Hypoglycemic Effect of Padina arborescens Extract in Streptozotocin-induced Diabetic Mice

This study investigated the hypoglycemic effect of the Padina arborescens extract in STZ-induced diabetic mice. Freeze-dried Padina arborescens were extracted with 80% methanol and concentrated for use in this study. The hypoglycemic effect was determined by inhibitory activities against α-glucosidase and α-amylase as well as the alleviation of postprandial blood glucose level. Padina arborescens extracts showed higher inhibitory activities than acarbose, a positive control against α-glucosidase and α-amylase. The IC50 values of Padina arborescens extracts against α-glucosidase and α-amylase were 0.26 and 0.23 mg/mL, respectively, which evidenced as more effective than observed with acarbose. The increase of postprandial blood glucose levels were significantly suppressed in the Padina arborescens extract administered group than the control group in the streptozotocin induced diabetic mice. Furthermore, the area under the curve (AUC) was significantly lowered via Padina arborescens extract administration in diabetic mice (p < 0.05). These results indicated that the Padina arborescens extract might be used as an inhibitor of α-glucosidase and α-amylase and delay absorption of dietary carbohydrates.

Quality assessment and anti-obesity activity of Stellaria media (Linn.) Vill

BackgroundObesity is recognized as a social problem, associated with serious health risks and increased mortality. Numerous trials have been conducted to find and develop new anti-obesity drugs through herbal sources to minimize side effects associated with the present anti-obesity drugs. The present study was designed to evaluate the quality control parameters, quantitative phytochemical analysis (total phenolic, total flavonoids and total saponin content), and the anti-obesity effect of lyophilized juice (LJ) of Stellaria media (Linn.) Vill. by employing in vitro and in vivo models.MethodsIn vitro studies were performed to evaluate the inhibitory activity of LJ on pancreatic amylase and lipase. The in vivo pancreatic lipase activity was evaluated by measurement of plasma triacylglycerol levels after oral administration of lipid emulsion to swiss albino mice. Furthermore, the anti-obesity effect of LJ was assessed at two doses, 400 mg/kg and 900 mg/kg body weight in mice fed a high-fat-diet with or without LJ for 6 weeks.ResultsThe LJ inhibited pancreatic amylase and lipase activity in vitro and elevated plasma triacylglycerol level in mice. LJ suppressed the increase in body weight, retroperitoneal adipose tissue, liver weights and serum parameters viz., total cholesterol, total triglyceride, LDL-cholesterol level at the dose of 900 mg/kg body weight of the mice fed with high fat diet. The total phenolic, flavonoid and saponin contents were found to be 0.26 mg/g, 1.4 mg/g and 1.19 μg/g respectively of LJ.ConclusionThe anti-obesity effects of LJ in high-fat-diet fed mice may be partly mediated through delaying the intestinal absorption of dietary fat and carbohydrate by inhibiting digestive enzymes.

Bioactive compounds extracted from Gamtae (Ecklonia cava) by using enzymatic hydrolysis, a potent α-glucosidase and α-amylase inhibitor, alleviates postprandial hyperglycemia in diabetic mice

This study was designed to investigate whether the brown alga gamtae (Ecklonia cava) may inhibit α-glucosidase and α-amylase activities, and alleviate postprandial hyperglycemia in streptozotocin-induced diabetic mice. For that purpose, we prepared an enzymatic hydrolysate from gamtae (EHG) by using the carbohydrase, Celluclast. EHG evidenced prominent inhibitory effect against α-glucosidase and α-amylase. The IC50 values of EHG against α-glucosidase and α-amylase were 0.62 and 0.59 mg/mL, respectively, which evidenced the higher activities than that of acarbose. EHG did not exert any cytotoxic effect in human umbilical vein endothelial cells (HUVECs) at various concentrations (from 0.25 to 2 mg/mL). The increase of postprandial blood glucose levels were significantly suppressed in the EHG administered group than those in the streptozotocin-induced diabetic or normal mice. Moreover, the area under curve (AUC) was significantly reduced via EHG administration (6,102 vs. 10,425 mg·min/dL) in the diabetic mice as well as it delays absorption of dietary carbohydrates. These result indicated that EHG might be a potent inhibitor for α-glucosidase and α-amylase.

