Dietary glycine supplementation enhances syntheses of creatine and glutathione by tissues of hybrid striped bass (Morone saxatilis ♀ × Morone chrysops ♂) fed soybean meal-based diets

BackgroundWe recently reported that supplementing glycine to soybean meal-based diets is necessary for the optimum growth of 5- to 40-g (Phase-I) and 110- to 240-g (Phase-II) hybrid striped bass (HSB), as well as their intestinal health. Although glycine serves as an essential substrate for syntheses of creatine and glutathione (GSH) in mammals (e.g., pigs), little is known about these metabolic pathways or their nutritional regulation in fish. This study tested the hypothesis that glycine supplementation enhances the activities of creatine- and GSH-forming enzymes as well as creatine and GSH availabilities in tissues of hybrid striped bass (HSB; Morone saxatilis♀ × Morone chrysops♂).MethodsPhase-I and Phase-II HSB were fed a soybean meal-based diet supplemented with 0%, 1%, or 2% glycine for 8 weeks. At the end of the 56-d feeding, tissues (liver, intestine, skeletal muscle, kidneys, and pancreas) were collected for biochemical analyses.ResultsIn contrast to terrestrial mammals and birds, creatine synthesis occurred primarily in skeletal muscle from all HSB. The liver was most active in GSH synthesis among the HSB tissues studied. In Phase-I HSB, supplementation with 1% or 2% glycine increased (P < 0.05) concentrations of intramuscular creatine (15%–19%) and hepatic GSH (8%–11%), while reducing (P < 0.05) hepatic GSH sulfide (GSSG)/GSH ratios by 14%–15%, compared with the 0-glycine group; there were no differences (P > 0.05) in these variables between the 1% and 2% glycine groups. In Phase-II HSB, supplementation with 1% and 2% glycine increased (P < 0.05) concentrations of creatine and GSH in the muscle (15%–27%) and liver (11%–20%) in a dose-dependent manner, with reduced ratios of hepatic GSSG/GSH in the 1% or 2% glycine group. In all HSB, supplementation with 1% and 2% glycine dose-dependently increased (P < 0.05) activities of intramuscular arginine:glycine amidinotransferase (22%–41%) and hepatic γ-glutamylcysteine synthetase (17%–37%), with elevated activities of intramuscular guanidinoacetate methyltransferase and hepatic GSH synthetase and GSH reductase in the 1% or 2% glycine group. Glycine supplementation also increased (P < 0.05) concentrations of creatine and activities of its synthetic enzymes in tail kidneys and pancreas, and concentrations of GSH and activities of its synthetic enzymes in the proximal intestine.ConclusionsSkeletal muscle and liver are the major organs for creatine and GSH syntheses in HSB, respectively. Dietary glycine intake regulates creatine and GSH syntheses by both Phase-I and Phase-II HSB in a tissue-specific manner. Based on the metabolic data, glycine is a conditionally essential amino acid for the growing fish.

Dietary glycine supplementation enhances glutathione availability in tissues of pigs with intrauterine growth restriction.

This study tested the hypothesis that dietary supplementation with glycine enhances the synthesis and concentrations of glutathione (GSH, a major antioxidant) in tissues of pigs with intrauterine growth restriction (IUGR). At weaning (21 d of age), IUGR pigs and litter mates with normal birth weights (NBW) were assigned randomly to one of two groups, representing supplementation with 1% glycine or 1.19% l-alanine (isonitrogenous control) to a corn- and soybean meal-based diet. Blood and other tissues were obtained from the pigs within 1wk after the feeding trial ended at 188 d of age to determine GSH, oxidized GSH (GSSG), and activities of GSH-metabolic enzymes. Results indicated that concentrations of GSH + GSSG or GSH in plasma, liver, and jejunum (P < 0.001) and concentrations of GSH in longissimus lumborum and gastrocnemius muscles (P < 0.05) were lower in IUGR pigs than in NBW pigs. In contrast, IUGR increased GSSG/GSH ratios (an indicator of oxidative stress) in plasma (P < 0.001), jejunum (P < 0.001), both muscles (P < 0.05), and pancreas (P = 0.001), while decreasing activities of γ-glutamylcysteine synthetase and GSH synthetase in liver (P < 0.001) and jejunum (P < 0.01); and GSH reductase in jejunum (P < 0.01), longissimus lumborum muscle (P < 0.01), gastrocnemius muscle (P < 0.05), and pancreas (P < 0.01). In addition, IUGR pigs had greater (P < 0.001) concentrations of thiobarbituric acid reactive substances (TBARS; an indicator of lipid peroxidation) in plasma, jejunum, muscles, and pancreas than NBW pigs. Compared with isonitrogenous controls, dietary glycine supplementation increased concentrations of GSH plus GSSG and GSH in plasma (P < 0.01), liver (P < 0.001), jejunum (P < 0.001), longissimus lumborum muscle (P = 0.001), and gastrocnemius muscle (P < 0.05); activities of GSH-synthetic enzymes in liver (P < 0.01) and jejunum (P < 0.05), while reducing GSSG/GSH ratios in plasma (P < 0.001), jejunum (P < 0.001), longissimus lumborum muscle (P < 0.001), gastrocnemius muscle (P = 0.01), pancreas (P < 0.05), and kidneys (P < 0.01). Concentrations of GSH plus GSSG, GSH, and GSSG/GSH ratios in kidneys were not affected (P > 0.05) by IUGR. Furthermore, glycine supplementation reduced (P < 0.001) TBARS concentrations in plasma, jejunum, muscles, and pancreas. Collectively, IUGR reduced GSH availability and induced oxidative stress in pig tissues, and these abnormalities were prevented by dietary glycine supplementation in a tissue-specific manner.

