Effect of Temperature on Photosynthetic Pigment Degradation during Freeze–Thaw Process of Postharvest of Celery Leaves

Celery (Apium graveolens L.) is a kind of green leaf vegetable with a large consumption demand in the food industry. It is a commonly used material in quick-frozen food stuffing such as dumplings and steamed stuffed. Fresh celery leaf blades and petioles are rich in photosynthetic pigments including chlorophyll and carotenoid, their contents are closely related to the quality of celery and its products. In order to explore the effects of freezing and thawing temperature and thawing time on the degradation of photosynthetic pigments in celery leaf blades and petioles, the changes in photosynthetic pigments during thawing storage were measured under different freezing and thawing temperatures. The results showed that lower freezing and thawing temperatures were beneficial to the preservation of photosynthetic pigments in celery leaf blades and petioles, and the loss of photosynthetic pigments enhanced with the increase in thawing temperature and thawing time. Under the cold storage condition of −80 °C, the loss rate of pigment substances can be reduced by nearly 20% compared with that of −18 °C, and −80 °C and 4 °C could be the best temperature combination of freezing and thawing. The content and degradation rate of photosynthetic pigments in celery leaf blades were higher than that in petioles during thawing, with a total chlorophyll loss rate reaching 35% during 6 to 12 h after thawing. The increase in temperature difference between freezing and thawing could aggravate the damage to the cell structure and the degradation of the pigment, as chlorophyll is more sensitive to temperature changes, and the degradation rate is significantly higher than that of carotenoids. From the perspective of delaying the degradation of photosynthetic pigments, the results of this study will provide potential references for the reasonable configuration of freezing and thawing temperatures in the process of storage and transportation of celery products.

Boosting algicidal efficiency of Alteromonas sp. FDHY-CJ against Skeletonema costatum through fermentation optimization

Algicidal bacteria exhibit promising potential against harmful algal blooms (HABs); however, their application has been limited due to their limited algicidal activity. This study demonstrates the enhanced algicidal activity of Alteromonas sp. FDHY-CJ bacteria against harmful Skeletonema costatum using a 5 L fermenter. Utilizing this refined framework increased the OD600 value and algal cell mortality by 6.50 and 2.88 times, respectively, compared to non-optimized culture cultivated in a flask using marine broth 2216E medium. The mechanism of action involves significant inhibition of algal photosynthetic efficiency with concurrent degradation of photosynthetic pigments. Relative to the non-optimized group, the optimized bacterial treatment led to a significant increase in H2O2 and MDA (malondialdehyde) by 19.54 and 4.22-fold, respectively, and resulted in membrane damage. The culture optimization procedure yielded effectual algicidal substances capable of considerably reducing the severity of S. costatum HABs through cell membrane disruption.

Efficient Inactivation and Removal of a Harmful Marine Algae—Heterosigma akashiwo—By UV-Assisted Permanganate Oxidation

Harmful algal blooms (HABs) caused by Heterosigma akashiwo are occurring in coastal waters frequently, posing a great risk to marine environments and subsequent treatment processes like desalination. UV-assisted permanganate oxidation (UV/KMnO4) is recognized as an innovative advanced oxidation process. This study investigated the inactivation and removal efficiencies of H. akashiwo cells by UV/KMnO4. Algal cells were effectively disintegrated into fragments by UV/KMnO4. Also, the degradation of photosynthetic pigments, membrane lipid peroxidation, and severe oxidative stress in algal cells was observed. The removal efficiency of algal cells reached 80.2% by 20 min of UV/KMnO4 oxidation, with a KMnO4 dosage of 5 mg L−1. In addition, the residual algal cells could be completely removed by a subsequent self-settling process, without an additional coagulation procedure. The fragmentation of algal cells caused by UV/KMnO4 may facilitate the formation of algal flocs, thereby improving the cell settleability. Furthermore, UV254 was significantly reduced by UV/KMnO4, which is expected to reduce the formation of disinfection byproducts and membrane fouling. This study elucidates that UV/KMnO4 can be a promising technique for the efficient treatment of harmful marine algae.

