Commentary for “antagonizing urotensin receptor is a novel therapeutic strategy for glucocorticoid‐induced skeletal muscle atrophy”

In 1950, the discovery of the glucocorticoid cortisone was awarded the Nobel Prize in Physiology and Medicine. Since then, glucocorticoids have been a mainstay in clinical practice. Endogenous glucocorticoids like cortisone are natural hormones released in response to stress or circadian stimuli, whereas synthetic glucocorticoids like dexamethasone are used as potent anti-inflammatory drugs. While synthetic glucocorticoids are the mainstay treatments for many clinical conditions, their chronic daily intake also induces a plethora of concerning adverse effects, including muscle loss. The debilitating effects of chronic daily glucocorticoids in muscle are generally evident quite rapidly and compromise muscle mass, structure and strength in the long term. Glucocorticoid-induced muscle wasting correlates with increased protein catabolism, muscle mass decrease and inhibition of endogenous regenerative potential.1, 2 The neuropeptide urotensin II (UII) and its receptor, UII receptor, are expressed in a variety of tissues, including the skeletal muscle. Impaired regulation of UII and UII receptor have been associated with various pathologies.3 Of relevance for this study, prior studies had reported on the correlation between UII and muscle atrophy and on the finding that UII promotes loss of muscle weight and strength.4 In the study published here, Yin et al., show a novel approach in which a UII receptor antagonist rescues the glucocorticoid-induced skeletal muscle atrophy in mice. The study reports a potentially clinically relevant effect of Palosuran, a potent and selective antagonist of the UII receptor, in preventing dexamethasone-induced muscle atrophy. This experimental study has a huge potential and, if confirmed in clinical studies, could alleviate the muscle wasting experienced by the many people who take glucocorticoids regularly. Though this is the first report using this drug to counteract muscle atrophy, Palosuran has been largely investigated in the context of kidney function, particularly diabetic nephropathy. Experimental studies in rodents have reported on the potential role of UII receptor antagonists in alleviating renal complications in diabetes.5 However, initial clinical studies in humans have provided mixed results with this drug in the treatment of diabetic nephropathy.6, 7 In the study presented here, the Authors investigate a novel context for this drug and detail a large set of compelling experiments to elucidate the muscle atrophy counteraction by Palosuran. Importantly, their pharmacological experiments were complemented by analyses in a transgenic mouse with body-wide germline knockout of the UII receptor gene. Taken together, the findings reported in this study show that UII antagonism improves muscle mass and function in the context of dexamethasone-induced muscle wasting. Several limitations and open questions need further discussion. While inhibition of the UII axis through UII receptor antagonists has shown promising results in experimental animals, a potent and safe antagonist that can be used in humans is yet to be found. While the results by Yin et al, are promising, additional mechanistic and clinical studies are needed to support the novel indication of Palosuran for glucocorticoid-induced muscle loss. For instance, the experiments presented here should be investigated in disease models where chronic glucocorticoid intake is the mainstay treatment. Moreover, many tissues in the body express the UII receptor and therefore a thorough evaluation of toxicity and off-target effects of UII antagonist regimens is warranted. Additionally, it should also be evaluated if Palosuran alters or interacts with the anti-inflammatory action of glucocorticoid drugs. The results presented in this study depict a rather convincing effect of Palosuran on counteracting the loss of muscle mass and strength. However, the study did not address the effects of UII signalling and its counteraction on individual myofiber types, particularly type 2A, 2B and 2X. Moreover, the effects of UII axis activation versus inhibition on muscle bioenergetics remain unknown. Nevertheless, the rich dataset presented in this study supports the novel idea of repurposing UII antagonists to treat or prevent glucocorticoid-induced muscle atrophy. This is a significant advancement not only to potentially reduce the iatrogenic burden of synthetic glucocorticoids but also to further decipher their multifaceted action in health and disease. Drs. One of them is the book of Rāhatu Ṣālihin wa Ṣowā‘iqu Al-Munafiqīn which is a religious script written in Arabic language and has several overarching articles for the advancement of islamic religious knowladge this research examined the manuscript in depth. This study revealed that the manuscript contains wich information about the articles conveering with worship procedures for muslim. Method of philology was used in the research by applying steps such as inventory of manuscript, description of manuscript, editing of manuscript, text translation and analysis of the manuscript contents.