Dietary glycine supplementation activates mechanistic target of rapamycin signaling pathway in tissues of pigs with intrauterine growth restriction.

The mechanistic target of rapamycin (mTOR) cell signaling pathway serves as the central mechanism for the regulation of tissue protein synthesis and growth. We recently reported that supplementing 1% glycine to corn- and soybean meal-based diets enhanced growth performance between weaning and market weights in pigs with intrauterine growth restriction (IUGR). Results of recent studies have revealed an important role for glycine in activating mTOR and protein synthesis in C2C12 muscle cells. Therefore, the present study tested the hypothesis that dietary glycine supplementation enhanced the mTOR cell signaling pathway in skeletal muscle and other tissues of IUGR pigs. At weaning (21 d of age), IUGR pigs and litter mates with normal birth weights (NBW) were assigned randomly to one of the two groups: supplementation with either 1% glycine or 1.19% l-alanine (isonitrogenous control) to a corn- and soybean meal-based diet. Tissues were obtained from the pigs within 1wk after the feeding trial ended at 188 d of age to determine the abundances of total and phosphorylated forms of mTOR and its two major downstream proteins: eukaryotic initiation factor 4E-binding protein-1 (4EBP1) and ribosomal protein S6 kinase-1 (p70S6K). Results showed that IUGR decreased (P < 0.05) the abundances of both total and phosphorylated mTOR, 4EBP1, and p70S6K in the gastrocnemius muscle and jejunum. In the longissimus lumborum muscle of IUGR pigs, the abundances of total mTOR did not differ (P > 0.05) but those for phosphorylated mTOR and both total and phosphorylated 4EBP1 and p70S6K were downregulated (P < 0.05), when compared to NBW pigs. These adverse effects of IUGR in the gastrocnemius muscle, longissimus lumborum muscle, and jejunum were prevented (P < 0.05) by dietary glycine supplementation. Interestingly, the abundances of total or phosphorylated mTOR, 4EBP1, and p70S6K in the liver were not affected (P > 0.05) by IUGR or glycine supplementation. Collectively, our findings indicate that IUGR impaired the mTOR cell signaling pathway in the tissues of pigs and that adequate glycine intake was crucial for maintaining active mTOR-dependent protein synthesis for the growth and development of skeletal muscle.

Excessive Substitution of Fish Meal with Fermented Soybean Meal Induces Oxidative Stress by Impairing Glutathione Metabolism in Largemouth Bass (Micropterus salmoides).

The application of fermented soybean meal (FSBM) is an effective strategy to alleviate the shortage of fish meal (FM) in aquaculture. However, an excessive substitution ratio often reduces fish growth and induces liver oxidative stress, while the mechanism remains poorly understood. Here, an 8-week feeding trial was conducted in largemouth bass (initial weight: 6.82 ± 0.09 g) to establish an oxidative stress model by replacing 50% of FM with FSBM (fermented by Bacillus subtilis). The results showed that FSBM substitution significantly reduced the growth performance of largemouth bass, including the weight gain rate and specific growth rate. Moreover, FSBM significantly reduced the contents of essential amino acids and total free amino acids in muscle, along with the mRNA expression of amino acids and small peptide transporters. Enzyme activity detection and liver sections showed that FSBM substitution caused liver oxidative stress, indicating the successful construction of an oxidative stress model. An integrated analysis of transcriptomic and metabolomic data revealed that FSBM substitution impaired glycine, serine and threonine metabolism, as well as glutathione metabolism. In addition, the ratio of reduced glutathione (GSH) to oxidized glutathione (GSSG) was decreased in the FSBM group, which may explain the mechanism of oxidative stress caused by FSBM substitution. Considering that glycine is an important component of glutathione synthesis, key genes involved in glycine metabolism (glya, gnmt and agxt) and dietary glycine supplementation should be valued to improve the availability of FSBM. This study reveals for the first time the importance of non-essential amino acids in improving the utilization of plant-based protein sources and provides original insight for the optimization of aquatic feeds.

