Isolation and structure elucidation of the compounds from Teucrium hyrcanicum L. and the investigation of cytotoxicity, antioxidant activity, and protective effect on hydrogen peroxide-induced oxidative stress

BackgroundTeucrium hyrcanicum L. (family Lamiaceae) is widely distributed in the North and Northwest of Iran. It has been used in the form of tea, tonic, and tincture for the treatment of various diseases such as cough, rheumatism, and fever.MethodsIn this study, the total phenolic and flavonoid contents, antioxidant and cytotoxic activities of methanol extract and different fractions of T. hyrcanicum were measured. Furthermore, the potential ability of T. hyrcanicum to protect against H2O2-induced oxidative stress was tested on the NIH3T3 cell line. Then, the isolation and structure elucidation of the compounds were performed on the most potent fractions. Finally, the quantification of isolated compounds in methanol extract (ME) was done by the HPLC method. Isolated phytochemicals were assessed for the cytotoxic and antioxidant activities.ResultsThe results indicated that the methanol fraction (MF) had the highest amount of phenolic and flavonoid contents (69.36 mg GAE/g extract and 68.95 mg QE/g extract). The highest radical scavenging activities were observed from MF and ME (IC50 44.32 and 61.12 μg.ml−1, respectively). The best cytotoxicity was obtained by ethyl acetate fraction (EF) against A431 and MCF7 cell lines (IC50 values of 235.4and 326.6 μg.ml−1, respectively). The pretreatment with MF exerts the highest reduction in malondialdehyde (MDA) formation (IC50 2.51 μM, p < 0.001) compared to the H2O2 group (5.77 μM). Also, MF significantly inhibited H2O2-induced Glutathione (GSH) oxidation (p < 0.001). Furthermore, two phenolic compounds, acteoside and quercetin, were isolated and identified in MF and EF, respectively. The IC50 values of acteoside and quercetin in the DPPH assay were 7.19 and 5.56 µg.ml−1, respectively. Both quercetin and acteoside significantly reduced the MDA formation and inhibited GSH oxidation, which was comparable with BHA (as a standard antioxidant) (p < 0.05). Acteoside demonstrated significant cytotoxicity against all tested cell lines (IC50 = 32 to 145 μg.ml−1). The HPLC quantification of isolated compounds revealed that the quantity of acteoside and quercetin in ME were 93.31 and 16.87 μg.mg−1, respectively.ConclusionThe isolated compounds (quercetin and acteoside) had significant antioxidant activities and revealed a protective effect on H2O2-induced oxidative stress which was comparable with BHA.

Potential Cytotoxic Activity of Methanol Extract, Ethyl Acetate, and n-Hexane Fraction from Clitoria ternatea L. on MCF-7 Breast Cancer Cell Line and Molecular Docking Study to P53

Breast cancer is a condition where the cells in breast tissue lose control and multiply uncontrollably. In this study, MCF-7 breast cancer cells were tested for cytotoxic activity using the MTT assay and the active compound's interaction with the p53 protein was tested in silico. The most active fraction was found to be the ethyl acetate fraction, with an IC50 value of 1.730 μg/mL and a selectivity index of 2.485. However, the selectivity index was less than 3, and Vero cells showed changes in morphology with the addition of the ethyl acetate fraction. GC-MS was used to identify 19 compounds in the ethyl acetate fraction, and in-silico tests were performed on 5 potential anticancer compounds. Lipinski's Rule of Five test showed that only 3 of these compounds could undergo molecular docking. The results indicated that Anethole compound can interact with p53 protein, while Cinnamaldehyde, (E)- can interact with p21 protein.

In Vitro Cytotoxic Activity of Methanol Extracts of Selected Medicinal Plants Traditionally Used in Mexico against Human Hepatocellular Carcinoma.

