Physicochemical and Morphological Properties of Litchi Seed Starch Oxidized by Different Levels of Sodium Hypochlorite

AbstractNon‐conventional starch sources have emerged as an interest due to their inherent physicochemical properties similar to conventional starches. This study aimed to enhance the value of non‐conventional litchi seed starch by employing anoxidizing process using different concentrations of sodium hypochlorite (NaOCl)(0.5, 1.5, 2.5, 3.5, 4.5%). The carbonyl and carboxyl content of oxidized starch was in the range of (0.019 – 0.323%) and (0.044 – 0.425%) showing an increase with the increasing concentration of NaOCl. The native and oxidized starches were further characterized for their functional and structural properties. The amylose content of native starch was 21.52% which was reduced to (19.34 – 13.43%) upon oxidation. The water and oil absorption capacities increased with theoxidation level. The swelling power(g/g) and solubility (g/100g) for native starch were 16.17 and 16.20 and 9.5 and 31.40 for modified starch. The oxidized starch produced clearer pastes. Scanning electron microscopy revealed surface erosion and cavities in oxidized starch granulesat higher concentrations. The oxidized starch showed higher relative crystallinity. FTIR was further used to assess structural changes in starches. Therefore, it can be concluded that oxidation significantly altered the characteristics of litchi seed starch, expanding its potential applications in various industries.

Antimicrobial Efficacy of Different Concentrations of Sodium Hypochlorite in the Elimination of Enterococcus Faecalis: An In-vitro Study

Introduction: Primary teeth are equally important as permanent teeth. Endodontic therapy aids in the preservation and maintenance of extremely carious primary teeth. For root canal therapy to be successful, root canals must be properly prepared and effectively irrigated. Sodium Hypochlorite (NaOCl) has been the gold standard for root canal treatment of primary teeth. However, there are a few drawbacks to using higher concentrations of sodium hypochlorite, such as toxic reactions from its apical penetration. In addition, an excess of irrigation fluid via the apical region could harm the permanent tooth underneath. Therefore, it is crucial to select the proper concentration to preserve the delicate balance between effectiveness and safety. Aim: The present in-vitro study was conducted in the city of Chennai, Tamil Nadu, between March and June 2022. The study aimed to evaluate the effectiveness of lower concentrations of sodium hypochlorite, such as 1%, 0.5%, and 0.25%, against Enterococcus faecalis. Materials and Methods: The antibacterial activity of different concentrations of sodium hypochlorite (1%, 0.5%, and 0.25%) against Enterococcus faecalis was performed using Muller Hinton agar. Three Mueller Hinton agar plates were prepared and sterilised for 15 minutes for each concentration. The volumes used were 25 μL, 50 μL, and 100 μL, respectively. E. faecalis was swabbed and poured into the sterilised plates. The plates were incubated for 24 hours at 37°C. After incubation, the plates were examined, and their zone of Inhibition was determined. ANOVA test was applied with Post-hoc Tukey’s analysis. Results: One-way ANOVA test showed that there was a significant difference with a p-value <0.001 in all three groups. Post-hoc Tukey’s analysis was done, and the test showed that the highest zone of inhibition was seen with 1% sodium hypochlorite at 100 μL, with zone diameter of 39 mm, and the least zone of inhibition was seen with 0.25% sodium hypochlorite solution at 25 μL, with zone diamter of 22 mm. Conclusion: The results of the present study show that a 1% sodium hypochlorite solution can be used to successfully disinfect root canals, indicating its potential as an effective antimicrobial agent in endodontic procedures.

Effect of sodium/calcium hypochlorite on adhesion and adaptation of fiber posts luted with a dual resin cement.

