Interaction Study of an Amorphous Solid Dispersion of Cyclosporin A in Poly-Alpha-Cyclodextrin with Model Membranes by (1)H-, (2)H-, (31)P-NMR and Electron Spin Resonance.

The properties of an amorphous solid dispersion of cyclosporine A (ASD) prepared with the copolymer alpha cyclodextrin (POLYA) and cyclosporine A (CYSP) were investigated by 1H-NMR in solution and its membrane interactions were studied by 1H-NMR in small unilamellar vesicles and by 31P 2H NMR in phospholipidic dispersions of DMPC (dimyristoylphosphatidylcholine) in comparison with those of POLYA and CYSP alone. 1H-NMR chemical shift variations showed that CYSP really interacts with POLYA, with possible adduct formation, dispersion in the solid matrix of the POLYA, and also complex formation. A coarse approach to the latter mechanism was tested using the continuous variations method, indicating an apparent 1 : 1 stoichiometry. Calculations gave an apparent association constant of log Ka = 4.5. A study of the interactions with phospholipidic dispersions of DMPC showed that only limited interactions occurred at the polar head group level (31P). Conversely, by comparison with the expected chain rigidification induced by CYSP, POLYA induced an increase in the fluidity of the layer while ASD formation led to these effects almost being overcome at 298 K. At higher temperature, while the effect of CYSP seems to vanish, a resulting global increase in chain fluidity was found in the presence of ASD.

EPR spin label study of walnut oil effects on phosphatidylcholine membranes

Effects of walnut oil (WO) on dynamic and thermodynamic properties of 0–50 wt% cholesterol (CH) containing dimyristoylphosphatidylcholine (DMPC) and 10 wt% CH containing dipalmitoylphosphatidylcholine (DPPC) membrane dispersions were studied by electron paramagnetic resonance (EPR), using 5-doxyl stearic acid (5-DSA) and 16-doxyl stearic acid (16-DSA). Incorporation of 10 wt% WO alone decreased the phase transition temperature and created depth-dependent effects at the gel phase. The order increased close to the head region and decreased in the hydrocarbon core of the DMPC bilayer. For DPPC, the order decreased both close to head region and in the hydrocarbon core. Ten weight percent WO did not have considerable effect at the fluid phase for both DMPC and DPPC. Incorporation of 40 wt% WO into DMPC created an abrupt decrease in the maximum hyperfine splitting values after 305 K. The effect of 10 wt% WO in CH containing DMPC dispersions was dependent on the CH concentration. An increase and a decrease in the order were observed at low and high CH concentrations, respectively. Incorporation of WO created different effects on fluidity of 10 wt% CH containing DMPC and DPPC dispersions. Close to the head group region, the order in DMPC increased both in the gel and fluid phases; but for DPPC, an opposite effect was observed in both of the phases. In the hydrocarbon core of the bilayer, addition of 10 wt% WO into 10 wt% CH containing DMPC decreased the order in the gel phase and WO did not affect the order in the fluid phase. For DPPC, WO effects were observed to alter with temperature. In the studied temperature range, order parameters, diffusion constants and effective tilt angles were obtained from simulations of the spectra using Microscopic Order Macroscopic Disorder (MOMD) and Vary Anisotropic Reorientation (VAR) models. For 16-DSA, spectra were also simulated using two domains with EPRSIM.

Time-Dependent Absorption Evidence of Phenylurea-Derived Herbicides on Model Biomembranes: A DSC Study

Environmental Context. The need to improve the quality of agricultural cultivation and to control the weed growth of non-cultivated area has brought herbicides to wide-scale use. Unfortunately, these compounds have been proved to be toxic for many organisms, humans included. With an aim to a better knowledge of the mobility and fate of environmental pollutants, it becomes very important to have an exact knowledge of the role that the medium in which these products are dissolved or dispersed plays on the absorption of these products by biological membranes as a complement to information on the interaction between these compounds and cells. Abstract. Phenylurea derivatives are used as herbicides that inhibit photosynthesis. These materials enter plants via their roots and are employed for selective control of germinating grass and broad-leaved weeds in many crops as well as for total weed control of non-cultivated areas such as roads, railways, and parks. The study of the interaction between the herbicide and lipid membrane is interesting in assessing the relevance of the dispersing medium in the absorption processes. Differential scanning calorimetry (DSC), which detects the effect of foreign molecules on the phase transition from an ordered to a disordered lipid structure when submitted to heating, was employed to study such an interaction. Effects exerted by four phenylurea herbicides (difenoxuron, diuron, metoxuron, and linuron) on the thermotropic behavior of model membranes were here investigated. Aqueous dispersions of dimyristoylphosphatidylcholine (DMPC) were used as model membranes to study herbicide–membrane interactions. Experiments carried out by leaving herbicides, in a powdered form, in contact with DMPC-based multilamellar or unilamellar vesicles show that all the examined herbicides are able, but in different ways, to migrate through the aqueous medium and interact with model membranes. Experiments on herbicide-loaded liposomes in contact with empty ones indicate that the compounds are able to migrate from a loaded membrane to an empty one. The obtained data seem to validate the use of the DSC in demonstrating that bioactive and potentially toxic compounds not only to interact with biological membranes but also adsorb into a cell when dispersed in a lipophilic medium.

