Jasmonate-insensitive mutant jar1b prevents petal elongation and flower opening coupling with parthenocarpic fruit development in Cucurbita pepo

Jasmonates are growth regulators that play a key role in flower development, fruit ripening, root growth, and plant defence. The study explores the coordination of floral organ maturation to ensure proper flower opening for pollination and fertilization. A new mutant (jar1b) was discovered, lacking petal elongation and flower opening but showing normal pistil and stamen development, leading to parthenocarpic fruit development. The mutation also enhanced the elongation of roots while reducing the formation of root hairs. BSA sequencing showed that jar1b is a missense mutation in the gene CpJAR1B, which encodes the enzyme that catalyzes the conjugation between JA and the amino acid isoleucine. The loss of function mutation in CpJAR1B produced a deficiency in biologically active (+) -7-iso-jasmonoyl-L-isoleucine (JA-Ile), which was not complemented by the paralogous gene CpJAR1A or any other redundant gene. Exogenous application of methyl jasmonate (MeJA) demonstrated that jar1b is partially insensitive to JA in both flowers and roots. Further experimentation involving the combination of JA-Ile deficient and ethylene-deficient, and ET insensitive mutations in double mutants revealed that CpJAR1B mediated ET action in female petal maturation and flower opening, but JA and ET have independent additive effects as negative regulators of the set and development of squash fruits. CpJAR1B also regulated the aperture of male flowers in an ethylene-independent manner. The root phenotype of jar1b and effects of external MeJA treatments indicated that CpJAR1B has a dual role in root development, inhibiting the elongation of primary and secondary roots, but promoting the formation of root hairs.

Cell wall invertase 3 plays critical roles in providing sugars during pollination and fertilization in cucumber.

In plants, pollen-pistil interactions during pollination and fertilization mediate pollen hydration and germination, pollen tube growth, and seed set and development. Cell wall invertases (CWINs) help provide the carbohydrates for pollen development; however, their roles in pollination and fertilization have not been well established. In cucumber (Cucumis sativus), CsCWIN3 showed the highest expression in flowers, and we further examined CsCWIN3 for functions during pollination to seed set. Both CsCWIN3 transcript and CsCWIN3 protein exhibited similar expression patterns in the sepals, petals, stamen filaments, anther tapetum, and pollen of male flowers, as well as in the stigma, style, transmitting tract, and ovule funiculus of female flowers. Notably, repression of CsCWIN3 in cucumber did not affect the formation of parthenocarpic fruit but resulted in an arrested growth of stigma integuments in female flowers and a partially delayed dehiscence of anthers with decreased pollen viability in male flowers. Consequently, the pollen tube grew poorly in the gynoecia after pollination. In addition, CsCWIN3-RNA interference plants also showed affected seed development. Considering that sugar transporters could function in cucumber fecundity, we highlight the role of CsCWIN3 and a potential close collaboration between CWIN and sugar transporters in these processes. Overall, we used molecular and physiological analyses to determine the CsCWIN3-mediated metabolism during pollen formation, pollen tube growth, and plant fecundity. CsCWIN3 has essential roles from pollination and fertilization to seed set but not parthenocarpic fruit development in cucumber.

Parthenocarpy-related genes induced by naphthalene acetic acid in oil palm interspecific O × G [Elaeis oleifera (Kunth) Cortés × Elaeis guineensis Jacq.] hybrids.