This study aimed to evaluate the effect of different concentrations of sodium hypochlorite (NaOCl) and calcium hypochlorite [Ca(OCl)2] on the bond strength and adaptation of glass fiber posts luted with a dual-curing resin cement. Fifty decoronated premolars were sectioned 14 mm from the apex and endodontically treated. The root canal filling was partially removed. The specimens were divided into 5 groups (n=10) according to the irrigant for post space irrigation: 0.9% sodium chloride (NaCl), (control); 2.5% NaOCl; 5.25% NaOCl; 2.5% Ca(OCl)2; and 5.25% Ca(OCl)2. For each group, irrigation was performed with 5 ml of solution. Afterward, the posts were luted with a dual-curing resin cement. One slice from each third was obtained and submitted to the push-out test and failure modes analysis. An additional slice from the middle third was submitted to confocal images for analysis of adaptation failures (gaps). Two-way ANOVA, Tukey's post-hoc, Kruskal-Wallis with Bonferroni adjusted, and chi-square tests, analyzed data. The group treated with 5.25% NaOCl showed lower bond strength values and generated more cohesive failures compared to the control (p < 0.05). Bond strength decreased from coronal to apical in the post space (p < 0.001). The groups treated with NaOCl had the highest percentages of gaps compared to the control (p < 0.05). Regardless of concentration, Ca(OCl)2 did not influence the bond strength and the occurrence of gaps (P > 0.05). Ca(OCl)2 is a good option for irrigating the post space before luting a fiber post with a dual-curing resin cement.

Physicochemical properties of potato starches oxidised by different concentrations of sodium hypochlorite, and their application in yogurt

Potato starch was modified by oxidation with different concentrations of active chlorine (0.5, 1.0, 2.0, 3.0, and 4.0%); and the physicochemical properties of the oxidised potato starch samples and the use of these starches in set yogurt were investigated as a function of their degree of oxidation. The carboxyl content, carbonyl content, and solubility of the oxidised starches increased with increasing sodium hypochlorite concentrations, while the swelling power decreased. Scanning electron microscopy showed that oxidation caused cracks or pores on the surfaces of the starch granules, and these morphological changes were enhanced by increased oxidant concentrations. Analysis with a Rapid Visco Analyser showed that high degrees of oxidation could improve the starch thermodynamic stability and resistance to retrogradation. Set yogurts made with oxidised potato starches had higher water-holding capacities, higher apparent viscosities, and better sensory qualities than those prepared with native starch.

Laser activated irrigation with SWEEPS modality reduces concentration of sodium hypochlorite in root canal irrigation

IntroductionShockwave enhanced emission photoacoustic streaming (SWEEPS) of Er: YAG laser has been proposed to improve the disinfecting efficacy of endodontic treatments. The purpose of this in vitro study was to compare the effects of different concentrations of sodium hypochlorite (NaOCl) combined with SWEEPS root canal irrigation on the removal of Enterococcus faecalis (E.faecalis) in infected bovine root canals. Materials and methodsForty-eight bovine root canals infected with E.faecalis were randomly divided into six groups. The root canals were irrigated by normal saline (NS) in combination with conventional needle irrigation (CI) , NS and different concentrations of NaOCl (0.5%, 1.0%, 2.0%, and 5.25%) in combination with SWEEPS. Then, the remaining live microbes from all specimens before and after root canal irrigation were collected and recovered via plate counting of the colony-forming unit (CFU). The CFU data were statistically analyzed by the Kruskal-Wallis H test. ResultsAfter root canal irrigation, the viable count in each group was significantly reduced (P < 0.05). The bacterial reduction in NS+CI and NS+SWEEPS groups was significantly lower than that in other groups (P < 0.05), and there was no significant difference among the other groups, or between these two groups (P > 0.05). ConclusionsThis study showed that SWEEPS used for root canal irrigation can enhance the effect of low-concentration NaOCl while ensuring the antimicrobial effect.

Effects of different concentrations of sodium hypochlorite on dentine adhesion and the recovery application of sodium erythorbate.

