Quantitative Proteomics Analysis of ABA- and GA3-Treated Malbec Berries Reveals Insights into H2O2 Scavenging and Anthocyanin Dynamics.

Abscisic acid (ABA) and gibberellic acid (GA3) are regulators of fruit color and sugar levels, and the application of these hormones is a common practice in commercial vineyards dedicated to the production of table grapes. However, the effects of exogenous ABA and GA3 on wine cultivars remain unclear. We investigated the impact of ABA and GA3 application on Malbec grapevine berries across three developmental stages. We found similar patterns of berry total anthocyanin accumulation induced by both treatments, closely associated with berry H2O2 levels. Quantitative proteomics from berry skins revealed that ABA and GA3 positively modulated antioxidant defense proteins, mitigating H2O2. Consequently, proteins involved in phenylpropanoid biosynthesis were downregulated, leading to decreased anthocyanin content at the almost ripe stage, particularly petunidin-3-G and peonidin-3-G. Additionally, we noted increased levels of the non-anthocyanins E-viniferin and quercetin in the treated berries, which may enhance H2O2 scavenging at the almost ripe stage. Using a linear mixed-effects model, we found statistical significance for fixed effects including the berry H2O2 and sugar contents, demonstrating their roles in anthocyanin accumulation. In conclusion, our findings suggest a common molecular mechanism by which ABA and GA3 influence berry H2O2 content, ultimately impacting anthocyanin dynamics during ripening.

Enhancing Sugarcane Seedling Resilience to Water Stress through Exogenous Abscisic Acid: A Study on Antioxidant Enzymes and Phytohormone Dynamics.

Exogenous hormones play a crucial role in regulating plant growth, development, and stress tolerance. However, the effects of exogenous abscisic acid (ABA) on sugarcane seedlings under water stress remain poorly understood. Here, in this study, a pot experiment was conducted on sugarcane seedlings 4 weeks after transplanting, employing three treatments: control (normal growth), drought (water stress), and drought + ABA (foliar application of 100 μM ABA before water stress). The main objectives of this research are to understand the effects of exogenous ABA on sugarcane seedlings under water stress conditions and to assess the changes in antioxidant enzyme activity and phytohormone levels in response to exogenous ABA. Water stress was induced in the solution culture by adding 25% (w/v) polyethylene glycol (PEG) 6000 to the Hoagland solution. Leaf samples were collected at 3, 6, and 9 days after treatment, and the photosynthetic and biochemical responses of ABA-treated plants to drought stress were investigated. The indole acetic acid (IAA) activity of the ABA-treated drought plants is compared to that of drought plants. Moreover, the endogenous ABA levels of the ABA-treated drought plants were significantly enhanced by 42.2, 39.9, and 42.3% at 3, 6, and 9 days, respectively, compared to those of drought plants. Additionally, the proline content of the ABA-treated drought plants significantly increased by 45 and 80% at 6 and 9 days, respectively, compared to that of drought plants. The expression of the catalase 1 (CAT1) gene was increased in the ABA-treated drought plants by 2.1-fold, 0.7-fold, and 1.37-fold at 3, 6, and 9 days, respectively, compared to that in drought plants. Similarly, the expression of superoxide dismutase, peroxidase, and ascorbate peroxidase genes of the ABA-treated drought plants also increased compared to those of the drought plants. In conclusion, foliar application of ABA mitigated the negative effects of water shortage of sugarcane plants under water stress. Applying ABA improved the antioxidant defense system of sugarcane plants under drought stress, thereby enhancing their photosynthetic activities and productivity.

Exogenous abscisic acid (ABA) improves the filling process of maize grains at different ear positions by promoting starch accumulation and regulating hormone levels under high planting density

