A rapid method for the determination of stable hydrogen isotope ratios of acetic acid in vinegar.

Vinegar is an everyday condiment made from fermented grains or fruits. It contains acetic acid which is the main organic material produced by fermentation. Vinegar suffers from the authenticity problem of exogenous adulteration due to the indistinguishability of low-cost chemical sources of synthetic acetic acid from acetic acid produced by fermentation. It is necessary to establish a simple and rapid measurement technique. Determination was according to the total acid content of vinegar diluted with acetone to a certain concentration. Online separation and determination of acetic acid δD in vinegar were carried out using gas chromatography-pyrolysis-isotope ratio mass spectrometry. An HP-Plot/U column was selected for online separation of acetic acid and water with molecular sieve characteristics. At the same time, combined with the instrument blowback function to remove water. Dilute solvent acetone was treated with a molecular sieve to remove trace water. The reproducibility of this method is less than 3‰. The long-term stability is within a reasonable error range. The accuracy correlation coefficient is greater than 0.99. The δD values of acetic acid in vinegar (-264.5 ± 20.3‰) and from chemical sources (-30.5 ± 90.8‰) were obtained. A rapid method was developed for identification of different sources of acetic acid. These different sources of acetic acid exhibited significant hydrogen isotope distribution characteristics. Additionally, it was observed that the carboxyl hydrogen of acetic acid exhibited facile exchange with water. In future investigations, we aim to mitigate this interference.

Employing 11-Ketotestosterone as a Target Analyte for Adrenosterone (11OXO) Administration in Doping Controls.

Adrenosterone (Androst-4-ene-3,11,17-trione, 11OXO) is forbidden in sports according to the Prohibited List of the World Anti-Doping Agency. The administration of 11OXO may be detected by monitoring the urinary concentrations of its main human metabolites 11β-hydroxy-androsterone and 11β-hydroxy-etiocholanolone. Preliminary urinary concentration and concentration ratio thresholds have been established for sports drug testing purposes, but adaptations are desirable as the suggested limits would result in numerous suspicious findings due to naturally elevated concentrations and ratios. Recently, the metabolism of 11-oxo-testosterone (KT) was investigated in the context of anti-doping research, resulting in a preliminary urinary concentration threshold and a confirmation procedure based on the determination of carbon isotope ratios (CIRs). Gas chromatography coupled to isotope ratio mass spectrometry was employed to investigate the CIRs of selected steroids. As KT is also a metabolite of 11OXO, the developed protocols for KT have been tested to elucidate their potential to detect the administration of 11OXO after a single oral dose of 100 mg. In order to further improve the analytical approach, the threshold for urinary concentrations of KT was re-investigated by employing a reference population of n = 5232 routine doping control samples. Quantification of urinary steroids was conducted by employing gas chromatography coupled to triple quadrupole mass spectrometry. Derived from these, a subset of n = 106 samples showing elevated concentrations of KT was investigated regarding their CIRs. By means of this, potentially positive samples due to the illicit administration of 11OXO or KT could be excluded, and the calculation of reference population-derived thresholds for the concentrations and CIR of KT was possible. Based on the results, the urinary concentration threshold for KT is suggested to be established at 130 ng/mL.

Coprecipitation as a One-Step Se Separation for Determination of Isotope Ratios Completed with Revised Uncertainty Evaluation.

This study introduces a simplified purification method for analyzing 82Se/78Se isotope ratios in diverse natural samples using hydride generation MC-ICP-MS. Unlike the thiol resin method, which is time-consuming and sensitive to the concentrations of reagents used at individual stages, our proposed alternative is quicker, simpler, and robust. The procedure involves coprecipitation of selenium with iron hydroxide and dissolution in hydrochloric acid. Combining hydride generation and a second cleanup stage achieves sufficient purification for Se isotope ratio measurements. The method is efficient, taking 3-4 h after sample decomposition, utilizing common reagents [HCl, Fe(NO3)3, NH4Cl] without evaporation or clean lab conditions. Results on 82Se/78Se isotope ratios in various matrices are presented, comparing them with literature data. All isotopic results have been subjected to a newly proposed state-of-the-art approach to uncertainty estimation dedicated to isotope ratio measurements. The approach is based on applying Monte Carlo simulations with consideration of different samples' results normalized by the expected value. By doing that, we obtained estimated uncertainty for any Se sample with the influence of particular measurements on the final estimation included. We employ a Monte Carlo simulation-based uncertainty estimation approach for isotope ratio measurements, providing estimated uncertainty for each selenium sample.

