This study collected SSR information from transcriptome samples of Pinellia ternata (P.T) from different regions in order to provide some reference for germplasm resources and genetic breeding of P.T. samples from six regions were selected as research objects. The frequency of SSR site, types of motifs, composition of motifs, and average distance was analyzed by Excel software. The results showed that there were 32,061–67,917 unigene sequences and 11,109–22,540 SSR site in the six P.T species, with a frequency of 32.7%–35.8% and an average distance of 2.65 kb–3.17 kb. The six SSR repeat types for P.T were mainly double base, triple base and Mononucleotide repeats. The number composition of dominant repeat motifs in the six P.T species from Mononucleotide to Tetranucleotide was basically the same, which were A/T, AG/CT, CCG/CGG and AGCT/AGCT, respectively. Most SSR sites were less than or equal to 30 bp in length, and the number of these SSRs accounted for 81.2%–82.3% of the total. It can be seen from the above results that, the transcriptome of P.T in different regions was rich in SSR types and high in polymorphisms, which can provide a basis for germplasm difference identification, genetic breeding and other research of P.T.
