Ethylene promotes fruit ripening initiation by downregulating photosynthesis, enhancing abscisic acid and suppressing jasmonic acid in blueberry (Vaccinium ashei).

Blueberry fruit exhibit atypical climacteric ripening with a non-auto-catalytic increase in ethylene coincident with initiation of ripening. Further, application of ethephon, an ethylene-releasing plant growth regulator, accelerates ripening by increasing the proportion of ripe (blue) fruit as compared to the control treatment. To investigate the mechanistic role of ethylene in regulating blueberry ripening, we performed transcriptome analysis on fruit treated with ethephon, an ethylene-releasing plant growth regulator. RNA-Sequencing was performed on two sets of rabbiteye blueberry ('Powderblue') fruit: (1) fruit from divergent developmental stages; and (2) fruit treated with ethephon, an ethylene-releasing compound. Differentially expressed genes (DEGs) from divergent developmental stages clustered into nine groups, among which cluster 1 displayed reduction in expression during ripening initiation and was enriched with photosynthesis related genes, while cluster 7 displayed increased expression during ripening and was enriched with aromatic-amino acid family catabolism genes, suggesting stimulation of anthocyanin biosynthesis. More DEGs were apparent at 1 day after ethephon treatment suggesting its early influence during ripening initiation. Overall, a higher number of genes were downregulated in response to ethylene. Many of these overlapped with cluster 1 genes, indicating that ethylene-mediated downregulation of photosynthesis is an important developmental event during the ripening transition. Analyses of DEGs in response to ethylene also indicated interplay among phytohormones. Ethylene positively regulated abscisic acid (ABA), negatively regulated jasmonates (JAs), and influenced auxin (IAA) metabolism and signaling genes. Phytohormone quantification supported these effects of ethylene, indicating coordination of blueberry fruit ripening by ethylene. This study provides insights into the role of ethylene in blueberry fruit ripening. Ethylene initiates blueberry ripening by downregulating photosynthesis-related genes. Also, ethylene regulates phytohormone-metabolism and signaling related genes, increases ABA, and decreases JA concentrations. Together, these results indicate that interplay among multiple phytohormones regulates the progression of ripening, and that ethylene is an important coordinator of such interactions during blueberry fruit ripening.

Melatonin seed priming improves early establishment and water stress tolerance of peanut

Water stress is a major cause of yield loss in peanut cultivation. Melatonin seed priming has been used to enhance stress tolerance in several crops, but not in peanut. We investigated the impact of seed priming with melatonin on the growth, development, and drought tolerance of two peanut cultivars, TUFRunner™ ‘511’, a drought tolerant cultivar, and New Mexico Valencia A, a drought sensitive cultivar. Peanut seed priming tests using variable rates of melatonin (0–200 μM), indicated that 50 μM of melatonin resulted in more uniform seed germination and improved seedling growth in both cultivars under non stress conditions. Seed priming with melatonin also promoted vegetative growth, as evidenced by higher whole-plant transpiration, net CO2 assimilation, and root water uptake under both well-watered and water stress conditions in both cultivars. Higher antioxidant activity and protective osmolyte accumulation, lower reactive oxygen species accumulation and membrane damage were observed in primed compared with non-primed plants. Seed priming with melatonin induced a growth promoting effect that was more evident under well-watered conditions for TUFRunnner™ ‘511’, whereas for New Mexico Valencia A, major differences in physiological responses were observed under water stress conditions. New Mexico Valencia A primed plants exhibited a more sensitized stress response, with faster down-regulation of photosynthesis and transpiration compared with non-primed plants. The results demonstrate that melatonin seed priming has significant potential to improve early establishment and promote growth of peanut under optimal conditions, while also improve stress tolerance during water stress.

Metabolic Rewiring in Tea Plants in Response to Gray Blight Disease Unveiled by Multi-Omics Analysis.

