receptor antagonist cetrorelix (ANT, 75 µg/kg, subcutaneously, 3 days) on the effects of LF. Male Wistar rats were used for the study and received HFHCD for 20 weeks. In obese rats, the level of the luteinizing hormone (LH) receptor in the testes was reduced and the expression of the Cyp11a1 gene, encoding the steroidogenic enzyme cytochrome P450scc, was compensatory increased. LF administration enhanced the effect of hCG on the testosterone level in the blood and the expression of the Star gene, encoding the cholesterol transport protein StAR, which indicates the ability of LF to positively modulate the activity of the HPT axis in obesity. Co-administration of ANT and LF, on the contrary, reduced the stimulating effect of hCG on testosterone levels and Star gene expression, which may be due to the testicular effects of LF. Our data indicate the ability of LF to influence various components of the male gonadal axis under conditions of diet-induced obesity.

Vitamin D3 reduces the symptoms of ovarian hyperstimulation syndrome in mice and inhibits the release of granulosa cell angiogenic factor through pentraxin 3

It has been reported that the effective inhibition of vascular endothelial growth factor (VEGF) can prevent the progression of ovarian hyperstimulation syndrome (OHSS). The present study aimed to investigate the mechanism underlying the effect of vitamin D3 (VD3) on OHSS in mouse models and granulosa cells. The effects of VD3 administration (16 and 24 IU) on ovarian permeability were determined using Evans blue. In addition, ovarian pathology, corpus luteum count, inflammatory responses, and hormone and VEGFA levels were assessed using pathological sections and ELISA. Molecular docking predicted that pentraxin 3 (PTX3) could be a potential target of VD3, and therefore, the effects of human chorionic gonadotropin (hCG) and VD3 as well as PTX3 overexpression on the production and secretion of VEGFA in granulosa cells were also investigated using western blotting and immunofluorescence. Twenty-four IU VD3 significantly reversed the increase in ovarian weight and permeability in mice with OHSS. Additionally, VD3 diminished congestion and the number of corpus luteum in the ovaries and reduced the secretion levels of inflammatory factors and those of estrogen and progesterone. Notably, VD3 downregulated VEGFA and CD31 in ovarian tissues, while the expression levels of PTX3 varied among different groups. Furthermore, VD3 restored the hCG-induced enhanced VEGFA and PTX3 expression levels in granulosa cells, whereas PTX3 overexpression abrogated the VD3-mediated inhibition of VEGFA production and secretion. The present study demonstrated that VD3 could inhibit the release of VEGFA through PTX3, thus supporting the beneficial effects of VD3 administration on ameliorating OHSS symptoms.

(192) Evaluating Effect of hCG and hCG+rFSH Therapy on Ejaculation Disorders in Middle-Aged Men: A Case Study