Phospholipid black foam films: dynamic contact angles and gas permeability of DMPC+DMPG black films

The behavior of black foam films from aqueous dispersions of dimyristoylphosphatidyl-choline (DMPC) with addition of the soluble phospholipid dimyristoylphosphatidylglycerol (DMPG) has been studied in dynamic conditions. The dynamic contact angles θ and the gas permeability coefficient K have been measured using the diminishing bubble method. The DMPC vesicle suspension in water is obtained through sonication and DMPG is dissolved in it. Two solutions with different NaCl concentrations (0.1 M and 0.5 M) have been studied. The behavior of the dynamic contact angles is very different for DMPC black films with, and without DMPG, respectively. They follow very different time dependence during spontaneous or forced variations of the bubble size. The gas permeability coefficient is significantly reduced by the DMPG addition. The NaCl concentration also influences this specific behavior. It seems that the electrically charged DMPG anions, which determine a significant electrostatic disjoining pressure, play an important role for this specific behavior. The results are discussed in connection with data regarding the thickness and structure of these black foam films. Films from DMPC+DMPG solutions in ethanol plus water mixed solvent have been studied as well, but no quantitative results could be obtained.

Phospholipid black foam films: dynamic film tension of DMPC bilayer films

Abstract The film tension of bilayer Newton black films (NBF) from aqueous dispersions of dimyristoylphosphatidylcholine (DMPC) has been studied in dynamic conditions. The dynamic film tension values γ have been measured using the capillary method for direct measurement of the film tension. Two different solutions have been used: DMPC vesicle suspension in water obtained through sonication, denoted as ‘DMPC(Son)’ (the DMPC adsorption layers are insoluble monolayers) and DMPC dissolved in ethanol plus water mixed solvent, denoted as ‘DMPC(EthW)’ (the DMPC adsorption layers are soluble). Both solutions contain 0.1 M NaCl. The behavior of the dynamic film tension is different for NBF from the two types of solutions. In the case DMPC(Son) γ strongly depends on the film area, while in the case DMPC(EthW) this dependence is less pronounced but still exists. The dependence of the film tension on the film area in case DMPC(Son) is well described by the Frumkin equation modified for bilayer films. Reasonable values of the parameters of Frumkin equation are determined from its fit to the experimental data.

Phospholipid black foam films: dynamic contact angles and gas permeability of DMPC bilayer films

In the case (i) the initially constant θ( t) sharply increase after approximately 2 h of the spontaneous diminishing of the bubble, they follow the gas pressure variation in the cell and depend on the film area. On the contrary in case (ii) the θ( t) values are almost constant during the spontaneous diminishing of the bubble as well as during the gas pressure variation in the cell and they do not depend on the film area. The gas permeability coefficient is larger in case (ii). The results are discussed in connection with the thickness and structure of the NBF from the two types of solutions, taking into account the solubility (or insolubility) and the hydration of the adsorption layers of the DMPC molecules.

Phospholipid matrix as a target for sulfur mustard (HD): NMR study in model membrane systems.