Evaluation of Food–drug Interaction of Guava Leaf Tea

Guava leaf tea (GLT) contains guava leaf polyphenol (Gvpp), which regulates the absorption of dietary carbohydrate from the intestines. Borderline diabetics, who are at high risk of development of diabetes, take GLT to suppress a rapid increase of blood sugar level after meals. However, patients with diabetes in whom diabetic drugs or warfarin as a blood thinner are prescribed also take GLT with the expectation of glycemic control. Therefore, we studied whether GLT had potential for inhibition or induction of cytochrome P450 (CYP) and an influence on the action of warfarin. Extract of guava leaf (GvEx) consists of carbohydrate and polyphenols, which are Gvpp, quercetin, and ellagic acid. These polyphenols, but not GvEx, showed a certain level of inhibition of human-cDNA-expressed CYPs. In a comparison of GLT and grapefruit juice, GLT showed weaker inhibition of CYP activities and of midazolam 1'-hydroxylation than grapefruit juice. Furthermore, neither liver weight nor CYP3A expression in the liver was changed in rats that received GvEx for 90 days compared with the control group. When rats were concomitantly treated with GLT and warfarin, the prolongation of clotting time of blood by warfarin was not influenced. These data suggest that GLT is unlikely to interact with drugs.

Hypoglycemic Effect of Sargassum ringgoldianum Extract in STZ-induced Diabetic Mice

This study was designed to investigate whether Sargassum ringgoldianum extract may inhibit α-glucosidase and α-amylase activities, and alleviate postprandial hyperglycemia in streptozotocin-induced diabetic mice. The IC50 values of Sargassum ringgoldianum extract against α-glucosidase and α-amylase were 0.12 mg/mL and 0.18 mg/mL, respectively, which evidenced higher activities than those of acarbose. The blood glucose levels of the Sargassum ringgoldianum extract administered group were significantly lower compared to the control group in the streptozotocin-induced diabetic mice. Moreover, the area under the two-hour blood glucose response curve was significantly reduced and the absorption of dietary carbohydrates was delayed after administration of Sargassum ringgoldianum extract in the diabetic mice. Therefore, these results indicated that Sargassum ringgoldianum extract may help decrease the postprandial blood glucose level via inhibiting α-glucosidase.

Hypoglycemic effect of fermented soymilk added with bokbunja (Rubus coreanus Miquel) in diabetic mice

This study investigated the hypoglycemic effect of fermented soymilk extracts added with bokbunja (FSB) in streptozotocin (STZ)-induced diabetic mice. FSB was prepared via fermentation with Bacillus subtilis of soymilk added with bokbunja extract and its extraction with methanol. The FSB showed higher (p<0.05) inhibitory activities than fermented soymilk extracts (FS) against α-glucosidase. The IC50 values of 5% FSB for α-glucosidase was 0.55 mg/mL, which was superior to that of FS (0.77 mg/mL). The increase of postprandial blood glucose levels were more significantly suppressed in the 5% FSB administered group than control or FS group in the streptozotocin induced diabetic mice. Moreover, the area under curve (AUC) was significantly reduced via 5% FSB administration in the diabetic mice as well as it delayed absorption of dietary carbohydrates. Results from this study suggest that FSB may help decrease the postprandial blood glucose level via inhibiting α-glucosidase.

