Adiponectin-targeted SERS immunoassay biosensing platform for early detection of gestational diabetes mellitus

The anisotropic gold nanotriangles (AuNTs) were synthesized by a fast seedless growth process. The high-yield monodispersed AuNT colloids were obtained through a purification process based on depletion-induced interactions. AuNTs were modulated with a localized surface plasmon resonance (LSPR) peak of 638 nm wavelength coherent with the Raman excitation light. However, from finite element computation results, the AuNT clusters showed better performance for the 785 nm laser source due to a red shift in their LSPR properties, hence it was selected for the surface-enhanced Raman scattering (SERS) immunoassay. A self-assembly strategy using a thiol group and ON-OFF strategy in the heat map was performed to ensure the stability of SERS immunoassay platform. The sandwich SERS immunoassay biosensor platform for adiponectin detection demonstrated a wide assay range (10–15–10–6 g/mL), good reliability (R2 = 0.994, clinically relevant range), femto-scale limit of detection (3.0 × 10–16 g/mL), and excellent selectivity without interference from other biomarkers. This showed the possibility of effectively detecting adiponectin levels in the biofluids of pregnant women. Therefore, our technology is the first to quantitatively detect adiponectin based on SERS technology for early detection of gestational diabetes mellitus and has the potential to be used as a clinical biosensor capable of diagnosing various obstetric diseases during early pregnancy.

Unfolding DNA condensates produced by DNA-like charged depletants: A force spectroscopy study.

In this work, we have measured, by means of optical tweezers, forces acting on depletion-induced DNA condensates due to the presence of the DNA-like charged protein bovine serum albumin (BSA). The stretching and unfolding measurements performed on the semi-flexible DNA chain reveal (1) the softening of the uncondensed DNA contour length and (2) a mechanical behavior strikingly different from those previously observed: the force-extension curves of BSA-induced DNA condensates lack the "saw-tooth" pattern and applied external forces as high as ≈80 pN are unable to fully unfold the condensed DNA contour length. This last mechanical experimental finding is in agreement with force-induced "unpacking" detailed Langevin dynamics simulations recently performed by Cortini et al. on model rod-like shaped condensates. Furthermore, a simple thermodynamics analysis of the unfolding process has enabled us to estimate the free energy involved in the DNA condensation: the estimated depletion-induced interactions vary linearly with both the condensed DNA contour length and the BSA concentration, in agreement with the analytical and numerical analysis performed on model DNA condensates. We hope that future additional experiments can decide whether the rod-like morphology is the actual one we are dealing with (e.g. pulling experiments coupled with super-resolution fluorescence microscopy).

Depletion-induced forces and crowding in polymer-nanoparticle mixtures: Role of polymer shape fluctuations and penetrability.

Depletion forces and macromolecular crowding govern the structure and function of biopolymers in biological cells and the properties of polymer nanocomposite materials. To isolate and analyze the influence of polymer shape fluctuations and penetrability on depletion-induced interactions and crowding by nanoparticles, we model polymers as effective penetrable ellipsoids, whose shapes fluctuate according to the probability distributions of the eigenvalues of the gyration tensor of an ideal random walk. Within this model, we apply Monte Carlo simulation methods to compute the depletion-induced potential of mean force between hard nanospheres and crowding-induced shape distributions of polymers in the protein limit, in which polymer coils can be easily penetrated by smaller nanospheres. By comparing depletion potentials from simulations of ellipsoidal and spherical polymer models with predictions of polymer field theory and free-volume theory, we show that polymer depletion-induced interactions and crowding depend sensitively on polymer shapes and penetrability, with important implications for bulk thermodynamic phase behavior.

Influence of polymer shape on depletion potentials and crowding in colloid-polymer mixtures.

Depletion-induced interactions between colloids in colloid-polymer mixtures depend in range and strength on size, shape, and concentration of depletants. Crowding by colloids in turn affects shapes of polymer coils, such as biopolymers in biological cells. By simulating hard-sphere colloids and random-walk polymers, modeled as fluctuating ellipsoids, we compute depletion-induced potentials and polymer shape distributions. Comparing results with exact density-functional theory calculations, molecular simulations, and experiments, we show that polymer shape fluctuations play an important role in depletion and crowding phenomena.

Highly Stable and Sensitive Paper-Based Bending Sensor Using Silver Nanowires/Layered Double Hydroxides Hybrids.

Highly sensitive flexible piezoresistive materials using silver nanowires (AgNWs) composites have been widely researched due to their excellent electrical, optical, and mechanical properties. Intrinsically, AgNWs tend to aggregate in polymer matrix because of the intense depletion-induced interactions, which seriously influence the percolation threshold of the composites. In this study, we report a highly stable and sensitive paper-based bending sensor using the AgNWs and layered double hydroxides (LDHs) to construct a hybrid conductive network in waterborne polyurethane that is easy to destruct and reconstruct under bending deformation. The nonconductive 2D LDH nanosheets are embedded into AgNWs network and assist dispersion of AgNWs, which depends on the hydrogen bonding between the two nanostructures. The percolation threshold of the composites decreases from 10.8 vol % (55 wt %) to 3.1 vol % (23.8 wt %), and the composites reaches a very low resistivity (10(-4) Ω·cm) with a small amount of AgNWs (8.3 vol %) due to the dispersion improvement of AgNWs with the effect of LDH nanosheets. The as-prepared conductive composites with low percolation threshold can be manufactured on paper via various methods such as rollerball pen writing, inkjet printing, or screen printing. The bending sensor prepared by manufacturing the composites on paper shows low-cost, excellent conductivity, flexibility (>3000 bending cycles), sensitivity (0.16 rad(-1)), fast response (120 ms) and relaxation time (105 ms), and nontoxicity. Therefore, a simple but efficient wearable sensor is developed to monitor the human motions (such as fingers and elbow joints movements) and presents good repeatability, stability, and responsiveness, making the bending sensor possibly able to meet the needs in numerous applications for robotic systems.

Coarse-grained molecular dynamics simulations of depletion-induced interactions for soft matter systems.

Given the ubiquity of depletion effects in biological and other soft matter systems, it is desirable to have coarse-grained Molecular Dynamics (MD) simulation approaches appropriate for the study of complex systems. This paper examines the use of two common truncated Lennard-Jones (Weeks-Chandler-Andersen (WCA)) potentials to describe a pair of colloidal particles in a thermal bath of depletants. The shifted-WCA model is the steeper of the two repulsive potentials considered, while the combinatorial-WCA model is the softer. It is found that the depletion-induced well depth for the combinatorial-WCA model is significantly deeper than the shifted-WCA model because the resulting overlap of the colloids yields extra accessible volume for depletants. For both shifted- and combinatorial-WCA simulations, the second virial coefficients and pair potentials between colloids are demonstrated to be well approximated by the Morphometric Thermodynamics (MT) model. This agreement suggests that the presence of depletants can be accurately modelled in MD simulations by implicitly including them through simple, analytical MT forms for depletion-induced interactions. Although both WCA potentials are found to be effective generic coarse-grained simulation approaches for studying depletion effects in complicated soft matter systems, combinatorial-WCA is the more efficient approach as depletion effects are enhanced at lower depletant densities. The findings indicate that for soft matter systems that are better modelled by potentials with some compressibility, predictions from hard-sphere systems could greatly underestimate the magnitude of depletion effects at a given depletant density.