Separation of Lipoproteins for Quantitative Analysis of 14C-Labeled Lipid-Soluble Compounds by Accelerator Mass Spectrometry.

To date, 14C tracer studies using accelerator mass spectrometry (AMS) have not yet resolved lipid-soluble analytes into individual lipoprotein density subclasses. The objective of this work was to develop a reliable method for lipoprotein separation and quantitative recovery for biokinetic modeling purposes. The novel method developed provides the means for use of small volumes (10-200 µL) of frozen plasma as a starting material for continuous isopycnic lipoprotein separation within a carbon- and pH-stable analyte matrix, which, following post-separation fraction clean up, created samples suitable for highly accurate 14C/12C isotope ratio determinations by AMS. Manual aspiration achieved 99.2 ± 0.41% recovery of [5-14CH3]-(2R, 4'R, 8'R)-α-tocopherol contained within 25 µL plasma recovered in triacylglycerol rich lipoproteins (TRL = Chylomicrons + VLDL), LDL, HDL, and infranatant (INF) from each of 10 different sampling times for one male and one female subject, n = 20 total samples. Small sample volumes of previously frozen plasma and high analyte recoveries make this an attractive method for AMS studies using newer, smaller footprint AMS equipment to develop genuine tracer analyses of lipophilic nutrients or compounds in all human age ranges.

Highly sensitive spectral measurement of rubidium isotopes using open multi-pass cell in tunable diode laser absorption spectroscopy

An open multi-pass cell was constructed to extend the interaction distance between the light and Rb atoms in the TDLAS measurement system. Compared with single-path direct absorption, the SNR of the system was improved by 3.8 times.

A new method was developed to separate In from geostandards of different matrices, and In recovery is estimated to 98% ± 2%. We measured In isotope compositions of these standards and found significant In isotopic variations in terrestrial samples.

Using Carbon Stable Isotopes to Study C3 and C4 Photosynthesis: Models and Calculations.

Stable carbon isotopes are a powerful tool to study photosynthesis. Initial applications consisted of determining isotope ratios of plant biomass using mass spectrometry. Subsequently, theoretical models relating C isotope values to gas exchange characteristics were introduced and tested against instantaneous online measurements of 13C photosynthetic discrimination. Beginning in the twenty-first century, laser absorption spectroscopes with sufficient precision for determining isotope mixing ratios became commercially available. This has allowed collection of large data sets at lower cost and with unprecedented temporal resolution. More data and accompanying knowledge have permitted refinement of 13C discrimination model equations, but often at the expense of increased model complexity and difficult parametrization. This chapter describes instantaneous online measurements of 13C photosynthetic discrimination, provides recommendations for experimental setup, and presents a thorough compilation of equations available to researchers. We update our previous 2018 version of this chapter by including recently improved descriptions of (photo)respiratory processes and associated fractionations. We discuss the capabilities and limitations of the diverse 13C discrimination model equations and provide guidance for selecting the model complexity needed for different applications.

Investigation of potential polyatomic interferences on uranium isotope ratio measurements for the LS-APGD-Orbitrap MS system

