Impact of chronic fluoxetine exposure on zebrafish: From fatty acid profile to behavior

The consumption of antidepressants, such as fluoxetine, has increased over the years and, as a result, they are increasingly found in aquatic systems. Given the increasing use of zebrafish as an animal model in toxicological studies, this work proposed to evaluate the effects of chronic exposure, for 21 days, to fluoxetine at environmentally relevant concentrations (1, 10, 100, and 1000 ng/L). The behavioral tests performed did not reveal significant effects of fluoxetine. However, oxidative stress and changes in energy metabolism were detected after exposure to the highest concentrations of fluoxetine tested, namely a decrease in glutathione S-transferase (GST) activity (decrease of ca. 31%), increase in catalase (CAT) activity (increase of ca. 71%), and decrease in lactate dehydrogenase (LDH) activity (decrease of ca. 53%). Analysis of the fatty acid profile (FA) revealed a decrease in the omega-3 FA, docosahexaenoic acid (DHA), C22:6 (decrease in relative abundance between 6% and 8% for both the head and body), an increase in omega-6 FA, linoleic acid (LA), C18:2, (increased relative abundance between 8% and 11% in the head and between 5% and 9% in the body), which may suggest changes in the inflammatory state of these organisms. The integrated analysis adopted proved to be useful in detecting subindividual effects of fluoxetine and modes of action in fish.

Herbicide application impacted soil microbial community composition and biochemical properties in a flooded rice field

Herbicide application is a common practice in intensive agriculture. However, accumulating herbicide residues in the ecosystem affects important soil attributes. The effect of two herbicides, pendimethalin and pretilachlor, on soil biochemical properties and microbial community composition was studied in a transplanted paddy field. Results reveal a gradual decline in herbicide residue up to 60 days after application. Changes in soil microbiological and biochemical properties (microbial biomass, enzymes, respiration, etc.) showed an inconsistent pattern across the treatments. Quantitative polymerase chain reaction analysis showed the archaeal, bacterial and fungal populations to be of higher order in control soil compared to the treated one. Amplicon sequencing (16S rRNA and ITS genes) exhibited that besides the unclassified genera, ammonia-oxidizing Crenarchaeota and the group represented by Candidatus Nitrososphaera were dominant in both the control and treated samples. Other archaeal genera viz. Methanosarcina and Bathyarchaeia showed a slight decrease in relative abundance of control (0.5 %) compared to the treated soil (0.7 %). Irrespective of treatments, the majority of bacterial genera comprised unclassified and uncultured species, accounting for >64–75 % in the control group and over 78.29 % in the treated samples. Members of Vicinamibacteraceae, Bacillus and Bryobacter were dominant in control samples. Dominant fungal genera belonging to unclassified groups comprised Curvularia, Aspergillus, and Emericellopsis in the control group, whereas Paraphysoderma and Emericellopsis in the herbicide-treated groups. Inconsistent response of soil properties and microbial community composition is evident from the present study, suggesting that the recommended dose of herbicides might not result in any significant change in microbial community composition. The findings of this investigation will help in the formulation of a framework for risk assessment and maintaining sustainable rice cultivation in herbicide- amended soils.

Alteration in the gut microbiome is associated with changes in bone metabolism after laparoscopic sleeve gastrectomy.

Laparoscopic sleeve gastrectomy (LSG), the most common bariatric surgical procedure, leads to durable weight loss and improves obesity-related comorbidities. However, it induces abnormalities in bone metabolism. One unexplored potential contributor is the gut microbiome, which influences bone metabolism and is altered after surgery. We characterized the relationship between the gut microbiome and skeletal health in severe obesity and after LSG. In a prospective cohort study, 23 adults with severe obesity underwent skeletal health assessment and stool collection preoperatively and 6mo after LSG. Gut microbial diversity and composition were characterized using 16S rRNA gene sequencing, and fecal concentrations of short-chain fatty acids (SCFA) were measured with LC-MS/MS. Spearman's correlations and PERMANOVA analyses were applied to assess relationships between the gut microbiome and bone health measures including serum bone turnover markers (C-terminal telopeptide of type 1 collagen [CTx] and procollagen type 1N-terminal propeptide [P1NP]), areal BMD, intestinal calcium absorption, and calciotropic hormones. Six months after LSG, CTx and P1NP increased (by median 188% and 61%, P< .01) and femoral neck BMD decreased (mean -3.3%, P< .01). Concurrently, there was a decrease in relative abundance of the phylum Firmicutes. Although there were no change in overall microbial diversity or fecal SCFA concentrations after LSG, those with greater within-subject change in gut community microbial composition (β-diversity) postoperatively had greater increases in P1NP level (ρ= 0.48, P= .02) and greater bone loss at the femoral neck (ρ= -0.43, P= .04). In addition, within-participant shifts in microbial richness/evenness (α-diversity) were associated with changes in IGF-1 levels (ρ= 0.56, P< .01). The lower the postoperative fecal butyrate concentration, the lower the IGF-1 level (ρ= 0.43, P= .04). Meanwhile, the larger the decrease in butyrate concentration, the higher the postoperative CTx (ρ= -0.43, P= .04). These findings suggest that LSG-induced gut microbiome alteration may influence skeletal outcomes postoperatively, and microbial influences on butyrate formation and IGF-1 are possible mechanisms.

