Study on the effects of DMF and FAM regimens in the treatment of advanced gastric cancer

Objective To study the effect and toxicity of DMF regimen and FAM regimen in the treatment of advanced gastric cancer.Methods From January 1995 to December 1998,56 patients were treated with DMF regimen and FAM regimen.Results In 28 patients treated with DMF regimen,the total response rate was (CR+PR)53 6%. In 28 patients treated with FAM regimen,the total response rate was (CR+PR)57 2%.The response rates in both groups were handled statistically with x 2( P 0 05).There were no apparent differences.The toxicity in DMF and FAM group all have maucea,vomithing and myelosupperession.Gastrointestinal toxicity in DMF group was more severe than that in FAM group and the myelosupperession was mild.There were apparent baldness in FAM group,and 2 patients hnd cardiac events.The nephrotoxicity in DMF group can be prevented by hydration.Conclusion The effects in DMF group were similar to the FAM group,and the toxicity was tolerable.There were no baldness,cardiac events and the price was low.So we suggest that the DMF regimen is worthy to be recommended to patients with advanced gastric cancer.

Different change patterns of the isozymes of cytochrome P450 and glutathione S-transferases in chemically induced liver damage in rat.

In this experiment, we studied the different changes in activities and protein levels of each subform of hepatic cytochrome P450 and glutathione S-transferase (GST), in chemical-induced liver injury in rats. Rats were administered 1,1-dichloroethylene (DCE), allyl alcohol (AA), bromobenzene (BB) and N,N-dimethylformamide (DMF) p.o. once every two days for 7 times, and decapitated 18 hr after the last administration. DCE and AA showed stronger hepatic toxicity than BB and DMF, as serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were higher in DCE and AA treated rats than in BB and DMF groups. Anti-cytochrome P450 inhibitable activity of toluene metabolism and/or immunoblot analysis showed that CYP2E1 and CYP2B1/2 were induced by BB and DMF, but not by the other two chemicals; CYP2C11 was greatly decreased by all of the four toxicants; and CYP1A1/2 was slightly reduced by the four treatments. These changes were reflected in testosterone metabolism. Formation of 6 beta- and 7 alpha-hydroxytestosterone from testosterone was enhanced only in DMF-treated rats, whereas that of 2 alpha- and 16 alpha-hydroxytestosterone was reduced by all of the four chemicals. Serum GST activity was increased only in BB and DMF treated rats, but liver cytosolic GST activity was enhanced by all of the four hepatotoxicants, with higher values in BB and DMF groups than in DCE and AA groups. Immunoblot analysis demonstrated that GST Yp was induced by BB and DMF treatments, and Ya and Yc were increased only by BB. GST Yk and Yb1 were not affected by the treatments. The different change patterns of enzymes by a specific toxin and the similar modifying effect on a specific enzyme by different toxins were discussed in relation to the liver damage and to the heterogeneous distribution of enzymes in liver.

Bis[μ-N,N'-bis(salicylidene)-1,3-propanediaminato]bis(dimethylformamide)di(μ-nitrito)copper(II)dinickel(II)–Dimethylformamide (1/2)

The unit cell of the title compound, [CuNi2(NO2)2(C17H16N2O2)2(C3H7NO)2].2C3H7NO, contains two molecules of the linear heterotrinuclear complex and four solvent molecules. The central CuII ion and the terminal NiII ions have distorted octahedral coordination spheres. The Cu–Ni pairs are each triply bridged, by the O atoms of an N,N'-bis(salicylidene)-1,3-propanediaminato (salpd2−) ligand and by an O and an N atom of a nitrite group. The central CuII ion, located on an inversion centre, has a total of six O atoms in its coordination sphere: two from each salpd2− ligand and one from each bridging nitrite group. The NiII ions are related by the inversion centre and each is coordinated by the two O and two N atoms of a salpd2− ligand and one N atom from a nitrite and an O atom from a dimethylformamide (dmf) ligand. The dmf and nitrite groups are mutually trans about the NiII ion. The Cu—Ni distance is 2.9967 (4) Å.