Parthenocarpy is the development without fertilization of seedless fruits. In the oil palm industry, the development of parthenocarpic fruits is considered an attractive option to increase palm oil production. Previous studies have shown the application of synthetic auxins in Elaeis guineensis, and interspecific O×G hybrids (Elaeis oleifera (Kunth) Cortés × E. guineensis Jacq.) induces parthenocarpy. The aim of this study was to identify the molecular mechanism through transcriptomics and biology system approach to responding to how the application of NAA induces parthenocarpic fruits in oil palm O×G hybrids. The transcriptome changes were studied in three phenological stages (PS) of the inflorescences: i) PS 603, pre-anthesis III, ii) PS 607, anthesis, and iii) PS 700, fertilized female flower. Each PS was treated with NAA, Pollen, and control (any application). The expression profile was studied at three separate times: five minutes (T0), 24 hours (T1), and 48h post-treatment (T2). The RNA sequencing (RNA seq) approach was used with 27 oil palm O×G hybrids for a total of 81 raw samples. RNA-Seq showed around 445,920 genes. Numerous differentially expressed genes (DEGs) were involved in pollination, flowering, seed development, hormone biosynthesis, and signal transduction. The expression of the most relevant transcription factors (TF) families was variable and dependent on the stage and time post-treatment. In general, NAA treatment expressed differentially more genes than Pollen. Indeed, the gene co-expression network of Pollen was built with fewer nodes than the NAA treatment. The transcriptional profiles of Auxin-responsive protein and Gibberellin-regulated genes involved in parthenocarpy phenomena agreed with those previously reported in other species. The expression of 13 DEGs was validated by RT-qPCR analysis. This detailed knowledge about the molecular mechanisms involved in parthenocarpy could be used to facilitate the future development of genome editing techniques that enable the production of parthenocarpic O×G hybrid cultivars without growth regulator application.

Propiconazole-induced brassinosteroid deficiency reduces female fertility by inhibiting female gametophyte development in woodland strawberry.

In woodland strawberry, a brassinosteroid biosynthesis inhibitor propiconazole induced typical brassinosteroid-deficient phenotypes and decreased female fertility due to attenuated female gametophyte development. Brassinosteroids (BRs) play roles in various aspects of plant development. We investigated the physiological roles of BRs in the woodland strawberry, Fragaria vesca. BR-level-dependent phenotypes were observed using a BR biosynthetic inhibitor, propiconazole (PCZ), and the most active natural BR, brassinolide (BL). Endogenous BL and castasterone, the active BRs, were below detectable levels in PCZ-treated woodland strawberry. The plants were typical BR-deficient phenotypes, and all phenotypes were restored by treatment with BL. These observations indicate that PCZ is an effective inhibitor of BR in woodland strawberry. Only one gene for each major step of BR biosynthesis in Arabidopsis is encoded in the woodland strawberry genome. BR biosynthetic genes are highly expressed during the early stage of fruit development. Emasculated flowers treated with BL failed to develop fruit, implying that BR is not involved in parthenocarpic fruit development. Similar to BR-deficient and BR-insensitive Arabidopsis mutants, female fertility was lower in PCZ-treated plants than in mock-treated plants due to failed attraction of the pollen tube to the ovule. In PCZ-treated plants, expression of FveMYB98, the homologous gene for Arabidopsis MYB98 (a marker for synergid cells), was downregulated. Ovules were smaller in PCZ-treated plants than in mock-treated plants, and histological analysis implied that the development of more than half of female gametophytes was arrested at the early stage in PCZ-treated plants. Our findings explain how BRs function during female gametophyte development in woodland strawberry.

Identification of a major QTL, Parth6.1 associated with parthenocarpic fruit development in slicing cucumber genotype, Pusa Parthenocarpic Cucumber-6.