Triclosan disturbs nitrogen removal in constructed wetlands: Responses of microbial structure and functions

This study investigated the influence of wetland types (vertical and tidal flow constructed wetlands [CWs] [VFCW and TFCW, respectively]) and concentrations of triclosan (TCS) on the removal of pollutants (TCS and nitrogen) and microbial characteristics. The efficiency of TCS removal was significantly higher with 5 μg/L TCS (Phase B) than with 30 μg/L (Phase C) in the two CWs. The efficiencies of removal of NH4+-N and NO3−-N were significantly inhibited in Phase C. Compared with the VFCW, the TFCW removed more NH4+-N at the same concentration of TCS, whereas less NO3−-N was removed, and it even accumulated. Saccharimondales, an important functional genus with the highest abundance and more node connections with other genera, had a sharp decrease in relative abundance as the increasing concentrations of TCS of the two CWs conformed with its relative abundance and significantly negatively correlated with the concentration of TCS. Differentiated Roseobacter_Clade_CHAB-I-5_Lineage and Sphaerotilus were enriched in the VFCW and TFCW, respectively. The abundance of enzymes that catalyzed nitritation was significantly inhibited by TCS, whereas nitrate reductase (EC 1.7.99.4) catalyzed both denitrification and dissimilatory nitrate reduction to ammonium (DNRA), and nitrite reductase (NADH) (EC 1.7.1.15) that catalyzed DNRA comprised a larger proportion in the two CWs. Simultaneously, the abundances of two enzymes were higher in the TFCW than in the VFCW. The network analysis indicated that the main genera were promoted more by TCS in the VFCW, while inhibited in the TFCW. Moreover, the concentrations of nitrogen (NH4+-N, NO3−-N, and TN) significantly positively correlated with TCS-resistant bacteria, and negatively correlated with most nitrogen-transforming bacteria with species that varied between the VFCW and TFCW. The results of this study provide a reference for the molecular biological mechanism of the simultaneous removal of nitrogen and TCS in the CWs.

Insight in molecular degradation patterns and co-metabolism during rose waste co-composting

Composting is recognized as a sustainable waste management strategy. However, little is known about green waste, and specifically rose waste, degradation patterns during composting. This study aimed (1) to gain insight in the underlying decomposition patterns during rose waste composting and (2) to identify co-metabolisms of ligneous material. Five different compost mixtures were tested ranging from pure rose waste to mixtures with tomato waste, kalanchoe waste or mature compost added. Samples were taken during a six-month experiment and analyzed by pyrolysis-GC/MS. The temporal trends in the relative abundance of 10 different compound groups were measured. Lignin and aliphatic compounds together accounted for ≥ 50% of the quantified pyrolysis products, but with changing contributions during composting. The relative abundance of polysaccharides and terpenes strongly decreased with more than 60% in the first 2 months. The simultaneous decrease in relative abundance of lignin and polysaccharides during initial composting phase indicated co-metabolism of lignin. The results from this study showed that while the presence of lignin is commonly regarded as a challenge in composting, it actually undergoes degradation through distinct mechanisms at the various composting stages.

Longitudinal Microbiome Changes in Supragingival Biofilm Transcriptomes Induced by Orthodontics.