The use of ethylene glycol for cryopreservation of sheep embryos

This study was aimed to compare different cryoprotectants for vitrification of sheep embryos produced in vivo. Blastocysts were obtained from superovulated Santa Ines ewes and randomized into three groups: conventional freezing using ethylene glycol (EG) (control group), vitrification with EG and dimethyl sulfoxide (DMSO vitrification), or vitrification with EG and dimethylformamide (DMF vitrification). All groups were analyzed for embryonic viability (propidium iodide staining), re-expansion rate after thawing (at morphological and ultrastructural levels) and pregnancy rate after embryo transfer (ET). Embryos of DMSO vitrification group showed lower cell viability (44.44%), compared to DMF group and control embryos (77.77% and 100%, respectively). The ultrastructural study showed similar cryopreservation damage among control and DMF embryos, and these were less damaged than DMSO vitrified embryos. Embryos vitrified with DMF had higher rates of re-expansion in vitro (53.33%) than DMSO (26.66%), and control (33.33%). After ET, similar pregnancy rates were obtained from all groups (DMF: 45%, DMSO: 30%, control: 40%). Collectively, DMF vitrification is more efficient than DMSO vitrification and is indistinguishable from conventional freezing of sheep embryos.

Synthesis, properties and structure of hexaaquo-tris( N,N-dimethylformamide)-lanthanide trifluoromethanesulfonates

The compounds of general formula [Ln(DMF) 3- (H 2O) 6](CF 3SO 3) 3 (Ln = LaEu, Tb, Dy) were synthesized and characterized by microanalysis, conductance measurements, IR absorption (Nd 3+) and emission (Eu 3+) spectra. The crystal structure of the neodymium compound was determined by X-ray diffraction techniques. The compound crystallizes in the triclinic system, space group P1, a = 8.589(4), b = 11.222(2), c = 12.271(2) Å, α = 56.83(2), β = 62.13(2), γ = 75.14(2)°, V = 875.2 Å 3, M = 918.4, Z = 1, D c = 1.73 g cm −3, λ(Mo Kα) = 0.71073 Å, μ = 1.65 mm −1, F(000) = 456, R = 0.056, R w = 0.057, for 2979 independent reflections with I > 3σ( I). Nd 3+ is coordinated to the oxygen atoms of six independent water molecules at a mean distance NdO = 2.52(1) Å, and to the oxygen atoms of three independent DMF groups at a mean distance NdO = 2.40(2) Å. The coordination polyhedron is a tricapped trigonal prism of point symmetry C 3 v .

Distribution of Substituted Anilines between N, N-Dimethylformamide and Macroreticular Sulfonated Ion-Exchange Resin

Distribution equilibria of three substituted anilines between N, N-dimethylformamide(DMF) and macroreticular sulfonic acid type ion-exchange resin, Amberlyst 15, has been investigated over a wide range of concentration. The reaction of the bases with the hydrogen resinate in DMF was regarded as acid-base equilibria which obey a classical mass law expression. The equilibrium constants(K) increased with an increase of acid dissociation constants of the bases in water and it was represented by equation log K=-0.85 + 0.752 pKa. On saturated sorption of the bases toward the resin in DMF, it was found that the degree of saturation of beses are related to pKa values and corresponds to 71% of the amounts of hydrogen ion in the resin for toluidine. This is considered to be the interaction between DMF and sulfonate group of the resin.Application of these results to chromatographic separation on ion exchanger in nonaqueous media was examined. The separation of o- and p-chloroanilines and of o- and m-chloroanilines were successfully achieved by using DMF or n-butylamine-DMF mixture as eluants.

Dental caries in relation to PTC taste sensitivity, secretor status, and salivary thiocyanate level

A previous study showed that tasters of phenylthiocarbamide (PTC) had significantly reduced risk of dental caries of the primary teeth. The present study is a further attempt to elucidate the association with 137 white children of ages from 4 to 17 yr divided into def and DMF groups. In addition, the secretor status of the ABO substances and the content of the whole saliva thiocyanate were studied relative to caries experience and PTC sensitivity. The net effects of these factors were estimated by the least squares method. There was an appreciable decrease of caries experience of the primary teeth (def) in tasters being not inconsistent with the previous finding There was no apparent relationship detected between the caries experience of the permanent teeth (DMF) and the PTC response. The secretor status was found to be unrelated to the caries scores. The level of the total salivary thiocyanate appears to be independent of PTC sensitivity and caries experience.
