Bacteriological evaluation and advanced SYBR-green multiplex real-time PCR assay for detection of minced meat adulteration.

Minced meat is a valuable source of nutrients, but it is vulnerable to contamination by microorganisms commonly present in the environment. In addition, there is a risk of adulteration with cheaper meat sources, which can be harmful to consumers. It is crucial to identify meat adulteration with distinct microbiological analysis for legal, economic, religious, and public health purposes. A total of 100 minced meat samples were collected from several markets in Sharkia Governorate, Egypt. These samples were then subjected to bacteriological testing and an advanced multiplex PCR method. This method enables the detection of bovine, equine, porcine, and dog species in meat samples with just one step. The adulterated samples had a higher total bacterial count and pH values compared to pure bovine meat. These differences in bacterial count and pH values were statistically significant, with p-values of 0.843 (log10) and 0.233, respectively. The frequency of Escherichia coli occurrence was 13%, and the O111 serotype was predominant in the adulterated samples. Listeria monocytogenes and Staphylococcus aureus were isolated with prevalence rates of 3% and 29%, respectively. Besides, the SYBR-green multiplex real-time PCR assay used in this study detected adulteration with dog, equine, and porcine meats in the examined samples at rates of 9%, 5%, and 4%, respectively. This method provides a sensitive and specific approach to detect issues related to well-being and safety.

Metagenomic analysis of fecal samples in colorectal cancer Egyptians patients post colectomy: A pilot study.

One of the most prevalent malignancies that significantly affects world health is colorectal cancer (CRC). While genetics are involved in a portion of CRC patients, most cases are sporadic. The microbiome composition could be a new source of tumor initiation and progression. This research was conducted to investigate the microbiota composition of CRC patients post colectomy at taxonomic and functional levels. Using a next-generation sequencing approach, using an Illumina Novaseq 6000, the fecal samples of 13 patients were analyzed and the obtained data was subjected to a bioinformatics analysis. The bacterial abundance and uniqueness varied in CRC patients alongside differences in bacterial counts between patients. Bacteroides fragilis, Bacteroides vulgatus, Escherichia coli, and Fusobacterium nucleatum were among the pro-cancerous microorganisms found. Concurrently, bacteria linked to CRC progression were detected that have been previously linked to metastasis and recurrence. At the same time, probiotic bacteria such as Bifidobacterium dentium, Bifidobacterium bifidum, and Akkermansia muciniphila increased in abundance after colectomies. Additionally, numerous pathways were deferentially enriched in CRC, which emerged from functional pathways based on bacterial shotgun data. CRC-specific microbiome signatures include an altered bacterial composition. Our research showed that microbial biomarkers could be more usefully employed to explore the link between gut microbiota and CRC using metagenomic techniques in the diagnosis, prognosis, and remission of CRC, thereby opening new avenues for CRC treatment.

In vitro comparison of methods for sampling copper-based antimicrobial surfaces.

Self-sanitizing surfaces such as copper (Cu) are increasingly used on high-touch surfaces to prevent the spread of harmful viruses and bacteria. Being able to monitor the antimicrobial properties of Cu is fundamental in measuring its antimicrobial efficacy. Thorough investigations into reliable methods to enumerate bacteria from self-sanitizing surfaces are lacking in the literature. This study demonstrates that direct use of Petrifilm on Cu surfaces most likely revives stressed and dying bacteria, which induces increased bacterial counts. This phenomenon was not observed with indirect collection methods. Studies assessing time-kill kinetics or long-term efficacy of Cu should consider the impact of the collection method chosen.

Agronomic Characteristics of Maize Grown in Paddy Soil, Along with Root Zone Soil Chemical Properties and Soil Bacterial Numbers