Differential transcriptome and metabolite profile with variable fluoride tolerance and altered genomic template stability in the identification of four fluoride-tolerant or fluoride-sensitive rice cultivars

Fluoride toxicity is the most commonly experienced challenge to rice plants due to irrigation with fluoride-polluted groundwater. In the present study, four hydroponically grown rice cultivars (Badshahbhog, Swarna, Sukumar and Shatabdi) were screened for fluoride tolerance on the basis of physiological and molecular parameters. Rice cultivars were maintained at 37 °C under 16/8 h light/dark photoperiodic cycle with 700 μmol photons m−2 s−1 intensity and relative humidity (50 %) in a plant growth chamber for 20 days. Treatment with NaF (50 mg L−1) caused considerable fluoride accumulation in Shatabdi which hindered its growth and triggered the formation of H2O2 and enhanced lipoxygenase and protease activity that eventually led to higher malondialdehyde formation and electrolyte leakage from the cells. This was accompanied with degradation of photosynthetic pigments (chlorophyll a, b, total chlorophyll, xanthophyll and carotenes) and inhibition in Hill activity. Higher formation of osmolytes (proline, amino acids and glycine betaine) and activity of enzymatic antioxidants (SOD, CAT, APX and GPX) reversed the effect of fluoride toxicity in Badshahbhog, concomitant with proper maintenance of the expression of gene concerned with photosynthetic mechanism (PsbA, PsbY, PsaH, PetC, PetH, Rbcs, Rca and rsp9) which preserved the photosynthetic machinery, providing sufficient energy to the seedlings to overcome the stressed conditions. RAPD profiling and GTS also established the superiority of Badshahbhog over the three cultivars, making it an efficient fluoride-tolerant variety.

UV-B radiation increased the sensitivity of Tibetan soil cyanobacterium Loriellopsis cavernicola to the herbicide glyphosate

The high concentrations of herbicide and UV-B radiation are two stresses for Tibetan soil microorganisms, but there is limited information about the combined effects of herbicide and UV-B radiation on their levels of stress. In this study, the Tibetan soil cyanobacterium Loriellopsis cavernicola was used to investigate the combined inhibitory effect of the herbicide glyphosate and UV-B radiation on the cyanobacterial photosynthetic electron transport through an analysis of the photosynthetic activity, photosynthetic pigments, chlorophyll fluorescence and antioxidant system activity. The results demonstrated that treatment with herbicide or UV-B radiation and the combination of both stresses caused a decrease in the photosynthetic activity, interfered with the photosynthetic electron transport, and caused the accumulation of oxygen radicals and the degradation of photosynthetic pigments. In contrast, the combined treatment of glyphosate and UV-B radiation had a synergistic effect, i.e., the sensitivity of cyanobacteria to glyphosate increased in the presence of UV-B radiation, which caused the photosynthesis of cyanobacteria to have a greater impact. Since cyanobacteria are the primary producers of soil ecosystems, a high intensity of UV-B radiation in the plateau areas could enhance the inhibition of glyphosate on cyanobacteria, which could affect the ecological health and sustainable development of plateau soils.

Physiological and Transcriptomic Analyses of the Effects of Exogenous Lauric Acid on Drought Resistance in Peach (Prunus persica (L.) Batsch).

