Cortical Metabolic Activity Matches the Pattern of Visual Suppression in Strabismus

When an eye becomes deviated in early childhood, a person does not experience double vision, although the globes are aimed at different targets. The extra image is prevented from reaching perception in subjects with alternating exotropia by suppression of each eye's peripheral temporal retina. To test the impact of visual suppression on neuronal activity in primary (striate) visual cortex, the pattern of cytochrome oxidase (CO) staining was examined in four macaques raised with exotropia by disinserting the medial rectus muscles shortly following birth. No ocular dominance columns were visible in opercular cortex, where the central visual field is represented, indicating that signals coming from the central retina in each eye were perceived. However, the border strips at the edges of ocular dominance columns appeared pale, reflecting a loss of activity in binocular cells from disruption of fusion. In calcarine cortex, where the peripheral visual field is represented, there were alternating pale and dark bands resembling ocular dominance columns. To interpret the CO staining pattern, [(3)H]proline was injected into the right eye in two monkeys. In the right calcarine cortex, the pale CO columns matched the labeled proline columns of the right eye. In the left calcarine cortex, the pale CO columns overlapped the unlabeled columns of the left eye in the autoradiograph. Therefore, metabolic activity was reduced in the ipsilateral eye's ocular dominance columns which serve peripheral temporal retina, in a fashion consistent with the topographic organization of suppression scotomas in humans with exotropia.

The Ferret Auditory Cortex: Descending Projections to the Inferior Colliculus

Descending corticofugal projections are thought to play a critical role in shaping the responses of subcortical neurons. Here, we examine the origins and targets of ferret auditory corticocollicular projections. We show that the ectosylvian gyrus (EG), where the auditory cortex is located, can be subdivided into middle, anterior, and posterior regions according to the pattern of cytochrome oxidase staining and immunoreactivity for the neurofilament antibody SMI32. Injection of retrograde tracers in the inferior colliculus (IC) labeled large layer V pyramidal cells throughout the EG and adjacent sulci. Each region of the EG has a different pattern of descending projections. Neurons in the primary auditory fields in the middle EG project to the lateral nucleus (LN) of the ipsilateral IC and bilaterally to the dorsal cortex and dorsal part of the central nucleus (CN). The projection to these dorsomedial regions of the IC is predominantly ipsilateral and topographically organized. The secondary cortical fields in the posterior EG target the same midbrain areas but exclude the CN of the IC. A smaller projection to the ipsilateral LN also arises from the anterior EG, which is the only region of auditory cortex to target tegmental areas surrounding the IC, including the superior colliculus, periaqueductal gray, intercollicular tegmentum, and cuneiform nucleus. This pattern of corticocollicular connectivity is consistent with regional differences in physiological properties and provides another basis for subdividing ferret auditory cortex into functionally distinct areas.

Experience-dependent regulation of the zincergic innervation of visual cortex in adult monkeys.

Zinc is packaged in, and released from, a subset of glutamatergic synapses in the mammalian telencephalon where it has been shown to act as a potent neuromodulator. In order to establish the functional role for zincergic neurons in visual cortical function and plasticity we have compared the topographic distribution of zincergic terminals in the primary visual cortex (V1) of normal adult vervet monkeys (Cercopithicus aethiops) to that in monkeys monocularly deprived of visual input for short (24 h) or long (3 months) survival times. In normal animals, staining levels for zinc were highest in layers 1-3, 4b, 5 and 6 and lowest in layers 4a and 4c. The laminar and tangential patterns of zinc staining were complementary to staining patterns demonstrated using cytochrome oxidase (CO) histochemistry. Following 3 months of monocular deprivation by enucleation, levels of zinc staining in layers 3, 4calpha and 6a were heterogeneously reduced, clearly revealing the ocular dominance pattern in V1. When compared with the pattern of CO staining, levels of both CO and zinc were reduced in cortical territory innervated by the enucleated eye. Zinc histochemistry also revealed the ocular dominance pattern after only 24 h of monocular impulse blockade induced by enucleation or intravitreal tetrodotoxin infusion. However, by either means of deprivation for 24 h, levels of zinc were increased in deprived-eye stripes relative to nondeprived-eye stripes. These results indicate that zincergic terminals demarcate distinct compartments in the primate visual cortex. Furthermore, levels of synaptic zinc are rapidly and dynamically regulated, suggesting that zinc and/or zincergic neurons participate in mediating activity-dependent changes in the organization of the adult neocortex.