Gray blight disease, which is caused by Pestalotiopsis-like species, poses significant challenges to global tea production. However, the comprehensive metabolic responses of tea plants during gray blight infection remain understudied. Here, we employed a multi-omics strategy to characterize the temporal transcriptomic and metabolomic changes in tea plants during infection by Pseudopestalotiopsis theae, the causal agent of gray blight. Untargeted metabolomic profiling with ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOFMS) revealed extensive metabolic rewiring over the course of infection, particularly within 24 h post-inoculation. A total of 64 differentially accumulated metabolites were identified, including elevated levels of antimicrobial compounds such as caffeine and (-)-epigallocatechin 3-gallate, as well as oxidative catechin polymers like theaflavins, theasinensins and theacitrins. Conversely, the synthesis of (+)-catechin, (-)-epicatechin, oligomeric proanthocyanidins and flavonol glycosides decreased. Integrated omics analyses uncovered up-regulation of phenylpropanoid, flavonoid, lignin biosynthesis and down-regulation of photosynthesis in response to the pathogen stress. This study provides novel insights into the defense strategies of tea plants against gray blight disease, offering potential targets for disease control and crop improvement.

Tracking autumn photosynthetic phenology on Tibetan plateau grassland with the green–red vegetation index

Accurate monitoring autumn photosynthetic phenology is essential for understanding carbon cycles. The broadband green-red vegetation index (GRVI) derived from broadband red and green reflectance has been increasingly used in this field. However, the performance of GRVI in large areas remains unclear. We evaluated the performance of the normalized vegetation index (NDVI), normalized difference greenness index (NDGI), the near-infrared reflectance of vegetation (NIRv), solar-induced chlorophyll fluorescence (SIF) and GRVI in tracking autumn photosynthetic phenology of alpine grassland at flux sites and the entire Tibetan Plateau (TP). The results revealed that GRVI (R2 = 0.42, RMSE = 7.68 d and Bias = 4.39 d) performed comparable with SIF (R2 = 0.45, RMSE = 5.79 d and Bias = 0.71 d) in extracting the end of the photosynthetically active season (EOS) with eddy covariance flux measurements as reference. On contrary, a systematically later EOS was estimated by NDVI (Bias = 20.35 d), NDGI (Bias = 13.62 d) and NIRv (Bias = 6.56 d). The application example on the TP collaborated these findings. The divergent performances between indicators were rooted in the photosynthesis downregulation in autumn which is jointly controlled by canopy structure and physiology. Due to the fact that NDVI, NDGI and NIRv primarily depict vegetation structure, while the use of SIF, which represents vegetation photosynthetic physiology, is influenced by the limited spatial resolution and temporal coverage. Our study highlights the unique advantage of GRVI over other existing satellite indicators for estimating autumn photosynthetic phenology with high resolution and long-time span. We suggest revisiting the dynamics of autumn photosynthetic phenology using GRVI, which has significant implications on carbon uptake studies.

Microplastics Weaken the Adaptability of Cyanobacterium Synechococcus sp. to Ocean Warming.

Ocean warming (OW) caused by anthropogenic activities threatens ocean ecosystems. Moreover, microplastic (MP) pollution in the global ocean is also increasing. However, the combined effects of OW and MPs on marine phytoplankton are unclear. Synechococcus sp., the most ubiquitous autotrophic cyanobacterium, was used to evaluate the response to OW + MPs under two warming scenarios (28 and 32 °C compared to 24 °C). The enhancement of the cell growth rate and carbon fixation under OW were weakened by MP exposure. Specifically, OW + MPs reduced carbon fixation by 10.9 and 15.4% at 28 and 32 °C, respectively. In addition, reduction in photosynthesis pigment contents of Synechococcus sp. under OW was intensified under OW + MPs, supporting the lower growth rate and carbon fixation under OW + MPs. Transcriptome plasticity (the evolutionary and adaptive potential of gene expression in response to changing environments) enabled Synechococcus sp. to develop a warming-adaptive transcriptional profile (downregulation of photosynthesis and CO2 fixation) under OW. Nevertheless, the downregulation of photosynthesis and CO2 fixation were alleviated under OW + MPs to increase responsiveness to the adverse effect. Due to the high abundances of Synechococcus sp. and its contributions to primary production, these findings are important for understanding the effects of MPs on carbon fixation and ocean carbon fluxes under global warming.