Although the interactions of sulfur mustard (HD) with nucleic acids and proteins have been well studied, the toxic interactions with the membrane matrix and specially the phospholipid bilayer have so far been poorly investigated. We have used several NMR techniques to study these interactions: 1H NMR to observe the localization of HD in membranes of small unilamellar vesicles (SUV) of lecithin; 31P NMR to verify the hypothesis of pore formation in membranes of large unilamellar vesicles (LUV); and pseudo solid state 31P and 2H NMR to analyze the dynamic consequences of the presence of HD in multilayer dispersions of dimyristoylphosphatidylcholine (DMPC). Immediate and late modifications of the DMPC-HD complexes have been observed at the macroscopic and microscopic levels. After intoxication, HD is spontaneously incorporated into the membrane and locates at the level of the chain methylene groups. This incorporation occurs without formation of pores in the membrane. The presence of HD in the phospholipid dispersion differentially increases the membrane fluidity depending upon the level involved. Weak at the superficial level (phosphate group), this increase is dose-dependent on progression into the membrane. This increase is related to a lowering of transition temperature when measured at the chain level. Macroscopically, HD induces dose- and time-dependent modifications of the DMPC-HD complexes, leading to the formation of an optically transparent gel. This gel formation is confirmed at a microscopic level, where all structures disappear after intoxication.

Singular behavior of three-phase contact between DMPC dispersion, air and quartz at the critical temperature of 29°C

The formation of wetting films on a horizontal quartz surface from diluted (10 −2 mg ml −1) dispersions of dimyristoylphosphatidylcholine (DMPC) multilamellar vesicles was studied in the temperature range 25–32°C. Film lifetimes, dynamic and equilibrium (static) three-phase contact angles were measured. The first two parameters exhibit singular behavior at the critical temperature for unilamellar state formation reported by Gershfeld et al. (1993) [N.L. Gershfeld, C. Muud, K. Tajima, R. Berger, Biophys. J. 65 (1993) 1174–1179]. The film lifetime is at a minimum at 29°C, whereas the dynamic contact angle displays a maximum at that temperature. The equilibrium contact angle of the wetting film increases with the temperature in the studied range and appears to be insensitive to the critical temperature. The surface tension of the DMPC dispersions at the air–water interface decreases with the temperature and displays a specific feature at 29°C manifested by a steeper decrease of this quantity above the critical temperature. These data demonstrate, by an independent method, the existence of a peculiar point at T*=29°C in DMPC dispersions.

Molecular dynamics of lipid association at the hydrophobic interface of gramicidin S

Gramicidin S was incorporated into dimyristoylphosphatidylcholine dispersions and the observed two-component EPR spectra of spin-labelled lipids at 30 °C were analysed by a two-stage algorithm, including spectral subtractions and two-site exchange simulations. A limited range of temperatures around 30 °C was found suitable for such measurements. It has been found that negatively charged labelled lipids display a selectivity towards the intramembranous part of the peptide. The relative association constants for spin-labelled stearic acid (14-SASL) and phosphatidylserine (14-PSSL) were Kr = 2.08± 0.10 and 1.18±0.08, respectively, when compared with the zwitterionic phosphatidylcholine label (14-PCSL, Kr≡ 1). The lateral diffusion of spin-labelled lipids in peptide-free regions causes exchange between those labels in the bulk fluid lipid phase and motionally restricted boundary labelled lipids at the apolar interface of gramicidin S. Owing to exchange, the spectral anisotropy of labelled lipids giving rise to the slow-motion spectral component was gradually decreased, and there was an augmentation of spectral intensity in between the motionally restricted (slow motion) and the fast tumbling (motionally averaged) labelled lipid components. Two-component exchange simulations allowed the determination of off-rates of labelled phospholipids, showing an inverse proportionality with lipid-protein selectivity. Spin-labelled procaine exhibited limited selectivity and fast exchange similar to the on-coming non-specifically associated lipids.

Phospholipid free thin liquid films with grafted poly(ethylene glycol)-2000: formation, interaction forces and phase states

Free thin liquid films (foam films) formed from aqueous dispersions of dimyristoylphosphatidylcholine (DMPC) and dipalmitoylphosphatidylethanolamine with covalently bound poly-(ethylene glycol) of molecular weight 2000 (DPPE-PEG-2000) were studied by the thin liquid film microinterferometric technique of Scheludko and Exerowa in the temperature range 14–36°C. The surface tension kinetics of the dispersions were studied in order to ensure equilibration of the foam films. These measurements showed that the rate of surface coverage depends slightly on the temperature and does not reach equilibrium values within reasonable time intervals for the dispersions containing only one amphiphile (DPPE-PEG-2000). The destruction of the vesicles at the air/(aqueous dispersion) interface was much faster for the dispersions containing DMPC/DPPE-PEG-2000 mixtures above 23°C, the temperature of the chain-melting phase transition of the main lipid component (DMPC). The dependence of the equilibrium thickness of the foam films on the electrolyte concentration was measured for 1 and 9 mol% DPPE-PEG-2000 at 28°C in the range 10 −3 to 0.5 M NaCl. These results indicate that at the low electrolyte concentrations the electrostatic and van der Waals interactions are dominant similar to the foam films stabilized with DMPC alone. At the high electrolyte concentrations the steric repulsion of the PEG layers becomes dominant. The temperature–composition dependence of the bilayer thickness was measured for the foam bilayers at 0.14 M NaCl. The data for the foam bilayer thickness and the comparison with the phase diagrams of PC/PE-PEG dispersions, show that the DMPC/DPPE-PEG-2000 foam bilayers are able to exist in two phase states characterised by different conformations (mushroom and brush) of the grafted polymer.

