Characterization of bacterial communities in ticks parasitizing cattle in a touristic location in southwestern China

The purpose of this study was to investigate tick species around Mount Fanjing and analyze bacterial communities in two species – Rhipicephalus microplus and Haemaphysalis longicornis – parasitizing cattle in Tongren, Guizhou province, Southwest China, using high-throughput sequencing methods. In April 2019, ticks were collected from five sites in Jiangkou County, Yinjiang County, and Songtao County. In total, 296 ticks were collected, comprising two genera and three species: H. longicornis, Haemaphysalis flava, and R. microplus. Rhipicephalus microplus was the most representative species (57.4%) within the collected group, being the dominant species in Tongren City, followed by H. longicornis (39.5%) and H. flava (3.0%). Beta-diversity analysis revealed differences in bacterial community composition among the tick species. The bacterial community structure of R. microplus collected in the three counties was highly similar. Chlorella and Bacillus were highly abundant in H. longicornis. Rickettsia was detected at high relative abundance in R. microplus but in low relative abundance in H. longicornis, suggesting that Rickettsia is more associated with R. microplus than with H. longicornis. More in-depth investigations are needed to determine the pathogenic risk of Rickettsia and its relationship with the host. This is the first survey on tick-borne bacterial communities in this area, which is of great significance for the prevention and control of tick-borne diseases locally.

Limited impacts of high doses of dietary copper on the gut bacterial metal resistome explain negligible co-selection of antibiotic resistance

High dietary intake of Cu has previously been linked to the selection of Cu resistance and co-selection of antibiotic resistance in specific gut bacteria. Based on a novel HT-qPCR metal resistance gene chip as combined with 16S rRNA gene amplicon sequencing and phenotypic resistance typing of Escherichia coli isolates, we here report the impacts of two contrasting Cu-based feed additives on the swine gut bacterial metal resistome and community assembly. DNA was extracted from fecal samples (n = 80) collected at day 26 and 116 of the experiment from 200 pigs allotted to five dietary treatments: negative control (NC) diet with 20 μg CuSO4 g−1 and four diets added 125 or 250 μg CuSO4 g−1 feed or 125 or 250 μg Cu2O g−1 feed to the NC diet. Dietary Cu supplementation reduced the relative abundance of Lactobacillus, but it had negligible impacts on bacterial community composition relative to the gut microbiome maturation effect (time). The relative importance of different bacterial community assembly processes was not markedly affected by the dietary Cu treatments, and differences in swine gut metal resistome composition could be explained primarily by differences in bacterial community composition rather than by dietary Cu treatments. High dietary Cu intake (250 μg Cu g−1) selected for phenotypic Cu resistance in E. coli isolates, but surprisingly it did not result in increased prevalence of the Cu resistance genes targeted by the HT-qPCR chip. In conclusion, the lacking impacts of dietary Cu on the gut bacterial metal resistome explain results from a previous study showing that even high therapeutic doses of dietary Cu did not cause co-selection of antibiotic resistance genes and mobile genetic elements known to harbor these genes.

Antibiotic resistance gene abundance and bacterial community composition in macroinvertebrates of an urban stream

Abstract Increase in antibiotic resistance genes (ARGs) in freshwater ecosystems as a consequence of anthropogenic activities is a matter of public health concern. Although researchers have looked at ARG abundance and the factors affecting them in biofilms, water and sediment, the role of freshwater macroinvertebrate gut microbiomes in harbouring and disseminating ARGs is unknown. In the current study, we tested whether: (a) macroinvertebrate gut microbiomes serve as hotspots of ARGs in comparison to their immediate environment, (b) functional feeding groups (FFGs) impact ARG abundance in macroinvertebrate guts, (c) bacterial community composition and ARG abundance are correlated, and (d) macroinvertebrates can be potential vectors of ARGs. Macroinvertebrates, sediment and water samples were collected from seven different sites along northeastern Ohio (U.S.A.) streams and abundances of three ARGs (sulI, sulII and tetW) were determined. Bacterial community composition also was examined for respective microbiomes and a microcosm study was designed for testing hypothesis 4. In many cases, the gut microbiome had higher relative ARG abundance (sulI and sulII) at sites along Tinker's Creek compared to the surrounding substrate and water. ARG abundances along with taxonomic differences in bacterial community composition were different among the FFGs (ARG abundance higher in omnivores). Certain bacterial taxa were significantly correlated with one or more ARGs. We did not find that freshwater macroinvertebrates can serve as vectors of ARGs (from the microcosm study). Overall, our site‐based results suggest that freshwater macroinvertebrate guts may serve as reservoirs of ARGs, differences in ARG abundance among FFGs was possibly a result of differences in bacteria ingested and gut morphophysiological conditions. Because this study indicates that macroinvertebrate guts can be potential reservoirs of ARGs, future studies can be designed to understand the role of macroinvertebrates in dissemination of ARGs in freshwater ecosystems.

