Golgi-localized APYRASE 1 is critical for Arabidopsis growth by affecting cell wall integrity under boron deficiency.

Many nucleoside triphosphate-diphosphohydrolases (NTPDases/APYRASEs, APYs) play a key role in modulating extracellular nucleotide levels. However, the Golgi-localized APYs, which help control glycosylation, have rarely been studied. Here, we identified AtAPY1, a gene encoding an NTPDase in the Golgi apparatus, which is required for cell wall integrity and plant growth under boron (B) limited availability. Loss of function in AtAPY1 hindered cell elongation and division in root tips while increasing the number of cortical cell layers, leading to swelling of the root tip and abundant root hairs under low B stress. Further, expression pattern analysis revealed that B deficiency significantly induced AtAPY1, especially in the root meristem and stele. Fluorescent-labeled AtAPY1-GFP localized to the Golgi stack. Biochemical analysis showed that AtAPY1 exhibited a preference of UDP and GDP hydrolysis activities. Consequently, the loss of function in AtAPY1 might disturb the homoeostasis of NMP-driven NDP-sugar transport, which was closely related to the synthesis of cell wall polysaccharides. Further, cell wall-composition analysis showed that pectin content increased and borate-dimerized RG-II decreased in apy1 mutants, along with a decrease in cellulose content. Eventually, altered polysaccharide characteristics presumably cause growth defects in apy1 mutants under B deficiency. Altogether, these data strongly support a novel role for AtAPY1 in mediating responses to low B availability by regulating cell wall integrity.

COBL7 is required for stomatal formation via regulation of cellulose deposition in Arabidopsis.

As a key regulator of plant photosynthesis, water use efficiency and immunity, stomata are specialized cellular structures that adopt defined shapes. However, our knowledge about the genetic players of stomatal pore formation and stomatal morphogenesis remains limited. Forward genetic screening, positional cloning, confocal and electron microscopy, physiological and pharmacological assays were employed for isolation and characterization of mutants and genes. We identified a mutant, dsm1, with impaired cytokinesis and deformed stomata. DSM1 is highly expressed in guard mother cells and guard cells, and encodes COBRA-LIKE 7 (COBL7), a plant-specific glycosylphosphatidylinositol (GPI)-anchored protein. COBRA-LIKE 7 and its closest homologue, COBL8, are first enriched on the forming cell plates during cytokinesis, and then their subcellular distribution and abundance change are correlated with the progressive stages of stomatal pore formation. Both COBL7 and COBL8 possess an ability to bind cellulose. Perturbing the expression of COBL7 and COBL8 leads to a decrease in cellulose content and inhibition of stomatal pore development. Moreover, we found that COBL7, COBL8 and CSLD5 have synergistic effects on stomatal development and plant growth. Our findings reveal that COBL7 plays a predominant and functionally redundant role with COBL8 in stomatal formation through regulating cellulose deposition and ventral wall modification in Arabidopsis.

Dynamic changes occur in the cell wall composition and related enzyme activities during flower development in Rosa damascena.

The flowering period of oil-bearing rose is short and many physiological processes occur during flower development. Changes in the cell wall composition and associated enzyme activities are important as they allow cells to divide, differentiate and grow. In the present study, changes in seven cell wall components and six cell wall-related enzyme activities at five flower development stages were investigated and the relationships between these parameters and flowering were examined. Ash content did not change between stages I to II but decreased at later stages. Neutral detergent fiber (NDF), acid detergent fiber (ADF) and hemicellulose contents increased after stage I but did not change significantly at the other developmental periods. Total pectin content increased throughout flower development. An "increase-decrease" trend was observed in total cellulose content and a "decrease-increase" pattern in uronic acid content. The activities of both glycosidases (β-galactosidase, β-glucosidase and endoglucanase) and pectinases (pectin lyase, pectin methyl esterase and polygalacturonase) increased until stage IV and decreased significantly at stage V of flower development. Correlation analysis revealed 14 positive and one negative correlation with the studied parameters. Cell wall enzymes showed positive correlations with each other. Principal component analysis (PCA) showed that ADF, NDF and cellulose content were significantly altered at stage II of flower development, and significant changes occurred in all cell wall enzyme activities between stages III and V. Overall, blooming is correlated closely with increased pectin and decreased cellulose contents, and changes in cell wall glucosidase and pectin hydrolysis enzyme activities. These results show that cell wall modifying enzymes are part of the flower development process in oil-bearing rose. Therefore, remodeling of cell wall components in petals is a process of flower development.

