Systemic Analyses of Anti-Cell-Senescence Active Compounds in Camellia Sect. Chrysantha Chang and Their Mechanisms.

Aging is an irreversible pathophysiological process for all organisms. The accumulation of senescent cells in pathological sites or tissues is recognized as the major cause of diseases and disorders during the aging process. Small molecules that reduce senescent cell burdens have gained increasing attention as promising intervention therapeutics against aging, but effective anti-senescence agents remain rare. Camellia Sect. Chrysantha Chang is documented as a traditional Chinese herbal medicine used by ethnic groups for many medical and health benefits, but its effect on aging is unclear. Here, we investigated the anti-senescence potential of eight C. Sect. Chrysantha Chang species. The results show that ethyl acetate fractions from these C. Sect. Chrysantha Chang species were able to delay the senescence of H9c2 cardiomyocytes except for C. pingguoensis (CPg). N-butanol fractions of C. multipetala (CM), C. petelotii var. grandiflora (CPt), and C. longzhouensis (CL) showed a senescent cell clearance effect by altering the expression levels of senescent-associated marker genes in the DNA-damage response (DDR) pathway and the senescent cell anti-apoptotic pathway (SCAPs). By using UPLC-QTOF-MS-based non-targeted metabolomics analyses, 27 metabolites from Sect. Chrysantha species were putatively identified. Among them, high levels of sanchakasaponin C and D in CM, CPt, and CL were recognized as the key bioactive compounds responsible for senescent cell clearance. This study is the first to disclose and compare the anti-cell-senescence effect of a group of C. Sect. Chrysantha Chang, including some rare species. The combination of senescent markers and metabolomics analyses helped us to reveal the differences in chemical constituents that target senescent cells. Significantly, contrary to the C. chrysantha var. longistyla (CCL), which is widely cultivated and commercialized for tea drinks, CM, CPt, and CL contain unique chemicals for managing aging and aging-related diseases. The results from this study provide a foundation for species selection in developing small-molecule-based drugs to alleviate diseases and age-related dysfunctions and may potentially be useful for advancing geroscience research.

Diversity in Pyracantha fortuneana fruits maturity stages enables discrepancy in the phenolic compounds, antioxidant activity, and tyrosinase inhibitory activity.

Pyracantha fortuneana (P. fortuneana) fruit is a wild fruit that is popular because of its delicious taste and numerous nutrients, and phenolic compounds are considered to be the main bioactive components in P. fortuneana fruits. However, the relationship between phenolic compounds and their antioxidant and tyrosinase (TYR) inhibitory activities during the ripening process is still unclear. The study compared the influence of the five developmental stages on the accumulation of phenolic compounds, antioxidant activity, and TYR inhibitory activity in the fruits of P. fortuneana. The compounds were identified by offline two-dimensional liquid chromatography-electrochemical detection (2D-LC-ECD) combined with liquid chromatography-tandem mass spectrometry, and the main active ingredients were quantified. The results showed that stage II had higher total phenolic and flavonoid content, as well as higher antioxidant and TYR inhibitory activity, but the total anthocyanin content was lowest at this stage. A total of 30 compounds were identified by 2D-LC-ECD. Orthogonal partial least squares discriminant analysis screened out six major potential markers, including phenolic acids, procyanidins, and flavonoids. In addition, it was found that caffeoylquinic acids, procyanidins, and flavonoids were higher in stage II than in stages I, III, IV, and V, whereas anthocyanins accumulated gradually from stages III to V. Therefore, this study suggests that the changes in antioxidant and TYR inhibitory activities of P. fortuneana during the five developmental stages may be due to the transformation of procyanidins, caffeoylquinic acids, and phenolic glycosides into other forms during the fruit maturation process. Practical Application: Differences in chemical constituents, antioxidant, and tyrosinase inhibitory activities in fruit maturity stages of P. fortuneana were elucidated to provide reference for rational harvesting and utilization of the fruits and their bioactive components. These findings are expected to provide a comprehensive assessment of the bioactive profile and guide the food industrial production.