Peach (Prunus persica (L.) Batsch) is a fruit tree of economic and nutritional importance, but it is very sensitive to drought stress, which affects its growth to a great extent. Lauric acid (LA) is a fatty acid produced in plants and associated with the response to abiotic stress, but the underlying mechanism remains unclear. In this study, physiological analysis showed that 50 ppm LA pretreatment under drought stress could alleviate the growth of peach seedlings. LA inhibits the degradation of photosynthetic pigments and the closing of pores under drought stress, increasing the photosynthetic rate. LA also reduces the content of O2-, H2O2, and MDA under drought stress; our results were confirmed by Evans Blue, nitroblue tetrazolium (NBT), and DAB(3,3-diaminobenzidine) staining experiments. It may be that, by directly removing reactive oxygen species (ROS) and improving enzyme activity, i.e., catalase (CAT) activity, peroxidase (POD) activity, superoxide dismutase (SOD) activity, and ascorbate peroxidase (APX) activity, the damage caused by reactive oxygen species to peach seedlings is reduced. Peach seedlings treated with LA showed a significant increase in osmoregulatory substances compared with those subjected to drought stress, thereby regulating osmoregulatory balance and reducing damage. RNA-Seq analysis identified 1876 DEGs (differentially expressed genes) in untreated and LA-pretreated plants under drought stress. In-depth analysis of these DEGs showed that, under drought stress, LA regulates the expression of genes related to plant-pathogen interaction, phenylpropanoid biosynthesis, the MAPK signaling pathway, cyanoamino acid metabolism, and sesquiterpenoid and triterpenoid biosynthesis. In addition, LA may activate the Ca2+ signaling pathway by increasing the expressions of CNGC, CAM/CML, and CPDK family genes, thereby improving the drought resistance of peaches. In summary, via physiological and transcriptome analyses, the mechanism of action of LA in drought resistance has been revealed. Our research results provide new insights into the molecular regulatory mechanism of the LA-mediated drought resistance of peach trees.

NO and GSH Alleviate the Inhibition of Low-Temperature Stress on Cowpea Seedlings.

Low-temperature stress in early spring seriously affects the growth and development of cowpea seedlings. To study the alleviative effect of the exogenous substances nitric oxide (NO) and glutathione (GSH) on cowpea (Vigna unguiculata (Linn.) Walp.) seedlings under 8 °C low-temperature stress, 200 μmol·L-1 NO and 5 mmol·L-1 GSH were sprayed on cowpea seedlings whose second true leaf was about to unfold to enhance the tolerance of cowpea seedlings to low temperature. Spraying NO and GSH can eliminate excess superoxide radicals (O2-) and hydrogen peroxide (H2O2) to varying degrees, reduce the content of malondialdehyde and relative conductivity, delay the degradation of photosynthetic pigments, increase the content of osmotic regulating substances such as soluble sugar, soluble protein, and proline, and improve the activity of antioxidant enzymes such as superoxide dismutase, peroxidase, catalase, ascorbate peroxidase, dehydroascorbate reductase, and monodehydroascorbate reductase. This study revealed that the mixed use of NO and GSH played an important role in alleviating low temperature stress, and the effect of spraying NO alone was better than that of spraying GSH.

A new targeted approach of postharvest accumulation of anthocyanin in fragrant leaves of Melissa officinalis L

Postharvest accumulation of anthocyanin in aromatic herbs is not well studied but holds potential for diverse application. Lemon balm (Melissa officinalis L.) an aromatic herb known for its lemon fragrance is widely used in traditional medicine as well as in herbal tea. M. officinalis is typically propagated by stem cutting. While attempting to induce roots in water from detached leaf bases, temporal changes in leaf colouration from green to red was observed, indicative of anthocyanin accumulation. Quantitative distinction of colouration as well as from the L* a* b* values generated support change of leaf colour from green to red. The detached leaves were observed to retard senescence during the colour-changing phase for up to eight weeks. However, the time-course accumulation of anthocyanin was found to be strictly leaf size dependent; highest accumulation of anthocyanin (1156.72 ± 36.92 µg/g) noted 49 days after postharvest treatment using leaf size of 5 cm. High-performance liquid chromatographic analysis revealed the presence of cyanidin glycosides as the major anthocyanin that accumulate in detached leaves upon postharvest treatment; presence of cyanidin moiety was further confirmed by acid hydrolysis. Degradation of photosynthetic pigments were also observed during the postharvest treatment process. Moreover, the content of citral, the major terpene volatile remained unchanged while enhanced accumulation of the phenolic marker compound rosmarinic acid was observed during the process. These anthocyanic leaves (49 days postharvest treated) showed higher tyrosine and elastase inhibitory activities and enhanced free-radical scavenging capacities as compared with green leaves. This new, yet-so-simple technology for value addition in lemon balm leaves holds promise especially in traditional medicine, colourant and fragrance sectors.

