Clinical observation on the therapeutic effect of Hematoporphyrin Monomethyl Ether

SignificanceHematoporphyrin Monomethyl Ether (HMME) is a promising photosensitizer for photodynamic therapy (PDT) and has been widely applied in the treatment of port-wine stains (PWS). ApproachPatients with PWS were treated with HMME-PDT, and efficacy was evaluated by comparing photographs and CDFI examination. Results9 of the 33 cases were cured (27.27%), 10 indicated a good efficacy (30.30%), 11 showed alleviation (33.33%), while 3 had no obvious efficacy (9.09%). The efficacy was related to the types of lesions and the total number of therapeutic sessions. More sessions the purple type and nodular thickening type received, better clinical cosmetic results were. CDFI may be an excellent technique to evaluate efficacy. ConclusionsHMME-PDT serves as a safe and effective approach for improving PWS. The clinical therapeutic effect is affected by the type of lesions and the total number of therapeutic sessions. CDFI examination may be an excellent evaluation method.

Study of hematoporphyrin monomethyl ether-mediated blue-light photodynamic therapy in the treatment of oral ulcers infected with Staphylococcus aureus: In vivo evaluation.

Clinical suppurative infection is mainly caused by Staphylococcus aureus. Although many antibiotics can be used to kill S. aureus, the resulting resistance problem is difficult to solve. Therefore, it is necessary to seek a new sterilizing method to solve the problem of drug resistance of S. aureus and improve the therapeutic effect of infectious diseases. Photodynamic therapy (PDT) has become an alternative for the treatment of a variety of drug-resistant infectious diseases due to its advantages of non-invasive, specific targeting, and no drug resistance. We have confirmed the advantages and experimental parameters of blue-light PDT sterilization in vitro experiments. This study aimed to treat buccal mucosa ulcer of hamster infected with S. aureus according to the parameters obtained in vitro experiment, and observe the bactericidal effect of hematoporphyrin monomethyl ether (HMME) mediated blue-light PDT in vivo and its therapeutic effect on tissue infection. The results indicated that HMME mediated blue-light PDT can effectively kill S. aureus in vivo and promote the healing of the oral infectious wound.The study findings lay a foundation for carrying out more HMME mediated blue-light PDT sterilizing therapy.

Hematoporphyrin monomethyl ether mediated photodynamic therapy inhibits oral squamous cell carcinoma by regulating the P53-miR-21-PDCD4 axis via singlet oxygen.

The objective of this study was to determine the mechanism and effect of hematoporphyrin monomethyl ether mediated photodynamic therapy (HMME-PDT) on oral squamous cell carcinoma (OSCC). Human OSCC CAL-27 cells were randomly divided into four groups: control group, HMME group, laser group, and HMME-PDT group. Cell viability was detected by the CCK-8 method. Cell cycle distribution was evaluated by flow cytometry. GEO database was used to screen differentially expressed microRNAs (DEMs), and TCGA database was performed to verify DEM expression in OSCC and normal tissues. The effects of HMME-PDT on DEM expression were assayed by real-time PCR, and the expressions of miRNAs target genes were measured by western blot. Fluorescence probes were used to determine the production of singlet oxygen (1O2). Compared with the other three groups, HMME-PDT dramatically inhibited CAL-27 cell proliferation and induced G0/G1 cycle arrest. The expressions of miR-21 and miR-155 were significantly upregulated in OSCC. HMME-PDT downregulated the expression of miR-21 but had no obvious effect on miR-155. HMME-PDT remarkably upregulated the levels of P53 and miR-21 target proteins, such as PDCD4, RECK, and SPRY2. 1O2 was generated during HMME-PDT, and inhibition of 1O2 production could reverse the regulation of HMME-PDT on P53, miR-21, and its target proteins, thus restoring cell viability. HMME-PDT can significantly inhibit the growth of OSCC cells, and the mechanism of this effect is related to the regulation of the P53-miR-21-PDCD4 axis via 1O2 induced by HMME-PDT.

The penetration effect of HMME-mediated low-frequency and low-intensity ultrasound against the Staphylococcus aureus bacterial biofilm

BackgroundThe purpose of this study was to observe the effect of hematoporphyrin monomethyl ether (HMME)-mediated low-frequency and low-intensity ultrasound on mature and stable Staphylococcus aureus (S. aureus) biofilms under different ultrasound parameters.MethodsThe biofilm was formed after 48-h culture with stable concentration of bacterial solution. Different types of ultrasound and time were applied to the biofilm, and the ultrasonic type and time of our experiments were determined when the biofilm was not damaged. The penetration effects of low-frequency and low-intensity ultrasound were decided by the amount of HMME that penetrated into the biofilm which was determined by fluorescence spectrometry.ResultsThe destruction of biofilms by pulse waveform was the strongest. Sinusoidal low-frequency and low-intensity ultrasound can enhance the biofilm permeability. For a period of time after the ultrasound was applied, the biofilm permeability increased, however, changes faded away over time.ConclusionsLow-frequency and low-intensity sinusoidal ultrasound significantly increased the permeability of the biofilms, which was positively correlated with the time and the intensity of ultrasound. Simultaneous action of ultrasound and HMME was the most effective way to increase the permeability of the biofilms.