The determination of actinide (e.g., U and Pu) content and isotopics is of importance to the nuclear forensics and safeguards communities. However, in the analysis of environmental samples, such as those collected by the International Atomic Energy Agency, uranium measurements can be complicated by isobaric interferences from polyatomic variants of heavy elements (e.g., Pb). This leads to complex, time-consuming sample manipulations (i.e., separations) before determining isotope ratios. An alternative strategy to sample pretreatment is to use high-resolution mass spectrometric platforms during the analysis to fully resolve the uranium isotopes from potential polyatomic interferences, negating the need for prior chemical separation. The liquid sampling - atmospheric pressure glow discharge (LS-APGD) coupled with an Orbitrap mass spectrometer provides a high-resolution (>70,000 m/Δm at m/z 200) inorganic mass spectrometry platform. As a demonstration of the power of this instrumental platform, and indeed, the high-resolution approach in general, uranium isotope ratios were determined in the presence of elemental impurities (e.g., Pb, Pt, Ta, W) commonly encountered with environmental sample swipe analysis, without any prior treatment. Even at elemental impurity concentrations of 1000–5000× relative to uranium, no interference was observed with the 235U or 238U signal. In addition, the 235U/238U isotope ratio for the samples with the concomitants present are within 2 standard deviations of the values obtained without their addition, indicating that these impurities do not impact the determined uranium isotope ratio. These findings represent a significant first step in leveraging the high resolution of the LS-APGD-Orbitrap-MS to overcome isobaric and molecular interferences instead of relying on chemical separations.

Application of the Compound-Specific Stable Isotopes (CSSI) Technique to Evaluate the Contributions of Sediment Sources in the Panama Canal Watershed

Sedimentation processes have negative socioeconomic and environmental consequences. The Compound-Specific Stable Isotopes (CSSI) technique allows for the evaluation of sediment inputs associated with different land use changes in a study region. In the present work, this technique was used in the Alhajuela Lake sub-basin, within the Panama Canal Watershed. The role of the main soil contributors to the landscape (land uses, river, runoff, slope) relevant to the sediment load within the sub-basin of Alhajuela Lake was evaluated, and the relevant indicators in the landscape were selected in order to obtain the best representative sample. The contribution levels of three (3) representative land uses (Forest, Pasture and Sediment) in the study area were evaluated for the sediments present in sixteen (16) selected mixing points. The samples collected were subjected to the standard laboratory process to obtain the carbon chain isotopic values present in the fatty acids. The results of the determinations of the carbon chain fatty acid isotope ratios were evaluated using a Bayesian mixing model that takes into account the uncertainty present in the identified source values. According to the results obtained, the source identified as Sediment has a prominent contribution in most of the mixing points. The contributions of Forest land use are important in the mixing points located north of the study area. The contributions associated with Pasture land use are relevant in the points located in the proximity of this land use. The results suggest that landslides caused by high rainfalls events (Forest and Pasture sources) cause strong sedimentation to the north of Alhajuela Lake. At the same time, a high distribution of soils deposition is observed in the area surrounding Alhajuela Lake due to the strong presence of soils with Sediment source in these places. The results obtained are consistent with observations and measurements of the sediments accumulated in Alhajuela Lake between the years 2008 and 2012.

Novel Methods for Concomitant Determination of the Stable Zr Isotope Composition, Lu‐Hf Isotope Systematics and U‐Pb Age of Individual Zircons

The stable Zr isotope ratios in zircon yield a novel geochemical tracer that, together with the Lu‐Hf and U‐Pb radiogenic isotope systems, allows for a better understanding of the magmatic evolution of silicate melts. We present a solution‐based procedure for coupled stable Zr, Lu‐Hf and U‐Pb isotope ratio determinations for individual zircons using a 91Zr‐96Zr tracer purified from Hf and a late‐spiking protocol for Zr. This method yields high‐precision Zr and Hf isotope results while maintaining low blank levels for U‐Pb isotope and Lu/Hf ratio determinations. With a two‐fold improvement on the precision relative to previous solution‐based work, we report δ94ZrIPGP‐Zr values (deviation of the 94Zr/90Zr ratio in the sample relative to the IPGP‐Zr reference material) and associated intermediate precisions (2s) for the zircon reference materials 91500, Mud Tank, Plešovice and Penglai of ‐0.041 ± 0.015‰, 0.018 ± 0.013‰, 0.089 ± 0.020‰ and ‐0.117 ± 0.021‰, respectively. Furthermore, this method yields un‐biased δ94ZrIPGP‐Zr and 176Hf/177Hf results for 25‐ng Zr and 0.56‐ng Hf aliquots of the Mud Tank zircon with intermediate precisions (2s) of 0.027‰ and 1.7 ε (parts per ten thousand), respectively. Thus, the presented method is applicable for the analysis of extremely small and rare zircon grains.