Common oral diseases are known to be associated with dysbiotic shifts in the supragingival microbiome, yet most oral microbiome associations with clinical end points emanate from cross-sectional studies. Orthodontic treatment is an elective procedure that can be exploited to prospectively examine clinically relevant longitudinal changes in the composition and function of the supragingival microbiome. A longitudinal cohort study was conducted among 24 adolescent orthodontic patients who underwent saliva and plaque sampling and clinical examinations at time points: before fixed appliance bonding and at 1, 6, and 12 wk thereafter. Clinical indices included bleeding on probing (BOP), mean gingival index (GI), probing depths (PDs), and plaque index (PI). To study the biologically (i.e., transcriptionally) active microbial communities, RNA was extracted from plaque and saliva for RNA sequencing and microbiome bioinformatics analysis. Longitudinal changes in microbiome beta diversity were examined using PERMANOVA tests, and the relative abundance of microbial taxa was measured using Kruskal-Wallis tests, Wilcoxon rank-sum tests, and negative binomial and zero-inflated mixed models. Clinical measures of oral health deteriorated over time-the proportion of sites with GI and PI ≥1 increased by over 70% between prebonding and 12 wk postbonding while the proportion of sites with PD ≥4 mm increased 2.5-fold. Streptococcus sanguinis, a health-associated species that antagonizes cariogenic pathogens, showed a lasting decrease in relative abundance during orthodontic treatment. Contrarily, caries- and periodontal disease-associated taxa, including Selenomonas sputigena, Leptotrichia wadei, and Lachnoanaerobaculum saburreum, increased in abundance after bonding. Relative abundances of Stomatobaculum longum and Mogibacterium diversum in prebonding saliva predicted elevated BOP 12 wk postbonding, whereas Neisseria subflava was associated with lower BOP. This study offers insights into longitudinal community and species-specific changes in the supragingival microbiome transcriptome during fixed orthodontic treatment, advancing our understanding of microbial dysbioses and identifying targets of future health-promoting clinical investigations. Bonding braces was associated with subsequent changes in the oral microbiome characterized by increases in disease-associated species, decreases in health-associated species, and worsened clinical measures of oral health.

Rhizosphere microbiome assembly mediated by consecutive monoculture triggers the replant disease of Rehmannia glutinosa

Consecutive monoculture of Rehmannia glutinosa results in severe replant disease and remarkably reduces the yield and quality. Rhizosphere microbiome plays a crucial role in soil health and plant performance. In this study, high-throughput pyrosequencing combined with a culture-dependent approach were applied to analyze the changes in rhizosphere microbiome structure and bacteria-fungi inter-kingdom associations under R. glutinosa consecutive monoculture. The results revealed a distinct separation between the newly planted (NP) and the two-year monocultured (SM). R. glutinosa consecutive monoculture resulted in a significant decline in the Shannon and Simpson diversity indices for the fungal community, and a significant decrease in relative abundances of the phyla Actinobacteria, Chloroflexi and Basidiomycota. The relative abundances of the genera Streptomyces, Arthrobacter, Bacillus, Nocardioides and Lysobacter were significantly higher in NP than in SM, while the opposite was true for Fusarium. Quantitative PCR confirmed a significant reduction in the phylum Actinobacteria and the genus Bacillus, and a significant increase in Fusarium oxysporum. Furthermore, the abundances of Actinobacteria and Bacillus strains with antagonistic activities against Fusarium oxysporum were lower in extended monoculture soils. Most of the isolated antagonistic bacteria were found to be positive for different plant growth promotion activities such as solubilization of phosphate and potassium, production of siderophore and indolic compounds. In addition, it was found that phenolic acid mixture and catalpol, identified in the root exudates of R. glutinosa, could selectively inhibit or stimulate different microbes. Moreover, the culture filtrate and volatile organic compounds produced by F. oxysporum could in turn suppress the growth of certain antagonistic bacteria. In conclusion, these findings highlight the close association between R. glutinosa replant disease and rhizosphere microbiome assembly, which was mediated by root exudates and microbe-microbe interactions.

Microbiome Alterations Driven by Trypanosoma cruzi Infection in Two Disjunctive Murine Models.