Fourier Transform Infrared Spectra and Molecular Orientation of Black Lipid Films in Air Interacting with Metal Ions

Polarized Fourier transform infrared spectra of black lipid films in air withdrawn from aqueous dispersions of dimyristoylphosphatidylcholine (DMPC) have been recorded at various electrolyte concentrations. The spectra revealed that monovalent cations gave little effect, whereas polyvalent cations except Mg2+ gave remarkable effects on vibrational wavenumbers of both the acyl chain and phosphate ester group of DMPC in black lipid films. In the case of Mg2+, the effect was discerned only for the phosphate ester group. By use of thin film optics, molecular orientation of DMPC in the black lipid film in air was quantitatively evaluated as a function of electrolyte concentration. In the case of monovalent and Mg2+ cations, the orientation angle of the acyl chain axes from the surface normal was little affected by the electrolyte addition. On the other hand, the other polyvalent cations caused decrease of the average orientation angle from ca. 40° to 30°. The affinity of the cations for the DMPC lipid films in air are in the order of Zn2+ > La3+ ∼ Ca2+ ∼ Pb2+ > Mg2+ > Na+ ∼ Li+.

Lipid mixing in dimyristoyl phosphatidylcholine-dimyristoyl glycerol dispersions: spin label ESR studies

The lipid transfer and mixing properties in hydrated dispersions of dimyristoyl phosphatidylcholine (DMPC)/dimyristoyl glycerol (DMG) binary mixtures have been investigated by using electron spin resonance spectroscopy of the spin-labelled lipid components. The assay system is based on the reduction in spectral broadening from spin–spin interactions that takes place by dilution of the spin-labelled lipid on transfer to a 9-fold excess of dispersions that contain no spin label. Lipid dispersions with DMPC:DMG compositions of 70:30, 40:60 and 20:80 mol/mol, for which the fluid phases have lamellar, inverted hexagonal and isotropic structures, respectively, have been studied. Essentially no transfer of spin-labelled lipid takes place for any of the lipid mixtures in the lamellar gel phase, or in dispersions of DMPC alone at all temperatures studied. The greatest degrees of transfer are found in the fluid phase of the DMPC/DMG mixtures. In general, the extent of lipid transfer is greater for the diacylglycerol component than for the phosphatidylcholine component. The extent of transfer of phosphatidylcholine is very low in the fluid lamellar phase of the 70:30 mol/mol DMPC/DMG, as compared with that of diacylglycerol. Only in the case of the 40:60 mol/mol DMPC/DMG mixture, in the inverted hexagonal phase, are the extents of transfer comparable for both phosphatidylcholine and diacylglycerol components, indicating a bulk transfer of lipid within the dispersions. The largest extent of transfer is found for diacylglyerol in the 20:80 mol/mol DMPC/DMG mixture in the isotropic phase.

Critical temperature for unilamellar vesicle formation in dimyristoylphosphatidylcholine dispersions from specific heat measurements