Development of cytochrome oxidase blobs in visual cortex of normal and visually deprived cats.

Cytochrome oxidase (CO) blobs are central to our understanding of the columnar organization and parallel pathways in primate and cat visual cortex. In primates, development of blobs and their relationship with other columnar features of the visual cortex begins pre-natally, before visual experience. In kittens, the supragranular layers differentiate post-natally, after eye opening, raising the possibility that visual experience may influence the development of blobs in cat V1. We have examined the development of blobs in unfolded and flattened sections through the visual cortex of normally reared, dark-reared, monocularly deprived and binocularly deprived kittens. Blobs were found in superficial layers of V1 of normally reared kittens as early as 2 weeks of age, although at this age the overall CO staining in V1 was lighter than in V2. By 6 weeks of age the blobs were adult-like. A patchy pattern of CO staining was also found in V2 of young kittens but not in adults. Visual experience was not necessary for expression of the blobs and monocularly deprived kittens had well developed blobs, indicating that strong Y cell drive is not necessary for the development of blobs in cat V1. CO blobs appear in kitten V1 very early in post-natal development and their expression is independent of visual experience, suggesting that they may be an intrinsic feature of V1 organization.

Differential expression of acetylcholinesterase in the brainstem, ventrobasal thalamus and primary somatosensory cortex of perinatal rats, mice, and hamsters

Acetylcholinesterase (AChE) is transiently expressed by thalamocortical axons in the rat, and staining for this enzyme has been used extensively to study the development of thalamocortical projections. In the present study, patterns of AChE staining were compared in the trigeminal brainstem, thalami and primary somatosensory cortices of perinatal rats, mice, and hamsters. As previously reported, the ventral posteromedial nucleus (VPM) of rats showed dense AChE staining from P-0 at least through P-8. The ventral posterolateral nucleus (VPL) contained heavy AChE staining at least through P-60. In the cortex, there was also dense AChE staining which was organized somatotopically in patches similar to those observed with other methods such as cytochrome oxidase (CO) staining. However, by adulthood, AChE staining revealed a negative image of the CO staining pattern in lamina IV. In the mouse and hamster, there was dense AChE staining in VPL from P-0 through adulthood, but VPM was much less heavily stained for this enzyme. Moreover, the staining in VPL of mice was markedly reduced after transection of axons that travel to the thalamus in the medial lemniscus, suggesting that much of it was contained in these afferent fibers. In the cortices of both perinatal and adult mice and hamsters, AChE staining yielded a negative image of the somatotopically organized patches demonstrable with CO staining. This negative image was apparent by P-2 in the mouse and P-4 in the hamster. These results document a dramatic species difference with respect to the expression of AChE in the thalami and cortices of developing rodents. The differences between the patterns observed in rats vs mice and hamsters probably reflect the fact that cortical AChE in the latter species is not contained in thalamocortical afferents arising from either VPM or VPL.

Organisation and Interrelationships of Functional Maps in Cat and Monkey Striate Cortex

Optical imaging of intrinsic signals allows mapping of the cortical functional architecture in vivo at high spatial resolution. The ability to image activity patterns evoked by many different stimuli in the same piece of cortex can provide information on the spatial relationships between different functional maps. Our findings on the organisation of multiple functional maps in cat and monkey striate cortex are reviewed. The main focus is on the recent finding in cat of two subsystems differing in their response to spatiotemporal aspects of the stimulus. We used grating stimuli of different spatial frequencies in an attempt to verify the existence of spatial frequency columns in cat area 17. Rather than observing a map of continuously changing spatial frequency across the cortical surface we found two distinct sets of domains, one preferring low and one preferring high spatial frequencies. By using different drift velocities we also found that the low-spatial-frequency domains preferred higher speeds than the high-spatial-frequency domains. Comparison of these spatiotemporal frequency domains with the cytochrome oxidase staining pattern revealed that the cytochrome oxidase blobs in cat striate cortex coincide with domains devoted to the processing of the low-spatial-frequency and high-temporal-frequency contents of the visual scene. Together with recent anatomical results these data suggest that spatiotemporal frequency domains are the manifestation of parallel streams in cat visual cortex with distinct patterns of thalamic inputs and extrastriate projections. In the same experiments we also imaged the orientation preference and ocular dominance maps. We investigated the relationships between these three columnar systems, and compared them to an earlier study of orientation, ocular dominance, and blobs in macaque striate cortex. We found systematic relationships between the three systems. While some of these relationships were much weaker than those found in monkey, the organisational principles are similar.