VILLIN2 regulates cotton defense against Verticillium dahliae by modulating actin cytoskeleton remodeling.

The active structural change of actin cytoskeleton is a general host response upon pathogen attack. This study characterized the function of the cotton (Gossypium hirsutum) actin-binding protein VILLIN2 (GhVLN2) in host defense against the soilborne fungus Verticillium dahliae. Biochemical analysis demonstrated that GhVLN2 possessed actin-binding, -bundling, and -severing activities. A low concentration of GhVLN2 could shift its activity from actin bundling to actin severing in the presence of Ca2+. Knockdown of GhVLN2 expression by virus-induced gene silencing reduced the extent of actin filament bundling and interfered with the growth of cotton plants, resulting in the formation of twisted organs and brittle stems with a decreased cellulose content of the cell wall. Upon V. dahliae infection, the expression of GhVLN2 was downregulated in root cells, and silencing of GhVLN2 enhanced the disease tolerance of cotton plants. The actin bundles were less abundant in root cells of GhVLN2-silenced plants than in control plants. However, upon infection by V. dahliae, the number of actin filaments and bundles in the cells of GhVLN2-silenced plants was raised to a comparable level as those in control plants, with the dynamic remodeling of the actin cytoskeleton appearing several hours in advance. GhVLN2-silenced plants exhibited a higher incidence of actin filament cleavage in the presence of Ca2+, suggesting that pathogen-responsive downregulation of GhVLN2 could activate its actin-severing activity. These data indicate that the regulated expression and functional shift of GhVLN2 contribute to modulating the dynamic remodeling of the actin cytoskeleton in host immune responses against V. dahliae.

A Golgi-localized glycosyltransferase, OsGT14;1, is required for growth of both roots and shoots in rice.

Glycosyltransferases (GTs) form a large family in plants and are important enzymes for the synthesis of various polysaccharides, but only a few members have been functionally characterized. Here, through mutant screening with gene mapping, we found that an Oryza sativa (rice) mutant with a short-root phenotype was caused by a frame-shift mutation of a gene (OsGT14;1) belonging to the glycosyltransferase gene family 14. Further analysis indicated that the mutant also had a brittle culm and produced lower grain yield compared with wild-type rice, but the roots showed similar root structure and function in terms of the uptake of mineral nutrients. OsGT14;1 was broadly expressed in all organs throughout the entire growth period, with a relatively high expression in the roots, stems, node I and husk. Furthermore, OsGT14;1 was expressed in all tissues of these organs. Subcellular observation revealed that OsGT14;1 encoded a Golgi-localized protein. Mutation of OsGT14;1 resulted in decreased cellulose content and increased hemicellulose, but did not alter pectin in the cell wall of roots and shoots. The knockout of OsGT14;1 did not affect the tolerance to toxic mineral elements, including Al, As, Cd and salt stress, but did increase the sensitivity to low pH. Taken together, OsGT14;1 located at the Golgi is required for growth of both roots and shoots in rice through affecting cellulose synthesis.

24-Epibrassinolide and 2,6-Dichlorobenzonitrile Promoted Celery Petioles and Hypocotyl Elongation by Altering Cellulose Accumulation and Cell Length