The effect of elevated CO2 on photosynthesis is modulated by nitrogen supply and reduced water availability in Picea abies.

It is assumed that the stimulatory effects of elevated CO2 concentration ([CO2]) on photosynthesis and growth may be substantially reduced by co-occurring environmental factors and the length of CO2 treatment. Here, we present the study exploring the interactive effects of three manipulated factors ([CO2], nitrogen supply and water availability) on physiological (gas-exchange and chlorophyll fluorescence), morphological and stoichiometric traits of Norway spruce (Picea abies) saplings after 2 and 3 years of the treatment under natural field conditions. Such multifactorial studies, going beyond two-way interactions, have received only limited attention until now. Our findings imply a significant reduction of [CO2]-enhanced rate of CO2 assimilation under reduced water availability which deepens with the severity of water depletion. Similarly, insufficient nitrogen availability leads to a down-regulation of photosynthesis under elevated [CO2] being particularly associated with reduced carboxylation efficiency of the Rubisco enzyme. Such adjustments in the photosynthesis machinery result in the stimulation of water-use efficiency under elevated [CO2] only when it is combined with a high nitrogen supply and reduced water availability. These findings indicate limited effects of elevated [CO2] on carbon uptake in temperate coniferous forests when combined with naturally low nitrogen availability and intensifying droughts during the summer periods. Such interactions have to be incorporated into the mechanistic models predicting changes in terrestrial carbon sequestration and forest growth in the future.

Differential Accumulation of sHSPs Isoforms during Desiccation of the Resurrection Plant Haberlea rhodopensis Friv. under Optimal and High Temperature.

Haberlea rhodopensis belongs to the small group of angiosperms that can survive desiccation to air-dry state and quickly restore their metabolism upon rehydration. In the present study, we investigated the accumulation of sHSPs and the extent of non-photochemical quenching during the downregulation of photosynthesis in H. rhodopensis leaves under desiccation at optimum (23 °C) and high temperature (38 °C). Desiccation of plants at 38 °C caused a stronger reduction in photosynthetic activity and corresponding enhancement in thermal energy dissipation. The accumulation of sHSPs was investigated by Western blot. While no expression of sHPSs was detected in the unstressed control sample, exposure of well-hydrated plants to high temperature induced an accumulation of sHSPs. Only a faint signal was observed at 50% RWC when dehydration was applied at 23 °C. Several cross-reacting polypeptide bands in the range of 16.5-19 kDa were observed in plants desiccated at high temperature. Two-dimensional electrophoresis and immunoblotting revealed the presence of several sHSPs with close molecular masses and pIs in the range of 5-8.0 that differed for each stage of treatment. At the latest stages of desiccation, fourteen different sHSPs could be distinguished, indicating that sHSPs might play a crucial role in H. rhodopensis under dehydration at high temperatures.

ABSTRACT The simultaneous effect of elevated CO2 concentration and drought on trees is still under investigation in the Amazon. We evaluated the effect of CO2 levels (400 and 800 ppm) and water regimes (50% and 100% soil field capacity) on photosynthetic traits, chlorophyll fluorescence, and total biomass accumulation in Ceiba pentandra. In well-watered plants, light-saturated photosynthesis (PN-sat) increased in plants exposed to elevated CO2, but both PN-sat and stomatal conductance decreased in response to water deficit. The maximum carboxylation rate of Rubisco declined under elevated CO2, which indicates down-regulation of photosynthesis at elevated CO2. The Fv/Fm ratio was not affected by treatments. Notwithstanding, total plant biomass and leaf area were reduced by 34-37% under water deficit, but they were not affected by CO2 levels. The PN-sat values measured in well-irrigated plants at ambient CO2 were similar to those observed in plants subjected to elevated CO2 and water deficit (p = 0.26). We concluded that the effect of water deficit on PN-sat was mitigated by elevated CO2. These results suggest that the increase of atmospheric CO2 concentrations associated to climate changes can at least partly offset the negative effect of drought in this multiuse and widely distributed species.