Medicinal plants are traditionally used in Mexico to treat diseases such as cancer. The present study aimed to evaluate the cytotoxic, antioxidant, and anti-hemolytic activity of 15 plants of ethnopharmacological use in Mexico. For this, plant methanol extracts were prepared by the Soxhlet method, after which their cytotoxic activity was evaluated against human hepatocellular carcinoma (HEP-G2) and monkey kidney epithelial (Vero) cells by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction colorimetric assay. The selectivity index (SI) of each extract was then determined by the IC50 ratio of normal to tumor cells. We showed that Ruta chalepensis extract possessed an IC50 of 1.79 µg/mL and 522.08 µg/mL against HEP-G2 and Vero cells, respectively, resulting in an SI of 291.50. Furthermore, antioxidant activity was evaluated by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging technique, where the best antioxidant potential was shown by the Heterotheca inuloides extract (IC50 = 19.24 µg/mL). Furthermore, the hemolytic potential was determined against human erythrocytes, which showed that the extracts with the highest anti-hemolytic activity were Smilax aspera (IC50 = 4.41 µg/mL) and Amphipterygium adstringens (IC50 = 5.35 µg/mL). In conclusion, we observed that R. chalepensis methanol extract possesses cytotoxic activity against HEP-G2 cells, without affecting non-tumorigenic Vero cells. Our results indicated the antitumor potential of medicinal plants used in Mexico.

Cytotoxic evaluation and LC-MS/MS analysis of aerial parts of Eryngium kotschyi Boiss. grown in Turkey

Increasing biological activity and phytochemical investigations on Eryngium species showed its potential as pharmaceutical approach. Eryngium kotschyi Boiss. is one of the species of Eryngium genus and is endemic to Turkey. It is known that this plant is traditionally used in the South-western part of Turkey for the treatment of various diseases. This study focuses on cytotoxic activities of methanol extract and ethyl acetate, n-butanol and water sub-extracts from E. kotschyi in A549, COLO 205 and MDA-MB-231 cell lines by Sulforhodamin B assay and qualitative and quantitative determination of phytochemical constituents in active extract by LC-MS/MS. From the result of the study, it was seen that E. kotschyi ethyl acetate (EKE) sub-extract showed the strongest cytotoxic effect with the low IC50 values (50.00; 31.96 and 22.26 μg/mL in A549; COLO 205 and MDA-MB-231 cells at 48 h, respectively). Preliminary examination of the mass spectrums revealed the presence of 15 phytochemical compounds in active sub-extract and 7 of them was quantified. According to quantitative analyses the main compounds of EKE sub-extract were rosmarinic acid (485.603 µg/mgextract), chlorogenic acid (62.355 µg/mgextract) and caffeic acid (59.266 µg/mgextract). Moreover, this preliminary study on inhibitory activity of EKE sub-extract suggests further toxicologic investigations and detailed investigation on cytotoxic effect of various combinations of determined compounds.

Antioxidant, Cytotoxic, Antidiabetic and Phytochemical Investigation of Root Extracts of Tecoma stans (Bignoniaceae)

Introduction: The plants are an alternative source for the management of a number of human disorders due to the presence of biologically active constituents Aims: Current study aims to investigate underexplored therapeutic potentials of roots of Tecoma stans through phytochemical and biological assays. Methods: The dried roots were extracted successively with methanol and dichloromethane by simple maceration and labeled with codes TSM and TSD respectively. Results: The phytochemical investigation revealed the presence of alkaloids, flavonoids, saponins, terpenoids, tannins, andphenols in the extracts. TSM fraction revealed the highest total phenolic, flavonoid and alkaloid contents159.79 mg GAE/g, 65.38to 13.04 mg QE/g and95.2 to 56.61 mg AT/g of extract respectively. As compared to TSD extract, TSM extract showed significant antioxidant activity using two models DPPH (79.8 %) and ABTS (71.06 %). Cytotoxic activity of methanol extracts against PC3 and HeLa cancer cell line exhibited 61.23% (IC50 22.89 µg/mL) and 74.03 % (17.78 µg/mL) inhibition of cancer cells respectively at concentration of 30 µg/mL. Extracts revealed antidiabetic activity through α-amylase and α-glucosidase inhibition with maximum inhibition 63.26% and 75.66% respectively was detected at the concentration of 1000 mg / mL by TSM extract. Conclusion: Methanolic extracts of roots of Tecoma stanscontainshigher amount plant secondary metabolites and revealed antioxidant, cytotoxic and α-amylase as well as α- glucosidase inhibition activities.