BRs (brassinosteroids), an endogenous hormone in plants, regulate cellulose accumulation, cell elongation and plant growth. Propiconazole (PCZ) is an effective inhibitor of BR biosynthesis. DCB (2,6-Dichlorobenzonitrile) can inhibit the synthesis of cellulose and affects the chemical composition of cell walls. Celery is one important leafy vegetable of the Apiaceae family with rich dietary fiber (including cellulose). The petioles length, leaf blades number and cellulose content determine the yield and quality of celery. The family members of AgCESAs are related to cellulose biosynthesis in higher plants. To investigate the effects of BRs, PCZ and DCB on the growth of celery, celery cv. ‘Jinnan Shiqin’ plants were treated with 24-epibrassinolide (24-EBL, most active form of BRs), PCZ and DCB, respectively. The results showed that exogenous application of BRs up-regulated the expression of AgCESAs genes and accumulated more cellulose in celery. The length of petioles and number of leaf blades in celery plants applied with exogenous BRs (1.24 × 10−6 mol/L 24-EBL) were increased 2.16 and 1.37 times of that in the control. The addition of PCZ inhibited the effects of exogenous BRs application. The lengths of hypocotyl and hypocotyl cells of celery plants treated with BRs were longer than that of the control. Under DCB treatments, the expression levels of AgCESAs genes in celery petioles and leaf blades were down-regulated compared with the control, and the celery plants showed decreased cellulose content, shorter petiole length and fewer leaf blades. The length of hypocotyl and hypocotyl cells of celery treated with DCB were shorter than that of the control. This study provided a reference for the functions of BRs and DCB on the growth and development of celery.

Cellulose accumulation mediated by PbrCSLD5, a cellulose synthase-like protein, results in cessation of pollen tube growth in Pyrus bretschneideri.

Cellulose, a key component of the cell wall, plays an important role in maintaining the growth of pollen tubes. However, the molecular mechanism of cellulose participating in the cessation of pear pollen tube growth remains unclear. Here, we reported that at 15 h post-cultured (HPC), the slow-growth pear pollen tubes showed thickened cell walls and cellulose accumulation in the inner wall. Transcriptome data and quantitative real-time PCR analysis showed that PbrCSLD5, a cellulose synthesis-like gene, was highly expressed in the 15 HPC pear pollen tubes. Knockdown of PbrCSLD5 caused a decrease in cellulose content in pear pollen tubes. Moreover, PbrCSLD5 overexpression in Arabidopsis resulted in the accumulation of cellulose and disruption of normal pollen tube growth. Transcription factor PbrMADS52 was found to bind to the promoter of PbrCSLD5 and enhanced its expression. Our results suggested that the PbrMADS52-PbrCSLD5 signaling pathway led to increased cellulose content in the pear pollen tube cell wall, thereby inhibiting pollen tube growth. These results provided new insights into the regulation of pollen tube growth.

CRISPR/Cas9‐mediated P‐CR domain‐specific engineering of CESA4 heterodimerization capacity alters cell wall architecture and improves saccharification efficiency in poplar

SummaryCellulose is the most abundant unique biopolymer in nature with widespread applications in bioenergy and high‐value bioproducts. The large transmembrane‐localized cellulose synthase (CESA) complexes (CSCs) play a pivotal role in the biosynthesis and orientation of the para‐crystalline cellulose microfibrils during secondary cell wall (SCW) deposition. However, the hub CESA subunit with high potential homo/heterodimerization capacity and its functional effects on cell wall architecture, cellulose crystallinity, and saccharification efficiency remains unclear. Here, we reported the highly potent binding site containing four residues of Pro435, Trp436, Pro437, and Gly438 in the plant‐conserved region (P‐CR) of PalCESA4 subunit, which are involved in the CESA4‐CESA8 heterodimerization. The CRISPR/Cas9‐knockout mutagenesis in the predicted binding site results in physiological abnormalities, stunt growth, and deficient roots. The homozygous double substitution of W436Q and P437S and heterozygous double deletions of W436 and P437 residues potentially reduced CESA4‐binding affinity resulting in normal roots, 1.5–2‐fold higher plant growth and cell wall regeneration rates, 1.7‐fold thinner cell wall, high hemicellulose content, 37%–67% decrease in cellulose content, high cellulose DP, 25%–37% decrease in cellulose crystallinity, and 50% increase in saccharification efficiency. The heterozygous deletion of W436 increases about 2‐fold CESA4 homo/heterodimerization capacity led to the 50% decrease in plant growth and increase in cell walls thickness, cellulose content (33%), cellulose DP (20%), and CrI (8%). Our findings provide a strategy for introducing commercial CRISPR/Cas9‐mediated bioengineered poplars with promising cellulose applications. We anticipate our results could create an engineering revolution in bioenergy and cellulose‐based nanomaterial technologies.

