Titanium dioxide nanoparticles seed priming as a remedy for nickel-induced stress in maize through antioxidant enhancement and ultrastructural optimization

Heavy metals (HMs) have emerged as a critical global concern, not only limiting crop productivity but also posing risks to public health. Among them, nickel (Ni) is an essential micronutrient for plant growth; however, it becomes toxic at higher concentrations. Nano-enabled approaches, on the other hand, have emerged as promising eco-friendly alternatives for mitigating the negative impact associated with HMs. Here, we investigated the potential of titanium dioxide nanoparticles (TiO2 NPs) against Ni-induced stress in maize. Our results showed that Ni stress caused negative changes in maize by the excessive production of reactive oxygen species (ROS), inhibiting photosynthetic attributes, and damaging cellular ultrastructure. In contrast, TiO2 NPs priming significantly enhanced the antioxidant mechanism, photosynthetic efficacy, and nutrient uptake while reducing ultrastructural damage caused by Ni stress. Furthermore, TiO2 NPs efficiently reduced Ni accumulation, MDA (28%/32%), H2O2 (23%/26%), and O2•‒ (31%/34%) levels in shoot/root tissues, respectively, compared to Ni treatment. Moreover, TiO2 NPs priming has modulated the expression of antioxidant and defense-related genes, thereby restoring cellular redox homeostasis. Collectively, this is the first piece of evidence demonstrating the potential of TiO2 NPs as an efficient and sustainable alternative for enhancing crop tolerance in Ni-contaminated areas.

A Zinc Polyphenolic Compound Increases Maize Resistance Against Infection by Bipolaris maydis.

Maize leaf blight (MLB), caused by the fungus Bipolaris maydis, is an important disease affecting maize production. In order to minimize the use of fungicides in agriculture, nutrient-based resistance inducers may become a promising alternative to manage MLB. The goal of this study was to investigate the potential of Semia® (zinc (20%) complexed with a plant-derived pool of polyphenols (10%)) to hamper the infection of maize leaves by B. maydis by analyzing their photosynthetic performance and carbohydrate and antioxidative metabolism, as well as the expression of defense-related genes. Plants were sprayed with water (control) or Semia® (referred to as induced resistance (IR) stimulus hereafter) and not inoculated or inoculated with B. maydis. The mycelial growth and conidium germination were significantly reduced by the IR stimulus in vitro. The MLB severity was significantly reduced by 76% for IR-stimulus-sprayed plants compared to plants from the control treatment. For infected and IR-stimulus-sprayed plants, the glucose, fructose, sucrose, and starch concentrations were significantly higher compared to inoculated plants from the control treatment. The activity levels of superoxide dismutase, ascorbate peroxidase, catalase, and glutathione reductase were significantly higher for the IR-stimulus-sprayed plants compared to plants from the control treatment. Less impairment on the photosynthetic apparatus (higher values for leaf gas exchange (rates of net CO2 assimilation, stomatal conductance to water vapor, and transpiration) and chlorophyll a fluorescence (variable-to-maximum Chl a fluorescence ratio, photochemical yield, and yield for dissipation by down-regulation) parameters)) along with a preserved pool of chlorophyll a+b and carotenoids were noticed for infected and IR-stimulus-sprayed plants compared to infected plants from the control treatment. The defense-related genes IGL, CHS02, PR1, PAL3, CHI, and GLU were strongly up-regulated in the leaves of IR-stimulus-sprayed and infected plants compared to infected plants from the control treatment. These findings highlight the potential of using this IR stimulus for MLB management.

Secreted Xylanase PstXyn1 Contributes to Stripe Rust Infection Possibly by Overcoming Cell Wall Barrier and Suppressing Defense Responses in Wheat.