Root canal therapy is the most effective and common method for pulpitis and periapical periodontitis. During the root canal preparation, chemical irrigation plays a key role. However, sodium hypochlorite (NaOCl), the widely used irrigation fluid, may impact the bonding strength between dentin and restorative material meanwhile sterilization and dissolving. Therefore, it's important to explore the influence of NaOCl on the adhesion between dentin and restoration materials to ensure clinical efficacy. This study aims to explore the effect of NaOCl on dentine adhesion and evaluate the effect of dentine adhesion induced by sodium erythorbate (ERY), and to provide clinical guidance on dentin bonding after root canal therapy. Seventy freshly complete extracted human third molars aged 18-33 years old, without caries and restorations were selected. A diamond saw was used under running water to achieve dentine fragments which were divided into 10 groups with 14 fragments in each group: 2 control [deionized water (DW)±10% ERY] and 8 experimental groups (0.5%, 1%, 2.5%, and 5.25% NaOCl±10% ERY). The dentine specimens in the control group (treated with DW) and the experimental groups (treated with 0.5% NaOCl, 1% NaOCl, 2.5% NaOCl, and 5.25% NaOCl) were immersed for 20 min using corresponding solutions which were renewed every 5 min. The other 5 groups were immersed in 10% ERY for 5 min after an initial washing with DW for 1 min. Then, we selected 4 dentine fragments from all 14 fragments in each group and the numbers and diameters of opening dentinal tubules were observed under scanning electron microscope (SEM). The other 10 dentine fragments from each group were used to make adhesive samples by using self-etch adhesive wand composite resin. All the above adhesive samples were sectioned perpendicular to the bonded interface into 20 slabs with a cross-sectional area of 1 mm×1 mm using a diamond saw under the cooling water, and then the morphology of 10 slabs in each group's bonding interface was observed from aspects of formation of resin tags, depth of tags in dentin, and formation of hybrid layer under SEM. The other 10 slabs of each group's microtensile bond strength and failure modes were also analyzed. Among the 0.5% NaOCl, 1% NaOCl, 2.5% NaOCl, and 5.25% NaOCl groups, the number and diameter of patent dentinal tubules gradually increased with the rise of concentration of NaOCl solution (all P<0.05). Among the DW, 0.5% NaOCl, 1% NaOCl, 2.5% NaOCl, and 5.25% NaOCl groups, the number and diameter of patent dentinal tubules increased after using ERY, but without significant difference (all P>0.05). Among the DW, 0.5% NaOCl, 1% NaOCl, and 2.5% NaOCl groups, the scores of formation of resin tags under SEM gradually increased with the increase of concentration of NaOCl solution, while the score in the 5.25% NaOCl group decreased significantly compared with the score of the 2.5% NaOCl group (P<0.05). There was no significant difference between using 10% ERY groups and without using 10% ERY groups (all P>0.05). The scores of length of the tags under SEM in the 5.25% NaOCl group was significantly higher than the scores of DW, 0.5% NaOCl, and 1% NaOCl groups (all P<0.05), and it was also higher than the score of the 2.5% NaOCl group, but without significant difference (P>0.05). There was no significant difference between using 10% ERY groups and without using 10% ERY groups (P>0.05). The scores of formation of hybrid layer under SEM in the 2.5% NaOCl and 5.25% NaOCl groups significantly decreased compared with the score of the DW group (all P<0.05). There were significant differences between the 2.5% NaOCl±10% ERY groups and between the 5.25% NaOCl±10% ERY groups (all P<0.05). Microtensile bond strength was greater in the 0.5% NaOCl, 1% NaOCl, and 2.5% NaOCl groups, but lower in the 5.25% NaOCl group than that in the DW group (all P<0.05). There were significant differences between the 2.5% NaOCl±10% ERY groups and between the 5.25% NaOCl±10% ERY groups (all P<0.05). The incidence of type "Adhesive" of failure modes in the 5.25% NaOCl group was significantly higher than that in other groups (all P<0.05), while the incidence of type "Adhesive" in the 5.25% NaOCl+10% ERY group was lower than that in the 5.25% NaOCl group (P<0.05). The bonding strength to dentine increases with the increase of NaOCl concentration when the concentration lower than 2.5%; whereas it is decreased at a higher concentration (such as 5.25%). 10% ERY has a definite recovery effect on attenuated bonding strength to 5.25% NaOCl-treated dentine.

