Exploring factors that affect Microcystis abundance in the sacramento san joaquin delta

Cyanobacteria harmful algal blooms (cHABs) are increasing in frequency, intensity and duration in estuaries worldwide. In the upper San Francisco Estuary, also known as the Sacramento San Joaquin Delta (Delta), cHABs have been a topic of concern over the past two decades. In response, managers are urgently working to understand the factors that drive cHABs and identify feasible management options to avert ecological and human health consequences. We used a six year data set to explore relationships between flow parameters, temperature, and Microcystis biovolume to determine the potential for managing large scale hydrodynamic conditions to address Delta cHABs. We also looked at the relationship between Microcystis biovolume and the low salinity zone to see if it could be used as a proxy for residence time, because residence time is positively related to cyanobacteria abundance. We found the low salinity zone is not a useful proxy for residence time in the area of the Delta that experiences the most severe cHABs. Our finding suggest that climatic conditions (i.e., temperature and water year type) have the greatest influence on Microcystis biovolume in the Delta, with higher biovolume during years with lower flow and higher temperatures. Further, there are interannual differences in Microcystis biovolume that cannot be fully explained by flow parameters or temperature, meaning other factors not included in our model may be involved. We conclude that management actions to increase flow may be ineffective at reducing Microcystis to desired levels if water temperatures remain high.

Environmentally Friendly and Cost-Effective Approaches to Reduce Toxin Content in Toxic Cyanobacterial Biomasses

Cyanobacterial outgrowths are naturally occurring processes in eutrophic aquatic ecosystems. Furthermore, as a result of climate change and anthropogenic pollution, cyanobacteria harmful algal blooms (CyanoHABs) are expanding worldwide. CyanoHABs are considered a threat to human health and environment due to the production of potent toxic substances, but at the same time, valuable products can be obtained from these microorganisms. The main objective of this study was to test straightforward and cost-effective methods to reduce the toxin content of cyanobacterial biomass for the exploitation of this important biological resource. To carry out this study, lyophilized or hydrated biomass from microcystin-LR (MC-LR) producing Microcystis aeruginosa and cylindrospermopsin (CYN) producing Chrysosporum ovalisporum strains were subjected to the following treatments: (1) thermal (50 °C); (2) ultraviolet (UV) radiation; (3) ozone; and (4) sunlight, for periods varying between 2 and 12 h. MC-LR and CYN concentrations were quantified by LC-MS and compared between experimental groups. The results show a significant reduction in the amount of MC-LR in M. aeruginosa biomass (lyophilized and hydrated) exposed to sunlight. Since no other treatment reduced MC-LR in M. aeruginosa biomass, this molecule was demonstrated to be very stable. Regarding CYN, the concentration of this toxin in C. ovalisporum biomass was significantly reduced with the exposure to UV radiation, to approximately 51% of the initial concentration after 2 h of exposure; 86% reduction after 5 h of exposure; and 77% reduction after 12 h of exposure. Overall, this study demonstrates that the toxicity of cyanobacterial biomass can be reduced by employing environmentally friendly and cost-effective treatments with sunlight and UV radiation.

The Role of Heterocysts in Cyanotoxin Production during Nitrogen Limitation.

Cyanobacteria harmful algal blooms (cyanoHABs) can have impacts on human health, aquatic ecosystems, and the economy. Nutrient management is an important mitigation and even remediation strategy. In this work, the paralytic shellfish toxin (PST)-producing Aphanizomenon (Aphan.) flos-aquae (Linnaeus) Ralfs ex Bornet & Flahault (now identified as Aphan. sp.) single filament isolate NH-5 was grown in P-depleted media, N-depleted media, and complete BG-11 media. Growth and heterocyst and vegetative cells were monitored using dry weight and cell counts. Ultrasonication was used to separate heterocysts from vegetative cells. HPLC-FLD with post-column derivatization was used to determine the saxitoxin (STX) and neosaxitoxin (NEOSTX) concentration per cell. Aphan. sp. NH-5 biomass was lower in the P-depleted media than in the N-depleted media and the control, though higher heterocyst counts were detected in the N-depleted media. The heterocyst toxin concentration was significantly higher compared to the vegetative cells for the N-depleted media, control, and P-depleted media. However, no significant differences were found among all preparations with regard to the STX-to-NEOSTX ratio. We conclude that N limitation induced higher heterocyst numbers and that N fixation activity is a factor behind the increase in the STX and NEOSTX production of Aphan. sp. NH-5.