Spatial variation and relationship between soil dissolved organic matter and bacterial community in urban greenspaces

Large-scale investigations of soil dissolved organic matter (DOM) and the relationship between bacterial diversity and DOM with climatic gradients are important to reveal the influence of climatic factors on soil DOM. The objectives of this study were to (1) investigate the distribution pattern of DOM at a large scale, (2) investigate the effects of climate change on DOM components and microbial communities, and (3) investigate the relationship between microbial communities and DOM components. This study involved the collection of 54 urban greenspaces samples from five climate zones in mainland China and utilization of FT-ICR-MS and 16S rRNA sequencing techniques to reveal the relationship between microbial communities and DOM. Our results showed that the DOM in soil was DOM inated by lignins, followed by proteins, unsaturated hydrocarbons and lipids. Structural equation modeling (SEM) indicated that lignins and proteins shared a negative response pattern with mean annual precipitation (MAP), while lipids and tannins showed a positive correlation with MAP. The differences in bacterial community composition among different climatic zones were not significant, and the DOM inant bacteria in the soil were Proteobacteria, followed by Acidobacteria and Actinobacteria. Bacterial diversity was highest in the subtropical monsoon climate, lowest in the tropical monsoon climate. In addition, SEM indicated that there was a significant correlation between the composition and diversity of DOM and the diversity and abundance of bacteria. Our results provided a deeper understanding of the impact of climatic factors on DOM components and the relationship between bacterial diversity and DOM in urban greenspaces.Graphical

The gut microbiota diversity of five Orthoptera (Insecta, Polyneoptera) insects determined by DNA metabarcoding.

Most orthopteran insects are phytophagous and some are important pests in agriculture and forests. Many intestinal microflora of Orthoptera insects have been reported, primarily from Acridoidea and there have been few reports of other taxa. In this study, we collected 15 individuals representing five species (Ruspolialineosa, Tetrixjaponica, Erianthusversicolor, Gryllotalpaorientalis and Teleogryllusemma) belonging to five orthopteran superfamilies (Tettigonioidea, Tetrigoidea, Eumastacoidea, Gryllotalpoidea and Grylloidea) to characterise and compare the gut microbiota with greater taxonomic width by performing sequencing analysis of the 16S rRNA V4 region in gut material. A total of 606,053 high-quality sequences and 3,105 OTUs were acquired from 15 gut samples representing 24 phyla, 48 classes, 69 orders, 133 families and 219 genera. Firmicutes and bacteria were the most abundant phyla, followed by Bacteroidetes, Cyanobacteria, Actinobacteria and Acidobacteria. At the genus level, Serratia, Citrobacter, Wolbachia, Lactobacillus and Parabacteroides were the most predominant genera in R.lineosa, T.japonica, E.versicolor, G.orientalis and T.emma, respectively. Both Principal Coordinates Analysis (PCoA) and heatmap results revealed significant differences in bacterial community composition across species. Additionally, alpha diversity analysis indicated the bacterial richness was significantly different amongst the five species.

Effects of supplementation of Bacillus amyloliquefaciens on performance, systemic immunity, and intestinal microbiota of weaned pigs experimentally infected with a pathogenic enterotoxigenic E. coli F18.