Metabolic energy capacity of dopaminergic grafts and the implanted striatum in parkinsonian nonhuman primates as visualized with cytochrome oxidase histochemistry

Histochemistry for visualization of the mitochondrial enzyme cytochrome oxidase has been used to detect cellular and regional differences in brain energy metabolism. We have examined the pattern of cytochrome oxidase (CO) staining in grafts of embryonic ventral mesencephalic tissue, and in the implanted striatum, of MPTP-treated monkeys as one index of the functional activity of grafted tissue and its influence on the host brain. Four monkeys were selected for study based on interesting variations in dopamine (DA) neuron content of their bilateral grafts as demonstrated with tyrosine-hydroxylase (TH) immunocytochemistry. The results suggest that grafts rich in DA neurons increase the metabolic activity of the implanted striatum of DA-depleted monkeys, and that this improvement of local energy metabolism is greater in the vicinity of grafts containing greater numbers of DA neurons. In addition, the pattern of CO staining within tissue transplants indicates that DA neurons exhibit the highest rate of metabolic activity among all cell types contained in the ventral mesencephalic grafts, and that the transplants receive metabolically active innervation from outside or within the grafted tissue.

Spatio-temporal frequency domains and their relation to cytochrome oxidase staining in cat visual cortex.

Spatial and temporal frequencies are important attributes of the visual scene. It is a long-standing question whether these attributes are represented in a spatially organized way in cat primary visual cortex. Using optical imaging of intrinsic signals, we show here that grating stimuli of different spatial frequencies drifting at various speeds produce distinct activity patterns. Rather than observing a map of continuously changing spatial frequency preference across the cortical surface, we found only two distinct sets of domains, one preferring low spatial frequency and high speed, and the other high spatial frequency and low speed. We compared the arrangement of these spatio-temporal frequency domains with the cytochrome oxidase staining pattern, which, based on work in primate striate cortex, is thought to reflect the partition of the visual cortex into different processing streams. We found that the cytochrome oxidase blobs in cat striate cortex coincide with domains engaged in the processing of low spatial and high temporal frequency contents of the visual scene. Together with other recent results, our data suggest that spatiotemporal frequency domains are a manifestation of parallel streams in cat visual cortex, with distinct patterns of thalamic inputs and extrastriate projections.

The distribution of callosal connections correlates with the pattern of cytochrome oxidase stripes in visual area V2 of macaque monkeys.

In visual area V2 of monkeys, cytochrome oxidase (CO) histochemistry reveals a system of stripe-like subregions where densely labeled thick and thin stripes and pale interstripes can be recognized. Several lines of evidence suggest that CO stripe-like subregions are associated with functional streams in the visual cortex. In the present study, the distribution of retrogradely labeled callosal cells in V2 and the pattern of CO staining were correlated using tangential sections through the flattened cortex. Spectral and coherency analyses of the callosal and CO patterns were performed to assess quantitatively the degree of spatial correlation between these two patterns. The results showed that labeled callosal cells accumulated along the V1/V2 border and in finger-like bands that protruded up to 7-8 mm into V2. These callosal bands were in register with thick and thin CO stripes, with relatively few labeled callosal cells found in interstripe regions. This finding supports the notion that the distribution of callosal connections in the visual cortex is dictated not only by the topography of visual areas, but also by the arrangement of cortical functional streams. Further, these results extend to interhemispheric pathways the notion of functional specificity currently associated mainly with some visual intrahemispheric pathways.

