Cholesterol drives enantiospecific effects of ibuprofen in biomimetic membranes

The interaction between chiral drugs and biomimetic membranes is of interest in biophysical research and biotechnological applications. There is a belief that the membrane composition, particularly the presence of cholesterol, could play a pivotal role in determining enantiospecific effects of pharmaceuticals. Our study explores this topic focusing on the interaction of ibuprofen enantiomers (S- and R-IBP) with cholesterol-containing model membranes. The effects of S- and R-IBP at 20 mol% on bilayer mixtures of dipalmitoylphosphatidylcholine (DPPC) with 0, 10, 20 and 50 mol% cholesterol were investigated using circular dichroism and spin-label electron spin resonance. Morphological changes due to IBP enantiomers were studied with atomic force microscopy on supported cholesterol-containing DPPC monolayers. The results reveal that IBP isoforms significantly and equally interact with pure DPPC lipid assemblies. Cholesterol content, besides modifying the structure and the morphology of the membranes, triggers the drug enantioselectivity at 10 and 20 mol%, with the enantiomers differently adsorbing on membranes and perturbing them. The spectroscopic and the microscopic data indicate that IBP stereospecificity is markedly reduced at equimolar content of Chol mixed with DPPC. This study provides new insights into the role of cholesterol in modulating enantiospecific effects of IBP in lipid membranes.

A Sigma1 Receptor Agonist Alters Fluidity and Stability of Lipid Monolayers.

Interactions between the sigma1 receptor agonist PRE-084 and various lipid monolayers, including dipalmitoylphosphatidylcholine (DPPC), DPP-ethanolamine (DPPE), DPP-glycerol (DPPG), DPP-serine (DPPS), palmitoylsphingomyelin (PSM), and cholesterol (Ch), were investigated to elucidate the effects of PRE-084 on membrane fluidity and stability. Their interactions with sigma1 receptor agonists have potential implications for neuroprotection, antidepressant, analgesic, and cognitive enhancement effects. In this study, we observed that the presence of PRE-084 in the subphase led to increased fluidity in DPPC and DPPE monolayers, whereas decreasing fluidity was observed in DPPG, DPPS, and PSM monolayers. The interaction of PRE-084 with Ch monolayers was found to be distinct from its interaction with other lipids. Fluorescence microscopy images revealed changes in the size and shape of liquid-condensed domains in the presence of PRE-084, supporting the notion of altered membrane fluidity. Our findings provide new insights into the interaction of PRE-084 with lipid monolayers and its potential implications for biological and membrane science.

The Interaction between Anesthetic Isoflurane and Model-Biomembrane Monolayer Using Simultaneous Quartz Crystal Microbalance (QCM) and Quartz Crystal Impedance (QCI) Methods.

The interaction between anesthetic Isoflurane (Iso) and model-biomembrane on the water surface has been investigated using quartz crystal microbalance (QCM) and quartz crystal impedance (QCI) methods. The model-biomembranes used were dipalmitoyl phosphatidyl choline (DPPC), DPPC-palmitic acid (PA) mixture (DPPC:PA = 8:2), DPPC-Alamethicin (Al) mixture (DPPC:Al = 39:1), and DPPC-β-Lactoglobulin (βLG) mixture (DPPC:βLG = 139:1) monolayers, respectively. The quartz crystal oscillator (QCO) was attached horizontally to each monolayer, and QCM and QCI measurements were performed simultaneously. It was found that Iso hydrate physisorbed on each monolayer/water interface from QCM and changed those interfacial viscosities from QCI. With an increase in Iso concentration, pure DPPC, DPPC-PA mixed, and DPPC-Al mixed monolayers showed a two-step process of Iso hydrate on both physisorption and viscosity, whereas it was a one-step for the DPPC-βLG mixed monolayer. The viscosity change in the DPPC-βLG mixed monolayer with the physisorption of Iso hydrate was much larger than that of other monolayers, in spite of the one-step process. From these results, the action mechanism of anesthetics and their relevance to the expression of anesthesia were discussed, based on the "release of interfacial hydrated water" hypothesis on the membrane/water interface.