UPLC-Q-TOF-MSE Based Profiling of the Chemical Composition of Different Parts of Tetrastigma hemsleyanum

Objectives: The differences in chemical constituents in different parts of Tetrastigma hemsleyanum were analyzed. Methods: The contents of total phenols, total flavonoids, and polysaccharides in methanol extracts from different parts of T. hemsleyanum in Zhejiang were determined, and the chemical components of different parts of T. hemsleyanum were collected by ultra-performance liquid chromatography-tandem quadrupole-time-of-flight mass spectrometry. The differential index components of different parts of T. hemsleyanum were identified and analyzed by accurate mass-to-charge ratio of primary mass spectrometry and fragment ion of secondary mass spectrometry with reference spectrum and software database search and related literature reports. Results: A total of 20 components were identified, including 13 identified in root tubers, 7 in fibrous roots and 8 in leaves, and the structural cleavage of its compounds was analyzed. Conclusion: It provides a possibility and theoretical basis for the future use of the whole plant of T. hemsleyanum as medicine.

Research progress in raw and processed Raphani Semen and prediction of quality markers

Raphani Semen, with both edible and medicinal values, is a typical Chinese herbal medicine with different effects before and after processing. The raw helps ascending and the cooked helps descending. This paper comprehensively summarizes the differences in chemical constituents and pharmacological effects between raw and processed Raphani Semen that are reported in recent years. Based on the principle of quality markers(Q-markers) of traditional Chinese medicines, the chemical constituent sources, chemical constituent detection techniques, and correlation between bidirectional regulatory efficacy and chemical constituents are compared between raw and processed Raphani Semen. The results suggest that sulforaphene and glucoraphanin could be used as candidate Q-markers of raw and processed Raphani Semen, respectively. This review is expected to provide a reference for further research on the processing, new drug development, and improvement of safety and effectiveness of Raphani Semen in clinical application.

Differences of Chemical Constituents and Pharmacological Activities of Traditional Herbs Before and After Honey-Frying

Processing is one of the important and crucial parts that has been applied in traditional oriental medicine for a long time. The traditional ingredients were processed on the basis of the Yin Yang theory, Five Elements theory, and folk experiences. Among many processing techniques, honey-frying is a process enhancing the effects of traditional herbs on the Spleen and Stomach organs. This report provides an overview of the differences between chemical constituents and pharmacological activities of some traditional ingredients before and after roasting with honey.

Quality consistency evaluation of chemical composition and pharmacology of Shaoyao-Gancao decoction dispensing granules and traditional decoction.

Dispensing granules of Chinese medicine (DGCM) have emerged as a more convenient alternative to traditional decoction (TD) of Chinese medicine, gaining popularity in recent years. However, the debate surrounding the consistency of DGCM compared to TD remains unresolved. In this study, three batches of Baishao and Gancao DGCM were obtained from manufacturers A, B, and C, and 15 batches of crude drugs were procured from hospital pharmacies for the preparation of dispensing granule decoction (DGD) and TD of Shaoyao-Gancao decoction (SGD). The HPLC-UV method was employed to determine the levels of gallic acid, paeoniflorin, albiflorin, liquiritin, liquiritin apioside, isoliquiritin apioside, isoliquiritin, glycyrrhizic acid, and isoliquiritigenin. The analgesic and antispasmodic effects were assessed using the hot plate and acetic acid writhing test in mice. To evaluate the consistency of chemical constituents and pharmacological effects between the two decoctions, the Criteria Importance Though Intercriteria Correlation (CRITIC) method combined with chemometrics was employed. Grey relation analysis (GRA) was used to assess the comprehensive quality consistency of the two decoctions. The CRITIC results revealed certain differences in chemical constituents and pharmacological effects between the selected DGCM and TD. Notably, DGD-A/C exhibited a significant difference from TD (p > 0.05), whereas DGD-B demonstrated no significant difference from TD (p > 0.05). The GRA analysis demonstrated that the overall quality consistency between DGD-B and TD was the highest among the three manufacturers. This study presents a method for evaluating the quality consistency of DGCM and TD of SGD, offering novel insights into the evaluation of consistency between DGCM and TD.

Chemical Variability of Hexane Extracts from Five Subspecies of Calendula suffruticosa from Spain.