A Comparative Study on Antioxidant and Cytotoxic Potentials of Methanol Extract of Jatropha curcas Seeds and Leaves

Many medicinal plants have been reported to exhibit protective effects against many physiological diseases as a result of their phytochemical components which are effective antioxidants. This study was aimed at comparing the phenolic and flavonoids contents, antioxidant and cytotoxic activities of methanol extract of Jatropha curcas seeds and leaves. The cytotoxicity level of J. curcas was assessed using the brine shrimp lethality test (BSLT). The antioxidant activity was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP), nitric oxide scavenging activities, 2, 2’-azino-bis-(3 ethyl benzothiazoline-6-sulphonic acid (ABTS) + scavenging activities and total antioxidant capacity. The total flavonoids and total phenols content (TPC) were carried out using aluminum chloride and Folin-Ciocalteu assay respectively. The results showed that J. curcas leaves have significant higher phenolic and flavonoids contents than the seeds. The total antioxidant capacity and FRAP were also significantly higher in the leaves than the seeds of J. curcas. Furthermore, the DPPH, ABTS and Nitric oxide scavenging activities were significantly higher in the leaves than the seeds. The J. curcas leaves have a higher LD50 than the seeds. In conclusion, this study suggests the antioxidant potency as well as the safety of the crude methanol extract of the leaves of J. curcas over the seeds.&#x0D; Keywords: Jatropha curcas, antioxidant, cytotoxicity, flavonoids, brine shrimp lethality test, total phenolic compounds

Cytotoxic Activities of Methanol Extract and Compounds of Porodaedalea pini Against Colorectal Cancer

Porodaedalea pini is a medicinally important mushroom with antioxidant, cytotoxic, immunostimulating, antitumor, antiviral and immunomodulating activities. Therefore, in this study, P. pini methanol extract and isolated compounds from the methanol extract were tested for cytotoxic activities against DLD-1 (colorectal cancer) and CCD-18Co (human colon fibroblast cell line) by using Alamar Blue assay. Cytotoxic activity on DLD-1 was decreased in the order of P. pini methanol extract&amp;gt; 4-(3,4-dihydroxyphenyl)but-3-en-2-one (3)&amp;gt; pinoresinol (2)&amp;gt; ergosta-7,24(28)-dien-3β-ol (1). P. pini methanol extract was determined to have the best cytotoxic activity with the lowest IC50 value on DLD-1 (IC50: 25.33±0.29 µg/mL) and the highest IC50 value on CCD-18Co (434.30±1.45 µg/mL). Within the scope of the findings, it is thought that P. pini mushroom can be used as a new and natural agent in the treatment of colorectal cancer.

Anti-inflammatory, antinociceptive, and cytotoxic activity of methanolic extract of Mansoa hymenaea (DC.) A.H. Gentry

ABSTRACT Mansoa hymenaea (DC.) A.H. Gentry is used in traditional medicine to treat pain, arthritis, and rheumatoid disease. It has also been demonstrated that species belonging to the Mansoa genus exert cytotoxic activity. Despite this data, biological properties and phytochemistry of M. hymenaea remain poorly understood. Therefore, we determined the metabolite profile of the methanolic extract of M. hymenaea leaves and evaluated its anti-inflammatory, antinociceptive, and cytotoxic activity. Phytochemical analysis was carried out by conventional methods and gas chromatography-mass spectrometry. Anti-inflammatory and antinociceptive effects were assessed in the TPA‑induced ear edema model and acetic acid-induced writhing test, respectively. Cytotoxic activity was determined in vitro against SW-620, MDA-MB231, SiHa, SKLU1, and PC-3 cancer cell lines. The results revealed that M. hymenaea methanolic extract at 2 mg/ear reduced inflammation significantly (p ≤ 0.05) by 50.4 ± 5.9 %, and decreased visceral pain with an effective dose 50 (ED50) of 148.9 ± 14.1 mg kg−1. Moreover, M. hymenaea exerted cytotoxic activity against the breast carcinoma cell line (MDA-MB231) with IC50 = 125.8 ± 16.7 µg mL−1. Chemical analyses revealed that M. hymenaea methanolic extract contained alkaloids, glycosides, tannins, sterols, and saponins. Likewise, trans-13-octadecenoic acid, n-hexadecanoic acid, diallyl disulfide, diallyl trisulfide, β-sitosterol, methyl ursolate, stigmasterol, and phytol were detected by CG-MS in the M. hymenaea methanolic extract. These compounds were previously reported with anti-inflammatory, antinociceptive, or cytotoxic effects. Therefore, the methanolic extract of M. hymenaea leaves is a good source of natural compounds with anti-inflammatory, antinociceptive, and cytotoxic properties.