Parthenocarpy is an extremely important trait that revolutionized the worldwide cultivation of cucumber under protected conditions. Pusa Parthenocarpic Cucumber-6 (PPC-6) is one of the important commercially cultivated varieties under protected conditions in India. Understanding the genetics of parthenocarpy, molecular mapping and the development of molecular markers closely associated with the trait will facilitate the introgression of parthenocarpic traits into non-conventional germplasm and elite varieties. The F1, F2 and back-crosses progenies with a non-parthenocarpic genotype, Pusa Uday indicated a single incomplete dominant gene controlling parthenocarpy in PPC-6. QTL-seq comprising of the early parthenocarpy and non-parthenocarpic bulks along with the parental lines identified two major genomic regions, one each in chromosome 3 and chromosome 6 spanning over a region of 2.7 Mb and 7.8 Mb, respectively. Conventional mapping using F2:3 population also identified two QTLs, Parth6.1 and Parth6.2 in chromosome 6 which indicated the presence of a major effect QTL in chromosome 6 determining parthenocarpy in PPC-6. The flanking markers, SSR01148 and SSR 01012 for Parth6.1 locus and SSR10476 and SSR 19174 for Parth6.2 locus were identified and can be used for introgression of parthenocarpy through the marker-assisted back-crossing programme. Functional annotation of the QTL-region identified two major genes, Csa_6G396640 and Csa_6G405890 designated as probable indole-3-pyruvate monooxygenase YUCCA11 and Auxin response factor 16, respectively associated with auxin biosynthesis as potential candidate genes. Csa_6G396640 showed only one insertion at position 2179 in the non-parthenocarpic parent. In the case of Csa_6G405890, more variations were observed between the two parents in the form of SNPs and InDels. The study provides insight about genomic regions, closely associated markers and possible candidate genes associated with parthenocarpy in PPC-6 which will be instrumental for functional genomics study and better understanding of parthenocarpy in cucumber.

Jasmonate-deficient mutant lox3a reveals crosstalk between jasmonate and ethylene in the differential regulation of male and female flower opening and early fruit development in Cucurbita pepo.

Jasmonate (JA) has been found to be a relevant hormone in floral development in numerous species, but its function in cucurbit floral development and sex determination is unknown. Crosstalk between JA and ethylene (ET) in the differential regulation of male and female floral development was investigated by using the novel JA-deficient mutant lox3a, and the ET-deficient and -insensitive mutants, aco1a and etr2b, respectively, of Cucurbita pepo. The lox3a mutation suppresses male and female flower opening and induces the development of parthenocarpic fruit. A bulked-segregant analysis coupled with whole genome sequencing and fine mapping approach allowed the identification of lox3a mutation in CpLOX3A, a LIPOXYGENASE gene involved in JA biosynthesis. The reduced JA content and expression of JA-signalling genes in male and female flowers of lox3a, and the rescue of lox3a phenotype by external application of methyl jasmonate (MeJA), demonstrated that JA controls petal elongation and flower opening, as well as fruit abortion in the absence of fertilization. JA also rescued the phenotype of ET mutants aco1a and etr2b, which are both specifically defective in female flower opening and fruit abortion. ET, the sex determining hormone of cucurbits, is induced in female flowers towards anthesis, activating JA production and promoting the aperture of the female flower, and the abortion of the unfertilized ovary. Given the close association between flower closure and parthenocarpic fruit development, we propose that flower opening can act as a switch that triggers fruit set and development in fertilized ovaries, but may alternatively induce the abortion of the unfertilized ovary. Both ET and JA from mature and senescent petals can serve as remote signals that determine the alternative development of the ovary and fruit.

Tomato POLLEN DEFICIENT 2 encodes a G-type lectin receptor kinase required for viable pollen grain formation.

Pollen development is a crucial biological process indispensable for seed set in flowering plants and for successful crop breeding. However, little is known about the molecular mechanisms regulating pollen development in crop species. This study reports a novel male-sterile tomato mutant, pollen deficient 2 (pod2), characterized by the production of non-viable pollen grains and resulting in the development of small parthenocarpic fruits. A combined strategy of mapping-by-sequencing and RNA interference-mediated gene silencing was used to prove that the pod2 phenotype is caused by the loss of Solanum lycopersicum G-type lectin receptor kinase II.9 (SlG-LecRK-II.9) activity. In situ hybridization of floral buds showed that POD2/SlG-LecRK-II.9 is specifically expressed in tapetal cells and microspores at the late tetrad stage. Accordingly, abnormalities in meiosis and tapetum programmed cell death in pod2 occurred during microsporogenesis, resulting in the formation of four dysfunctional microspores leading to an aberrant microgametogenesis process. RNA-seq analyses supported the existence of alterations at the final stage of microsporogenesis, since we found tomato deregulated genes whose counterparts in Arabidopsis are essential for the normal progression of male meiosis and cytokinesis. Collectively, our results revealed the essential role of POD2/SlG-LecRK-II.9 in regulating tomato pollen development.

