Evaluation of Spirulina spp. Crude Extract Revealed Antimicrobial, Antioxidant and UV Photoprotective Properties

The antimicrobial, UV photoprotective and antioxidant capabilities of Spirulina spp. methanolic extracts were determined using Fourier transform infrared spectroscopy (FTIR) and gas chromatography-mass spectrometry (GC-MS). Notable compounds were reported to be 5 fatty acid methyl esters, 4- methoxyallocinnamic acid, 2-ethylhexyl esters and bumetrizole. Moreover, these metabolites contained functionalities including hydroxyl, alkyl and carbonyl, as supported by the FTIR result. After 24 h, there was no zone of inhibition in the antibacterial activity as measured by the agar well diffusion method at doses of 30, 50 and 70 g/mL. The ABTS radical scavenging assay revealed that the methanolic Spirulina spp. exhibited an inhibitory concentration of 1,047.91 g/mL for antioxidant activity and SPF values of 0.40, 0.23 and 0.93 were calculated using Mansur’s equation for concentrations of 30, 50 and 70 g/mL, respectively. The results of the study showed the potential of cyanobacterial extracts as a bioactive ingredient in cosmetic product development. HIGHLIGHTS Methanolic extraction of Spirulina spp. comprises 5 fatty acid methyl esters, 4-ethoxyallocinnamic acid 2-ethylhexyl esters and bumetrizole characterized by GC-MS. Constituents of the extract contain functional groups including –OH, –C–H and C=O. The extracts were classified as weak radical stabilizers, confirmed by ABTS assay and possessed UV photoprotective capacity confirmed by the in vitro SPF assay. GRAPHICAL ABSTRACT

Potential Bioactivities of Tamarind Seed Jellose at the Cellular Level for Cosmetic Product Development

In recent years, the utilization of tamarind seeds as a potential and sustainable ingredient in green cosmetics has gained significant interest. These seeds, previously considered by-products in various food industries, are now being recognized for their interesting value and wide range of bioactive compounds. This study aimed to deeply examine the potential biological activities and underlying molecular mechanisms of tamarind seed jellose (TJ), a natural polysaccharide derived from Tamarindus indica seeds, for various cosmetic applications. Tyrosinase, a key regulator of melanin synthesis and skin color, was the main focus of this study. Through a series of in vitro experiments on skin fibroblasts and B16 melanoma cells, the cytotoxicity, antioxidant activity, and melanogenesis inhibitory potential of tamarind seed jellose were evaluated. Notably, the results revealed that TJ had no obvious cytotoxic effects on skin fibroblast cells at any tested concentrations, ranging from 0 to 10 mg/mL. Interestingly, tamarind seed jellose effectively reduced melanin synthesis by inhibiting tyrosinase expression in a dose-dependent manner. In addition, TJ exhibited a promising antioxidant activity. Collectively, these findings highlight that TJ has the potential to serve as a safe and multi-functional ingredient for green cosmetic applications, offering the potential opportunity to repurpose waste for the beauty industry.

Patchouli Alcohol: A Potent Tyrosinase Inhibitor Derived from Patchouli Essential Oil with Potential in the Development of a Skin-Lightening Agent

The inhibitory effects of Pogostemon cablin essential oil (patchouli essential oil, PEO) and its primary bioactive compound, patchouli alcohol (PA), on tyrosinase and melanin were investigated in vitro and ex vivo. Treatment with PEO and PA significantly, as well as dose-dependently, reduced forskolin (FRK)-induced melanin biosynthesis, cellular tyrosinase activity, and tyrosinase (TYR) protein expression. However, the transcriptional levels of TYR and tyrosinase-related proteins (TRP-1 and TRP-2) remained unaffected. These results suggest that PEO and PA may directly interrupt tyrosinase enzyme activity, leading to a reduction in melanin biosynthesis. Further experiments supported this notion, revealing that both PEO and PA significantly and dose-dependently inhibited mushroom tyrosinase activity in both the monophenolase and diphenolase phases. Additionally, an in silico molecular docking analysis was performed, utilizing a homology model of human tyrosinase. In conclusion, these findings strongly suggest that patchouli essential oil and its primary bioactive component, patchouli alcohol, hold promise as potential treatments for hyperpigmentary skin conditions and in the development of cosmetic products designed to lighten the skin.