BackgroundHigher planting densities typically cause a decline in grain weight, limiting the potential for high maize yield. Additionally, variations in grain filling occur at different positions within the maize ear. Abscisic acid (ABA) is important for grain filling and regulates grain weight. However, the effects of exogenous ABA on the filling process of maize grains at different ear positions under high planting density are poorly understood. In this study, two summer maize hybrids (DengHai605 (DH605) and ZhengDan958 (ZD958)) commonly grown in China were used to examine the effects of ABA application during the flowering stage on grain filling properties, starch accumulation, starch biosynthesis associated enzyme activities, and hormone levels of maize grain (including inferior grain (IG) and superior grain (SG)) under high planting density.ResultsOur results showed that exogenous ABA significantly increased maize yield, primarily owing to a higher grain weight resulting from an accelerated grain filling rate relative to the control. There was no significant difference in yield between DH605 and ZD958 in the control and ABA treatments. Moreover, applying ABA promoted starch accumulation by raising the activities of sucrose synthase, ADP-glucose pyrophosphorylase, granule-bound starch synthases, soluble starch synthase, and starch branching enzyme in grains. It also increased the levels of zeatin riboside, indole-3-acetic acid, and ABA and decreased the level of gibberellin in grains, resulting in more efficient grain filling. Notably, IG exhibited a less efficient filling process compared to SG, probably due to lower starch biosynthesis associated enzyme activities and an imbalance in hormone contents. Nevertheless, IG displayed greater sensitivity to exogenous ABA than SG, suggesting that appropriate cultural measures to improve IG filling may be a viable strategy to further increase maize yield.ConclusionsAccording to our results, spraying exogenous ABA could effectively improve grain filling properties, accelerate starch accumulation by increasing relevant enzyme activities, and regulate hormone levels in grains, resulting in higher grain weight and yield of maize under high planting density. Our findings offer more evidence for using exogenous hormones to improve maize yield under high planting density.

Effects of Exogenous Abscisic Acid on the Physiological and Biochemical Responses of Camellia oleifera Seedlings under Drought Stress.

This study comprehensively investigates the physiological and molecular regulatory mechanisms of Camellia oleifera seedlings under drought stress with a soil moisture content of about 30%, where exogenous abscisic acid (ABA) was applied via foliar spraying at concentrations of 50 µg/L, 100 µg/L, and 200 µg/L. The results demonstrated that appropriate concentrations of ABA treatment can regulate the physiological state of the seedlings through multiple pathways, including photosynthesis, oxidative stress response, and osmotic balance, thereby aiding in the restructuring of their drought response strategy. ABA treatment effectively activated the antioxidant system by reducing stomatal conductance and moderately inhibiting the photosynthetic rate, thus alleviating oxidative damage caused by drought stress. Additionally, ABA treatment promoted the synthesis of osmotic regulators such as proline, maintaining cellular turgor stability and enhancing the plant's drought adaptability. The real-time quantitative PCR results of related genes indicated that ABA treatment enhanced the plant's response to the ABA signaling pathway and improved disease resistance by regulating the expression of related genes, while also enhancing membrane lipid stability. A comprehensive evaluation using a membership function approach suggested that 50 µg/L ABA treatment may be the most-effective in mitigating drought effects in practical applications, followed by 100 µg/L ABA. The application of 50 µg/L ABA for 7 h induced significant changes in various biochemical parameters, compared to a foliar water spray. Notably, superoxide dismutase activity increased by 17.94%, peroxidase activity by 30.27%, glutathione content by 12.41%, and proline levels by 25.76%. The content of soluble sugars and soluble proteins rose by 14.79% and 87.95%, respectively. Additionally, there was a significant decrease of 31.15% in the malondialdehyde levels.

Quality analysis and metabolomic profiling of the effects of exogenous abscisic acid on rabbiteye blueberry.

Blueberry is a characteristic berry fruit shrub of the genus Vaccinium in the Rhododendron family. The fruit is rich in anthocyanins and has a variety of nutritional and health functions. This study aimed to systematically study the effect of exogenous abscisic acid (ABA) application on ripening and metabolites in blueberry fruits. Blueberry fruit ripening was divided into six stages for further analysis. In this study, nontarget metabolomics was performed to demonstrate the effect on metabolite levels. The results showed that 1000 mg/L ABA significantly promoted fruit ripening and increased anthocyanin content. Moreover, exogenous ABA treatment can affect endogenous ABA levels and improve its antioxidant capacity. Important metabolites of the flavonoid pathway were detected, and the results showed that anthocyanin synthesis increased, and some other bioactive metabolite levels decreased. After comprehensive assessments, we believe that 1000 mg/L exogenous ABA application will have positive impacts on blueberry fruit quality and economic benefits.