Real-time topography inspection of DPPC monolayers using a surface-plasmon resonance sensor

This paper presents an alternative optical characterization of biosensors based on supported lipid monolayers (SLMs). Developing these biosensors requires precise thickness characterization of the films to understand their structure and dynamics. This paper proposes an optical technique to measure the thickness, optical properties, and location of a dipalmitoyl-phosphatidylcholine (DPPC) SLM on top of a metallic thin film. DPPC SLMs are of interest for biosensing applications, such as detecting pulmonary-related infections like SARS-CoV-2, Avian Influenza, and the H1N1 influenza virus. The monolayer was fabricated using the Langmuir–Blodgett technique, and the experimental characterization consisted of measuring the surface-plasmon resonance angle in the Kretschmann configuration. This technique provides an alternative option for real-time visual inspection and determination of the location and shape of DPPC monolayers in large areas. Therefore, it offers a useful tool for further developing SLM-based biosensors.

Numerical investigations of translocation characteristics of non-spherical silica nanoparticles across pulmonary surfactant monolayer under different respiratory states using coarse-grained molecular dynamics method

Understanding translocation characteristics of non-spherical silica nanoparticles (SiNPs) across pulmonary surfactant (PS) monolayer is of great significance for their applications in inhaled nanoparticles (NPs) drugs, as their shapes may impact drug delivery efficiency. Therefore, this study has performed coarse-grained molecular dynamics (CGMD) simulation to study the characteristics of SiNPs across the PS monolayer and adsorption of dipalmitoylphosphatidylcholine (DPPC) molecules. It is found that SiNPs smaller than 8 nm are more likely to cross the PS monolayer in expansion state than other states and crossing times required for ellipsoidal SiNPs are the shortest, and when SiNPs are in penetrating and embedding states, the number of DPPC molecules adsorbed by bulged-spherical SiNPs is the highest. In conclusion, ellipsoidal SiNPs have the strongest crossing ability and bulged-spherical SiNPs cause the greatest damage. Therefore, it is suggested to adopt ellipsoidal SiNPs and avoid bulged-spherical SiNPs when improving transportation efficiencies of spherical SiNP drug carriers.

Bioinspired nanoarchitectonics at the air-water interface to understand the interaction of lipids with a Europium-coordinated quinoline derivative

5SO3H-8-hydroxyquinoline coordinated to Europium (Eu-5SO3-HQ) was incorporated in biomembrane models using Langmuir monolayers. Dipalmitoyl phosphatidylcholine (DPPC) and dipalmitoyl phosphatidylserine (DPPS) were employed, representing mammalian cells and dioctadecyldimethylammonium bromide (DODAB) as a positively charged lipid to study the contrast with negatively charged lipids. Tensiometric, rheological and spectroscopic techniques were employed to characterize Eu-5SO3-HQ- lipid monolayer interactions. The complex condenses all the monolayer indicating interactions with the lipids' polar heads, but with distinctive effects on the mechanical and rheological properties. While the complex decreases the compression and elastic moduli of DPPC and DPPS monolayers, it increases for DODAB, also decreasing its lateral viscosity. Infrared spectroscopy shows that the interaction of Eu-5-SO3-HQ alters the ordering of the lipids' alkyl chains, impacting the monolayer's molecular packing. These results show that the interaction of Eu-5SO3-HQ with lipid monolayers at the air-water is modulated by the composition of the polar head, which can be supportive in the preparation of nanodevices for molecular probing.

Effect of partial O-methylation in dehydrodieugenol on its antitrypanosomal activity - correlation with the toxicity using cell membrane models