The Influence Of Perceived Quality On Repurchase Intention With The Mediating Customer Satisfaction And Perceived Value Of Cosmetic Products In Jabodetabek

The increasing development of cosmetic products leads to fierce business competition. Cosmetic products are a basic necessity for women and part of a woman's lifestyle to look beautiful and confident. Almost all women depend on cosmetic products. This study aims to determine the effect of perceived quality, customer satisfaction and perceived value on repurchase intention. This research was conducted on female customers aged 17 years and over who have experience more than 2 times buying cosmetic products for at least the last 6 months in Jabodetabek. The sampling method used purposive sampling with a total sample of 95 respondents. The number of statements given to respondents was 19 statement items. The results of this study indicate a significant effect on perceived quality on customer satisfaction and perceived quality on perceived value, as well as an insignificant effect on customer satisfaction on repurchase intention, perceived value on repurchase intention and perceived quality on repurchase intention.

Impact of a novel sugarcane straw extract-based ingredient on skin microbiota via a new preclinical in vitro model

During cosmetic product development, the impact on the skin microbiota needs to be evaluated, as it plays an important role in skin health. Clinical studies are frequently used to assess cosmetic effects, but are expensive, time-consuming and require finished ingredients, which may be a limiting factor. Thus, this study had two main objectives, the development of a preclinical in vitro model for the evaluation of the effect of cosmetic ingredients on the skin microbiota, and its use to evaluate a novel sugarcane straw extract-based ingredient. In the development of the preclinical in vitro model, the microbiota incubation time and atmospheric conditions were optimized. The model was validated using a benchmark ingredient with reported in vivo effects on skin microbiota. Then, the new sugarcane straw extract-based ingredient was tested in the model, using skin microbiota samples from 12 healthy female volunteers. The impact on microbial groups was analyzed using qPCR and next-generation sequencing. The best conditions for the in vitro model were 24 h incubation under aerobic conditions. Furthermore, the results obtained with the benchmark ingredient agreed with those obtained in vivo, thus validating our model. Sugarcane straw extract-based ingredient was found to have no effect on community α-diversity, however it appears to affect S. epidermidis. In conclusion, the developed model can be used as a tool to assess the impact of novel cosmetic ingredients on skin microbiota. Also, the novel ingredient seems to have an impact on commensal S. epidermidis.

Development of Anti-Aging Cream Preparations with Active Substances from Plant Extracts: Physicochemical Review and Potential Applications

Indonesia is known to be rich in biodiversity, including various types of plants that can be used as traditional medicines to treat various skin problems, including acne, skin lightening, and anti-aging. In optimizing efforts to overcome aging caused by free radicals, it is important to develop cosmetic formulations that are effective and comfortable to use. Cosmetics can be in various dosage forms, including gels, creams, powders, ointments, and lotions. It is necessary for the formulation and evaluation of anti-aging cream preparations containing active substances from plant extracts. The method used in this study is comparative research, which involves collecting data from various research journals via the internet. Literature research is conducted online through platforms such as Science Direct, ResearchGate, Google Scholar, and other journal sites. In addition, this study provides information on formulations of anti-aging creams containing active substances from plant extracts based on available scientific evidence in the literature. These findings indicate that the formula used in the development of this cream has the potential to be used in anti-aging products containing active substances from plant extracts. Although the results of this review show success in physicochemical stability, it is important to continue further research to test the effectiveness and safety of using this cream on human skin. More in-depth clinical trials will provide stronger evidence of the benefits and safety of this product. Thus, the development of cream preparations based on the results of this review provides a solid foundation for the development of cosmetic products that combine the natural properties of plants with excellence.

The aim was to identify and analyze the success factors of cosmetic product development and market launch processes and to provide a solution model. Identified factors prevailing in the development and launch of new products in the cosmetics industry sector and the connections between them. Investigated the influence of these factors on the success of new products in the market. The paper presents the results of a survey of companies in the Baltic cosmetics industry. Based on the results of the research, a model for the development and launch of new products for the cosmetics industry is presented.

Correlation between sensory and instrumental characterization of developed sunscreens containing grape seed extract and a commercial product.