Puccinia striiformis f. sp. tritici (Pst) secretes a plethora of cell wall-degrading enzymes (CWDEs) to facilitate fungal invasion during infection. However, the functions and molecular mechanisms of the CWDEs from Pst remain unclear. In this study, we identified a secreted xylanase, named PstXyn1, with the GH10 domain. PstXyn1 was significantly up-regulated at the early infection stage of Pst. The signal peptide of PstXyn1 was confirmed to be functional. The purified PstXyn1 showed detectable xylanase activity. In addition, we found that PstXyn1-silenced wheat plants exhibited broad-spectrum resistance against multiple Pst pathotypes. Colloidal gold labeling and transcriptome sequencing analyses revealed that PstXyn1 contributed to xylan degradation in host cell walls and suppressed the expression of defense-related genes. Conclusively, our results indicate that PstXyn1 is secreted as an important virulence factor to overcome host cell wall barriers and compromise immune responses for fungal invasion, providing potential targets for improving wheat resistance to stripe rust.

Nei 6 You 7075, a hybrid rice cultivar, exhibits enhanced disease resistance and drought tolerance traits

BackgroundRice is the main food crop for much of the population in China. Therefore, selecting and breeding new disease resistance and drought tolerance in rice is essential to ensure national food security. The utilization of heterosis has significantly enhanced rice productivity, yet many of the molecular mechanisms underlying this phenomenon remain largely unexplored. ‘Nei 6 You 7075’ (‘N6Y7075’) is a novel hybrid rice cultivar with exceptional quality, developed through the crossbreeding of ‘Fuhui 7075’ (‘FH7075’) and ‘Neixiang 6 A’ (‘NX6A’). However, the precise mechanisms underlying the disease resistance and drought tolerance in ‘N6Y7075’ are poorly understood. In this study, we investigated the resistance of hybrid rice ‘N6Y7075’ to bacterial blight (Xanthomonas oryzae pv. oryzae), rice blast (Magnaporthe oryzae), and drought and identified differentially expressed genes between hybrid rice ‘N6Y7075’ and its parents through RNA-seq analysis.ResultsOur research found that the hybrid ‘N6Y7075’ and its female parent ‘NX6A’ were less susceptible to bacterial blight and rice blast than the male parent ‘FH7075’, while ‘FH7075’ showed better drought tolerance than ‘NX6A’. The hybrid ‘N6Y7075’ exhibited heterosis. Clustering results revealed that the expression profiles of the F1 hybrid closely resembled those of its parental lines rather than exhibiting an intermediate profile between the two parental lines. The disease resistance of hybrid rice ‘N6Y7075’ may be attributed to the plant-pathogen interaction pathways involving Xa21, CDPK, and RPM1-mediated hypersensitive response and WRKY1-induced defense-related gene expression and programmed cell death. The MAPK signaling pathway PR1 could also be associated with plant defense responses. Hybrid rice ‘N6Y7075’ may enhance drought tolerance by regulating MAPKKK17 and WAK60 in the MAPK signaling pathway. These proteins affect ABA stress adaptation and stomatal development in plants, respectively.ConclusionsOur results provide a preliminary exploration of ‘N6Y7075’ disease resistance and drought tolerance and provide a relevant theoretical basis for its further study and use. This study provides insights into the molecular mechanisms of heterosis in hybrid rice and identifies potential associated genes.

Investigation of the anti-Huanglongbing effects using antimicrobial lipopeptide and phytohormone complex powder prepared from Bacillus amyloliquefaciens MG-2 fermentation