The objective of this study was to investigate the effects of dietary supplementation of Bacillus (B.) amyloliquefaciens on growth performance, diarrhea, systemic immunity, and intestinal microbiota of weaned pigs experimentally infected with F18 enterotoxigenic Escherichia coli (ETEC). A total of 50 weaned pigs (7.41 ± 1.35 kg BW) were individually housed and randomly allotted to one of the following five treatments: sham control (CON-), sham B. amyloliquefaciens (BAM-), challenged control (CON+), challenged B. amyloliquefaciens (BAM+), and challenged carbadox (AGP+). The experiment lasted 28 days, with 7 days of adaptation and 21 days after the first ETEC inoculation. ETEC challenge reduced (P < 0.05) average daily gain (ADG) of pigs. Compared with CON+, AGP+ enhanced (P < 0.05) ADG, while B. amyloliquefaciens supplementation tended (P < 0.10) to increase ADG in pigs from days 0 to 21 post-inoculation (PI). The ETEC challenge increased (P < 0.05) white blood cell (WBC) count on days 7 and 21 PI, while BAM+ pigs tended (P < 0.10) to have low WBC on day 7 PI and had lower (P < 0.05) WBC on day 21 PI compared with CON+. In comparison to AGP+ fecal microbiota, BAM+ had a lower (P < 0.05) relative abundance of Lachnospiraceae on day 0 and Clostridiaceae on day 21 PI, but a higher (P < 0.05) relative abundance of Enterobacyeriaceae on day 0. In ileal digesta, the Shannon index was higher (P < 0.05) in BAM+ than in AGP+. Bray-Curtis PCoA displayed a difference in bacterial community composition in ileal digesta collected from sham pigs vs. ETEC-infected pigs on day 21 PI. Pigs in BAM+ had a greater (P < 0.05) relative abundance of Firmicutes, but a lower (P < 0.05) relative abundance of Actinomycetota and Bacteroidota in ileal digesta than pigs in AGP+. Ileal digesta from AGP+ had a greater (P < 0.05) abundance of Clostridium sensu stricto 1 but lower (P < 0.05) Bifidobacterium than pigs in BAM+. In conclusion, supplementation of B. amyloliquefaciens tended to increase ADG and had limited effects on the diarrhea of ETEC-infected pigs. However, pigs fed with B. amyloliquefaciens exhibit milder systemic inflammation than controls. B. amyloliquefaciens differently modified the intestinal microbiota of weaned pigs compared with carbadox.

Niche-mediated bacterial community composition in continental glacier alluvial valleys under cold and arid environments.

Bacteria are an essential component of glacier-fed ecosystems and play a dominant role in driving elemental cycling in the hydrosphere and pedosphere. However, studies of bacterial community composition mechanisms and their potential ecological functions from the alluvial valley of mountain glaciers are extremely scarce under cold and arid environments. Here, we analyzed the effects of major physicochemical parameters related to soil on the bacterial community compositions in an alluvial valley of the Laohugou Glacier No. 12 from the perspective of core, other, and unique taxa and explored their functional composition characteristics. The different characteristics of core, other, and unique taxa highlighted the conservation and difference in bacterial community composition. The bacterial community structure of the glacial alluvial valley was mainly affected by the above sea level, soil organic carbon, and water holding capacity. In addition, the most common and active carbon metabolic pathways and their spatial distribution patterns along the glacial alluvial valley were revealed by FAPTOTAX. Collectively, this study provides new insights into the comprehensive assessment of glacier-fed ecosystems in glacial meltwater ceasing or glacier disappearance.

Anthropogenic Nitrate Contamination Impacts Nitrous Oxide Emissions and Microbial Communities in the Marchica Lagoon (Morocco)

Lagoon systems are often confined, and their waters are poorly renewed, which makes them vulnerable to pollutants’ accumulation. Here, the impact of different sources of anthropogenic contamination (domestic, urban, industrial, and agricultural) on the nitrate (NO3−) content, emission of the greenhouse gas nitrous oxide (N2O), abundance of total bacterial archaeal, nitrifying, and denitrifying communities, and diversity and composition of bacterial communities in the sediments of the RAMSAR-protected Marchica lagoon (Nador, Morocco) was investigated. Six lake sites differing in NO3− concentration were selected. Wastewater coming from industrial activities results in the greatest concentration of NO3− in sediments and emissions of N2O. Increased carbon to nitrogen content in sites near domestic activities resulted in an increase in the abundance of total bacterial and archaeal communities, as well as nitrification and denitrification genes, but low N2O emissions due to a greater presence of microorganisms involved in N2O production over those able to reduce N2O. Significant differences in bacterial community composition between sites were observed, with the NO3− content being the main driver of these changes. Increased NO3− content in the sampling sites significantly reduced bacterial diversity. Bacterial genera involved in the degradation of organic and inorganic pollutants and nitrous oxide reduction, such as Robiginitalea, Symbiobacterium, Bacillus, Fusibacter, Neptunomonas, Colwellia, and Alteromonas, were the most abundant in the lagoon. The results suggest that the type of anthropogenic contamination directly impacts the nitrate content in the sediments of the Marchica lagoon, which determines variations in nitrous oxide emissions, nitrogen-cycling gene abundances, and bacterial diversity.