Effects of exogenous abscisic acid on antioxidant system of salt tolerant and salt sensitive cotton cultivars

Summary Salinity is one of the most imperative global problems that affect crop productivity on a large scale. Salinity impairs plant growth and development by imposing various stresses. Therefore it is vital to decode those stress factors and identify possible solutions to improve agriculture productivity. However, the adaptive mechanisms under saline conditions of glycophytes have not been studied. The present study was undertaken to determine the effects of exogenous abscisic acid (ABA) on salinity tolerance in cotton plants. Some patterns of resistance development were revealed on the seedlings of two cotton cultivars, a salt-tolerant (Gulistan) and a salt-sensitive one (C-4727). Moreover, the antioxidant potentials of these cultivars were compared. The activities of ascorbate peroxidase (APX), super-oxide dismutase (SOD), as well as the quantity of endogenous ABA, malondialdehyde (MDA), and free proline (Pro) were determined in control and post treatment. Our results demonstrate significant differences between the salt- tolerant and sensitive cotton seedlings in response to saline stress, i.e., high levels of Pro and endogenous ABA, but lower MDA concentrations, and higher activity of APX and SOD for the salt-tolerant cultivar, Gulistan, as compared to the salt stress-sensitive cultivar C-4727.

Exogenous 1',4'-trans-Diol-ABA Induces Stress Tolerance by Affecting the Level of Gene Expression in Tobacco (Nicotiana tabacum L.).

1′,4′-trans-diol-ABA is a key precursor of the biosynthesis of abscisic acid (ABA) biosynthesis in fungi. We successfully obtained the pure compound from a mutant of Botrytis cinerea and explored its function and possible mechanism on plants by spraying 2 mg/L 1′,4′-trans-diol-ABA on tobacco leaves. Our results showed that this compound enhanced the drought tolerance of tobacco seedlings. A comparative transcriptome analysis showed that a large number of genes responded to the compound, exhibiting 1523 genes that were differentially expressed at 12 h, which increased to 1993 at 24 h and 3074 at 48 h, respectively. The enrichment analysis demonstrated that the differentially expressed genes (DEGs) were primarily enriched in pathways related to hormones and resistance. The DEGs of transcription factors were generally up-regulated and included the bHLH, bZIP, ERF, MYB, NAC, WRKY and HSF families. Moreover, the levels of expression of PYL/PYR, PP2C, SnRK2, and ABF at the ABA signaling pathway responded positively to exogenous 1′,4′-trans-diol-ABA. Among them, seven ABF transcripts that were detected were significantly up-regulated. In addition, the genes involved in salicylic acid, ethylene and jasmonic acid pathways, reactive oxygen species scavenging system, and other resistance related genes were primarily induced by 1′,4′-trans-diol-ABA. These findings indicated that treatment with 1′,4′-trans-diol-ABA could improve tolerance to plant abiotic stress and potential biotic resistance by regulating gene expression, similar to the effects of exogenous ABA.

In order to evaluate the effects of exogenous abscisic acid (ABA) in alleviating nickel (Ni) stress in wheat plants. We studied the changes of biochemical and physiological in wheat seedlings exposed to 250 μM Ni with or without different treatments of ABA. Exposed to Ni (250 μM) caused adverse effect on growth of wheat seedlings, which was accompanied by increased the concentrations of superoxide anion（O2−） and malondialdehyde (MDA). However, exogenous application of ABA (2.5 and 5 μM) alleviated the Ni-induced inhibition of plant growth, decreased the concentrations of O2− and MDA in wheat shoots. Further, application of ABA significantly modulated the activities of antioxidant enzymes and enhanced content of proline and soluble sugar in Ni-stressed wheats, but the application of 20 μM of ABA had no different significantly response for these parameters. The results indicated that application of ABA enhanced the antioxidant defense activities in Ni-stressed wheats, thus alleviating Ni-induced oxidative injury and enhancing Ni tolerance.

Previous studies in Siberian apricot (Prunus sibirica) seed kernel (SASK) have suggested the involvement of abscisic acid (ABA) signaling pathway in oil accumulation. However, there are few reports on the effects of ABA on the metabolism of fatty acids (FA) in seed development. Here, we first evaluated the response of developing SASK to ABA treatment, with a focus on oil content, FA composition, biodiesel properties, lipid compounds and gene expressions. Compared with control samples, the application of exogenous ABA increased the total oil content by 6.55% in mature SASK. The C18:1 content markedly increased in ABA treatment, and conversely C16:0 decreased. Exogenous ABA also improved the biodiesel properties of SASK oil, making it better suited to the specifications of biodiesel standards. Furthermore, the molecular species of phosphatidylcholine (PC), phosphatidic acid (PA), diacylglycerol (DAG) and triacylglycerol (TAG) were detected using lipidomics analysis. The 18:1/18:1 was the main component in PA, PC and DAG, while the main components of 18:1/18:1/18:2, 18:1/18:1/18:3, 18:2/18:2/18:2 and 18:1/18:1/18:1 in TAG. Most lipid species gradually increased with SASK maturity. In addition, the relative contents of TAG-18:1/18:1/18:2 and TAG-18:1/18:1/18:1 in developing SASK increased with the application of exogenous ABA. We also detected elevated gene expression of key genes involved in ABA chemical pathway, which likely affected FA biosynthesis and accumulation. Our results provide insight into the effects of ABA on the oil accumulation in developing SASK, which has direct applications to improving the quality of SASK-derived biodiesel.