Maize is being considered as an alternative crop for off-season rice production in Thailand due to increasing water scarcity. The objective of this study was to evaluate the agronomic characteristics and root-zone soil properties of 4 hybrid maize varieties grown in paddy soil during off-season rice cultivation, in order to assess their growth and development after the flooded-rice harvest and the associated soil microorganisms. The randomized complete block design with 4 treatments and 4 replications was used. The 4 treatments were 2 new maize varieties (Suwan 5731 and Suwan 5819), 1 check variety (Suwan 4452), and 1 local use variety (PAC 789). Suwan 5819 showed significantly higher plant height than Suwan 4452, Suwan 5731 and PAC 789, with the highest plant height recorded for Suwan 5819 (221 cm). Similarly, Suwan 5819 exhibited significantly higher estimated leaf area index than Suwan 4452, Suwan 5731 and PAC 789, with differences of 32.67, 42.57 and 29.29 % at VT, respectively. These superior performances of Suwan 5819 could be attributed in part to increased dry matter and nutrient accumulation at the VT stage. The grain yield for PAC 789 was 13.71 t ha–1, Suwan 4452 was 13.23 t ha–1, Suwan 5731 was 12.83 t ha–1, and Suwan 5819 was 12.25 t ha–1. However, there was no significant difference in the grain yields among the 4 varieties. The soil in the root zone of Suwan 5819 at VT was significantly less acidic (pH 5.08) than Suwan 4452 (pH 4.58), PAC 789 (pH 4.61) and Suwan 5731 (pH 4.75). The nitrogen-fixing bacteria were most abundant in PAC 789 (84 %), phosphate-solubilizing bacteria in Suwan 4452 (15 %), and potassium-solubilizing bacteria in Suwan 5819 (16 %). Overall, the study suggests that Suwan 5819 is a promising maize variety for agronomic performance in the tested region. HIGHLIGHTS Maize is a viable alternative to rice for off-season cultivation in water-scarce regions. One new maize hybrid (Suwan 5819) showed better growth and nutrient accumulation during the vegetative transition stage. Soil properties, including beneficial microorganisms, did not differ significantly among the four maize varieties studied. Suwan 5819 new maize hybrid had less acidic root zone soil conditions than the other tested varieties. Commercial maize hybrid had the highest counts of N-fixing and K-solubilizing bacteria in the root zone soil, but there was no significant difference in bacterial count compared to the Suwan maize GRAPHICAL ABSTRACT

Assessment of Pre-operative Vaginal Preparation for Laparoscopic Hysterectomy.

Determine the difference in microbial growth from the vagina and uterine manipulator among patients undergoing laparoscopic hysterectomy after randomization to one of three vaginal preparation solutions (10% Povidone-iodine, 2% Chlorhexidine, or 4% Chlorhexidine). This was a prospective randomized controlled trial in an academic community hospital. Patients were ≥ 18 years old and scheduled for laparoscopic hysterectomy for benign and malignant indications. Fifty patients were identified and randomized into each arm. Prior to surgery, the surgical team prepared the vaginal field using 10% Povidone-iodine, 2% Chlorhexidine, or 4% Chlorhexidine, according to group assignment. Cultures were collected from the vagina after initial preparation, prior to the colpotomy, and on surfaces of the uterine manipulator. Bacterial count from the baseline vaginal fornix/cervical canal cultures did not differ significantly among the three groups. There was a difference in bacterial count among the second cervical canal/vaginal fornix cultures (p < 0.01), with the Povidone-iodine arm demonstrating the highest level of growth of cultures (93.8%), followed by 2% Chlorhexidine (47.4%), and 4% Chlorhexidine (20%). There was no difference in growth on the uterine manipulator handle and no difference in vaginal itching or burning was found across the three arms postoperatively. Bacterial growth prior to colpotomy was the lowest with 4% Chlorhexidine followed by 2% Chlorhexidine, the Povidone-iodine group exhibited the highest bacterial growth. There was no difference in moderate to severe vaginal itching or burning. This showed that 4% Chlorhexidine is superior in reducing bacterial growth when used in laparoscopic hysterectomy.

Effect of calcium fluoride nanoparticles in prevention of demineralization during orthodontic fixed appliance treatment: a randomized clinical trial.