Factors causing different CO2 responses in shoot regrowth after defoliation between two grassland species in Mongolia

To address the interspecific differences in CO2 responses of plant regrowth after grazing, we grew two Mongolian grasses, Agropyron cristatum and Stipa krylovii, which are highly palatable to livestock, under ambient or elevated CO2 concentration ([CO2]) and subjected them to shoot clipping. We found two major differences under elevated [CO2]: a larger increase in shoot regrowth in A. cristatum than in S. krylovii and a decrease in the degree of biomass compensation (the ratio of shoot mass of clipped plants plus mass of clippings to shoot mass of non-clipped plants) in A. cristatum but not in S. krylovii. The former resulted from a decrease in net assimilation rate in S. krylovii. The latter was caused by a larger increase in shoot growth in non-clipped than in clipped plants under elevated [CO2] in A. cristatum, which resulted from an increase in biomass partitioning to shoots caused by elevated [CO2] in non-clipped plants. Our results imply that the productivity increase caused by elevated [CO2] would be higher in A. cristatum-dominated than in S. krylovii-dominated grasslands under grazing. Downregulation of photosynthesis at elevated [CO2] and CO2-induced changes in biomass partitioning may contribute to the interspecific differences in CO2 responses in the regrowth of grass species.

The effect of nut growth limitation on triose phosphate utilization and downregulation of photosynthesis in almond.

There is a controversy regarding when it is appropriate to apply the irrigation restriction in almond trees (Prunus dulcis Mill.) to save water without penalizing yield. We hypothesized that knowing when plants demand fewer photoassimilates would be a good indicator of less sensitivity of the crop to water deficit. One parameter that defines the photosynthetic capacity is the triose phosphate utilization (TPU). Due to its connection to the export of sugars from the leaves to other sink organs, it is a good candidate for being such an indicator. The objective was to analyze the seasonal evolution of the photosynthetic capacity of three almond cultivars (cvs Guara, Marta and Lauranne) subjected to water stress during vegetative, kernel-filling and postharvest stages. Two sustained deficit irrigation (SDI) treatments (SDI75 and SDI65 with water reductions of 25 and 35%, respectively) and a control treatment (FI) consisting of fully irrigated trees were applied. The response of curves AN-Ci was analyzed to assess the maximum carboxylation rate (Vcmax), maximum rate of electron transport (Jmax), TPU and mesophyll conductance to CO2. In addition, leaf water potential and yield were measured. Our experimental findings showed any significant differences in the variables analyzed among cultivars and irrigation treatments. However, consistent differences arose when the results were compared among the phenological stages. During the kernel-filling and the postharvest stages, a progressive limitation by TPU was measured, suggesting that the demand for photoassimilates by the plant was reduced. This result was supported by the correlation found between TPU and fruit growth rate. As a consequence, a downregulation in Jmax and Vcmax was also measured. This study confirms that the kernel-filling stage might be a good time to apply a reduction in the irrigation and suggests a method to detect the best moments to apply a regulated deficit irrigation in almond trees.

Histopathology of the Shoot Apex of Sugarcane Colonized by Leifsonia xyli subsp. xyli.

Colonization of the xylem of sugarcane by Leifsonia xyli subsp. xyli results in the occlusion of the vessels by a gum-like compound and compromises the elongation of the stalk leading to stunted plants. However, no study has been performed in the apical tissue where the elongation of the stalks initiates at the intercalary meristem (IM). Microscopic and histochemical analyses were performed in plants with lower and higher bacterial titers and revealed that in both cases L. xyli subsp. xyli is present in this tissue and colonizes the forming xylem vessels in a similar way as observed in developed internodes. In both cases, it was observed adhering to the secondary walls, but only in plants with higher titers were a mild degradation of the walls and a granular material filling the vessels observed. The mixed composition of lipids, proteins, and pectin indicates that the filling is not a bacterial extracellular polymeric substance. Plants with higher bacterial populations also presented lower starch content in the ground parenchyma at the node elements, possibly resulting from the reported downregulation of photosynthesis and increased accumulation of phenolics. Their second and third IMs presented fewer cells and reduced expression of genes related to the cell cycle and to the synthesis of ABA in the apical tissue. These results indicate that increased L. xyli subsp. xyli colonization affects the development of the IM, which ultimately would reduce the length of the internodes, resulting in the main symptom of the disease.

