Novel synthesis of mannosamine conjugated magnetic nanoparticles for purification and stabilization of human lysosomal β-mannosidase

Human β-mannosidase (MANB) was purified to homogeneity directly from lysosomes by using mannosamine conjugated magnetic (Fe3O4) nanoparticles, DE-52 cellulose, and sephadex G-200 chromatography. Fe3O4 nanoparticles were synthesized and utilized ammonia to attach the amino group on the nanoparticles. The particles were covalently attached with D-mannosamine by cross linker glutaraldehyde and confirmed by FTIR spectroscopy. In FTIR analysis, the peaks appeared at 2,356.6 cm−1 for −N = CH linkage and at 3,378.4 cm−1, 3,664.9 cm−1 for −OH groups confirmed the conjugation of D-mannosamine with Fe3O4 nanoparticles. Results showed a single band of 97 kDa of purified MANB in SDS-PAGE. The isoelectric point was 4.5 and the Km and Vmax values were 2.51 mM and 0.315 µM/min/mg, respectively. The purification fold was 329 with 68% yield. The optimal activity was at pH 5.0 and 75% activity was stable in 20% glycerol at 4°C. The enzyme activity was inhibited by Ni2+, Zn2+, Cd2+, Cu2+, Mo2+, Ag+1, iodoacetate, SDS, DMF, DMSO, ethanol, and acetone; slightly reduced by Pb2+, Co2+, EDTA, DTT, and β-mercaptoethanol. The activity was not affected by Mg2+, Mn2+, Sn2+, Ca2+, Fe3+, PMSF, Triton X-100, D-mannosamine, D-mannose, D-mannitol, D-glucose, and D-fructose. The homogeneity of MANB enzyme was further confirmed by 2D-PAGE and immunoblot. This is the first novel report of conjugation of D-mannosamine with Fe3O4 nanoparticles for purification of human MANB enzyme.

A Novel Immunomodulatory Protein from Poria cocos Induces Toll-like Receptor 4-Dependent Activation within Mouse Peritoneal Macrophages

Poria cocos is an important Oriental medical fungus with multiple functionalities, yet its bioactive substances and the mechanisms involved have not been fully characterized. A novel immunomodulatory protein (P. cocos immunomodulatory protein; PCP) was purified from the dried sclerotium of P. cocos (Schw.) Wolf using DE-52 cellulose and gel filtration chromatography. Chromatography and electrophoresis results indicated that the native PCP (35.6 kDa) is a disulfide-linked heterodimeric glycoprotein consisting of 14.3 and 21.3 kDa subunits with N- and O-glycosylation. PCP was capable of stimulating RAW 264.7 macrophages in vitro through the induction of tumor necrosis factor-alpha (TNF-alpha) and interleukin-1 beta (IL-1beta) as well as the regulation of nuclear factor-kappa B (NF-kappaB)-related gene expression. In primary mouse macrophages, PCP directly activated peritoneal cavity macrophages to induce Toll-like receptor 4 (TLR4)-mediated myeloid differentiation factor 88 (MyD88)-dependent signaling. This study demonstrated the cell surface interactions of PCP with TLR4 and the capacity of PCP for TLR4 tyrosine phosphorylation. Results obtained with peritoneal macrophages from TLR4-deficient C57BL/10ScN mice revealed that PCP-induced activation and PCP cell surface binding were significantly attenuated. Moreover, enzymatic deglycosylation decreased PCP-mediated responses, indicating that the glycosylated portion of PCP was a key factor in PCP signaling through TLR4 in peritoneal macrophages. These findings suggest that PCP is a new potential immune stimulator within P. cocos and that TLR4 is primarily responsible for PCP signaling in murine macrophages.

Purification and characterization of a cysteine-like protease from the body wall of the sea cucumber Stichopus japonicus

The sea cucumber (Stichopus japonicus) is able to undergo autolysis in response to a variety of environmental and mechanical cues. Within the framework of a long-term study of this phenomenon we have purified a protease from the body wall of the sea cucumber by means of ion-exchange chromatography with DE-52 cellulose and gel filtration chromatography with Sephadex G-100. The final enzyme preparation was nearly homogeneous on polyacrylamide gel electrophoresis, and its molecular weight was estimated to be approximately 35.5 kDa. The purified enzyme exhibited a maximum activity for the hydrolysis of casein at pH 7.0 and 50°C and a remarkable stability at pH 4.0–7.0 and 40–60°C. Based on the inhibition and activation profiles obtained with numerous specific protease inhibitors and an activator, the protease purified from the body wall of the sea cucumber was defined to be a cysteine-like protease.