Leveraging multimission satellite data for spatiotemporally coherent cyanoHAB monitoring

Cyanobacteria harmful algal blooms (cyanoHABs) present a critical public health challenge for aquatic resource and public health managers. Satellite remote sensing is well-positioned to aid in the identification and mapping of cyanoHABs and their dynamics, giving freshwater resource managers a tool for both rapid and long-term protection of public health. Monitoring cyanoHABs in lakes and reservoirs with remote sensing requires robust processing techniques for generating accurate and consistent products across local and global scales at high revisit rates. We leveraged the high spatial and temporal resolution chlorophyll-a (Chla) and phycocyanin (PC) maps from two multispectral satellite sensors, the Sentinel-2 (S2) MultiSpectral Instrument (MSI) and the Sentinel-3 (S3) Ocean Land Colour Instrument (OLCI) respectively, to study bloom dynamics in Utah Lake, United States, for 2018. We used established Mixture Density Networks (MDNs) to map Chla from MSI and train new MDNs for PC retrieval from OLCI, using the same architecture and training dataset previously proven for PC retrieval from hyperspectral imagery. Our assessment suggests lower median uncertainties and biases (i.e., 42% and -4%, respectively) than that of existing top-performing PC algorithms. Additionally, we compared bloom trends in MDN-based PC and Chla products to those from a satellite-derived cyanobacteria cell density estimator, the cyanobacteria index (CI-cyano), to evaluate their utility in the context of public health risk management. Our comprehensive analyses indicate increased spatiotemporal coherence of bloom magnitude, frequency, occurrence, and extent of MDN-based maps compared to CI-cyano and potential for use in cyanoHAB monitoring for public health and aquatic resource managers.

A Review of Cyanophage-Host Relationships: Highlighting Cyanophages as a Potential Cyanobacteria Control Strategy.

Harmful algal blooms (HABs) are naturally occurring phenomena, and cyanobacteria are the most commonly occurring HABs in freshwater systems. Cyanobacteria HABs (cyanoHABs) negatively affect ecosystems and drinking water resources through the production of potent toxins. Furthermore, the frequency, duration, and distribution of cyanoHABs are increasing, and conditions that favor cyanobacteria growth are predicted to increase in the coming years. Current methods for mitigating cyanoHABs are generally short-lived and resource-intensive, and have negative impacts on non-target species. Cyanophages (viruses that specifically target cyanobacteria) have the potential to provide a highly specific control strategy with minimal impacts on non-target species and propagation in the environment. A detailed review (primarily up to 2020) of cyanophage lifecycle, diversity, and factors influencing infectivity is provided in this paper, along with a discussion of cyanophage and host cyanobacteria relationships for seven prominent cyanoHAB-forming genera in North America, including: Synechococcus, Microcystis, Dolichospermum, Aphanizomenon, Cylindrospermopsis, Planktothrix, and Lyngbya. Lastly, factors affecting the potential application of cyanophages as a cyanoHAB control strategy are discussed, including efficacy considerations, optimization, and scalability for large-scale applications.

A validation of satellite derived cyanobacteria detections with state reported events and recreation advisories across U.S. lakes

Cyanobacteria harmful algal blooms (cyanoHABs) negatively affect ecological, human, and animal health. Traditional methods of validating satellite algorithms with data from water samples are often inhibited by the expense of quantifying cyanobacteria indicators in the field and the lack of public data. However, state recreation advisories and other recorded events of cyanoHAB occurrence reported by local authorities can serve as an independent and publicly available dataset for validation. State recreation advisories were defined as a period delimited by a start and end date where a warning was issued due to detections of cyanoHABs over a state's risk threshold. State reported events were defined as any event that was documented with a single date related to cyanoHABs. This study examined the presence-absence agreement between 160 state reported cyanoHAB advisories and 1,343 events and cyanobacteria biomass estimated by a satellite algorithm called the Cyanobacteria Index (CIcyano). The true positive rate of agreement with state recreation advisories was 69% and 60% with state reported events. CIcyano detected a reduction or absence in cyanobacteria after 76% of the recreation advisories ended. CIcyano was used to quantify the magnitude, spatial extent, and temporal frequency of cyanoHABs; each of these three metrics were greater (r > 0.2) during state recreation advisories compared to non-advisory times with effect sizes ranging from small to large. This is the first study to quantitatively evaluate satellite algorithm performance for detecting cyanoHABs with state reported events and advisories and supports informed management decisions with satellite technologies that complement traditional field observations.