Observation of unexpected neo like-fruit development from Cakile maritima calli

Parthenocarpy, the ability of some plants to undergo fruit growth in absence of fertilization, is an important question of basic science and the subject of much interest due to its possible agricultural benefits. In the context of our cellular biology studies on a halophyte of interest, Cakile maritima, we generated calli, pluripotent cell masses, that unexpectedly allowed the appearance of parthenocarpic fruits without any floral tissues. These observations raise the hope to develop an in vitro model to study parthenocarpic fruit development.

Application of naphthalene acetic acid and gibberellic acid favours fruit induction and development in oil palm hybrid (Elaeis oleiferaxElaeis guineensis)

SummaryThe OxG hybrid bunches contain more parthenocarpic fruits (PF) than normal fruits (NF) and present problems of development and ripening due to either an asynchronous opening of flowers or insufficient pollination. The objective of this study was to compare the effect of α-naphthaleneacetic acid (NAA) and gibberellic acid (GA3) in the induction and development of PF and NF, the fatty acid profile (FAP), and the oil potential in the ‘Coari x La Mé’ oil palm hybrid. NAA and GA3induced parthenocarpy in the fruits and did not alter the FAP of the mesocarp oil. The commercial dose of pollen (0.9 g talc + 0.1 g pollen) resulted in increased bunch weight (BW) (20.8 kg) and lower percentage of PF in the bunch (65.4%). The most effective hormonal doses to induce the formation of PF in the bunch were NAA 300 and 600 mg L-1. GA3alone or in mixture with NAA increased the percentage of PF but did not increase the BW, indicating that GA3had no synergistic effect on BW. The NAA applications represent alternatives to complement assisted pollination of OxG hybrids to increase bunch production and oil yield.

Induced Genetic Variations in Fruit Trees Using New Breeding Tools: Food Security and Climate Resilience.

Fruit trees provide essential nutrients to humans by contributing to major agricultural outputs and economic growth globally. However, major constraints to sustainable agricultural productivity are the uncontrolled proliferation of the population, and biotic and abiotic stresses. Tree mutation breeding has been substantially improved using different physical and chemical mutagens. Nonetheless, tree plant breeding has certain crucial bottlenecks including a long life cycle, ploidy level, occurrence of sequence polymorphisms, nature of parthenocarpic fruit development and linkage. Genetic engineering of trees has focused on boosting quality traits such as productivity, wood quality, and resistance to biotic and abiotic stresses. Recent technological advances in genome editing provide a unique opportunity for the genetic improvement of woody plants. This review examines application of the CRISPR-Cas system to reduce disease susceptibility, alter plant architecture, enhance fruit quality, and improve yields. Examples are discussed of the contemporary CRISPR-Cas system to engineer easily scorable PDS genes, modify lignin, and to alter the flowering onset, fertility, tree architecture and certain biotic stresses.