White spot lesions (WSLs) are the most common complications of fixed appliance orthodontic treatment. To evaluate the effectiveness of calcium fluoride nanoparticles-containing orthodontic primer (nCaF2-primer) in preventing the incidence of WSLs during orthodontic treatment. Single-centre, double-blinded, split-mouth, randomized clinical trial. The sample involved 31 orthodontic patients (≥12 years). Participants were recruited using a simple nonstratified randomization. Data collection, measurements, and analysis were performed blindly. Outcome measures included comparing the effect of nCaF2-primer with control primer (Transbond) regarding the degree of demineralization (DIAGNOdent pen), Streptococcus mutans (S. mutans) bacterial counting [real-time polymerase chain reaction device (PCR)], and WSLs incidence (pre- and post-operative photographs). The measurements were performed before bonding, 1, 3, and 6 months after bonding and after appliance removal. A two-way repeated measure analysis of variance test (for DIAGNOdent pen scores), and Wilcoxon signed-rank test (for the difference between bacterial counting and WSLs incidence) were used (P < 0.05). Thirty-one patients were recruited and randomized (mean age 17.9 ± 2.45 years). For the primary outcome (DIAGNOdent pen scores) and secondary outcome of S. mutans counting: 31 patients (310 teeth for each group) were included in scoring at T1 and T3, and 30 patients (300 teeth) were included at T6. While for the photographic scores, 26 patients were included after bracket bonding. The demineralization scores showed significant differences at all-time intervals within the 6 months after bracket bonding which was more noticeable after the first month. There was a significant difference in bacterial count between the two primer groups at the T1 only. Regarding photographic scores, there were no significant differences in the WSLs incidence between the two primers groups after brackets removal. No harm was detected during treatment, except the usual pain/gingival irritation. nCaF2-primer effectively decreased demineralization scores within the 6 months after bracket bonding. Moreover, it significantly reduced S. mutans colonization after the first month. However, the tested primer did not have an extra advantage in preventing WSLs development at the clinical level after appliance removal. The trial was registered with ClinicalTrials.gov on 8 May 2021 (registration number: NCT04994314).

Stop Drill, Make a Change: An In Vivo Study.

BackgroundThe chemomechanical method of caries removal is a modality that is non-invasive, cost-effective, and if some caries preventive measures could be added to them, then it might render the remaining dentin more resistant to future caries attacks also. Thereby, this study is conducted to evaluate the in vivo aspect of newly developed “one man army material—Apacaries gel”.Aim and objectiveTo evaluate and compare the antimicrobial efficacy, efficiency, and pain perception of a chemomechanical caries removal agent (Apacaries gel) with a conventional method.DesignTwenty children (aged 6–12 years) with bilateral occlusal caries making a sample size of 40 were included in the study. Group I: caries removal using rotary instruments and Group II: caries removal using Apacaries gel (n = 20 each). Caries removal time and pain perception were measured using a stopwatch and Wong–Baker Pain Scale. Bacterial removal in flattened root canals with different tapers of ProDesign Logic single-file system

Objective: This study evaluated the bacterial removal in flattened root canals after preparation with different tapers of ProDesign Logic instruments. Methodology: Fifty-five mandibular incisors with flattened root canals were contaminated with a pure culture of Enterococcus faecalis ATCC 29212 for 21 days at 37ºC. Root canals preparation was performed using ProDesign Logic and 0.9% sterile saline solution for irrigation. The specimens were randomly allocated to three experimental groups (n = 15) according to the taper of the instrument used: 25.03, 25.04, or 25.06. Intracanal bacteriological samples were taken before (S1) and after (S2) preparation and the number of colony-forming units per milliliter (CFU/mL) was calculated. Differences in bacterial counts before and after treatment, within each group, were analyzed by the paired t-test. Groups were compared by ANOVA with a post hoc test. The level of significance was set at p &lt;0.05. Results: Mature biofilm formation was confirmed at root canal walls by scanning electron microscopy. Bacterial colonies were significantly reduced in all experimental groups at S2 compared to S1 (p &lt;0.001). No significant difference was observed between groups regarding the percentage of bacterial reduction (p &gt;0.05). Conclusion: Preparation of flattened root canals using ProDesign Logic instruments with tapers of 0.03, 0.04 and 0.06 resulted in similar planktonic and bacterial biofilm reduction. None of the instruments rendered root canals completely free of bacteria.

Semen quality and total microbial load: An association study in important Indian Goat breeds during different seasons.