Abstract Introduction The prevalence of various ejaculation disorders affecting middle-aged and older men has gained increasing attention due to their significant impact on quality of life. One of them, Late-Onset Hypogonadism (LOH) syndrome, can result in a decline in overall well-being. Testosterone replacement therapy (TRT) is commonly used to address LOH; however, it presents challenges such as diminishing endogenous testosterone secretion and incomplete symptom relief. Objective This study aimed to investigate the effects of human chorionic gonadotropin (hCG) or hCG plus recombinant follicle-stimulating hormone (rFSH) therapy in patients experiencing ejaculation disorders characterized by low semen volume and decreased pleasure during ejaculation. Methods Four male patients, aged 55 to 59 years, complaining of decreased ejaculatory pleasure and low semen volume, were enrolled in the study. One patient received hCG+rFSH therapy, while three patients received hCG alone. The initial dose of hCG was 5,000 units administered twice a week, and if deemed insufficient, it was increased to three times a week. rFSH was administered at a dosage of 150 units three times a week. Levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone, and estradiol (E2) were measured monthly. Anastrozole (1 mg) was added for patients exhibiting elevated E2 levels. Results Case 1: A 58-year-old male. Prior to treatment, testosterone and E2 levels were 4.84 ng/mL and 34.2 pg/mL, respectively. hCG+rFSH therapy resulted in increased testosterone levels (10.2 ng/mL) and elevated E2 levels (109.0 pg/mL) after three months of treatment. Addition of anastrozole resulted in testosterone levels of 10.90 ng/mL and E2 levels of 57.9 pg/mL in the following month. The patient experienced improved sexual desire, increased semen volume, and hair loss as a side effect. Treatment was discontinued after six months. Case 2: A 57-year-old male with diabetes mellitus. Pre-treatment testosterone and E2 levels were 2.19 ng-mL and 21.9 pg/mL, respectively. After changing the hCG dosage to 5,000 units three times a week, testosterone levels reached 5.33 ng/mL and E2 levels increased to 72.8 pg/mL after two months. With the addition of anastrozole four months into treatment, E2 levels were 44.4 pg/mL after five months. The patient reported improved morning erections, increased libido, increased semen volume, and improved glycemic control (HbA1c decreased from 9.0 to 7.0). Case 3: A 55-year-old male. Pre-treatment testosterone and E2 levels were 4.99 ng/mL and 32.3 pg/mL, respectively. In the first month of hCG therapy, testosterone levels increased to 13.6 ng/mL, while E2 levels rose to 75.5 pg/mL. Then, despite testosterone remaining in the 9.0–10.0 ng/mL range, E2 levels increased to 100 pg/mL. With the addition of anastrozole in the fifth month, the patient reported increased semen volume thereafter. Case 4: A 59-year-old male with a history of bipolar disorder. Prior to treatment, testosterone and E2 levels were 2.45 ng/mL and 23.0 pg/mL, respectively. hCG therapy resulted in testosterone levels of 6.71 ng/ml and E2 levels of 51.0 pg/mL after two months. Conclusions This case study highlights the outcomes of hCG or hCG+rFSH therapy in middle-aged men experiencing ejaculation disorders. Disclosure No.

The Effect of Human Chorionic Gonadotropin and Ketoprofen Applications on Pregnancy Rates in Dairy Cows After Artificial Insemination

The aim of the presented study is to investigate the effect of providing progesterone (P4) support for the formation and maintenance of pregnancy or suppressing the luteolysis mechanism during the maternal acceptance process on pregnancy rates. For this purpose, 96 Holstein cows, aged between 2 and 8 years, were synchronized using the Ovsynch protocol to align their estrous cycles. After artificial insemination, the cows were divided into four groups. The group receiving Human Chorionic Gonadotropn (hCG) on the 6th day after artificial insemination (H; n=20), the group receiving Ketoprofen on the evening of the 15th day and the morning of the 16th day after artificial insemination (K; n=24), the group receiving both hCG on the 6th day and Ketoprofen on the 15th day evening and 16th day morning after artificial insemination (HK; n=25), and the control group (C; n=27) that did not receive any post-insemination treatment were established. Pregnancy checks were conducted on the 30th, 45th, and 60th days through rectal and ultrasonographic (USG) examinations. According to the results of the pregnancy examinations, it was determined that 9 out of 20 cows (45.00%) in Group H, 10 out of 24 cows (41.67%) in Group K, 16 out of 25 cows (64.00%) in Group HK, and 12 out of 27 cows (44.44%) in Group C were pregnant. No statistically significant difference (P>0.05) was observed among the groups. Our results suggest that a multi-purpose treatment may be required to reduce pregnancy loss during the early pregnancy period in cows due to an approximately 20% increased pregnancy rate after administering hCG and Ketoprofen. However, further studies, especially involving more cows, are necessary to optimize and use these combined treatments and potentially increase pregnancy rates.

Effect of human chorionic gonadotropin injection before frozen-thawed embryo transfer: A retrospective cohort study.

The aim of this study is to investigate the effect of human chorionic gonadotropin (hCG) in hormone replacement regimen for frozen-thawed embryos. We performed a retrospective cohort study and included patients who underwent frozen embryo transfer (FET) between January 1, 2020 and May 31, 2022. According to the protocols for the FET cycle, the patients were divided into control (n = 238) and hCG groups (n = 216). The clinical pregnancy rate, live birth rate, early abortion rate, late abortion rate, and ectopic pregnancy rate were compared between the 2 groups. There was a significant difference in clinical pregnancy rate between the hCG and control groups (55.1% vs 45.8%, P = .048). The ectopic pregnancy rate decreased (5.0% vs 6.4%, P = .654), while the live birth rate increased (36.1% vs 29.0%, P = .105) in the hCG group. However, these differences were not statistically significant. The administration of hCG injection in HRT-FET cycles alone was also found to be associated with clinical pregnancy by logistic regressive analysis. HCG injection in the hormone replacement regimen for FET increased the clinical pregnancy rate.