In the development of cosmetic products, sensory evaluation is an important step in determining consumer acceptance before it is released on a market but is often time-consuming and costly. However, correlating sensory characteristics with instrumental parameters using multivariate techniques is a potential way to facilitate the development of cosmetic products. Sunscreen formulations varied in the content of grape seed extract (GSE) and ultraviolet (UV) filters, and benchmark products were characterized using sensory descriptive analysis and instrumental analysis. Principal Component Analysis (PCA) was applied to the panel's performance data to study how well the panelists performed compared to each other and to find an association between rheological and textural instrumental parameters of cream samples. Further, applying Partial Least Squares (PLS) regression analysis, the association between sensory attributes and instrumental parameters was analyzed. In addition, a preference for the sensory properties of the studied sunscreen products that are important for consumers living in Southeast Asia was assessed by PLS. In this study, both the sensory and instrumental properties of all tested formulations were described well by PCA. The practicality of PLS was confirmed by an established correlation between sensory attributes from the categories of appearance (glossiness), pick-up (integrity of shape, firmness, glossiness, stringiness), and after-feel (glossiness, spreadability, stickiness) with both rheological and textural parameters. Although the instrumental analysis could not completely replace sensory evaluation, a described method applying PLS can be used as an additional cost-effective and time-saving method during the development of cosmetic products. Moreover, PLS revealed that sunscreens with a light texture and glossy appearance providing smooth skin after-feel are likely to be preferred over the thicker formulations having a residual color appearance in Southeast Asia. Until a universal model is created, the cosmetics developers and companies can apply a described method of determining sensory properties from the instrumental parameters of their own products. Future studies will be worth exploiting the applicability of the PLS regression model on instrumental datasets predicting sensory characteristics of other sunscreen products.

Production of bacterial cellulose with antioxidant additive from grape residue with promising cosmetic applications

AbstractThe present study describes the development of a polymeric matrix based on bacterial cellulose (BC) additivated with grape skin residue extract by in situ and ex situ methods, to obtain a material with antioxidant and moisturizing characteristics for possible cosmetic applications. The samples developed by the different methods showed antioxidant activity, and this property was intensified with the increase of the extracted content used. Among the samples added by the in situ method, the produced with a fermentation medium containing only pure grape skin residue extract (100%v/v—G100) presented the highest value of antioxidant activity 35.6%. While for the produced by the ex situ method (HSG“ex situ”) the value was approximately 30.3%. Both antioxidant activity results were close to those found for the pure extract (38.5%), emphasizing its stability even after incorporation into the membrane. In addition, it was observed that other properties, such as high water retention capacities (around 98%), high porosity (around 85%), and low crystallinity, favor the development of cosmetic products, because they increase the moisturizing effect and the ability to incorporate active compounds. In addition, the use of residue from the grape juice extraction process is valuable inputs for BC additive with antioxidant properties.

Impacts of sun protection and skin care habits in the biophysical and morphological properties of young men skin.

Previous studies have demonstrated that UVB radiation may cause changes in the epidermal permeability barrier and in the stratum corneum hydration. It is also well known that sun exposure causes erythema, skin cancer, and other physiologic alterations. Furthermore, because of the cultural barrier, men usually apply less sunscreen. The objective of this study was to evaluate the cosmetic use and sun protection habits of young men, correlating their lifestyle with the biophysical and morphological skin characteristics, as well as to study how skin unprotected sun exposure can affect these features even in young people. 60 participants between 18 and 28years old were divided into two groups: with photoprotection habits (PP habits) and without photoprotection habits (No PP habits). They were questioned about their sunscreen and cosmetic products use. The skin parameters were evaluated using biophysical and skin imaging techniques. 60% of the participants did not apply sunscreen often, and 80% did not consume other skin care products. No PP habits group presented an increase in the transepidermal water loss-TEWL and sebum level, as well as a reduction in the stratum corneum water content and dermis echogenicity. The skin characteristics evaluated in this study were correlated with the sun protection habits and skin care behavior showing that men face some sociocultural barrier to start the use of cosmetic products. Thus, it is important to consider this information for the dermatological clinical and development of cosmetic products for men's skin.