VviFSK and VviFTK, Two Novel Genes Encoding Putative Non-RD Receptor Kinases Associated with Reproductive Development in Grapevine

Although several genes homologous to those involved in the modulation of reproductive development in the model plant Arabidopsis thaliana have been identified in the Vitis vinifera genome, the regulatory network associated with pollen development, pollen tube elongation, and fecundation in grapevine is largely unknown. In Arabidopsis, receptor kinases play essential roles in pollen tube growth and guidance, leading to proper fertilization and fruit initiation. Comparing the transcriptomic profiles of flowers and early developing berries, two grapevine genes encoding proteins with structural domains corresponding to non-RD receptor kinases were identified. The first of them, VviFTK (Vitis vinifera Flower and Tendril Kinase), is transcribed in flowers at pre-anthesis and in tendrils and shares high sequence homology with At3g03770 gene from Arabidopsis which encodes a putative phloem-specific receptor kinase of unknown function. The second gene, VviFSK (Vitis vinifera Flower and Seed Kinase), is mainly expressed in flowers at anthesis stage and in immature seeds and codes for a protein with high similarity to ScORK17, a receptor kinase involved in the ovule and seed development regulation in Solanum chacoense. VviFSK shows different expression patterns in two cultivars with opposite tendency to parthenocarpic fruit development (PFD) and its transcription is induced in response to exogenously added sucrose. In concordance with the expression mode of VviFSK, the in silico analysis of its promoter region indicates the presence of cis regulatory sequences recognized by floral homeotic transcription factors as well as elements associated with seed-specific expression and sugar induction of gene transcription. These results suggest that VviFSK protein plays a role in the seed development process in grapevine.

Effect of the application of plant hormones on the formation of parthenocarpic fruits and oil production in oil palm interspecific hybrids (Elaeis oleifera Cortes x Elaeis guineensis Jacq.)

ABSTRACT Oil palm is the most important oil crop globally, accounting for almost 40% of fats and oil consumption worldwide. The OxG interspecific hybrid results from the crossing of African oil palm Elaeis guineensis Jacq. and the American oil palm Elaeis oleifera Cortes. It is a promising genotype for its high productivity, oil quality, and partial resistance to some diseases. These hybrids require assisted pollination, an activity that is time-consuming and expensive. Oil production in the OxG hybrids depends on the formation of normal (fertile) fruits and seedless (parthenocarpic) fruits. Thus, it is appealing to stimulate parthenocarpic fruits without assisted pollination. This research evaluated artificial methods of inducing parthenocarpy by applying plant hormones, and the effect on bunch formation and oil quality and its potential implementation at an industrial scale. The study was carried out in a 6-year-old hybrid (OxG) Coari x La Mé. The effects of the plant hormones indoleacetic acid (IAA), 2,4-dichlorophenoxyacetic acid (2,4-D), naphthalene acetic acid (NAA), and an ethylene precursor (ethephon) were evaluated. IAA and ethephon did not result in consistent bunch production of parthenocarpic fruits. The plants presented abnormal fruit growth, low-weight bunches, and high abortion rates. 2,4-D and NAA promoted proper parthenocarpic fruit development with oil-to-bunch (O/B) values similar to those obtained with assisted pollination. Thus, NAA and 2,4-D are an excellent alternative for oil production in the OxG hybrids that can be implemented in the African oil palm, with particular relevance that pollinators are endangered, and highly female genotypes are widely used.

The inhibition of SlIAA9 mimics an increase in endogenous auxin and mediates changes in auxin and gibberellin signalling during parthenocarpic fruit development in tomato

Parthenocarpic fruit formation can be achieved through the inhibition of SlIAA9, a negative regulator of auxin signalling in tomato plant. During early fruit development under SlIAA9 inhibition, cell division and cell expansion were observed. Bioactive gibberellin (GA) accumulated, but indole-3-acetic acid (IAA) and trans-zeatin did not accumulate substantially. Furthermore, under SlIAA9 inhibition, auxin-responsive genes such as SlIAA2, -3, and -14 were upregulated, and SlARF7 was downregulated. These results indicate that SlIAA9 inhibition mimics an increase in auxin. The auxin biosynthesis genes SlTAR1, ToFZY, and ToFZY5 were stimulated by an increase in auxin and by auxin mimicking under SlIAA9 inhibition. However, SlTAR2 and ToFZY2 were upregulated only by pollination followed by high IAA accumulation. These results suggest that SlTAR2 and ToFZY2 play an important role in IAA synthesis in growing ovaries. GA synthesis was also activated by SlIAA9 inhibition through both the early-13-hydroxylation (for GA1 synthesis) and non-13-hydroxylation (GA4) pathways, indicating that fruit set caused by SlIAA9 inhibition was partially mediated by the GA pathway. SlIAA9 inhibition induced the expression of GA inactivation genes as well as GA biosynthesis genes except SlCPS during early parthenocarpic fruit development in tomato. This result suggests that inactivation genes play a role in fine-tuning the regulation of bioactive GA accumulation.

