Disruption of bacterial biofilms by a green synthesized artemisinin nano-copper nanomaterial.

Bacterial biofilms are associated with antibiotic resistance and account for ∼80% of all bacterial infections. In this study, we explored novel nanomaterials for combating bacteria and their biofilms. Artemisinin nano-copper (ANC) was synthesized using a green synthesis strategy, and its shape, size, structure, elemental composition, chemical valence, zeta potential, and conductivity were characterized using transmission electron microscopy, X-ray diffractometer, X-ray photoelectron spectroscopy, zeta potential, and dynamic light scattering. The results showed that ANC was successfully synthesized utilizing a liquid phase chemical reduction method using chitosan as a modified protectant and l-ascorbic acid as a green reducing agent. The stability of ANC was evaluated using dynamic light scattering. The results showed that the particle size of ANC at different concentrations was comparable to that of the original solution after 7 days of storage, and there was no significant change in the polydispersity index (P>0.05). The antibacterial effects of ANC on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) were determined by disc diffusion and broth dilution methods. The results demonstrated that ANC inhibited and killed E. coli and S. aureus. The effect of ANC on bacterial biofilms was investigated using crystal violet staining, scanning electron microscopy, laser confocal microscopy, and quantitative polymerase chain reaction. The results showed that ANC treatment was able to destroy bacterial biofilms and downregulate biofilm- and virulence-related genes in E. coli (HlyA, gyrA, and F17) and S. aureus (cna, PVL, ClfA, and femB). Green-synthesized ANC possesses excellent antibiofilm properties and is expected to exhibit antibacterial and antibiofilm properties.

Kinetics analysis of PAHs degradation using SiO2–ZnO nanoparticles and evaluating their antibacterial and antibiofilm efficacy

The adsorption of Polycyclic aromatic hydrocarbons (PAHs) using nanoparticles is gaining significant attention due to the rapid removal or treatment rates. In this study, Silicon Dioxide-Zinc Oxide nanoparticles (SiO2–ZnO NPs) were synthesized to adsorb pyrene. Physicochemical characterization of SiO2–ZnO NPs showed plasmon resonance at 323 nm, agglomeration, irregular dispersion, and diameters of 90–100 nm. FT-IR analysis identified major functional groups on SiO2–ZnO NPs, including alkyne, amine, and isothiocyanate. SiO2–ZnO NPs demonstrated significant pyrene adsorption at pH 5, with 10 μg/mL of SiO2–ZnO NPs and 2 μg/mL of PAHs, performing better under UV irradiation. Two isotherm models, adsorption isotherm and kinetics adsorption, were used to analyze the PAHs adsorption by SiO2–ZnO NPs. Additionally, SiO2–ZnO NPs were tested for antibacterial and antibiofilm activities against both Gram-negative and Gram-positive bacteria. At a concentration of 150 μg/mL, SiO2–ZnO NPs produced inhibition zones of 21.57 mm, 20.30 mm, 19.30 mm, and 11.30 mm against Staphylococcus aureus, Escherichia coli, Bacillus subtilis, and Klebsiella pneumoniae, respectively. They also inhibited and disrupted biofilms of Micrococcus luteus and Acinetobacter baumannii. Furthermore, SiO2–ZnO NPs exhibited photocatalytic degradation of lead, achieving 68.24% degradation within 5 h of treatment. Therefore, SiO2–ZnO NPs are efficient candidates for multiple applications, including pyrene adsorption, bacterial biofilm disruption, and lead degradation under sunlight.

Physical parametric study of bacterial biofilm disruption and removal by jet impingement: A CFD investigation