Bacterial Communities and Diversity of Western Ghats Soil: A Study of a Biodiversity Hotspot

The Western Ghats is one of India's mega-diversity hotspots and an ecologically and geologically important area for the diversity of endemic plants and animals. The present study provides insights into the aerobic bacterial diversity and composition of the soils of North Western Ghats located in Maharashtra state (NWGM), India. The samples for the culture-dependent study were collected from 6 different locations namely Malshej Ghat, Bhimashankar, Lonavala, Mulshi, Tail-Baila, and Mahabaleshwar. A total of 173 isolates were obtained from the different samples, which belonged to Proteobacteria (43%), Firmicutes (36%), and Actinobacteria (19%). Sequences of 15 strains shared ≤ 98.7% similarity (a species cut-off) which represent potential novel species. Metagenomic analysis revealed the presence of Actinobacteria and Proteobacteria as the most dominant phyla at both MB and MG. However, both sites showed variation in the composition of rare phyla and other dominant phyla. This difference in bacterial community composition could be due to differences in altitude or other physicochemical properties. The functional prediction from the amplicon sequencing showed the abundance of carbohydrate, protein, and lipid metabolism which was corroborated by screening the isolated bacterial strains for the same. The present study has a unique take on microbial diversity and defines the importance of community assembly processes such as drift, dispersal, and selection. Such processes are relatively important in controlling community diversity, distribution, as well as succession. This study has shown that the microbial community of NWGM is a rich source of polysaccharide degrading bacteria having biotechnological potential.

Differences in bacterial community composition, structure and function between sediments in waterways and non-navigable channels in a plain river network area.

Bacterial communities greatly help maintain the balance of river ecosystems and are highly sensitive to changes in environmental conditions. Plain river network areas (PRNs) are characterized by dense river networks, low-lying terrain, and slow water flow, where the bottom sediment is frequently disturbed by ship navigation due to the limited water depth and width of waterways, providing a unique ecological niche for bacterial growth. Hence, understanding how bacterial communities in PRNs respond to changes in hydrodynamic conditions, physicochemical parameters, and pollutants under ship navigation is essential to maintaining the stability of inland waterway ecosystems. The Taihu Lake Basin, a typical PRN, was selected to explore the differences in bacterial community composition, structure and function between sediments in waterways (WS) and non-navigable channels (NS). The results indicate that the sediment from NS possessed more diverse and complex bacterial communities than WS. NMDS and ANOSIM analyses further verified the significant differences in bacterial community structure between WS and NS. Combined with LEfSe, we observed the highly differential taxonomy between WS and NS from phylum to order. Moreover, a comparison of beta diversity dissimilarity indices revealed that although species replacement dominated both the WS and NS beta-diversity patterns, species loss caused the differences in the overall beta diversity between them. Variance partitioning analysis revealed that physicochemical parameters (clay content, pH, ORP, and others) and ship traffic volume (STV) were the main driving factors for bacterial community distribution between WS and NS, while pollutants (heavy metals, perfluoroalkyl acids, and others) had a relatively minor influence. PICRUSt2 analysis revealed that the changes in pH, ORP, and STV under ship navigation might inhibit the bacterial ability to metabolize carbohydrates. The results reveal the comprehensive effects of ship navigation disturbance on sediment bacterial communities in the PRN and contribute to further understanding of inland waterway ecosystems.

Spatiotemporal variations, assembly processes, and co-occurrence patterns of particle-attached and free-living bacteria in a large drinking water reservoir in China.