Biseugenol (1), a neolignan with antiprotozoal activity against Trypanosoma cruzi, was partially methylated, and the compound obtained – methyl biseugenol (2) – had its activity evaluated against the extracellular (trypomastigotes) and intracellular (amastigotes) forms of T. cruzi. It was observed that both compounds 1 and 2 exhibited similar effects against trypomastigotes (IC50 of 11.7 and 16.2 μM, respectively), whereas compound 2 displayed higher activity against amastigotes (IC50 = 8.2 μM) in comparison with biseugenol (IC50 = 15.4 μM). Additionally, reduced toxicity against NCTC cells for compound 2 was observed (CC50 > 200 μM), differently from compound 1 with CC50 = 58.0 μM. Aiming to understand better the molecular mechanism of the biological action of compound 2, the prodrug was incorporated into cellular membrane models constituted of Langmuir monolayers of the lipids dipalmitoylphosphatidylcholine (DPPC), dipalmitoylphosphatidylethanolamine (DPPE), dipalmitoylphosphatidylserine (DPPS), and dipalmitoylphosphatidylglycerol (DPPG). The lipid-drug interaction was inferred through tensiometry, surface potential, infrared spectroscopy (PM-IRRAS), and Brewster angle microscopy (BAM). The prodrug expanded DPPC and DPPG monolayers and condensed DPPE ones, as well as presented characteristic behaviors regarding the chemical structure of the lipid considering expansion-compression curves, surface potential-area isotherms, and stability of previously compressed monolayers to relevant-biological surface pressures. PM-IRRAS indicated a molecular disorder for DPPC and DPPS alkyl chains in the presence of the drug. BAM revealed the presence of domains in the DPPG and DPPE monolayers, which was probably induced by the prodrug. These data suggest, in general, that the lipid composition modulates the interaction of compound 2, whose results are expected to correlate to its trypanocidal activity, which involves the plasma membrane of T. cruzi as the primary target, i.e., the first barrier that the compound should encounter to interact with the microorganism.

Heterogeneous Nanostructures Cause Anomalous Diffusion in Lipid Monolayers.

The diffusion and mobility in biomembranes are crucial for various cell functions; however, the mechanisms involved in such processes remain ambiguous due to the complex membrane structures. Herein, we investigate how the heterogeneous nanostructures cause anomalous diffusion in dipalmitoylphosphatidylcholine (DPPC) monolayers. By identifying the existence of condensed nanodomains and clarifying their impact, our findings renew the understanding of the hydrodynamic description and the statistical feature of the diffusion in the monolayers. We find a universal characteristic of the multistage mean square displacement (MSD) with an intermediate crossover, signifying two membrane viscosities at different scales: the short-time scale describes the local fluidity and is independent of the nominal DPPC density, and the long-time scale represents the global continuous phase taking into account nanodomains and increases with DPPC density. The constant short-time viscosity reflects a dynamic equilibrium between the continuous fluid phase and the condensed nanodomains in the molecular scale. Notably, we observe an "anomalous yet Brownian" phenomenon exhibiting an unusual double-peaked displacement probability distribution (DPD), which is attributed to the net dipolar repulsive force from the heterogeneous nanodomains around the microdomains. The findings provide physical insights into the transport of membrane inclusions that underpin various biological functions and drug deliveries.

Incorporation of dehydrodieugenol, a neolignan isolated from Nectandra leucantha (Lauraceae), in lipid Langmuir monolayers as biomembrane models

Dehydrodieugenol, a neolignan isolated from the Brazilian plant Nectandra leucantha (Lauraceae) with reported antiprotozoal and anticancer activity, was incorporated in Langmuir monolayers of selected lipids as cell membrane models, aiming to comprehend its action mechanism at the molecular level. The interaction of this compound with the lipids dipalmitoylphosphatidylcholine (DPPC), dipalmitoylphosphatidylethanolamine (DPPE), dipalmitoylphosphatidylserine (DPPS), and dipalmitoylphosphatidylglycerol (DPPG) was inferred through tensiometry, infrared spectroscopy, and Brewster angle microscopy. The interactions had different effects depending on the chemical nature of the lipid polar head, with expansion for DPPC monolayers, condensation for DPPE, and expansion (at low surface pressures) followed by the overlap of the isotherms (at high surface pressure values) for DPPS and DPPG. Effects caused by dehydrodieugenol in the negatively charged lipids were distinctive, which was also reflected in the hysteresis assays, surface potential-area isotherms, and rheological measurements. Infrared spectroscopy indicated that the drug interaction with the monolayer affects not only the polar groups, but also the acyl lipid chains for all lipids. These results pointed to the fact that the interaction of the drug with lipid monolayers at the air-water interface is modulated by the lipid composition, mainly considering the polar head of the lipids, as well as the hydrophobicity of the lipids and the drug. As negatively charged lipids pointed to distinctive interaction, we believe this can be related to the antiprotozoal and anticancer properties of the compound.