Bacterial biofilm formation is one of the most critical challenges in industrial and health systems. It is the leading cause of nearly 80 % of clinical infections. Biofilms are usually evaluated from the physiological point of view as microbial cells enclosed in an extracellular polymer and represent a complex, dynamic bacterial community. Cohesive mechanical forces hold the bacteria within the biofilm together. These forces contribute to the biofilm's mechanical and structural stability, resulting in its viscoelastic properties. Studying the biofilm's mechanical properties as a viscoelastic matter can provide a better understanding of the biofilm's characteristics and the survival strategies of bacteria in the biofilm state of life. In the present study, a 2D-axisymmetric RANS-CFD simulation model using the Volume Of Fluid (VOF) method was developed to track changes in biofilm surface and ripple formation across six biofilm thicknesses ranging from 15 μm to 115 μm, two substrate geometries including flat and curved, and two substrate roughnesses of 0.4 μm and 0.9 μm height. A modified Herschel-Bulkley model was employed to capture the biofilm's non-Newtonian shear-thinning behavior under high-velocity gas jet impingement. The results revealed the importance of biofilm thickness, substrate geometries, and properties like roughness in biofilm's mechanical behavior. Higher thickness of biofilm resulted in smaller jet-impingement region and more significant ripples formation. Biofilm on curved substrates decreased in thickness more rapidly than those on flat substrates. The substrate's geometry impacted biofilm folding patterns, removal, and mechanical behavior. Selected substrate roughnesses did not create any resistance against the mechanical removal of biofilms. Here, we successfully captured biofilms' non-Newtonian and shear-thinning behavior with numerical simulations consistent with previously reported experimental results. This could benefit industrial and health systems by tackling biofilm-related challenges and developing more accurate and detailed models of biofilms to optimize tools for drug testing or screening and enhance the models for in-vitro and in-vivo analysis.

Biological Activity of Acinetobacter calcoaceticus IMV B-7241 Surfactants Synthesized in the Presence of Competitive Bacteria Bacillus subtilis BT-2

Currently, the effectiveness of technologies for microbial surfactants, which are characterized by a complex of practically valuable physicochemical and biological properties is lower than that of synthetic analogues. To reduce the cost of these products of microbial synthesis, industrial waste is used as substrates for their biosynthesis. In previous studies, it has been established that surfactants synthesized by Acinetobacter calcoaceticus IMV B-7241 on crude glycerol have lower antimicrobial activity compared to that obtained on purified glycerol. The main approaches to the regulation of the biological activity of microbial surfactants are their post-fermentation chemical modification, as well as the improvement of producer strains by methods of metabolic and genetic engineering. In recent years, numerous studies have appeared on the co-cultivation of producers of antimicrobial compounds with competitive microorganisms (biological inductors), in response to the presence of which the antimicrobial activity of the final product increases. Aim. To study the effect of live and inactivated cells of Bacillus subtilis BT-2, as well as the corresponding supernatant, on the antimicrobial and anti-adhesive activity and the ability to destroy biofilms of A. calcoaceticus IМV B-7241 surfactants synthesized in a medium with glycerol of different degrees of purification. Methods. The A. calcoaceticus IMV B-7241 strain was grown in a liquid mineral medium with purified and crude glycerol, into which live and inactivated B. subtilis BT-2 cells as well as the supernatant after growing the B. subtilis BT-2 strain (2.5—10%, v/v) were added. Surfactants were extracted from the supernatant of the culture liquid with Folch’s mixture. Anti-adhesive activity and the degree of destruction of biofilms were determined by the spectrophotometric method, and antimicrobial activity — by the indicator of the minimum inhibitory concentration. The activity of enzymes of surface-active aminolipids biosynthesis (NADP+-dependent glutamate dehydrogenase) and glycolipids (phosphoenolpyruvate (PEP)-carboxylase, PEP-synthetase, PEP-carboxykinas, trehalose-phosphate synthase) was analyzed in cell-free extracts obtained after сells sonication. Results. It was established that the introduction of inactivated B. subtilis BT-2 cells and supernatant into the medium with both substrates did not affect the indicators of the surfactant synthesis, while in the presence of live cells of the B. subtilis BT-2 strain in the medium with purified glycerol, a decrease in the concentration of the final product by 1.5 times, and in the culture medium with crude glycerol — an increase by1.4 times were observed compared to the indicators with no inductor. The study of the antimicrobial activity of surfactants showed that the most effective of the used inductors (live, inactivated cells, supernatant) were live cells of B. subtilis BT-2. The introduction of B. subtilis BT-2 strain live cells into the culture medium with both substrates was accompanied by the formation of surfactants, the minimum inhibitory concentrations of which in relation to bacterial (Bacillus subtilis BT-2, Staphylococcus aureus BMS-1, Proteus vulgaris PA-12, Enterobacter cloacae С-8) and yeast (Candida albicans D-6, Candida tropicalis PE-2) test-cultures were 3—23 times lower than established for those synthesized on the medium with no inductor. Anti-adhesive activity of surfactants obtained on purified and crude glycerol in the presence of all types of inductors was higher compared to those synthesized in the culture medium without inductors (cells adhesion of bacterial and yeast test-cultures on polyvinyl chloride was 13—70 and 33—96%, respectively). Introduction of live and inactivated B. subtilis BT-2 cells or the supernatant into A. calcoaceticus IMV B-7241 cultivation medium was accompanied by the synthesis of surfactants, in the presence of which the disruption of bacterial biofilms was on average 10-20% higher compared to using surfactants synthesized without an inductor. In the presence of B. subtilis BT-2 in the medium, in the cells of the A. calcoaceticus IMV B-7241 strain, the activity of NADP+-dependent glutamate dehydrogenase (a key enzyme of aminolipids biosynthesis) increased by 1.5—2 times, while the activity of biosynthesis of glycolipids enzymes remained practically at the same level as without an inductor. Such data indicate that the higher biological activity of surfactants obtained by A. calcoaceticus IMV B-7241 in the presence of biological inductors might be due to an increase in the content of aminolipids in their composition. Conclusions. This research has established the possibility of regulating the antimicrobial and anti-adhesive activity as well as the ability to disrupt biofilms of A. calcoaceticus IМV B-7241 surfactants by introducing competitive bacteria B. subtilis BT-2 into the culture medium. It is important that under such cultivation conditions, the antimicrobial activity of surfactants synthesized on toxic crude glycerol significantly increases.