Effect of dietary glycine supplementation on productive performance, egg quality, stress response, and fatty liver incidence in laying hens raised under heat stress conditions

The current experiment aimed to investigate the effect of dietary glycine (Gly) supplementation on productive performance, egg quality, stress response, and fatty liver incidence in laying hens raised under heat stress (HS) conditions. A total of two hundred eighty 24-wk-old Lohmann Brown-Lite laying hens were randomly allotted to 1 of 4 dietary treatments with 7 replicates. The negative control (NC) diet was prepared to meet or exceed the nutrient and energy requirement for Lohmann Brown laying hens, whereas the positive control (PC) diet was formulated to increase AMEn by 100 kcal/kg compared with the NC diet. Two additional diets were prepared by supplementing 0.341% and 0.683% Gly to the NC diet. All hens were exposed to cyclic HS at 31.4 ± 1.17°C for 8 h/d and 26.7 ± 1.10°C for the remaining time for a 12-wk trial. Results indicated that increasing supplementation of Gly in diets tended (linear, P=0.088) to decrease the FCR of laying hens. Increasing supplementation of Gly in diets increased (linear, P < 0.05) eggshell lightness and decreased (linear, P < 0.05) egg yolk color. Moreover, a tendency for a quadratic association (P < 0.10) of serum aspartate aminotransferase and alanine aminotransferase concentrations with increasing supplementation of Gly was observed. Increasing supplementation of Gly in diets decreased (linear, P < 0.05) blood heterophil:lymphocyte ratio of laying hens. Hens fed the NC diet showed higher fatty liver incidence (P < 0.05) than those fed the PC diet, but increasing supplementation of Gly decreased (linear, P < 0.05) fatty liver incidence of laying hens. In conclusion, increasing supplementation of Gly up to 0.683% in diets decreases FCR, stress response, and fatty liver incidence in laying hens raised under HS conditions.

Glycine: The Smallest Anti-Inflammatory Micronutrient.

Glycine is a non-essential amino acid with many functions and effects. Glycine can bind to specific receptors and transporters that are expressed in many types of cells throughout an organism to exert its effects. There have been many studies focused on the anti-inflammatory effects of glycine, including its abilities to decrease pro-inflammatory cytokines and the concentration of free fatty acids, to improve the insulin response, and to mediate other changes. However, the mechanism through which glycine acts is not clear. In this review, we emphasize that glycine exerts its anti-inflammatory effects throughout the modulation of the expression of nuclear factor kappa B (NF-κB) in many cells. Although glycine is a non-essential amino acid, we highlight how dietary glycine supplementation is important in avoiding the development of chronic inflammation.

Effects of Dietary Glycine Supplementation on Growth Performance, Immunological, and Erythrocyte Antioxidant Parameters in Common Carp, Cyprinus carpio.

The effects of dietary glycine supplementation, 0 (control), 5 (5 GL), and 10 (10 GL) g/kg, have been investigated on growth performance, hematological parameters, erythrocyte antioxidant capacity, humoral and mucosal immunity in common carp, Cyprinus carpio. After eight weeks feeding, the 5 GL treatment exhibited significant improvement in growth performance and feed efficacy, compared to the control treatment. Red blood cell (RBC) and white blood cell (WBC) counts, hemoglobin, hematocrit, neutrophil and monocyte counts/percentages, RBC reduced glutathione (GSH) content, and skin mucosal alkaline phosphatase, peroxidase, protease, and lysozyme activities were similar in the glycine-treated fish and significantly higher than the control treatment. Blood lymphocyte percentage decreased in the glycine-treated fish, but lymphocyte count increased, compared to the control fish. RBC glutathione reductase activities in the glycine-treated fish were similar and significantly lower than the control treatment. The highest plasma lysozyme and alternative complement activities were observed in GL treatment. The glycine-treated fish, particularly 5 GL, exhibited significant improvement in RBC osmotic fragility resistance. Dietary glycine had no significant effects on RBC glutathione peroxidase activity, plasma immunoglobulin, eosinophil percentage/count, and hematological indices. In conclusion, most of the benefits of dietary glycine supplementation may be mediated by increased glutathione synthesis and antioxidant power.

Dietary glycine supplementation enhances postweaning growth and meat quality of pigs with intrauterine growth restriction.