The invasion of the male urogenital tract by microorganisms, and its subsequent effects on sperm fertilising ability, has not been well discussed in bucks. The present study was conducted to assess the bacterial load in fresh semen of the 2-6years old bucks. For conducting the experiment, semen ejaculates from 18 bucks (6 from each breed namely Jakhrana, Jamunapari and Barbari) were used. We collected 5 ejaculates from each buck in each season (Summer-April to June, Rainy-July to Sept and Winter-November to January). Semen was collected with the artificial vagina (AV) method, and separate AV was used for each buck every time. The semen collection frequency was once in a week. Immediately after initial evaluation, collected semen samples were transferred to the microbiology laboratory of the institute. Thereafter, the semen samples were subjected to bacteriological examination to assess the microbial load. The results of the current study indicate that the microbial load in the semen was significantly (p<0.05) higher in the Jamunapari bucks and in aged bucks. Bacteriospermia in different seasons was not significantly varied; however, nonsignificant increase in microbial load during the rainy season was observed. Overall, the average bacterial load in the semen of Jamunapari, Barbari and Jakhrana bucks was found 540.50±55.88CFU/ml, 391.81±46.33CFU/ml and 388.93±44.71CFU/ml respectively. No significant difference in bacterial counts in the subsequent ejaculates among bucks was observed. Moreover, correlation analysis revealed that the proportions of motility, viability, plasma membrane integrity and acrosomal integrity were negatively influenced by the increased bacterial contamination of buck semen.

Comparison of Periodontopathic Bacterial Profiles of Different Periodontal Disease Severity Using Multiplex Real-Time Polymerase Chain Reaction.

Periodontopathic bacteria are known to have a pivotal role in the pathogenesis of periodontitis. The aim of the study was to quantitatively compare bacterial profile of patients with different severity of periodontal disease using samples from mouthwash and the subgingival area. Further analysis was performed to evaluate the correlation between mouthwash and two subgingival sampling methods: paperpoint and gingival retraction cord; 114 subjects enrolled in the study, and were divided equally into three groups according to disease severity. Mouthwash and subgingival sampling were conducted, and the samples were quantitatively analyzed for 11 target periodontopathic bacteria using multiplex real-time PCR. There were statistically significant differences in bacterial counts and prevalence of several species between the study groups. Mouthwash sampling showed significant correlations with two different subgingival sampling methods in regard to the detection of several bacteria (e.g., ρ = 0.793 for Porphyromonas gingivalis in severe periodontitis), implying that mouthwash sampling can reflect subgingival microbiota. However, the correlation was more prominent as disease severity increased. Although bacteria in mouthwash have potential to become a biomarker, it may be more suitable for the diagnosis of severe periodontitis, rather than early diagnosis. Further research is required for the discovery of biomarkers for early diagnosis of periodontitis.

The application possibility of acellular dermal matrix decorated with nano-silver in the reconstruction of contaminated abdominal wall

Acellular dermal matrix (ADM) is a biomaterial, which commonly used for repair of tissue defects; however, infection is the main factor underlying the failure of treatments involving ADM. To enhance the anti-infection ability of ADM, we constructed a new form of ADM that was decorated with nano-silver (‘NS-ADM’). The introduction of nano-silver did not destroy the decellularized structure of ADM, and no significant difference was detected with regards to the maximum tensile force when compared between NS-ADM and ADM (P = 0.351). NS-ADM was not cytotoxic to cell growth when the concentration of nano-silver solution ≤ 25 ppm and exhibited strong antibacterial activity in vitro. Besides, when rats were inoculated with 104 CFU/mL, there were significantly lower bacterial counts in the NS-ADM group than in the ADM group when assessed seven days after surgery (P = 0.047); no significant differences were detected on days 14 and 28. Although there were no significant differences in bacterial counts on days 7, 14, or 21 between the two groups (rats were inoculated with 106 CFU/mL), the number of rats showing reduced bacterial counts or clearing was higher in the NS-ADM group than in the ADM group. Rats that were inoculated with 108 CFU/mL showed repair failure. Overall, NS-ADM is a promising antibacterial biomaterial for repairing contaminated soft-tissue defects, in which antibacterial properties are superior to ADM. The antibacterial activity of NS-ADM was limited for severe infections, and further in vivo studies are needed to evaluate its efficacy and biosafety.