Deciphering the Pharmacological Properties of Methanol Extract of Psychotria calocarpa Leaves by In Vivo, In Vitro and In Silico Approaches.

The present study explores the neuropharmacological, antinociceptive, antidiarrheal, antioxidant, thrombolytic and cytotoxic activity of methanol extract of Psychotria calocarpa leaves (MEPC). In anxiolytic activity testing of MEPC by elevated plus maze test, hole–board test and light–dark test, the extract exhibited a dose-dependent reduction of anxiety while the open field test observed a decreased locomotion. The administration of MEPC revealed a significant dose-dependent reduction of depressant behavior in forced swimming and tail suspension test. Additionally, the antinociceptive and antidiarrheal activity exposed a significant reduction of nociception and diarrheal behavior at the highest dose. In addition, a strong antioxidant activity was observed in DPPH-free radical-scavenging assay (IC50 = 461.05 μg/mL), total phenol content (118.31 ± 1.12 mg) and total flavonoid content (100.85 ± 0.97 mg). The significant clot–lysis activity was also observed with moderate toxicity (LC50 = 247.92 μg/mL) level in the lethality assay of brine shrimp. Moreover, in silico molecular docking study showed that the compound Psychotriasine could offer promising active site interactions for binding proteins. Furthermore, ADME/T and toxicological properties of the compound satisfied the Lipinski’s rule of five and Veber rules for drug-like potential and toxicity level. Overall, MEPC had a potential neuropharmacological, antinociceptive, antidiarrheal and antioxidant activity that warranted further investigation.

Chemical Composition and Biological Activities of Red beetroot (Beta Vulgaris Linnaeus) Roots

Plants have been used for many years as a source of traditional medicine to treat various diseases and conditions. Beta Vulgaris Linnaeus ranks among the 10 most powerful vegetables as excellent sources for phytochemicals, which showed potent antioxidant and anticancer activities. The aim of this study is to characterize the chemical composition and determine the cytotoxicity and antibacterial activity of B.Vulgaris roots extract. The results of GC-Ms analysis of the unsaponifiable matter of B.Vulgaris roots showed the presence of 18 components. The undecane is the major hydrocarbon compound and α-citronellol is the major oxygenated compound. The methanolic extract of B.Vulgaris roots were enriched with a complicated mixture of phenolic acids and flavoniod which elucidated for the first time from this plant , including polyphenolic acid; ellagic acid (1); pyrogallol(2); salicylic(3); catechol(4); benzoic(5); protocatchuic (6); chlorogenic(7); p-hydroxy benzoic acids (8) . The molecular components of a flavoniod fraction, obtained from liquid chromatography extract, were identified using HPLC -ESI -MS. The major components were vitexin -2 -O-rhamnoside (1); demethylated -2-xylosylvitexin (2); isorhamnetin -3-gentiobioside (3); rutin(4); beside minor components , luteolin-6-arabinose-8-glucose(1); hesperidin (2); acacetin (3); kaempferol-3-(2 -p -comaroyl) glucose (4); apigenin 6 -arabinose -8-glactose (5); quercetin (6); naringin (7). The structures of all isolated compounds were elucidated by conventional methods, spectroscopic analysis alongside their mass spectrometric investigations. The search for new, potentially biologically active extract becomes much more efficient after identification of all compounds in that mixture. The cytotoxic activity of methanolic extract was evaluated against pancreatic carcinoma (PANC-1) , hepatocellular carcinoma cell line HEPG-2 and , Lung carcinoma (A549) using MTT assay and vinblastine as a reference drug. methanolic extract showed higher activity with (IC50=3.69 µg/ml), mild cytotoxic activity against HepG2 with (IC50=4.43 µg/ml) ,Lung carcinoma (A549) with (IC50=4.9 µg/ml) and a weak activity against prostate carcinoma PC-3 with (IC50=6.1 µg/ml). The antimicrobial activity of B.Vulgaris roots methanolic extract was evaluated using agar well diffusion method towards two representatives for each of Gram positive and Gram negative bacteria. The extract demonstrated inhibitory effect against pathogenic microorganisms: Staphylococcus aureus, ATCC 29213; (ZI=24mm) strains and Pseudomonas aerogenasa ATCC 27853); (ZI=19.3 mm) at 100 µL concentration.