Inactivation Mechanism of Algal Chlorophyll by Allelochemical Quercetin.

Cyanobacteria harmful algal blooms (CyanoHABs) are a global concern. Application of allelochemicals is a promising solution for cyanobacteria control, due to its high efficiency, low cost and ecological safety. Flavonoids (natural polyphenols produced by aquatic plants) are reported capable of effectively inhibiting the growth of algae; however, the molecular mechanism of algae chlorophyll inactivation is still unclear. In this study, quercetin was used as a typical flavonoid, to investigate the inactivation effect of allelochemical on Microcystis aeruginosa chlorophyll a. The absorption and fluorescence spectra showed that chlorophyll reacted with quercetin to form pheophytin, and the formation rate of pheophytin increased with increasing quercetin concentration (1 × 10-5-1 × 10-2M). FTIR spectra and DFT calculation showed that Mg2+ complexed with the 3-OH and 4-C = O groups in the quercetin ring C so that chlorophyll was inactivated due to the loss of Mg2+ ions. Overall, this study revealed that quercetin inactivated chlorophyll a of cyanobacteria by capturing Mg2+ ions, providing insights into the molecular mechanisms of algal bloom control by allelochemicals.

Satellite-derived cyanobacteria frequency and magnitude in headwaters & near-dam reservoir surface waters of the Southern U.S.

Reservoirs are dominant features of the modern hydrologic landscape and provide vital services. However, the unique morphology of reservoirs can create suitable conditions for excessive algae growth and associated cyanobacteria blooms in shallow in-flow reservoir locations by providing warm water environments with relatively high nutrient inputs, deposition, and nutrient storage. Cyanobacteria harmful algal blooms (cyanoHAB) are costly water management issues and bloom recurrence is associated with economic costs and negative impacts to human, animal, and environmental health. As cyanoHAB occurrence varies substantially within different regions of a water body, understanding in-lake cyanoHAB spatial dynamics is essential to guide reservoir monitoring and mitigate potential public exposure to cyanotoxins. Cloud-based computational processing power and high temporal frequency of satellites enables advanced pixel-based spatial analysis of cyanoHAB frequency and quantitative assessment of reservoir headwater in-flows compared to near-dam surface waters of individual reservoirs. Additionally, extensive spatial coverage of satellite imagery allows for evaluation of spatial trends across many dozens of reservoir sites. Surface water cyanobacteria concentrations for sixty reservoirs in the southern U.S. were estimated using 300 m resolution European Space Agency (ESA) Ocean and Land Colour Instrument (OLCI) satellite sensor for a five year period (May 2016–April 2021). Of the reservoirs studied, spatial analysis of OLCI data revealed 98% had more frequent cyanoHAB occurrence above the concentration of >100,000 cells/mL in headwaters compared to near-dam surface waters (P < 0.001). Headwaters exhibited greater seasonal variability with more frequent and higher magnitude cyanoHABs occurring mid-summer to fall. Examination of reservoirs identified extremely high concentration cyanobacteria events (>1,000,000 cells/mL) occurring in 70% of headwater locations while only 30% of near-dam locations exceeded this threshold. Wilcoxon signed-rank tests of cyanoHAB magnitudes using paired-observations (dates with observations in both a reservoir's headwater and near-dam locations) confirmed significantly higher concentrations in headwater versus near-dam locations (p < 0.001).