Particle-attached (PA) and free-living (FL) bacterial communities are sensitive to pollutant concentrations and play an essential role in biogeochemical processes and water quality maintenance in aquatic ecosystems. However, the spatiotemporal variations, assembly processes, co-occurrence patterns, and environmental interactions of PA and FL bacteria in drinking water reservoirs remain as yet unexplored. To bridge this gap, we collected samples from 10 sites across four seasons in Lake Tianmu, a large drinking water reservoir in China. Analysis of 16S rRNA gene libraries demonstrated spatiotemporal variations in bacterial diversity and identified differences in bacterial community composition (BCC) between PA and FL lifestyles. Capacity for nitrogen respiration, nitrogen fixation, and nitrate denitrification was enriched in the PA lifestyle, while photosynthesis, methylotrophy, and methanol oxidation were enriched in the FL lifestyle. Deterministic processes, including interspecies interactions and environmental filtration, dominated the assembly of both PA and FL bacterial communities. The influence of environmental filtration on the FL community was stronger than that on the PA community, indicating that bacteria in the FL lifestyle were more sensitive to environmental variation. Co-occurrence patterns and keystone taxa differed between PA and FL lifestyles. The ecological functions of keystone taxa in the PA lifestyle were associated with the supply and recycling of nutrients, while those in FL were associated with the degradation of complex pollutants. PA communities were more stable than FL communities in the face of changing environmental conditions. Nutrients (e.g., TDN and NO3 -) and abiotic and biotic factors (e.g., WT and Chl-a) exerted positive and negative effects, respectively, on the co-occurrence networks of both lifestyles. These results improve our understanding of assembly processes, co-occurrence patterns, and environmental interactions within PA and FL communities in a drinking water reservoir.

Bacterial Diversity and Community Structure Characteristics in Rhizosphere Soil of Coconut (Cocos nucifera L.) under Intercropping Pineapple

To explore the bacterial diversity and community structure characteristics of the rhizosphere soil of fruit type coconuts of different varieties under the intercropping pineapple model, the Illumina high throughput sequencing technology was used to detect the composition and diversity of soil microorganisms. Furthermore, the correlation between the physical and chemical properties of soil and the characteristics of the rhizosphere soil community was analyzed. The contents of OM, EC, AN, AP, AK, ACa, and AMg in coconut rhizosphere soil under intercropping were significantly higher than those under coconut monocropping. However, the soil pH value of the intercropping mode decreased significantly. The diversity of bacteria in the rhizosphere soil of coconut intercropped pineapple was higher than that of coconut monocropped. The diversity of bacteria in the rhizosphere soil of different varieties of coconuts was also different: The order of bacterial diversity was JG > JYT > JYO > JH. The dominant bacteria of the rhizosphere soil, in order, were Proteobacteria, Acidobacteriota, Bacteroidota, Verrucomicrobiota, Actinobacteria, Planctomycota, Firmicutes, Gemmatimonadota, Myxococcota, Nitrospirota, which indicated that the composition of bacterial species between intercropping and monocropping was very different. JG, JYO, and JYT had a high similarity of bacterial community through principal component analysis and cluster analysis, and the bacterial community composition of these three treatments was significantly different from that of CH and JH. In addition, JH had significant differences in bacterial community composition compared to CH. This also showed that different varieties of coconut intercropping pineapple have a great impact on the bacterial composition of coconut rhizosphere soil.

Soil bacterial community structure in the habitats with different levels of heavy metal pollution at an abandoned polymetallic mine.

Heavy metal pollution caused by mining activities can be harmful to soil microbiota, which are highly sensitive to heavy metal stress. This study aimed to investigate the response of soil bacterial communities to varying levels of heavy metal pollution in four types of habitats (i.e., tailing, remediation, natural recovery, and undisturbed areas) at an abandoned polymetallic mine by high-throughput 16S rRNA gene sequencing, and to determine the dominant ecological processes and major factors driving the variations in bacterial community composition. The diversity and composition of bacterial communities varied significantly between soil habitats (p<0.05). Heterogeneous selection played a crucial role in shaping the difference of bacterial community composition between distinct soil habitats. Redundancy analysis and Pearson correlation analysis revealed that the total contents of Cu and Zn were key factors causing the difference in bacterial community composition in the tailing and remediation areas, whereas bioavailable Mn and Cd, total nitrogen, available nitrogen, soil organic carbon, vegetation coverage, and plant diversity were key factors shaping the soil bacterial structure in the undisturbed and natural recovery areas. These findings provide insights into the distribution patterns of bacterial communities in soil habitats with different levels of heavy metal pollution, and the dominant ecological processes and the corresponding environmental drivers, and expand knowledge in bacterial assembly mechanisms in mining regions.

Does plastic type matter? Insights into non-indigenous marine larvae recruitment under controlled conditions.