Photocatalytic degradation effect and mechanism of Karenia mikimotoi by non-noble metal modified TiO2 loading onto copper metal organic framework (SNP-TiO2@Cu-MOF) under visible light.

In this study, the SNP-TiO2@Cu-MOF composite was prepared successfully by loading non-noble metal modified TiO2 (SNP-TiO2) on the surface of copper metal organic skeleton (Cu-MOF), and compared the inactivation efficiency of different photocatalysts to Karenia mikimotoi (K. mikimotoi) under visible light. The obtained photocatalyst had the characteristic crystal faces of Cu-MOF and SNP- TiO2, and contained functional groups such as Cu-O, -COOH, N-O, P-O, etc., which indicated the structural stability of the photocatalyst. The band gap of SNP-TiO2@Cu-MOF composite was 2.82eV, and it had great light absorption ability in visible light region. It was proved to be a mesoporous adsorption material, which had a huge specific surface area (245m2/g). Compared with other photocatalysts, SNP-TiO2@Cu-MOF composite showed the strongest photocatalytic activity. When the concentration of composite material was set to 100mg/L and the exposure time was 6h, the visible light photocatalytic inactivation efficiency of K. mikimotoi was 93.75 %. By measuring various metabolic indexes of K. mikimotoi under the action of different photocatalysts for 1h, it was confirmed that cell inactivation was due to the increased membrane permeability and degradation of photosynthetic pigments and main life proteins. This research showed that SNP-TiO2@Cu-MOF composite material was full of great potential and application prospect in controlling the outbreak of eutrophication.

Comparison of lauric acid and 12-hydroxylauric acid in the alleviation of drought stress in peach (Prunus persica (L.) Batsch).

Water shortage is a key factor that can restrict peach tree growth. Plants produce fatty acids and the fatty acid derivatives lauric acid (LA) and 12-hydroxylauric acid (LA-OH), which are involved in abiotic stress responses, but the underlying stress response mechanisms remain unclear. In this study, physiological examination revealed that in Prunus persica (L.) Batsch, pretreatment with 50 ppm LA-OH and LA reduced drought stress, efficiently maintained the leaf relative water content, and controlled the relative conductivity increase. Under drought stress, LA-OH and LA treatments prevented the degradation of photosynthetic pigments, increased the degree of leaf stomatal opening and enhanced the net photosynthetic rate. Compared with drought stress, LA-OH and LA treatment effectively increased the net photosynthetic rate by 204.55% and 115.91%, respectively, while increasing the Fv/Fm by 2.75% and 7.75%, respectively, but NPQ decreased by 7.67% and 37.54%, respectively. In addition, the level of reactive oxygen species increased under drought stress. The content of O2 - in LA-OH and LA treatment decreased by 12.91% and 11.24% compared to CK-D, respectively, and the content of H2O2 decreased by 13.73% and 19.94%, respectively. At the same time, the content of malondialdehyde (MDA) decreased by 55.56% and 58.48%, respectively. We believe that the main reason is that LA-OH and LA treatment have improved the activity of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). The application of exogenous LA increased the levels of soluble sugars, soluble proteins, proline and free amino acids under drought stress, and maintained the osmotic balance of cells. Compared with CK-D treatment, it increased by 24.11%, 16.89%, 29.3% and 15.04%, respectively. At the same time, the application of exogenous LA-OH also obtained similar results. In conclusion, exogenous LA-OH and LA can alleviate the damage to peach seedlings caused by drought stress by enhancing the photosynthetic and antioxidant capacities, increasing the activities of protective enzymes and regulating the contents of osmotic regulators, but the molecular mechanism is still in need of further exploration.

Recovery from Salinity and Drought Stress in the Perennial Sarcocornia fruticosa vs. the Annual Salicornia europaea and S. veneta.