Risk Management of New Cosmetic Product Development Based on Data Management of Visualization in Scientific Computing

With the continuous development of the social economy, people pay more and more attention to their own protection and maintenance, especially the quality of commodities in terms of usage of the commodity. As commonly used commodities, the quality of their new products in cosmetics is extremely important. In the rapidly developing cosmetics industry, in view of these needs and limitations, the data management method of visualization in scientific computing is introduced in this paper. Multiple risk factors in cosmetics development are established and sorted from the three perspectives of cosmetics development, production, and sales, by combining the new cosmetic product development process. The biggest risks are clarified, the corresponding cosmetic product risk assessment model is constructed, and the new cosmetic product development risk strategy and related management are realized. The simulation experiment results show that the data management method of visualization in scientific computing is effective and can support the risk management of new cosmetic product development.

Evaluation of the Bioactivities of Natural Phenolics from Mango (Mangifera indica Linn) Leaves for Cosmetic Industry Applications

Mango is one of the most important crops in the Philippines but there had been no local study on the possible utilization of its non-food parts such as the barks and leaves, which were reported to be rich in compounds with biological activities that are of significance in the development of cosmetic products (Masibo and He 2008; DA 2018). In this study, aqueous acetone extracts from leaves of the Philippine mango cultivars (“carabao,” “pico,” “apple mango, “sinaging,” and “sipsipin”) were investigated for their total phenolics content (TPC), phenolics composition, and biological activities – specifically, antioxidant as well as tyrosinase and elastase inhibitory properties. Results show that all mango leaf extracts had significant levels of TPC. Phenolic compounds such as mangiferin, gallic acid, quercetin 3-β-D-glucopyranoside, and kaempferol were all found to be present in the extracts with mangiferin as the predominant compound. All the extracts exhibited greater antioxidant capacity than the standard ascorbic acid, implying greater protection against skin damages due to free radicals. Also, all extracts exhibited greater inhibition on elastase than tocopherol, suggesting a greater anti-aging property. While only some extracts showed greater inhibition on tyrosinase than ascorbic acid, it did not surpass but gave comparable inhibitory activity with that of kojic acid. This implies that the extracts and kojic acid have comparable whitening capacities. Overall, the results affirm the great potential of the extracts of the local mango leaves as a cosmetic active ingredient

Development of a skin microbiome diagnostic method to assess skin condition in healthy individuals: Application of research on skin microbiomes and skin condition.

Skin microbiomes vary across individuals. They are known to play essential roles in maintaining homeostasis and preventing infectious pathogens. In recent years, cosmetic product development has begun to focus on the relationship between skin microbiomes and skin conditions. However, the statistical methods used in many studies include the standard t-test and small-scale correlation analysis, which do not take into account the internal correlation structure in data on skin microbiomes and skin features. In this study, we aimed to understand the relationship between skin microbiomes and skin features by analysing complex microbiomes and skin data. We obtained data on 19skin characteristics and skin microbiomes based on 16s ribosomal RNA (16S rRNA) gene analysis of 276healthy Japanese women. We then performed the principal component analysis (PCA), a method that takes into account the internal correlation structure, on 234 panels of them that did not contain outliers or missing values. We confirmed the relationship between skin microbiomes and skin features with principal component regression analysis and hierarchical clustering analysis (HCA). The principal component regression analysis showed strong relationships between skin microbiomes and sebum-related skin characteristics and skin pH. In the HCA, the female panel was classified into two major groups based on the skin microbiome. Furthermore, there were significant differences in sebum-related skin characteristics and the way skin condition changes with ageing between those groups, suggesting the possibility of measuring skin condition and age-related skin risk based on microbiome data. In addition, sebum-related characteristics differed significantly among middle-aged participants, suggesting a strong relationship between skin microbiomes and sebum-related characteristics. Analysis of skin condition and skin microbiome in Japanese women, taking into account the correlation between variables, showed that skin microbiome was significantly related to the number of pores and the amount of sebum. Furthermore, it was suggested that the skin condition and the way the skin condition changes with ageing may differ depending on the type of skin microbiome. The finding of a relationship between skin condition and skin microbiome suggests the possibility of proposing a new beauty method focusing on the skin microbiome in the future.