Purification of recombinant wheat cytochrome P450 monooxygenase expressed in yeast and its properties

To investigate the properties of wheat cytochrome P450 and the characteristics of herbicide metabolism by cytochrome P450 in vitro, deeply understand the mechanisms of herbicide selectivity, recombinant wheat cytochrome P450 monooxygenase (CYP71Cv1) heterologously expressed in yeast was purified by DE-52 cellulose chromatography and fast protein liquid chromatography (FPLC) with Mono-Q column. The degree of purification was 1366-fold. The specific activity of purified cytochrome P450 reached to 512 nmol min −1 mg −1 protein with herbicide chlorsulfuron as substrate. The purified cytochrome P450 exhibited one band in sodium dodecyl sulfate (SDS)–polyacrylamide gel electrophoresis, and the molecular mass was 52.5 kDa. Kinetic parameter was determined in vitro. The K m values for chlorsulfuron and triasulfuron were 57 (±15) and 38 (±16) μM, respectively; and V max for chlorsulfuron and triasulfuron were 4.1 (±0.7) and 2.7 (±0.5) nmol min −1 mg −1 protein in vitro, respectively.

N-acetylglucosamine kinase and N-acetylglucosamine 6-phosphate deacetylase in normal human erythrocytes and Plasmodium falciparum.

The major pathways of glucose metabolism in the malaria parasite, Plasmodium falciparum, have now been elucidated, and the structures and properties of parasite-specific enzymes are presently being investigated. Little is known, however, about the enzymes catalysing monosaccharide interconversions in the parasite. In the present investigation we have examined the pathway of N-acetylglucosamine catabolism which, in higher organisms, involves the following reaction sequence: N-acetylglucosamine -->N-acetylglucosamine 6-phosphate-->glucosamine 6-phosphate-->fructose 6-phosphate. Assay of the specific kinase (E.C. 2.7.1.59) catalysing the phosphorylation of the sugar showed that the enzyme is present in Plasmodium extracts as well as in normal human erythrocytes; specific activities of 7.2 and 5.3 nmol/h/mg protein were measured for the parasite and erythrocyte extracts, respectively, N-Acetylglucosamine 6-phosphate deacetylase (E.C. 3.5.1.25), catalysing the second reaction, was also detected in both normal and Plasmodium-infected erythrocytes. At 75% parasitaemia, the deacetylase activity was close to 3 times higher than that of normal control cells. The erythrocyte deacetylase was purified approximately 16,000-fold by chromatography on DE52 cellulose, chromatofocusing, and size exclusion chromatography. Attempts to purify the parasite enzyme by the same procedures were unsuccessful due to loss of activity. A partially purified erythrocyte deacetylase preparation (eluted from DE52 cellulose) had a pH optimum of 7.5, a pI of 6.0, as indicated by chromatofocusing, and a K(m) of 29 microM. In conjunction with previous investigations, the present study indicated that all three enzymes required for N-acetylglucosamine utilization are present in Plasmodium parasites as well as in normal erythrocytes.

Characterization of a deaminated neuraminic acid-containing glycoprotein from the skin mucus of the loach, Misgurnus anguillicaudatus.