Influência do fumo na atividade da amilase salivar e na curva glicêmica

OBJETIVO: Determinar a atividade da amilase salivar e a relação com a glicemia, antes e após a ingestão de carboidratos em fumantes e não fumantes, uma vez que in vitro a exposição da saliva à fumaça do cigarro induz à redução da atividade da amilase salivar e poderia influenciar na absorção dos carboidratos da dieta. MÉTODOS: Foram avaliados voluntários fumantes (n=10) e não fumantes (n=10). Realizou-se coletas da saliva antes e após o fumo e determinou-se a glicemia antes e após a ingestão de 72g de carboidratos. Para glicemia usaram-se tempos de 0, 15, 30, 60, 120 minutos. A determinação da atividade da amilase salivar foi realizada por meio de kits comerciais. A glicemia foi determinada utilizando o aparelho Glicomiter (Accu-Chek-Roche). As análises estatísticas foram realizadas no software Sigmastat, utilizou-se o método Teste t pareado (p&lt;0,05). RESULTADOS: O aumento da glicemia aos 15, 30, 60 e 90 minutos foi de 3,9; 11,9; 34,8 e 22,7% para os não fumantes e 4,9; 6,5; 13,8 e 9,7% para os fumantes, respectivamente. No pico máximo de absorção tem-se uma glicemia de 21,0 % maior nos pacientes não fumantes. A atividade da amilase salivar antes e após alimentação apresentou-se 75,0% menor nos indivíduos fumantes. CONCLUSÃO: Estes resultados sugerem que o fumo inibe a amilase e influencia na diminuição da digestão/absorção de carboidratos, consequentemente na concentração de glicose sanguínea, reduzindo assim o aporte energético ingerido.

Dieckol isolated from Ecklonia cava inhibits α-glucosidase and α-amylase in vitro and alleviates postprandial hyperglycemia in streptozotocin-induced diabetic mice

This study was designed to investigate whether dieckol may inhibit α-glucosidase and α-amylase activities, and alleviate postprandial hyperglycemia in streptozotocin-induced diabetic mice. Dieckol isolated from Ecklonia cava, brown algae, evidenced prominent inhibitory effect against α-glucosidase and α-amylase. The IC50 values of dieckol against α-glucosidase and α-amylase were 0.24 and 0.66mM, respectively, which evidenced the higher activities than that of acarbose. Dieckol did not exert any cytotoxic effect in human umbilical vein endothelial cells (HUVECs) at various concentrations (from 0.33 to 2.69mM). The increase of postprandial blood glucose levels were significantly suppressed in the dieckol administered group than those in the streptozotocin-induced diabetic or normal mice. Moreover, the area under curve (AUC) was significantly reduced via dieckol administration (259 versus 483mmolmin/l) in the diabetic mice as well as it delays absorption of dietary carbohydrates. Therefore, these result indicated that dieckol might be a potent inhibitor for α-glucosidase and α-amylase.

The chemical and digestive properties of a soluble glucan from Agrobacterium sp. ZX09

A salt-tolerant strain Agrobacterium sp. ZX09 produced a high molecular mass, water-soluble extracellular polysaccharide (EPS) composed of d-glucose. By examining periodate oxidation, 1H NMR and 13C NMR spectra, it was proven that the EPS consisted of the following repeating unit: →3)-β- d-Glc p-(1 → 3)-[β- d-Glc p-(1 → 3)-β- d-Glc p-(1 → 3)]3-α- d-Glc p-(1 → 3)-α- d-Glc p-(1→. In vitro the EPS was indigestible by α-amylase, and strongly inhibited pancreatic α-amylase activity. Fasting mice fed on the EPS failed to increase blood glucose levels. Experimental diets containing the EPS suppressed steep increase in blood glucose concentration. These results suggest that the novel water-soluble glucan could be potentially useful for decreasing absorption of dietary carbohydrate and postprandial blood glucose level.