Pigs with intrauterine growth restriction (IUGR) have suboptimum growth performance and impaired synthesis of glycine (the most abundant amino acid in the body). Conventional corn- and soybean meal-based diets for postweaning pigs contain relatively low amounts of glycine and may not provide sufficient glycine to meet requirements for IUGR pigs. This hypothesis was tested using 52 IUGR pigs and 52 litter mates with normal birth weights (NBW). At weaning (21 d of age), IUGR or NBW pigs were assigned randomly to one of two nutritional groups: supplementation of a corn-soybean meal-based diet with either 1% glycine plus 0.19% cornstarch or 1.19% L-alanine (isonitrogenous control). Feed consumption and body weight (BW) of pigs were recorded daily and every 2 or 4 wks, respectively. All pigs had free access to their respective diets and clean drinking water. Within 1 wk after the feeding trial ended at 188 d of age, blood and other tissue samples were obtained from pigs to determine concentrations of amino acids and meat quality. Neither IUGR nor glycine supplementation affected (P > 0.05) feed intakes of pigs per kg BW. The final BW, gain:feed ratio, carcass dressing percentages, and four-lean-cuts percentages of IUGR pigs were 13.4 kg, 4.4%, 2%, and 15% lower (P < 0.05) for IUGR pigs than NBW pigs, respectively. Compared with pigs in the alanine group, dietary glycine supplementation increased (P < 0.05) final BW, gain:feed ratio, and meat a* value (a redness score) by 3.8 kg, 11%, and 10%, respectively, while reducing (P < 0.05) backfat thickness by 18%. IUGR pigs had lower (P < 0.05) concentrations of glycine in plasma (-45%), liver (-25%), jejunum (-19%), longissimus dorsi muscle (-23%), gastrocnemius muscle (-26%), kidney (-15%), and pancreas (-6%), as compared to NBW pigs. In addition, dietary glycine supplementation increased (P < 0.05) concentrations of glycine in plasma and all analyzed tissues. Thus, supplementing 1% of glycine to corn-soybean meal-based diets improves the growth performance, feed efficiency, and meat quality of IUGR pigs.

Dietary glycine supplementation improves the growth performance of 110- to 240-g (phase II) hybrid striped bass (Morone saxatilis ♀× Morone chrysops ♂) fed soybean meal-based diets.

We recently reported that supplementing glycine to soybean meal (SBM)-based diets is necessary for optimum growth of 5- to 40-g (phase I) hybrid striped bass (HSB). The present study tested the hypothesis that supplementing glycine to SBM-based diets may enhance the growth of 110- to 240-g (phase II) HSB. HSB (the initial body weight of approximately 110 g) were fed an SBM (58%)-based diet supplemented with 0%, 1%, or 2% of glycine, with l-alanine serving as the isonitrogenous control. There were four tanks per dietary group, with four fish per tank. The fish were fed their respective diets to apparent satiation twice daily. The feed intake and body weight of fish were recorded daily and every 2 wk, respectively. At the end of the 56-d feeding trial, plasma and tissue samples were collected to determine amino acid concentrations and histological alterations, and tissues were used to measure the oxidation of l-glutamate, l-glutamine, l-aspartate, and glycine. Results showed that dietary supplementation with 1% and 2% glycine dose-dependently increased (P < 0.05) the concentration of glycine in the plasma of HSB by 48% and 99%, respectively. Compared with the 0%-glycine group, dietary supplementation with 1% glycine did not affect (P > 0.05) the feed intake of HSB but increased (P < 0.05) their final body weight, weight gain, and gain:feed ratio during the whole period by 13%, 29%, and 21%, respectively. Compared with the 1% glycine group, dietary supplementation with 2% glycine increased (P < 0.05) the feed intake, final body weight, and weight gain of HSB by 13%, 7%, and 14%, respectively. Compared with the 0%-glycine group, fish fed with the 1%-glycine and 2%-glycine diets had a greater (P < 0.05) villus height in the proximal intestine, when compared with the 0%-glycine group. Collectively, these results indicated that SBM-based diets did not provide sufficient glycine for phase II HSB (110 to 240 g) and that dietary glycine supplementation is essential for their optimum growth and intestinal structure.

Dietary glycine supplementation enhances the growth performance of hybrid striped bass (Morone saxatilis ♀× Morone chrysops ♂) fed soybean meal-based diets.