GABAA receptor subunit immunoreactivity in primate visual cortex: distribution in macaques and humans and regulation by visual input in adulthood

Subunit proteins that make up functional GABAA receptors were localized immunocytochemistry in the primary visual cortex (area 17) of adult monkeys and humans. Immunoreactivity for the alpha 1, beta 2/3, and gamma 2 subunits is greatest in layers (II-III, IVA and IVC) of monkey area 17 that contain the highest density of GABA neurons and terminals. Immunostaining for each subunit is unevenly distributed in layers II and III, where patches of immunoreactivity correspond to regions of intense cytochrome oxidase (CO) staining, and in layer IVA, where intense immunoreactivity forms a honeycomb pattern identical to the CO staining pattern. Immunoreactivity for the subunits is localized principally within the neuropil, which, by simultaneous comparison with the distribution of microtubule-associated protein immunostaining, was found to include bundles of thin dendrites and zones of numerous dendritic segments. In addition, gamma 2 immunostaining surrounds the somata of a subpopulation of GABAergic neurons, immunoreactive for the calcium-binding protein parvalbumin. All three subunits are present in the somata and processes of neurons that occupy the white matter subjacent to monkey area 17. In human visual cortex, the alpha 1, beta 2/3, and gamma 2 subunits are distributed in a manner similar to that found in monkeys, with relatively intense immunostaining in layers IVC and IVA. In layer IVC, vertical stripes of intense receptor immunostaining (20-30 microns wide) alternate with wider stripes of pale immunostaining (30-60 microns wide). In the upper and lower halves of IVC beta, these stripes form lattices similar to those in layers IVC and IVA of monkeys. Following monocular deprivation by intravitreal injections of TTX in adult monkeys, immunoreactivity for each subunit in layer IVC consists of alternating intensely and lightly stained stripes. Comparison with the pattern of CO staining indicates that intense immunostaining for alpha 1, beta 2/3, and gamma 2 occurs in normal-eye stripes while abnormally light immunostaining is present in deprived-eye stripes. For all three subunits, immunoreactivity in deprived-eye stripes is reduced within 5 d of monocular deprivation and remains abnormally low for deprivations that extend to at least 30 d. These findings indicate that each of several GABAA receptor subunits adopt similar laminar and compartmental distributions in monkey and human area 17 and are likely to be expressed by the same neurons. The deprivation-dependent reduction in immunoreactivity for alpha 1, beta 2/3, and gamma 2 subunits suggests that all are regulated by visually driven activity.(ABSTRACT TRUNCATED AT 400 WORDS)

Differential effects of peripheral damage on vibrissa-related patterns in trigeminal nucleus principalis, subnucleus interpolaris, and subnucleus caudalis

Histochemistry for cytochrome oxidase reveals a vibrissa-related pattern in trigeminal nucleus principalis, subnucleus interpolaris, and the magnocellular portion of subnucleus caudalis. This pattern is apparent in late fetal animals and is disrupted by transection of the infraorbital nerve on the day of birth. We recently reported results suggesting that the cytochrome oxidase pattern reflects primary afferent-induced clustering of second order neurons in all of these nuclei. If this conclusion is correct, it should follow that primary afferent lesions made after the cytochrome oxidase pattern became established in the brainstem might have little effect upon it. Accordingly, we transected the infraorbital nerve (the trigeminal branch that supplies the vibrissae) on postnatal days 0–10 and evaluated the vibrissa-related pattern in the brainstem with cytochrome oxidase histochemistry at varying intervals after these lesions. If the infraorbital nerve was sectioned on postnatal days 0–2, the vibrissa-related pattern was absent in trigeminal nucleus principalis, and both subnucleus interpolaris and caudalis. If such lesions were made after postnatal day 9, there was no appreciable effect upon the cytochrome oxidase pattern in any portion of the trigeminal brainstem complex. However, if lesions were made between postnatal days 3 and 8, the density and clarity of the cytochrome oxidase staining pattern were reduced in interpolaris and caudalis, but not in principalis. This difference was not due to differential transganglionic degeneration in these nuclei. Tracing with horseradish peroxidase demonstrated qualitatively equivalent primary afferent losses in principalis, interpolaris, and caudalis. Immunocytochemistry with a monoclonal antibody directed against parvalbumin also demonstrated a vibrissa-related pattern of cell bodies in principalis and interpolaris in rats killed on postnatal day 9 or later ages. The combination of retrograde tracing and immunocytochemistry revealed that the parvalbuminimmunoreactive neurons in principalis projected to thalamus while those in interpolaris were not labelled by tracer injections into the thalamus, midbrain, cerebellum or spinal cord. Infraorbital nerve transections made as late as postnatal day 8 resulted in a sharp decrease in the staining of parvalbumin-positive neurons in interpolaris, but not in principalis. Lesions made on postnatal day 10 had no qualitative effect upon parvalbumin-positive neurons in any portion of the trigeminal brainstem complex. The results of this study support the conclusion that the vibrissa-related cytochrome oxidase pattern in principalis becomes independent of primary afferent input at a very short interval after its initial appearance. In contrast, the patterns in more caudal portions of the trigeminal brainstem complex require maintenance of primary afferent input for a much longer postnatal period. The present results also provide further support for the conclusion that postsynaptic elements in the trigeminal brainstem complex may underlie the primary afferent-induced cytochrome oxidase pattern.