Using NMR spectroscopy and mass spectrometry, the major sialic acid of the skin mucus of the loach, Misgurnus anguillicaudatus was found to be 3-deoxy-D-glycero-D-galacto-2-nonulosonic acid (KDN). We have subsequently devised a method to isolate a KDN-containing glycoprotein preparation from loach skin mucus. The method involves the sonication of the skin mucus with 0.05 M Tris-HCl, pH 8.0, to solubilize the glycoprotein, followed by DE52-cellulose chromatography of the extract, Nuclease P1 treatment, and Sepharose CL-4B gel filtration. The purified glycoprotein preparation was found to contain 38.5% KDN, 0.4% NeuAc, 24.6% GalNAc, 3.3% Gal, and 28.2% amino acids (w/w). The amino acid composition of this glycoprotein preparation revealed that it is unusually rich in Thr, and 6 amino acids, Thr, Ser, Glu (or Gln), Pro, Gly and Ala, account for 83% of the total amino acids. This glycoprotein is extremely poor in Cys, Met, Tyr, Phe, Arg, and Trp. Treatment of this glycoprotein with alkali resulted in the destruction of 83% of Thr suggesting that most of the sugar chains in this glycoprotein are linked through Thr. Alkaline borohydride treatment of 100 mg of the glycoprotein preparation, followed by Sephadex G-25 (superfine) gel filtration, yielded two major oligosaccharide alditols, I (15.4 mg) and II (15.6 mg). Using liquid secondary ion mass spectrometry and methylation analysis, I was identified to be KDN alpha 2-->6GalNAc-ol and II, KDN alpha 2-->6(KDN alpha 2-->3)-GalNAc-o1. KDN alpha 2-->6GalNAc is structurally similar to NeuAc alpha 2-->GalNAc found in ovine submaxillary glycoprotein while II represents a novel structure which contains two sialic acids linked to a GalNAc through both alpha 2-->3 and alpha 2-->6 linkages. This structure has never been found in mucin type glycoproteins including mammalian epithelial mucin glycoproteins. This is the first report of the presence of a mucin type glycoprotein which contains KDN instead of NeuAc or NeuGc in fish skin mucus.

Copper Induced Polymerization of Hemoglobin from the Ocean Pout, Macrozoarces americanus

Hemoglobins that polymerize are not uncommon among amphibians, reptiles, and elasmobranchs, but are rare among teleosts and humans. They are useful models for the study of protein structure, function, and molecular evolutionary relationships. The freshwater teleost Hoplias malabarica (I), the marine teleost Lophius americanus (2), and the clown fish, Amphiprion (3) are the only bony fishes reported to have hemoglobins that polymerize. Hoplias and Lophius polymerize by forming intermolecular disulfide bonds, while the polymerization mechanism for the clown fish is, as yet, unknown. Among humans, hemoglobins Ta-Li, Mississippi, and Pot-to Alegre (4, 5, 6) have a single mutation involving the substitution of a cysteine residue at different loci on the globin chain. We now report that the ocean pout has a single hemoglobin with six sullhydryl groups per tetramer and polymerizes, in vitro, after oxidation with potassium ferricyanide (K,Fe(CN),) or cupric chloride (CuClJ. Procedures for hemoglobin purification by DE-52 cellulose chromatography, molecular weight estimations by gel filtration on G100 columns, and urea gel electrophoresis for globin subunit analysis have been described (2). Hemoglobin was incubated with a twoto threefold molar excess of CuCIZ for 15 min in an ice bucket. The excess copper was removed by dialysis or by gel filtration from Sephadex G-25 columns equilibrated with 0.1 M K-PO., buffer, pH 7.4. Thereafter 20-40 mg of the hemoglobin was applied to and eluted from G100 columns. One-ml fractions were collected and the absorbance of each measured at 540 nm. The approximate polymer molecular weight was estimated from the elution volume of the peak tube. Hemoglobin solutions (0.52.0 g/%) were also oxidized by adding l-3 mg of potassium ferricyanide per ml of hemoglobin. Iodoacetamide (IAA)-alkylated hemoglobin ( 15 rmoles/mg hemoglobin) was dialyzed overnight, centrifuged, and subsequently treated with CuC12. In a separate xperiment, a 60-fold molar excess of beta-mercaptoethanol (B-ME) was added to CuCl*-treated hemoglobin, incubated for 1 h, and dialyzed for 3 h before G100 chromatography. Untreated hemoglobin and hemoglobin treated with CuCIZ alone served as controls and were dialyzed for the same period. The gel filtration profile of known molecular weight proteins is shown in Figure 1 A. The inset graph is a plot of the log of the molecular weight of each standard versus the elution volume of the peak tube. The linear relationship indicates that the peak tube elution volume provides a reasonable stimate of the protein molecular weight between 16,000 and 150,000 Da. (The molecular weight fractionation range for Sephadex G-100 is 4 X 10’ and 1.5 X 105.) .The effect of CuCl* on pout hemoglobin is shown in Figure 1 B. In the absence of the cation, the peak tube eluted at a volume comparable to human hemoglobin, suggesting anormal molecular weight for pout hemoglobin of approximately 60,000. The addition of CuC12 results in a polymer with a minimum molecular mass of 100 to 155 kDa, which corresponds to aggregates of, perhaps, two to three tetramers. Cupric-chloride-induced polymer formation may be inhibited by prior incubation with IAA (Fig. 1C). Hemoglobin alkylated with IAA elutes at the same volume as normal pout and human hemoglobin, but in the absence of IAA, cupric-chloride-induced

Purification, amino-terminal sequence and functional properties of a 64 kDa cytosolic protein from heart muscle capable of modulating calcium transport across the sarcoplasmic reticulum in vitro.