This study was conducted to test the hypothesis that supplementing 1% and 2% glycine to soybean meal (SBM)-based diets can improve the growth performance of juvenile hybrid striped bass (HSB). The basal diets contained 15% fishmeal and 58% SBM (DM basis). Alanine was used as the isonitrogenous control in different diets. All diets contained 44% CP and 10% lipids (DM basis). There were 4 tanks (15 fish per tank) per dietary group, with the mean of the initial body weight (BW) of fish being 5.3g. Fish were fed to apparent satiation twice daily, and their BW was recorded every 2wk. The trial lasted for 8wk. Results indicated that the BW, weight gain, protein efficiency ratio (PER), and retention of dietary lipids in fish were enhanced (P < 0.05) by dietary supplementation with 1% or 2% glycine. In addition, dietary supplementation with glycine did not affect (P > 0.05) the feed intake of fish but increased (P < 0.05) the retention of dietary nitrogen, most amino acids, and phosphorus in the body, compared to the 0% glycine group. Dietary supplementation with 1% and 2% glycine dose-dependently augmented (P < 0.05) the villus height of the intestine and reduced the submucosal thickness of the gut, while preventing submucosal and lamina propria hemorrhages. Compared with the 0% glycine group, dietary supplementation with 1% or 2% glycine decreased (P < 0.05) the proportion of skeletal-muscle fibers with diameters of 40-60 µm but increased (P < 0.05) the proportion of skeletal-muscle fibers with diameters of 80-100 µm and > 100 µm. Collectively, these findings indicate that glycine in SBM-based diets is inadequate for maximum growth of juvenile HSB and that dietary supplementation with 1% or 2% glycine is required to improve their weight gain and feed efficiency. Glycine is a conditionally essential amino acid for this fish.

Dietary glycine supplementation prevents heat stress-induced impairment of antioxidant status and intestinal barrier function in broilers

This study tested the hypothesis that glycine improves intestinal barrier function through regulating oxidative stress in broilers exposed to heat stress. A total of 300 twenty-one-day-old female Arbor Acres broilers (600 ± 2.5g) was randomly allocated to 5 treatments (6 replicate of 10 birds each). The 5 treatments were as follows: the control group (CON) was kept under thermoneutral condition (24 ± 1°C) and was fed a basal diet. Broilers fed a basal diet and reared under high ambient temperature (HT) were considered as the HT group (34 ± 1°C for 8 h/d). Broilers fed a basal diet supplemented with 0.5%, 1.0%, and 2.0% glycine and exposed to HT were regarded as the HT+glycine treatments. The results exhibited that heat stress reduced growth performance, serum total antioxidant capacity (T-AOC), and glutathione (GSH) concentration (P < 0.05); increased activity of serum catalase (CAT) and the contents of hydrogen peroxide (H2O2) and malondialdehyde (MDA) (P < 0.05). HT exposure led to downregulating the mRNA expression of NAD(P)H quinone dehydrogenase 1 (NQO1), Occludin, and zonula occludens-1 (ZO-1) (P < 0.05); enhanced the mRNA levels of Kelch-like ECH-associated protein 1 (Keap1), CAT, glutathione synthetase (GSS), and glutamate-cysteine ligase modifier subunit (GCLM) (P < 0.05); impaired the intestinal morphology (P < 0.05); and altered the diversity and community of gut microbiota (P < 0.05). The final body weight (FBW), ADFI, ADG, and gain-to-feed ratio (G: F) increased linearly or quadratically, and the antioxidant capacity was improved (P < 0.05) with glycine supplementation. Glycine treatment increased the villus height (VH), and villus height to crypt depth ratio (V/C) of the duodenum linearly or quadratically, and linearly increased the VH of jejunum and ileum. The mRNA expression of Occludin, and ZO-1 were increased linearly in the ileum mucosa of broilers subjected to HT. Collectively, these results demonstrated that glycine supplementation alleviates heat stress-induced dysfunction of antioxidant status and intestinal barrier in broilers.

Glycine Alleviated Intestinal Injury by Inhibiting Ferroptosis in Piglets Challenged with Diquat.

The purpose of this research was to examine the impact of glycine on intestinal injury caused by oxidative stress in piglets. A 2 × 2 factorial experiment with diets (basic diet vs. 1% glycine diet) and oxidative stress (saline vs. diquat) was conducted on 32 weanling piglets. On day 21, all piglets received an injection of either saline or diquat. After 7 days, all pigs were slaughtered and intestinal samples were collected. Dietary glycine supplementation improved intestinal mucosal morphology, increased the activities of disaccharidases and enhanced intestinal mucosal antioxidant capacity, while regulating the expression of ferroptosis mediators in the piglets under oxidative stress. These findings suggested that dietary glycine supplementation improved the morphology and function of the intestinal mucosa, which was involved in regulating antioxidant capacity and ferroptosis.

Dietary glycine supplementation modulates antioxidant and immune responses of beluga, Huso huso, juveniles