Differential Gene Expression of Brachypodium distachyon Roots Colonized by Gluconacetobacter diazotrophicus and the Role of BdCESA8 in the Colonization

Alternatives to synthetic nitrogen fertilizer are needed to reduce the costs of crop production and offset environmental damage. Nitrogen-fixing bacterium Gluconacetobacter diazotrophicus has been proposed as a possible biofertilizer for monocot crop production. However, the colonization of G. diazotrophicus in most monocot crops is limited and deep understanding of the response of host plants to G. diazotrophicus colonization is still lacking. In this study, the molecular response of the monocot plant model Brachypodium distachyon was studied during G. diazotrophicus root colonization. The gene expression profiles of B. distachyon root tissues colonized by G. diazotrophicus were generated via next-generation RNA sequencing, and investigated through gene ontology and metabolic pathway analysis. The RNA sequencing results indicated that Brachypodium is actively involved in G. diazotrophicus colonization via cell wall synthesis. Jasmonic acid, ethylene, gibberellin biosynthesis. nitrogen assimilation, and primary and secondary metabolite pathways are also modulated to accommodate and control the extent of G. diazotrophicus colonization. Cellulose synthesis is significantly downregulated during colonization. The loss of function mutant for Brachypodium cellulose synthase 8 (BdCESA8) showed decreased cellulose content in xylem and increased resistance to G. diazotrophicus colonization. This result suggested that the cellulose synthesis of the secondary cell wall is involved in G. diazotrophicus colonization. The results of this study provide insights for future research in regard to gene manipulation for efficient colonization of nitrogen-fixing bacteria in Brachypodium and monocot crops.[Formula: see text] Copyright © 2021 The Author(s). The HD-Zip IV transcription factor SlHDZIV8 controls multicellular trichome morphology by regulating the expression of Hairless-2.

Trichomes are specialized epidermal appendages that serve as excellent models to study cell morphogenesis. Although the molecular mechanism underlying trichome morphogenesis in Arabidopsis has been well characterized, most of the regulators essential for multicellular trichome morphology remain unknown in tomato. In this study, we determined that the recessive hairless-2 (hl-2) mutation in tomato causes severe distortion of all trichome types, along with increased stem fragility. Using map-based cloning, we found that the hl-2 phenotype was associated with a 100 bp insertion in the coding region of Nck-associated protein 1, a component of the SCAR/WAVE complex. Direct protein-protein interaction was detected between Hl-2 and Hl (SRA1, specifically Rac1-associated protein) using yeast two-hybrid and co-immunoprecipitation assays, suggesting that these proteins may work together during trichome formation. In addition, knock-down of a HD-Zip IV transcription factor, HDZIPIV8, distorted trichomes similar to the hl-2 mutant. HDZIPIV8 regulates the expression of Hl-2 by binding to the L1-box in the Hl-2 promoter region, and is involved in organizing actin filaments. The brittleness of hl-2 stems was found to result from decreased cellulose content. Taken together, these findings suggest that the Hl-2 gene plays an important role in controlling multicellular trichome morphogenesis and mechanical properties of stems in tomato plants.

Isolation and identification of fungi inhabiting rubber-impregnated wood, and their role of quality changing the impregnated wood

Knowledge of the types of fungi inhabiting wood, their growth conditions, and level of damage is needed to design an effective and efficient preservation process. Researchers have tried to identify fungi that play a role in attacking wood products (impregnated wood), including the type and the role of these fungi in changing the quality of processed wood. Isolation was completed on fungal-attacked wood. The first isolation resulted in approximately 26 isolates, which were then grouped into 8 groups based on the macroscopic appearance of the fungus. Molecular identification of the fungi produced types of Aspergillus tamarii, Penicillium citrinum, Trichoderma longibrachiatum, Trichoderma reesei, and Fusarium solani. Chemical analysis using the Chasson Datta method was performed on attacked and non-attacked wood. A decrease was found in cellulose content and an increase in lignin content for impregnated wood attacked by fungi. It was suspected that there were cellulose-attacking fungi that consumed cellulose, which caused a decrease in cellulose content. An increasing lignin content was thought to be caused by phenols, produced by the delignification process by fungi and additional phenols from the impregnation process. Fourier transform infrared results revealed the presence of carbohydrates, showing that the degradation of cellulose by fungi produced simple carbohydrates.