Effect of Sodium Hypochlorite Concentration on Electronic Apex Locator Reliability.

This ex vivo study aimed to measure the performance of an electronic apex locator (EAL) in the presence of sodium hypochlorite irrigants with different concentrations. Two EALs (Root ZX Mini and Locapex 6) were used to locate the apical foramen in 10 extracted single-rooted teeth in the presence of 0.5%, 2.5% and 5% sodium hypochlorite. Visual working lengths were also determined using #10 K-file under a microscope before the electronic measurements were made. The performance of both EALs was compared for the electronic working lengths determined under the different concentrations of sodium hypochlorite. A multiple-way ANOVA and PLSD Fisher’s test with an α risk fixed at 5% were conducted. There were no statistical differences in the working lengths determined by both EALs between the three groups with different concentrations of sodium hypochlorite and their visual control measurements. When a ± 0.5 mm margin was applied, the Root ZX Mini and the Locapex 6 presented 88% and 83% accuracy, respectively. Sodium hypochlorite concentration in irrigants does not affect the accuracy and reliability of either the Root ZX Mini or the Locapex 6. Electronic apex locators are reliable with any concentration of sodium hypochlorite irrigants.

Micropropagation of Feverfew (Tanacetum parthenium) and Quantification of Parthenolide Content in Its Micropropagated and Conventionally Grown Plants

Feverfew (Tanacetum parthenium) is a well-known multi-functional plant with anti-inflammatory, cardiotonic, antiangiogenic, and anticancer effects. The therapeutic value of this plant is due to its phytochemical constitutes, especially parthenolide. Tissue culture techniques have been applied to improve the bioactive components of many herbal plants. Hence, this study, was carried out to establish a protocol for micropropagation of the feverfew plant and to quantify parthenolide content in its micropropagated and conventionally grown plants. To establish an aseptic culture, different concentrations of sodium hypochlorite (NaOCl) were investigated for seed surface sterilization. Besides, the effects of plant growth regulators (PGRs) on the callus induction, shoot organogenesis from callus and in vitro rooting were evaluated. Additionally, the parthenolide yield of the micropropagated and conventionally grown plants was determined by using high-performance liquid chromatography (HPLC). The results showed that surface sterilization of feverfew seeds with 6% NaOCl for 15 min obtained 65.00 ± 2.69% aseptic seeds. Murashige and Skoog (MS) medium supplemented with 0.4 mg/L thidiazuron (TDZ) and 2 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) resulted in 86.00 ± 1.72% callus induction. The highest number of shoots (5.00 ± 0.15) per explant was obtained in the treatment of MS medium supplemented with 5 mg/L zeatin. MS medium fortified with 3 mg/L indole-3-butyric acid (IBA) produced the maximum number of roots per plantlet (8.90 ± 0.35). A total of 90% of the micropropagated plantlets survived when planted in perlite + peat moss (1:1 v/v); the micropropagated plantlets were successfully established in the ex vitro conditions. According to parthenolide analysis, its level was significantly higher in the micropropagated plants than conventionally grown plants. Among different solvents, ethanolic extraction obtained the highest parthenolide content of the feverfew plant. Hence, it can be concluded that micropropagation of feverfew could be applied to produce disease-free planting materials and to improve the parthenolide content of the feverfew plant.