Global citrus production has been severely affected by citrus Huanglongbing (HLB) disease, caused by Candidatus Liberibacter asiaticus (Clas), and the development of effective control methods are crucial. This study employed antimicrobial lipopeptide and phytohormone complex powder (L1) prepared from the fermentation broth of the endophytic plant growth promoting bacterium (PGPB) of Bacillus amyloliquefaciens strain MG-2 to treat Candidatus Liberibacter asiaticus (CLas)-infected ‘Citrus reticulata ‘Chun Jian’ plants. Real-time fluorescence quantitative polymerase chain reaction (qPCR) and PCR were employed for disease detection. The results revealed that after 15 spray-drench treatments with L1 solution, the HLB infection rate decreased from 100 to 50%, the bacterial titer decreased by 51.9% compared with a 27.9% decrease in the control group. L1 treatment triggered the production of reactive oxygen species, increased lignin content, and increased defense enzyme activities (p < 0.05). Defense-related gene expression significantly increased within 12 h of treatment. In addition, L1 application also promoted plant growth, as evidenced by higher transpiration rates and net photosynthetic rates as well as increased leave or root density. Root flora analysis revealed that the abundances of Burkholderia_thailandensis, unclassified_g_Burkholderia-Caballeronia-Paraburkholderia, unclassified_g__Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium, and Pseudomonas_mosselii were 1.64, 1.46, 5.84, and 6.93 times greater, respectively, than those in the control group. The levels of phenylpropanoids, polyketides, lipids, lipid-like molecules, organic acids, and derivatives, significantly increased following L1 treatment (FC > 2, p < 0.05). Additionally, salicylic acid, dihydrojasmonic acid, and isopentenyl adenosine levels in leaves markedly increased. High-performance liquid chromatography (HPLC) confirmed that L1 contained surfactin, iturin and fengycin cyclic-lipopeptides (CLPs) as well as indole-3-acetic acid (IAA), 3-indolebutyric acid (IBA), indole-3-carboxylic acid and indole-3-carboxaldehyde auxins, N6-entopentenyladenine and t-zeatin-riboside cytokinins, abscisic acid, 1-aminocyclicpanecarboxylic acid, salicylic acid, and gibberellin A1, A3 and A4 phytohormones. These findings provide insight into multiple mechanisms by which endophytic Bacillus PGPB L1 is able to combat HLB disease, to promote citrus plant growth, and to optimize the root flora for soil health which offering an innovative strategy for sustainable management of this severe disease and improving citrus plant growth and productivity

Caffeic acid improves biocontrol effect of Pantoea jilinensis D25 against tomato gray mold.

Gray mold in tomato caused by Botrytis cinerea is a destructive disease, which can be treated using biocontrol agents. Pretreatment of biocontrol strains with oxidative-stress-ameliorating compounds can enhance their tolerance to oxidative microenvironment in infected fruit wounds. In this study, we aimed to determine the effect of caffeic acid (CA) on the biocontrol efficacy of Pantoea jilinensis D25 in gray mold in cherry tomato. D25 strain pretreated with CA exhibited enhanced oxidative stress response in cherry tomato wounds. CA-treated D25 cells recovered from cherry tomato wounds had lower ROS level and cellular oxidative damage than D25 cells not treated with CA. The antioxidation-related genes (CAT and GPX) were upregulated and colonization ability of D25 strain was improved in cherry tomato wounds after CA application. Furthermore, the biocontrol effects of CA-treated D25 strain were improved. CA pretreatment to D25 strain could significantly improve the expression of defense-related genes (SlMYC-2, SlLoxD, SlEIN2, and SlEIN3) and activity of defense-related enzymes in cherry tomato wounds. Application of CA to D25 strain could improve its ability to combat antioxidant stress and biocontrol efficacy against postharvest tomato gray mold. © 2024 Society of Chemical Industry.

Transcriptome Comparison between Resistant and Susceptible Soybean Cultivars in Response to Inoculation of Phytophthora sojae.