Alkaline pretreatment of sugarcane bagasse on pilot scale reactor

Sugarcane bagasse is one of the potential raw materials for producing bioethanol based on lignocellulosic biomass.The aim of this study is to determine the percentage of lignocellulose over the duration of the pretreatment delignification process.The delignification of pilot scale in the form of a reactor batch has a volume capacity of 147 L with a working volume capacity of 120 L. Sugarcane bagasse used in this process was 3 kg per batch with a size of 50 mesh. The pretreatment method was carried out chemically using NaOH at a concentration of 1 N, a working temperature of 120°C with a pressure of 2 bar and the variation of the delignification time was 1,1.5, 2, 2.5, 3, and 3.5 hours.The highest cellulose content from this study was found at delignification time of 1.5 hours which was 77.26%, greater than the sugarcane bagasse without pretreatment, with a cellulose content of 43.48%. At reaction time of 2 to 3.5 hours, the results showed a decrease in cellulose content with an average gain of 74.63%.Thus increasing the time in the chemical delignification pretreatment process did not affect the increase in cellulose percentage acquisition.

The effectiveness of various compositions lignolytic and cellulolytic microbes in composting empty fruit bunch palm oil and sugar cane biomass

In sustainable agriculture, the balance of the use of inorganic fertilizers, organic matter and biofertilizers are highly recommended to increase crop production and at the same time maintaining or even improving the soil health and quality. Farm waste such as palm oil empty bunches and chopped sugar cane biomass are abundant and is a useful material to be returned to the soil to fertilize the plants. Constraints of both the material utilization is a very long process of decomposed because it contains lignin and cellulose and difficult to decomposed by microbes. Organic materials palm oil empty bunches and sugar cane biomass inoculated cellulolytic fungi, cellulolytic bacteria, lignolytic fungi, lignolytic bacteria consortia 1%, 2%, 3%, 4%, and without decomposers. The experiments were conducted in randomized block design (RBD) with four replications. Composting each 1 m3 of organic matter carried out aerobically for one month. Largest decrease in cellulose content of palm oil empty bunches and sugar cane biomass obtain on inoculation cellulolytic and lignolytic microbes that was 4% respectively to 64.92% and 61.12%, while the largest decrease in lignin content on palm oil empty bunches contained of 3% decomposer with cellulolytic and lignolytic microbes. While on sugar cane biomass obtained by 4% decomposers cellulolytic and lignolytic microbes. The addition of containing 2% cellulolytic and lignolytic most effective in decreasing the C/N either palm oil empty bunches or sugar cane biomass.

Synergistic sorption performance of cellulose-modified La0.9Sr0.1FeO3 for organic pollutants

La0.9Sr0.1FeO3 was successfully prepared by the cellulose-modified microwave-assisted citrate method at relatively low calcination temperatures of 450 and 600 °C. Prepared unmodified and cellulose-modified samples were characterized by X-ray diffraction, scanning electron microscope, FTIR, and BET surface area measurements. Characterization techniques ascertained the formation of LaFeO3 perovskite phase with the coexistence of the cellulosic phase for modified samples. The cellulose modification resulted in smaller particle size and higher BET values. The adsorption performance of cellulose-modified perovskite for congo red dye removal from aqueous solutions, and wastewater was investigated. The adsorption followed pseudo 1st order and intra-particle diffusion kinetic models, the maximum adsorption capacity, qm of cellulose-modified La0.9Sr0.1FeO3, 38.46 mg g−1, was higher than those of either individual cellulose and unmodified La0.9Sr0.1FeO3, 18.2 and 13.89 mg g−1, respectively. The value of qm was decreased by increasing the calcination temperature due to the decreased cellulose content. The proposed sorbent could be regenerated and reused, with an excellent performance, compared to the freshly prepared one.