Alterations caused by Trypanosoma cruzi in the composition of gut microbiome may play a vital role in the host-parasite interactions that shapes physiology and immune responses against infection. Thus, a better understanding of this parasite-host-microbiome interaction may yield relevant information in the comprehension of the pathophysiology of the disease and the development of new prophylactic and therapeutic alternatives. Therefore, we implemented a murine model with two mice strains (BALB/c and C57BL/6) to evaluate the impact of Trypanosoma cruzi (Tulahuen strain) infection on the gut microbiome utilizing cytokine profiling and shotgun metagenomics. Higher parasite burdens were observed in cardiac and intestinal tissues, including changes in anti-inflammatory (interleukin-4 [IL-4] and IL-10) and proinflammatory (gamma interferon, tumor necrosis factor alpha, and IL-6) cytokines. Bacterial species such as Bacteroides thetaiotaomicron, Faecalibaculum rodentium, and Lactobacillus johnsonii showed a decrease in relative abundance, while Akkermansia muciniphila and Staphylococcus xylosus increased. Likewise, as infection progressed, there was a decrease in gene abundances related to metabolic processes such as lipid synthesis (including short-chain fatty acids) and amino acid synthesis (including branched-chain amino acids). High-quality metagenomic assembled genomes of L. johnsonii and A. muciniphila among other species were reconstructed, confirming, functional changes associated with metabolic pathways that are directly affected by the loss of abundance of specific bacterial taxa. IMPORTANCE Chagas disease (CD) is caused by the protozoan Trypanosoma cruzi, presenting acute and chronic phases where cardiomyopathy, megaesophagus, and/or megacolon stand out. During the course of its life cycle, the parasite has an important gastrointestinal tract transit that leads to severe forms of CD. The intestinal microbiome plays an essential role in the immunological, physiological, and metabolic homeostasis of the host. Therefore, parasite-host-intestinal microbiome interactions may provide information on certain biological and pathophysiological aspects related to CD. The present study proposes a comprehensive evaluation of the potential effects of this interaction based on metagenomic and immunological data from two mice models with different genetic, immunological, and microbiome backgrounds. Our findings suggest that there are alterations in the immune and microbiome profiles that affect several metabolic pathways that can potentially promote the infection's establishment, progression, and persistence. In addition, this information may prove essential in the research of new prophylactic and therapeutic alternatives for CD.

Effect of Citrus aurantium juice as a disinfecting agent on quality and bacterial communities of striped catfish steaks stored at -20 °C.

Sodium hypochlorite is generally used as a disinfectant in washing of freshwater fishes where the safety aspect of health is of concern. Although plant-based essential oils and synthetic chemical agents have been applied, they might contain toxic substances, are expensive and can cause undesirable quality. This research aims to fill the knowledge gap necessary to validate Citrus aurantium juice as a disinfecting agent for preserving striped catfish steaks at -20 °C for 28 days. Fifty (50) ppm sodium hypochlorite was used as a commercial disinfectant (control). The results showed that a negative color characteristic (higher a* and increased b*) was found in the control but not in striped catfish steaks immersed in C. aurantium juice (TM) on days 14 and 28. No significant differences were found in the peroxide value among the treatments on days 14 and 28 (P > 0.05). A lower accumulation of trichloroacetic acid soluble peptides was detected in TM but not in control, while total volatile basic nitrogen of all treatments was up to standard of fish quality during storage. Contrastingly, the total viable count of both treatments increased to >7.0 log CFU/g on day 28 which did not meet the edible limit of standard for freshwater fishes. The spoilage microbial community was observed on days 0 and 28 of storage which showed a decrease in relative abundance of Acinetobacter, Pseudomonas, Brochothrix, Lactococcus, Carnobacterium, Psychrobacter, and Vagococcus as found in TM on day 28, when compared to the control. Thus, these results implied that C. aurantium juice could replace sodium hypochlorite as an alternative disinfecting agent to control the microbiological spoilage and physico-chemical quality of striped catfish steaks.

Candidatus Scalindua, a Biological Solution to Treat Saline Recirculating Aquaculture System Wastewater

Recirculating aquaculture systems (RAS) are promising candidates for the sustainable development of the aquaculture industry. A current limitation of RAS is the production and potential accumulation of nitrogenous wastes, ammonium (NH4+), nitrite (NO2−) and nitrate (NO3−), which could affect fish health and welfare. In a previous experiment, we have demonstrated that the marine anammox bacteria Candidatus Scalindua was a promising candidate to treat the wastewater (WW) of marine, cold-water RAS. However, the activity of the bacteria was negatively impacted after a direct exposure to RAS WW. In the current study, we have further investigated the potential of Ca. Scalindua to treat marine RAS WW in a three-phase experiment. In the first phase (control, 83 days), Ca. Scalindua was fed a synthetic feed, enriched in NH4+, NO2− and trace element (TE) mix. Removal rates of 98.9% and 99.6% for NH4+ and NO2−, respectively, were achieved. In the second phase (116 days), we gradually increased the exposure of Ca. Scalindua to nitrogen-enriched RAS WW over a period of about 80 days. In the last phase (79 days), we investigated the needs of TE supplementation for the Ca. Scalindua after they were fully acclimated to 100% RAS WW. Our results show that the gradual exposure of Ca. Scalindua resulted in a successful acclimation to 100% RAS WW, with maintained high removal rates of both NH4+ and NO2− throughout the experiment. Despite a slight decrease in relative abundance (from 21.4% to 16.7%), Ca. Scalindua remained the dominant species in the granules throughout the whole experiment. We conclude that Ca. Scalindua can be successfully used to treat marine RAS WW, without the addition of TE, once given enough time to acclimate to its new substrate. Future studies need to determine the specific needs for optimal RAS WW treatment by Ca. Scalindua at pilot scale.