Current agricultural problems, such as the decline of freshwater and fertile land, foster saline agriculture development. Salicornia and Sarcocornia species, with a long history of human consumption, are ideal models for developing halophyte crops. A greenhouse experiment was set up to compare the response of the perennial Sarcocornia fruticosa and the two annual Salicornia europaea and S. veneta to 30 days of salt stress (watering with 700 mM NaCl) and water deficit (complete withholding of irrigation) separate treatments, followed by 15 days of recovery. The three species showed high tolerance to salt stress, based on the accumulation of ions (Na+, Cl−, Ca2+) in the shoots and the synthesis of organic osmolytes. These defence mechanisms were partly constitutive, as active ion transport to the shoots and high levels of glycine betaine were also observed in non-stressed plants. The three halophytes were sensitive to water stress, albeit S. fruticosa to a lesser extent. In fact, S. fruticosa showed a lower reduction in shoot fresh weight than S. europaea or S. veneta, no degradation of photosynthetic pigments, a significant increase in glycine betaine contents, and full recovery after the water stress treatment. The observed differences could be due to a better adaptation of S. fruticosa to a drier natural habitat, as compared to the two Salicornia species. However, a more gradual stress-induced senescence in the perennial S. fruticosa may contribute to greater drought tolerance in this species.

Physiological and Biochemical Traits of Two Major Arabidopsis Accessions, Col-0 and Ws, Under Salinity.

Salinity affects plant growth and development as shown with the glycophyte model plant, Arabidopsis thaliana (Arabidopsis). Two Arabidopsis accessions, Wassilewskija (Ws) and Columbia (Col-0), are widely used to generate mutants available from various Arabidopsis seed resources. However, these two ecotypes are known to be salt-sensitive with different degrees of tolerance. In our study, 3-week-old Col-0 and Ws plants were treated with and without 150 mM NaCl for 48, 72, or 96 h, and several physiological and biochemical traits were characterized on shoots to identify any specific traits in their tolerance to salinity. Before salt treatment was carried out, a different phenotype was observed between Col-0 and Ws, whose main inflorescence stem became elongated in contrast to Col-0, which only displayed rosette leaves. Our results showed that Col-0 and Ws were both affected by salt stress with limited growth associated with a reduction in nutrient uptake, a degradation of photosynthetic pigments, an increase in protein degradation, as well as showing changes in carbohydrate metabolism and cell wall composition. These traits were often more pronounced in Col-0 and occurred usually earlier than in Ws. Tandem Mass Tags quantitative proteomics data correlated well with the physiological and biochemical results. The Col-0 response to salt stress was specifically characterized by a greater accumulation of osmoprotectants such as anthocyanin, galactinol, and raffinose; a lower reactive oxygen detoxification capacity; and a transient reduction in galacturonic acid content. Pectin degradation was associated with an overaccumulation of the wall-associated kinase 1, WAK1, which plays a role in cell wall integrity (CWI) upon salt stress exposure. Under control conditions, Ws produced more antioxidant enzymes than Col-0. Fewer specific changes occurred in Ws in response to salt stress apart from a higher number of different fascilin-like arabinogalactan proteins and a greater abundance of expansin-like proteins, which could participate in CWI. Altogether, these data indicate that Col-0 and Ws trigger similar mechanisms to cope with salt stress, and specific changes are more likely related to the developmental stage than to their respective genetic background.

Lipoic Acid Combined with Melatonin Mitigates Oxidative Stress and Promotes Root Formation and Growth in Salt-Stressed Canola Seedlings (Brassica napus L.).