Glycine is a non-essential amino acid used in fish nutrition, and possess several important roles in physiological process. Despite such functions, little is known about the glycine roles in fish, particularly in sturgeons. Therefore, the present study aimed at assessing the effects of dietary glycine supplementation on growth performance, antioxidant, and immunological parameters in beluga, Huso huso. Four hundred eighty fish (~ 48 g) were randomly distributed in 12 fiberglass tanks (1200 L) at a stocking density of 40 fish per tank. Four diets supplemented with 0% (control), 0.25% (Gly0.25), 0.5% (Gly0.5), and 1% (Gly1) glycine were offered to the fish (each diet to a batch of three tanks) for eight weeks at water temperature of 23.2 ± 0.70 °C. At the end of the feeding trial, the fish were blood-sampled for blood leukocyte count, plasma cortisol, glucose, triglyceride, cholesterol, reduced glutathione (GSH), glutathione reductase (GR), glutathione peroxidase (GPx), malondialdehyde (MDA), lysozyme, and alternative complement activity (ACH50) analysis. The results indicated there were no significant differences in the fish final weight (197–213 g; P = 0.511), weight gain (299–311%; P = 0.956) specific growth rate (2.46–2.52%/d; P = 0.944), feed conversion ratio (0.97–1.03; P = 0.863), protein efficiency ratio (2.29–2.45; P = 0.775), and survival (91.7–100%; P = 0.306) among the treatments. Plasma cortisol level significantly decreased in Gly0.25 treatment, compared to the control (24.7 vs. 28.8 ng/mL; P = 0.035). Plasma glucose levels exhibited significant (P < 0.001) elevation in Gly0.5 (51.0 mg/dL) and Gly1 (53.0 mg/dL) treatments, compared to the control (42.0 mg/dL). The treatment Gly0.5 showed significant elevations in blood leukocyte count (11.4 vs. 8.23 ×1000cell/µL; P = 0.032), plasma triglyceride (581 vs. 410 mg/dL; P = 0.017), cholesterol (71.3 vs. 59.8 mg/dL; P = 0.011), GSH (2.67 vs. 2.21 mM/mL; P = 0.003), GPx (243 vs. 217 U/mL; P < 0.001), and MDA (123 vs. 99 mM/L; P < 0.001), compared to the control treatment. Plasma GR (P < 0.001) showed significant decrease in Gly0.5 (1154 vs. 1784 U/mL) and Gly1 (1171 vs. 1784 U/mL) treatments. Besides, Gly0.25 and Gly0.5 treatments showed significantly higher plasma lysozyme (45.8–45.9 vs. 36.5 U/mL; P = 0.001) and ACH50 (138–139 vs. 130 U/mL; P < 0.001), compared to the control. Based on the results, dietary glycine supplementation at 0.25–0.5% could suppress plasma cortisol levels and improve antioxidant and immunological parameters on beluga, although has no beneficial effects on the fish growth performance.

Dietary glycine supplementation alleviates transportation-induced stress in common carp, Cyprinus carpio

Fish transportation is a stressful event in aquaculture industry, which causes crowding stress, oxidative stress, hypoxia, and water pollution stress. Therefore, it is necessary to work on the methods for mitigation of such stresses. Glycine has been known for its roles in the antioxidant system and ammonia detoxification in fish. Therefore, the present study aimed at assessing the effects of dietary glycine on growth performance, stress and antioxidant responses of common carp, Cyprinus carpio, during transportation. The fish were fed diets supplemented with 0 (G0), 2.5 (G2.5), 5 (G5), and 10 (G10) g/kg glycine for 8 weeks, then transported in plastic bags for 6 h. According to the results, dietary glycine induced no significant changes in growth performance of the fish. Plasma ammonia decreased, but plasma urea increased by dietary glycine supplementation before and/or after transportation. Dietary glycine significantly increased plasma glutathione (GSH) levels before transportation and suppressed GSH decrease after transportation, particularly at 5 and 10 g/kg. Glycine supplementation significantly suppressed plasma cortisol, glucose, superoxide dismutase, catalase, glutathione peroxidase, glutathione-S-transferase, and malondialdehyde after transportation, particularly at 5 and 10 g/kg. The present study demonstrates that glycine is a useful dietary supplement for common carp, as it improves antioxidant capacity and helps the fish during transportation. Such effects can be achieved by dietary glycine supplementation at 5 g/kg in common carp.

Effects of dietary glycine administration on biochemical responses to ammonia toxicity in common carp, Cyprinus carpio

The benefits of dietary glycine supplementation were investigated in common carp, Cyprinus carpio, following an acute ammonia challenge. Fish were fed diets supplemented with glycine (0, 2.5, 5 and 10 g/kg; named C, 2.5G, 5G and 10G, respectively) for 3 weeks, followed by 3-h exposure to 0.5 mg/L unionized ammonia nitrogen. Plasma immunoglobulin levels showed no changes in relation to ammonia challenge or dietary glycine supplementation. Dietary glycine significantly decreased plasma glucose, but increased plasma total antioxidant capacity (TAC) and glutathione (GSH). Ammonia exposure significantly increased plasma glucose and alternative complement, but decreased plasma GSH and TAC levels. Dietary glycine and ammonia exposure induced interaction effects on plasma ammonia, urea, cortisol, lysozyme, alanine aminotransferase (ALT), aspartate aminotransferase (AST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione-S-transferase (GST) and malondialdehyde (MDA). Before ammonia exposure, glycine administration significantly decreased plasma ammonia and urea; after ammonia exposure, glycine supplementation significantly mitigated hyperammonaemia, but stimulated uraemia. Dietary glycine induced no changes in plasma cortisol before ammonia challenge, but decreased it after the ammonia challenge. Dietary glycine had no significant effects on ALT, AST, SOD and MDA, decreased CAT, but increased GPx and GST, before ammonia exposure. Moreover, dietary glycine mitigated the ammonia-induced elevations in ALT, AST, SOD, CAT, GPx, GST and MDA. Among the treatments, 10G exhibited the lowest ammonia-induced changes in the tested parameters; thus, it is recommended to use 10 g glycine per kg diet for 3 weeks to mitigate ammonia-induced stress, oxidative stress and hepatic damages in common carp.