Influence of Lactobacillus reuteri, Bifidobacterium animalis subsp. lactis, and prebiotic inulin on dysbiotic dental biofilm composition ex vivo.

Probiotic bacterial supplementation has shown promising results in the treatment of periodontitis and the maintenance of periodontal health. The purpose of this investigation was to evaluate the influence of Lactobacillus reuteri or Bifidobacterium animalis subsp. lactis supplementation with and without prebiotic inulin on biofilm composition using an ex vivo biofilm model. Subgingival plaque specimens from three periodontitis-affected human donors were used to grow biofilms on hydroxyapatite disks in media supplemented with varying combinations of prebiotic inulin, Lactobacillus reuteri, and Bifidobacterium animalis subsp. lactis. Relative abundances of bacterial genera present in mature biofilms were evaluated using 16S rRNA next-generation sequencing. Diversity metrics of microbial communities were evaluated using a next-generation microbiome bioinformatics platform. Inulin supplementation produced statistically significant dose-dependent increases in relative abundances of Lactobacillus and Bifidobacterium species (p<0.001) with concomitant decreases in relative abundances of Streptococcus, Veillonella, Fusobacterium, Parvimonas, and Prevotella species (p<0.001). Inoculation with L. reuteri or B. animalis subsp. lactis increased the relative abundance of only the supplemented probiotic genera (p<0.05). Supplemental inulin led to a statistically significant decrease in biofilm alpha diversity (p<0.001). The described ex vivo model appears suitable for investigating the effects of probiotic bacteria, prebiotic oligosaccharides, and combinations thereof on biofilm composition and complexity. Within the limitations imposed by this model, results from the present study underscore the potential for prebiotic inulin to modify biofilm composition favorably. Additional research further elucidating biologic rationale and controlled clinical research defining therapeutic benefits is warranted.

The moderate substitution of Astragalus sinicus returning for chemical fertilizer improves the N cycle function of key ecological bacterial clusters in soil.

Astragalus sinicus (Chinese milk vetch) is a well-established resource of organic fertilizer widely used in paddy soil to partially replace chemical fertilizers. However, the influence of returning A. sinicus to fields on the soil bacterial community remains poorly understood. Here, we used different amounts of A. sinicus partially replacing chemical fertilizers and investigated the changes in soil physicochemical factors and the soil bacterial community structure responses. Returning A. sinicus to the field significantly increased the soil total nitrogen and available phosphorus content (p < 0.05). Weighted gene correlation network analysis (WGCNA) was applied to detect significant associations between the soil microbiome data and physicochemical factors. Two key ecological bacterial clusters (MEturquoise and MEgreen), mainly containing Acidobacteria, Proteobacteria, and Chloroflexi, were significantly correlated with soil nitrogen (N) levels. A. sinicus partially replacing chemical fertilizers reduced the normalized stochasticity ratio (NST) of rare amplicon sequence variants (ASVs), abundant ASVs, MEturquoise, and MEgreen (p < 0.05). Our results further indicated that a moderate amount of A. sinicus returned to the soil effectively mitigated the trend of reduced relative abundance of N fixation function of key ecological clusters caused by chemical fertilizer. However, a large amount of A. sinicus led to a significant increase in relative abundance of denitrification function and a significant decrease in relative abundance of N fixation function of key ecological clusters. This implies that the moderate substitution of A. sinicus returning for chemical fertilizer improves the N cycling function of key ecological bacterial clusters in soil. From the perspective of the bacterial community in paddy soil, this study provides new insight and a reference on how to find a good balance between the amount of A. sinicus returned to the soil and ecological safety.

Response of Pacific halibut (Hippoglossus stenolepis) to future climate scenarios in the Northeast Pacific Ocean

