Deep-tissue photoacoustic imaging of photoswitchable transgenic mouse models

Compared with visible light, near-infrared (NIR) light (700–1300 nm) is capable of deep-tissue penetration because of low tissue attenuation, and thus holds great promise for deep-tissue optical manipulation and imaging. In this work, we take advantages of the NIR Rhodopseudomonas palustris bacterial phytochrome photoreceptors (BphP1) and generate a loxP-BphP1 photoswitchable transgenic mouse model that enables Cre-dependent temporal and spatial targeting of BphP1 expression at the whole-body scale in vivo. This BphP1 phytochrome incorporates biliverdin chromophore and reversibly switches between the ground state and activated state, which enabled the light-controlled manipulation of gene expression and differential detection in photoacoustic tomography (PAT). We first validated the optogenetic performance of BphP1 that binds its engineered protein partner QPAS1 to trigger gene transcription in primary cells and living mice, and demonstrated its superior capability of deep-tissue optogenetics over the existing visible-light proteins. Then, by taking advantage of PAT's deep-tissue non-invasive imaging ability and the BphP1's photoswitching properties, we can suppress the non-switching signals from background blood and improve the molecular detection sensitivity by three orders of magnitude. This study shows the BphP1-encoded photoswitchable transgenic mouse model can be used for both NIR optogenetic manipulation and deep-tissue PAT.

Activity-based Photoacoustic Probes Reveal Elevated Intestinal MGL and FAAH Activity in a Murine Model of Obesity.

Obesity is a chronic health condition characterized by the accumulation of excessive body fat which can lead to and exacerbate cardiovascular disease, type-II diabetes, high blood pressure, and cancer through systemic inflammation. Unfortunately, visualizing key mediators of the inflammatory response, such as monoacylglycerol lipase (MGL) and fatty acid amide hydrolase (FAAH), in a selective manner is a profound challenge owing to an overlapping substrate scope that involves arachidonic acid (AA). Specifically, these enzymes work in concert to generate AA, which in the context of obesity, has been implicated to control appetite and energy metabolism. In this study, we developed the first selective activity-based sensing probes to detect MGL (PA-HD-MGL) and FAAH (PA-HD-FAAH) activity via photoacoustic imaging. Activation of PA-HD-MGL and PA-HD-FAAH by their target enzymes resulted in 1.74-fold and 1.59-fold signal enhancements, respectively. Due to their exceptional selectivity profiles and deep-tissue photoacoustic imaging capabilities, these probes were employed to measure MGL and FAAH activity in a murine model of obesity. Contrary to conflicting reports suggesting levels of MGL can be attenuated or elevated, our results support the latter. Indeed, we discovered a marked increase of both targets in the gastrointestinal tract. These key findings set the stage to uncover the role of the endocannabinoid pathway in obesity-mediated inflammation.

Side-viewing photoacoustic waveguide endoscopy

Side-viewing hollow optical waveguides allow for minimally invasive endoscopy by concentrically guiding light and sound for photoacoustic generation and detection. Here, we characterize the side-viewing photoacoustic waveguide (PWG) endoscope by scanning 7.2 μm diameter carbon fiber threads within phantom tissues and animal tissues. Photoacoustic signals are carried along the 5.5 and 10.0 cm length of the PWG with minimal attenuation. Thus, this technology enables 360°, deep-tissue photoacoustic imaging. Photoacoustic signals were identified up to 8.0 mm from the PWG imaging window in an optically clear medium. The outer diameter of this device is measured as just over 1.0 mm, with the potential to be further miniaturized due to its unique design. The PWG is an ideal candidate for a myriad of pre-clinical and clinical applications where typical photoacoustic endoscopy systems are impractical, due to their size. Presented here, is the first side-viewing photoacoustic waveguide endoscope.

Deep tissue photoacoustic imaging of nickel(II) dithiolene-containing polymeric nanoparticles in the second near-infrared window.

Photoacoustic imaging is gaining great attention in the medical world due to its significant potential for clinical translation. Light excitation in the second near-infrared (NIR-II) window (1000-1350 nm) has resolution and penetration depth suitable for several clinical applications. However, the significant challenge exists for clinical translation because of the absence of notable intrinsic chromophores in this clinically significant optical range to generate diagnostic images.Methods: We present newly developed a biocompatible nickel dithiolene-based polymeric nanoparticle (NiPNP), which have a strong and sharp absorption peak at 1064 nm, as a photoacoustic contrast agent to boost specific absorbance in the NIR-II window for in vivo deep tissue imaging.Results: We confirm the enhanced PA signal by NiPNP's strong light absorption in the NIR-II window (287% higher than that of NIR-I) and deep tissue imaging capability (~5.1 cm) through in vitro experiment. We have successfully acquired diagnostic-quality in vivo photoacoustic images in deep tissue (~3.4 cm) of sentinel lymph nodes, gastrointestinal tracts, and bladders of live rats by using clinically viable imaging system.Conclusions: Our results prove that with strong absorption in the NIR-II window and with deeper imaging depth, the clinical translation of photoacoustic imaging with NiPNP is feasible for preclinical studies and thus would facilitate further clinical investigations.