Effect of Different Concentrations of Sodium Hypochlorite on the Compressive Strength of Endodontically Treated Roots

IntroductionThe aim was to compare the compressive strength of dentin after irrigation with different concentrations of sodium hypochlorite (NaOCl). MethodsA total of 259 extracted human tooth roots with no root caries or previous endodontic treatment were collected. They were sorted by size and length, and assigned to 7 groups (n = 37): negative control (no instrumentation, no irrigant), positive control (saline irrigant), 1%, 2.5%, 4%, 5.25%, and 10% NaOCl. A total of 13 mL NaOCl or saline (in 1-mL increments) was used per canal while being instrumented to #35 files (approximately 13 minutes). EDTAC (15%) was then used to remove the smear layer (2 minutes), followed by NaOCl as a final rinse. The roots were sectioned into 2-mm-thick disks and subjected to compression testing. ResultsThere was a significant difference between the negative control group and all instrumented groups (P < .05). The positive control group was significantly different from the 5.25% group (P < .05) and had a low P value when compared with the 2.5% group and the other higher concentrations (P ≅ .1). When the NaOCl groups were compared with each other, there was a significant difference between 1% NaOCl and all higher concentrations (P < .05), but no significant difference when 2.5% NaOCl was compared with higher concentrations. ConclusionA concentration of 1% NaOCl had the least effect on the compressive strength of dentin and this was significantly different from all higher concentrations tested. Therefore, the use of concentrations of NaOCl above 1% should be reconsidered.

Purpose. Development of a method for the regeneration of highly porous fillers for biofilters after slagging of their pores with biofilm residues in the process of growing aquaculture objects in recirculating aquaculture systems (RAS). Methodology. Regeneration of fillers was performed with solutions of sodium hypochlorite of different concentrations (2.5%, 1%, 0.5% of active chlorine) after their exploitation for 6 months. The cleaning efficiency was monitored by the difference in fillers weights after cleaning. Findings. According to the study, the use of sodium hypochlorite for cleaning highly porous fillers for biofilters (using an example of porous ceramic filler and foamed glass «JBL Micromec») is efficient and cost-effective. A reduction in the weight of the test samples averaged 12.95% for porous ceramics and 15.01% for foamed glass. Based on the obtained data, aqueous solutions with different concentrations of sodium hypochlorite do not have a pronounced difference in efficiency, respectively; the optimal for use is a 0.5% solution. Originality. The use of highly porous fillers for biofilters has long been unprofitable in fish farming due to the rapid loss of their main advantages - a high specific area per unit volume as a result of micropore slagging with biofilm residues. As a result, complete regeneration of these filter materials was required annually, which, given the significantly higher cost compared to polymeric fillers, made their use unprofitable. The described technique allows the use of porous ceramics and foamed glass, after their restoration, for a long time, with replacement only after mechanical wear. Practical value. The described method allows using highly porous fillers for biofilters in the process of growing aquaculture objects without their main disadvantage - a rapid decrease in efficiency as a result of pore slagging with biofilm residues. Antimicrobial Effect of Acetic Acid, Sodium Hypochlorite, and Thermal Treatments against Psychrotolerant Bacillus cereus Group Isolated from Lettuce (Lactuca sativa L.).

Various food products distributed throughout the cold chain can present a health risk for consumers due to the presence of psychrotolerant B. cereus group species that possess enterotoxin genes and antibiotic resistance. As these bacteria can grow at the low temperatures used in the food industry, this study evaluated the antimicrobial efficacy of acetic acid, sodium hypochlorite, and thermal treatments for inhibition of psychrotolerant strains and the effect that differences in activation temperature (30 °C and 10 °C) have on their efficacy. The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and bacterial growth assay of acetic acid and thermal treatment showed an equal or higher antimicrobial efficacy in isolates activated at 10 °C than in those activated at 30 °C. In particular, psychrotolerant strains from the B. cereus group were completely eliminated with 0.25% acetic acid, regardless of the activation temperature. The possibility of tolerance was determined by observing responses in cells activated at 10 and 30 °C when exposed to different concentrations of sodium hypochlorite. Five isolates activated at 10 °C exhibited enhanced survivability in sodium hypochlorite compared to isolates activated at 30 °C, and these isolates were able to grow in sodium hypochlorite at concentrations of 250 ppm or higher. Although a significant difference in antimicrobial efficacy was observed for psychrotolerant B. cereus group strains depending on the activation temperature, acetic acid may be the most effective antimicrobial agent against psychrotolerant B. cereus species isolated from food products distributed in a cold chain.