In this study, water solutions with different concentrations of abscisic acid (ABA) (10, 20, and 40 μmol L-1) were sprayed on leaves of drought-stressed tobacco seedlings. The results showed that under drought stress, both water content and photosynthetic carbon assimilation ability were reduced, PSII activity was also inhibited. Drought stress also led to the outbreak of reactive oxygen species in tobacco leaves and increased the degree of membrane peroxidation. However, spraying exogenous ABA significantly alleviated the damage caused by drought stress on the leaves. The potential mechanisms by which drought resistance in tobacco is improved by spraying exogenous ABA include: (1) regulation of the stomatal limitation of leaves, which is beneficial to water retention ability and can enhance photosynthetic function; (2) reduction of the energy pressure of the PSII reaction center by increasing nonphotochemical quenching (NPQ); and (3) effective reduction of the production of reactive oxygen species in the leaves and reducing the degree of membrane peroxidation.

Effects of exogenous abscisic acid (ABA) on growth and physiological characteristics of Machilus yunnanensis seedlings under drought stress

In this study, the effects of exogenous abscisic acid (ABA) on the growth and physiolo-gical characteristics of Machilus yunnanensis seedlings were examined under well water supply conditions (70%-75% field capacity, FC) and different drought stress conditions, i.e., light drought stress (50%-55% FC) and severe drought stress (30%-35% FC), respectively. The results showed that drought stress reduced leaf relative water content, plant height, and total biomass of seedlings significantly decreased, as well as net photosynthetic rate and maximal photochemistry efficiency (Fv/Fm), but enhanced root/shoot and malondialdehyde contents (MDA). Exogenous ABA improved the adaptability of seedlings under drought stress, especially under severe drought stress, with leaf relative water content being improved 21.0%. Plant height and biomass allocation were improved by exogenous ABA under drought, while root/shoot was improved by 1.1 times compared with the well watered plants. The accumulation of MDA was decreased, the activities of CAT and SOD were significantly increased, and the proline content was increased 6.7 times by exogenous ABA under drought. Exogenous ABA application alleviated the negative effect of drought on photosynthetic organs, reduced the decrease of net photosynthetic rate and stomatal conductance under drought, and enhanced Fv/Fm value. The results suggested that exogenous ABA treatment could enhance the resistance of M. yunnanensis to drought stress.

Effects of Exogenous Abscisic Acid on Bioactive Components and Antioxidant Capacity of Postharvest Tomato during Ripening.

Abscisic acid (ABA) is a phytohormone which is involved in the regulation of tomato ripening. In this research, the effects of exogenous ABA on the bioactive components and antioxidant capacity of the tomato during postharvest ripening were evaluated. Mature green cherry tomatoes were infiltrated with either ABA (1.0 mM) or deionized water (control) and stored in the dark for 15 days at 20 °C with 90% relative humidity. Fruit colour, firmness, total phenolic and flavonoid contents, phenolic compounds, lycopene, ascorbic acid, enzymatic activities, and antioxidant capacity, as well as the expression of major genes related to phenolic compounds, were periodically monitored. The results revealed that exogenous ABA accelerated the accumulations of total phenolic and flavonoid contents; mostly increased the contents of detected phenolic compounds; enhanced FRAP and DPPH activity; and promoted the activities of PAL, POD, PPO, CAT, and APX during tomato ripening. Meanwhile, the expressions of the major genes (PAL1, C4H, 4CL2, CHS2, F3H, and FLS) involved in the phenylpropanoid pathway were up-regulated (1.13- to 26.95-fold) in the tomato during the first seven days after treatment. These findings indicated that ABA promoted the accumulation of bioactive components and the antioxidant capacity via the regulation of gene expression during tomato ripening.