Dominant hydrophobic interactions with β-glucan in nanoarchitectonics with mixed Langmuir monolayers of cholesterol/dipalmitoyl phosphatidyl choline.

The polysaccharide β-glucan, found in the cell wall of cereals such as wheat, oats, and barley, is believed to lower the concentration of bad cholesterol in humans, but the molecular-level mechanisms responsible for such an action are unknown. In this study, we use Langmuir monolayers of cholesterol and dipalmitoyl phosphatidyl choline (DPPC) as cell membrane models that are made to interact with β-glucan. Neat cholesterol and mixed cholesterol/DPPC monolayers were expanded upon incorporating β-glucan from the aqueous subphase. This incorporation was found to induce ordering in mixed monolayers and dehydration of the carbonyl group at higher cholesterol concentrations. These effects are attributed to hydrophobic interactions as identified with polarization-modulated infrared reflection-absorption spectroscopy. They correlate well with the hypothesis that cholesterol levels can be lowered by the formation of soluble fibers with β-glucan through hydrophobic interactions, blocking cholesterol absorption by the organism.

Spontaneous evolution of equilibrium morphology in phospholipid-cholesterol monolayers.

Competition between intradomain electrostatic repulsions and interdomain line tension leads to domain shape transitions in phase-separating lipid monolayers. The question remains if these morphologies are energy minima or are kinetically trapped metastable states. We show the reversible evolution of uniform width stripe domains from polydisperse semicircular domains in monolayers of dipalmitoylphosphatidylcholine (DPPC), hexadecanol (HD) or palmitic acid (PA), and dihydrocholesterol (DChol). The initial semicircular domains grow at a fixed 2:1 DPPC:HD (or PA) stoichiometry, depleting the liquid phase of HD, leaving behind a liquid enriched in DPPC and DChol. At higher surface pressures, the remaining DPPC precipitates onto existing domains, decreasing the ratio of line tension to the square of the dipole density difference, λ/μ2. Theory predicts that, as λ/μ2 decreases, circular domains reversibly transform to uniform width stripes as the minimum energy structure. Measuring the stripe width provides the first estimates of λ/μ2 at liquid condensed–liquid expanded phase coexistence.

In situ nonlinear optical spectroscopic study of the structural chirality in DPPC Langmuir monolayers at the air/water interface.

We conduct a molecular study on the structural chirality in Langmuir monolayers composed of dipalmitoylphosphatidylcholine (DPPC) using in situ nonlinear optical spectroscopies, including second harmonic generation (SHG) and sum frequency generation (SFG). Chiral SHG response is observed from L-DPPC monolayers at moderate surface pressures and almost vanishes at a high surface pressure. SFG spectra of L-DPPC monolayers show chiral features that can be assigned to the terminal CH3 groups and the CH2 groups attached to the chiral center atom. This means that these achiral moieties form chiral superstructures at the interface. Along with increasing surface pressure, the structural chirality of CH3 groups shows a similar trend as that of chiral SHG, but CH2 chirality increases monotonically. Furthermore, in a racemic DPPC monolayer with a moderate surface pressure, both chiral SHG and chiral SFG of CH3 groups are absent, whereas chiral SFG of CH2 groups is clearly present, indicating that L- and D-DPPC are diastereomers at the air/water interface and interfacial CH2 prefers a certain orientation regardless of the molecular handedness. A molecular mechanism is proposed to explain the origin of the structural chirality in DPPC monolayers.