Effect of Evening Exposure to Bright or Dim Light after Daytime Bright Light on Absorption of Dietary Carbohydrates the Following Morning

We had previously reported on the effect of exposure to light on the human digestive system: daytime bright light exposure has a positive effect, whereas, evening bright light exposure has a negative effect on the efficiency of dietary carbohydrate absorption from the evening meal. These results prompted us to examine whether the light intensity to which subjects are exposed in the evening affects the efficiency of dietary carbohydrate absorption the following morning. In this study, subjects were exposed to either 50 lux (dim light conditions) or 2,000 lux (bright light conditions) in the evening for 9 h (from 15:00 to 24:00) after staying under bright light in the daytime (under 2,000 lux from 07:00 to 15:00). We measured unabsorbed dietary carbohydrates using the breath-hydrogen test the morning after exposure to either bright light or dim light the previous evening. Results showed that there was no significant difference between the two conditions in the amount of breath hydrogen. This indicates that evening exposure to bright or dim light after bright light exposure in the daytime has no varying effect on digestion or absorption of dietary carbohydrates in the following morning's breakfast.

NAPDH oxidase mediates glucolipotoxicity-induced beta cell dysfunction – Clinical implications

An impairment of glucose-stimulated insulin secretion--reflecting decreased glucokinase expression--and a moderate decrease in beta cell mass attributable to increased apoptosis, constitute the key features of beta cell failure in type 2 diabetes. Oxidative stress, provoked by prolonged exposure to excessive levels of glucose and/or fatty acids (glucolipotoxicity), appears to be a key mediator of these defects. Oxidant-provoked JNK activation induces nuclear export of the PDX-1 transcription factor, required for expression of glucokinase and other beta cell proteins. Conversely, increases in cAMP induced by incretin hormones promote the nuclear importation of PDX-1, counteracting the diabetogenic impact of oxidant stress; this may explain the utility of measures that slow dietary carbohydrate absorption for diabetes prevention. The ability of oxidative stress to boost apoptosis in beta cells is poorly understood, but may also entail JNK activation. Recent work establishes a phagocyte-type NADPH oxidase as the chief source of glucotoxicity-mediated oxidative stress in beta cells. Since bilirubin is now known to function physiologically as an inhibitor of NADPH oxidase, and phycocyanobilin (PCB) derived from spirulina likewise can inhibit this enzyme complex, supplemental PCB may have utility in the prevention and control of diabetes, and Gilbert syndrome, associated with chronically elevated free bilirubin, may be associated with decreased diabetes risk.

Diphlorethohydroxycarmalol isolated from Ishige okamurae, a brown algae, a potent α-glucosidase and α-amylase inhibitor, alleviates postprandial hyperglycemia in diabetic mice

This study was designed to investigate whether diphlorethohydroxycarmalol (DPHC) may inhibit α-glucosidase and α-amylase activities, and alleviate postprandial hyperglycemia in streptozotocin-induced diabetic mice. DPHC isolated from Ishige okamurae, a brown algae, evidenced prominent inhibitory effect against α-glucosidase and α-amylase. The IC 50 values of DPHC against α-glucosidase and α-amylase were 0.16 and 0.53 mM, respectively, which evidenced the higher activities than that of acarbose. DPHC did not exert any cytotoxic effect in human umbilical vein endothelial cells (HUVECs) at various concentrations (from 0.49 to 3.91 mM). The increase of postprandial blood glucose levels were significantly suppressed in the DPHC-administered group than those in the streptozotocin-induced diabetic or normal mice. Moreover, the area under curve (AUC) was significantly reduced via DPHC administration (2022 versus 2210 mmol∙min/l) in the diabetic mice as well as it delays absorption of dietary carbohydrates. Therefore, these result indicated that DPHC might be a potent inhibitor for α-glucosidase and α-amylase.