DNA vaccination with the Mycobacterium marinum MMAR_4110 antigen inhibits reactivation of a latent mycobacterial infection in the adult Zebrafish

BackgroundTuberculosis is a major challenge for health care, as options for its treatment and prevention are limited. Therefore, novel approaches, such as DNA vaccination, to both prevent primary infections and the reactivation of latent infections need to be developed. A Mycobacterium marinum infection in adult zebrafish (Danio rerio) recapitulates features of the human Mycobacterium tuberculosis infection, providing a convenient preclinical animal model for studying tuberculosis. MethodsHypoxic M. marinum cultures were produced with the Wayne model, and further reaerated to replicate the in vivo reactivation in vitro. Expression levels of M. marinum genes were studied with mRNA sequencing from exponentially growing bacteria, anaerobic cultures and at 2 and 12 h after reaeration. Seven reactivation-associated genes were selected for further studies, where their antigen potentiality as DNA-vaccines to prevent reactivation of a latent mycobacterial infection was investigated in the adult zebrafish model. The Mann-Whitney test was used to evaluate differences in bacterial counts between the groups. ResultsThe mRNA sequencing data showed that, seven M. marinum genes, MMAR_0444, MMAR_0514, MMAR_0552, MMAR_0641, MMAR_1093, MMAR_4110 and MMAR_4524, were upregulated during reactivation when compared to both dormant and logarithmic growing bacteria. Four different MMAR_4110 antigens prevented the reactivation of a latent mycobacterial infection in the adult zebrafish. ConclusionThis study provides novel information about reactivation-related M. marinum genes. One of the antigens, MMAR_4110, inhibited the reactivation of a latent M. marinum infection in zebrafish, implicating that the characterized genes could be potential targets for further vaccine and drug development against mycobacterial diseases.

Bacteriospermia and Sperm Quality of Cryopreserved Bull Semen Used in Artificial Insemination of Cows in South Wollo Zone, Ethiopia.

The objectives of this trial were to estimate prevalence of bacteriospermia, to determine the bacterial load, and to isolate the types of bacteria as well as to assess the association between bacterial load and sperm quality traits in cryopreserved bull semen in field conditions in the South Wollo Zone. A total of 309 cryopreserved straws of semen from the Holstein Friesian (HF)-cross bull (n = 180 straws) and pure Jersey bull (n = 129 straws) were investigated. Bacteriological assessments of the presence of aerobic bacteria, estimation of bacterial count and bacterial isolation, as well as semen quality were performed. Aerobic bacterial contamination was prevalent in 38.8% of the semen straws. No significant difference in the prevalence of bacteriospermia was observed among bulls although the HF-cross bull had a higher prevalence (40.0%). But, significant difference in prevalence of bacteriospermia was found among semen ejaculates of the same bull. The risk of bacteriospermia in the HF-cross bull was higher (Odds ratio = 1.86, 95% CI = 0.168–20.26) compared to Jersey although not significant. Overall average bacterial load of 50.38 ± 16.29 colony-forming units (CFU)/ml (from nil to 1318.20 CFU/ml) was found. No significant difference in bacterial count among bulls and their ejaculates was observed. Moreover, correlation analysis revealed that the proportions of motility, live, and normal morphology were negatively influenced by an increase in the bacterial contamination of semen. In this study, three isolates of coagualse-negative Staphylococcus species and one isolate of Corynebacterium species were found. Average percentages of sperm motility (48.35 ± 1.23), live (66.08 ± 1.0), and normal morphology (80.62 ± 1.24) were observed. It was concluded that cryopreservation does not guarantee the quality of semen from bacterial contamination. Hence, meticulous care should be adopted to prevent contamination of semen by bacteria during collection, transportation, processing, and storage times.

Antimicrobial Efficacy of Charcoal vs. Non-charcoal Toothbrushes: A Randomized Controlled Study.