Development of Citrate-based Dual-Imaging Enabled Biodegradable Electroactive Polymers.

Increasing occurrences of degenerative diseases, defective tissues and severe cancers heighten the importance of advanced biomedical treatments, which in turn enhance the need for improved biomaterials with versatile theranostic functionalities yet using minimal design complexity. Leveraging the advantages of citrate chemistry, we developed a multifunctional citrate-based biomaterial platform with both imaging and therapeutic capabilities utilizing a facile and efficient one-pot synthesis. The resulting aniline tetramer doped biodegradable photoluminescent polymers (BPLPATs) not only possess programmable degradation profiles (<1 to >6 months) and mechanical strengths (~20 MPa to > 400 MPa), but also present a combination of intrinsic fluorescence, photoacoustic (PA) and electrical conductivity properties. BPLPAT nanoparticles are able to label cells for fluorescence imaging and perform deep tissue detection with PA imaging. Coupled with significant photothermal performance, BPLPAT nanoparticles demonstrate great potential for thermal treatment and in vivo real-time detection of cancers. Our results on BPLPAT scaffolds demonstrate three-dimensional (3D) high-spatial-resolution deep tissue PA imaging (23 mm), as well as promote growth and differentiation of PC-12 nerve cells. We envision that the biodegradable dual-imaging-enabled electroactive citrate-based biomaterial platform will expand the currently available theranostic material systems and open new avenues for diversified biomedical and biological applications via the demonstrated multi-functionality.

Vacancy engineering of Cu2-xSe nanoparticles with tunable LSPR and magnetism for dual-modal imaging guided photothermal therapy of cancer.

The vacancies in the semiconductor nanocrystals not only induce unique properties, but also provide spaces for engineering them with multifunctions by the introduction of other elements. Herein, the vacancy of Cu2-xSe nanoparticles was tuned by doping with magnetic ferric ions (Fe3+) at room temperature, and the position and intensity of the near-infrared localized surface plasmon resonance (LSPR) in the resultant nanostructure can be finely controlled by altering the feeding amount of Fe3+ ions. The results of the density-functional theory (DFT) calculations show that both doping and replacement reactions are favourable. Owing to its tunable near-infrared absorption and magnetic property, the obtained hybrid nanostructure was demonstrated to be a novel nanotheranostic agent for effective deep-tissue photoacoustic imaging, magnetic resonance imaging, and photothermal therapy of cancer.

Super-resolution photoacoustic imaging via flow-induced absorption fluctuations

In deep-tissue photoacoustic imaging, optical-contrast images of deep-lying structures are formed by recording acoustic waves that are generated by optical absorption. Although photoacoustics is perhaps the leading technique for high-resolution deep-tissue optical imaging, its spatial resolution is fundamentally limited by the acoustic wavelength, which is orders of magnitude longer than the optical diffraction limit. Here, we present an approach for surpassing the acoustic diffraction limit in photoacoustics by exploiting inherent temporal fluctuations in the photoacoustic signals due to sample dynamics, such as those induced by the flow of absorbing red blood cells. This was achieved using a conventional photoacoustic imaging system by adapting concepts from super-resolution fluorescence fluctuation microscopy to the statistical analysis of acoustic signals from flowing acoustic emitters. Specifically, we experimentally demonstrate that flow of absorbing particles and whole human blood yields super-resolved photoacoustic images, and provides static background reduction. By generalizing the statistical analysis to complex-valued signals, we demonstrate super-resolved photoacoustic images that are free from common photoacoustic imaging artifacts caused by band-limited acoustic detection. The presented technique holds potential for contrast-agent-free microvessel imaging, as red blood cells provide a strong endogenous source of naturally fluctuating absorption.

Photoacoustic imaging beyond the acoustic diffraction-limit with dynamic speckle illumination and sparse joint support recovery.

In deep tissue photoacoustic imaging the spatial resolution is inherently limited by the acoustic wavelength. Recently, it was demonstrated that it is possible to surpass the acoustic diffraction limit by analyzing fluctuations in a set of photoacoustic images obtained under unknown speckle illumination patterns. Here, we purpose an approach to boost reconstruction fidelity and resolution, while reducing the number of acquired images by utilizing a compressed sensing computational reconstruction framework. The approach takes into account prior knowledge of the system response and sparsity of the target structure. We provide proof of principle experiments of the approach and demonstrate that improved performance is obtained when both speckle fluctuations and object priors are used. We numerically study the expected performance as a function of the measurement's signal to noise ratio and sample spatial-sparsity. The presented reconstruction framework can be applied to analyze existing photoacoustic experimental data sets containing dynamic fluctuations.

Quantitative photoacoustic image reconstruction improves accuracy in deep tissue structures.

