Feeding yearling Angus bulls low-level ergot daily for 9 weeks decreased serum prolactin concentrations and had subtle effects on sperm end points

Our objective was to determine whether feeding yearling bulls with the higher recommended Canadian limit of ergot alkaloids (∼3 mg/kg dry matter intake, DMI) would affect sperm characteristics and plasma prolactin concentrations. Aberdeen Angus bulls (12–13 mo old, n = 7/group) allocated by blocking for sperm concentration and body weight, were fed placebo or ergot alkaloids in gelatin capsules (60 μg/kg body weight daily, 3.4 mg/kg of DMI) for 9 wk. Semen samples were collected weekly by electroejaculation and examined with a computer assisted semen analyzer (CASA) and flow cytometry, for the intervals 5 wk before (Pre-exposure period), 9 wk during (Exposure period) and 9 wk after (Post-exposure period) treatment. Weekly plasma samples were analyzed for prolactin by radioimmunoassay. Plasma prolactin concentrations decreased markedly (mean ± SEM, 16.74 ± 3.70 in Exposure and 33.42 ± 3.08 ng/mL in Post-Exposure periods; P < 0.01) compared to Control (67.54 ± 21.47 and 42.59 ± 15.06 ng/mL). Treatment did not affect (P ≥ 0.17) body weight gain, sperm concentration, sperm count/ejaculate, motility or percent live sperm. Averaged over the exposure and post-exposure durations, the scrotal circumference was smaller (P = 0.02) by 2.7% in the Ergot group. Progressive motility remained unchanged from 59.92 ± 2.31% in Exposure to 59.61 ± 2.59% in Post-Exposure periods, compared to marked increase in Control (61.42 ± 1.60% to 67.52 ± 1.47%; P = 0.02). Straight-line sperm velocity decreased (−3.15 ± 1.53 μm/s) from exposure to post-exposure periods in Ergot group (P = 0.04) versus an increase (2.96 ± 2.17 μm/s) in Control. Midpiece defects decreased from Exposure to Post-exposure periods in Control group but remained unchanged in Ergot group (trt∗age, P < 0.01). Ergot feeding resulted in a smaller proportion of sperm with medium mitochondrial potential (Ergot: 22.65 ± 0.98%, Control: 24.35 ± 1.05%, P = 0.04). In conclusion, feeding ergot at Canadian permissible limit for 9-wk resulted in a 4-fold decrease in plasma prolactin concentrations. Semen end points were not significantly affected, although there were subtle effects on progressive motility, midpiece defects and mitochondrial membrane potential. Clinical relevance of observed changes requires further evaluation. Results supported our hypothesis that prolonged low-level ergot will adversely affect plasma prolactin. However, semen parameters were partially affected, supporting similar work on fescue toxicosis.

Changes in prolactin and testosterone levels induced by acute physical exertion in young female athletes.

In order to establish the influence of acute atypical exercise on prolactin and testosterone levels in young female athletes the authors selected a group of 13 short distance runners who were subjected to exercise on a cycle ergometer. Hormone levels were measured before the test, just after the end of test, 30 and 90 minutes thereafter. Significant increase in testosterone concentration appeared immediately after the exercise and 90 minutes after completion of a test testosterone level returned to preexercise levels. Unlike the results of other authors statistically significant decrease in plasma prolactin concentration was found 90 minutes after the end of test.

Pituitary and ovarian response to transient doe-litter separation in nursing rabbits

The effects of a transient doe-litter separation on plasma prolactin, FSH and oestradiol concentrations, as well as the effect on LH response to exogenous GnRH administered at the time of artificial insemination, were determined in nursing rabbits. The effects on fertility, and litter size after parturition, as well as litter survival after doe-litter separation, were also studied. Control does (n = 12) had free access to nursing, whereas biostimulated does (n = 12) were separated from their litters for 48 h before artificial insemination. Plasma prolactin concentrations were decreased 24 h after the doe-litter separation (P < 0.05). The response of prolactin to suckling reached 10 times the basal values measured on day 10 after parturition (P < 0.0001). Increased oestradiol concentrations were found during the 48 h after the doe-litter separation: at 0 h, before artificial insemination (P< 0. 0001), 1.0-2.0 h after artificial insemination (P < 0.001), at 2.5 h (P < 0.05), 3.0 h (P < 0.01), and at 3.5 h (P < 0.05) after artificial insemination. Exogenous GnRH administered at the time of artificial insemination caused a greater LH response in does previously separated from their litters during 48 h (P < 0.01). The transient doe-litter separation did not affect plasma FSH concentrations, fertility, litter size or litter survival. These results suggest that a transient separation of nursing does from their litters before artificial insemination results in a decrease in plasma prolactin concentrations that could promote growth of follicular waves, and high steroidogenesis activity, leading to increased oestradiol concentrations and inducing higher sensitivity of the pituitary gland to exogenous GnRH. These findings associated to the absence of suckling episodes would lead to higher LH response and, therefore, exert a major effect on fertility.