Phytophthora root and stem rot, caused by Phytophthora sojae, considerably reduces soybean yield worldwide. Our previous study identified two genomic regions on chromosome 18 (2.1-2.6 and 53.1-53.3 Mbp) that confer resistance to the P. sojae isolate 2457, through linkage analysis using progenies derived from the Daepung × Socheong2 population. These two regions contained 51 and 19 annotated genes, respectively. However, the specific gene responsible for resistance to P. sojae isolate 2457 has yet to be identified. In this study, we performed a comparative transcriptomic analysis of Socheong2 and Daepung, two Korean soybean varieties identified as resistant and susceptible to P. sojae isolate 2457, respectively. RNA sequencing was conducted on tissue samples collected at 0, 6, and 12 hours after inoculation (HAI), and significant differences in the expression of defense-related genes were observed across time points and between the two cultivars. Genes associated with the jasmonic acid, salicylic acid, ethylene, and systemic acquired resistance pathways were upregulated in both cultivars at 6 and 12 HAI compared to 0 HAI, with these biological processes were more strongly upregulated in Socheong2 compared to Daepung at 6 and 12 HAI. A comparison of differentially expressed genes (DEGs) and candidate genes within the previously identified QTL regions revealed an ortholog of the HS1 PRO-1 2 gene from Arabidopsis thaliana among the upregulated DEGs in Socheong2, particularly at 12 HAI compared to 0 HAI. This study will aid in targeted breeding efforts to develop soybean varieties with improved resistance to P. sojae.

Histone H4K8hib modification promotes gene expression and regulates rice immunity.

This study uncovers the role of H4K8hib as an active epigenetic mark in rice, which positively correlates with gene expression and enhances immune responses. Furthermore, HDA705 was identified as the key enzyme regulating H4K8hib. Mutation of HDA705 led to hyper-H4K8hib, which in turn activated the expression of defense-related genes and enhanced rice resistance to pathogens.

The secreted isochorismatases derived from certain filamentous pathogens play vital roles in the infection of host plants by lowering salicylic acid (SA) levels and suppressing SA-mediated defense pathway. However, it remains unclear whether the fungus Fusarium oxysporum f. sp. cubense tropical race 4 (FocTR4), which causes vascular wilt in bananas, utilizes isochorismatases to modulate SA levels in the host and subvert the banana defense system for successful infection. In the current study, we selected and functionally characterized the foisc1 gene, one of 10 putative isochorismatase-encoding genes in FocTR4 that showed significant upregulation during early stages of infection. Deletion of foisc1 resulted in enhanced vegetative growth and conidiation, increased sensitivity to SA, reduced colonization within host plants, as well as impaired pathogenicity. Conversely, complementation restored phenotypes similar to those observed in the wild-type strain. Furthermore, deletion of foisc1 led to a notable rise in activities of defense-related enzymes such as catalase, peroxidase, and phenylalnine ammonialyase; along with an upregulated expression of several defense-related genes including PR genes and NPR1 genes within hosts' tissues. The non-secretory nature of Foisc1 protein was confirmed and its absence did not affect SA levels within host plants. Transcriptome analysis revealed that deletion of foisc1 resulted in decreased expression levels for numerous genes associated with pathogenicity including those involved in fusaric acid biosynthesis and effector genes as well as a catechol 1,2-dioxygenase gene essential for SA degradation; while increasing expression levels for numerous genes associated with hyphal growth and conidiation were observed instead. Therefore, our findings suggest that Foisc1 may influence hyphal growth, conidiation, sensitivity to SA, and pathogenicity of FocTR4 through modulation of various genes implicated in these processes. These findings provide valuable insights into the pathogenesis of FocTR4, and create a groundwork for the future development of innovative control strategies targeting vascular wilt disease of banana.

Sugar beet root susceptibility to storage rots and downregulation of plant defense genes increases with time in storage