Using a heat conduction calorimeter with very high resolution (+/- 0.00005 J/degrees C.cm3), we have measured the specific heat CpL between 25 and 35 degrees C of dimyristoylphosphatidylcholine (DMPC) in aqueous dispersions. Previous studies of the temperature dependence of the chemical potential of DMPC in the L alpha phase (lamellar, liquid crystalline) indicated that a dispersion consisting only of unilamellar vesicles forms spontaneously at a critical temperature T* of 29.0 degrees C. Our present measurements show an anomaly in CpL between 28.70 and 29.50 degrees C: the curve for CpL versus T first decreases and then exhibits an inflection point at 28.96 degrees C before it flattens. This anomaly is attributed to the transformation from multilamellar dispersion to unilamellar vesicles at T* = 28.96 degrees C. Two independent properties of the CpL data also indicate T* is a critical point for the formation of unilamellar vesicles: (a) the time to reach equilibrium upon changing temperature increased dramatically between 28.7 and 28.96 degrees C, increasing as (T* - T)-1; at T > T* the dramatic "slowing-down" phenomenon was not observed. This slowing-down near T* is a general characteristic of critical phenomena. (b) The free energy change for the multilamellar-unilamellar transformation was obtained from the CpL-T data over this temperature interval and found to be 3.2 J/mol or 0.016 ergs/cm2 of bilayer, in agreement with other estimates of the interaction energy between neutral bilayers. We conclude with a discussion of the implications for membrane bilayer stability of these newly identified dynamic properties of the transformation.

Two-dimensional rotational-echo double resonance of Val1-[1-13C]Gly2-[15N]Ala3-gramicidin A in multilamellar dimyristoylphosphatidylcholine dispersions.

The dipolar coupling between the Gly2 13C-1 carbon and Ala3 15N-amide nitrogen was used to investigate the conformation and dynamics of the Gly2-Ala3 13C-15N peptide bond in Val1-[1-13C]Gly2-[15N]Ala3-gramicidin A incorporated into multilamellar dispersions of dimyristoylphosphatidylcholine. Measurement of the 13C-15N dipolar coupling constant D of the labeled gramicidin in a powder and the effective dipolar coupling constant De in a multilamellar dispersion was accomplished by two-dimensional rotational-echo double-resonance (2D REDOR) NMR, a magic-angle spinning experiment designed to measure weak dipolar coupling constants. The magnitudes of D and De were measured by the mirror-symmetric form of 2D REDOR, and the signs of D and De were determined relative to the sign of the isotropic indirect spin-spin coupling constant J by the mirror-asymmetric form of 2D REDOR. From knowledge of the magnitudes of D and De, four possible values were calculated for the angle between the Gly2-Ala3 13C-15N peptide bond and the gramicidin helical axis. Additional knowledge of the signs of D and De permitted the set of possible values for the peptide bond angle to be reduced to a single angle and its supplement (64 degrees, 116 degrees). This information about the Gly2-Ala3 13C-15N peptide bond angle eliminates the double-stranded, helical dimers and the left-handed, single-stranded, beta 6.3 helical dimer but supports the right-handed, single-stranded, beta 6.3 helical dimer as the structural model for gramicidin in multilamellar dispersions.

Conformation and Orientation of Gramicidin a in Oriented Phospholipid Bilayers Measured by Solid State Carbon-13 NMR

Three analogues of the helical ionophore gramicidin A have been synthesized with (13)C-labeled carbonyls ((13)C=O) incorporated at either Gly(2), Ala(3), or Val(7). A fourth compound incorporated (13)C at both the carbonyl and alpha-carbon of Gly(2) within the same molecule. These labels were studied using solid-state, proton-enhanced, (13)C nuclear magnetic resonance (NMR) in hydrated dispersions of dimyristoylphosphatidylcholine (DMPC)-gramicidin A. The dispersions were aligned on glass coverslips whose orientation to the magnetic field could be varied through 180 degrees . The orientation dependence of the NMR spectrum was used to obtain an accurate measurement of the (13)C chemical shift anisotropy (CSA), and in the case of the fourth compound, the (13)C-(13)C dipolar coupling constant. From the measured CSA and estimates of the orientation of the (13)C shielding tensor, we are able to determine the direction of the (13)C=O bonds and to compare these with the predictions of the various reported models for the configuration of gramicidin A in phospholipid bilayers. Our results are consistent with the left-handed pipi(6.3) (LD) single-stranded helix (Urry, D. W., J. T. Walker, and T. L. Trapane. 1982. J. Membr. Biol. 69:225-231). The right-handed pipi(6.3) (LD) single-stranded helix observed for gramicidin A in sodium dodecyl sulfate micelles (Arseniev, A. S., I. L. Barsukov, V. F. Bystrov, A. L. Loize, and Yu A. Ovchinnikov. 1985. FEBS (Fed. Eur. Biochem. Soc.) Lett. 186:168-174) yields a poorer fit to the data. However, the width of the carbonyl resonances suggests a distribution of molecular geometries possibly resulting from a spread in the helix pitch and handedness. Double-stranded helices and beta sheet structures are excluded. In dispersions in which the lipid is in the L(alpha) phase, the gramicidin A undergoes rapid reorientation about an axis which is centered on the normal to the plane of the coverslips. When the supporting lipid is in the L(beta') phase the helices are rigid on the timescale of (13)C-NMR. The configuration of gramicidin A is unaltered by L(alpha)-L(beta') phase transition of the bilayer lipid.