Implantation of multiscale silk fibers on poly (lactic acid) fibrous membrane for biomedical applications

Despite that poly (lactic acid) (PLA) has satisfactory biodegradation in vivo, its application in biomedicine is restricted due to its unsatisfactory cytocompatibility. Silk fiber (SF) has outstanding biocompatibility and silk fibroin protein obtained from silk by degumming has good hydrophilicity. Therefore, combining the PLA and silk can improve hydrophilicity of PLA to apply as biomedical materials. In this study, different concentrations of sodium hypochlorite (NaClO) were used to separate the silk to obtain multiscale silk fibers (MSFs), which were implanted into the PLA electrospun fibrous membranes (EFMs). The morphology and structure of silk fibers separated by different concentrations of NaClO were studied by Zetasizer Nano ZS, UV spectroscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and scanning electron microscopy. Moreover, the biocompatibility of the surface-modified PLA composite membranes by MSFs was investigated by cell cultivation and proliferation. The results showed that the surface-modified PLA EFMs through MSF bundles obtained from NaClO split silk exhibited a certain improvement on PLA hydrophilicity and enhancement on cellular compatibility, which could have a broad prospect in the practical application of biomedical materials.

Effectiveness of photon-initiated photoacoustic streaming in root canal models with different diameters or tapers

BackgroundThis study aimed to compare the use of photon-initiated photoacoustic streaming (PIPS) and conventional needle irrigation (CNI) in conjunction with different concentrations of sodium hypochlorite (NaOCl) to remove Enterococcus faecalis (E. faecalis) suspended bacteria and biofilms from root canal systems with different diameters or tapers.MethodsArtificial root canal samples (n = 480) were randomly divided into three groups (n = 160/group). The canals were prepared to fit file sizes #10/.02, #25/.02, or #25/.06. The size #10/.02 group was incubated for seven days. The size #25/.02 or #25/.06 group was incubated for 2 days. A stable biological model of E. faecalis infection was established. The root canals were washed with distilled water or with 1%, 2%, or 5.25% NaOCl combined with CNI or PIPS. Bacterial suspensions and biofilms were assessed using an ATP assay kit and fluorescence microscopy. Image-Pro Plus was used to analyse the average fluorescence intensity to determine the most suitable root canal irrigation solution.ResultsIn the CNI and PIPS groups, the ATP value of the 5.25% NaOCl subgroup was the lowest, followed by that of the 2% and 1% NaOCl subgroups. The ATP value of the distilled water subgroup was the highest (P < 0.05). When the root canal taper was 0.02, the ATP value of the #10/.02 + PIPS group was significantly lower than that of the #25/.02 + CNI group (P < 0.05). The average fluorescence intensity of the #10/.02 + PIPS group was lower than that of the #25/.02 + CNI group (P < 0.05). When the apical diameter was #25, the ATP value of the 0.02 taper in the PIPS group was lower than that of the 0.06 taper in the CNI group (P < 0.05), and the average fluorescence intensity of the 0.02 taper + PIPS group was lower than that of the 0.06 taper + CNI group (P < 0.05). PIPS combined with 2% and 5.25% NaOCl effectively improved the long-term antibacterial effect after irrigation and re-culture for 6 h.ConclusionsCompared with CNI, PIPS has greater ability to remove bacteria in root canals with a small preparation diameter and a small taper. PIPS with 2% and 5.25% NaOCl exhibited superior antibacterial and bacteriostatic effects.