The chemical profile of the hexane extracts of the subspecies carbonellii, greuteri, marginata, trialata, and vejerensis of Calendula suffruticosa growing in Spain, herein described for the first time, were studied to access their value as a chemo taxonomical tool and search for potentially useful compounds. The subsp. greuteri and carbonellii showed higher extract yields. Terpenoids were the most abundant chemical class in subsp. carbonellii, greuteri, trialata, and vejerensis, while alkanes were the most abundant in subsp. marginata. Differences in chemical constituents were identified among the subspecies of C. suffruticosa analysed, which the PCA can prove. The subsp. trialata and greuteri showed more significant phytochemical similarity, which might indicate genetic proximity between these two subspecies. C. suffruticosa subsp. marginata presented the fewest number of compounds and in the smallest quantities, and C. suffruticosa subsp. vejerensis presented the largest number, however, both showed no alcohols. Furthermore, some of the compounds found in significant amounts are known for their pharmacological and nutraceutical properties, denoting potential use.

Differences in Chemical Constituents between Dalbergia oliveri Heartwood and Sapwood and Their Effect on Wood Color.

The purpose of this study was to characterize and quantify the chemical constituents of heartwood and sapwood of Dalbergia oliveri extract in order to investigate the chemical components that determine the formation of heartwood's color. In this work, the types of pigments in heartwood and sapwood extract were analyzed using UV-Visible (UV) Spectrophotometer, and the main pigment components of heartwood and sapwood extract were identified and quantified using ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS). The results showed that the difference in content of the main components between heartwood and sapwood of Dalbergia oliveri was slight, and the lignin structure between heartwood and sapwood is basically identical; flavonoid pigments were found to be the primary chromophoric components of heartwood and sapwood extract. However, a total of 21 flavonoids were identified in heartwood and sapwood, of which the unique substances to heartwood were vitexin, isorhamnetin, and pelargonidin, and the content of isoliquiritigenin, formononetin, and biochanin A were 253, 37, and 583 times higher in the heartwood than in the sapwood, respectively, which could be the main pigment components affecting the significant color difference between heartwood and sapwood of Dalbergia oliveri. These results will provide a foundation for revealing the underlying mechanism of color difference between heartwood and sapwood and provide a theoretical basis for wood coloring.

Effect of Different Processing Methods on the Chemical Constituents of Scrophulariae Radix as Revealed by 2D NMR-Based Metabolomics.

Scrophulariae Radix (SR) is one of the oldest and most frequently used Chinese herbs for oriental medicine in China. Before clinical use, the SR should be processed using different methods after harvest, such as steaming, “sweating”, and traditional fire-drying. In order to investigate the difference in chemical constituents using different processing methods, the two-dimensional (2D) 1H-13C heteronuclear single quantum correlation (1H-13C HSQC)-based metabolomics approach was applied to extensively characterize the difference in the chemical components in the extracts of SR processed using different processing methods. In total, 20 compounds were identified as potential chemical markers that changed significantly with different steaming durations. Seven compounds can be used as potential chemical markers to differentiate processing by sweating, hot-air drying, and steaming for 4 h. These findings could elucidate the change of chemical constituents of the processed SR and provide a guide for the processing. In addition, our protocol may represent a general approach to characterizing chemical compounds of traditional Chinese medicine (TCM) and therefore might be considered as a promising approach to exploring the scientific basis of traditional processing of TCM.

Classification of Pu-erh ripened teas and their differences in chemical constituents and antioxidant capacity

For the scientific classification of Pu-erh ripened teas (PRTs), phytochemical compositions including 14 polyphenolic compounds, 2 purine alkaloids and 18 free amino acids in 24 PRTs were determined by HPLC, respectively. The in vitro antioxidant activities were evaluated by 5 various assays. PCA explaining 38.7% of total variance, HCA and OPLS-DA (R2Y = 0.887, Q2 = 0.873, respectively) could classify PRTs into Puer and Banna groups by production place basically, which caused significant (P < 0.05), highly significant (P < 0.01) or extremely significant (P < 0.001) differences of 29 chemical components. The 19 relevant components used for the identification of storage location through OPLS-DA (R2Y = 0.625, Q2 = 0.601, respectively) were selected. PRTs of various origins, such as production place and storage location, demonstrated significant (P < 0.05) differences in antioxidant activities. Several polyphenolic compounds like (−)-epicatechin, (−)-gallocatechin, rutin and kaempferol contributed to the antioxidant activities. This study helps to understand chemical profile and antioxidant activities of PRTs.