Morphological Behavior of DPPC-DMPC Two-dimensional Mixed Monolayer on Water Surface by Dropping Method

We have investigated the morphology of dipalmitoyl phosphatidyl choline (DPPC)-dimyristoyl PC (DMPC) mixed monolayer formed on the water surface by the dropping method using surface tension measurement (STm), Brewster angle microscopy (BAM), and infrared external reflection spectroscopy (IERS). The limiting molecular area (A0) obtained by STm showed that the negative deviation at < mole fraction (xDMPC)= 0.4 whereas the positive deviation at > xDMPC =0.6 when DMPC was added to the DPPC monolayer. BAM images also showed that the condensed rigid monolayer at xDMPC < 0.4 changed to the expandable monolayer at xDMPC > 0.5. Moreover, IER spectra showed that the alkyl chains of both DPPC and DMPC molecules are in trans conformation in mixed monolayer at xDMPC =0.2 whereas gauche conformation existed at xDMPC = 0.8. Above findings have justified the transition of flexible gauche conformation alkyl chains of DMPC molecule into rigid trans conformation by the addition of DPPC molecule at xDMPC < 0.4 whereas alkyl chains retained flexible gauche conformation in both DPPC and DMPC molecules at xDMPC > 0.6.

Penetration of C60 into lung surfactant membranes: Molecular dynamics simulation studies

AbstractWe performed molecular dynamics simulations of the systems consisting of C60 molecules and dipalmitoylphosphatidylcholine (DPPC) monolayer membranes to study the penetration of C60 into lung surfactant (LS) membranes. The potential of mean force of the C60 penetration through the LS membrane was calculated as a function of the distance of a C60 molecule from the DPPC monolayer membrane. The free energy minimum of around 43 kcal/mol is located in the DPPC tail region, indicating that the C60 molecules can accumulate in the LS membrane region. The energy decomposition shows the main driving force of the C60 accumulation in the lipid tail region is the van der Waals interaction with the hydrocarbon tails of DPPC lipids. Finally, we observed that the water evaporation rate can be significantly enhanced by the accumulation of C60 molecules in the membrane tail region.

Novel cytotoxic amphiphilic nitro-compounds derived from a synthetic route for paraconic acids

A series of precursors for bioactive paraconic acids (PA) were synthesized and their cytotoxicity assessed on human cells in vitro. Two amphiphilic nitro-containing precursors, Nitro-C15-EED and the butanolide Nitro-C12-GBL, were cytotoxic at the micromolar scale, with higher activity on tumor HeLa cells than on HEK-293T of non-tumor origin. The structure of these molecules is simple but different from reported bioactive nitro compounds. Nitro-C12-GBL was generally more cytotoxic, but after short-term (2 h) exposure both compounds reached maximum cytotoxicity. At 72 h post-treatments of HeLa cells the final dose-response for Nitro-C12-GBL (LC50 = 21.9 µmol L−1) was close to that for Nitro-C15-EED (LC50 = 25.3 µmol L−1), corresponding to LC50s ~ 3–3.6 times lower than those on HEK-293T. Short-term treatments with 50 µmol L−1 of these compounds promoted comparable outcomes, reducing tumor cells viability up to 27–36% of the controls and preserving ~70% of HEK-293T viability at 72 h post-treatments. Reduced cytotoxicity was observed in cultures continuously exposed to the compounds for longer periods (24–72 h), especially on tumor cells, underlining short-term treatments as alternatives to antiproliferative strategies. Due to their amphiphilic nature, these compounds show spontaneous surface activity and adsorption onto Langmuir monolayers of dipalmitoyl phosphatidyl choline (DPPC), especially Nitro-C12-GBL. The effects on DPPC monolayers are indicative of a possible physiological action that depends on the interaction with the cell membranes. Coarse-grained molecular dynamics indicate that individualized molecules of Nitro-C15-EED and the less toxic PA precursors are susceptible to trapping into phospholipid films. In contrast, Nitro-C12-GBL consistently forms large aggregates with outward polar domains, which could favor interaction with phospholipid polar heads of biological membranes.

Wetting Dynamics of a Lipid Monolayer.