Lipoic acid (LA) and melatonin (MT) are pleiotropic molecules participating in plant stress resistance by modulating cellular biochemical changes, ion homeostasis, and antioxidant enzyme activities. However, the combined role of these two molecules in counteracting the detrimental impacts of salinity stress is still unknown. In the present study, we determined the effects of exogenous LA (0.5 µM), MT (1 µM) and their combination (LA + MT) on growth performance and biomass accumulation, photosynthetic pigments, enzymatic and non-enzymatic antioxidant activities, and ions homeostatic in canola (Brassica napus L.) seedlings under salinity stress (0, 100 mM) for 40 days. The results indicate that exogenous application of LA + MT improved the phenotypic growth (by 25 to 45%), root thickness (by 68%), number of later lateral roots (by 52%), root viability (by 44%), and root length (by 50%) under salinity stress. Moreover, total soluble protein, chlorophyll pigments, the concentration of superoxide dismutase (SOD), catalase peroxidase (CAT), and ascorbic peroxidase (ASA) increased with the presence of salt concentration into the growth media and then decreased with the addition of LA + MT to saline solution. Leaf protein contents and the degradation of photosynthetic pigments were lower when LA + MT treatments were added into NaCl media. The proline and phenol contents decreased in the exogenous application of LA + MT treatments more than individual LA or MT treatments under the salinity stress. The incorporation of LA or MT or a combination of LA + MT to saline solution decreased salinity-induced malondialdehyde and electrolyte leakage. In conclusion, the alteration of metabolic pathways, redox modulation, and ions homeostasis in plant tissues by the combined LA and MT application are helpful towards the adaptation of Brassica napus L. seedlings in a saline environment. The results of this study provide, for the first time, conclusive evidence about the protective role of exogenous LA + MT in canola seedlings under salinity stress.

Arabidopsis Phototropins Participate in the Regulation of Dark-Induced Leaf Senescence.

Senescence is the final stage of plant development, affecting individual organs or the whole organism, and it can be induced by several environmental factors, including shading or darkness. Although inevitable, senescence is a complex and tightly regulated process, ensuring optimal remobilization of nutrients and cellular components from senescing organs. Photoreceptors such as phytochromes and cryptochromes are known to participate in the process of senescence, but the involvement of phototropins has not been studied to date. We investigated the role of these blue light photoreceptors in the senescence of individually darkened Arabidopsis thaliana leaves. We compared several physiological and molecular senescence markers in darkened leaves of wild-type plants and phototropin mutants (phot1, phot2, and phot1phot2). In general, all the symptoms of senescence (lower photochemical activity of photosystem II, photosynthetic pigment degradation, down-regulation of photosynthetic genes, and up-regulation of senescence-associated genes) were less pronounced in phot1phot2, as compared to the wild type, and some also in one of the single mutants, indicating delayed senescence. This points to different mechanisms of phototropin operation in the regulation of senescence-associated processes, either with both photoreceptors acting redundantly, or only one of them, phot1, playing a dominant role.

The floral repressors TEMPRANILLO1 and 2 modulate salt tolerance by regulating hormonal components and photo-protection in Arabidopsis.

Members of the plant specific RAV family of transcription factors regulate several developmental and physiological processes. In the model plant Arabidopsis thaliana, the RAV TEMPRANILLO 1 (TEM1) and TEM2 control important phase changes such as the juvenile to adult and the vegetative to reproductive transitions. Besides their known regulatory function in plant development, a transcriptomics analysis of transgenic plants overexpressing TEM1 also revealed overrepresentation of Gene Ontology (GO) categories related to abiotic stress responses. Therefore, to investigate the biological relevance of these TEM-dependent transcriptomic changes and elucidate whether TEMs contribute to the modulation of plant growth in response to salinity, we analyzed the behavior of TEM gain and loss of function mutants subjected to mild and high salt stresses at different development stages. With respect to increasing salinity, TEM overexpressing plants were hypersensitive whereas the tem1 tem2 double mutants were more tolerant. Precisely, tem1 tem2 mutants germinated and flowered faster than the wild-type plants under salt stress conditions. Also, tem1 tem2 plants showed a delay in salt-induced leaf senescence, possibly as a consequence of downregulation of jasmonic acid biosynthesis genes. Besides a shorter life cycle and delayed senescence, tem1 tem2 mutants appeared to be better suited to withstand oxidative stress as they accumulated higher levels of α-tocopherol (an important antioxidant metabolite) and displayed a slower degradation of photosynthetic pigments. Taken together, our studies suggest novel and crucial roles for TEM in adaptive growth as they modulate plant development in response to environmental changes such as increasing soil salinity.