Strategies to improve hematopoietic stem and progenitor cell (HSPC) mobilization from the bone marrow can have a pivotal role in addressing iatrogenic bone-marrow insufficiency from chemo(radio)therapy and overcoming peripheral blood stem cell transplantation (PBSCT) limitations such as insufficient mobilization. Granulocyte-colony stimulating factor (G-CSF) represents the standard mobilization strategy for HSPC and has done so for more than three decades since its FDA approval. Its association with non-G-CSF agents is often employed for difficult HSPC mobilization. However, obtaining a synergistic effect between the two classes is limited by different timing and mechanisms of action. Based on our previous in vitro results, we tested the mobilization potential of human chorionic gonadotropin (HCG), alone and in combination with G-CSF in vivo in a murine study. Our results show an improved mobilization capability of the combination, which seems to act synergistically in stimulating hematopoiesis. With the current understanding of the dynamics of HSPCs and their origins in more primitive cells related to the germline, new strategies to employ the mobilization of hematopoietic progenitors using chorionic gonadotropins could soon become clinical practice.

Effect of human chorionic gonadotrophin (HCG) and luteinizing hormone releasing hormone A3 (LHRH-A3) on reproduction and development of brood pouch in Hippocampus erectus

Syngnathid fishes (seahorses and pipefish) are unique in reproduction, with males becoming pregnant and giving birth. The brood pouch provides osmoregulation, gas exchange, waste transport, immunoprotection and nutrition supply. However, the function of hormone trafficking is rarely studied. By injecting the mixture liquor of human chorionic gonadotrophin (HCG, 2 IU/g) and luteinizing hormone-releasing hormone A3 (LHRH-A3, 5 ng/g) into the brood pouch of Hippocampus erectus, we found the reproductive interactions accelerated dramatically, and 4 pairs were successfully mated. After 24 h, the inner epithelium was highly folded and the loose connective tissue layer was highly distended. In the inner tissue layer of the brood pouch, fibroblasts secreted vigorously, blood vessels multiplied and expanded, indicating the physiological structure of the brood pouch modified hastily from the unpregnant stage to the progravid phase. Correspondingly, transcriptome results revealed the gene expression of circadian, epitheliogenesis, angiogenesis, lipid transfer, and spermiogenesis were significantly up-regulated. We innovatively found that macromolecule exogenous hormone (HCG) can be not only efficiently transported from brood pouch to body, but also promote mating and the physiological changes of the brood pouch.

Objective: To compare the effect of human chorionic gonadotropin (hCG)-producing peripheral blood mononuclear cells (PBMCs) and PBMCs activated by hCG in vitro and expressions of related immune genes in mouse implantation. Methods: hCG-producing PBMCs (transfected PBMC) and PBMCs activated by hCG in vitro were introduced into isolated mouse endometrial cells, while cell cultures were divided into four groups: the control, PBMC, transfected, and activated PBMC groups. The expression of studied genes (IL-lβ, IL-6, Lif, and Vegf) was evaluated and blastocyst attachment on the cocultured cells (isolated endometrial cells and PBMC cells) was monitored in all four groups. Results: Data showed that expression decreased in the PBMC group compared to the treated PBMC (transfected and activated PBMCs) and increased in transfected PBMC compared to the activated PBMC. Attachment and migration of blastocysts were dramatically enhanced in the transfected PBMC group compared to the activated PBMC group (P<0.05). Conclusions: Use of hCG-producing PBMCs (transfected PBMC) has more influence on endometrial receptivity.

Comparative Effects of Human Chorionic Gonadotropin and Recombinant Luteinizing Hormone Supplementation during the Late Follicular Phase

In selected patients, addition of LH activity can improve the outcome of ovarian stimulation. Two common LH activity supplementation strategies are recombinant human LH (rLH) and human chorionic gonadotropin (hCG). The respective effectiveness of hCG versus rLH using various daily dosages and modes of administration is still discussed. In this study, we compared the outcomes of hCG versus rLH supplementation among patients with slow response to recombinant follicle stimulating hormone (rFSH).