Disruption of Staphylococcus aureus Biofilms with Purified Moringa oleifera Leaf Extract Protein.

A major cause of economic losses in the medical implant sector has been bacterial biofilms due to their ability to persist on various surfaces and their tolerance against endogenous defences, antibiotics, or other anti-microbial agents. The quest for potential sources causing inhibition or disruption of bacterial biofilms has been taken up to alleviate the loss. Plantderived extracts such as essential oils, bioactive compounds and other solvent extracts are regularly being used instead of antibiotics and other synthetic compounds as they are safer, economical, and in many instances, have an elaborate history of traditional medicinal usage. As a plant that has been traditionally used over the centuries, the Moringa oleifera Lam., or more commonly the drumstick tree, is being tapped for myriad pharmaceutical applications. The protein-rich leaf of this tree has not only proved to be of great nutritional value but also divulged numerous potential therapeutic applications. While reports of proteinaceous components are rare, here we report the efficacy of the aqueous extract of the leaf of M. oleifera and a 62 kDa protein component in the disruption of staphylococcal biofilms, which are largely implicated in nosocomial infections. The application of the M. oleifera leaf extract protein had a marked effect on the biofilm growth or formation by Staphylococcus aureus. While the crude extract itself showed considerable disruption of biofilm formation, the application of the purified protein obtained after a two-step process led to a significant increase in the anti-biofilm activity.

Triple-Function Au-Ag-Stuffed Nanopancakes for SERS Detection, Discrimination, and Inactivation of Multiple Bacteria.

New strategies combining sensitive pathogenic bacterial detection and high antimicrobial efficacy are urgently desirable. Here, we report smart triple-functional Au-Ag-stuffed nanopancakes (AAS-NPs) exhibiting (1) controllably oxidative Ag-etching thickness for simultaneously obtaining the best surface-enhanced Raman scattering (SERS) enhancement and high Ag-loading antibacterial drug delivery, (2) expressive Ag+-accelerated releasing capability under neutral phosphate-buffered saline (PBS) (pH ∼ 7.4) stimulus and robust antibacterial effectiveness involving sustainable Ag+ release, and (3) three-in-one features combining specific discrimination, sensitive detection, and inactivation of different pathogenic bacteria. Originally, AAS-NPs were synthesized by particle growth of the selective Ag-etched Au@Ag nanoparticles with K3[Fe(CN)6], followed by the formation of an unstable Prussian blue analogue for specifically binding with bacteria through the cyano group. Using specific bacterial "fingerprints" resulting from the introduction of dual-function 4-mercaptophenylboronic acid (4-MPBA, serving as both the SERS tag and internal standard) and a SERS sandwich nanostructure that was made of bacteria/SERS tags/AAS-NPs, three bacteria (E. coli, S. aureus, and P. aeruginosa) were highly sensitively discriminated and detected, with a limit of detection of 7 CFU mL-1. Meanwhile, AAS-NPs killed 99% of 1 × 105 CFU mL-1 bacteria within 60 min under PBS (pH ∼ 7.4) pretreatment. Antibacterial activities of PBS-stimulated AAS-NPs against S. aureus, E. coli, and P. aeruginosa were extraordinarily increased by 64-fold, 72-fold, and 72-fold versus PBS-untreated AAS-NPs, respectively. The multiple functions of PBS-stimulated AAS-NPs were validated by bacterial sensing, inactivation in human blood samples, and bacterial biofilm disruption. Our work exhibits an effective strategy for simultaneous bacterial sensing and inactivation.