Marine plastic debris (MPD) are a global threat to marine ecosystems. Among countless ecosystem impacts, MPD can serve as a vector for marine 'hitchhikers' by facilitating transport and subsequent spread of unwanted pests and pathogens. The transport and spread of these non-indigenous species (NIS) can have substantial impacts on native biodiversity, ecosystem services/functions and hence, important economic consequences. Over the past decade, increasing research interest has been directed towards the characterization of biological communities colonizing plastic debris, the so called Plastisphere. Despite remarkable advances in this field, little is known regarding the recruitment patterns of NIS larvae and propagules on MPD, and the factors influencing these patterns. To address this knowledge gap, we used custom-made bioassay chambers and ran four consecutive bioassays to compare the settlement patterns of four distinct model biofouling organisms' larvae, including the three notorious invaders Crassostrea gigas, Ciona savignyi and Mytilus galloprovincialis, along with one sessile macro-invertebrate Spirobranchus cariniferus, on three different types of polymers, namely Low-Linear Density Polyethylene (LLDPE), Polylactic Acid (PLA), Nylon-6, and a glass control. Control bioassay chambers were included to investigate the microbial community composition colonizing the different substrates using 16S rRNA metabarcoding. We observed species-specific settlement patterns, with larvae aggregating on different locations on the substrates. Furthermore, our results revealed that C. savignyi and S. cariniferus generally favoured Nylon and PLA, whereas no specific preferences were observed for C. gigas and M. galloprovincialis. We did not detect significant differences in bacterial community composition between the tested substrates. Taken together, our results highlight the complexity of interactions between NIS larvae and plastic polymers. We conclude that several factors and their potential interactions influenced the results of this investigation, including: (i) species-specific larval biological traits and ecology; (ii) physical and chemical composition of the substrates; and (iii) biological cues emitted by bacterial biofilm and the level of chemosensitivity of the different NIS larvae. To mitigate the biosecurity risks associated with drifting plastic debris, additional research effort is critical to effectively decipher the mechanisms involved in the recruitment of NIS on MPD.

Response of soil bacterial communities in wheat rhizosphere to straw mulching and N fertilization.

Straw mulching and N fertilization are effective in augmenting crop yields. Since their combined effects on wheat rhizosphere bacterial communities remain largely unknown, our aim was to assess how the bacterial communities respond to these agricultural measures. We studied wheat rhizosphere microbiomes in a split-plot design experiment with maize straw mulching (0 and 8,000 kg straw ha-1) as the main-plot treatment and N fertilization (0, 120 and 180 kg N ha-1) as the sub-plot treatment. Bacterial communities in the rhizosphere were analyzed using 16S rRNA gene amplicon sequencing and quantitative PCR. Most of the differences in soil physicochemical properties and rhizosphere bacterial communities were detected between the straw mulching (SM) and no straw mulching (NSM) treatments. The contents of soil organic C (SOC), total N (TN), NH4 +-N, available N (AN), available P (AP) and available K (AK) were higher with than without mulching. Straw mulching led to greater abundance, diversity and richness of the rhizosphere bacterial communities. The differences in bacterial community composition were related to differences in soil temperature and SOC, AP and AK contents. Straw mulching altered the soil physiochemical properties, leading to greater bacterial diversity and richness of the rhizosphere bacterial communities, likely mostly due to the increase in SOC content that provided an effective C source for the bacteria. The relative abundance of Proteobacteria was high in all treatments and most of the differentially abundant OTUs were proteobacterial. Multiple OTUs assigned to Acidobacteria, Chloroflexi and Actinobacteria were enriched in the SM treatment. Putative plant growth promoters were enriched both in the SM and NSM treatments. These findings indicate potential strategies for the agricultural management of soil microbiomes.

Although insect development is known to be a driver of changes of bacterial communities, the fluctuations along BSF life cycle in terms of composition and diversity of bacterial communities are still unknown. In this work, we used a metabarcoding approach to explore the differences in bacterial diversity along all four BSF developmental stages: eggs, larvae, pupae, and adult. We detected not only significant differences in bacterial community composition and species richness along the development of BSF, but also nine prevalent amplicon single variants (ASVs) forming the core microbiota. Out of the 2010 ASVs identified, 160 were significantly more abundant in one of the life stages. Moreover, using PICRUSt2, we inferred 27 potential metabolic pathways differentially used among the BSF life cycle. This distribution of metabolic pathways was congruent with the bacterial taxonomic distribution among life stages, demonstrating that the functional requirements of each phase of development are drivers of bacterial composition and diversity. This study provides a better understanding of the different metabolic processes occurring during BSF development and their links to changes in bacterial taxa. This information has important implications for improving bio-waste processing in such an economically important insect species.