Evaluation of Antioxidant, Antibacterial and Cytotoxic Activity of Methanol Extract from Leaves and Fruits of Iranian Squirting Cucumber (Ecballium elaterium (L.) A. Rich)

Background and objectives: Ecballium elaterium (L.) A. Rich (squirting cucumber) has been used traditionally as a remedy for different disorders such as fever, sinusitis, and rheumatic disease. In the present study, antibacterial, antioxidant, and cytotoxic activities of leaves and fruits were evaluated. Also, total flavonoid and phenolic contents were measured were measured. Methods: The fruits and leaves of E. elaterium were extracted by percolation method with methanol. The antioxidant activity was assessed by 2, 2'-diphenyl-1-picrylhydrazyl (DPPH) methods. Then, the total phenolics and flavonoids contents were measured. The cytotoxicity was tested against three cancerous cell lines (MCF-7, MDA-MB-468 and MKN-45) and a normal cell line (HDF). Antibacterial activity was investigated against Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa, and Escherichia coli. Results: The total phenolics content of leaves and fruits were 39.97 and 30.90 mg gallic acid equivalents/g of dry extract, respectively. Also, the total flavonoids content in leaf extract was 49.17 mg quercetin as equivalents/g of dry extract while flavonoids were not detected in the fruit extract. In the DPPH assay, the IC50 values were 1.15 and 1.18 mg/mL for leaves and fruits, respectively.The fruit extract showed the most considerable antibacterial activity (MIC 37.5 mg/mL) against P. aeruginosa. Both extracts indicated cytotoxicity on MDA-MB-468 cells (IC50 264 and 50 µg/mL, respectively). Conclusion: The antioxidant, antibacterial, and cytotoxicity of extracts may be due to some secondary metabolites like phenolic compounds and flavonoids. This study suggests that this plant could be considered for further investigations as a natural source of biological compounds.

Ripening of ‘Hass’ avocado mesocarp alters its phytochemical profile and the in vitro cytotoxic activity of its methanolic extracts

Avocado is an excellent source of phytochemicals with remarkable bioactive properties. This study evaluated the effect of ripeness stage of ‘Hass’ avocado on the profile and content of lipophilic and hydrophilic phytochemicals and their in vitro cytotoxic activity. Qualitative and quantitative analyses of the major phytochemicals were conducted in four stages of ripeness, using gas and liquid chromatography analyses. In general, phenolics, carotenoids, tocopherols, and phytosterols tended to increase during ripening. A decrease of individual phytochemicals was observed, namely, protocatechuic acid (0.36 to 0.03 mg 100 g−1 dw) and β-carotene (0.9 to 0.4 mg 100 g−1 dw), while quercetin and δ-tocopherol were only detected in a single ripeness stage (1 and 3, respectively). The major phytochemicals quantified at the end of ripening were epicatechin (12.66 mg 100 g−1 dw), lutein (0.5 mg 100 g−1 dw), α-tocopherol (12.4 mg 100 g−1 oil) and β-sitosterol (35.6 mg 100 g−1 dw) for phenolics, carotenoids, tocopherols and phytosterols, respectively. The cytotoxic activity of methanolic extracts of avocado was tested against RAW 264.7 and HeLa cells, where all RS showed a dose-dependent effect. We conclude that ripeness stage influences the concentration of specific compounds in each group of phytochemicals, as well as its in vitro cytotoxic activity, something that must be considered in future studies on the potential health benefits of avocado.

Mangifera indica L. Leaves Extract Induced Intrinsic Apoptotic Pathway in MCF-7 Cells by Decreasing Bcl-2 Expression and Inducing Bax Expression

Extract of Mangifera indica L. has been investigated to have cytotoxic activity against MCF-7 cells. Current research was conducted to determine the cytotoxic activity of methanol extract of M. indica leaves (MEML) on MCF-7 breast cancer cells and to investigate on the MEML's effect on Bcl-2 and Bax expressions. M. indica leaves were extracted using soxhletation with methanol. The cytotoxic test was perfomed using MTT assay with various MEML concentrations: 15.625, 31.25, 62.5, 125, 250 and 1000 μg/mL. Meanwhile, Bax and Bcl-2 expressions of MCF-7 breast cancer cells were detected with immunocytochemical staining after treatment of MEML with concentrations of ¼ IC50, ½ IC50 and IC50. The results showed that IC50 of cytotoxic activity was 308.12 µg/mL. MEML could influence Bax and Bcl-2 protein expressions. After treatment of MEML, Bcl-2 expression was decreased while Bax expression was increased.Keywords : Mangifera indica L., Cytotoxic, Bax, Bcl-2, MCF-7

Bioactivities of leaf and root extract of Ceriscoids turgida (Roxb.)