Direct measurement and control of the dynamic wetting properties of a lipid-coated water-air interface over a wide range of surface tension variations have many important applications. However, the wetting dynamics of the interface near its partial-to-complete wetting transition has not been fully understood. Here, we report a systematic study of the wetting dynamics of a lipid-coated water-air interface around a thin glass fiber of diameter 1-5 μm and length 100-300 μm. The glass fiber is glued onto the front end of a rectangular cantilever to form a "long-needle" atomic-force-microscope probe. Three surface modifications are applied to the glass fiber to change its wetting properties from hydrophilic to hydrophobic. A monolayer of phospholipid dipalmitoylphosphatidylcholine (DPPC) is deposited on the water-air interface in a homemade Langmuir-Blodgett trough, and the surface tension γL of the DPPC-coated water-air interface is varied in the range of 2.5 ≲ γL ≲ 72 mN/m. From the measured hysteresis loop of the capillary force for the three coated fiber surfaces with varying γL, we observe a sharp transition from partial to complete wetting when γL is reduced to a critical value (γL)c. The obtained values of (γL)c are 27 ± 1 mN/m for a DPPC-coated fiber surface and 23 ± 1 mN/m for an trichloro(1H,1H,2H,2H-perfluorooctyl) silane (FTS)-coated surface. Below (γL)c, the contact angle θ0 of the liquid interface is found to be zero for both hydrophobic fiber surfaces and the corresponding spreading parameter S becomes positive. For the FTS-coated fiber surface, the height of capillary rise exhibits a jump when γL is reduced to (γL)c, which indicates that a rapidly advancing liquid film is formed on the fiber surface when the partial-to-complete wetting transition takes place. Our experiment thus establishes a quantitative method by which many other liquid interfaces coated with polymers, surfactants, and biomolecules (such as proteins and lipids) may be characterized dynamically.

Surfactant Driven Marangoni Spreading in the Presence of Predeposited Insoluble Surfactant Monolayers.

When an insoluble surfactant is deposited on the surface of a thin fluid film, stresses induced by surface tension gradients drive Marangoni spreading across the subphase surface. The presence of a predeposited layer of an insoluble surfactant alters that spreading. In this study, the fluid film was aqueous, the predeposited insoluble surfactant was dipalmitoylphosphatidylcholine (DPPC), and the deposited insoluble surfactant was oleic acid. An optical density-based method was used to measure subphase surface distortion, called the Marangoni ridge, associated with propagation of the spreading front. The movement of the Marangoni ridge was correlated with movement of surface tracer particles that indicated both the boundary between the two surfactant layers and the surface fluid velocities. As the deposited oleic acid monolayer spread, it compressed the predeposited DPPC monolayer. During spreading, the surface tension gradient extended into the predeposited monolayer, which was compressed nonuniformly, from the deposited monolayer. The spreading was so rapid that the compressed predeposited surfactant could not have been in quasi-equilibrium states during the spreading. As the initial concentrations of the predeposited surfactant were increased, the shape of the Marangoni ridge deformed. When the initial concentration of the predeposited surfactant reached about 70 A2/molecule, there was no longer a Marangoni ridge but rather a broadly distributed excess of fluid above the initial fluid height. The nonuniform compression of the annulus of the predeposited monolayer also caused tangential motion ahead of both the Marangoni ridge and the boundary between the two monolayers. Spreading ceased when the two monolayers reached the same final surface tension. The final area per molecule of the DPPC monolayer matched that expected from the equilibrium DPPC isotherm at the same final surface tension. Thus, at the end of spreading, there was a simple surface tension balance between the two distinct monolayers.

First quantitative assessment of the adsorption of a fluorocarbon gas on phospholipid monolayers at the air/water interface