Sonodynamic effect of hematoporphyrin monomethyl ether on ligature-induced periodontitis in rats.

ObjectivesThe aim of this study was to perform a histological evaluation of sonodynamic therapy (SDT) of hematoporphyrin monomethyl ether (HMME) on artificially induced periodontal disease in rats.MethodsSubmerging ligatures were placed at the subgingival region of the first maxillary molar in rats. Eighty rats were randomly assigned into four groups: group 1 received no treatment; group 2 was subjected to 50 μg/mL HMME alone; group 3 was treated with low-intensity ultrasound alone (1 W/cm2); and group 4 was treated with 50 μg/mL HMME plus ultrasound irradiation (1 MHz, 30 minutes). Ten rats in each group were euthanized at 7 and 15 days, and periodontal tissue samples were taken for histological examination.ResultsThe animals treated by SDT showed less bone loss (P<0.05) at all experimental periods than the other three groups. No significant differences were found between the control and HMME groups (P>0.05).ConclusionOur results suggest that HMME-mediated SDT can effectively alleviate the periodontal tissue destruction in artificially induced periodontitis in rats. Hence, SDT may have good clinic potential as a noninvasive treatment of periodontal diseases.

Sonodynamic effects of hematoporphyrin monomethyl ether on Staphylococcus aureus in vitro.

This study aims to investigate the effect of hematoporphyrin monomethyl ether (HMME)-mediated sonodynamic antimicrobial chemotherapy (SACT) on Staphylococcus aureus. SACT was carried out using HMME and 1 MHz ultrasound irradiation. The bactericidal effect was evaluated by the counting colony-forming units (CFU), and important SACT parameters including ultrasound intensity and HMME concentration were determined. More than 95% of the bacteria colonies were effectively killed in the SACT group by 50 μg mL(-1) HMME combined with 6 W cm(-2) tone-burst ultrasound at 1 MHz, but this ultrasound level without HMME only reduced CFU by 38%. In the sonodynamic treatment, higher HMME concentrations and higher ultrasound intensities caused more death of bacteria. Incubation with different HMME concentrations without ultrasound showed no effect. Our results show that the HMME-mediated SACT can be significantly in killing S. aureus.

Study of the mechanism of sonodynamic therapy in a rat glioma model.

PurposeThe study reported here examined the effect of hematoporphyrin monomethyl ether (HMME)-mediated sonodynamic therapy (SDT) on C6 gliomas implanted in rat brains.MethodsTwo weeks after inoculation, glioma development was evaluated by measuring tumor volume using a 1.5 T magnetic resonance imager. Rats that had a well-developed C6 glioma (usually when the tumor diameter reached 3–5 mm) were used to test SDT, ultrasound-alone, and HMME-alone treatments. Rats both administered and not administered intravenous HMME 10 μg/mL were insonated by a 1 MHz ultrasound at a dose of 0.5 W/cm2.ResultsSDT treatment could effectively inhibit the expansion of intracranial gliomas in vivo. The treatment with ultrasound alone could inhibit glioma growth within 1 week; however, 1 week later, the tumor started growing again. In contrast, the effect of SDT could last at least 2 weeks. Injection of HMME alone had no effects on inhibiting glioma growth, suggesting the sonosensitizer HMME has no antitumor effect. Both SDT and ultrasound-alone treatment could extend the survival of rats implanted with a C6 glioma. Pathological and electron microscopic examinations suggested SDT and ultrasound-alone treatment could induce glioma necrosis by way of triggering glioma-cell apoptosis, which was confirmed by immunohistological examination with cytochrome-c and caspase-3 antibodies. Most importantly, we found that the sonosensitizer HMME could enhance the ultrasound-induced antitumor effect by selectively assisting ultrasound targeting of glioma angiogenesis inhibition.ConclusionThis study with a rat C6 glioma experimental model showed that SDT can potentially be useful in the treatment of deep-seated malignant gliomas.

Effects of hematoporphyrin monomethyl ether (HMME) on Worker Behavior of red imported fire ant, Solenopsis invcita

The effect of hematoporphyrin monomethyl ether (HMME) activated under visible light on worker behavior of Solenopsis invcita was studied with the potter spray tower method. The results showed that greater than 10 mg/L HMME activated under visible light could reduce the walking, grasping, aggregation, and water and food recognition abilities of red imported fire worker ant significantly, but 100 mg/L HMME in darkness could not affect their abilities or behaviors significantly. Therefore, HMME may be a potential novel insecticide that can be used as a substitute for toxic insecticides for controlling red imported fire ants.
