In vitro assessment of the effect of magnetic fields on efficacy of biosynthesized selenium nanoparticles by Alborzia kermanshahica

Cyanobacteria represent a rich resource of a wide array of unique bioactive compounds that are proving to be potent sources of anticancer drugs. Selenium nanoparticles (SeNPs) have shown an increasing potential as major therapeutic platforms and led to the production of higher levels of ROS that can present desirable anticancer properties. Chitosan–SeNPs have also presented antitumor properties against hepatic cancer cell lines, especially the Cht-NP (Chitosan–NPs), promoting ROS generation and mitochondria dysfunction. It is proposed that magnetic fields can add new dimensions to nanoparticle applications. Hence, in this study, the biosynthesis of SeNPs using Alborzia kermanshahica and chitosan (CS) as stabilizers has been developed. The SeNPs synthesis was performed at different cyanobacterial cultivation conditions, including control (without magnetic field) and magnetic fields of 30 mT and 60 mT. The SeNPs were characterized by uv-visible spectroscopy, Fourier-transform infrared spectroscopy (FT-IR), Dynamic light scattering (DLS), zeta potential, and TEM. In addition, the antibacterial activity, inhibition of bacterial growth, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC), as well as the antifungal activity and cytotoxicity of SeNPs, were performed. The results of uv-visible spectrometry, DLS, and zeta potential showed that 60 mT had the highest value regarding the adsorption, size, and stabilization in compared to the control. FTIR spectroscopy results showed consistent spectra, but the increased intensity of peaks indicates an increase in bond number after exposure to 30 mT and 60 mT. The results of the antibacterial activity and the inhibition zone diameter of synthesized nanoparticles showed that Staphylococcus aureus was more sensitive to nanoparticles produced under 60 mT. Se-NPs produced by Alborzia kermanshahica cultured under a 60 mT magnetic field exhibit potent antimicrobial and anticancer properties, making them a promising natural agent for use in the pharmaceutical and biomedical industries.

Green and ecofriendly biosynthesis of selenium nanoparticles using Urtica dioica (stinging nettle) leaf extract: Antimicrobial and anticancer activity.

Plant extract is affordable and does not require any particular conditions; rapid production of nanoparticles using plants offers more advantages than other approaches. Selenium nanoparticles (SeNPs) have received much attention in the last decade due to SeNPs diverse and different applications. Herein, this study aimed to biosynthesize SeNPs using aqueous extract of Urtica dioica leaf through green and ecofriendly method. Moreover to fully characterize SeNPs using different techniques, and to evaluate it for antimicrobial activity as well as anticancer activity. SeNPs were biosynthesis using aqueous leaf extract of U. dioica (stinging nettle). The biosynthesized SeNPs were characterized using UV-visible spectroscopy (UV-Vis), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), energy dispersive electron spectroscopy (EDX), transmission electron microscopy (TEM), and thermal-gravimetric analysis (TGA). Antimicrobial and anticancer activities of biosynthesized SeNPs were assessed. Results illustrated that SeNPs exhibited promising antibacterial activity against Gram-positive and Gram-negative bacteria, as well as unicellular and multi-cellular fungi. Moreover, minimal-inhibitory concentration (MIC) of SeNPs against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus were 250, 31.25, and 500μgmL-1 , respectively, while were 62.5, 15.62, 31.25, and 7.81μgmL-1 against Candida albicans, Aspergillus fumigatus, Aspergillus niger, and Aspergillus flavus, respectively. The cytotoxicity of SeNPs was performed on Vero normal-cell line CCL-81, where IC50 was 173.2μgmL-1 . For the first time, aqueous stinging nettle leaf extract was utilized to biosynthesize SeNPs in a green method. SeNPs have outstanding antimicrobial-activity against pathogenic bacterial and fungal strains. Moreover, SeNPs have promising anticancer activity against HepG2 cancerous cell line without cytotoxicity on Vero normal cell line. Finally, the biosynthesized SeNPs via aqueous extract of stinging nettle leaf exhibited potential antibacterial, antifungal, and anticancer action, making them useful in the medical field.

Selenium nanoparticle synthesis from endangered medicinal herb (Enicostema axillare).