In traditional medicine leaf and root of Ceriscoides turgida (Roxb.) are commonly used to treat indigestion in child, leucorrhoea, dog bite, snake bite, scorpion string, epilepsy, stomachache and a number of ailments. The present study was carried out to investigate antioxidant, analgesic, antibacterial and cytotoxic activities of methanol extract of leaves and roots of C. turgida. Total flavonoids content was spectrophotometrically determined using aluminum chloride colorimetric assay while total phenolic and tannin content by Folin Chiocalteu’s reagent. Antioxidant activity was determined by DPPH free radical scavenging and reducing power assay. Analgesic activity was assessed by acetic acid induced writhing method. Antibacterial activity was evaluated by disc diffusion assay while brine shrimp lethality bioassay was carried out to verify the cytotoxic potential. Root extract contains greater amount of total phenolic (79.99 mg GAE/g dry extract) and flavonids content (35.27 mg QE/g dry extract) in comparison to leaf extract (30.75 mg GAE/g and 25.93 mg QE/g dry extract respectively). Root extracts showed DPPH scavenging (157.45 μg/mL) that was comparable to standard ascorbic acid (14.15 μg/mL). Reducing power of root extract was prominent than leaf extract in comparison to ascorbic acid. Both extract showed significant (P < 0.01) analgesic activity. In disc diffusion assay leaf extract showed greater antibacterial activity than root extract. Furthermore, extracts showed moderate cytotoxic effect. The present study demonstrates that methanol extract of C. turgida leaf and root has significant antioxidant and analgesic effect. The extracts also showed moderate antibacterial and cytotoxic activities.

Characterization of Cnidoscolus quercifolius Pohl bark root extract and evaluation of cytotoxic effect on human tumor cell lines

Objective: To evaluate the chemical components of active extract from Cnidoscolus quercifolius root bark and its cytotoxic potential against several tumor strains. Methods: The high-performance liquid chromatography with diode-array detection and 1H and 13C nuclear magnetic resonance spectroscopy of the extract were used to distinguish the existence of possible functional groups in the root bark extract. The in vitro cytotoxic activity of methanol extract on human colon cancer cell lines was evaluated using OVCAR-8, SF-295, HCT-116, HL-60 strains and the samples were assessed by 3-(4,5-dimethylthiazol2-yl)-2,5-diphenyltetrazolium bromide method. Results: The analysis of nuclear magnetic spectra of the active chloroform fraction revealed the presence of absorptions bands correspondent to a mixture of favelines such as neofavelanone, deoxofaveline or methyl-faveline, which structures were confirmed by ultraviolet spectra upon high-performance liquid chromatography with diode-array detection analysis. The active fraction showed cytotoxic effects in the tested strains, HCT-116, SF-295, OVCAR-8 and HL-60 cells with IC50 of 72 hours ranging from 4.95 to 15.23 μg/mL. Conclusions: The results suggest that the substances present in faveleira (Cnidoscolus quercifolius) root bark extract have a cytotoxic potential against several tumor lines, showing a broader antitumour potential, and in addition no adverse to healthy cells. Therefore, the root bark extract of Cnidoscolus quercifolius has a possibility of use for anticarcinogenic therapies.

Chemical Composition and Cytotoxic Activity of Methanol Extract and its Fractions of Streblus asper Leaves on Human Cancer Cell Lines.