Storage rots are a significant cause of postharvest losses for the sugar beet crop, however, intrinsic physiological and genetic factors that determine the susceptibility of roots to pathogen infection and disease development are unknown. Research, therefore, was carried out to evaluate the disease development in sugar beet roots caused by two common storage pathogens as a function of storage duration and storage temperature, and to identify changes in the expression of defense genes that may be influencing the root susceptibility to disease. To evaluate root susceptibility to disease, freshly harvested roots were inoculated with Botrytis cinerea or Penicillium vulpinum on the day of harvest or after 12, 40, or 120 d storage at 5 or 12 °C and the weight of rotted tissue present in the roots after incubation for 35 d after inoculation were determined. Disease susceptibility and progression to B. cinerea and P. vulpinum increased with storage duration with elevations in susceptibility occurring more rapidly to B. cinerea than P. vulpinum. Also, B. cinerea was more aggressive than P. vulpinum and caused greater rotting and tissue damage in postharvest sugar beet roots. Storage temperature had minimal effect on root susceptibility to these rot-causing pathogens. Changes in defense gene expression were determined by sequencing mRNA isolated from uninoculated roots that were similarly stored for 12, 40 or 120 d at 5 or 12 °C. As susceptibility to rot increased during storage, concurrent changes in defense-related gene expression were identified, including the differential expression of 425 pathogen receptor and 275 phytohormone signal transduction pathway-related genes. Furthermore, plant resistance and hormonal signaling genes that were significantly altered in expression coincident with the change in root susceptibility to storage rots were identified. Further investigation into the function of these genes may ultimately elucidate methods by which storage rot resistance in sugar beet roots may be improved in the future.

Regulation of maize growth and immunity by ZmSKI3-mediated RNA decay and post-transcriptional gene silencing.

Disease resistance is often associated with compromised plant growth and yield due to defense-growth tradeoffs. However, key components and mechanisms underlying the defense-growth tradeoffs are rarely explored in maize. In this study, we find that ZmSKI3, a putative subunit of the SUPERKILLER (SKI) complex that mediates the 3'-5' degradation of RNA, regulates both plant development and disease resistance in maize. The Zmski3 mutants showed retarded plant growth and constitutively activated defense responses, while the ZmSKI3 overexpression lines are more susceptible to Curvularia lunata and Bipolaris maydis. Consistently, the expression of defense-related genes was generally up-regulated, while expressions of growth-related genes were mostly down-regulated in leaves of the Zmski3-1 mutant compared to that of wild type. In addition, 223 differentially expressed genes that are up-regulated in Zmski3-1 mutant but down-regulated in the ZmSKI3 overexpression line are identified as potential target genes of ZmSKI3. Moreover, small interfering RNAs targeting the transcripts of the defense- and growth-related genes are differentially accumulated, likely to combat the increase of defense-related transcripts but decrease of growth-related transcripts in Zmski3-1 mutant. Taken together, our study indicates that plant growth and immunity could be regulated by both ZmSKI3-mediated RNA decay and post-transcriptional gene silencing in maize.

Investigating the Role of OsHDT701 and Other Blast-Associated Negative Regulatory Genes in Indica Rice Cultivar Ranjit Using Combined Wet Lab and Computational Approaches.

Rice blast, caused by Magnaporthe oryzae, severely impacts global rice production. Understanding the role of the host's blast negative regulatory genes is crucial for combating this disease. We studied the expression of seven rice blast negative regulatory genes (previously characterized in ssp. japonica) in susceptible (Ranjit and Mahsuri) and tolerant/resistant (Shahsarang 1 and IR64) indica rice cultivars during M. oryzae infection. The selected genes showed differential expression patterns, with OsHDT701 (histone H4 deacetylase) downregulated in tolerant/resistant and upregulated in susceptible cultivars. Promoter analysis of OsHDT701 in cv. Ranjit revealed biotic stress-responsive and M. oryzae effector binding motifs (MoHTR2 and MoSPAB1). Protein-DNA docking showed an interaction for both the effectors and promoter sequences. Furthermore, a strong interaction between the OsERF922 transcription factor (a negative regulator of rice blast) and GCC box at the promoter suggests OsERF922-mediated regulation of OsHDT701. Next, a comparative analysis of ssp. japonica (Nipponbare) OsHDT701 CDS to 11 indica rice cultivars, including Ranjit, detected an SNP (T-A) at position 591. Moreover, the structure modeling and protein-protein interaction reveal that OsHDT701 may form a complex with other histone deacetylases to regulate defense-related gene expression. Taken together, our study unveils new possibilities for OsHDT701 regulation mechanisms during M. oryzae infection.