In previous studies we have described the inhibitory action of a cytosolic protein fraction from heart muscle on ATP-dependent Ca2+ uptake by the sarcoplasmic reticulum (SR); further this inhibition was shown to be blocked by an inhibitor antagonist, also derived from the cytosol (Narayanan et al., Biochim. Biophys. Acta. 735: 53-66, 1983; Can. J. Physiol. Pharmacol. 67: 999-1006, 1989). Here we report the complete purification of the antagonist protein (AP) and characterization of its functional properties. AP was purified to homogeneity from rabbit heart cytosol using two procedures, one utilizing sequential DE52-cellulose and hydroxylapatite chromatography, and the other utilizing anion exchange chromatography on Mono Q HR 5/5 column in a Pharmacia FPLC system. The purified AP has an apparent molecular weight of 64 kDa; it is made up of about 43% hydrophobic and 57% hydrophilic residues with the following amino-terminal sequence: E-A-H-K-S-E-I-A-H-R-F-N-D-V-G-E-E-H-F-I-G-L-V-L-I-T-F-S-Q-Y-L-Q-K-X-P-Y- E-E-H-A . This partial amino acid sequence data indicate strong sequence homology to serum albumin (sequence homology: 85% to rat serum albumin and 74% to sheep and bovine serum albumin). The purified AP caused concentration-dependent-blockade of the inhibition of Ca2+ uptake by SR observed in the presence of the cytosolic Ca2+ uptake inhibitor protein.(ABSTRACT TRUNCATED AT 250 WORDS)

Efficient production of biologically active human salivary cystatins in Escherichia coli

Different Escherichia coli expression systems were used for expression of cDNA clones encoding the human salivary cysteine proteinase (CysP) inhibitors, cystatins SN and S (CsnSN and CsnS). These included pOTSNco 12 that expresses foreign sequences as authentic (nonfusion) proteins, and pGEX-2T that directs the synthesis of foreign polypeptides as fusion proteins with glutathione S-transferase (GST). The pOTS vector produced low levels of recombinant CsnSN (reCsnSN) that was localized in the soluble fraction, but not easily purified. The pGEX vector, on the other hand, produced much higher yields of the fusion protein, GST::CsnSN, that was localized almost entirely in the insoluble protein fraction. Solubilized and refolded GST::CsnSN inhibited the CysP, papain, more efficiently than chicken egg white Csn, indicating that the recombinant product was biologically active and that the GST carrier did not interfere with the biological activity. The pGEX-2T vector was subsequently used for the large-scale production of reCsnSN and reCsnS that were cleaved from the GST by thrombin and purified by DE-52 cellulose chromatography. ReCsnSN inhibited papain almost as efficiently as salivary CsnSN, while the reCsnS showed lower inhibitory activity as compared to both salivary CsnS and reCsnSN.

Purification and properties of peroxidase from Pinus pinaster needles

Peroxidase (Ec 1.11.1.7) was purified from needles of Pinus pinaster to apparent homogeneity by DE-52 cellulose chromatography with a final recovery of enzyme activity of about 85%. The purified enzyme ( A 402/ A 275= 1.05) had a specific activity of about 948 U/mg of protein and ran as a single protein band both on SDS-PAGE and native PAGE with M r of 37 000 and 151 000, respectively. Both native PAGE and isoelectric focusing gels of the purified enzyme were stained for activity which coincided with the protein band. The p I of the purified enzyme was found to be 3.2 by isoelectric focusing on an ultrathin polyacrylamide gel. The enzyme has an optimum pH of activity of 5.0 and temperature optimum of 30°C. Stability studies of the enzyme as a function of pH and temperature suggest that it is most stable at pH 5.0 and 0–40°C, respectively.

Altered type I protein kinase in adhesion defective CHO cell variants.