Green synthesis of selenium nanoparticles (SeNPs) was achieved by a simple biological procedure using the reducing power of E. axillare leaf extract. This method is capable of producing SeNPs in a size range of around 56.23-98.18nm, under ambient lab condition. The synthesized nanoparticles were characterized by UV-Vis spectroscopy, Fourier-transform infrared spectroscopy (FTIR), Scanning electron microscopy (SEM) and Energy-dispersive X-Ray (EDX). To check the potential hazards of nanotoxicity median lethal concentration (LC50) and Na + /K + -ATPase activity was assessed on zebra fish which showed the LC50 of258.72ppm for 96h and Na + /K + -ATPase seems to significantly decrease with increase in SeNPs concentration. Antibactericidal activity explored the use of SeNPs against wide range of pathogens. Cytotoxicity of SeNPs was assayed against human lung cancer cells (A549) confirmed that SeNPs are able to inhibit the cell growth by dose-dependent manner. In conclusion, green synthesized selenium nanoparticles are less toxic and harmless. This is the first report on green synthesized selenium nanoparticles using E. axillare possessing anti-bacterial and anti-cancer activity.

Mutual influence of selenium nanoparticles and FGF2-STAB\xae on biocompatible properties of collagen/chitosan 3D scaffolds: in vitro and ex ovo evaluation

In a biological system, nanoparticles (NPs) may interact with biomolecules. Specifically, the adsorption of proteins on the nanoparticle surface may influence both the nanoparticles' and proteins' overall bio-reactivity. Nevertheless, our knowledge of the biocompatibility and risk of exposure to nanomaterials is limited. Here, in vitro and ex ovo biocompatibility of naturally based crosslinked freeze-dried 3D porous collagen/chitosan scaffolds, modified with thermostable fibroblast growth factor 2 (FGF2-STAB®), to enhance healing and selenium nanoparticles (SeNPs) to provide antibacterial activity, were evaluated. Biocompatibility and cytotoxicity were tested in vitro using normal human dermal fibroblasts (NHDF) with scaffolds and SeNPs and FGF2-STAB® solutions. Metabolic activity assays indicated an antagonistic effect of SeNPs and FGF2-STAB® at high concentrations of SeNPs. The half-maximal inhibitory concentration (IC50) of SeNPs for NHDF was 18.9 µg/ml and IC80 was 5.6 µg/ml. The angiogenic properties of the scaffolds were monitored ex ovo using a chick chorioallantoic membrane (CAM) assay and the cytotoxicity of SeNPs over IC80 value was confirmed. Furthermore, the positive effect of FGF2-STAB® at very low concentrations (0.01 µg/ml) on NHDF metabolic activity was observed. Based on detailed in vitro testing, the optimal concentrations of additives in the scaffolds were determined, specifically 1 µg/ml of FGF2-STAB® and 1 µg/ml of SeNPs. The scaffolds were further subjected to antimicrobial tests, where an increase in selenium concentration in the collagen/chitosan scaffolds increased the antibacterial activity. This work highlights the antimicrobial ability and biocompatibility of newly developed crosslinked collagen/chitosan scaffolds involving FGF2-STAB® and SeNPs. Moreover, we suggest that these sponges could be used as scaffolds for growing cells in systems with low mechanical loading in tissue engineering, especially in dermis replacement, where neovascularization is a crucial parameter for successful skin regeneration. Due to their antimicrobial properties, these scaffolds are also highly promising for tissue replacement requiring the prevention of infection.

Ecofriendly novel synthesis of tertiary composite based on cellulose and myco-synthesized selenium nanoparticles: Characterization, antibiofilm and biocompatibility

Microbial infections are considered common and dangerous for humans among other infections; therefore the synthesis of high efficacy antimicrobial and anti-biofilm composites is continuous to fight microbial resistance. In our study, a new and novel tertiary composite (TC) was synthesized, it composed of TEMPO cellulose (TOC), chitosan, starch, and myco-synthesized Se-NPs. Myco-synthesized Se-NPs and TC were fully characterized using UV, FT-IR, XRD, SEM with EDX, particle distribution, and mapping. The antimicrobial and anti-biofilm properties of selenium nanoparticles (Se-NPs) were effectively established for Pseudomonas aeruginosa and Staphylococcus aureus biofilms. The possible impact of myco-synthesized novel cellulose-based selenium nanoparticles tertiary composite on the biofilm formation of P. aeruginosa, S. aureus, and Candida albicans was evaluated in this study. TC exhibited constant biofilm inhibition against P. aeruginosa, S. aureus, and C. albicans, while the results obtained from cytotoxicity of Se-NPs and TC showed that, alteration occurred in the normal cell line of lung fibroblast cells (Wi-38) was shown as loss of their typical cell shape, granulation, loss of monolayer, shrinking or rounding of Wi-38 cell with an IC50 value of where 461 and 550 ppm respectively.

Comparative Study of the Antimicrobial Activity of Selenium Nanoparticles With Different Surface Chemistry and Structure.