Exploring nonanol from Bacillus velezensis (YEBBR6) for root-knot nematode management in bananas: Integrating experimental and computational approaches

Nematode infestations pose a severe threat to global banana production, leading to substantial yield losses and economic damage. Among these, the root-knot nematode Meloidogyne incognita is particularly detrimental, causing root galls that severely impair water and nutrient uptake, leading to stunted growth, reduced fruit quality, and significant economic losses for farmers. In the face of increasing agricultural challenges, bacterial endophytes have emerged as essential tools for sustainable agriculture, offering ecofriendly solutions to enhance plant health and manage pests. The present study aims to identify and characterise biomolecules from bacterial endophyte Bacillus velezensis (YEBBR6) with potential nematicidal properties against root-knot nematode Meloidogyne incognita infecting banana. The analysis of volatile and non-volatile organic compounds (VOCs/NVOCs) from the inhibition zone during the ditrophic interaction between B. velezensis (YEBBR6) and Fusarium oxysporum f.sp. cubense (Foc) revealed the presence of several distinctive biomolecules with antifungal and antimicrobial properties. These compounds included linoelaedic acid, nonanol, acetylvaleryl, 5–hydroxyl methyl furfural, clindamycin, allobarbital, 3-thiazolidine carboxamide, azulene, aminomorpholine, procyclidine, campholic acid, 3 amino-4 hydroxy phenyl sulfone, 3–deoxy mannoic lactone, hexadecanoic acid, oleic acid, and dihydroacridine. The in-silico analysis revealed that nonanol, exhibited strong binding efficacy against M. incognita protein targets, including cytochrome C oxidase subunit 1, calreticulin, neuropeptide G-protein coupled receptor, chorismate mutase 1, venom allergen-like proteins, and β-1,4-endoglucanase. Notably, nonanol proved more effective than the commercially used nematicide carbofuran 3G. Molecular dynamics studies further supported the potential of nonanol, and its nematicidal properties were validated through in vitro and pot culture studies. Moreover, nonanol was found to induce the expression of defense-related genes, such as MAPK10, WRKY22, ascorbate peroxidase, catalase, and NADPH oxidase, in banana seedlings, thus enhancing the plant's immune response. These findings highlight the potential of nonanol, derived from B. velezensis (YEBBR6), as a potent and environmentally friendly alternative for controlling nematode infestations in banana cultivation.

GmWRKY33A positively regulates disease resistance in soybean (Glycine max)

The WRKY transcription factor gene family is a plant-specific transcription factor that plays important roles defense responses. Studies in model plant Arabidopsis demonstrated that WRKYs function downstream of mitogen activated-protein kinase (MAPK) signaling cascade and participate in defense responses through activating the expression of defense-related genes. However, the roles of WRKYs in defense responses have not been previously investigated in paleopolyploidy soybean. Bioinfomatic analysis revealed that there are three pair of GmWRKY33 genes in the soybean genome. The identity of first two pair of GmWRKY33 genes is greater than 84% (named as GmWRKY33A). The identity of genes within the same pair is greater than 95%. A 300 bp fragment highly homologous to these four GmWRKY33A was chosen to clone into bean pod mosaic virus (BPMV)-based silencing vector (BPMV-VIGS) to achieve the goal of silencing four GmWRKY33A genes simultaneously. In this study, we simultaneously silenced four homologous genes of GmWRKY33A using a bean pod mottle virus (BPMV) vector carrying a single fragment of GmWRKY33A. Comparing the silenced plants with the vector control plants, no evident morphological phenotypes were observed. However, the GmWRKY33A-silenced plants exhibited significantly reduced resistance to Pseudomonas syringae pv. glycinea (Psg), Xanthomonas axonopodis pv. glycine (Xag), as well as to soybean mosaic virus (SMV). Furthermore, we demonstrated that silencing these GmWRKY33A genes significantly inhibited the activation of GmMPK3/GmMPK6 induced by Psg infection. Collectively, our results suggest that GmWRKY33As are involved in soybean immunity through regulating the transcription of GmMPK3/6 genes or activating the kinase activities of GmMPK3/6. Taken together, our results demonstrated that GmWRKY33As are positive regulators of soybean immune responses.