Background:Streblus asper, family Moraceae is well-known important medicinal plant used in the Indian system of medicine. In Ayurveda, stem bark of S. asper is recommended against elephantiasis for which there is still no any other effective medicine in the modern system of medicine.Objectives:In the present work, methanol extract (SAM) and its fractions of S. asper leave tested for in vitro anticancer activity against cancer cell lines (MCF-7, A-549, Hep-G2, and K-562) which claims its folklore importance in cancer and gas chromatography-mass spectrometry identification of extracts was also performed.Materials and Methods:Shade dried plant material was extracted with methanol and fractionated sequentially with hexane, chloroform, and butanol.Results:All tested extracts found highly effective against human lung cancer cell line (A-549) with IC50 <10 μg/mL. On Hep-G2 cancer cell line, only chloroform fraction are highly active with IC50 <10 μg/mL. Methanol and hexane fraction showed potent anticancer activity on K-562 cancer cell line with IC50 <10 μg/mL.Conclusion:Qualitative phytochemical analysis confirmed the presence of fatty acids, phytosterol, triterpenoids, polyol, sugar acid, aldehyde, diterpene, terpene, carboxylic compounds, acid and sugar in S. asper leaves extract. Topmost abundant compounds in SAM are α-D-glucopyranoside (10.60%), glycerol (7.96%), myo-inositol (4.90%), and butanedioic acid (3.30%). Hexane consists of the higher amount of hexadecanoic acid (18.07%), octadecanoic acid (7.39%), β-sitosterol (4.50%), and α-D-glucopyranoside (4.03%). Determination of Antiemetic, Antimicrobial, Anti-Radical and Cytotoxic Activity of Methanolic Extracts of &amp;lt;i&amp;gt;Centella asiatica&amp;lt;/i&amp;gt;

This study was conducted to investigate the antiemetic, antimicrobial, antioxidant and cytotoxic activity of methanolic extracts of Centella asiatica . The antiemetic assay was carried out by using chick emetic model with minor modifications by calculating the mean decrease in the number of retching. The antimicrobial activity of the crude extract was performed by Disc Diffusion method. The anti-radical activity was determined by the 2,2-diphenyl-2-picrylhydrazyl hydrate (DPPH) method. Brine shrimp lethality bioassay is done to determine cytotoxic activity. The anti-emetic activity of Centella asiatica leaves on young chicks revealed that these extracts have a less anti-emetic effect. The group of chicks treated with Chlorpromazine was found to have 38.4 retches as compared to the 60.4 retches of the control group, thus Chlorpromazine reduced the retches by 36.96%. The chickens treated with leaves extracts inhibited the retches up to 15.67%. The manimum antimicrobial effect was found in this methanol crude extract. The extract did not appear potent in terms of both zones of inhibition and spectrum of activity. In anti-radical activity test, the extract showed moderate free radical scavenging activity with IC50 value 241.71 μg/ml. while compared to that of the reference standard ascorbic acid. Moreover, the methanolic crude extracts also possess moderate cytotoxic principles potential (LC50 value of 39.06 μg/ml) comparing with that of standard vincristine (0.839 μg/ml).

THE N-HEXANE FRACTION OF MYRMECODIA PENDANS INHIBITS CELL SURVIVAL AND PROLIFERATION IN COLON CANCER CELL LINE

Objective: Despite advanced treatment options available for colorectal cancer, many reported resistance and unresponsiveness to conventional chemotherapeutic agents. Therefore, it is urgent to discover a novel drug for colon cancer. Sarang Semut (Myrmecodia pendans), an Indonesian native plant, has been studied extensively due to its anti-cancer profiles. This study aimed to evaluate the anti-tumour activity of Sarang Semut in colon cancer cells.Methods: We evaluated cytotoxic activity of methanol extract as well as n-hexane and ethyl acetate fraction towards colon cancer cell lines (Caco-2 and HCT-116 cells) utilizing 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay. The most potent fraction was evaluated further in inhibiting cell survival using MTT assay and cell proliferation using trypan blue exclusion assay as well as a clonogenic assay.Results: Our data showed that the n-hexane fraction of Sarang Semut induces more cell death than the methanol extract and ethyl acetate fraction. Therefore, we analyzed the n-hexane fraction further and found that the inhibitory concentration 50% (IC50) of the n-hexane fraction was 24 and 30 parts per million (ppm) for Caco-2 and HCT-116 cells, respectively. Moreover, it inhibited cell growth as well as cell colony formation, in particular, shown by the plating efficiency (P&lt;0.05) and colony area per seed (P&lt;0.01) of the control group were different to the treatment group.Conclusion: The n-hexane fraction of Sarang Semut demonstrates a high potential antitumor activity in colon cancer cell line.