Although selenium nanoparticles (SeNPs) have gained attention in the scientific community mostly through investigation of their anticancer activity, a great potential of this nanomaterial was recognized recently regarding its antimicrobial activity. The particle form, size, and surface chemistry have been recognized as crucial parameters determining the interaction of nanomaterials with biological entities. Furthermore, considering a narrow boundary between beneficial and toxic effects for selenium per se, it is clear that investigations of biomedical applications of SeNPs are very demanding and must be done with great precautions. The goal of this work is to evaluate the effects of SeNPs surface chemistry and structure on antimicrobial activity against several common bacterial strains, including Staphylococcus aureus (ATCC 6538), Enterococcus faecalis (ATCC 29212), Bacillus subtilis (ATCC 6633), and Kocuria rhizophila (ATCC 9341), as well as Escherichia coli (ATCC 8739), Salmonella Abony (NCTC 6017), Klebsiella pneumoniae (NCIMB 9111) and Pseudomonas aeruginosa (ATCC 9027), and the standard yeast strain Candida albicans (ATCC 10231). Three types of SeNPs were synthesized by chemical reduction approach using different stabilizers and reducing agents: (i) bovine serum albumin (BSA) + ascorbic acid, (ii) chitosan + ascorbic acid, and (iii) with glucose. A thorough physicochemical characterization of the obtained SeNPs was performed to determine the effects of varying synthesis parameters on their morphology, size, structure, and surface chemistry. All SeNPs were amorphous, with spherical morphology and size in the range 70–300 nm. However, the SeNPs obtained under different synthesis conditions, i.e. by using different stabilizers as well as reducing agents, exhibited different antimicrobial activity as well as cytotoxicity which are crucial for their applications. In this paper, the antimicrobial screening of the selected systems is presented, which was determined by the broth microdilution method, and inhibitory influence on the production of monomicrobial and dual-species biofilm was evaluated. The potential mechanism of action of different systems is proposed. Additionally, the cytotoxicity of SeNPs was examined on the MRC-5 cell line, in the same concentration interval as for antimicrobial testing. It was shown that formulation SeNPs-BSA expressed a significantly lower cytotoxic effect than the other two formulations.

Selenium nanoparticles: synthesis, characterization and study of their cytotoxicity, antioxidant and antibacterial activity

This report concerns the green synthesis of selenium nanoparticles (Se-NPs) through chemical reduction method using ascorbic acid (Vitamin C) as a reluctant and polyvinyl alcohol (PVA) or chitosan (CS) as stabilizers. Ultraviolet-visible (UV–vis) absorption spectroscopy, dynamic light scattering (DLS), fourier transform infrared spectroscopy (FT-IR) and transmission electron microscopy (TEM) were applied as the main characterization tools. The cytotoxicity of Se-NPs on fibroblast L929 cells was determined by MTT assay. The nanoparticles showed low toxicity toward the investigated cell line which remained more than 50% viable at 170 ppm for 24 h. The in-vitro antioxidant property of the synthesized Se nanoparticles was evaluated by radical scavenging (DPPH) assay. The IC50 values of the nanoparticles were 26.56 and 530 ppm for PVA coated SeNPs (PVA-SeNPs) and CS coated SeNPs (CS–SeNPs), respectively. Finally, the antibacterial activity of Se-NPs was determined on the gram-negative bacteria Escherichia coli, Pseudomonas aeruginosa, and the gram positive bacteria Staphylococcus aureus, Streptococcus epidermidis. The results showed that Se nanoparticles exhibited dose-dependent antibacterial activity against all the four bacterial strains tested. Noticeably, PVA-SeNPs exhibited significant effect against S. epidermidis (MIC 125 ppm) and S. aureus (MIC 125 ppm). The results of this study indicate that Se nanoparticles can be potentially used as antimicrobial and antioxidant agents.

Biosynthesis, Characterization and Cytotoxicity of Selenium Nanoparticles Using Bacillus tropicus Ism 2

Green biosynthesis of selenium nanoparticles (SeNPs) using Bacillus tropicus Ism 2 (MK332444) isolated from soil polluted with industrial wastewater near the Ismailia Canal, Egypt, was carried out. SeNPs were biogenically synthesized using bacterial pellets mixed with 1 mM Na2SeO3. Characterization of biosynthesized SeNPs was carried out by UV-Vis spectrum where SeNPs solution exhibited characteristic peak at 270 nm. Dynamic Light Scattering (DLS) analysis and High-Resolution Transmission Electron Microscope (HRTEM) showed spherical shaped SeNPs with size ranged from 60 to 125 nm with PDI= 0.544 and zeta potential value of -30.6. Biologically synthesized SeNPs showed no antibacterial activity against both Gram-positive and Gram-negative bacteria. Cytotoxicity of SeNPs was studied against Huh-7 human liver cell line and showed low cytotoxicity with IC50 value of367±5.5 μg/ml. In conclusion in this work Bacillus tropicus Ism 2 locally isolated bacterium showed the capability to biosynthesize SeNPs with spherical shape and low cytotoxicity.