Effect of the degree of milling on the physicochemical properties, pasting properties and in vitro digestibility of Simiao rice

Simiao rice is a variety of long-grain Indica rice popular in South China. However, researches on the milling technology of Simiao rice are limited. The effect of different degrees of milling (DOMs) of Simiao rice (0–12%) were evaluated in terms of the physicochemical properties and in vitro starch digestibility of the rice. The results showed that, as the DOM increased from 0 to 12%, the head rice yield (HRY) and the content of protein, lipid, dietary fiber, vitamin B1, vitamin E and niacin nonlinearly decreased, while the content of total starch and amylose and paste viscosity parameters nonlinearly increased. The differences in chemical constituents between the brown and milled Simiao rice with different DOMs lead to the variation in their pasting properties and in vitro digestibility. Because of lower content of slowly digestible starch (SDS) and resistant starch (RS), milled Simiao rice exhibited greater digestibility and a higher estimated glycaemic index (EGI). The EGI of Simiao rice ranged from 77.98 to 85.55, and remained almost constant (approximately 84) when the DOM was above 4%. Correlation analysis indicated that the EGI had strong negative correlations with lipid, protein, dietary fiber and RS, but a positive correlation with amylose. These results provide theoretical guidance for the production of Simiao rice with desired physicochemical properties and digestibility by adjusting the DOM.

UHPLC−QTOF−MS based metabolite profiling analysis and the correlation with biological properties of wild and artificial agarwood

To date, the agarwood has been over exploited worldwide in the wild due to high demand. As an alternative, the agarwood obtained through artificial methods has greatly resolved the shortage of agarwood supply in the global market. However, little information about the difference in chemical constituents and bioactivities of the wild agarwood and the artificial agarwood is available. This study aims to systematically compare the chemical composition and the bioactivities between wild and artificial agarwood on the basis of the integrated method of ultra-high-performance liquid chromatography coupled with time-of-flight mass spectrometry (UHPLC-TOF-MS) and multivariate statistical analysis. The invitro antioxidant activity was determined using the 2,2′-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl hydrate (DPPH) radical scavenging activity assays. The cytotoxic activity of agarwood from different origin against three human cancer cell lines (i.e., A375, U251, and Skov3) were compared using the MTT assay. Fifty metabolites from UPLC-QTOF-MS spectra were identified and included in the multivariate analysis. Among these metabolites, 2-(2-phenylethyl) chromone derivatives (PECs), bi-2-(2-phenylethyl) chromone derivatives (BPECs) and sesquiterpene-2-(2-phenylethyl) chromone conjugates (SPECs) were found to be the major metabolites and acted as discriminant compounds in agarwood from different origin. The antioxidant activity study showed that the wild agarwood displayed significant antioxidant capacity compared with the artificial agarwood. Particularly, the content of secondary metabolites of SPEC analogs shown a positive effect on the radical scavenging activities, whereas the PECs were negatively correlated. Interestingly, no significant difference was observed between wild and artificial agarwood in terms of cytotoxic activity against three human cancer cell lines (i.e., A375, U251, and Skov3). To the best of our knowledge, this research is the first to study the metabolite profiles and bioactivities of the wild and the artificial agarwood in a holistic approach, and is expected to provide a rational basis for the quality assessment of artificial agarwood as a substitute for wild agarwood.

Untargeted Metabolomics Profiling and Global Semi-quantitation of a Prescription Chinese Herbal Medicine Formula Yinqiaosan Using UPLC-QTOF-MS with a Single Exogenous Reference Internal Standard