Molecular insights into dietary induced colic in the horse

Equine colic, a disorder manifested in abdominal pain, is the most frequent cause of emergency treatment and death in horses. Colic often requires intestinal surgery, subsequent hospitalisation and post operative care, with a strong risk of complications arising from surgery. Therefore strategies that explore approaches for preventing the condition are essential. To this end, a better understanding of the factors and mechanisms that lead to the development of colic and related intestinal diseases in the horse allows the design of preventive procedures. Colic is a multifactorial disorder that appears to be induced by environmental factors and possibly a genetic predisposition. One factor that seems to influence the risk of developing colic is the excessive consumption of diets containing high levels of carbohydrates. Therefore, major efforts have been made by various laboratories and institutions across the world to study the type and digestibility of various feed in order to formulate accurate and safe feed components and proportions. However, relatively little work has been carried out to characterise, in detail, the carbohydrate digestive and absorptive capacity and mechanisms underlying the potential adaptive response of equine gut epithelium to a changing diet. This review focuses on advances made towards understanding the molecular and cellular mechanisms involved in digestion and absorption of dietary carbohydrates in the equine gastrointestinal tract and the implication of these processes for the whole body physiology. It addresses the underlying mechanisms that may govern the adaptive response of equine small intestine to increased dietary hydrolysable carbohydrates. Furthermore, it describes changes that occur in the equine large intestinal microbiology and host tissue biology brought about by alterations in diet and in colic. It is hoped that a better understanding of the molecular and cellular processes that play important roles in the physiology and pathology of the equine gastrointestinal tract will assist the development of effective strategies to prevent equine colic.

Seasonal variation in gastric myoelectrical activity in young Japanese females

In order to test our hypothesis that there is seasonal variation in digestion and absorption of dietary carbohydrate from the intestine, we previously determined the amount of unabsorbed carbohydrate after breakfast by the breath hydrogen test in the four seasons. In pursuing our hypothesis further, we also recorded gastric myoelectrical activity before and after the breakfasts. In the current report, we describe the seasonality of gastric myoelectrical activity. Twenty-six Japanese female subjects were studied in winter, spring, summer and autumn. The cutaneous electrogastrogram was analysed by spectral analysis to compute the pre- and post-prandial dominant slow wave frequency (DF), and percentage of the 2 – 4 cpm gastric slow wave (Normal %). Two-factor ANOVA indicated that there was no significant seasonal variation in DF and Normal %. These results indicate that seasonal variations in digestion and absorption of dietary carbohydrate are caused by factors other than gastric and small intestinal motility.

Seasonal Variation in the Amount of Dietary Carbohydrate Not Absorbed from the Intestine after Breakfast in Elderly Japanese Females

It was previously shown that there is seasonality in the amount of dietary carbohydrate not absorbed from the intestine after breakfast, the amount of carbohydrate in winter being significantly larger than that in autumn in young Japanese subjects. In order to investigate this phenomenon further, the experiment was repeated on 22 elderly Japanese female subjects (61–78 yrs of age) during the four seasons of the year. The amount of unabsorbed dietary carbohydrate by the breath hydrogen test, which measures the amount of hydrogen in exhaled air, was then esitmated. A 6 g solution of lactosucrose, an indigestible trisaccharide, was used for comparison. Two groups of subjects, 16 subjects in Osaka and 6 subjects in Nagano, were studied in the summer (July to August) and autumn (October to November) of 2005 and the winter (January to February) and spring (April to May) of 2006. The following results were found using the pooled data of the total of 22 subjects. With regard to the amount of breath hydrogen excretion of the lactosucrose solution, there was no significant difference between the four seasons. There was a significant seasonal change in the efficiency of dietary carbohydrate absorption from the intestine after breakfast. The percentage of total carbohydrate that was not absorbed was lowest in the spring and highest in the winter. A comparison of the results from studies on the elderly and young subjects revealed the percentage of total carbohydrate that was not absorbed in the elderly was significantly lower than in the young in the winter, spring, and summer. These results indicate that there is seasonal variation in the efficiency of dietary carbohydrate absorption from the intestine among elderly female Japanese subjects as well as young female Japanese subjects. They also suggest that the efficiency of dietary carbohydrate absorption from the intestine after breakfast is retained in these naturally active and healthy elderly subjects.

