Spectrophotometric determination of Rauwolfia alkaloids: estimation of reserpine in pharmaceuticals.

A simple, sensitive and economically viable spectrophotometric method for the determination of some Rauwolfia alkaloids (ajmaline, ajmalicine, reserpine and yohimbine-HCl) has been developed. The method involves the oxidation of Rauwolfia alkaloids by iron(III) and subsequent complexation of iron(II) with 1,10-phenanthroline, forming a red-colored complex having the maximum absorbance at 510 nm. The method is applied to the determination of reserpine in tablets of pharmaceutical formulations. The common excipients do not interfere with the proposed method. A statistical comparison of these results with those of a reported method shows good agreement and indicates no significant difference in the precision.

Flow injection spectrofluorimetric determination of reserpine in tablets by on-line acetone sensitized photochemical reaction

On-line photochemical reaction of reserpine in the presence of acetone was investigated. Acetone was found to speed up the on-line photochemical conversion of reserpine into an intensively fluorescent compound. Not only reaction acidity but also the acetate buffer concentration affected the on-line photochemical induced fluorescence signal. Based on the observation an automated flow injection photochemical fluorimetric approach was developed. An injected sample zone was carried by a water stream to be merged with a acetate buffer (pH 3.4) solution containing 0.02% acetone in a knotted PTFE reactor (KR), which was freely coiled around a 6-W low pressure mercury lamp. While passing the KR, reserpine was transformed into an intensively fluorescent compound. It was on-line detected in a flow-through cell at the emission wavelength of 490 nm and excitation wavelength of 386 nm. At optimized conditions, a detection limit 0.45 μg l −1 was achieved at a sampling rate of 90 h −1. Eleven determinations of a 0.5 mg l −1 reserpine standard solution gave a R.S.D. of 0.3%. The linear dynamic range of reserpine calibration curve was 0.01–0.75 mg l −1. The proposed method was applied to assay the reserpine content in tablets and to monitor the dissolution profile of reserpine tablets. Satisfactory results were obtained for both the assays and dissolution studies.

Determination of Reserpine in Tablets by Liquid Chromatography with Fluorescence Detection: Revised Procedure

A procedure is presented for the determination of reserpine in tablets by liquid chromatography (LC) that is a slight modification of a method presented in a previous publication. The sample is extracted with methanol, and solutions are filtered through filter paper. For LC, a 7.5 cm column is used; the mobile phase is methanol containing a small volume of an aqueous solution of the sodium salt of 1-pentanesulfonic acid. Detection is by fluorescence with 280 nm excitation and 360 nm emission. Two commercial samples containing 0.1 and 0.25 mg reserpine were analyzed. For each sample, 2 determinations were made on a group composite. Ten tablets were also analyzed individually. A linearity study was conducted, with solutions ranging in concentration from 80 to 120% of the amount present in the reference solution.

Collaborative Study of a Semiautomated Fluorometric Method for the Determination of Reserpine in Tablets

A semiautomated fluorometric method for the determination of resperpine in tablets was collaboratively studied by 7 laboratories. The method is a modification of the semiautomated method of Urbányi and Stober, which involves formation of a fluorogen with vanadium pentoxide. Collaborators were supplied with 3 composites, each from a different dosage level of commercial tablets. The results obtained agreed well with the AOAC manual fluorometric method; coefficients of variation ranged from 0.45 to 2.70%. The method has been adopted as official first action.

Semiautomated Method for the Determination of Reserpine in Tablets

Abstract A chloroform solution of reserpine tablets is automatically sampled, washed successively with aqueous solutions of citric acid and sodium bicarbonate, and diluted with methanol. The chloroform-methanol solution is split into 3 streams: one flows sequentially through 2 UV spectrophotometers, recording the absorbances at 268 and 295 nm; the other 2 streams are used for the colorimetric assay (addition of hydrochloric acid and sodium nitrite) and blank (hydrochloric acid only). Day-to-day relative standard deviations were 0.40–0.95%. Comparisons with Barkan’s chromatographic procedure and the USP XVII method are presented.

Abstract A method is described for the quantitative determination of reserpine in tablet formulations. A suitable amount of tablet material is triturated with dimethylsulfoxide (DMSO), incorporated onto Celite, and transferred to a chromatographic column containing acid, base, and water layers. The reserpine is eluted with chloroform and quantitated spectrophotometrically. Duplicate results from 16 collaborators averaged 100.7% recovery as determined by UV, 98.8% recovery determined by colorimetric analysis. The method is recommended for adoption as official first action.

Abstract A column partition procedure for the analysis of reserpine in tahlets is presented. The sample formulation is triturated with dimethyl sulfoxide (DM SO), incorporated on Celite, and transferred to a column containing hase, acid, and water layers. Recovery is quantitative; no degradation of reserpine by DM SO is evidenced.

A rapid and specific method for the analysis of reserpine was needed. It was ob-served that a fluorescent, greenish-yellow color was obtained by treating an alcoholic solution of reserpine with dilute sulfuric acid and sodium nitrite. This reaction has been adapted to the quantitative determination of reserpine. The method is applicable to the determination of reserpine in tablets, solutions, and elixirs. Similar alkaloids such as alstonine, rauwolscine, sarpagine, raunescine, deserpidine, and yohimbine do not interfere.

The Identification and Determination of Reserpine in Tablets