Abstract Yinqiaosan is a classic Chinese herbal medicine formula that has been used to treat various bacterial and viral infections by Chinese medicine doctors for over two centuries. In this work, we developed a comprehensive qualitative and quantitative method for identification, quantitation, and quality assessment of chemical constituents of Yinqiaosan formula in four different preparation forms (i.e., decoction, granule, pill, and tablet), which employed ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry with a single exogenous reference internal standard for untargeted metabolomics profiling and global semiquantitative analysis. The use of a single exogenous reference internal standard permitted not only qualitative and quantitative analyses of multiple herbal components in a single instrument run, but also cross-comparison of chemical contents in between all four Yinqiaosan preparation forms. The acquired mass chromatograms were analyzed, quantitated, and compared using multivariate data analysis for similarities and differences of chemical constituents in four Yinqiaosan preparation forms. For the first time, we were able to identify over 100 chemical constituents from each preparation form using the available database. Among the 49 commonly identified compounds in the 4 Yinqiaosan preparation forms, 16 have been reported to have pharmacological activities, which may be used in a network pharmacology study of Yinqiaosan for exploring the underlying mechanism of the herbal formula.

Mechanism of anti-inflammatory effects of volatile compounds of Ai pian based on network pharmacology, in vivo animal experiments, and GC–MS

Ai pian (AP) is a well-known Miao national herb with resuscitative effects. However, pharmacological and clinical applications of AP are limited because its precise molecular mechanism remains unclear. This study was conducted to evaluate the anti-inflammatory activities of the volatile compounds of AP in in vivo animal models and determine the molecular mechanism underlying the anti-inflammatory effects based on network pharmacology and molecular docking. We performed gas chromatography-mass spectrometric analysis of volatile compounds with chemometric methods, including hierarchical clustering analysis and principal component analysis, to identify AP from different origins. Mouse models of xylene-induced ear edema were used to examine the in vivo anti-inflammatory activities of AP with cotton ball-granulation test. The mechanism of AP was determined by network pharmacology analysis and molecular docking. Significant differences in chemical constituents and percentage contents were observed among different habitats. We found that AP exerted potent anti-inflammatory effect, and that multiple targets and pathways were involved in this effect. These results provided a foundation for further comprehensive development and application of AP from Miao national herb.

Productivity of Summer Savory (Satureja hortensis L.) In Different Locations of Egypt

Field experiment was conducted to evaluate the growth, yield and essential oil constituents of savory at three different locations across Egypt (El-Minya, El-Sharkia and Sinai). Plants cultivated in El-Sharkia showed significantly higher values of plant height, number of branches and herb fresh and dry weights. Also, the highest quality was observed on plant cultivated in El-Sharkia region. Thirty one main compounds were detected in GC-MS analysis of essential oil. There were obvious differences in chemical constituents of essential oil of plants cultivated under different locations. The main components were a-Terpinene and Carvacrol. The highest a-Terpinene and Carvacrol were observed also in plants cultivated in El-Sharkia. These results suggested that agro-climatic zone is very important for maximizing the content and composition of essential oils.Further studies are still needed to determine the precise factors of agro-climatic zoning which could be important strategies for optimizing the yield and quality.

Quality evaluation of multi-components simultaneous determination and fingerprint of Gardeniae Fructus from different regions

An analysis method was established by UPLC fingerprint and then applied to simultaneous determination of multiple compounds in Gardeniae Fructus from different areas in China. Samples were separated on a Waters Acquity UPLC BEH C18( 2. 1 mm×50 mm,1. 7 μm) column with 0. 1% formic acid-water and acetonitrile solution as gradient mobile phase at a flow rate of 0. 4 m L·min-1 at various wavelengths. The similarity of samples was over 0. 95 with ″Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine( 2012 edition) ″. The UPLC common fingerprints for 32 batches were established with 19 common peaks identified. The samples were divided into 3 groups analyzed by HCA and PCA. Five components were identified as the main compositions which caused the differences of chemical constituents in the samples from different areas with partial least squares discriminant analysis( PLS-DA). The content of the total components in each area was Zhejiang > Fujian > Jiangxi > Sichuan. This method was accurate and viable,could be used to evaluate the quality of Gardeniae Fructus.

Simultaneous Determination and Analysis of Major Ginsenosides in Wild American Ginseng Grown in Tennessee.