Antibiofilm and Antifungal Activities of Laurelia sempervirens (Chilean laurel) Essential Oil

Background: Laurelia sempervirens, better known as Chilean laurel, is endemic to Chile. For many years, the leaves and branches of this plant have been popularly used as medicinal agents. However, its antifungal properties and antibiofilm activity against bacterial strains have not been studied. Objectives: To determine antibiofilm and antimicrobial properties of Chilean laurel essential oil on human pathogenic strains. Methods: The antifungal and antibacterial activities of Chilean laurel essential oil were evaluated through the agar diffusion method, and its minimum inhibitory concentration was determined using the standard microdilution method. Antibiofilm activity was examined based on the formation and disruption of bacterial biofilms and evidenced by the crystal violet technique. Results: The results showed important antimicrobial activity against human pathogenic yeast strains, and the minimum inhibitory concentration of Chilean laurel essential oil was 64 µg mL-1 against Candida albicans. The essential oil also showed an important inhibitory effect against the formation of biofilms produced by Staphylococcus aureus, since it inhibited the formation of biofilms by over 50% at the concentration of 64 µg mL-1. With increasing the essential oil’s concentration to 128 µg mL-1, its antibiofilm activity increased by 60%. Conclusions: These results approve the domestic use of Chilean laurel essential oil as an antimicrobial agent and provide knowledge about the antibiofilm and antifungal properties of L. sempervirens.

Combined Confocal Microscope and Brandaris 128 Ultra-High-Speed Camera

Controlling microbubble-mediated drug delivery requires the underlying biological and physical mechanisms to be unraveled. To image both microbubble oscillation upon ultrasound insonification and the resulting cellular response, we developed an optical imaging system that can achieve the necessary nanosecond temporal and nanometer spatial resolutions. We coupled the Brandaris 128 ultra-high-speed camera (up to 25 million frames per second) to a custom-built Nikon A1R+ confocal microscope. The unique capabilities of this combined system are demonstrated with three experiments showing microbubble oscillation leading to either endothelial drug delivery, bacterial biofilm disruption, or structural changes in the microbubble coating. In conclusion, using this state-of-the-art optical imaging system, microbubble-mediated drug delivery can be studied with high temporal resolution to resolve microbubble oscillation and high spatial resolution and detector sensitivity to discern cellular response. Combining these two imaging technologies will substantially advance our knowledge on microbubble behavior and its role in drug delivery.

NIR-Light-Active ZnO-Based Nanohybrids for Bacterial Biofilm Treatment.

Nanomaterials with antimicrobial properties triggered by external stimuli appear to be a promising and innovative substitute for the destruction of antibiotic-resistant superbugs as they can induce multiple disruptions in the cellular mechanism. This study demonstrates the use of squaraine (SQ) dye as the photosensitive material, activated in the near-infrared tissue-transparent therapeutic window. The dye has been covalently attached to the ZnO nanoparticle surface, forming ZnO-SQ nanohybrids. The formation of the nanohybrids is confirmed using Fourier transform infrared and other optical spectroscopic methods. The photoinduced interfacial electron transfer process (as confirmed using the time-resolved fluorescence technique) from the excited state of SQ to the conduction band of ZnO is responsible for the greater reactive oxygen species (ROS) generation ability of the nanohybrid. The production of photoactivated ROS (especially singlet oxygen species) by ZnO-SQ provides remarkable antimicrobial action against clinically significant Staphylococcus aureus. Detailed investigations suggest synergistic involvement of cell membrane disruption and nanoparticle internalization followed by photoinduced intracellular ROS generation, which result in an unprecedented 95% bacterial killing activity by the nanohybrid. Moreover, the efficacy of the nanohybrid for disruption of bacterial biofilms has been examined. The electron microscopic images suggest significant bacterial cell death following structural alteration and reduced adherence property of the biofilms. Nanodimension-driven greater internalization of ZnO-SQ followed by an improved dissolution of ZnO in an acidic environment of the biofilm as well as red-light-driven interfacial charge separation and ROS generation improves the efficacy of the material for biofilm destruction. An artificial medical implant mimicking titanium sheets coated with ZnO-SQ depicts light-triggered disruption in the adherence property of matured biofilms. The cytotoxicity and hemolysis assays show inherent biocompatibility of the photoactive nanohybrid. This study is notably promising for the treatment of life-threatening drug-resistant infections and eradication of biofilms formed within artificial implants.