ABSTRACTAim:To assess the efficacy of the antimicrobial properties of charcoal vs. non-charcoal toothbrushes and the level of bacterial contamination in the oral cavity using a charcoal toothbrush.Materials and Methods:This was a randomized, double-blind controlled study in which both male and female subjects aged from 18 to 35 were included (n = 30; 15 males and 15 females). The subjects were selected from (students) of Riyadh Elm University in Riyadh, Saudi Arabia. Subjects were informed about the study and signed the consent form before participation. From January to April 2019, subjects were given charcoal and non-charcoal toothbrushes.Results:The Wilcoxon signed rank test showed that there was a significant difference in bacterial counts between non-charcoal and charcoal toothbrushes (P = 0.000). Of the subjects, 70% showed a decrease in the number of bacterial counts while 30% showed no increase in bacterial counts. There was a statistically significant decrease in the number of bacteria in the gingival crevicular fluid (GCF) with charcoal treatment (P < 0.001). Of the subjects, 96.6% showed a decrease in the count of bacteria in GCF after using a charcoal toothbrush. Only 3.3% of the subjects had the similar counts of bacteria in GCF after using the charcoal toothbrush.Conclusion:The study demonstrates that charcoal toothbrushes reduce bacterial contamination and the poor effects on oral health after 1 week of use.

Effect of Infectious Bursal Disease (Ibd) Vaccines on Infection of Salmonella Heidelberg in Broiler Chickens

ABSTRACT Salmonella Heidelberg (SH) has represented a great concern to the Brazilian poultry industry in the last years. It is known that immunosuppression in poultry is a contributing factor to increase Salmonella faecal shedding and to disturb control programmes. Not only infectious bursal disease (IBD) virus but also some live vaccines have been reported to induce immunosuppression. In the present study we assessed the effects of two live vaccines against IBD on SH-infected broiler chicks. At 7 days of age, birds of three groups (vaccinated with recombinant HVT-IBD vector, with immune complex-IBD vaccine and unvaccinated) were orally challenged with 1 x 108 CFU of SH. A group of hatchmates remained unvaccinated/unchallenged to serve as negative controls. Caecal colonization and systemic invasion were evaluated by bacterial enumeration at 1, 3, 5, 7 and 14 days post-infection (Dpi) and SH faecal shedding assessed by cloacal swabs at 3, 7, 10 and 14 Dpi. The counts of SH in caecal contents were higher in birds vaccinated with immune complex-IBD than in those that received the HVT-IBD vector vaccine at 5, 7 and 14 dpi (p<0.01). There were no statistical differences in bacterial counts in liver and spleen among birds of different groups. Cloacal swabs also indicated that the birds vaccinated with immune complex-IBD shed more SH than those vaccinated with HVT-IBD vector or those unvaccinated (p<0.01). The results of the present study suggested that the immunosuppressive effect of the immune complex-IBD vaccine helped to increase the SH-faecal shedding in the infected birds.

Bacterial Flora and Proximate Composition of Edible Frogs (Ptychadena mascareniensis and Ptychadena pumilio) from Some Locations in Rivers State, Nigeria