Infraorbital nerve blockade from birth does not disrupt central trigeminal pattern formation in the rat

We tested the hypothesis that patterned primary afferent impulse activity during early postnatal periods is necessary for central trigeminal pattern formation. Newborn rats had their whiskers trimmed daily and new slices of slow release polymer containing the sodium channel blocker, tetrodotoxin, were placed under the infraorbital nerve every 8 h for up to 9 days. Electrophysiological recordings indicated that trigeminal ganglion cells were unresponsive to peripheral stimuli and chronically silenced. Trigeminal ganglion cell numbers were unaffected by nerve blockade. Cytochrome oxidase staining patterns in the trigeminal brainstem complex, thalamus, and barrel cortex were normal on postnatal day 1, 3, 5, 7, or 9 ( n = 4 each). Whisker-related patches were of normal sizes and staining densities. Similar negative results were obtained in 9 rats in which whiskers were trimmed daily and the long-acting local anesthetic bupivicaine was injected into the whisker pad at 2.5- to 4-h intervals from birth to sacrifice on postnatal day 5–9. Cytochrome oxidase staining patterns and patch properties again did not differ from normal. Thus, trigeminal pattern formation occurs even when the entire infraorbital nerve is silenced from birth.

Calcium‐binding proteins as markers for subpopulations of GABAergic neurons in monkey striate cortex

Recent studies have shown that the presence of immunoreactivity for parvalbumin (PV-IR) and calbindin-D 28k (Cal-IR) can be used as markers for certain types of gamma-aminobutyric acid (GABA) immunoreactive interneurons in monkey cerebral cortex. Little quantitative information is available regarding the features that distinguish these two subpopulations, however. Therefore, in this study we localized PV-IR and Cal-IR neurons in Macaca monkey striate cortex and analyzed quantitatively their laminar distribution, cell morphology, and co-localization with GABA by double-labeling immunocytochemistry. PV-IR was found in nonpyramidal cells in all layers of the cortex, although PV-IR cells in layer 1 were rare. In contrast, Cal-IR was found mainly in nonpyramidal cells in two bands corresponding to layers 2-3 and 5-6. We found very few double-labeled PV-IR/Cal-IR cells but confirmed that almost all PV-IR and Cal-IR cells are GABAergic. Overall, 74% of GABA neurons in striate cortex displayed PV-IR compared to only 12% that displayed Cal-IR and 14% that were GABA-IR only. Quantitative analysis indicated that the relative proportion of GABA cells that displayed PV-IR or Cal-IR showed conspicuous laminar differences, which were often complementary. Cell size measurements indicated that PV-IR/GABA cells in layers 2-3 and 5-6 were significantly larger than Cal-IR/GABA cells. Analysis of the size, shape, and orientation of stained cell bodies and proximal dendrites further demonstrated that each subpopulation contained several different types of smooth stellate cells, suggesting that Cal-IR and PV-IR are found in functionally and morphologically heterogeneous subpopulations of GABA neurons. There was a thick bundle of PV-IR axons in the white matter underlying the striate but not prestriate cortex. PV-IR punctate labeling matched the cytochrome oxidase staining pattern in layers 4A and 4C, suggesting that PV-IR is present in geniculocortical afferents as well as intrinsic neurons. Cal-IR neuropil staining was high in layers 1, 2, 4B, and 5, where cytochrome oxidase staining is relatively low. We did not find a preferential localization of either PV-IR or Cal-IR cell bodies in any cytochrome oxidase compartments in layers 2-3 of the cortex. These findings indicate that PV and Cal are distributed into different neuronal circuits.