In‐depth analysis of antibacterial mechanisms of laser generated shockwave treatment

Background and Objectives Laser generated shockwave (LGS) is a novel modality for minimally invasive disruption of bacterial biofilms. The objectives of this study are to determine the mechanisms behind LGS treatment and non-biofilm effects on bacterial disruption, including (1) comparing bacterial load with and without LGS in its planktonic form and (2) estimating bacterial cell permeability following LGS. Study Design/Materials and Methods For the first study, planktonic S. epidermidis were treated with gentamicin (0, 8, 16, 32, 64 μg/ml) with and without LGS (1064 nm Nd:YAG laser, 110.14 mJ/mm2 , pulse duration 9 ns, spot size 3 mm, n = 8/group), and absorbances at 600 nm compared. For the second study, four samples of planktonic S. epidermidis were treated with LGS (same settings). Propidium iodide (PI) uptake via flow cytometry as a measure of cell permeability was measured at 0, 10, and 20 minutes following LGS. RESULTS: In comparing corresponding gentamicin concentrations within both LGS-treated samples and controls at 0 hours, there were no differences in absorbance (P = 0.923 and P = 0.814, respectively). Flow cytometry found modest PI uptake (10.4 ± 2.5%) immediately following LGS treatment, with time-dependent increase and persistence of the signal at 20 minutes (R2 = 0.449, P = 0.048). CONCLUSION: Taken together, LGS does not appear to have direct bacteriocidal properties, but rather by allowing for biofilm disruption and bacterial cell membrane permeabilization, both of which likely increase topical antibiotic delivery to pathogenic organisms. Insight into the mechanisms of LGS will allow for improved clinical applications and facilitate safe and effective translation of this technology. Lasers Surg. Med. © 2018 Wiley Periodicals, Inc.

Evaluation of the kinetics and mechanism of action of anti-integration host factor-mediated disruption of bacterial biofilms.

The extracellular polymeric substance produced by many human pathogens during biofilm formation often contains extracellular DNA (eDNA). Strands of bacterial eDNA within the biofilm matrix can occur in a lattice-like network wherein a member of the DNABII family of DNA-binding proteins is positioned at the vertex of each crossed strand. To date, treatment of all biofilms tested with antibodies directed against one DNABII protein, Integration Host Factor (IHF), results in significant disruption. Here, using non-typeable Haemophilus influenzae as a model organism, we report that this effect was rapid, IHF-specific and mediated by binding of transiently dissociated IHF by anti-IHF even when physically separated from the biofilm by a nucleopore membrane. Further, biofilm disruption fostered killing of resident bacteria by previously ineffective antibiotics. We propose the mechanism of action to be the sequestration of IHF upon dissociation from the biofilm eDNA, forcing an equilibrium shift and ultimately, collapse of the biofilm. Further, antibodies against a peptide positioned at the DNA-binding tips of IHF were as effective as antibodies directed against the native protein. As incorporating eDNA and associated DNABII proteins is a common strategy for biofilms formed by multiple human pathogens, this novel therapeutic approach is likely to have broad utility.

Lucilia sericata Chymotrypsin Disrupts Protein Adhesin-Mediated Staphylococcal Biofilm Formation

Staphylococcus aureus and Staphylococcus epidermidis biofilms cause chronic infections due to their ability to form biofilms. The excretions/secretions of Lucilia sericata larvae (maggots) have effective activity for debridement and disruption of bacterial biofilms. In this paper, we demonstrate how chymotrypsin derived from maggot excretions/secretions disrupts protein-dependent bacterial biofilm formation mechanisms.

Microbubble acoustic surface cleaning

Surface cleaning is accomplished by fluid mechanical forcing, often assisted by chemical and acoustical activation. Examples include the removal of nanoparticles from IC semiconductor substrates and the disruption of bacterial biofilms in dentistry. The presence of bubbles is known to greatly enhance the cleaning efficiency as it promotes mixing of the chemicals and it yields higher stresses through acoustic streaming and jetting following asymmetric collapse of the bubbles. With smaller nanofabricated structures and more delicate surgical therapies, there is a growing demand for precision cleaning with minimum damage to the surrounding media. Here we explore the concept of micro-machined cylindrical pits acting as cavitation nuclei for a continuous source of microbubbles, thereby localizing the cavitation phenomena and suppressing its inherent chaotic nature. The micropit bubble was found to be stable against dissolution, and the resonance behavior of surface mode vibrations of the cap was modeled with the unsteady Stokes equation combined with a Fourier-Bessel expansion. Above a pressure threshold, destabilization of the micromeniscus results in bubble pinch-off which was studied using high-speed imaging down to nanoseconds timescales. It was also found that the acoustic coupling and merging of cavitation clouds from neighboring micropits increasingly promote acoustic surface cleaning.

