Event Abstract Back to Event Early seizures following hemorrhagic stroke in the rat collagenase model Charlotte Germonpré1*, Robrecht Raedt1, Paul Boon1 and Veerle De Herdt1 1 Ghent University, Belgium AIM Intracerebral hemorrhage (ICH) is a known risk factor for the development of seizures, but little is known about the incidence and consequence of seizures in the acute phase post-ICH. The aim of this study was to optimize the rat collagenase model for the detection of early seizures. Therefore, ICH was induced by injecting collagenase in left striatum and the occurrence of seizures was assessed. METHODS Thirteen male Sprague-Dawley rats were implanted with scalp electrodes (4 AP, +/-2 ML and -4.5 AP, +/-3 ML, coordinates in mm and relative to bregma) and a craniotomy (0.5 AP, 3.5 ML) was made for later injection of collagenase and filled with bone wax. After one week of baseline video-EEG recording, bone wax was removed and rats were injected at a depth of 6 mm with 0.6 U collagenase dissolved in 0.7 µl saline (n=11) or saline (n=2) and immediately reconnected to the video-EEG setup during 7 days. Afterwards animals were euthanized to visualize the hemorrhage by histology and video-EEG recordings were analyzed. RESULTS Collagenase induced an intracerebral hemorrhage in all animals with a mean volume of 26.5 ±7.4 mm³. Epileptic seizures occurred in 5/11 animals, starting between 1h and 61h after ICH induction, with a mean seizure frequency of 3 seizures/animal (±0.84). Saline injection did not result in hemorrhage nor epileptic seizures. CONCLUSION Injecting rats with 0.6 U of collagenase is a useful model to study early post-stroke seizures as it results in hemorrhagic stroke in all animals without mortality and a 45% incidence rate of stroke-induced early seizures.