Temperature-Dependent Compatible and Incompatible Pollen-Style Interactions in Citrus clementina Hort. ex Tan. Show Different Transglutaminase Features and Polyamine Pattern.

In clementine, failure of fertilization can result in parthenocarpic fruit development, which has several advantages, such as seedless fruit, longer shelf-life, and greater consumer appeal. Recently, S-RNases have been identified in Citrus grandis, thus revealing that the self-incompatibility (SI) reaction relies on the S-RNase gametophytic mechanism. The fundamental role of environmental factors, mostly temperature, in determining the numbers of pollen tubes reaching the ovary is also well established in Citrus. In the present work, temperature-dependent pollen–pistil interactions in C. clementina were analyzed, focusing on several morphological aspects, as well as on polyamine (PA) content and the activity and distribution of transglutaminase (TGase), both reported to be involved in the SI response in pear and in pummelo. Results clearly indicate that temperature contributed to a different activation of the SI response, which occurs at optimal temperature of 25°C but was by-passed at 15°C. TGase activity was stimulated during the SI response, and it localized differently in the compatible and incompatible interaction: in compatible pollinated styles, TGase localized inside the style canal, while it was detected all around it in incompatible crosses. TGase localization and activity were congruent with the levels of soluble and insoluble conjugated PAs and with morphological evidences, which highlighted cell wall modification occurring as a result of SI.

Pollination techniques in zucchini production in the presence of boron fertilization

Zucchini (Curcubita pepo L.), known in Brazil as “abóbora de moita”, belongs to the Cucurbitaceae family and is among the top ten vegetables produced and of highest economic value in Brazil. The present study aims to evaluate the efficiency of different forms of pollination in zucchini plantations subject to boron fertilization. The experimental design used was randomized blocks in a 4 x 2 factorial arrangement, with four replications. The treatments consisted of a combination of four pollination methods (fruiting induced with 2,4-dichlorophenoxyacetic acid (2,4-D); manual pollination; natural pollination by bees and control) and boron fertilization (absent and in the soil at a dose of 2 kg ha-1). The percentage of flowering was evaluated and, of the harvested squashes, the diameter, length and average weight of the commercial fruits (those uniform in terms of shape, size, and color) were recorded. The application of 2,4-D promoted the greatest fruit setting rate, diameter, length and mass in the squash. 2,4-D promoted the development of parthenocarpic fruits and can be used to promote yield gains in zucchini production.

Loss of function of the Pad-1 aminotransferase gene, which is involved in auxin homeostasis, induces parthenocarpy in Solanaceae plants