Organization of cytochrome oxidase staining in the visual cortex of nocturnal primates (Galago crassicaudatus and Galago senegalensis): I. Adult Patterns

The distribution and differential staining patterns of cytochrome oxidase (CO) activity in visual cortical areas have provided useful anatomical markers for the modular organization of area 17 (striate cortex) and area 18 in primates. In macaque and squirrel monkeys, previous studies have shown that the majority of cells that lie in areas of high CO activity are color selective, are nonoriented, and project to adjacent zones of high CO activity in area 17 and to stripes of high CO activity in area 18. By contrast, most cells in zones with weak CO activity in area 17 have relatively narrow orientation tuning and are not color selective (Livingstone and Hubel: J. Neurosci. 4:309-356, 2830-2835, '84; 7:3371-3377, '87). The periodic organization of CO activity in area 17, the "blobs," and the stripe-like organization in area 18 thus seem to define visual cortical processing modules and/or channels in primates. We have investigated the organization of CO activity in areas 17 and 18 in two species of nocturnal prosimian primates [Galago crassicaudatus (GCC) and Galago senegalensis (GSS)] in order to evaluate CO staining patterns in primates that have been reported to possess almost exclusively rod retinae and no color vision. In area 17 of both species, our results show that, as in diurnal and nocturnal simian primates, the darkest CO staining occurs in layers III and IV, with clear periodicity in layer III (i.e., CO blobs) and homogeneous staining in layer IV beta, the cortical recipient sublayer of the geniculate parvocellular layers. In GCC, individual blobs in layer III appear to be larger and less frequent than has been reported for the macaque monkey. Unlike simian primates, both galago species exhibit clear CO periodicities within layer IV alpha, the cortical recipient sublayer of the magnocellular geniculate layers. In addition, faint CO periodicities are apparent in layer VI and scattered large darkly CO stained pyramidal cells are visible throughout layer V. Quantitative analysis suggests that CO periodicities are more frequent in GSS than in GCC, suggesting that there may be evolutionary pressure to maintain the same number of CO modules within the smaller striate cortex of the lesser galago, although this is not the trend found across distantly related species. CO activity in area 18 is less well-developed than reported in other primates. In fact, we could not reliably identify discontinuities in CO staining in area 18 of GSS.(ABSTRACT TRUNCATED AT 400 WORDS)

The LectinVicia VillosaLabels a Distinct Subset of GABAergic Cells in Macaque Visual Cortex

The morphology and distribution of neurons labeled specifically by the lectin, Vicia villosa (VVA), were examined in striate cortex of adult macaque monkeys. Following incubation with VVA conjugated to histochemical markers, fine punctate reaction product appears to cover the surface of the soma and proximal dendrites of a population of cortical neurons. Although a small number of VVA-labeled cells are located in layers 2, 3A, 5, and 6, approximately 75% are located in a strip of cortex overlying layers 3B through 4Ca. Layers 1 and 4C beta are virtually devoid of labeled cells. The morphology of labeled cells varies throughout the layers. In the supragranular layers, the labeled cells generally display a round or multipolar soma with a small number of radially disposed dendrites. In deeper layers, labeled cells are multipolar or horizontal, and their proximal dendrites are often more densely labeled. There is no clear correlation between the distribution of labeled cells and the pattern of cytochrome oxidase staining in supragranular layers. Double labeling of single sections for VVA and for GABA (gamma-aminobutyric acid) immunoreactivity revealed that most VVA-labeled cells are also immunoreactive for GABA. The double-labeled cells comprise approximately 30% of all GABA immunoreactive cells. Soma size analysis of double-labeled cells shows that medium-to-large GABA cells in each layer are labeled by VVA. The soma size, laminar distribution, and morphology of the VVA-labeled GABA cells suggest that they include the large basket cells originally observed in Golgi preparations.