Fluorescence characteristics of E. faecalis in dentine following treatment with oxidizing endodontic irrigants

IntroductionThis study aimed to assess changes in the fluorescence characteristics of Enterococcus faecalis in human dentine over a period of 24 h following treatment with endodontic irrigants. MethodSterilised, non-functional extracted third molars were embedded in acrylic resin and uniformly sectioned into 2 mm thick dentine sections. After the removal of smear layer, the dentine sections were inoculated with E. faecalis and cultured for 7 days. The infected dentine sections were subsequently treated with different concentrations of sodium hypochlorite (NaOCl) and hydrogen peroxide (H2O2). Bacterial fluorescence readings were assessed at different time points using a calibrated laser device. All data were assessed for normality (Kolmogorov Smirnoff test) and analysed using ANOVA with Bonferroni post-hoc tests. ResultsFluorescence readings were quenched when E. faecalis infected human dentine sections were treated with oxidizing irrigants in vitro. Throughout a 24-hour period, fluorescence recovered in part but did not return to baseline level. ConclusionThe fluorescence quenching effect of these oxidizing agents needs to be considered when using laser fluorescence in assessing the quality of root canal debridement or disinfection.

Dentro de la familia de las Musaceas, el género Musa ha adquirido importancia en la alimentacion a nivel mundial, ya sea por el consumo directo (fruta) o indirecto (previa cocción) como es en caso de Bolivia, sin embargo por cambio de actividades agroeconómicas este rubro esta siendo afectado, y tambien carecen de un mecanismo de regeneracion de los plantines libre de enfermedades. El presente estudio tuvo como objetivo estandarizar la desinfección de hijuelos de Gran enano (banano) y Harton (plátano) con diferentes concentraciones de hipoclorito de sodio (NaClO) e inducir la multiplicación con la aplicación de bencilaminopurina. Los hijuelos de ambas variedades fueron sometidas a dos tiempos de desinfección, el primero consistió en seleccionar los hijuelos de 10 a 12 cm de longitud y sumergirlas en hipocloriro de sodio al 8 % durante 12 min y posteriormente, se dividio en dos grupos para la segunda desinfeccion de 2 y 4 % con hipoclorito de sodio durante 7 min, luego fueron enjuagadas y sembradas en el medio Murashige y Skoog (MS) con 2 mg L-1 BAP (bencilaminopurina) + 1 mg L-1 ANA (ácido naftalenacético). En la fase de multiplicación los brotes se sembraron en medios de cultivo (MC1 = MS + 2.5 mg L-1 BAP + 1 mg L-1 ANA) y (MC2 = MS + 5 mg L-1 BAP + 1 mg L-1 ANA). Se determinó que la combinacion de una primera y segunda desinfección 8 y 4 % de NaClO desinfectó un 80 % en Gran enano y 40 % en Hartón respectivamente. En la fase de multiplicación se determinó que el medio de cultivo MS suplementado con 5 mg L-1 de BAP favoreció en la formación de 8 brotes en banano en ambas variedades.

Controlling the source of green tides in the Yellow Sea: NaClO treatment of Ulva attached on Pyropia aquaculture rafts

Green macroalgal blooms have occurred in the Yellow Sea for 13 consecutive years since 2007. However, effective control methods have not yet been successfully developed. Ulva prolifera attached on Pyropia aquaculture rafts was identified as the main source of the blooms, thus, eliminating the attached Ulva from the rafts should be the most efficient method to prevent and control these blooms. In this study, we designed a new molecular marker (rps2-trnL) to identify U. prolifera collected from the aquaculture site. Samples identified as U. prolifera were cultured, and different concentrations of sodium hypochlorite (NaClO) solution (0.2, 0.4, 0.6, 0.8, and 1.0%) were used to kill the alga. The measured chlorophyll content, light quantum yield, and photosynthetic oxygen release rate data from the laboratory experiment suggested that the appropriate concentration of NaClO solution should be 0.6–1.0%. To carry out a small-scale field trial, we developed a device that can spray NaClO on Ulva blades and recycle the residual liquid. Results showed that only the 1.0% NaClO solution could quickly kill the Ulva in outdoor experiments. During the treatment, very little NaClO dripped on the tidal flat, which indicated that the recycling device avoided a potential negative impact of residual solution on the marine ecosystem. By the beginning of November 2019, the Pyropia aquaculture area treated by NaClO was about 13,000 ha, covering about one-third of the total area at the coast of Jiangsu Province. Monitoring by an unmanned aerial vehicle showed that very little attached Ulva was present on the ropes after NaClO treatment, which indicated that it was an effective method to control the green tide. Also, the range of residual active chlorine within seawater during the treatment was only from 0.003 to 0.012 mg/L, which complied with Chinese national standards. However, further monitoring of the formation process of green tides should be conducted in future years to evaluate the effect of NaClO treatment.