Fruit development normally occurs after pollination and fertilization; however, in parthenocarpic plants, the ovary grows into the fruit without pollination and/or fertilization. Parthenocarpy has been recognized as a highly attractive agronomic trait because it could stabilize fruit yield under unfavorable environmental conditions. Although natural parthenocarpic varieties are useful for breeding Solanaceae plants, their use has been limited, and little is known about their molecular and biochemical mechanisms. Here, we report a parthenocarpic eggplant mutant, pad-1, which accumulates high levels of auxin in the ovaries. Map-based cloning showed that the wild-type (WT) Pad-1 gene encoded an aminotransferase with similarity to Arabidopsis VAS1 gene, which is involved in auxin homeostasis. Recombinant Pad-1 protein catalyzed the conversion of indole-3-pyruvic acid (IPyA) to tryptophan (Trp), which is a reverse reaction of auxin biosynthetic enzymes, tryptophan aminotransferases (TAA1/TARs). The RNA level of Pad-1 gene increased during ovary development and reached its highest level at anthesis stage in WT. This suggests that the role of Pad-1 in WT unpollinated ovary is to prevent overaccumulation of IAA resulting in precocious fruit-set. Furthermore, suppression of the orthologous genes of Pad-1 induced parthenocarpic fruit development in tomato and pepper plants. Our results demonstrated that the use of pad-1 genes would be powerful tools to improve fruit production of Solanaceae plants.

PbGA20ox2 Regulates Fruit Set and Induces Parthenocarpy by Enhancing GA4 Content.

Fruit set and development occur following successful fertilization. Parthenocarpy, a valuable trait in some self-incompatible species, produces seedless fruit without fertilization. Gibberellin (GA) is a crucial hormone in fruit-set regulation and development. While investigating the development of parthenocarpy in pear (Pyrus bretschneideri Rehd.), we determined that GA 20-oxidases (GA20ox) may play key roles in seedless pear fruit development. Sequence analysis revealed three PbGA20ox genes: PbGA20ox1, PbGA20ox2, and PbGA20ox3. We analyzed the expression patterns of candidate genes and found that PbGA20ox2 levels significantly changed in pollinated fruits. Tissue-specific expression assays revealed that PbGA20ox2 is highly expressed in young fruit and leaves. Subcellular localization assays showed it was located in the cytoplasm, nucleus, and plasma membrane. Overexpressed PbGA20ox2 tomato plants were taller and had longer hypocotyls and internodes, and the emasculated flowers produced parthenocarpic fruit. In pear, the transient overexpression of PbGA20ox2 promoted fruit development and delayed the drop of nonpollinated fruit. Furthermore, the fruit of PbGA20ox2-overexpressing tomato and transient PbGA20ox2-overexpressing pear had increased GA4 (but not GA3 and GA1) contents. This result provided evidence that PbGA20ox2 was necessary for GA4-dependent pear fruit development. Our study revealed that PbGA20ox2 altered the GA biosynthetic pathway and enhanced GA4 synthesis, thereby promoting fruit set and parthenocarpic fruit development.

Morphological Features of the Anther Development in Tomato Plants with Non-Specific Male Sterility.

The study was devoted to morphological and cytoembryological analysis of disorders in the anther and pollen development of transgenic tomato plants with a normal and abnormal phenotype, which is characterized by the impaired development of generative organs. Various abnormalities in the structural organization of anthers and microspores were revealed. Such abnormalities in microspores lead to the blocking of asymmetric cell division and, accordingly, the male gametophyte formation. Some of the non-degenerated microspores accumulate a large number of storage inclusions, forming sterile mononuclear pseudo-pollen, which is similar in size and appearance to fertile pollen grain (looks like pollen grain). It was discussed that the growth of tapetal cells in abnormal anthers by increasing the size and ploidy level of nuclei contributes to this process. It has been shown that in transgenic plants with a normal phenotype, individual disturbances are also observed in the development of both male and female gametophytes. The reason for the developmental arrest of some ovules was the death of endosperm at different stages of the globular embryo. At the same time, noticeable hypertrophy of endothelial cells performing a secretory function was observed. In the ovules of transgenic plants with abnormalities, the endothelium forms a pseudo-embryo instead of the embryo sac, stimulating the development of parthenocarpic fruits. The data obtained in this study can be useful for a better understanding of the genetic and molecular mechanisms of cytoplasmic male sterility and parthenocarpic fruit development in tomatoes.