ADv cells are Chinese hamster ovary (CHO) cell variants which cannot adhere to fibronectin coated substrata (Harper & Juliano: J. Cell Biol., 1980; Nature 1981a,b). We have shown that the defect in some clones of ADv cells is distal to the initial interaction between fibronectin and its cell surface receptors (Cheung and Juliano: Exp. Cell Res., 1984), and that it extends to fibronectin mediated aggregation and endocytosis. The adhesion defect in some ADv clones can be corrected by raising intracellular cAMP levels (Cheung & Juliano: J. Cell Physiol., 1985). Here we examine the protein kinase activities and phosphorylation patterns in an adhesion defective variant clone ADv F11CA11. Analysis of the cAMP dependent protein kinase activity (cAdPK) in crude extracts of F11CA11 cells shows an apparent increase in K (activation) as compared to wild type (WT) CHO cell extracts. Further, the DE-52 cellulose chromatography profile of cAdPK in the F11CA11 variant is markedly different from WT in that the type I cAdPK peak elicited by 1 microM cAMP is essentially missing in F11CA11, while the type II cAdPK peak is similar to that in WT. Raising the cAMP level to 100 microM elicits a type I peak in F11CA11 with about 45% of the activity of the WT peak. Binding studies with 3H-cAMP reveal that the type I peak in F11CA11 has a Kd of 1.7 X 10(-8) M as compared to 2.0 X 10(-9) M for WT, whereas the type II peak Kd is approximately 1 X 10(-9) M for both WT and F11CA11. Two-dimensional polyacrylamide gel analysis of 32Pi labeled WT cells and F11CA11 cells with or without cAMP treatment reveals the presence of a protein(s) of 50 kilodaltons which is phosphorylated in WT cells and in cAMP treated F11CA11 cells but not in untreated F11CA11 cells. These findings, coupled with our previous observations, strongly indicate that the adhesion defect in ADvF11CA11 cells is associated with an altered type I cAdPK having lower affinity for cAMP. At normal cellular cAMP levels this enzyme fails to phosphorylate one or more critical protein substrates; however, by raising internal cAMP levels, the defect can be overcome. Thus type I cAdPK seems to play an important role in the regulation of fibronectin mediated cell adhesion, cell aggregation, and endocytosis.

Gonadotropin release and redistribution of calcium-activated, phospholipid-dependent protein kinase in phorbol-stimulated rat pituitary cells

The effect of phorbol esters on calcium-activated, phospholipid-dependent kinase (protein kinase C) and luteinizing hormone (LH) secretion was examined in cultured rat anterior pituitary cells. The potent tumor promoter 12-O-tetra-decanoylphorbol-13-acetate (TPA) stimulated LH secretion and activated pituitary protein kinase C in the presence of calcium and phosphatidylserine. The enzyme activity present in cytosol and particulate fractions was eluted at about 0.05 m NaCl during DE52-cellulose chromatography. Preincubation of pituitary cells with TPA markedly decreased cytosolic protein kinase C activity and increased enzyme activity in the particulate fraction. The maximal TPA-induced change in enzyme activity, with a 76% decrease in cytosol and a 4.3-fold increase in the particulate fraction, occurred within 10 min. The dose-dependent changes in protein kinase C redistribution in TPA-treated cells were correlated with the stimulation of LH release by the phorbol ester. These results suggest that activation of protein kinase C by TPA is associated with intracellular redistribution of the enzyme and is related to the process of secretory granule release from gonadotrophs.

Occurrence of two l-threonine (l-serine) dehydratases in the thermophile Chloroflexus aurantiacus

The thermophilic phototrophic prokaryote, Chloroflexus aurantiacus was shown to contain high constitutive l-threonine (l-serine) deaminating activity. Separation of cellular proteins by DE 52-cellulose chromatography and by polyacrylamide gel electrophoresis with subsequent activity staining of the gels yielded two bands, one representing an isoleucine-sensitive, the other one an isoleucine-insensitive form of l-threonine dehydratase. Both enzymes had a molecular weight of 120,000 but were distinguished by their different affinities to the two substrates, l-threonine and l-serine.

Purification of the 300K intermediate filament-associated protein and its in vitro recombination with intermediate filaments.