Role of Transition Metals in Metal-Organic Frameworks as Nanoporous Ion Emitters for Thermal Ionization Mass Spectrometry.

Thermal ionization mass spectrometry is a powerful analytical technique that allows for precise determination of isotopic ratios. Analysis of low abundance samples, however, can be limited by the ionization efficiency. Following an investigation into a new type of metal-organic hybrid material, nanoporous ion emitters (nano-PIEs), devised to promote the emission of analyte ions and reduce traditional sample loading challenges, this work evaluates the impact that changing the metal in the material has on the ionization of uranium (U). Being derived from metal-organic frameworks (MOFs), nano-PIEs inherit the tunability of their parent MOFs. The MOF-74 series has been well studied for probing the impact various framework metals (i.e., Mg, Mn, Co, Ni, Cu, Zn, and Cd) have on material properties, and thus, a series of nano-PIEs with different metals were derived from this isoreticular MOF series. Trends in ionization efficiency were studied as a function of ionization potential, volatility, and work function of the framework metals to gain a better understanding of the mechanism of analyte ionization. This study finds a correlation between the analyte ionization efficiency and nano-PIE framework metal volatility that is attributed to its tunable thermal stability and degradation behavior.

Faraday detector uranium isotope ratio measurement: Insights from solution- and laser ablation-based sampling methodologies on the Neoma MC-ICP-MS

In this study we utilize an all-faraday cup detector configuration on the Neoma MC-ICP-MS to perform uranium isotope ratio determinations from certified reference materials and a nuclear fuel precursor material via solution and laser ablation based sampling methodologies. The goal in performing these measurements is primarily to demonstrate that the all-faraday cup method can produce highly precise and accurate isotope ratios even at relatively low signal intensity levels while also developing a preliminary understanding of how the complex interplay between different amplifier resistor levels (e.g. 1011 vs 1013 Ω) and integration times impacts the precision and accuracy of different types of measurements. Our results show that the faraday cups with 1013 Ω resistors in their amplifier feedback loops can be used to produce accurate and relatively precise isotope ratios for ion beams down to ∼5 K cps. However, at the lower end of the signal intensities observed in this study, longer integration times are necessary to mitigate the impact of ion beam instability that can occur during laser ablation based sampling. Despite this potential limitation, the all-faraday cup method is highly versatile as exemplified by the analysis of a collection of uranium reference materials with highly variable uranium isotopic compositions.

Determination of intermolecular and intramolecular isotopic compositions of low-abundance gaseous hydrocarbons using an online hydrocarbon gas concentration method

The stable isotopic composition of natural gas can be used to identify its origin and source. However, low concentrations of gaseous hydrocarbons in high-mature natural and shale gases hinder accurate determination of their compound- and position-specific isotopic compositions. In this study, an online C2+ hydrocarbon gas concentration system combined with gas chromatography–isotope ratio mass spectrometry (GC–IRMS) or gas chromatography–pyrolysis–gas chromatography–isotope ratio mass spectrometry (GC–Py–GC–IRMS) was developed to determine compound- and position-specific isotopic compositions of low-abundance gaseous hydrocarbons. The lower limit of the gas concentration required for isotope ratio determination using the online concentration system is 0.001% (0.003%) for compound-specific carbon (hydrogen) isotopes and 0.005% for position-specific carbon isotopes and is thus applicable to most natural gas samples. The online concentration technique does not cause significant isotopic fractionation effects, and the combination with GC–IRMS and GC–Py–GC–IRMS can accurately and precisely determine the compound-specific δ13C and δD values of low-content C2+ gaseous hydrocarbons and the position-specific δ13C values (δ13Ca, δ13Cb, and SP values) of propane in low-content propane samples, respectively. The application of our method to two natural gas samples from the Ordos and Sichuan basins further confirms that the online concentration method allows simple and rapid determination of the compound- and position-specific isotopic compositions of low-abundance gaseous hydrocarbons.

Initial Characterization and Optimization of the Liquid Sampling-Atmospheric Pressure Glow Discharge Ionization Source Coupled to an Orbitrap Mass Spectrometer for the Determination of Plutonium.