Cap-snatching as a possible contributor to photosynthesis shut-off.

Cap-snatching is a mechanism applied by segmented, negative strand (-) RNA viruses (NSVs) to initiate genome transcription. So far, the cap donor source of cytoplasmic-replicating NSVs has remained elusive. Recently, studies pointed to processing body (P body, PB) as the potential source for providing capped RNAs but conclusive evidence is still lacking. To attempt identifying these sources, here the 5' non-viral leader sequences of Tomato spotted wilt virus (TSWV) N mRNAs were analysed by high-throughput sequencing (HTS) from plants subjected to normal and heat-stress conditions, and subsequently mapped on host donor transcripts. The majority of non-viral heterogenous, host-derived leader sequences ranged in size between ~10-20 nt and contained A or AG residues at the cleavage site and the presence of certain sequence motifs. Mapping the capped-leader sequences to the 5' UTR region of genes encoded by the Nicotiana tabacum genome, identified 348 donor genes and which were specifically enriched in cellular photosynthesis pathway. Nineteen of those were clearly expressed differentially at normal condition versus heat-stress conditions. Although the results did not point towards snatching of capped-RNA leader sequences from certain cytoplasmic RNA granules in particular, they indicated photosynthesis downregulation (and development of disease symptoms) partially result from cap-snatching.

Genome-Wide Transcriptomic Analysis of the Effects of Infection with the Hemibiotrophic Fungus Colletotrichum lindemuthianum on Common Bean.

Bean anthracnose caused by the hemibiotrophic fungus Colletotrichum lindemuthianum is one of the most important diseases of common bean (Phaseolus vulgaris) in the world. In the present study, the whole transcriptome of common bean infected with C. lindemuthianum during compatible and incompatible interactions was characterized at 48 and 72 hpi, corresponding to the biotrophy phase of the infection cycle. Our results highlight the prominent role of pathogenesis-related (PR) genes from the PR10/Bet vI family as well as a complex interplay of different plant hormone pathways including Ethylene, Salicylic acid (SA) and Jasmonic acid pathways. Gene Ontology enrichment analysis reveals that infected common bean seedlings responded by down-regulation of photosynthesis, ubiquitination-mediated proteolysis and cell wall modifications. In infected common bean, SA biosynthesis seems to be based on the PAL pathway instead of the ICS pathway, contrarily to what is described in Arabidopsis. Interestingly, ~30 NLR were up-regulated in both contexts. Overall, our results suggest that the difference between the compatible and incompatible reaction is more a question of timing and strength, than a massive difference in differentially expressed genes between these two contexts. Finally, we used RT-qPCR to validate the expression patterns of several genes, and the results showed an excellent agreement with deep sequencing.

Silicon Enhances Plant Vegetative Growth and Soil Water Retention of Soybean (Glycine max) Plants under Water-Limiting Conditions.

Silicon has been implicated as a factor affecting the degree of resistance to abiotic stresses in several plant species. However, the role of silicon in soybean (Glycine max) under water-limiting conditions is not yet fully understood. This study was conducted to evaluate the effects of silicon application on the vegetative growth of two soybean cultivars (Asgrow 5332 and Progeny 5333) grown under water-limiting conditions. Silicon was provided by adding silicate to the soil. Water-limiting treatments were imposed on plants at two vegetative growth stages for 20 days by irrigating with a reduced amount of water (66% or 33% of the required water). Silicate application enhanced plant height, leaf area, and total dry weight of soybean plants. Significant increases in root volumes were observed in both the silicate-treated cultivars compared to the control plants under water-limiting conditions (33% irrigation). Net photosynthesis and stomatal conductance were decreased, but the quantum efficiency of photosystem II (Fv’/Fm’) did not change under the same irrigation condition, which indicates photosynthesis downregulation through stomatal limitation. Silicate-treated plants in both cultivars had higher water use efficiency as compared to control plants under water-limiting conditions (irrigated with 66% or 33% of required water). Under water-limiting conditions, the soil moisture content was significantly higher in pots containing silicate than in those without added silicate, suggesting that silicon application improves water holding capacity. Taken together, the results from this study indicate that silicon application can improve the vegetative growth of soybeans under low water conditions by increasing the water use efficiency of plants and enhancing the soil’s ability to retain moisture.