Transatlantic Visions and Revisions of Race: Hawthorne, Joseph Jenkins Roberts, and the Editing ofJournal of an African Cruiser

A strong argument can be made that Journal of an African Cruiser, by an Officer of U.S. Navy (1845) should be included in The Centenary Edition of Works of Nathaniel Hawthorne, for evidence indicates that his editing of manuscript of his friend Horatio Bridge was a major creative effort. When Hawthorne submitted edited text to Evert Duyckinck, who published book as first volume of Wiley and Putnam's of America Books, he admitted, My own share of it is so amalgamated with substance of work, that I cannot very well define what it is. (1) one of few substantial studies of project, Patrick Brancaccio observes that Hawthorne served more as a ghostwriter than editor, and points out that the thematic organization and ironic and morally ambiguous point of view clearly betray Hawthorne's (Brancaccio 33). A comparison of a portion of Bridge's 1843-44 manuscript journal (in Berg Collection at New York Public Library) with published book has revealed that Hawthorne ameliorated Bridge's racism, softened his callousness, and made him a more introspective and peace-loving author, much like Hawthorne himself. (2) Hawthorne, however, I have found, was not only editor of Journal of an African Cruiser. Joseph Jenkins Roberts, black Governor of Liberia, edited book as well, at request of Bridge, who took published book with him to Africa in 1846, on his third cruise to region, and asked his friend Roberts to offer corrections and revisions for a second edition, which Roberts did. The bound 1845 book contains Roberts' marginalia in ink on pages and his commentary in Bridge's hand on pieces of paper of various sizes, tipped onto printed pages, with quotation marks and Roberts' initials. The expanded and revised second edition of Journal of an African Cruiser was never published (a reprint of first edition appeared in 1853, with Bridge's name on title page for first time); however, materials Bridge collected exist in Bridge's personal copy of book and in 1846 manuscript journal Bridge kept on his voyage (both preserved in Bowdoin College Library Special Collections). front and back of 1846 journal, Bridge wrote following note: In case of my death I wish this journal and my copy of 'Journal of an African Cruiser' in which I have made notes, given to my friend Nath. Hawthorne Esq of Salem Mass. For his own use & benefit. (3) Bridge obviously hoped that someday he or Hawthorne could prepare new volume for publication. Some of topics Bridge elaborated upon in his new material were price of slaves, forms of government of native tribes, native religion, trial of witchcraft, racial mixing of Britains and natives on Gold Coast, vices of civilization acquired by natives, and nature of fever and sickness visited upon outsiders to region. For most part, Governor Roberts added little on these topics. What he addressed most frequently and firmly with his comments and corrections were racial issues, specifically assumptions Bridge and Hawthorne made about importance of race among whites, black settlers, and West African natives. As I hope to show in this essay, an examination of Roberts' diplomatic critique of Bridge-Hawthorne book provides a rare opportunity to see a transatlantic social text being created. The process features a self-taught American-African trying to educate two white Bowdoin College graduates living across Atlantic. I his 1846 journal, Bridge acknowledged help of both friends in his ongoing authorship: When I returned from an African cruise and gave my journal and memoranda to a close friend and classmate [Hawthorne] to be edited and published, it was with little expectation of producing, by our joint labors, so successful a volume. Richard Wright's search for a counter-hegemonic genre: the anamorphic and matrixial potential of haiku

Examining Richard Wright's vast haiku oeuvre, this article shows how he used haiku to reinvent the form in English by repeating playfully anamorphic imagery so as to construct a poetic matrix modelled on, but distinct from, the categories he saw used in Japanese haiku. Writing haiku in this productive manner produced Nietzschean jouissance rather than resentment, so that the affective dimension of a multitude of socio-economic relations, including race, could be demonstrably reframed in this matrix of anamorphic imagery which maintained political allegories and critical consciousness in the landscapes of his invention. He asserted measured displacements and anamorphic transvaluations of how one sees (in Jacques Rancière's sense), and as such presented a modernist haiku constructed not in isolate verses (as haiku are often read), but in refrains and nodes, suggesting an intertextual matrix asserting new commonplaces (locus communis) and self-evident ways of seeing. This article also points out the lack of evidence for the current consensus in literary criticism which mistakenly asserts that Wright discovered a Zen spirit which elevated his spirit and brought him closer to nature. Moreover, the editing of the book manuscript is shown to be not only flawed in its critical framing, but in the very ordering of haiku, presentation, and even title.

DAMASCENE and Meta-Ecological Research: A Bridge Too Far.

Population level studies commonly induce the ecological fallacy, i.e., the belief that a relationship identified at the population level implies a cause and effect relationship at the level of the individual. A recent research report (DAMASCENE) that related clinical trial effect sizes in cardiovascular cell therapy trials to reporting discrepancies in the trials themselves has compounded meta-analysis difficulties with ecological fallacy issues. This dangerous combination has produced misleading conclusions and speculation that the research community should set aside.