The Endocrinology of Extrapyramidal System Disorders

The study of endocrinologic changes in Parkinson's disease, Huntington's disease, and tardive dyskinesia may elucidate the pathophysiology of these disorders, especially the presence of hypothalamic lesions. There is probably a decrease in PRL concentrations in Parkinson's disease, and there may be an increase in TSH response to TRH stimulation. It is not clear if there is any change in GH concentrations in Parkinson's disease. There appears to be a robust increase in GH concentrations in Huntington's disease, and there may be a small increase in PRL as well. At present no endocrinologic abnormality has been well documented in tardive dyskinesia.

Seasonal and circadian patterns of circulating prolactin during lactation and seasonal quiescence in the tammar, Macropus eugenii.

Prolactin was measured in peripheral plasma of female tammars once a week for 14 months and every 2 months it was measured in a subgroup of 7 tammars at hourly intervals for 28 h to determine short-term changes in concentration. During the course of the study 6 females gave birth in January/February and reared their young to weaning in October-November, while another 4 gave birth 1.5-3.0 months later and weaned their young in November-December. Concentrations of prolactin were less than 40 ng/ml in a non-lactating animal and for the first 140 days of lactation in the others. Between 140 and 180 days of lactation the concentrations of prolactin fluctuated between 10 and greater than 100 ng/ml and thereafter remained high until the young vacated the pouch for the last time at about 250 days. After pouch exit the concentrations declined to about 40 ng/ml even though lactation continued for a further 21-35 days. Although the short-term patterns of prolactin concentrations showed no consistent relationship between light-dark phases there was an increase in the magnitude and duration of peaks of plasma prolactin which correlated with the stage of lactation. Removal of pouch young aged between 203 and 242 days for 24 h resulted in a rapid decrease in plasma prolactin concentration which was reversed when the young was returned to the pouch, indicating that the high levels of prolactin in the second half of lactation may be maintained by the sucking stimulus of the pouch young. This period of lactation, therefore, may be equated with lactation in eutherians, but there appears to be no eutherian equivalent to early lactation in the tammar.

Role of prolactin in the photoperiodic control of moulting in the mink (Mustela vison).

Beginning at the summer solstice adult female mink were maintained in long- or short-day photoperiods and treated with bromocriptine or prolactin. In control females kept under natural lighting conditions the moult coincided with the seasonal decrease in prolactin and resulted in the growth of a dense winter coat which was completed by the end of November. Long days, which slowed the decrease in plasma prolactin relative to animals in the natural photoperiod, induced a more or less complete moult followed by growth of a thin summer coat. On the contrary we observed an accelerated decrease in plasma prolactin concentrations followed by an early and brief moult in females kept under long days but treated with bromocriptine and in females under short days. The growth of a dense winter coat was completed by the end of September in all the females of the short-day group and in six of eleven females treated with bromocriptine. In the other five females, moult was followed by the growth of a summer coat. These results may suggest that the decline of prolactin after the summer solstice is responsible for the onset of the autumn moult, but the early, abbreviated moult and the growth of a winter coat observed in females kept under short days and treated with prolactin do not seem to support this hypothesis. However, the huge non-physiological levels of prolactin measured in the plasma of these females and the appearance of abnormal white under-hairs might suggest that the hormonal balance in this group was completely disturbed by the treatment. The physiological role of prolactin in the seasonal moulting cycle in mink is discussed.

THE HYPOTHALAMIC CENTRES INVOLVED IN THE CONTROL OF PRODUCTION AND RELEASE OF PROLACTIN IN SHEEP

Lesions made in the anterior medial basal hypothalamus (MBH) in pregnant or lactating ewes caused lack of development of the mammary gland and depressed milk yield, a decrease in plasma prolactin concentration and structural changes in the prolactin cells as manifested by a lack of the expected degranulation (lack of the hormone release) or by atrophic changes and diminished cell granulation (or granule synthesis). Lesions made in the caudal MBH during pregnancy advanced development of the mammary gland, induced a rapid increase in the plasma prolactin concentration and caused extensive degranulation of prolactin cells. These results indicate that at least two functional systems controlling the secretion and production of prolactin exist in the hypothalamus of sheep: an anterior system which is stimulatory and a caudal which is inhibitory.

The influence of surgery, time of day, blood volume reduction and anaesthetics on plasma prolactin in ovariectomized rats.

SUMMARY Plasma prolactin was measured by double antibody radioimmunoassay in ovariectomized rats bearing indwelling aortic catheters. Samples were obtained during recovery from ovariectomy and catheterization, at various times during the day, after blood volume reduction, and after administration of various anaesthetics. Prolactin levels were stabilized by day 3 after the operation but not by day 1. There was a significant (P &lt; 0·01) increase in plasma prolactin during the afternoon in both ovariectomized rats and ovariectomized rats treated with 1 mg polyoestradiol phosphate. The magnitude of the response was more pronounced in the animals treated with oestrogen. Withdrawal of blood samples without subsequent replacement of fluid volume induced a significant (P &lt; 0·05) decrease in plasma prolactin concentration after removal of only 1·2 ml blood within 20 min. No change was observed, however, when blood was replaced with saline. Ether and the intraperitoneal injection of sodium pentobarbitone produced prolonged increases in plasma prolactin whereas the intra-arterial administration of sodium pentobarbitone produced no change. Urethane and chloral hydrate caused only transient increases in plasma levels of prolactin regardless of the route of administration. Ketamine did not significantly alter plasma levels of prolactin.