IFAP-300K is a 300,000-mol-wt intermediate filament-associated protein previously identified in the baby hamster kidney fibroblastic cell line (BHK-21) by a monoclonal antibody (Yang H.-Y., N. Lieska, A. E. Goldman, and R. D. Goldman, 1985, J. Cell Biol., 100: 620-631). In the present study, this molecule was purified from the high salt/detergent-insoluble cytoskeletal preparation of these cells. Gel filtration on Sephacryl S-400 in the presence of 7.2 M urea allowed separation of the high molecular weight fraction from the structural intermediate filament (IF) subunits desmin and vimentin, designated 54K and 55K, respectively, and other low molecular weight polypeptides. DE-52 cellulose chromatography of the high molecular weight fraction using a linear NaCl gradient in 8 M urea yielded a pure 300,000-mol-wt species which was confirmed to be IFAP-300K by immunological and peptide mapping criteria. Two-dimensional PAGE of native BHK IF preparations followed by immunoblot analysis demonstrated the inability of the IFAP-300K-immunoreactive material to enter the first dimensional gel except as a 200,000-mol-wt doublet which presumably represented a major proteolytic derivative of IFAP-300K. The molecule's pl of 5.35, as determined by chromatofocusing, and its amino acid composition were extremely similar to those of BHK cell vimentin/desmin despite their non-identity. Ultrastructurally, IFAP-300K preparations in low salt buffers existed as particles composed of one or two elliptical units measuring 16 X 20 nm. In physiological salt buffers, the predominant entities were large, elongated aggregates of the elliptical units, which were able to be decorated by using the immunogold technique with monoclonal anti-IFAP-300K. Compared with the morphology of homopolymer vimentin IF, in vitro recombination studies using column-purified vimentin and IFAP-300K demonstrated the additional presence of aggregates similar in appearance to IFAP-300K at points of contact between IFs. Antibody decoration and immunogold labeling of these recombined preparations using rabbit antidesmin/vimentin and monoclonal anti-IFAP-300K confirmed the identity of the inter-filament, amorphous material as IFAP-300K. The presence of IFAP-300K at many points of intersection and lateral contact between IFs, as well as at apparent inter-filament "bridges," in these recombined specimens was identical to that seen both in situ and in native IF preparations. No such co-sedimentation was found in vitro between actin and IFAP-300K. No effects of IFAP-300K upon the kinetics of IF polymerization were detected by turbidimetric measurements.

Purification and properties of the activating enzyme for iron protein of nitrogenase from the photosynthetic bacterium Rhodospirillum rubrum.

The oxygen-labile, activating enzyme for iron protein from the photosynthetic bacterium, Rhodospirillum rubrum, was purified 11,800-fold using a combination of chromatophore washing, DE52-cellulose chromatography, hydroxylapatite chromatography, reactive red-120 cross-linked agarose chromatography, reactive red-120 cross-linked agarose chromatography, and Sephadex G-75 gel filtration. Activating enzyme appeared homogeneous on silver-stained sodium dodecyl sulfate-polyacrylamide gels, and the staining intensity of the activating-enzyme band was correlated with the activating-enzyme activity observed in in vitro assays. Either formaldehyde fixation or higher acrylamide concentration was required to accurately assess the purity of activating enzyme on silver-stained gels. Activating enzyme was stable for 30 days at 4 degrees C. Dithiothreitol was a necessary component for the stability of partially purified activating enzyme. NaCl inhibited the coupled assay for activating enzyme. The pI of activating enzyme was determined to be 6.5. Activating enzyme is composed of a minimum of 336 amino acids and a minimum calculated Mr is 32,032. The Mr of activating enzyme was estimated to be 21,700 by analytical gel filtration and 32,800 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. An absorption maximum at 280 nm was observed for the activating enzyme.

Chromatographic behavior of cyclic AMP dependent protein kinase and its subunits from Dictyostelium discoideum.

An adenosine cyclic 3',5'-monophosphate (cAMP) dependent protein kinase has recently been shown to exist in Dictyostelium discoideum and to be developmentally regulated. In this report we have followed the chromatographic behavior of both the holoenzyme and its subunits. A cAMP-dependent holoenzyme could be obtained from the 100000 g soluble fraction after passage through DE-52 cellulose (pH 7.5) and Sephacryl S300. Under conditions of low pH the holoenzyme could be further purified by flat-bed electrofocusing (pI = 6.8). Application of the holoenzyme to electrofocusing at high pH resulted in dissociation of the holoenzyme into a cAMP binding component (pI = 6.1) and a cAMP-independent catalytic activity (pI = 7.4). Dissociation of the holoenzyme into subunits also occurred during histone affinity chromatography and gel filtration chromatography (S300) in the presence of a dissociating buffer. Although the subunit structure was clearly evident during chromatography, the holoenzyme could not be dissociated by simple addition of cAMP to the extract. The catalytic subunit could be purified further by CM-Sephadex, DE-52 cellulose (pH 8.5), histone affinity, and hydrophobic chromatography. The regulatory subunit was further purified by DE-52 cellulose (pH 8.5) and cAMP affinity chromatography. Proof that the cAMP binding activity and the cAMP-independent catalytic activity were in fact the regulatory and catalytic subunits was shown by reconstitution of the cAMP-dependent holoenzyme from the purified subunits. By using these separation procedures, one can obtain from extracts of Dictyostelium the subunits that are free of each other as well as free of any endogenous protein substrates.