Dependence of cytochrome oxidase activity in the rat lateral geniculate nucleus on retinal innervation

Patterns of cytochrome oxidase (CO) activity were examined histochemically in the dorsal lateral geniculate nucleus (LGNd) and retina of pigmented rats. CO staining was not uniform and was distributed in a pattern similar to that of retinal afferents. Portions of the LGNd receiving an exclusively crossed projection were moderately reactive whereas regions receiving an uncrossed or overlapping crossed and uncrossed projection were darkly reactive. The dependence of oxidative metabolic activity in the LGNd on retinal innervation was verified in animals with unilateral enucleation. In adults, chronic monocular enucleation led to a decrease in CO staining in portions of the LGNd deprived of retinal input; in animals enucleated at birth, normal patterns of CO reactivity failed to develop and both LGNds had a more uniform pattern of moderate CO staining. Most neurons in the ganglion cell layer of the retina were moderately reactive for CO. However, there were approximately 3,000 darkly reactive cells, most of which appear to be ganglion cells. The darkly reactive cells were more numerous in the peripheral temporal retina. The laminar pattern of CO staining in the retina was similar to that described previously for carnivores and primates. The most reactive laminae were the inner and outer plexiform layers and the photoreceptor inner segments. Within the inner plexiform layer, sublamina a was more darkly stained than sublamina b. These results suggest that the physiological properties of crossed and uncrossed visual pathways in rats are functionally dissimilar at the level of both the retina and the LGNd.

Development of cytochrome oxidase staining in the retina and lateral geniculate nucleus: a possible correlate of ON- and OFF-center channel maturation

A histochemical stain for cytochrome oxidase (CO) activity was used to examine the maturation of a neurochemical correlate of ON and OFF channels in the retina and dorsal lateral geniculate nucleus (LGN) of the tree shrew. In the adult tree shrew, the CO staining pattern can be used as a histochemical marker of segregated ON- and OFF-center channels in the retina, LGN, and striate cortex. Our previous studies have shown that the retina is immature and the LGN unlaminated at birth. In the present study, we show that the laminar development of CO reactivity emerges during the first postnatal week in the LGN, while the maturation of CO staining in the presumed ON and OFF sublaminae of the retinal inner plexiform layer develops slowly, well after the appearance of differential laminar CO staining in the LGN.

Cytochrome oxidase activity of postsurgical bullous keratopathy endothelium

Proper corneal hydration is normally maintained by a pump-leak mechanism located on the lateral cell membranes of the endothelium (1,2). When endothelial cells become sufficiently dysfunctional in response to trauma, the cornea becomes irreversibly edematous and opaque. Previous studies in our laboratory have shown that mitochondrial cytochrome oxidase (CO) activity, an enzyme important in respiratory activities, is correlated with endothelial cell functional activity in corneas with Fuchs' endothelial dystrophy. In this study we investigated cytochrome oxidase activity in corneas with postsurgical bullous keratopathy. Corneas with aphakic and pseudophakic bullous keratopathy (ABK, PBK) were incubated in diaminobenzidine-cytochrome C media. Results showed that the staining pattern of ABK and PBK corneas was uniformly low compared to keratoconus corneas, and in contrast to previous studies on Fuchs' dystrophy corneas which demonstrated a regional staining pattern. This suggests that CO staining patterns correlate with functional activity and may be a useful technique to detect dysfunctional cells in various disease categories.

The organization of the neonatal rat's brainstem trigeminal complex and its role in the formation of central trigeminal patterns

The present study delimits the relationship of primary trigeminal afferents to their targets, the brainstem trigeminal nuclei of the neonatal rat. Previously, the brainstem trigeminal complex of the rat has been subdivided on the basis of either cytoarchitectonics or patterns of succinic dehydrogenase activity into the principal sensory nucleus and the three subnuclei of the spinal trigeminal nucleus, oralis, interpolaris, and caudalis. In this paper, we demonstrate that each of these subdivisions can also be identified by its pattern of primary trigeminal afferents. In addition, we demonstrate that the terminations of these afferents are distributed in a punctate fashion which correlates with vibrissae-related patterns of histochemical staining. Further, vibrissae removal in the neonatal rat at any age studied results in a corresponding deafferentation of both the principal sensory nucleus and all subnuclei of the spinal trigeminal nucleus. This same procedure has a graded, age-dependent effect on the vibrissae-related pattern of cytochrome oxidase staining in somatosensory cortex. On this basis, we conclude that vibrissae-related pattern formation in the central trigeminal system can be best understood in terms of a single "sensitive" period for the entire system. We hypothesize that this is the period during which an interaction normally occurs between primary trigeminal afferents and target neurons of the principal sensory nucleus.