Preparation of pH-Responsive Kas@ZnO Quantum Dots for Synergistic Control of Rice Blast and Enhanced Disease Resistance in Rice.

The construction of controlled-release formulations improves the sustained-release performance and utilization efficiency of pesticides, which are important aspects in plant protection and environmental chemistry. The current study employs kasugamycin (Kas), which is widely used to control Magnaporthe oryzae, conjugated with carboxyl-functionalized ZnO quantum dots via amide linkages to yield a pH-responsive pesticide delivery system (Kas@ZnO). Physicochemical characterizations indicated the successful preparation of the Kas@ZnO nanoparticles. In vitro drug release assessments indicated that Kas@ZnO exhibited a loading capacity of 21.05% and could effect the controlled release of Kas in an acidic environment, which is beneficial given the unique acidic microenvironment of M. oryzae. Bioactivity assays demonstrated that Kas@ZnO could simultaneously inhibit mycelial growth and spore germination. Additionally, bioactivity tests showed that the control efficacy of Kas@ZnO against rice blast reached 67.43% after 14 days of in vivo spray inoculation, which was higher than that obtained with Kas (55.50%), suggesting improved beneficial effects of Kas@ZnO application over a prolonged duration. Moreover, Kas@ZnO enhanced the activity of defense-related enzymes in rice and upregulated the expression of defense-related genes, such as OsPR2, OsPR3, OsPR5, OsWRKY45, OsLYP6, and OsNAC4. Ultimately, the biosafety assessments revealed that Kas@ZnO did not exert any harmful effects on rice and demonstrated slight toxicity toward zebrafish. These findings indicate that Kas@ZnO can function as a pH-sensitive pesticide delivery system, allowing for targeted release of the pesticide within plant tissues. This technology demonstrates significant potential for eco-friendly plant disease management.

Forest tree extracts induce resistance to Pseudomonas syringae pv. tomato in Arabidopsis.

The widespread use of conventional pesticides for plant pathogen control poses significant risks to human health and the environment, and it is therefore crucial to develop environmentally friendly, human-safe alternatives to these products that offer a sustainable approach for crop protection. Here, we examined the potential of ethanolic extracts from four forest tree species for their antibacterial activity against the bacterial pathogenPseudomonas syringaepv.tomato (Pst) and their ability to trigger effective defense responses in the model plantArabidopsis thaliana. The extracts exhibited direct toxic effects against Pst and triggered the expression of defense-related genes naturally induced by oxidative stress cues or the defense elicitor salicylic acid in leaf tissue. The direct antibacterial effects of the tree extracts, together with their defense gene-inducing effectsin planta, resulted in a strong host plant-protecting effect againstPst. These findings suggest the eventual effectiveness of forest tree extracts as plant protectants against the bacterial pathogenPst. They also suggest the potential of these extracts as a sustainable, eco-friendly alternative to conventional pesticides for the management of economically important plant pathogens.

Chitosan, Methyl Jasmonate, and Silicon Induce Resistance to Angular Leaf Spot in Common Bean, Caused by Pseudocercospora griseola, with Expression of Defense-Related Genes and Enzyme Activities