Effects of Foliar Application of Indol Butyric Acid(IBA), Gibberellic Acid (GA3) and Zinc (Zn) on Yield and Quality of Tomato

The yield and quality of tomato largely depend on soil and climatic conditions and also on variety. Plant growth regulators function as chemical messengers for intercellular communication. In tomato, different growth regulators and zinc play a pivotal role in germination, root development, branching, flower initiation, fruiting, lycopene development, synchronization and early maturation, parthenocarpic fruit development, ripening, TSS, acidity, seed production etcetera. The field experiment was conducted at Sher-e-Bangla Agricultural University Farm, Dhaka, Bangladesh during October 2017 to April, 2018 to evaluate the effect of foliar application of IBA, GA3 and Zn on yield and quality of tomato [1]. Single variety BARI Tomato-14 and foliar application of IBA 5 ml/L, GA3 5 ml/l and Zn were used to conduct this experiment. The experiment was laid out in Randomized Complete Block Design (RCBD) having three factors and replicated three times. Data were collected on chlorophyll content of leaf, number of flowers cluster per pant, number of fruit per plant, yield per hectare, Vitamin-C content and TSS% content. A statistically significant variation was recorded in terms of the above characters about yield and quality of tomato. All the characters shows maximum result in treatment of T7 (IBA+GA3+Zn).The treatment combination of T7 (IBA 5 ml / L, GA3 5 ml / L and Zn 1 kg/ha) and T4 (IBA 5 ml / L, GA3 5 ml / L) gave the maximum yield (100.00 t/ha) and the minimum yield (39.75 t/ha) was found from the treatment combination on T0 (control). The highest Vitamin C content (113.10 mg/100 gm) and highest TSS (%) (7.000) were found from T7 (IBA 5 ml / L, GA3 5 ml / L and Zn 1 kg/ha) than that of control under present experiment. So, it can be concluded that treatment combination of T7 (IBA 5 ml / L, GA3 5 ml / L and Zn 1 kg/ha) is the best for yield and quality of tomato and can be tested further under different field conditions.

Transcriptome analysis of oil palm inflorescences revealed candidate genes for an auxin signaling pathway involved in parthenocarpy.

Oil palm parthenocarpic fruits, which are produced without fertilization, can be targeted to increase oil content because the majority of the fruit is occupied by mesocarp, the part in which palm oil is stored. Consequently, gaining an understanding of the parthenocarpic mechanism would be instrumental for producing parthenocarpic oil palm. This study aims to determine effects of auxin treatment and analyze differentially expressed genes in oil palm pistils at the pollination/anthesis stage, using an RNA sequencing (RNA seq) approach. The auxin treatment caused 100% parthenocarpy when auxin was sprayed before stigmas opened. The parthenocarpy decreased to 55%, 8% and 5% when the auxin was sprayed 1, 2 and 3 days after the opening of stigmas, respectively. Oil palm plants used for RNA seq were plants untreated with auxin as controls and auxin-treated plants on the day before pollination and 1 day after pollination. The number of raw reads ranged from 8,425,859 to 11,811,166 reads, with an average size ranging from 99 to 137 base pairs (bp). When compared with the oil palm transcriptome, the mapped reads ranged from 8,179,948 to 11,320,799 reads, representing 95.85–98.01% of the oil palm matching. Based on five comparisons between RNA seq of treatments and controls, and confirmation using reverse transcription polymerase chain reaction and quantitative real-time RT-PCR expression, five candidate genes, including probable indole-3-acetic acid (IAA)-amido synthetase GH3.8 (EgGH3.8), IAA-amido synthetase GH3.1 (EgGH3.1), IAA induced ARG7 like (EgARG7), tryptophan amino transferase-related protein 3-like (EgTAA3) and flavin-containing monooxygenase 1 (EgFMO1), were differentially expressed between auxin-treated and untreated samples. This evidence suggests a pathway of parthenocarpic fruit development at the beginning of fruit development. However, more research is needed to identify which genes are definitely involved in parthenocarpy.