Ginsenosides are the major constituent that is responsible for the health effects of American ginseng. The ginsenoside profile of wild American ginseng is ultimately the result of germplasm, climate, geography, vegetation species, water, and soil conditions. This is the first report to address the ginsenoside profile of wild American ginseng grown in Tennessee (TN), the third leading state for production of wild American ginseng. In the present study, ten major ginsenosides in wild American ginseng roots grown in TN, including Rb1, Rb2, Rb3, Rc, Rd, Re, Rf, Rg1, Rg2, and Rg3, were determined simultaneously. The chemotypic differences among TN wild ginseng, cultivated American ginseng, and Asian ginseng were assessed based on the widely used markers of ginsenoside profiling, including the top three ginsenosides, ratios of PPD/PPT, Rg1/Rb1, Rg1/Re, and Rb2/Rc. Our findings showed marked variation in ginsenoside profile for TN wild ginseng populations. Nevertheless, TN wild ginseng has significant higher ginsenoside content and more ginsenoside diversity than the cultivated ginseng. The total ginsenoside content in TN wild ginseng, as well as ginsenosides Rg1 and Re, increases with the age of the roots. Marked chemotypic differences between TN wild ginseng and cultivated American ginseng were observed based on the chemotypic markers. Surprisingly, we found that TN wild ginseng is close to Asian ginseng with regard to these characteristics in chemical composition. This study verified an accessible method to scientifically elucidate the difference in chemical constituents to distinguish wild from the cultivated American ginseng. This work is critical for the ecological and biological assessments of wild American ginseng so as to facilitate long-term sustainability of the wild population.

Chemical Constituents of Essential Oil in Chervil (Anthriscus cerefolium L. Hoffm.) Cultivated in Different Locations

Climate plays an important role in crop production, and it is the main factor limiting the success of crop production in specific area. Medicinal plants are characterized by its contents of biological active substances. Among those, essential oil is considered as one of the most important natural substances. This field experiment was conducted during the two successive seasons 2016-2017 and 2017-2018 in order to evaluate the growth, yield and essential oil constituents of chervil plants at four different locations across Egypt (El-Fayoum, El-Minya, El-Sharkia, and El-Baharia Oasis). Plants cultivated in El-Fayoum showed the highest growth rate (plant height) and the highest yield (fresh and dry herb) followed by El-Sharkia. Also, the highest quality (0.180 and 0.185 % essential oil with 0.158 and 0.120 ml/plant in first and second season, respectively) was observed on plant cultivated in El-Fayoum and the lowest observed in El-Baharia Oasis plants. Thirteen main compounds were detected in GC/MS analysis of essential oil. There were obvious differences in chemical constituents of essential oil of plants cultivated under different locations. The lowest Methyleugenol, Estragoleand 2-Allyl-1,4-dimethoxybenzenecontents were observed in plant cultivated in El-Minya location. The highest Methyleugenol content (47.21 %) was observed in El-Sharkia, the highest Estragole content (18.04) were observed in El-Fayoum, and the highest 2-Allyl-1,4-dimethoxybenzene content (7.89) was observed in El-Baharia Oasis. The results of this study suggested that agro-climatic zone and suitable location selection could be as important strategies for maximizing the content and composition of essential oils.

AbstractLitsea glutinosa samples collected from Thai Nguyen and Thua Thien‐Hue province were investigated chemically and biologically. As results: There were remarkable differences in chemical constituents and biological activities between two Litsea glutinosa samples. Flavonoid glycosides, megastigmanes and aporphine alkaloids were isolated and identified from the sample collected in Thua Thien Hue province, while no flavonoid glycosides, only three aporphine alkaloids and some fatty acids and fatty acid esters were isolated from the sample collected in Thai Nguyen province. The ethanol extract of sample from Thua Thien‐Hue province showed a stronger α‐ glucosidase inhibition than that of the sample from Thai Nguyen province. The in vivo antidiabetic activity of Thua Thien‐Hue sample was evaluated. The result was promising.

Chemical Constituents of Sweetpotato Genotypes in Relation to Textural Characteristics of Processed French Fries.

In recent years, sweetpotato French fries (SPFF) have grown in popularity, but limited information is available on SPFF textural properties in relation to the differences in chemical constituents among sweetpotato varieties. This study demonstrated that sensory texture attributes of SPFF varied widely and were significantly correlated with chemical components such as dry matter, starch, and total sugar contents of raw sweetpotatoes and instrumental texture measurements of SPFF.