Green synthesis and characterization of selenium nanoparticles and its augmented cytotoxicity with doxorubicin on cancer cells

Green synthesis of selenium nanoparticles (SeNPs) was achieved by a simple biological procedure using the reducing power of fenugreek seed extract. This method is capable of producing SeNPs in a size range of about 50-150 nm, under ambient conditions. The synthesized nanoparticles can be separated easily from the aqueous sols by a high-speed centrifuge. These selenium nanoparticles were characterized by UV-Vis spectroscopy, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and elemental analysis by X-ray fluorescence spectrometer (XRF). Nanocrystalline SeNPs were obtained without post-annealing treatment. FTIR spectrum confirms the presence of various functional groups in the plant extract, which may possibly influence the reduction process and stabilization of nanoparticles. The cytotoxicity of SeNPs was assayed against human breast-cancer cells (MCF-7). It was found that SeNPs are able to inhibit the cell growth by dose-dependent manner. In addition, combination of SeNPs and doxorubicin shows better anticancer effect than individual treatments.

Cytotoxicity of selenium nanoparticles in rat dermal fibroblasts

Background:Ventilator-associated pneumonia is a deadly nosocomial infection caused by contaminated endotracheal tubes. It has been shown that polyvinyl chloride (PVC, the endotracheal tube substrate) coated with elemental selenium nanoparticles reduces bacterial adherence and proliferation on PVC by over 99%. However, it is not known if selenium nanoparticles elicit a cytotoxic effect in vitro. The purpose of this study was to investigate the cytotoxic effects of PVC coated with selenium nanoparticles on fibroblasts, which are mammalian cells central to endotracheal tube intubation.Methods:Different concentrations of selenium nanoparticles were precipitated onto the PVC surface by reduction of selenium salts using glutathione. Characterization of PVC coated with selenium nanoparticles was done by scanning electron microscopy, energy dispersive x-ray, and contact angle measurements. For the cytotoxicity experiments, fibroblasts were seeded at a density of 5000 cm2 onto PVC coated with three different concentrations of selenium nanoparticles (high, medium, low) and incubated for 4 hours (adhesion) as well as for 24 hours and 72 hours (proliferation). The half-maximal inhibitory concentration (IC50) value was determined after 72 hours using an ultrahigh concentration. MTT assays were used to assess cell viability at the indicated time points.Results:The three concentrations of selenium nanoparticles did not elicit a cytotoxic effect after 72 hours (P < 0.01, n = 3). It was found that the IC50value was at the ultrahigh concentration of selenium nanoparticles. The nanoparticulate elemental selenium concentration previously shown to decrease the function of bacteria was shown not to cause a cytotoxic effect on fibroblasts in vitro.Conclusion:These findings demonstrate great selectivity between bacteria and healthy cells, and are a viable option for coating endotracheal tubes in order to prevent ventilator-associated pneumonia.

Surface decoration by Spirulina polysaccharide enhances the cellular uptake and anticancer efficacy of selenium nanoparticles

A simple and solution-phase method for functionalization of selenium nanoparticles (SeNPs) with Spirulina polysaccharides (SPS) has been developed in the present study. The cellular uptake and anticancer activity of SPS-SeNPs were also evaluated. Monodisperse and homogeneous spherical SPS-SeNPs with diameters ranging from 20 nm to 50 nm were achieved under optimized conditions, which were stable in the solution phase for at least 3 months. SPS surface decoration significantly enhanced the cellular uptake and cytotoxicity of SeNPs toward several human cancer cell lines. A375 human melanoma cells were found extremely susceptible to SPS-SeNPs with half maximal (50%) inhibitory concentration value of 7.94 μM. Investigation of the underlying mechanisms revealed that SPS-SeNPs inhibited cancer cell growth through induction of apoptosis, as evidenced by an increase in sub-G1 cell population, deoxyribonucleic acid fragmentation, chromatin condensation, and phosphatidylserine translocation. Results suggest that the strategy to use SPS as a surface decorator could be an effective way to enhance the cellular uptake and anticancer efficacy of nanomaterials. SPS-SeNPs may be a potential candidate for further evaluation as a chemopreventive and chemotherapeutic agent against human cancers.