Comparative analysis of human facial skin microbiome between topical sites compared to entire face.

Skin is an essential outer barrier and supports the growth of commensal microorganisms that protects a host from the offense of foreign toxic organisms. With the rapid development of next-generation sequencing (NGS)-based applications, skin microbiome research for facial health care has reached industry growth, such as therapy and cosmetic product development. Despite the acceleration of skin microbiome research, experimental standardization protocol has not yet been established in the facial site and method of sampling. Thus, we aimed to investigate the differences in microbial composition at each facial site (cheek, mouth, forehead, and entire face) using comprehensive microbiome analysis. Twelve specimens from three men (four specimens per one person) were collected. The hypervariable regions (V3-V4) of the bacterial 16S rRNA gene were targeted for 16S amplicon library construction and classification of bacterial taxonomy. Skin microbial composition for all specimens was investigated, and the differences site-by-site in skin microbial composition were analyzed and evaluated by the various statistical tests. We were able to validate the independent correlation between the skin microbiome composition and the facial sites. The cheek site showed the highest alpha-diversity in richness and evenness scores compared to the forehead and mouth. The cheek and mouth sites showed a positive correlation (R2 value > 0.93) with the entire face, while the forehead sites were negatively correlated (R2 value < 0.2). Given the relative abundance based on statistical correlation analysis, we estimated that the cheek site could be considered an optimal topical site to replace the entire face. Our study suggests that skin microbiome profiling of four facial sites confirms that the cheek shows the most similar skin flora with the entire face. This study would be informative for preventing bias caused by sampling methods before researching and understanding skin cosmetics development or skin diseases.

Profiling cosmeceutical effects of various herbal extracts through elastase, collagenase, tyrosinase inhibitory and antioxidant assays

• Cotinus coggygria is one of the plant species in terms of cosmeceutical effect. • Active plant species usually have records against skin disorders. • Use of active extracts with different antioxidant mechanisms will be more effective. Plants have been a very rich resource for the development of potentially new drug candidates and also the development of cosmetic products for skincare. The skin produces free radicals or reactive oxygen species through repeated exposure to the sun and these molecules cause oxidative stress and inflammatory reactions in the dermal and epidermal layers of the skin. Matrix metalloproteinases are triggered by aging and in inflamed skin, such as photo-aged skin, skin damage, and wrinkles occurred by collapsing dermal proteins such as collagen and elastin. In the development of plant-based skin whitening products, the first target is to investigate the inhibitors of the tyrosinase enzyme, which provides melanin production. The ethanol extracts of 92 herbal extracts and propolis were tested for their elastase, collagenase, and tyrosinase inhibitory effects using ELISA microtiter assays. Their antioxidant effect was determined using DPPH, DMPD, and NO radical scavenging activity, metal-chelation capacity, FRAP, PRAP, and CUPRAC assays. Total phenolic and flavonoid contents of the extracts were also determined spectrophotometrically. Our results from the screening studies showed that the ethanol extracts prepared from the aerial parts of Lamium purpureum subsp. purpureum, the pericarpium of Garcinia mangostana and Pistacia vera , the flowers of Punica granatum, and the seeds of Vitis vinifera were effective against at least two of these enzymes and therefore, the plant species were selected for further studies.

Extracellular Vesicles from Korean Codium fragile and Sargassum fusiforme Negatively Regulate Melanin Synthesis.

Although various marine ingredients have been exploited for the development of cosmetic products, no previous study has examined the potential of seaweed extracellular vesicles (EV) in such applications. Our results revealed that EV from Codium fragile and Sargassum fusiforme effectively decreased α-MSH-mediated melanin synthesis in MNT-1 human melanoma cells, associated with downregulation of MITF (microphthalmia-associated transcription factor), tyrosinase and TRP1 (tyrosinase-related proteins 1). The most effective inhibitory concentrations of EV were 250 μg/ml for S. fusiforme and 25 μg/ml for C. fragile, without affecting the viability of MNT-1 cells. Both EV reduced melanin synthesis in the epidermal basal layer of a three-dimensional model of human epidermis. Moreover, the application of the prototype cream containing C. fragile EV (final 5 μg/ml) yielded 1.31% improvement in skin brightness in a clinical trial. Together, these results suggest that EV from C. fragile and S. fusiforme reduce melanin synthesis and may be potential therapeutic and/or supplementary whitening agents.