Over the past 2 years, multiple clinical trials have reported results that will influence the treatment of patients with non-ST-segment elevation acute coronary syndromes (ACS) for many years to come. However, large-scale clinical trials take years to complete, during which time the underlying landscape may shift. Thus, while clinical trials provide baseline information to help physicians make evidence-based decisions regarding patient care, trials must be interpreted in the context of current treatment guidelines and practices. In addition, regulatory and advisory board decisions, case reports, clinician experience, and patient factors have a clear but difficult-to-measure impact on practice patterns. In recognition of the wide range of information affecting physicians' decision-making processes in patients with ACS, a roundtable was convened on June 5, 2009, in New York City to discuss the implications of recent data for clinical practice. Eight clinicians from the disciplines of cardiology and emergency medicine shared information and opinions on recent advances in antiplatelet treatment, clinical trial results, guidelines, and other issues related to patient care. This article is derived from transcripts of their presentations and the surrounding discussions at this meeting, with the intent of reporting both quantitative information on recent advances in the management of ACS and qualitative information and opinions from participants. Each author held primary responsibility for the writing and editing of his or her section, and participated in the editing of the entire manuscript.

BJOG trainee editors

Do the editors of BJOG misunderstand your work? Are they slow to deal with your papers? Do they send them to unsuitable referees? Could you do better? Now is your chance. Next year BJOG will create two new posts of trainee editor. Each editorship will be for one year, renewable for a further year subject to satisfactory performance. The positions are open to any member of the RCOG in a sub-consultant grade. The closing date for applications is 31 June 2004 and interviews will be held in September. We are looking for clinician/scientists who can also write well. We will judge candidates' writing skills on the basis of a 1000-word editorial on a topic of their choosing and on their editing of a real manuscript. We want people who can make poorly written manuscripts clear, without distorting the sense. We will judge applicants' scientific achievements from their curriculum vitae. The BJOG Editors will make the appointment. Each successful candidate will work with one Editor as mentor. They will select referees, present manuscripts at editors' meetings and revise accepted ones. Like the main Editors, trainees need not attend all editors meetings; the expectation will be six per year. These are all held at the College. The posts are honorary but expenses will be paid. Trainees will be encouraged to undertake a research project on peer review or some other aspect of scientific publishing. Electronic manuscript tracking and publishing provide good data for analysis and many opportunities to introduce experimental interventions into the publishing process. The Species of Ciborinia Pathogenic to Salix, Magnolia, and Quercus

Illus. 1960.There are 2 species of Ciborinia that attack various species of Salix. One of these, C. wisconsinensis, is reported as new and its phytopathology and life history are discussed. This species forms sclerotia in the leaves along the midrib. The sclerotia germinate in the year following infection and form apothecia in late June. In culture, sclerotia formed from mass ascospore-shootings produce apothecia. Ciborinia candolleana is reported for the first time attacking Quercus spp. in the United States. A key to the known species of the genus Ciborinia is provided. THIS study of the genus Ciborinia Whetzel is a continuation of previous work (Batra and Korf, 1959). Most of the species of Ciborinia attacking woody angiosperms form sclerotia along the vascular bundles, primarily encircling the midrib and the major veins. Generally the selerotia are formed late in the season, after the leaves have fallen on the ground. With the exception of one species, C. violae Batra and Korf, the species of Ciborinia attacking herbaceous angiosperms form sclerotia scattered all over the leaves while they are still green and attached to the plant. The materials investigated and the methods employed are essentially the same as reported earlier (Batra and Korf, 1959). Starbick's (1895) concept of discomycete tissue anatomy elaborated by Korf (1951, 1958) has proved to be very useful in classifying the species of Ciborinia. Most of the work reported here was done at Cornell University during the period 1956-59. Although no extensive study was made of the species attacking, Populus L., they are being included in the following key and the reader is referred to the critical study done by Groves and Bowerman (1955). Four species, Ciborinia foliicola (Cash and Davidson) Whet., C. wisoonsinensis, a new species, C. gracilipes (Cooke) Seaver, and C. candolleana (Lev.) Whet. are described in detail. Ciborinia hirtella (Boud.) Batra and Korf, not so far recorded from the United States, is also included in the key. All the material pertaining to this research is deposited in the herbarium of the Department of Plant Pathology, Cornell University (CUP), Ithaca, N. Y. Names of the herbaria where the specimens referred to in this paper are located are taken from Lanjouw and Stafleu