Pacific halibut (Hippoglossus stenolepis) are a large-bodied species of flatfish that are important culturally, economically, and as a key predator in marine systems in the USA and Canada. The species has a wide distribution, and complex life history including large-scale migrations to spawn and feed, making it potentially susceptible to climate change impacts. We examined the potential changes in halibut distribution and relative abundance that may arise from changing temperature and dissolved oxygen concentrations using species distribution models (SDMs) and future climate scenarios downscaled by two regional ocean models. SDMs were fit with both environmental variables (depth, near-bottom temperature and dissolved oxygen concentration) and spatial random field components (representing unknown habitat-related variables). The best-fitting models, trained on data from 2009 to 2013, were able to account for 33 % and 53 % of the variation in small and large halibut catch-per-unit-effort in the annual set-line survey data from 2014 to 2020. The results suggest that the response of Pacific halibut to climate change in British Columbia and Washington waters is likely to depend on changes in dissolved oxygen concentration. Pacific halibut appear sensitive to changes in dissolved oxygen, yet relatively tolerant to increases in temperature. Projections for 2046–2065 period suggest that future decreases in near-bottom dissolved oxygen in shallow waters that small halibut inhabit are likely to result in moderate decreases in relative abundance. Projected changes in relative abundance are less certain for large Pacific halibut, due to disagreement in the regional ocean models near-bottom dissolved oxygen projections at mid depths (300–600 m) where large Pacific halibut are more common. Overall, the relative abundance of Pacific halibut is expected to decrease in most areas of British Columbia. Future management strategies will need to account for the projected changes in distribution and abundance and their uncertainty.

Intracellular autolytic whole cell Salmonella vaccine prevents colonization of pathogenic Salmonella Typhimurium in chicken

Salmonella enterica (SE) is a major foodborne bacterial pathogen in the United States, commonly found as the normal flora of various animals that is attributed to causing at least 1.2 million infections annually. Poultry plays a major role in disseminating SE through direct contact with live animals and consumption of contaminated products. Vaccinating poultry against SE is a sustainable approach that can reduce SE in the host, preventing future infections in humans. An intracellular autolytic SE serovar Typhimurium vaccine (STLT2+P13+19) was developed by integrating genes 13 (holin) and 19 (lysozyme) of bacteriophage P22 into the bacterial chromosome. These were inserted downstream of sseA, an SPI-2 chaperone in SE that expresses during the intracellular phase of SE. Intracellular viability of STLT2+P13+19 reduced by 94.42% at 24 hr compared to the wild type in chicken macrophage cells (HD-11), whereas growth rate and adhesion ability remained unchanged. Inoculating STLT2+P13+19 in HD-11 significantly enhanced the relative log fold expression of genes associated to production of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, IL-10, IL-12 p40, IL-18, and GM-CSF) and Toll-like-receptors (TRL-3 and 7). Vaccination of an in vivo chicken model demonstrated significant changes in secretion of iNOS, IL-6, IL-8, IL-12, and TNF-α, as well as a reduction in the intestinal colonization of SE serovar Typhimurium. Microbiome analysis of cecal fluid using 16S rRNA gene sequencing also showed modulation of intestinal microbial composition, specifically a decrease in relative abundance of Proteobacteria and increasing Firmicutes. This study provides insight into a novel vaccine design that could make food products safer without the use of synthetic compounds.

Short-term response of soil enzyme activities and bacterial communities in black soil to a herbicide mixture: Atrazine and Acetochlor

Combined application of atrazine (ATZ) and acetochlor (ACE) was used as an emerging herbicidal method in black soil zone of northeastern China recently. However, the interaction toxic effect of the two herbicides on the soil biological characteristics has not been investigated yet. In this study, the alterations of soil enzyme activities and bacterial communities in black soil were assayed during a short term (28 days) exposure period by spiking ATZ, ACE or ATZ + ACE (1:1, w: w) at 0×, 1×, 2×, 5× and 10× the recommended dose in a microcosm experiment conducted under greenhouse condition. The results showed that ATZ and ACE had synergistic inhibitory effects on soil enzymes activities. Higher inhibition rates of urease, acid phosphatase, sucrase and catalase were observed in ATZ + ACE, and the enzyme activities could not recover to the level without herbicide application after 28 days. The higher IBRv2 value of ATZ + ACE also indicated that the comprehensive toxicity was stronger than that of ATZ or ACE. The microbial community structure was affected differently by ATZ, ACE and ATZ + ACE as the PLFAs analysis displayed, and ATZ + ACE was more harmful to bacteria than ATZ or ACE. The decrease in relative abundance of Proteobacteria was more sensitive to ATZ, and the decrease of Acidobacteriota was more sensitive to ACE, and the relative abundance of these two phyla decreased more obviously in ATZ + ACE. In addition, the PICRUSt analysis showed that the functions of the replication and repair, energy metabolism, cell motility and cellular processes and signaling were inhibited by ATZ and ACE, especially in the ATZ + ACE treatment. This means that combined application of ATZ and ACE has a greater potential risk on soil microbes and consequently the nutrient cycling process.

