Bueno1* 1 Assistant professor, Physiology and Biochemistry, Colombia 2 Faculty of Agricultural Sciences, School of Veterinary Medicine, Colombia 3 Universidad Cooperativa de Colombia- Sede Medellin, Facultad de Medicina, Colombia Background: Cell invasion and placenta development involve variations of oxygen supply that supports phenotypic changes of tumoral and trophoblast cells, including the glycocalix and membrane glycoprotein expression. Methods: JEG-3 and the extravillous trophoblastic cell line HTR8/Svneo were incubated for 12, 24 and 48 h with 150 μM CoCl2 (cobalt cloride) . A panel of multiple lectins labeled with FITC (Datura stramonium agglutinin- DSA, Helix pomattia agglutinin-HPA, Maackia amurensis agglutinin-MAA, Sambucus nigra agglutinin-SNA, Peanut agglutinin-PSA) was used to determine glycotopes of N- and O-glycoproteins using confocal microscopy and flow cytometry. Protein extracts from these cells were assessed for lectin blot analysis and enzymatic digestion with PNGase. nvasion assays of HTR8/Svneo cells were determined using Matrigel-coated 5 μm pore size polycarbonate membrane Transwell inserts and blocking N-glycosylation with tunicamycin and castanospermine. Results: After 48 h of treatment with CoCl2, an increase the expression of Tn antigen and N-acetyl-galactosamine in JEG-3 cells was visible by lectin-binding assays with HPA and DBA respectively (P < 0,001). Furthermore, increased expression of the terminal sialic acid linked to galactose (α2-6 and α2-3) was observed in glycoproteins 60-70 kD long. The percentage of invasion observed in HTR8/Svneo cells was reduced by blocking N-glycosylation with tunicamycin and castanospermine (P < 0,05).