The effect of human chorionic gonadotropin day serum progesterone level on the live birth rate of fresh embryo transfer with GnRH antagonist protocols

Objective: To investigate the effect of human chorionic gonadotropin (HCG)day serum progesterone (P) level on the live birth rate (LBR) of fresh embryo transfer with GnRH antagonist protocols. Methods: Patients who underwent the first IVF/ICSI in the Reproductive Center of the Third Affiliated Hospital of Zhengzhou University from January 2018 to December 2020 were included for analysis. The patients with normal ovarian response with GnRH antagonist protocols were included (n=765). The receiver operating characteristic curve (ROC) was used to select the optimal cut-off value of serum P on HCG day (0.83 μg/L), and the included cycles were divided into two groups: P<0.83 μg/L (n=444) and P≥0.83 μg/L (n=321). The primary outcome measure was LBR. Secondary outcome measures included clinical pregnancy rate (CPR) and early miscarriage rate. The difference of the above indexes between the two groups was compared. Multivariate logistic regression model was used to analyze the effect of serum P level on LBR in fresh embryo transfer cycles. Results: The maternal ages in P<0.83 μg/L group and P≥0.83 μg/L group were (32.40±5.49) years and (32.53±5.51) yeas, respectively. The paternal ages were (33.35±6.34) years and (33.43±6.38) years, respectively of which, the difference was not statistically significant (P>0.05). The CPR in the P<0.83 μg/L group was 45.9% (n=204), which was significantly higher than that in the P≥0.83 μg/L group (37.1%) (n=119) (P=0.014). There was no significant difference in the early miscarriage rate between the two groups [14.2% (n=29) vs 14.3% (n=17), P=0.986]. The LBR in the P<0.83 μg/L group was significantly higher than that in the P≥0.83 μg/L group [36.3% (n=161) vs 28.0% (n=90), P=0.017]. By multivariate logistic regression model analysis, the maternal age, type of embryo transferred, number of embryos transferred, endometrial thickness on HCG day and serum P level on HCG day were independent risk factors of LBR. The adjust OR(95%CI) were 0.91(0.88-0.94), 2.36(1.04-5.35), 1.84(1.14-2.95), 1.16(1.07-1.25)and 0.63(0.44-0.89), all P<0.05. Conclusion: When the GnRH antagonist protocol is applied in the normal ovarian response population, as the serum P on the HCG trigger day≥0.83 μg/L, the CPR and LBR of fresh embryo transfer are decreased.

Effects of human chorionic gonadotropin-producing peripheral blood mononuclear cells on the endometrial receptivity and implantation sites of the mouse uterus.

This research investigated the effects of human chorionic gonadotropin (HCG)-producing peripheral blood mononuclear cells (PBMCs) on the implantation rate and embryo attachment in mice. In this experimental study, a DNA fragment of the HCG gene was cloned into an expression vector, which was transfected into PBMCs. The concentration of the produced HCG was measured using enzyme-linked immunosorbent assay. Embryo attachment was investigated on the co-cultured endometrial cells and PBMCs in vitro. As an in vivo experiment, intrauterine administration of PBMCs was done in plaque-positive female mice. Studied mice were distributed into five groups: control, embryo implantation dysfunction (EID), EID with produced HCG, EID with PBMCs, and EID with HCG-producing PBMCs. Uterine horns were excised to characterize the number of implantation sites and pregnancy rate on day 7.5 post-coitum. During an implantation window, the mRNA expression of genes was evaluated using real-time polymerase chain reaction. DNA fragments were cloned between the BamHI and EcoRI sites in the vector. About 465 pg/mL of HCG was produced in the transfected PBMCs. The attachment rate, pregnancy rate, and the number of implantation sites were substantially higher in the HCG-producing PBMCs group than in the other groups. Significantly elevated expression of the target genes was observed in the EID with HCG-producing PBMCs group. Alterations in gene expression following the intrauterine injection of HCG-producing PBMCs, could be considered a possible cause of increased embryo attachment rate, pregnancy rate, and the number of implantation sites.