ABSTRACTAbscisic acid (ABA) is an important hormone that can regulate the heavy-metal uptake of plants. In this study, exogenous ABA was applied to lettuce (Lactuca sativa) grown under cadmium (Cd)-stress conditions, and the effects of exogenous ABA on the growth and Cd accumulation of lettuce were evaluated. The results showed that treatment of Cd significantly decreased biomass and increased Cd content of lettuce compared with the control. The shoot biomass increased after spraying 1, 5 and 10 μmol/L ABA, and the root biomass increased after spraying 1–20 μmol/L ABA compared with the treatment of Cd. The Cd content in shoots significantly decreased when exposed to different ABA concentrations. The lowest Cd content in shoots of lettuce was produced by the ABA concentration of 5 μmol/L, with decreased by 23.60% compared with the treatment of Cd. Therefore, spraying ABA effectively alleviated Cd toxicity and reduced Cd uptake in the edible parts of lettuce, and 5 μmol/L ABA was the optimal dose.

Leaf rolling observed in some crops such as maize, rice, wheat and sorghum is an indicator of decreased water status. Moderate leaf rolling not tightly or early increases the photosynthesis and grain yield of crop cultivars under environmental stresses. Moreover, the effects of exogenous abscisic acid (ABA) on stomatal conductance, water status and synthesis of osmotic compounds are a well-known issue in plants subjected to water deficit. However, it is not clear how the cross-talk of ABA with H2O2 and osmolyte compounds affects the leaf rolling mechanism. Regulation mechanism of leaf rolling by ABA has been first studied in maize seedlings under drought stress induced by polyethylene glycol 6000 (PEG 6000) in this study. ABA treatment under drought stress reduced hydrogen peroxide (H2O2) content and the degree of leaf rolling (%) while the treatment-induced ABA synthesis, osmolyte levels (proline, polyamine and total soluble sugars) and some antioxidant enzyme activities in comparison to the plants that were not treated with ABA. Furthermore, exogenous ABA up-regulated the expression levels of arginine decarboxylase (ADC) and pyrroline-5-carboxylate synthase (P5CS) genes and down-regulated polyamine oxidase (PAO), diamine oxidase (DAO) and proline dehydrogenase (ProDH) gene expressions. When endogenous ABA content was decreased by the treatment of fluoridone (FLU) that is an ABA inhibitor, leaf rolling degree (%), H2O2 content and antioxidant enzyme activities increased, but osmolyte levels, ADC and P5CS gene expressions decreased. Finally, the treatment of ABA to maize seedlings exposed to drought stress resulted in the stimulation of the antioxidant system, osmotic adjustment and reduction of leaf rolling. We concluded that ABA can be a signal compound cross-talking H2O2, proline and polyamines and thus involved in the leaf rolling mechanism by providing osmotic adjustment. The results of this study can be used to provide data for the molecular breeding of maize hybrids with high grain yield by means of moderately rolled leaves.

Contribution of abscisic acid to aromatic volatiles in cherry tomato (Solanum lycopersicum L.) fruit during postharvest ripening

Fruit aroma development depends on ripening. Abscisic acid (ABA) has been reported to be involved in the regulation of tomato fruit ripening. In the present study, the effects of exogenous ABA on aromatic volatiles in tomato fruit during postharvest ripening were studied. The results showed that exogenous ABA accelerated color development and ethylene production as well as the accumulation of carotenoids, total phenolics and linoleic acid in tomato fruit during ripening. Moreover, exogenous ABA increased the accumulation of volatile compounds such as 1-peten-3-one (2.06-fold), β-damascenone (1.64-fold), benzaldehyde (3.29-fold) and benzyl cyanide (4.15-fold); induced the expression of key genes implicated in the biosynthesis pathways of aromatic volatiles, including TomloxC, HPL, ADH2, LeCCD1B and SlBCAT1 (the values of the log2 fold changes ranged from −3.02 to 2.97); and promoted the activities of lipoxygenase (LOX), hydroperoxide lyase (HPL) and alcohol dehydrogenase (ADH). In addition, the results of promoter analyses revealed that cis-acting elements involved in ABA responsiveness (ABREs) exist in 8 of the 12 key genes involved in volatile biosynthesis, suggesting that ABA potentially affects aromatic volatile emissions via the regulation of gene expression profiles.