Effects of three Polyphenolic compounds on the intestinal flora of mice exposed simulated intermittent plateau hypoxia

To investigate the protective effects of three Polyphenolic compounds on intestinal microbial communities in mice exposed intermittent plateau hypoxia. In this study, 60 healthy male Balb/c mice were randomly divided into plain control group, plateau control group, primary anthocyanin intervention group, quercetin intervention group and resveratrol intervention group, 12 mice in each group. Primary anthocyanin, quercetin and resveratrol were administrated by gavage at the doses of 50, 100 and 20 mg/kg in pharmacological intervention group, respectively. After exposure of the mice to simulation plateau-condition for 30 days, the serum samples were collected for DAO testing, sterile feces were collected in mice, and the diversity and genus level of the mouse gut bacteria were detected by using 16S rRNA technology. Ileum tissue was fixed and stained with HE. HE staining showed that the plateau control group had significant damage to the intestinal tissue structure compared to the plain control group, and the serum DAO concentration was increased (P＜0.05), but there was no statistical difference in the abundance and diversity of intestinal flora species. Contrast to simulated intermittent plateau hypoxia group, the structure of the intestine tissue and the level of DAO in the quercetin intervention group and resveratrol intervention group were improved(P＜0.05), the abundance and α diversity of the intestinal flora were decreased, the relative abundance of Bacteroidetes was reduced(P＜0.05), and the Firmicutes was increased. Concomitantly, significant decreases in relative abundance were observed for Corynebacterium glutamicum and Lactobacillus reuteri(P＜ 0.05). Quercetin and resveratrol showed some degree of protection to mice intestinal microbial communities, and increased the diversity and the abundance of the dominant flora and inhibited the growth of conditional pathogenic bacteria.

Antibiotic and antifungal use in pediatric leukemia and lymphoma patients are associated with increasing opportunistic pathogens and decreasing bacteria responsible for activities that enhance colonic defense.

Due to decreased immunity, both antibiotics and antifungals are regularly used in pediatric hematologic-cancer patients as a means to prevent severe infections and febrile neutropenia. The general effect of antibiotics on the human gut microbiome is profound, yielding decreased diversity and changes in community structure. However, the specific effect on pediatric oncology patients is not well-studied. The effect of antifungal use is even less understood, having been studied only in mouse models. Because the composition of the gut microbiome is associated with regulation of hematopoiesis, immune function and gastrointestinal integrity, changes within the patient gut can have implications for the clinical management of hematologic malignancies. The pediatric population is particularly challenging because the composition of the microbiome is age dependent, with some of the most pronounced changes occurring in the first three years of life. We investigated how antibiotic and antifungal use shapes the taxonomic composition of the stool microbiome in pediatric patients with leukemia and lymphoma, as inferred from both 16S rRNA and metagenome data. Associations with age, antibiotic use and antifungal use were investigated using multiple analysis methods. In addition, multivariable differential abundance was used to identify and assess specific taxa that were associated with multiple variables. Both antibiotics and antifungals were linked to a general decline in diversity in stool samples, which included a decrease in relative abundance in butyrate producers that play a critical role in host gut physiology (e.g., Faecalibacterium, Anaerostipes, Dorea, Blautia),. Furthermore, antifungal use was associated with a significant increase in relative abundance of opportunistic pathogens. Collectively, these findings have important implications for the treatment of leukemia and lymphoma patients. Butyrate is important for gastrointestinal integrity; it inhibits inflammation, reinforces colonic defense, mucosal immunity. and decreases oxidative stress. The routine use of broad-spectrum anti-infectives in pediatric oncology patients could simultaneously contribute to a decline in gastrointestinal integrity and colonic defense while promoting increases in opportunistic pathogens within the patient gut. Because the gut microbiome has been linked to both short-term clinical outcomes, and longer-lasting health effects, systematic characterization of the gut microbiome in pediatric patients during, and beyond, treatment is warranted.

Intermittent Microaeration Technology to Enhance the Carbon Source Release of Particulate Organic Matter in Domestic Sewage