A calorimetric examination of the effect of myotoxin a on the thermotropic phase behavior of model lipid membranes

The effect of myotoxin a on the thermotropic phase behavior of aqueous dispersions of dimyristoyl phosphatidylcholine (DMPC) and dimyristoyl phosphatidylserine (DMPS) was examined using differential scanning calorimetry (DSC). Myotoxin a significantly altered the normal phase behvior of DMPC in a concentration dependent fashion. This effect is perturbed by Ca e+ and is sensitive to ionic strength and pH. High concentrations of toxin eliminate the characteristic pretransition associated with the polar head group of DMPC. They also increase the temperature of the main gel-to-liquid crystal transition from 23°C to 32–235°C. At low concentrations of toxin, the first visible effect is upon the pretransition which is split into two components that diminish with time. The main transition is less affected at low toxin concentrations, although the magnitude of the transition is reduced while it is simultaneously shifted to higher temperatures. The main transition is also split into multiple components. The toxin also had pH specific effects on the phase behavior of DMPS. Above physiological pH (8.5) the normal transition of DMPS at 36–38°C was split in the presence of myotoxin a and new components appeared centered at 31°C and 35°C. These observations are consistent with reports that the skeletal muscle membrane system is the major site of the myonecrotic effect of myotoxin a.

Raman-spectroscopic evidence for the incorporation of alamethicin into dimyristoylphosphatidylcholine bilayers

The interaction of alamethicin with dimyristoylphosphatidylcholine dispersions was investigated by Raman spectroscopy. The temperature dependence of the CH stretching spectra demonstrates the incorporation of the polypeptide into the hydrophobic core even in the absence of a transmembrane potential gradient.

Phospholipid surface bilayers at the air-water interface. I. Thermodynamic properties

Dispersions of dimyristoylphosphatidylcholine (DMPC) in water spontaneously form a surface bilayer at the equilibrium air/water surface (Gershfeld, N. L., and K. Tajima, 1979, Nature [Lond.]. 279: 708-709). This phenomenon has now been demonstrated with dispersions of dioleoylphosphatidylcholine (DOPC), dipalmitoylphosphatidylcholine (DPPC), and with a mixture of DMPC and DOPC. Each of these dispersions forms a surface bilayer at a singularity in temperature that is a characteristic of the phospholipid. The surface bilayer formed by the lipid mixture is shown to have the same composition as the bulk liquid-crystal phase of the dispersion, and the surface components have identical partial molar entropies as the bulk lipid components. These properties indicate that the surface bilayer has the same structure as the bilayer in the liquid-crystal phase of the bulk dispersion.

Dispersions of dimyristoylphosphatidylcholine (DMPC) in water have been reported to form a structure at 29 degrees C at the equilibrium air/water surface with a molecular density equal to that of a typical bilayer. In this study, the water permeability of this structure has been evaluated by measuring the rate of water evaporation from DMPC dispersions in water in the temperature range where the surface film density exceeds that of a monolayer. Evaporation rates for the lipid dispersions did not deviate from those for lipid-free systems throughout the entire temperature range examined (20-35 degrees C) except at 29 degrees C, where a barrier to evaporation was detected. This strengthens the view that the structure that forms at this temperature has the properties of a typical bilayer.

Phase diagrams of bilayers of dimyristoyl lecithin plus heteroacid lecithins

Aqueous dispersions of dimyristoyl phosphatidylcholine (DMPC) mixed with 1-palmitoyl-2-oleoyl phosphatidylcholine (POPC) or with 1-palmitoyl-2-stearoyl phosphatidylcholine (PSPC) have been studied by differential scanning calorimetry. Analysis of phase diagrams of the resulting bilayers indicated that both POPC-DMPC were miscible, but not ideally mixed, in the gel and in the liquid-crystalline states. In both states, estimated values for excess interaction energies were more positive for PSPC-DMPC than for POPC-DMPC, indicating that mixing was less ideal in the former system.