Comparison of Carbohydrate Digestion between Japanese and Polish Healthy Subjects

We have revealed that light environment affects digestion and absorption of dietary carbohydrates in the gastrointestinal tract. This experimental result supposes that the efficiency of carbohydrate absorption may differ among people who live in different latitudes, such as Japanese and Polish people, at the same calendar season. In order to prove this hypothesis, we have been comparing the efficiency of carbohydrate absorption using the breath hydrogen test in Japan and Poland. Here, we report the comparison of the result obtained in the summer of 2004 as the following; (1) Orocecal transit time (OCTT) for indigestible trisaccharide of Japanese subjects was significantly longer than that in Poland (p = 0.043). (2) On the ingestion of minestrone, the amount of unabsorbed carbohydrate of Japanese subjects (which was estimated as trisaccharide equivalent) was significantly larger than that of Polish subjects (p = 0.006).

Seasonal Variation in Amount of Unabsorbed Dietary Carbohydrate from the Intestine after Breakfast in Japanese Subjects

We previously showed that daytime dim‐light exposure has a negative effect on the efficiency of dietary carbohydrate absorption in the evening, whereas evening‐time dim‐light exposure has a beneficial effect. These results suggest that seasonal changes in the environmental light may affect gastrointestinal activity, and that there might, therefore, be seasonal variation in the efficiency of dietary carbohydrate absorption from the intestine. In order to prove this hypothesis, we measured the amount of dietary carbohydrate unabsorbed from the intestine after a breakfast in healthy female Japanese subjects during the four seasons of the year. We estimated the amount of unabsorbed dietary carbohydrate by the breath hydrogen test, which measures the amount of hydrogen in exhaled air. A 6 g solution of lactosucrose, an indigestible trisaccharide, was used for comparison. Two groups of subjects, 12 subjects in Osaka and 14 subjects in Nagano, were studied in the winter (January to February), spring (April to May), summer (July to August), and autumn (October to November) of 2004. We found the following results: (1) In no season were there any significant differences between the two subgroups in the orocecal transit time of the breakfast and the lactosucrose solution. Nor were there any significant differences in the amount of unabsorbed dietary carbohydrate from the breakfast. (2) Using the pooled data of the total of 26 subjects, there was no significant seasonal variation in the orocecal transit time of the breakfast or the lactosucrose solution. (3) There was a significant seasonal variation in the amount of unabsorbed dietary carbohydrate from the breakfast. (4) The amount of unabsorbed dietary carbohydrate from the breakfast was largest in winter and smallest in autumn. Results in spring and in summer were similar and intermediate between those in winter and autumn. Post hoc multiple comparison tests showed that the amount of unabsorbed dietary carbohydrate in winter was significantly larger than in autumn. (5) In winter, the average ratio of the amount of unabsorbed dietary carbohydrate to the total amount of carbohydrate in the breakfast was about 12%; in autumn it was about 6%. These results clearly show that there is seasonal variation in the efficiency of intestinal dietary carbohydrate absorption among young female Japanese subjects.

A chlorogenic acid-induced increase in GLP-1 production may mediate the impact of heavy coffee consumption on diabetes risk

Recent prospective epidemiology links heavy coffee consumption to a substantial reduction in risk for type 2 diabetes. Yet there is no evidence that coffee improves insulin sensitivity and, at least in acute studies, caffeine has a negative impact in this regard. Thus, it is reasonable to suspect that coffee influences the risk for beta cell “failure” that precipitates diabetes in subjects who are already insulin resistant. Indeed, there is recent evidence that coffee increases production of the incretin hormone glucagon-like peptide-1 (GLP-1), possibly owing to an inhibitory effect of chlorogenic acid (CGA – the chief polyphenol in coffee) on glucose absorption. GLP-1 acts on beta cells, via cAMP-dependent mechanisms, to promote the synthesis and activity of the transcription factor IDX-1, crucial for maintaining the responsiveness of beta cells to an increase in plasma glucose. Conversely, the “glucolipotoxicity” thought to initiate and sustain beta cell dysfunction in diabetics can suppress expression of this transcription factor. The increased production of GLP-1 associated with frequent coffee consumption could thus be expected to counteract the adverse impact of chronic free fatty acid overexposure on beta cell function in overweight insulin resistant subjects. CGA's putative impact on glucose absorption may reflect the ability of this compound to inhibit glucose-6-phosphate translocase 1, now known to play a role in intestinal glucose transport. Delayed glucose absorption may itself protect beta cells by limiting postprandial hyperglycemia – though, owing to countervailing effects of caffeine on plasma glucose, and a paucity of relevant research studies, it is still unclear whether coffee ingestion blunts the postprandial rise in plasma glucose. More generally, diets high in “lente carbohydrate”, or administration of nutraceuticals/pharmaceuticals which slow the absorption of dietary carbohydrate, should help preserve efficient beta cell function by boosting GLP-1 production, as well as by blunting the glucotoxic impact of postprandial hyperglycemia on beta cell function.