Aim: To determine and compare the bacterial flora and proximate composition of edible frogs from some selected Local Government Areas (LGA) of Rivers State.&#x0D; Place and Duration of Study: Live adult edible frogs were collected from six locations in six Local Government Areas in Rivers State which includes; Biara (E 7º29’ 2.097’’, N 4º78’ 70.608’’) in Gokana, Rumuodumaya (E 7º0’ 57.16.908’’, N 4º52’ 40.398’’) in Obio/Akpo, Umuikere Oyigbo (E 6º55’ 50.606’’, N 9º1’ 13.831’’) in Oyigbo, Diobu (E 6º59’ 6.882’’, N 4º48’ 29.514’’) in Port Harcourt, Igwuruta (E 4º55’ 57.006’’, N 7º1’13.692’’) in Ikwerre and Akpajo (E 5º50’ 67.306’’, N 9º1’ 43.112’’) in Eleme Local Government Areas of Rivers State. This study was conducted from July 2017 to February 2018, covering both the wet and dry seasons.&#x0D; Methodology: The microbiological analyses were carried out using standard microbiological methods. Conventional and molecular identification methods were used to identify the bacteria isolated. Proximate compositions of the two species of edible frogs were also determined using standard methods. Analysis of variance (ANOVA) was used to test for significant difference between the data obtained from the various locations using a computer based program SPSS version 22.&#x0D; Results: Results revealed that Ptychadena pumilios had high bacteria counts across the six Local Government areas. Counts of total heterotrophic bacteria for Ptychadena pumilios ranged from 1.0 x108cfu/g to 9.0 x108cfu/g, Total Staphylococcus count ranged from 1.0 x105cfu/g to 9.4 x105cfu/g, Total coliform counts ranged 1.0 x105cfu/g to 8.9 x105 cfu/g, Total Feacal Coliform counts ranged from 1.4 x104cfu/g to 9.0 x104cfu/g. while for Ptychadena mascareniensis Total heterotrophic bacterial counts ranged from 2.2 x108 to 4.5 x107cfu/g, Staphylococcal counts ranged from 1.8 x105cfu/g to 9.5 x105cfu/g, Total coliform counts ranged 1.0 x105cfu/g to 9.8 x105cfu/g, Total Feacal Coliform ranged from 1.2 x104cfu/g 9.8 x104cfu/g. Statistically there were significant differences in bacterial counts in the six studied LGAs. Statistically there were significant differences in the bacterial counts from the six studied locations. A total of 259 bacterial isolate belonging to the following genera: Escherichia, Staphylococcus, Bacillus, Klebsiella, and Pseudomonas were isolated during this study. The proportion of nutritional composition obtained in this study ranged as follow: carbohydrate from 1.82 to 2.24, crude fibre 2.28 to 3.00, ash 3.99 to 4.89%, lipid 11.90 to 12.32%, protein13.74 to14.00%, moisture 64.45 to 65.74%, for the species of the edible frogs tested.&#x0D; Conclusion: The presence of Escherichia coli identified indicates faecal contamination and indicates the possible presence of potential pathogens. Proper processing and cooking of the frog meat will destroy these organisms, thereby preventing food borne infections. Improper handling and cooking may lead to food borne infections with greater effect observed in immunocompromised patents, the elderly and children. Ptychadena mascareniensis and Ptychadena pumilio have high protein contents therefore, they could be considered as an alternative source of protein despite the bacterial load.

Microbial Contamination of Organically and Conventionally Produced Fresh Vegetable Salads and Herbs from Retail Markets in Southwest Germany.

A total of 189 samples of fresh products (leafy salads, ready-to-eat mixed salads, and fresh herbs) bought in retail in Southwest Germany were investigated for their microbiological quality and the presence of pathogenic bacteria, including Salmonella spp., Listeria monocytogenes, and presumptive Bacillus cereus. Total aerobic mesophilic plate counts (TAC) ranged from 5.5 to 9.6 log colony-forming units (CFUs) per gram. Enterobacteria and pseudomonads were the predominant microorganisms and were detected in all samples with counts between 5.0 and 9.2 log CFU/g. Strains of Escherichia coli were detected in 9 salad (7.9%) and 25 herb samples (33.3%). Significant differences in bacterial counts were found between conventionally and organically-grown products: in herbs the counts of moulds were significantly higher in organically-grown products, while E. coli was only detected in conventionally-grown products. In conventionally-grown salad samples, yeast counts were significantly higher. Salmonella Enteritidis was only detected in two conventionally- and in one organically-produced salad samples (2.6%). No coagulase-positive staphylococci were detected in fresh salads as well as in herbs. High levels of B. cereus sensu lato (≥3 log CFU/g) were detected in 19 vegetable salads (16.7%) and even in 55 samples of fresh herbs (73.3%). Listeria monocytogenes could not be detected in fresh herbs; however, three L. monocytogenes strains were isolated from two conventionally-produced salad samples and belonged to PCR serogroup IIa. Although our results indicate a high microbial load in fresh salads and herbs in Southwest Germany in 2015, the incidences of human pathogenic bacteria, that is, L. monocytogenes, Salmonella spp., and coagulase-positive staphylococci strains, were low.