Methyltrioctylammonium chloride mediated removal of lignin from sugarcane bagasse for themostable cellulase production

The present study was aimed to evaluate the Methyltrioctylammonium Chloride (IL) and Sodium hydroxide effect on sugarcane bagasse (SB) structure and its subsequent utilization to produce cellulase from a thermophilic bacterium Bacillus aestuarii UE25. The strain was isolated from a crocodile pond of Manghopir, Karachi. Ten different factors affecting IL pretreatment of SB and cellulase production by UE25 were evaluated by Plackett-Burman design and three significant factors were optimized by employing Box Behnken design. Under optimum conditions, the strain produced 118.4 IU mL−1 of EG and 75.01 IU mL−1 of BGL that corroborated well with the predicted values by the model. Scanning electron microscopy, gravimetric analysis, Fourier transform infrared spectroscopy and NMR of SB revealed removal of lignin, decrease in cellulose content and structural changes in the SB after pretreatment and fermentation. The data provide prospects of utilizing this IL in comparison to imidazolium based IL for pretreatment of biomass.

Involvement of CesA4, CesA7-A/B and CesA8-A/B in secondary wall formation in Populus trichocarpa wood.

Cellulose synthase A genes (CesAs) are responsible for cellulose biosynthesis in plant cell walls. In this study, functions of secondary wall cellulose synthases PtrCesA4, PtrCesA7-A/B and PtrCesA8-A/B were characterized during wood formation in Populus trichocarpa (Torr. & Gray). CesA RNAi knockdown transgenic plants exhibited stunted growth, narrow leaves, early necrosis, reduced stature, collapsed vessels, thinner fiber cell walls and extended fiber lumen diameters. In the RNAi knockdown transgenics, stems exhibited reduced mechanical strength, with reduced modulus of rupture (MOR) and modulus of elasticity (MOE). The reduced mechanical strength may be due to thinner fiber cell walls. Vessels in the xylem of the transgenics were collapsed, indicating that water transport in xylem may be affected and thus causing early necrosis in leaves. A dramatic decrease in cellulose content was observed in the RNAi knockdown transgenics. Compared with wildtype, the cellulose content was significantly decreased in the PtrCesA4, PtrCesA7 and PtrCesA8 RNAi knockdown transgenics. As a result, lignin and xylem contents were proportionally increased. The wood composition changes were confirmed by solid-state NMR, two-dimensional solution-state NMR and sum-frequency-generation vibration (SFG) analyses. Both solid-state nuclear magnetic resonance (NMR) and SFG analyses demonstrated that knockdown of PtrCesAs did not affect cellulose crystallinity index. Our results provided the evidence for the involvement of PtrCesA4, PtrCesA7-A/B and PtrCesA8-A/B in secondary cell wall formation in wood and demonstrated the pleiotropic effects of their perturbations on wood formation.

PagMYB216 is involved in the regulation of cellulose synthesis in Populus

Cellulose accounts for most of the structural material in secondary cell walls. Although the synthesis of cellulose has been extensively investigated, the regulation of cellulose synthesis in woody plants remains elusive. In this study, we showed PagMYB216 to be involved in the regulation of cellulose synthesis in Populus. Subcellular localization and transcriptional activation analyses suggest that PagMYB216 is a nuclear-localized transcriptional activator. Transient transcriptional assays revealed that PagMYB216 could activate the expression of secondary cell wall cellulose synthesis genes (CESAs) through binding to the GAMYB motif. Overexpression of PagMYB216 in Populus resulted in enhanced expression of secondary wall CESAs and increased cellulose content in stems. In contrast, the dominant repression of PagMYB216 transgenic poplar plants led to the downregulation of secondary wall CESAs and decreased cellulose content. In addition, cellulose deposition and secondary wall thickness were increased in stems from PagMYB216 overexpression poplar plants and decreased in stems from PagMYB216 dominant repression poplar plants. Taken together, our results suggest that PagMYB216 is involved in the modulation of cellulose synthesis in Populus by controlling the expression of secondary wall CESAs.

Brittle Culm 1 Encodes a COBRA-Like Protein Involved in Secondary Cell Wall Cellulose Biosynthesis in Sorghum.