The effect of progesterone injection combined with prostaglandineF2α, GnRH and hCG administrations on the pregnancy and lambing rate of lactating and non-lactating fat-tailed ewes during the breeding season.

The objective was to compare the effect of human chorionic gonadotropin (hCG) and gonadotropin-releasing hormone (GnRH) on the pregnancy and lambing rate of treated ewes with a short-term progesterone (P4) injection during the breeding season. In Exp 1, non-lactating ewes (n=158) were used and received P4 , three times every 48h and received PGF2α along with the last dose of P4. Ewe received hCG (n=79, 3PHCG, IM) or GnRH (n=79, 3PGnRH, IM) 24 h after the last dose of P4 treatment. In the Exp 2, lactating ewes (n=62) received P4 and hCG (n=24, 3PHCG) or GnRH (n=24, 3PGnRH) similar to Exp 1, or considered as control (n=14) and received PGF2α 48 h before ram release. Rams were released into the studied ewe's flock 24 h after the hCG or GnRH administration, and pregnancy diagnosis was performed on day 50 after ram release. In Exp 1, all reproductive indices were not significant between groups except twin lambing rate, that was higher in the 3PHCG (11.3%) compared with the 3PGnRH (1.9%) group (p=.05). There were no significant differences in overall pregnancy and lambing rates between 3PHCG (70.9% and 67.1%) and 3PGnRH (69.6% and 68.4%) groups (p > .05). In Exp 2, all reproductive parameters were not significant between 3PHCG and 3PGnRH groups (p > .05). There was significant difference in overall oestrous rate between control (35.7%) and treatment (3PHCG, 70.9% and 3PGnRH, 79.2%) groups. To conclude, administration of GnRH can be a good alternative to hCG injection under field conditions.

Graduate Student Literature Review: Effects of human chorionic gonadotropin on follicular and luteal dynamics and fertility in cattle.

Circulating progesterone concentrations during the growth of the ovulatory follicle and early embryo development have been positively associated with embryo quality and survival and pregnancy success. As a potent luteotropic agent with LH-like activity, human chorionic gonadotropin (hCG) has been tested in different studies to improve pregnancy outcomes by increasing circulating progesterone concentrations during the growth of the ovulatory follicle or early embryonic development. Nevertheless, hCG has produced inconsistent, contradictory, and intriguing results. Furthermore, recent research indicates that hCG, when used before artificial insemination, may affect physiological events necessary for the ovulation of a viable oocyte. In addition, the use of hCG-inducing accessory corpus luteum during the estrous cycle seems to disturb luteolysis and follicle and luteal dynamics during the estrous cycle. This literature review discusses past and current research exploring the effects of hCG on the estrous cycle characteristics and pregnancy per artificial insemination and embryo transfer.

Human chorionic gonadotropin promotes murine Treg cells and restricts pregnancy-harmful proinflammatory Th17 responses.

An equilibrium between proinflammatory and anti-inflammatory immune responses is essential for maternal tolerance of the fetus throughout gestation. To study the participation of fetal tissue-derived factors in this delicate immune balance, we analyzed the effects of human chorionic gonadotropin (hCG) on murine Treg cells and Th17 cells in vitro, and on pregnancy outcomes, fetal and placental growth, blood flow velocities and remodeling of the uterine vascular bed in vivo. Compared with untreated CD4+CD25+ T cells, hCG increased the frequency of Treg cells upon activation of the LH/CG receptor. hCG, with the involvement of IL-2, also interfered with induced differentiation of CD4+ T cells into proinflammatory Th17 cells. In already differentiated Th17 cells, hCG induced an anti-inflammatory profile. Transfer of proinflammatory Th17 cells into healthy pregnant mice promoted fetal rejection, impaired fetal growth and resulted in insufficient remodeling of uterine spiral arteries, and abnormal flow velocities. Our works show that proinflammatory Th17 cells have a negative influence on pregnancy that can be partly avoided by in vitro re-programming of proinflammatory Th17 cells with hCG.