Endogenous substrates for in vitro phosphorylation by cyclic AMP-dependent protein kinase from Dictyostelium discoideum

Endogenous proteins which could serve as substrates for cyclic AMP-dependent protein kinase in vitro were measured in cytosolic fractions at four stages of development. A peak of cyclic AMP-dependent phosphorylation occurred at the slug stage, coincident with the appearance of cyclic AMP-dependent protein kinase. After partial purification of the slug-stage extracts by DE-52 cellulose and Sephacryl S-300 chromatography, cyclic AMP dependency of six proteins was observed. The apparent subunit molecular weights of the proteins were >200,000, 110,000, 107,000, 91,000, 75,000, and 69,000. Upon further purification of the cyclic AMP-dependent protein kinase by chromatofocusing, the endogenous substrates were separated from the enzyme. In addition, the enzyme separated into catalytic and regulatory subunits. If the purified catalytic subunit was added to heated S300 fractions, proteins with apparent molecular weights of 91,000 and 107,000 were specificity phosphorylated. The results show the stage-dependent appearance of a cyclic AMP-dependent protein kinase and point out several in vitro substrates for the enzyme.

Separation, Purification, and Characterization of Two Isoforms of Glutamine Synthetase from Chlamydomonas reinhardii

Two isoforms of glutamine synthetase. GS1 and GS2, have been separated from Chlamydomonas reinhardii cells grown autotrophically with nitrate. The intracellular level of GS2 was higher than that of GS1. In cells under darkness, the GS1 peak increased markedly, whereas that of GS2 became negligible. The two isoenzymes were purified to electrophoretic homogeneity by a method which included: a) DE-52 cellulose chromatography; b) ammonium sulphate fractionation: and c) affinity chromatography on ADP-sepharose. The specific activity was 114 and 63 U/mg for GS1 and GS2 respectively, and both enzymes (Mr = 380 000 and 373 000) are oligomeric proteins composed by 8 subunits of similar size (Mr = 48 000 in GS1, and 46 000 in GS2). The basic differences between GS1 and GS2 are: a) the effect of light on their intracellular level: b) their Km for ammonium (83 and 244 μᴍ, respectively). Both isoenzymes were inhibited in a similar extent by ʟ-alanine, ʟ-glycine and ʟ-arginine.

Purification and Mn2+ activation of Rhodospirillum rubrum nitrogenase activating enzyme.

The Fe protein activating enzyme for Rhodospirillum rubrum nitrogenase was purified to approximately 90% homogeneity, using DE52-cellulose chromatography and sucrose density gradient centrifugation. Activating enzyme consists of a single polypeptide of molecular weight approximately 24,000. ATP was required for catalytic activity, but was relatively ineffective in the absence of Mg2+. When the concentration of MgATP2- was held in excess, there was an additional requirement for a free divalent metal ion (Mn2+) for enzyme activity. Kinetic experiments showed that the presence of Mg2+ influenced the apparent binding of Mn2+ by the enzyme, resulting in a lowering of the concentration of Mn2+ required to give half-maximum activity (K alpha) as the free Mg2+ concentration was increased. A low concentration of Mn2+ had a sparing effect on the requirement for free Mg2+. There is apparently a single metal-binding site on activating enzyme which preferentially binds Mn2+ as a positive effector, and free Mg2+ can compete for this site.

The isolation of a family of cysteine-containing phosphoproteins from human submandibular-sublingual saliva.

A family of acidic proteins has been isolated from human submandibular-sublingual saliva using Sephadex G-200 gel filtration, DE-52 cellulose chromatography, and immuno-affinity chromatography. These molecules were homogenous in size and by immunological criteria, but exhibited charge heterogeneity. The heterogeneity displayed was not due to gustatory stimulation or degradation during isolation procedures.