Quantitation of Acetyl Hexapeptide-8 in Cosmetics by Hydrophilic Interaction Liquid Chromatography Coupled to Photo Diode Array Detection

Bioactive peptides are gaining more and more popularity in the research and development of cosmetic products with anti-aging effect. Acetyl hexapeptide-8 is a hydrophilic peptide incorporated in cosmetics to reduce the under-eye wrinkles and the forehead furrows. Hydrophilic interaction liquid chromatography (HILIC) is the separation technique of choice for analyzing peptides. In this work, a rapid HILIC method coupled to photodiode array detection operated at 214 nm was developed, validated and used to determine acetyl-hexapeptide-8 in cosmetics. Chromatography was performed on a Xbridge® HILIC BEH analytical column using as mobile phase a 40 mM ammonium formate water solution (pH 6.5)-acetonitrile mixture 30:70, v/v at flow rate 0.25 mL min−1. The assay was linear over the concentration range 20 to 30 μg mL−1 for the cosmetic formulations and 0.004 to 0.007% (w/w) for the cosmetic cream. The limits of quantitation for acetyl hexapeptide-8 were 1.5 μg mL−1 and 0.002% (w/w) for the assay of cosmetic formulations and cosmetic creams, respectively. The method was applied to the analysis of cosmetic formulations and anti-wrinkle cosmetic creams.

169 First-in-human safety and mechanism of action (MOA) analyses of repeatedly dosed in vivo gene delivery for directed human type III collagen (COL3) expression in aesthetics

Due to the essential role collagen plays in skin biorejuvenation, collagen stimulation has been a focus of recent cosmetic product development. However, directed expression of full-length human COL3, produced by and secreted from the subject’s own cells, has not been explored clinically. To this end, we engineered KB301, a replication-defective HSV-1 gene therapy vector, for delivery of COL3. Preclinically, KB301 was shown to transduce clinically relevant skin cells and secrete COL3 in vitro and express dermally localized COL3 without toxicity in vivo.

Skin Moisturizing Effects of a Microneedle Patch Containing Hyaluronic Acid and Lonicerae flos

The utilization of cosmetic raw materials for medicinal purposes in a microneedle patch containing hyaluronic acid (HA) and Lonicerae flos ethanol extract (LEE) has increased based on their eco-friendly characteristics. The moisturizing effects of this microneedle patch were assessed on the forearms of 20 female participants, and our results showed an increase between 29.60% and 88.54% in skin moisture content at the same time points relative to the untreated group. Apart from confirming the safety of HA and Lonicerae flos, our findings may also promote the use of HA and medicinal herbs in relevant industries, indicating their potential as raw ingredients for the development of cosmetic products and topical dermatological drugs. The results suggested that HA, Lonicerae flos, and other combinations of chemical and natural products based on traditional oriental medicine could beneficially affect skin health.

Isolated isoquercitrin from Green ball apple peel inhibits photoaging in CCD-986Sk fibroblasts cells via modulation of the MMPs signaling.

Ultraviolet B (UVB) irradiation is viewed as the main factor of skin aging, associated with acceleration of elastin, collagen degradation and expression of matrix metalloproteinases (MMPs). Apples are one of the most commonly eaten fruits in the world, and isoquercitrin is the main active ingredient in new bred varieties "Green ball" apple. Therefore, we are studying the functionality of the active ingredient of apple, a natural raw material that does not have toxicity or sensitivity problems. The aim of this study, we scrutinized the effects of isoquercitrin on skin photoaging in UVB-exposed human fibroblasts (CCD-986Sk). To investigate the inhibition effect on photoaging factor regulation, isolated isoquercitrin were treated with UVB, which induces photoaging-related factors in CCD-986Sk fibroblast cells. Pro-inflammatory factors were measured by ELISA, Western blotting and real-time PCR. Isoquercitrin exhibited antioxidant activity and UVB-induced generation of photoaging-related factor inhibition without showing any toxicity. Anti-photoaging effect for protein levels using Isoquercitin was competent, of both the combate MMP-1 and MMP-9.