Plutonium measurements are essential to the nuclear forensics and safeguards community. The liquid sampling-atmospheric pressure glow discharge (LS-APGD) microplasma ionization source coupled with an Orbitrap mass spectrometer is a proven platform for uranium isotope ratio determinations. This work expands the LS-APGD-Orbitrap platform capabilities by reporting the first-ever analysis of plutonium with the LS-APGD and the first-ever measurement of elemental plutonium with an Orbitrap mass spectrometer. This coupling has the potential to dramatically reduce the complex sample manipulations required for traditional analysis techniques employed for actinide isotope ratio determinations. As a first step toward the goal of simultaneous uranium and plutonium isotope ratio determinations, the initial characterization and optimization of the platform for the detection of plutonium are reported. Collision-induced dissociation modality settings were optimized to reduce water-related and other molecular clusters containing plutonium, maximizing 242Pu16O2+ responses. A design of experiments study was conducted to optimize the discharge conditions of the dual-electrode LS-APGD toward the responsivity of 242Pu16O2+. The measurement sensitivity was determined from a Pu response curve, yielding a limit of detection of 10 fg (absolute) of total analyte when data was collected and processed with a Spectroswiss FTMS Booster X2 data acquisition system. Additionally, plutonium and uranium were measured in a simultaneous acquisition, and each analyte remained unaffected by the other. It is believed that the LS-APGD-Orbitrap platform could be a valuable addition to the nuclear forensics' toolbox and, indeed, other scientific disciplines and regulatory communities in which rapid, high-resolution plutonium determinations are paramount.

U mobilization and associated U isotope fractionation by sulfur-oxidizing bacteria.

Uranium (U) contamination of the environment causes high risk to health, demanding for effective and sustainable remediation. Bioremediation via microbial reduction of soluble U(VI) is generating high fractions (>50%) of insoluble non-crystalline U(IV) which, however, might be remobilized by sulfur-oxidizing bacteria. In this study, the efficacy of Acidithiobacillus (At.) ferrooxidans and Thiobacillus (T.) denitrificans to mobilize non-crystalline U(IV) and associated U isotope fractionation were investigated. At. ferrooxidans mobilized between 74 and 91% U after 1 week, and U mobilization was observed for both, living and inactive cells. Contrary to previous observations, no mobilization by T. denitrificans could be observed. Uranium mobilization by At. ferrooxidans did not cause U isotope fractionation suggesting that U isotope ratio determination is unsuitable as a direct proxy for bacterial U remobilization. The similar mobilization capability of active and inactive At. ferrooxidans cells suggests that the mobilization is based on the reaction with the cell biomass. This study raises doubts about the long-term sustainability of in-situ bioremediation measures at U-contaminated sites, especially with regard to non-crystalline U(IV) being the main component of U bioremediation.

A Marble Bust Newly Discovered by the Trapani Lombardo Family of Reggio Calabria (Southern Italy)

This research concerns a stylistic and archaeometric study of an ancient marble female bust recently discovered by the Trapani Lombardo family of Reggio Calabria (Southern Italy) and delivered to the Superintendency of Archaeology, Fine Arts, and Landscape for the metropolitan city of Reggio Calabria and Vibo Valentia (SABAP). Based on the first technical, stylistic, and iconographic observations made by the competent bodies, it is a half-length portrait bust from the Roman era, which precisely had the function of faithfully reproducing the physiognomy of the depicted subject. The research aimed to establish the authenticity of the artwork and the origin of the raw material, providing indications about the textural and compositional features and of the alteration products as well as identifying traces of any previous restoration interventions. For these purposes, after a preliminary assessment of the state of conservation of the bust using visual inspections supported by a handheld digital microscope, different analytical techniques, including polarized optical microscopy (OM), scanning electron microscopy coupled with energy dispersive spectrometry (SEM-EDX), carbon and oxygen stable isotope ratio determinations (δ13C and δ18O), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FT-IR), were used. The results highlighted the originality of the artifact, thus remarking on the importance of the precious archaeological find to be included in the cataloging of tangible assets in the panorama of Italian cultural heritage.