Dynamics of morphological and biochemical parameters in the blood of white mice under the action of the drug “Vitosept”

The paper presents the results of the study of the drug toxicity “Vitosept” in its preclinical studies in intact white mice. It was found that according to the assessment of toxicological parameters of the studied biocidal product is low-toxic. Oral administration of even its highest concentrations (500 mg/l) did not show a pronounced toxic effect. During the action of the drug for 20 days no deviations in the clinical condition, behavioral reactions, reflex activity did not occur. It was found that most of the values that characterize the functional state of the hematopoietic and hepatobiliary systems, against the background of different concentrations of sodium hypochlorite, did not undergo significant changes compared with the control. The hemoglobin content in the blood of white mice in the experimental groups increased compared with the control in the first three experimental groups (concentration of GHN 100–300 mg/l). With increasing concentration of the drug, when administered intraperitoneally, there was a greater number of leukocytes in the blood of laboratory animals. At the same time, a probable increase in this indicator was found only in mice of the II experimental group when administered orally to animals of the lowest test concentration (100 mg/l). The concentration of total protein, urea and glucose in the blood of white mice under the action of “Vitosept” was similar in animals of the control and experimental groups. The revealed tendency to increase the activity of certain enzymes (LF, ALT, AST) was short-lived, and is obviously a compensatory response of animals to the xenobiotic effect of the drug. On the 20th day of the experiment, the studied white mice of the experimental groups were close to those in the control group.

Use and Efficiency of Disinfectants within a Hydrocooler System for Postharvest Disease Control in Sweet Cherry

ABSTRACT Commercial disinfectants were used within a hydrocooler to prevent postharvest diseases of sweet cherries and to extend the storage and shelflife durations. Different concentrations of sodium hypochlorite, peracetic acid, hydrogen peroxide and Citrox® were used in in vitro experiments whereby the toxicity of disinfectants on B. cinerea and P. expansum spore germination and germ tube elongation was investigated. Citrox® was the only disinfectant containing organic acids and flavonoids. The in vitro experiments demonstrated that disinfectants were effective in inhibiting spore germination and germ tube elongation. In in vivo experiments, fruits were inoculated with a suspension containing 1 × 106 cfu/mL B. cinerea and P. expansum spores and hydrocooled with appropriate disinfectant concentrations. Total microbial, fungal, and bacterial populations on fruit surfaces and in the water used for processing were reduced by all disinfectant applications. Fruits were packed in modified atmosphere packages and stored at 1°C for 30 days and for 4 additional days on a shelf at 20°C. During cold storage, fruits were evaluated in terms of phytotoxicity, and it was observed that Citrox®, hydrogen peroxide, and ozonated water applications cause phytotoxicity in the both the stem and fruit. In contrast, no phytotoxic reaction was observed in sodium hypochlorite and peracetic acid applications. Fruit decay did not occur during cold storage but was seen during the shelflife period. Fruit decay was significantly reduced by sodium hypochlorite and peracetic acid applications. Sodium hypochlorite and peracetic acid applications within a hydrocooler were found to be efficient in preventing postharvest diseases of sweet cherries.