Effect of Zinc on Microcystis aeruginosa UTEX LB 2385 and Its Toxin Production

Cyanobacteria harmful algal blooms (CHABs) are primarily caused by man-made eutrophication and increasing climate-change conditions. The presence of heavy metal runoff in affected water systems may result in CHABs alteration to their ecological interactions. Certain CHABs produce by-products, such as microcystin (MC) cyanotoxins, that have detrimentally affected humans through contact via recreation activities within implicated water bodies, directly drinking contaminated water, ingesting biomagnified cyanotoxins in seafood, and/or contact through miscellaneous water treatment. Metallothionein (MT) is a small, metal-sequestration cysteine rich protein often upregulated within the stress response mechanism. This study focused on zinc metal resistance and stress response in a toxigenic cyanobacterium, Microcystis aeruginosa UTEX LB 2385, by monitoring cells with (0, 0.1, 0.25, and 0.5 mg/L) ZnCl2 treatment. Flow cytometry and phase contrast microscopy were used to evaluate physiological responses in cultures. Molecular assays and an immunosorbent assay were used to characterize the expression of MT and MC under zinc stress. The results showed that the half maximal inhibitory concentration (IC50) was 0.25 mg/L ZnCl2. Flow cytometry and phase contrast microscopy showed morphological changes occurred in cultures exposed to 0.25 and 0.5 mg/L ZnCl2. Quantitative PCR (qPCR) analysis of selected cDNA samples showed significant upregulation of Mmt through all time points, significant upregulation of mcyC at a later time point. ELISA MC-LR analysis showed extracellular MC-LR (µg/L) and intracellular MC-LR (µg/cell) quota measurements persisted through 15 days, although 0.25 mg/L ZnCl2 treatment produced half the normal cell biomass and 0.5 mg/L treatment largely inhibited growth. The 0.25 and 0.5 mg/L ZnCl2 treated cells demonstrated a ~40% and 33% increase of extracellular MC-LR(µg/L) equivalents, respectively, as early as Day 5 compared to control cells. The 0.5 mg/L ZnCl2 treated cells showed higher total MC-LR (µg/cell) quota yield by Day 8 than both 0 mg/L ZnCl2 control cells and 0.1 mg/L ZnCl2 treated cells, indicating release of MCs upon cell lysis. This study showed this Microcystis aeruginosa strain is able to survive in 0.25 mg/L ZnCl2 concentration. Certain morphological zinc stress responses and the upregulation of mt and mcy genes, as well as periodical increased extracellular MC-LR concentration with ZnCl2 treatment were observed.

Impact of extreme wet and dry years on the persistence of Microcystis harmful algal blooms in San Francisco Estuary

Cyanobacteria harmful algal blooms (CHABs) became a concern in the upper San Francisco Estuary, California beginning in 1999, when yearly blooms of Microcystis began in the Delta region. Subsequent research identified that the increase in the magnitude, duration and toxicity of Microcystis blooms was associated with drought related conditions of elevated water temperature and low streamflow. However, the impact of extreme conditions on the resilience of the bloom was unknown. The 2014 and 2017 water years provided a unique opportunity to determine the effect of climatic “whiplash” produced by the occurrence of extreme wet conditions following extreme dry conditions on the Microcystis bloom. We hypothesized that the period of record wet conditions in 2017 (1906-2018) would eliminate the Microcystis bloom for that year and perhaps revert the estuary phytoplankton community back to pre-bloom conditions due to extreme flushing, despite the increase in magnitude and spatial and temporal distribution of the Microcystis bloom during the 2014 extreme dry year. Field sampling was conducted at 2-week or 4-week intervals between July and November at stations throughout the Delta for both years and included a suite of physical, chemical and biological factors. Using PRIMER-e DISTLM, we determined that retention time in the upper estuary and water temperature were key environmental correlates with the Microcystis bloom amplitude and in regression models described 58-78% of the variation of the bloom surface biovolume or subsurface abundance. The period of record high streamflow in 2017 was not enough to eliminate the Microcystis bloom. However, the bloom was small in 2017, with a low abundance, late initiation, short duration, narrow distribution and low toxin production. Warm water temperature enabled the bloom to flower in late summer despite streamflow many times those measured previously. In addition, although conditions early in the summer of 2017 favored diatoms, the summer was characterized by an abundance of other non-Microcystis cyanobacteria. We conclude that once established, Microcystis is likely to be resistant to extreme wet conditions, as long as water temperature and other key water quality conditions are favorable.

Spatio-temporal variation of cyanobacteria and cyanotoxins in public supply reservoirs of the semi-arid region of Brazil