Highly effective reduction of phosphate and harmful bacterial community in shrimp wastewater using short-term biological treatment with immobilized engineering microalgae

Shrimp farming wastewater includes high amounts of phosphate and microbiological contaminants, necessitating further treatment before release into receiving water bodies. After 24 h of shrimp wastewater treatment, alginate beads containing the blue-green algal Synechocystis strain lacking the phosphate regulator gene (mutant strain ΔSphU) at 150 mg L−1 reduced phosphate content from 17.5 mg L−1 to 5.0 mg L−1, representing 71.5% removal efficiency, with phosphate removal rate reaching 6.9 mg gDW−1 h−1 during photobioreactor operation. For short-term treatment, removal rates of nitrate, ammonium and nitrite were 42.7, 48.5 and 92.9%, respectively. Microalgal encapsulated beads also impacted the bacterial community composition dynamics in shrimp wastewater. Next-generation sequencing targeting the V3–V4 region of the 16S rDNA gene showed significant differences in bacterial community composition after 24 h of treatment. Proteobacteria are the most abundant phylum in shrimp wastewater. After 24 h of bioremediation, reductions of harmful bacteria in the Cellvibrionaceae and Pseudomonadaceae families were recorded at 5.85 and 3.18%, respectively. Engineered microalgal immobilization under optimal conditions can be applied as an alternative short-term bioremediation strategy to remove phosphate and other harmful microbial contamination from shrimp farming wastewater.

Effects of common microplastics on aerobic composting of cow manure: Physiochemical characteristics, humification and microbial community

With the detection of microplastics in organic/biomass wastes, the risks of utilizing aerobic composting technology to treat wastes were increasingly recognized, but the impacts of microplastics on composting remain largely unidentified. The objective of this study was to investigate the effect of common microplastics (polypropylene (PP) and polyethylene (PE)) during 55 days composting. In this study, 0.5% PP and 0.5% PE were separately added to the mixture of cow manure, corn straw and bean dregs. The treatment without microplastics was regarded as control (CK). Microplastics significantly reduced the seed germination index values and the NO3−-N contents by 32.40%− 42.78% and 30.37%− 37.04%, respectively. Moreover, the degree of polymerization and humification index of CK were maximum at the end of composting (3.64% and 27.75%) compared to the microplastics added treatments. The alpha and beta diversity analyses showed that the exposure of microplastics reduced the abundance and diversity of bacterial communities. On day 3, the differences in bacterial community composition were mainly caused by time, while the effect of microplastics was more pronounced in the long-term composting. The relative abundance of potential bacterial pathogens at the end of composting in the CK was 2.14%− 2.96% lower than that in the microplastics added treatments. In summary, the results indicated that microplastics reduced compost quality while increased the potential pathogenic risk of composted products.

Periodontitis induced by orthodontic wire ligature drives oral microflora dysbiosis and aggravates alveolar bone loss in an improved murine model.

AimTo assess the contribution of polymicrobial disruption of host homeostasis to periodontitis progression in orthodontic wire ligation murine model.MethodsOrthodontic wire rings were inserted between the first and second molars of mice for 18 days for the orthodontic wire ligation mouse model, and Pg injection model and Pg-LPS injection model were used as controls. Alveolar bone loss and periodontal inflammation were analyzed by micro-CT, histological staining and qRT-PCR. Further, pyrosequencing of 16S rRNA gene amplicon was used to analyze the development of oral microorganism dysbiosis in the mice.ResultsMicro-CT, TRAP staining and qRT-PCR showed that orthodontic wire ligation model led to more severe alveolar bone loss than Pg and Pg-LPS models.H&E staining and qRT-PCR demonstrated that stronger inflammatory response was induced by the orthodontic wire treatment compared to the other models. In addition, pyrosequencing of 16S rRNA gene amplicons revealed that the composition of oral microbiota presented a transition as the disease progressed and significant differences emerged in oral microbiota communities between orthodontic ligature mice and healthy controls. Furthermore, antibiotic treatment decreased both inflammation and alveolar bone loss in response to microbial community dysbiosis. However, no significant difference in bacterial community composition was observed in Pg and Pg-LPS models.ConclusionsOrthodontic wire ligation drove oral microbial community transitions that mimicked polymicrobial communities characterized by polymicrobial synergy and dysbiosis. Our improved model is suitable for further study of pathogenesis of periodontitis and exploration of corresponding treatment strategies.