The effectiveness of human chorionic gonadotropin in stimulation of second spermiation in pikeperch Sander lucioperca during the spawning season

The aim of the present study was to evaluate the effectiveness of human chorionic gonadotropin (hCG) in stimulation of second spermiation in already reproduced pikeperch males during the same spawning season. Fish (mean weight 919.9 ± 236.9 g) were divided into the control saline-injected group, and two groups injected with 200 and 400 IU hCG/kg body weight. Forty-eight hours following injection (at a temperature of 16.7 °C) the percentage of fish producing useable amounts of semen increased from 30% in the control group up to 80% in the treated fish and the highest volume of semen was recorded in fish treated with 400 IU/kg BW hCG. Spermatocrit (50.7 ± 9.1%) and sperm concentration (17.5 ± 5.0 × 109 spermatozoa/mL) were significantly higher in control fish than those recorded in hCG-treated fish (24.8 ± 7.7% and 8.1 ± 2.4 × 109 spermatozoa/mL in 200 IU/kg; 32.3 ± 17.6% and 10.9 ± 7.5 × 109 spermatozoa/mL in 400 IU/kg). No significant differences were observed between the groups with respect to sperm motility (as a spermatozoa activity time and percentage); however, a change in plasma alkaline phosphatase activity was found in the group injected with 400 IU/kg of hCG. The lack of significant differences in plasma metabolites and cortisol between control and hCG-treated fish indicated that the hCG had no effect on the stress response. Testosterone levels were significantly increased in the hCG-treated fish, whereas no significant differences in 17β-estradiol were recorded. No differences between the groups in plasma levels of thyroid hormones suggested lack of hCG-induced effect on hypothalamus-pituitary-thyroid axis. The results of this study demonstrated that the spermiation of already reproduced pikeperch males could be induced by hCG during the same spawning season. Based on the results on semen volume and concentration, injection with hCG could improve the efficiency of pikeperch reproduction to reduce the number of brooders and costs of juvenile pikeperch production.

Evaluation of hormonal protocols for induction of synchronized estrus on reproductive indices in Kangal-Akkaraman ewes during the outbreeding season

The objective of this study was to assess the effect of human chorionic gonadotropin (hCG) and gonadotropin-releasing hormone (GnRH) treatment before sponge insertion and anti-Müllerian hormone (AMH) concentration in ewes which were either pregnant or non-pregnant during the previous breeding season. In the study, 375 (3–6-year-old) Kangal-Akkaraman ewes that conceived or did not conceive in the preceding breeding season, and 36 rams (3–6-year-old) were used. The sheep were randomly divided into 5 equal groups. Blood samples were taken before stimulation (day −4), the day stimulation was initiated (day 0), after stimulation (day 9), on the mating day (days 11–12). AMH concentrations were measured in the samples taken on − 4th day. As a conventional method, a sponge containing 20 mg of flugeston acetate (Chronogest® CR, MSD, Turkey) was placed intravaginally (day 0) into to study ewes. The ewes were intramuscularly administered 131.5 µg prostaglandin F2α (Estrumate, MSD, Turkey) and 600 IU equine chorionic gonadotropin (Chronogest® PMSG 6000, MSD, Turkey) simultaneously with sponge removal 9 days after insertion. No other treatment was applied to group 5 which served as control. Ewes in groups 1, 2, and 3 received an intramuscular injection of GnRH (4.2 µg of Buserelin acetate, Receptal, MSD, Turkey) at the time of insertion of progesterone sponge. GnRH or 600 IU hCG (Chorulon®, MSD, UK) was administered 4 days prior to sponge insertion for the ewes in the groups 2, and 3, respectively. As for the group 5 group, hCG (instead of GnRH) was administered to the ewes at the beginning of the progesterone treatment. Ewes in estrus were detected with a teaser ram and taken into a different pen with rams. Transrectal ultrasonography (Mindray DP50/Vet/US, rectal probe) was achieved during days the 30–35 following mating. Transabdominal ultrasonography was achieved during days 60–65 to detect late embryonic and early fetal death. The estrus rate was 71.19 % out of breeding season for all ewes. The pregnancy rate on day 30 post-mating was 32 % in the control group and was 31.00 %. No significant differences were observed between groups for pregnancy rates on day 30. The mean pregnancy rate in sheep that conceived and did not conceived in the preceding breeding season of the study was 32.03 % and 32.06 % respectively (P > 0.05). After treatments, AMH concentrations in non-pregnant were significantly higher than pregnant ewes (P < 0.005). In conclusion, although the AMH concentration is significantly higher in non-pregnant ewes, the AMH concentration may not be a suitable biomarker to predict induction success outside breeding season. Progesterone-based protocols may achieve an acceptable reproductive outcome in lactating ewes and the ones which experienced infertility.