Cyanobacteria harmful algal blooms (CyanoHABs) have become increasingly frequent and intense in public supply reservoirs as a result of eutrophication and global climate change. The semi-arid region of Brazil has a well documented history of CyanoHABs but the underlying factors that control the excessive proliferation of these organisms and the production of their bioactive secondary metabolites are not comprehensively understood. This study aimed to identify the environmental factors that explain the spatial and temporal variations in the abundance of cyanobacteria and the concentration of cyanotoxins (microcystins, saxitoxins, and cylindrospermopsin) in semi-arid reservoirs. The following hypotheses were tested: (a) the largest biovolumes of potential toxin producing cyanobacteria occur when cyanotoxin concentrations are highest; and (b) the environmental factors that explain variations in biovolume of cyanobacteria also explain changes in cyanotoxins concentrations. Samples were taken from four reservoirs located in the Northeast region of Brazil, over a three-month period (October 2016 and February and June 2017). Of the 24 species of cyanobacteria identified, 13 were potentially toxin-producing. Physicochemical variables such as water volume of the reservoir, water transparency, soluble reactive phosphorus, and total phosphorus explained the abundance of cyanobacteria and the levels of cyanotoxins. These results corroborate the hypothesis that similar physicochemical conditions influence the abundance and diversity of cyanobacteria and cyanotoxins. Cyanobacterial blooms composed of more than one potential toxin producing species were observed in the studied reservoirs, where potential microcystin-producing species were the most common. Microcystins and saxitoxins were detected in all the reservoirs studied, while cylindrospermopsin and the cyanobacterium Cylindrospermopsis raciborskii were simultaneously recorded in only one reservoir (Camalaú Reservoir). Cylindrospermopsin was only detected in a reservoir for the first time in the State of Paraíba. Canonical redundancy analysis showed that the cyanotoxins were related to potential toxin producing species. These results corroborate the proposed hypothesis that there is a correlation between cyanotoxins and the biomass of potential producers. Also, there were situations where cyanotoxins were detected without the presence of potential producers. These results demonstrate the need for reassessment of potential toxin producing species of cyanobacteria in semi-arid reservoirs. This may lead to the identification and characterization of novel producers of these bioactive secondary metabolites.

Synthesis and Activity of 1,2,3-Triazole Aminopyrimidines against Cyanobacteria as PDHc-E1 Competitive Inhibitors.

Cyanobacteria harmful algal blooms are of global concern, but all currently available algicides in the market are nonselective and have potential side effects on nontarget species. In the present work, two series of compounds (4 and 6) comprising 16 novel 1,2,3-triazole aminopyrimidines were rationally designed and synthesized as control agent for cyanobacteria. Our design focus was the inhibiting cyanobacteria by inhibition against pyruvate dehydrogenase complex E1 (PDHc-E1). Compounds 4 and 6 showed potent inhibition against Escherichia coli PDHc-E1 (IC50 = 4.13-23.76 μM) and also strong algicidal activities against Synechocystis sp. PCC 6803 (EC50 = 1.7-8.1 μM) and Microcystis sp. FACHB905 (EC50 = 2.1-11.8 μM). In particular, the algicidal activities of 6d against four algal species were not only higher than that of prometryn; they were also comparable to or higher than that of copper sulfate. The analogues 4c, 4d, 6d, and 6e displayed potent algicidal activities and inhibition of E. coli PDHc-E1 but exhibited negligible inhibition of porcine PDHc-E1. As revealed by molecular docking, site-directed mutagenesis, enzymatic assays, and an inhibition kinetic analysis, 4c and 6d inhibited PDHc-E1 in a competitive manner. Our results suggest that highly selective, effective algicides can be developed by rationally designing competitive PDHc-E1 inhibitors.

A Comprehensive Review: Development of Electrochemical Biosensors for Detection of Cyanotoxins in Freshwater.

Cyanobacteria harmful algal blooms are increasing in frequency and cyanotoxins have become an environmental and public concern in the U.S. and worldwide. In this Review, the majority of reported studies and developments of electrochemical affinity biosensors for cyanotoxins are critically reviewed and discussed. Essential background information about cyanobacterial toxins and electrochemical biosensors is combined with the rapidly moving development of electrochemical biosensors for these toxins. Current issues and future challenges for the development of useful electrochemical biosensors for cyanotoxin detection that meet the demands for applications in field freshwater samples are discussed. The major aspects of the entire review article in a prescribed sequence include (i) the state-of-the-art knowledge of the toxicity of cyanotoxins, (ii) important harmful algal bloom events, (iii) advisories, guidelines, and regulations, (iv) conventional analytical methods for determination of cyanotoxins, (v) electrochemical transduction, (vi) recognition receptors, (vii) reported electrochemical biosensors for cyanotoxins, (viii) summary of analytical performance, and (ix) recent advances and future trends. Discussion includes electrochemical techniques and devices, biomolecules with high affinity, numerous array designs, various detection approaches, and research strategies in tailoring the properties of the transducer-biomolecule interface. Scientific and engineering aspects are presented in depth. This review aims to serve as a valuable source to scientists and engineers entering the interdisciplinary field of electrochemical biosensors for detection of cyanotoxins in freshwaters.