Inquiry-based examination of chemical disruption of bacterial biofilms

Inquiry-based instruction in the sciences has been demonstrated as a successful educational strategy to use for both high school and college science classrooms. As participants in the NSF Graduate STEM Fellows in K-12 Education (GK-12) Program, we were tasked with creating novel inquiry-based activities for high school classrooms. As a way to introduce microbiology, molecular biology, ecology, and human health to students, we created a laboratory activity involving formation of biofilms composed of environmental bacteria from pond water and investigation into the resistance of these biofilms to antimicrobial agents. Two high schools participated in this study in different ways. Pike High School biology and advanced environmental science classrooms obtained pond water samples and grew biofilms from the bacteria in the pond water on plastic plates. They also observed killing of these biofilms by common household antimicrobial agents. As a senior capstone project, students at Arsenal Technical High School built on these research findings by isolating two different bacterial strains from the pond water and demonstrating the stimulatory effect of ethanol on biofilms formed by isolated bacterial strains. These activities were successful at introducing complex biological topics to high school students in a unique and exciting way. The students scored significantly higher on postactivity surveys compared with preactivity surveys that measured microbiology knowledge and experimental design knowledge. Furthermore, these projects seemed to elicit an excitement for science in the students who participated.

Heptosyltransferase I Lipid a Structure Activity Relationships (SAR)

The increasing incidence of antibiotic resistant bacterial infections has necessitated the search for novel targets and mechanisms for therapeutic intervention. Formation of a biofilm enables survival of bacteria in hostile environments, including in the presence of antimicrobials, thereby prompting research toward the disruption of bacterial biofilms. Lipopolysaccharide (LPS), a major constituent of the outer membrane of gram-negative bacteria, plays a key role in the formation and stability of biofilms. Therefore, disrupting the biosynthesis of the LPS is an attractive mechanism for development of anti-biofilm agents. One such LPS biosynthetic enzyme is heptosyltransferase I (Hep I), which is responsible for the transfer of the first L-glycero-D-manno-heptose to a 3-deoxy-α-D-manno-oct-2-ulopyranosonic acid (KDO) of the growing Lipid A portion of LPS. Previous attempts to co-crystallize Hep I with both substrates have been unsuccessful, hindering the ability to effectively visualize the binding interactions important for specificity. Substrate analogues for E. coli Hep I Lipid A are being used to identify which portions are necessary and sufficient for binding. Determination of the SAR of Hep I for these analogues may lead to the discovery of a Lipid A analogue that is able to be co-crystallized with Hep I. Locally Delivered Antimicrobials in the Management of Periodontitis: A Critical Review of the Evidence for their Use in Practice

Plaque bacteria are the primary initiators of periodontal disease in susceptible persons and therapy is largely based on mechanical bacterial biofilm disruption. Patients' response to periodontal treatment is unpredictable and periodontal stability is not always achieved. Locally delivered antimicrobials (LDAs) may be used as adjuncts to mechanical therapy in treatment of recalcitrant deep (> or = 5mm), active, non-responding sites, providing the patient's oral hygiene is adequate. Their use as a monotherapy cannot be justified. The literature reveals that LDAs are safe and that they achieve statistically significant, yet clinically modest, gains in clinical attachment and reductions in pocket depths. It has been suggested that LDAs may improve the clinical outcome in the treatment of recurrent and refractory cases of periodontitis when used as an adjunct to scaling and root surface instrumentation. This paper examines and discusses the evidence.

Macrolides et infection à Pseudomonas aeruginosa

Several unique effects of subinhibitory concentrations of C14 and C15 macrolides on Pseudomonas aeruginosa infection are described in vitro: P. aeruginosa virulence factors inhibition, bacterial biofilm disruption, P. glycoprotein expression upregulation, anti-inflammatory and immunoregulatory effects. Clinical trials in vivo are warranted to assess the potential usefulness of macrolides for treatment of chronic infections caused by P. aeruginosa.