Physiological Response and Multi-Elemental Content in Lichens Growing on Agricultural Fences: A Pilot Study Comparing No-Tillage and Organic Cropping.

The objective of this study is to compare the physiological response (content and degradation of photosynthetic pigments, membrane oxidation products and soluble proteins) and multi-element content of Ramalina celastri (lichenized fungi) growing on agricultural fences with no-tillage (associated with transgenic crops and agrochemical application), organic cropping and a non-cultivated area. We found that R. celastri did not differ in its physiological response to agricultural practices, except for the contents of chlorophyll b and phaeophytin a which were high in both cultivated areas. Lichens growing in organic cropping fields have higher arsenic, chromium, uranium and internal transition elements common in the earth's crust, possibly due to the greater resuspension of the material during soil tillage. Lichens that grow on posts close to no-tillage field had higher bromine contents (present in numerous pesticides). We found evidence that R. celastri behaves as a tolerant species to air pollution in agricultural environments.

Nitrogen Deprivation-Induced Production of Volatile Organic Compounds in the Arachidonic-Acid-Accumulating Microalga Lobosphaera incisa Underpins Their Role as ROS Scavengers and Chemical Messengers

The green microalga Lobosphaera incisa accumulates long-chain polyunsaturated arachidonic acid sequestered in triacylglycerols under nitrogen (N)-starvation conditions. Many of L. incisa’s physiological and metabolic responses to N-starvation have been previously investigated. However, the temporal dynamics of the volatile organic compounds (VOCs) under different N availability and their role in L. incisa stress responses have yet to be elucidated. Here, we investigated the VOC profiles of L. incisa to reveal their emission patterns, and proposed their physiological roles under N-starvation. Using gas chromatography-mass spectrometry, 42 and 19 VOCs were identified in the algal biomass (AVOCs) and in the medium (MVOCs), respectively, belonging to alkanes, alkenes, benzenoids, esters, fatty alcohols, fatty aldehydes, fatty acids (FAs), FA esters, ketones, and terpenoids; most of these are the oxidative products of FAs or photosynthetic pigment degradation. The discriminant AVOCs and MVOCs produced under N-starvation were identified by principal component analyses and hierarchical clustering. A significant treatment- and time-dependent increase in volatile FAs and their oxidative products was observed in the algal biomass and medium under N-starvation as compared to N-replete controls, following a similar pattern as reactive oxygen species (ROS) production. This suggests that VOCs may be involved in ROS scavenging. Despite the increase in total VOCs, terpenoids and alkenes decreased significantly with N-starvation duration, along with chloroplast degradation. Transcriptomic data supported the VOC patterns and revealed the upregulation of genes involved in fatty aldehyde, fatty alcohol, and ketone synthesis, including lipoxygenase, thioesterase, and fatty acyl CoA reductase, along with a decrease in the expression of genes putatively implicated in alkene biosynthesis. Several VOCs were identified as potential biotechnological targets, and their putative biosynthetic pathways were proposed, which could be genetically manipulated to enhance the yields of high value products. We conclude that VOCs may play an important role in stress metabolism, ROS scavenging, and the amelioration of oxidative stress under N-starvation. Also, VOCs released in the medium may communicate the signal to neighboring cells, thus priming them to adjust their metabolic activities to adapt to N-starvation conditions, indicating that the function of VOCs as chemical signaling messengers is conserved from microalgae to higher plants.