Effects of human chorionic gonadotropin on the last day of a 5-day CIDR Synch protocol and 5 days later on circulating progesterone concentrations and luteal area in Holstein heifers

The objective was to determine the effect of replacing GnRH with human chorionic gonadotropin (hCG) on the last day of a 5-d CIDR Synch protocol (d 0) and inducing accessory corpus luteum (CL) formation with hCG 5 d later (d 5) on serum progesterone (P4) concentrations and luteal dynamics in dairy heifers. Holstein heifers (n = 207) were synchronized with a 5-d controlled internal drug release (CIDR) Synch protocol (d −8: used CIDR inserted; d −3: CIDR removed and PGF2α). Heifers were randomly assigned to 1 of 4 treatments on d 0: control (G; n = 55), H (n = 50), GH (n = 53), and HH (n = 49). Heifers in G were treated with 100 μg of GnRH on d 0, while H heifers received hCG (3,300 IU) on d 0. Heifers enrolled in GH were treated with GnRH on d 0 and hCG on d 5, while HH received hCG on d 0 and 5. Ovaries were scanned by ultrasound on d 0, 5, and 12, and blood was collected on d 0, 5, 7, and 12. Heifers that ovulated before or after the hCG or GnRH d 0 treatment and had P4 ≤ 0.50 ng/mL on d 0 were considered as synchronized. Overall protocol synchronization response was 93.8%, with no differences among treatments. Only synchronized heifers (n = 193) were included in the analyses of luteal dynamics after d 0. Serum P4 concentration and original CL luteal area on d 5 in heifers treated with hCG on d 0 (H + HH) were greater than in heifers treated with GnRH on d 0 (G + GH). Almost all heifers treated with hCG on d 5 had ≥2 CL on d 12 (98.6%). Ovulatory response for d 5 hCG treatment did not differ for GH versus HH (97.2 vs. 94.7%). Heifers in HH had the highest serum P4 on d 7, and G had the lowest serum P4 on d 7 and 12. In contrast, serum P4 on d 7 did not differ for H versus GH. On d 12, serum P4 and total luteal area were not different for GH versus HH. In summary, heifers that received hCG on d 0 had a larger total luteal area and greater serum P4 concentration on d 5 than heifers treated with GnRH on d 0. Moreover, hCG on d 5 promoted a greater proportion of heifers with ≥2 CL on d 12 and a larger luteal area of the original CL, which resulted in a larger total luteal area on d 12. The HH treatment successfully increased serum P4 concentrations in heifers on d 7 compared with the other treatments.

Comparative Study of the Restoring Effect of Metformin, Gonadotropin, and Allosteric Agonist of Luteinizing Hormone Receptor on Spermatogenesis in Male Rats with Streptozotocin-Induced Type 2 Diabetes Mellitus.

We compared the effectiveness of human chorionic gonadotropin (hCG; 5 days, 20 IU/rat/day), allosteric luteinizing hormone receptor agonist TP04 (5 days, 20 mg/kg/day), and metformin (28 days, 120 mg/kg/day) in restoring spermatogenesis in male rats with type 2 diabetes mellitus. hCG and TP04 increased the levels of testosterone and expression of the steroidogenic protein StAR, the number of spermatogenic cells, thickness of the seminal epithelium, and the number and motility of mature sperm that were reduced in diabetic rats, though they did not reduce the number of defective spermatozoa. Metformin had a weak effect on steroidogenesis, but was not inferior to luteinizing hormone receptor agonist by its restorative effect on spermatogenesis and also reduced the number of defective forms of spermatozoa. Thus, the spermatogenesis-restoring effect of metformin and luteinizing hormone receptor agonist in type 2 diabetes mellitus are comparable, despite different mechanisms of action.