Genome-Wide Investigation of the PtrCHLP Family Reveals That PtrCHLP3 Actively Mediates Poplar Growth and Development by Regulating Photosynthesis

Chlorophyll (Chl) plays a crucial role in plant photosynthesis. The geranylgeraniol reductase gene (CHLP) participates in the terminal hydrogenation of chlorophyll biosynthesis. Although there are many studies related to the genome-wide analysis of Populus trichocarpa, little research has been conducted on CHLP family genes, especially those concerning growth and photosynthesis. In this study, three CHLP genes were identified in Populus. The evolutionary tree indicated that the CHLP family genes were divided into six groups. Moreover, one pair of genes was derived from segmental duplications in Populus. Many elements related to growth were detected by cis-acting element analysis of the promoters of diverse PtrCHLPs. Furthermore, PtrCHLPs exhibit different tissue expression patterns. In addition, PtrCHLP3 is preferentially expressed in the leaves and plays an important role in regulating chlorophyll biosynthesis. Silencing of PtrCHLP3 in poplar resulted in a decrease in chlorophyll synthesis in plants, thus blocking electron transport during photosynthesis. Furthermore, inhibition of PtrCHLP3 expression in poplar can inhibit plant growth through the downregulation of photosynthesis. Ultimately, PtrCHLP3 formed a co-expression network with photosynthesis and chlorophyll biosynthesis-related genes, which synergistically affected the growth and photosynthesis of poplars. Thus, this study provides genetic resources for the improved breeding of fast-growing tree traits.

Diurnal and Seasonal Dynamics of Solar‐Induced Chlorophyll Fluorescence, Vegetation Indices, and Gross Primary Productivity in the Boreal Forest

AbstractRemote sensing of solar‐induced chlorophyll fluorescence (SIF) provides a powerful proxy for gross primary productivity (GPP). It is particularly promising in boreal ecosystems where seasonal downregulation of photosynthesis occurs without significant changes in canopy structure or chlorophyll content. The use of SIF as a proxy for GPP is complicated by inherent non‐linearities due to both physical (illumination effects) and ecophysiological (light use efficiencies) controls at fine spatial (tower/leaf) and temporal (half‐hourly) scales. To study the SIF‐GPP relationship, we investigated the diurnal and seasonal dynamics of continuous tower‐based measurements of SIF, GPP, and common vegetation indices at the Southern Old Black Spruce Site (SOBS) in Saskatchewan, CA over the course of two years. We find that SIF outperforms other vegetation indices as a proxy for GPP at all temporal scales but shows a non‐linear relationship with GPP at a half‐hourly resolution. At small temporal scales, SIF and GPP are predominantly driven by light and non‐linearity between SIF and GPP is due to the light saturation of GPP. Averaged over daily and monthly scales, the relationship between SIF and GPP is linear due to a reduction in the observed PAR range. Seasonal changes in the light responses of SIF and GPP are driven by changes in light use efficiency which co‐vary with changes in temperature, while illumination and canopy structure partially linearize the SIF‐GPP relationship. Additionally, we find that the SIF‐GPP relationship has a seasonal dependency. Our results help clarify the utility of SIF for estimating carbon assimilation in boreal forests.