Comparisons of Chlorophyll Fluorescence and Physiological Characteristics of Wheat Seedlings Influenced by Iso-Osmotic Stresses from Polyethylene Glycol and Sodium Chloride

Wheat (Triticum aestivum) cultivar Taichung SEL.2 (TCS2) is a salt-tolerance variety, but the mechanism involved remains unclear. This study aims to distinguish between the non-ionic osmotic and salt-mediated physiological effects on TCS2. Osmotic agents polyethylene glycol (PEG) and sodium chloride (NaCl) were applied at three iso-osmotic levels, level 1 containing 24% (w/v) PEG and 200 mM NaCl, level 2 containing 26.5% (w/v) PEG and 250 mM NaCl), and level 3 containing 29% (w/v) PEG and 300 mM NaCl, respectively. According to the investigation of chlorophyll fluorescence in the better NaCl-treated seedlings, maximal quantum yield of photosystem II (PSII) (Fv/Fm) and significant higher effective quantum yield of PSII (ΦPSII) at level 3 were observed. Meanwhile, the non-photochemical quenching of PSII (NPQ) and the quantum yield of regulated energy dissipation of PSII [Y(NPQ)] were significantly higher in the NaCl-treated seedlings, and the quantum yield of non-regulated energy dissipation of PSII [Y(NO)] in the NaCl-treated seedlings was lower than the PEG-treated ones at level 2 and level 3. Furthermore, the less extensive degradation of photosynthetic pigments, the better ascorbate peroxidase (APX) activity and the less accumulation of malondialdehyde (MDA) were also observed in NaCl-treated seedlings. In the morphological traits, shoot elongation in NaCl-treated seedlings was also preserved. These results suggest that TCS2 is more resistant to NaCl-induced osmotic stress than to the PEG-induced stress. This study contributes to plant breeder interest in drought- and/or salt-tolerant wheat varieties.

Nano-ZnO alleviates drought stress via modulating the plant water use and carbohydrate metabolism in maize

ABSTRACT The wide application of zinc oxide (ZnO) nanoparticles inevitably facilities their release into the terrestrial and agricultural ecosystems, severely affecting crop production. The effects of nano-ZnO (100 mg L−1) on stomatal morphology, gas exchange and key carbon metabolism enzyme activities were investigated in drought-stressed and well-watered maize. Drought stress limited the plant growth, caused stomatal closure, decreased the net photosynthetic rate, while increased the osmolyte concentrations. In drought-stressed maize, the application of nano-ZnO (100 mg L−1) alleviated photosynthetic pigment degradation and benefited the stomatal movement, maintained a higher net photosynthetic rate, and enhanced water use efficiency, promoting the drought tolerance in maize. In addition, the nano-ZnO increased the activities of UDP-glucose pyrophosphorylase, phosphoglucoisomerase and cytoplasmic invertase by 17.8%, 391.5% and 126%, respectively, which enhanced the starch and sucrose biosynthesis and glycolysis metabolism in leaves under drought stress. Manipulation of primary carbohydrate metabolism through nano-ZnO induced regulation of the key enzymes activity benefited the drought tolerance in maize. The nano-ZnO application (100 mg L−1) alleviates the negative effect of drought stress via improving photosynthetic carbon assimilation of maize.

Physiological and transcriptome analyses of the effects of exogenous dopamine on drought tolerance in apple

Water shortage is one of the main limiting factors in apple (Malus domestica Borkh.) production. Although dopamine is produced in plants and has been linked with response to abiotic stress, the underlying mechanism remains unknown. In this study, physiological analyses revealed that pretreatment with 100 μM dopamine alleviated drought stress in apple seedlings. Dopamine inhibited the degradation of photosynthetic pigments and increased net photosynthetic rate under drought stress. Dopamine also reduced H2O2 content, possibly through direct scavenging and by mediating the antioxidant enzyme activity. Seedlings pretreated with dopamine had higher sucrose and malic acid contents but lower starch accumulation in their leaves. RNA-Seq analysis identified 1052 differentially expressed genes (DEGs) between non-treated and dopamine-pretreated plants under drought. An in-depth analysis of these DEGs revealed that dopamine regulated the expression of genes related to metabolism of nitrogen, secondary compounds, and amino acids under drought stress. In addition, dopamine may improve apple drought tolerance by activating Ca2+ signaling pathways through increased expression of CNGC and CAM/CML family genes. Moreover, analysis of transcription factor expression suggested that dopamine affected drought tolerance mainly through the regulation of WRKY, ERF, and NAC transcription factors.