This study assessed the efficacy of chitosan, methyl jasmonate, and silicon in the reduction of disease severity and the induction of defense responses in common bean plants against angular leaf spot caused by Pseudocercospora griseola. The expression level of several pathogenesis-related (PR) proteins, PR1, PR2 (β-1,3-glucanase), and PR3 (chitinase), and defense-related enzymes, phenylalanine ammonia-lyase, peroxidase, and lipoxygenase, was analyzed at different time points in common bean plants after different treatments. Elicitor treatments significantly reduced disease severity 21 days after inoculation, with silicon at a 2 mM concentration proving most effective with 38.93% disease control, followed by 1 mM MeJA and 2% chitosan, respectively. Treatments with chitosan, methyl jasmonate, and silicon, regardless of pathogen infection, significantly elevated PR1, PR2, and PR3 gene expressions at 48 h after inoculation (hpi). PAL and POD activities were similarly increased following elicitor treatments and pathogen infection, especially at 48 hpi. Chemical elicitors applied post-inoculation induced PR proteins, PAL, and POD enzyme activities at 48 hpi, while LOX activity exhibited a variable fluctuation with treatments. These findings suggested that chemical elicitors, especially silicon, were effective in reducing ALS disease severity in common beans, with improved resistance associated with the expression of pathogen-responsive genes. This study is the first to analyze the expression profiles of defense-related genes in common beans treated with chemical elicitors prior to P. griseola infection.

Biodegradable soy protein isolate carvacrol coating controls soft rot on nectarines by enhancing defense response

Nectarine fruits are rich in nutrients, which are susceptible to fungal infection during postharvest, resulting in quality deterioration and decay. This study aimed to assess the effect of different concentrations of carvacrol and soy protein isolate coatings on the fungal infection and quality characteristics of nectarines during postharvest storage. The results showed that soy protein isolate carvacrol (SPI-C) coating inhibited the growth of Rhizopus stolonifer in vitro. The SPI-C coating significantly reduced the incidence of postharvest soft rot, maintained the postharvest quality of nectarines during storage. SPI-C coating induces the defense response to soft rot by enhancing the accumulation of phenolic, flavonoids and lignin, and up-regulating the expression of defense-related genes in nectarines. The SPI-C coating activated the transcription of antioxidant enzymes (CAT, SOD and POD) and eliminated ROS damage. The disease resistance response promotes the production of defense-related substances by upregulating the gene expression of PpPPO and PpPAL. In summary, SPI-C coating significantly reduces the incidence of postharvest soft rot, maintains the postharvest quality and induces defense response to soft rot by enhancing the expression of genes involved in phenylpropanoid metabolism in nectarines during storage.

Influences of Ipomoea batatas Anti-Cancer Peptide on Tomato Defense Genes.

This study investigates the impact of IbACP (Ipomoea batatas anti-cancer peptide) on defense-related gene expression in tomato leaves, focusing on its role in plant defense mechanisms. Previously, IbACP was isolated from sweet potato leaves, and it was identified as a peptide capable of inducing an alkalinization response in tomato suspension culture media. Additionally, IbACP was found to regulate the proliferation of human pancreatic adenocarcinoma cells. Elucidate IbACP's molecular influence on defense-related gene expression in tomato leaves using next-generation sequencing analysis. To assess the impact of IbACP on defense-related gene expression, transcriptome data were analyzed, encompassing various functional categories such as photosynthesis, metabolic processes, and plant defense. Semi-quantitative reverse-transcription polymerase chain reaction analysis was employed to verify transcription levels of defense-related genes in tomato leaves treated with IbACP for durations ranging from 0 h (control) to 24 h. IbACP induced jasmonic acid-related genes (LoxD and AOS) at 2 h, with a significant up-regulation of salicylic acid-dependent gene NPR1 at 24 h. This suggested a temporal antagonistic effect between jasmonic acid and salicylic acid during the early hours of IbACP treatment. Downstream ethylene-responsive regulator genes (ACO1, ETR4, and ERF1) were consistently down-regulated by IbACP at all times. Additionally, IbACP significantly up-regulated the gene expressions of suberization-associated anionic peroxidases (TMP1 and TAP2) at all time points, indicating enhanced suberization of the plant cell wall to prevent pathogen invasion. IbACP enhances the synthesis of defense hormones and up-regulates downstream defense genes, improving the plant's resistance to biotic stresses.