Surgical masks as source of bacterial contamination during operative procedures

BackgroundSurgical masks (SMs) are used to reduce bacterial shedding from the mouth, nose and face. This study aimed to investigate whether SMs may be a potential source of bacterial shedding leading to an increased risk of surgical site infection. MethodsBacterial contamination of the SMs was tested by making an impression of the external surface of the mask on sterile culture media immediately. We investigated the difference in bacterial counts between the SMs worn by surgeons and those placed unused in the operating room (OR), and the bacterial count variation with indicated wearing time. Moreover, the difference in bacterial counts on the external surface between the first and second layers of double-layered SMs was also assessed. ResultsThe bacterial count on the surface of SMs increased with extended operating times; significant difference was found between the 4- to 6-hour and 0-hour groups (p < 0.05). When we analysed the bacterial counts from the same surgeon, a significant increase was noted in the 2-hours group. Moreover, the bacterial counts were significantly higher among the surgeons than the OR. Additionally, the bacterial count of the external surface of the second mask was significantly higher than that of the first one. ConclusionsThe source of bacterial contamination in SMs was the body surface of the surgeons rather than the OR environment. Moreover, we recommend that surgeons should change the mask after each operation, especially those beyond 2 hours. Double-layered SMs or those with excellent filtration function may also be a better alternative. The translational potential of this articleThis study provides strong evidence for the identification that SMs as source of bacterial contamination during operative procedures, which should be a cause for alarm and attention in the prevention of surgical site infection in clinical practice.

Evaluation of Petrifilm Lactic Acid Bacteria Plates for Counting Lactic Acid Bacteria in Food

Although lactic acid bacteria (LAB) are used widely as starter cultures in the production of fermented foods, they are also responsible for food decay and deterioration. The undesirable growth of LAB in food causes spoilage, discoloration, and slime formation. Because of these adverse effects, food companies test for the presence of LAB in production areas and processed foods and consistently monitor the behavior of these bacteria. The 3M Petrifilm LAB Count Plates have recently been launched as a time-saving and simple-to-use plate designed for detecting and quantifying LAB. This study compares the abilities of Petrifilm LAB Count Plates and the de Man Rogosa Sharpe (MRS) agar medium to determine the LAB count in a variety of foods and swab samples collected from a food production area. Bacterial strains isolated from Petrifilm LAB Count Plates were identified by 16S rDNA sequence analysis to confirm the specificity of these plates for LAB. The results showed no significant difference in bacterial counts measured by using Petrifilm LAB Count Plates and MRS medium. Furthermore, all colonies growing on Petrifilm LAB Count Plates were confirmed to be LAB, while yeast colonies also formed in MRS medium. Petrifilm LAB Count Plates eliminated the plate preparation and plate inoculation steps, and the cultures could be started as soon as a diluted food sample was available. Food companies are required to establish quality controls and perform tests to check the quality of food products; the use of Petrifilm LAB Count Plates can simplify this testing process for food companies.

Evidence of variable bacterial colonization on coloured elastomeric ligatures during orthodontic treatment: An intermodular comparative study.

BackgroundBesides, other factors, the choice of materials used as orthodontic ligatures could be one of the many tools to counter the effects of microbial adhesion, that culminates into dental ailments. Therefore, we assessed bacterial adhesion on elastomeric ligatures with special reference to coloured elastomeric rings during orthodontic treatment.Material and MethodsA split mouth study, involving 240 samples of different elastomeric ligatures from forty orthodontic patients possessing good oral hygiene was carried out. The archwire was ligated to the brackets on both arches with elastomeric rings (superslick, clear transparent , blue and pink) at predetermined quadrants. After six weeks, ligatures from second premolars were removed and processed for bacterial enumeration using standard techniques. Bacterial counts were also determined for stimulated saliva samples taken at 0 and 6 weeks.ResultsA statistically significant difference in bacterial counts was obtained amongst different elastomeric modules used. Maximum bacterial counts were found on conventional pigmented elastomeric modules, followed by Superslick module and clear module. More number of bacteria associated with the conventional pink as compared to the conventional blue pigmented modules, however it was not statistically significant. The three bacterial genera Streptococcus Staphylococcus and Aerobic Lactobacilli adhered to elastomeric modules in following predominant pattern i.e. Conventional pink>Conventional Blue>Superslick>Clear.ConclusionsThe studies evidenced colour and material dependent bacterial colonization on orthodontic modules and could be an indicator of bacterial biofilm forming potential based on surface chemistries and a clinically efficacious tool to redesign conventional and modified elastomeric rings as orthodontic ligation accessories.