HypothesisFluorocarbon gases introduced above monolayers of phospholipids at the air/water interface were recently found to promote the adsorption of diverse molecular compounds, with potential application in drug-loaded microbubble design. Quantitative determination of the fluorocarbon present in the monolayers is strongly needed for the development of such applications. We hypothesized that neutron reflectometry (NR) and ellipsometry experiments would allow quantification of the fluorocarbon trapped in the monolayers. ExperimentsWe report the first quantitative determination of the extents of adsorption of perfluorohexane (F-hexane) on different phospholipid monolayers with respect to both their phase and isotopic form. To this aim, we applied an approach based on co-modeling the data obtained from NR and ellipsometry. FindingsWe found that F-hexane adsorbs strongly in monolayers of dipalmitoylphosphatidylcholine (DPPC) when they are both in the liquid expanded (LE) and liquid condensed (LC) phases, but to different extents according to the isotopic form of the phospholipid. Kinetic resolution of the interfacial composition from data on both isotopic contrasts (assuming chemical identicality) was therefore not possible using NR alone, so an alternative NR/ellipsometry co-modeling treatment was applied to data from each isotopic contrast. F-hexane adsorbs more abundantly on monolayers of hydrogenous DPPC than chain-deuterated DPPC when they are in the LE phase, whilst the opposite was observed when they monolayers are in the LC phase. The extents of adsorption of F-hexane in monolayers of dimyristoylphosphatidylcholine (DMPC, LE phase) and distearoylphosphatidylcholine (DSPC, LC phase) concurs with the strong dependence of those with phospholipids of different isotopic contrasts according to the monolayer phase. This new methodology can lead to advances in the novel characterization of fluorocarbons interacting with phospholipid monolayers of relevance to applications such as in the shells of fluorocarbon-stabilized medically-oriented microbubbles.

Synthesis and physicochemical evaluation of fluorinated lipopeptide precursors of ligands for microbubble targeting.

Ligand-targeted microbubbles are focusing interest for molecular imaging and delivery of chemotherapeutics. Lipid–peptide conjugates (lipopeptides) that feature alternating serine–glycine (SG)n segments rather than classical poly(oxyethylene) linkers between the lipid polar head and a targeting ligand were proposed for the liposome-mediated, selective delivery of anticancer drugs. Here, we report the synthesis of perfluoroalkylated lipopeptides (F-lipopeptides) bearing two hydrophobic chains (CnF2n+1, n = 6, 7, 8, 1–3) grafted through a lysine moiety on a hydrophilic chain composed of a lysine–serine–serine (KSS) sequence followed by 5 SG sequences. These F-lipopeptides are precursors of targeting lipopeptide conjugates. A hydrocarbon counterpart with a C10H21 chain (4) was synthesized for comparison. The capacity for the F-lipopeptides to spontaneously adsorb at the air/water interface and form monolayers when combined with dipalmitoylphosphatidylcholine (DPPC) was investigated. The F-lipopeptides 1–3 demonstrated a markedly enhanced tendency to form monolayers at the air/water interface, with equilibrium surface pressures reaching ≈7–10 mN m−1 versus less than 1 mN m−1 only for their hydrocarbon analog 4. The F-lipopeptides penetrate in the DPPC monolayers in both liquid expanded (LE) and liquid condensed (LC) phases without interfacial film destabilization. By contrast, 4 provokes delipidation of the interfacial film. The incorporation of the F-lipopeptides 1–3 in microbubbles with a shell of DPPC and dipalmitoylphosphatidylethanolamine-PEG2000 decreased their mean diameter and increased their stability, the best results being obtained for the C8F17-bearing lipopeptide 3. By contrast, the hydrocarbon lipopeptide led to microbubbles with a larger mean diameter and a significantly lower stability.

The interaction of chondroitin sulfate with a lipid monolayer observed by using nonlinear vibrational spectroscopy.

The first vibrational sum-frequency generation (VSFG) spectra of chondroitin sulfate (CS) interacting with dipalmitoyl phosphatidylcholine (DPPC) at air–liquid interface are reported here, collected at a laser repetition rate of 100 kHz. By studying the VSFG spectra in the regions of 1050–1450 cm−1, 2750–3180 cm−1, and 3200–3825 cm−1, it was concluded that in the presence of Ca2+ ions, the head groups together with the head-group-bound water molecules in the DPPC monolayer are strongly influenced by the interaction with CS, while the organization of the phospholipid tails remains mostly unchanged. The interactions were observed at a CS concentration below 200 nM, which exemplifies the potential of VSFG in studying biomolecular interactions at low physiological concentrations. The VSFG spectra recorded in the O–H stretching region at chiral polarization combination imply that CS molecules are organized into ordered macromolecular superstructures with a chiral secondary structure.