Plant mechanical strength contributes to lodging resistance and grain yield, making it an agronomically important trait in sorghum (Sorghum bicolor). In this study, we isolated the brittle culm 1 (bc1) mutant and identified SbBC1 through map-based cloning. SbBC1, a homolog of rice OsBC1 and Arabidopsis thaliana AtCOBL4, encodes a COBRA-like protein that exhibits typical structural features of a glycosylphosphatidylinositol-anchored protein. A single-nucleotide mutation in SbBC1 led to reduced mechanical strength, decreased cellulose content, and increased lignin content without obviously altering plant morphology. Transmission electron microscopy revealed reduced cell wall thickness in sclerenchyma cells of the bc1 mutant. SbBC1 is primarily expressed in developing sclerenchyma cells and vascular bundles in sorghum. RNA-seq analysis further suggested a possible mechanism by which SbBC1 mediates cellulose biosynthesis and cell wall remodeling. Our results demonstrate that SbBC1 participates in the biosynthesis of cellulose in the secondary cell wall and affects the mechanical strength of sorghum plants, providing additional genetic evidence for the roles of COBRA-like genes in cellulose biosynthesis in grasses.

THE ROLE OF VEGETABLE BREEDING AND MODERN RESEARCHES IN FOOD STABILITY

The vegetable production is one of the main factors for food security, as vegetables play an important role in the structure of nutrition. The Federal Scientific Vegetable Center creates competitive varieties and hybrids of vegetable crops that meet market requirements to meet the needs of domestic producers and thereby contributes to food security. As a result of national breeding program, the varieties and hybrids adapted to different growing conditions, with resistance to local races of pathogens, temperature stresses, and ground frosts were developed to provide the sustainable production of vegetables with high nutritional and medicinal qualities. The varieties and hybrids F1 of white head cabbage that have been created for the last 5 years are distinguished from foreign ones by taste qualities, appropriate pickling characteristics with increased sugar, and decreased cellulose contents. The local onion varieties combine long shelf life, early maturing, well bulb formation, high dry matter content (18-20%) and ability to form the bulb for one year. Breeders have developed bee-pollinated and parthenocarpic hybrids of cucumber well adapted to local growing conditions and suitable for open field cultivation in different regions of Russia. Parthenocarpic heterotic hybrids of the multi-propose use, corresponding to the modern variety model with high productivity, early-ripening, bunch ovary disposition, resistance to abiotic stresses, and most harmful diseases. The carrot and red beet varieties with high nutritional qualities, long shelf-life, ecological plasticity that are widely used for seed production have been created. The varieties of Solanaceae crops have been developed to cultivate in Non Chernozem zone, Far East, Western Siberia, the Middle Belt of Russia and the south of Russia, are also suitable for different greenhouse complexes located in different light zones of the Russia. Wide application of varieties produced through national vegetable breeding programs can provide the population with our own products of high quality.

OsSND2, a NAC family transcription factor, is involved in secondary cell wall biosynthesis through regulating MYBs expression in rice

BackgroundAs one of the most important staple food crops, rice produces huge agronomic biomass residues that contain lots of secondary cell walls (SCWs) comprising cellulose, hemicelluloses and lignin. The transcriptional regulation mechanism underlying SCWs biosynthesis remains elusive.ResultsIn this study, we isolated a NAC family transcription factor (TF), OsSND2 through yeast one-hybrid screening using the secondary wall NAC-binding element (SNBE) on the promoter region of OsMYB61 which is known transcription factor for regulation of SCWs biosynthesis as bait. We used an electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation analysis (ChIP) to further confirm that OsSND2 can directly bind to the promoter of OsMYB61 both in vitro and in vivo. OsSND2, a close homolog of AtSND2, is localized in the nucleus and has transcriptional activation activity. Expression pattern analysis indicated that OsSND2 was mainly expressed in internodes and panicles. Overexpression of OsSND2 resulted in rolled leaf, increased cellulose content and up-regulated expression of SCWs related genes. The knockout of OsSND2 using CRISPR/Cas9 system decreased cellulose content and down-regulated the expression of SCWs related genes. Furthermore, OsSND2 can also directly bind to the promoters of other MYB family TFs by transactivation analysis in yeast cells and rice protoplasts. Altogether, our findings suggest that OsSND2 may function as a master regulator to mediate SCWs biosynthesis.ConclusionOsSND2 was identified as a positive regulator of cellulose biosynthesis in rice. An increase in the expression level of this gene can improve the SCWs cellulose content. Therefore, the study of the function of OsSND2 can provide a strategy for manipulating plant biomass production.