511 Late-Breaking: Dietary Glycine Supplementation During Growing and Finishing Phases Increases Body Weight and Improves Meat Quality of Intrauterine Growth Restricted Pigs

Abstract Glycine is required for maximum growth and development for growing pigs. Traditional swine diets are deficient in glycine. This is concerning for intrauterine growth restricted (IUGR) pigs (20–25% of all pigs born) as they are born with low bodyweights and underdeveloped skeletomuscular systems. Therefore, IUGR piglets are culled at birth, as they are unlikely to survive and unable to reach market weight at the same rate as normal birthweight (NBW) littermates. Additionally, IUGR pigs are not fed to market weight due to undesirable carcass attributes at slaughter such as increased subcutaneous fat deposition and decreased overall desirable protein yield. This study was designed to test the hypothesis that providing dietary glycine supplementation in traditional corn- and-soybean meal-based diets would improve the growth rate of IUGR pigs between weaning (age 21d) and market weight (age 188d). Fourteen IUGR pigs (birthweight = 0.98±0.03 kg, mean ± SEM) and 20 NBW pigs (birthweight = 1.44±0.02 kg, mean ± SEM) were obtained at weaning and used for this study. Pigs from each birthweight group were randomly assigned to 1% glycine + 0.19% corn starch treatment group or 1.19% alanine group (isonitrogenous control). Data were analyzed by using 2-way ANOVA and Duncan multiple comparison test. At 188 d of age, glycine supplemented IUGR pigs were 4.4 kg heavier (P &amp;lt; 0.05) than control IUGR pigs, and had similar (P &amp;gt;0.05) body weights to those NBW counterparts. Feed intake did not differ among the treatment groups (P &amp;gt;0.05). Additionally, glycine supplementation improved markers of meat quality in IUGR pig carcasses compared to control IUGR carcasses, including decreased backfat thickness, increased loineye area, and increased overall muscle score (P &amp;lt; 0.05). Overall, results of this study show that dietary glycine supplementation to IUGR pigs post-weaning has positive effects on lean tissue gain and meat quality. (Supported by USDA-NIFA)

510 Late-Breaking: Dietary Glycine Supplementation During Growing and Finishing Phases Increases Tissue Concentrations of Total Creatine and Gene Expression of Creatine-synthetic Enzymes in Low-birthweight Pigs

Abstract Dietary glycine is required for maximum growth and development in animals by stimulating muscle protein synthesis and as a component of creatine. Creatine is synthesized from glycine, arginine, and S-adenosylmethionine by arginine:glycine amidinotransferase (AGAT) and guanidinoacetate N-methyltransferase (GAMT). Sufficient creatine synthesis for growth requires adequate substrate supply. However, swine diets are deficient in glycine. Additionally, intrauterine growth restricted (IUGR) pigs have reduced glycine synthesis. This results in decreased creatine synthesis and lower total creatine content in tissues, leading to reduced cellular energy metabolism and diminished muscle protein accretion. This study was designed to test the hypothesis that dietary glycine supplementation in corn-and-soybean-meal-based diets would improve overall growth and skeletal muscle accretion in post-weaning IUGR pigs by increasing the expression of creatine-synthetic enzymes and tissue concentrations of total creatine. Fourteen IUGR pigs (birthweight = 0.98±0.03 kg, mean±SEM) and 20 normal birthweight pigs (birthweight = 1.44±0.02 kg, mean±SEM) were obtained at weaning for this study. Pigs from each birthweight group were randomly assigned to 1% glycine + 0.19% corn starch treatment group or 1.19% alanine group (isonitrogenous control) for the study (21 d to 188 d of age); tissues were collected at d 188. Glycine-supplemented IUGR pigs had greater tissue concentrations of creatine, creatinine, and creatine phosphate than control IUGR in all tissues measured (P&amp;lt; 0.05). Control IUGR pigs showed diminished activity and mRNA expression of creatine-synthetic enzymes (P &amp;lt; 0.05); this was mitigated by glycine supplementation as glycine supplemented IUGR pigs showed normal levels of enzyme activity and mRNA expression. Overall, results of this study indicate dietary glycine supplementation to IUGR pigs between weaning and market weight effectively restores creatine-synthetic enzyme activities and increase tissue concentrations of total creatine, leading to increased lean tissue growth. Muscle-Liver Trafficking of BCAA-Derived Nitrogen Underlies Obesity-Related Glycine Depletion