Biofertilization with photosynthetic bacteria as a new strategy for mitigating photosynthetic acclimation to elevated CO2 on cherry tomato

The stimulated effects of elevated CO2 concentration decline over time in many C3 plants. However, the phenomenon is a rare occurrence on plants hosting some symbiosis bacteria. The photosynthesis acclimation associated with an extended period of high CO2 concentration was therefore studied in cherry tomato that treated with a kind of photosynthetic bacteria. Bio-fertilization with photosynthetic bacteria, which can fix nitrogen for host crops and contains a variety of growth promoting factors, can play an important role in maintaining photosynthetic activity. In this research, we conducted a CO2 elevation experiment combined different water regimes and inoculation with photosynthetic bacterium, to evaluate the improvement of photosynthesis acclimation by using photosynthesis bacteria, and to explore associated mechanisms for artificial chamber grown cherry tomatoes. As a result of these experiments, we found that elevated CO2 had a positive effect on cherry tomato photosynthetic activity and yield of all treatment. After 55 days of CO2 injection, photosynthetic acclimation was found in the plant of uninoculated photosynthetic bacteria, which were most pronounced in high water treatments. Reduced irrigation amounts decreased the magnitude of photosynthetic acclimation, but relative fruit yield gradually decreased after achieving a maximum. In contrast, the photosynthetic bacteria inoculated plants just showed a slightly down-regulation of photosynthesis under durative high CO2 condition. Because inoculation of photosynthetic bacteria increased the photosynthetic pigment content, mesophyll conductance, maximum carboxylation rate and maximum electron transport rate. Furthermore, inoculation with the photosynthetic bacteria diminished the yield penalties caused by the reduced irrigation treatment through promoting nitrogen content and relieve starch accumulation in tomato leaves compared to those of the uninoculated control. And enhanced yield of inoculated plants grown under durative high CO2 concentration were observed. The results suggest that photosynthesis bacteria application could be a potential means to further improve the CO2 stimulation effect on cherry tomato production.

Response of Poplar Leaf Transcriptome to Changed Management and Environmental Conditions in Pure and Mixed with Black Locust Stands

Mixed cropping in short rotation coppice can be an alternative to monocultures. To design optimized mixtures, field trials are needed. Poplar, as an economically important and fast-growing species, and black locust, as a nitrogen-fixing species, are promising candidates for such studies. RNA sequencing (RNA-seq) was used to monitor effects of mixed and pure cultivations on the gene expression of poplar along with growth measurements during 2017 and 2018. Both biomass production and leaf transcriptomes revealed a strong competition pressure of black locust and the abiotic environment on poplar trees. Gene expression differed between the two study sites and pure and mixed stands. Shading effects from black locust caused the downregulation of photosynthesis and upregulation of shade avoidance genes in mixed stands in 2017. As a result of higher light availability after cutting black locust, plant organ development genes were upregulated in mixed stands in 2018. Drought conditions during the summer of 2018 and competition for water between the two species caused the upregulation of drought stress response genes in mixed stands and at the unfavorable growing site. Further investigations are required to discover the mechanisms of interspecific competition and to develop stand designs, which could increase the success and productivity of mixed plantations.

A reduction in metabolism explains the tradeoffs associated with the long-term adaptation of phytoplankton to high CO2 concentrations.

Phytoplankton are responsible for nearly half of global primary productivity and play crucial roles in the Earth's biogeochemical cycles. However, the long-term adaptive responses of phytoplankton to rising CO2 remains unknown. Here we examine the physiological and proteomics responses of a marine diatom, Phaeodactylum tricornutum, following long-term (c. 900 generations) selection to high CO2 conditions. Our results show that this diatom responds to long-term high CO2 selection by downregulating proteins involved in energy production (Calvin cycle, tricarboxylic acid cycle, glycolysis, oxidative pentose phosphate pathway), with a subsequent decrease in photosynthesis and respiration. Nearly similar extents of downregulation of photosynthesis and respiration allow the high CO2 -adapted populations to allocate the same fraction of carbon to growth, thereby maintaining their fitness during the long-term high CO2 selection. These results indicate an important role of metabolism reduction under high CO2 and shed new light on the adaptive mechanisms of phytoplankton in response to climate change.