Photoacoustic imaging (PAI) is emerging as a potentially powerful imaging tool with multiple applications. Image reconstruction for PAI has been relatively limited because of limited or no modeling of light delivery to deep tissues. This work demonstrates a numerical approach to quantitative photoacoustic image reconstruction that minimizes depth and spectrally derived artifacts. We present the first time-domain quantitative photoacoustic image reconstruction algorithm that models optical sources through acoustic data to create quantitative images of absorption coefficients. We demonstrate quantitative accuracy of less than 5% error in large 3 cm diameter 2D geometries with multiple targets and within 22% error in the largest size quantitative photoacoustic studies to date (6cm diameter). We extend the algorithm to spectral data, reconstructing 6 varying chromophores to within 17% of the true values. This quantitiative PA tomography method was able to improve considerably on filtered-back projection from the standpoint of image quality, absolute, and relative quantification in all our simulation geometries. We characterize the effects of time step size, initial guess, and source configuration on final accuracy. This work could help to generate accurate quantitative images from both endogenous absorbers and exogenous photoacoustic dyes in both preclinical and clinical work, thereby increasing the information content obtained especially from deep-tissue photoacoustic imaging studies.

Ambient Aqueous Synthesis of Ultrasmall PEGylated Cu2-x Se Nanoparticles as a Multifunctional Theranostic Agent for Multimodal Imaging Guided Photothermal Therapy of Cancer.

Ultrasmall PEGylated Cu2-x Se nanoparticles with strong near-infrared absorption have been prepared by an ambient aqueous method. The resultant water-soluble and biocompatible nanoparticles are demonstrated to be a novel nanotheranostic agent for effective deep-tissue photoacoustic imaging, computed tomography imaging, single-photon emission computed tomography imaging, and photothermal therapy of cancer.

In vivo demonstration of reflection artifact reduction in photoacoustic imaging using synthetic aperture photoacoustic-guided focused ultrasound (PAFUSion).

Reflection artifacts caused by acoustic inhomogeneities are a critical problem in epi-mode biomedical photoacoustic imaging. High light fluence beneath the probe results in photoacoustic transients, which propagate into the tissue and reflect back from echogenic structures. These reflection artifacts cause problems in image interpretation and significantly impact the contrast and imaging depth. We recently proposed a method called PAFUSion (Photoacoustic-guided focused ultrasound) to identify such reflection artifacts in photoacoustic imaging. In its initial version, PAFUSion mimics the inward-travelling wavefield from small blood vessel-like PA sources by applying ultrasound pulses focused towards these sources, and thus provides a way to identify the resulting reflection artifacts. In this work, we demonstrate reduction of reflection artifacts in phantoms and in vivo measurements on human volunteers. In view of the spatially distributed PA sources that are found in clinical applications, we implemented an improved version of PAFUSion where photoacoustic signals are backpropagated to imitate the inward travelling wavefield and thus the reflection artifacts. The backpropagation is performed in a synthetic way based on the pulse-echo acquisitions after transmission on each single element of the transducer array. The results provide a direct confirmation that reflection artifacts are prominent in clinical epi-photoacoustic imaging, and that PAFUSion can strongly reduce these artifacts to improve deep-tissue photoacoustic imaging.

Super-resolution photoacoustic fluctuation imaging with multiple speckle illumination

In deep tissue photoacoustic imaging, the spatial resolution is inherently limited by acoustic diffraction. Moreover, as the ultrasound attenuation increases with frequency, resolution is often traded-off for penetration depth. Here we report on super-resolution photoacoustic imaging by use of multiple speckle illumination. Specifically, we show that the analysis of second-order fluctuations of the photoacoustic images combined with image deconvolution enables resolving optically absorbing structures beyond the acoustic diffraction limit. A resolution increase of almost a factor 2 is demonstrated experimentally. Our method introduces a new framework that could potentially lead to deep tissue photoacoustic imaging with sub-acoustic resolution.

A new design of light illumination scheme for deep tissue photoacoustic imaging

A new light illumination scheme to increase imaging depth in photoacoustic (PA) imaging was designed and evaluated by in silico simulations and tested by in vitro experiments. A relatively large portion of the light energy shining into the body of a human reflects off the skin surfaces. Collecting the reflected light and redirecting it onto skin surfaces will increase the effective input energy, resulting in an increase of light penetration depth for the same light source. Its performance in PA imaging was evaluated using a finite element (FE)-based numerical simulation model composed of four modules. In the in vitro experiments with the light catcher, PA image of multiple targets at different locations exhibited an enhancement both in uniformity and in depth of the light illumination.

Deep Tissue Photoacoustic Imaging Using a Miniaturized 2-D Capacitive Micromachined Ultrasonic Transducer Array

In this paper, we demonstrate 3-D photoacoustic imaging (PAI) of light absorbing objects embedded as deep as 5 cm inside strong optically scattering phantoms using a miniaturized (4 mm × 4 mm × 500 μm), 2-D capacitive micromachined ultrasonic transducer (CMUT) array of 16 × 16 elements with a center frequency of 5.5 MHz. Two-dimensional tomographic images and 3-D volumetric images of the objects placed at different depths are presented. In addition, we studied the sensitivity of CMUT-based PAI to the concentration of indocyanine green dye at 5 cm depth inside the phantom. Under optimized experimental conditions, the objects at 5 cm depth can be imaged with SNR of about 35 dB and a spatial resolution of approximately 500 μm. Results demonstrate that CMUTs with integrated front-end amplifier circuits are an attractive choice for achieving relatively high depth sensitivity for PAI.