Glycine levels are inversely associated with branched-chain amino acids (BCAAs) and cardiometabolic disease phenotypes, but biochemical mechanisms that explain these relationships remain uncharted. Metabolites and genes related to BCAA metabolism and nitrogen handling were strongly associated with glycine in correlation analyses. Stable isotope labeling in Zucker fatty rats (ZFRs) shows that glycine acts as a carbon donor for the pyruvate-alanine cycle in a BCAA-regulated manner. Inhibition of the BCAA transaminase (BCAT) enzymes depletes plasma pools of alanine and raises glycine levels. In high-fat-fed ZFRs, dietary glycine supplementation raises urinary acyl-glycine content and lowers circulating triglycerides but also results in accumulation of long-chain acyl-coenzyme As (acyl-CoAs), lower 5' adenosine monophosphate-activated protein kinase (AMPK) phosphorylation in muscle, and no improvement in glucose tolerance. Collectively, these studies frame a mechanism for explaining obesity-related glycine depletion and also provide insight into the impact of glycine supplementation on systemic glucose, lipid, and amino acid metabolism.

Obesity increases hepatic glycine dehydrogenase and aminomethyltransferase expression while dietary glycine supplementation reduces white adipose tissue in Zucker diabetic fatty rats.

Obesity is associated with altered glycine metabolism in humans. This study investigated the mechanisms regulating glycine metabolism in obese rats. Eight-week-old Zucker diabetic fatty rats (ZDF; a type-II diabetic animal model) received either 1% glycine or 1.19% L-alanine (isonitrogenous control) in drinking water for 6weeks. An additional group of lean Zucker rats also received 1.19% L-alanine as a lean control. Glycine concentrations in serum and liver were markedly lower in obese versus lean rats. Enteral glycine supplementation restored both serum and hepatic glycine levels, while reducing mesenteric and internal white fat mass compared with alanine-treated ZDF rats. Blood glucose and non-esterified fatty acid (NEFA) concentrations did not differ between the control and glycine-supplemented ZDF rats (P > 0.10). Both mRNA and protein expression of aminomethyltransferase (AMT) and glycine dehydrogenase, decarboxylating (GLDC) were increased in the livers of obese versus lean rats (P < 0.05). In contrast, glycine cleavage system H (GCSH) hepatic mRNA expression was downregulated in obese versus lean rats, although there was no change in protein expression. These findings indicate that reduced quantities of glycine observed in obese subjects likely results from an upregulation of the hepatic glycine cleavage system and that dietary glycine supplementation potentially reduces obesity in ZDF rats.

Glycine improves peritoneal vasoreactivity to dialysis solutions in the elderly.

Background: Peritoneal dialysis solution (PDS) dilates peritoneal microvessels predominantly by the activation of the endothelial nitric oxide (NO) pathway. We made an incidental observation of decreased PDS-induced, NO-dependent peritoneal microvascular vasoreactivity in elderly rats naïve to PDS exposure. We hypothesized that this subordinate NO-mediated peritoneal microvascular vasoreactivity is caused by increased oxidative stress in the aged endothelium, which compromises NO bioavailability in the elderly, and that peritoneal microvascular vasoreactivity can be improved by the supplementation of antioxidant glycine to PDS. Methods: We studied PDS-mediated vasoreactivity of four intestinal visceral arterioles of different orders by in vivo intravital microscopy in weaned, adult, and elderly rats to (i) confirm subordinate vasoreactivity to PDS in elderly rats; (ii) restore vasoreactivity by glycine supplementation; and (iii) establish age as an independent risk factor for endothelial cell dysfunction. Results: In a crossover series, peritoneal microvascular vasoreactivity to PDS exposure was remarkably decreased in elderly rats. This subordinate vasoreactivity was completely restored by the supplementation of glycine to PDS. In a separate series, we assessed in situ endothelial cell function in weaned and adult rats using the cumulative acetylcholine concentration–response curves. Unlike the adults, the weaned rats demonstrated remarkable sensitivity and reactivity to cumulative acetylcholine concentrations, suggesting the dependency of endothelial cell function on age. Conclusion: Aging is an independent risk factor for peritoneal microvascular endothelial cell dysfunction. Endothelial function in the elderly can be recovered by reinforcing the bioavailability of endothelial-derived NO through glycine. Dietary glycine supplementation is a potential therapeutic strategy to decrease the burden of oxidative stress on the aged endothelium.