A Soybean Peptide Isolate Diet Promotes Postprandial Carbohydrate Oxidation and Energy Expenditure in Type II Diabetic Mice

The aim of the present study was to determine the effects of dietary proteins on the oxidation of dietary carbohydrate and lipids in type II diabetic mice. KK-A(y) strain mice were provided free access to a high fat diet (30% of energy as fat) for an initial 4-wk period to induce diabetes. To reduce body weight gain, the mice were subsequently fed restrictive isoenergetic and isonitrogenous diets (35% of energy as protein and 5% as fat) based on either casein or soy protein isolate hydrolysate (SPI-H) for 4 wk. To measure exogenous carbohydrate and lipid oxidation, the mice were fed a diet containing (13)C-glucose or (13)C-triolein while they were in a respiratory chamber for 72 h. Postprandial energy expenditure was higher in the SPI-H than in the casein group; this difference was due to an increase in postprandial exogenous and endogenous carbohydrate oxidation. There were no differences in 24-h energy expenditure between dietary groups. Oxidation of exogenous carbohydrate tended to be higher (P = 0.054) in the SPI-H group during the 24 h of measurement. Fecal excretion of (13)C-glucose was lower but the excretion of lipid was higher in mice fed the SPI-H diet than in casein-fed mice. These results indicate that in type II diabetic mice, dietary SPI-H not only inhibits the absorption of dietary lipids and increases the absorption of dietary carbohydrates but also augments postprandial energy expenditure, which is accompanied by a postprandial increase in oxidation of dietary carbohydrates.

3-0 methylglucose absorption in vivo correlates with nutrient absorption and intestinal surface area in experimental short bowel syndrome.

Inert carbohydrate probes are widely used to study intestinal permeability and examine the passive uptake of markers. This study examined the use of quantifying 3-0 methylglucose (3-0 MG) absorption as a marker of intestinal surface area and active nutrient transport capability. Using a rat model, varying degrees of short bowel syndrome (SBS) were induced: sham operation (intestinal transection only), 50% resection (R50), 75% resection (R75), and 90% resection (R90; n = 6 to 8 in each group). Animals were pair fed, and over days 5 and 6 postoperation, a balance study was done to quantify absorption of dietary fat, protein, and carbohydrate. On day 7, animals were gavaged with the test solution containing 3-0 MG, lactulose, and mannitol, followed by a 16-hour urine collection. Urine recovery of probes was quantified using high-performance liquid chromotography. Animals were then killed, and the gross and microscopic intestinal morphology was determined. As expected, increased resection resulted in reduced absorption of dietary nutrients and 3-0 MG, which reached significance in the R90 resected animals. 3-0 MG absorption was significantly correlated with intestinal surface area and the absorption of dietary protein and fat (p < .01 for all comparisons). Interestingly, 3-0 MG absorption was not significantly correlated with the absorption of dietary carbohydrate. 3-0 MG absorption is a useful marker of functional intestinal surface area and of the absorption of dietary fat and protein in experimental SBS. 3-0 MG may be a useful marker of nutrient absorptive capacity in patients with SBS; further study is indicated.