Domestic sewage treatment plants often have insufficient carbon sources in the influent water. To solve this problem, the commonly used technical means include an additional carbon source, primary sludge fermentation, and excess sludge fermentation, but these methods are uneconomical, unsustainable, and not applicable to small-scale wastewater treatment plants. Intermittent microaeration technology has the advantages of low energy-consumption, ease of application, and low cost, and can effectively promote anaerobic digestion of municipal sludge; however little research has been reported on its use to enhance the carbon sources release of particulate organic matter (POM) from domestic wastewater. Therefore, the effect of intermittent microaeration on the carbon source release of POM was evaluated in this study, with POM as the control test. The results showed that the release concentration of soluble chemical oxygen demand (SCOD) was the highest on day 4 under microaerobic conditions, and the concentrations of SCOD, NH4+-N, and PO43−-P in the liquid phase were 1153, 137.1, and 13 mg/L, respectively. Compared with the control group, the SCOD concentration increased by 34.2%, and the NH4+-N and PO43−-P concentrations decreased by 18.65% and 17.09%, respectively. Intermittent microaeration can effectively promote the growth of Paludibacter, Actinomyces, and Trichococcus hydrolytic fermentation functional bacteria. Their relative abundances increased by 282.83%, 21.77%, and 23.47%, respectively, compared with the control group. It can simultaneously inhibit the growth of acetate-type methanogenic archaea, Methanosaeta and Methanosarcina, with a decrease in relative abundances of 16.81% and 6.63%, respectively. The aforementioned data show that intermittent microaeration can not only promote the hydrolysis of POM, but can also reduce the loss of acetic acid carbon source, which is a cost-effective technical way to enhance the release of a carbon source of particulate organic matter in domestic sewage.

Continuous Bioleaching of Arsenic-Containing Copper-Zinc Concentrate and Shift of Microbial Population under Various Conditions

The goal of this work was to study the bioleaching of arsenic-containing polymetallic concentrate that contained 6.2% Cu, 7.3% Zn and 1.7% As, depending on different temperatures and in the presence of CO2 and molasses in the medium, as well as the difference in the composition of microbial population formed under various conditions. A mixed population of moderately thermophilic and thermotolerant acidophilic microorganisms formed during the continuous bioleaching of copper concentrate was used as an inoculum. The experiments were carried out in a continuous mode in laboratory scale reactors, with a temperature range of 40 °C to 60 °C. To assess the effect of CO2 and molasses on metal leaching and microbial population composition, the experiments were carried out in three reactors: CO2 (~0.01 L/min) was supplied into the first reactor; 0.02% molasses were added to the pulp of the second reactor; and no additional carbon sources were supplied into the control reactor. The highest copper recovery (27%) was achieved at 50°C in the experiment with molasses, while the highest zinc recovery (82.1%) was reached at 45°C in the control experiment. Additional carbon sources affected the extraction of non-ferrous metals only at 60 °C and increased the extraction of copper and zinc by 12.6% and 24.2%, respectively. Both the temperature and carbon source used affected the microbial population composition. The main microbial genera revealed in the populations by next generation sequencing (NGS) were bacteria of the genera Sulfobacillus and Acidithiobacillus, as well as archaea of the genera Ferroplasma, Acidiplasma, and Cuniculiplasma. At low temperatures (40 and 45 °C), Acidithiobacillus, Sulfobacillus, and Ferroplasma predominated, while at temperatures 50–55 °C, the decrease in relative abundance of these genera occurred, and the predominance of Acidiplasma archaea was observed. The usage of both CO2 and molasses led to the increase in Sulfobacillus and Acidiplasma in relative abundance.

Vaginal estrogen treats atrophy thereby restoring flora, which prevents UTIs. While estrogen is effective, some patients have contraindications or are unwilling to use it. Vaginal testosterone cream (VT) has been shown to improve atrophy, however, its effects on flora and rUTIs are unknown. The purpose of this study was to determine the effect of VT versus placebo on vaginal flora in postmenopausal women with rUTI. METHODS: This randomized, double- blind pilot study enrolled postmenopausal women with rUTIs. Patients were randomized to VT or placebo. Vaginal health index (VHI) and vaginal swab to assess flora via DNA sequencing were collected at baseline and at one month follow up. RESULTS: A total of 21 patients were enrolled. Average VHI at enrollment was 16.68±4. There was no difference between groups at baseline with regard to VHI. There was diversity of vaginal flora in both treatment arms at baseline (Figure 1). Among patients who completed follow up, almost half of those in the VT arm had an increase in relative abundance of lactobacilli, which was not seen in the placebo patients. As compared to the placebo arm, a greater number of patients in the VT arm had a decrease in relative abundance of uropathogens such as Bacteroides and Pseudomonas. The majority of patients in the VT arm had improvement in VHI, which was not seen with placebo at one month. Four patients developed UTIs during the study, two in each arm. CONCLUSIONS: Initial data demonstrates increased abundance of lactobacilli and improvement in VHI at one-month post treatment with VT. Although there was no difference in the number of patients who developed UTIs between groups, longer treatment follow up and increased patient enrollment will be valuable in determining if VT is effective for UTI prevention.