Impacts of the 2014 severe drought on the Microcystis bloom in San Francisco Estuary

The increased frequency and intensity of drought with climate change may cause an increase in the magnitude and toxicity of freshwater cyanobacteria harmful algal blooms (CHABs), including Microcystis blooms, in San Francisco Estuary, California. As the fourth driest year on record in San Francisco Estuary, the 2014 drought provided an opportunity to directly test the impact of severe drought on cyanobacteria blooms in SFE. A field sampling program was conducted between July and December 2014 to sample a suite of physical, chemical, and biological variables at 10 stations in the freshwater and brackish reaches of the estuary. The 2014 Microcystis bloom had the highest biomass and toxin concentration, earliest initiation, and the longest duration, since the blooms began in 1999. Median chlorophyll a concentration increased by 9 and 12 times over previous dry and wet years, respectively. Total microcystin concentration also exceeded that in previous dry and wet years by a factor of 11 and 65, respectively. Cell abundance determined by quantitative PCR indicated the bloom contained multiple potentially toxic cyanobacteria species, toxic Microcystis and relatively high total cyanobacteria abundance. The bloom was associated with extreme nutrient concentrations, including a 20-year high in soluble reactive phosphorus concentration and low to below detection levels of ammonium. Stable isotope analysis suggested the bloom varied with both inorganic and organic nutrient concentration, and used ammonium as the primary nitrogen source. Water temperature was a primary controlling factor for the bloom and was positively correlated with the increase in both total and toxic Microcystis abundance. In addition, the early initiation and persistence of warm water temperature coincided with the increased intensity and duration of the Microcystis bloom from the usual 3 to 4 months to 8 months. Long residence time was also a primary factor controlling the magnitude and persistence of the bloom, and was created by a 66% to 85% reduction in both the water inflow and diversion of water for agriculture during the summer. We concluded that severe drought conditions can lead to a significant increase in the abundance of Microcystis and other cyanobacteria, as well as their associated toxins.

Associations between chlorophyll a and various microcystin health advisory concentrations.

Cyanobacteria harmful algal blooms (cHABs) are associated with a wide range of adverse health effects that stem mostly from the presence of cyanotoxins. To help protect against these impacts, several health advisory levels have been set for some toxins. In particular, one of the more common toxins, microcystin, has several advisory levels set for drinking water and recreational use. However, compared to other water quality measures, field measurements of microcystin are not commonly available due to cost and advanced understanding required to interpret results. Addressing these issues will take time and resources. Thus, there is utility in finding indicators of microcystin that are already widely available, can be estimated quickly and in situ, and used as a first defense against high levels of microcystin. Chlorophyll a is commonly measured, can be estimated in situ, and has been shown to be positively associated with microcystin. In this paper, we use this association to provide estimates of chlorophyll a concentrations that are indicative of a higher probability of exceeding select health advisory concentrations for microcystin. Using the 2007 National Lakes Assessment and a conditional probability approach, we identify chlorophyll a concentrations that are more likely than not to be associated with an exceedance of a microcystin health advisory level. We look at the recent US EPA health advisories for drinking water as well as the World Health Organization levels for drinking water and recreational use and identify a range of chlorophyll a thresholds. A 50% chance of exceeding one of the specific advisory microcystin concentrations of 0.3, 1, 1.6, and 2 μg/L is associated with chlorophyll aconcentration thresholds of 23, 68, 84, and 104 μg/L, respectively. When managing for these various microcystin levels, exceeding these reported chlorophyll aconcentrations should be a trigger for further testing and possible management action.

Cyanobacteria harmful algal blooms (cHABs) are associated with a wide range of adverse health effects that stem mostly from the presence of cyanotoxins. To help protect against these impacts, several health advisory levels have been set for some toxins. In particular, one of the more common toxins, microcystin, has several advisory levels set for drinking water and recreational use. However, compared to other water quality measures, field measurements of microcystin are not commonly available due to cost and advanced understanding required to interpret results. Addressing these issues will take time and resources. Thus, there is utility in finding indicators of microcystin that are already widely available, can be estimated quickly and in situ, and used as a first defense against high levels of microcystin. Chlorophyll a is commonly measured, can be estimated in situ, and has been shown to be positively associated with microcystin. In this paper, we use this association to provide estimates of chlorophyll a concentrations that are indicative of a higher probability of exceeding select health advisory concentrations for microcystin. Using the 2007 National Lakes Assessment and a conditional probability approach, we identify chlorophyll a concentrations that are more likely than not to be associated with an exceedance of a microcystin health advisory level. We look at the recent US EPA health advisories for drinking water as well as the World Health Organization levels for drinking water and recreational use and identify a range of chlorophyll a thresholds. A 50% chance of exceeding one of the specific advisory microcystin concentrations of 0.3, 1, 1.6, and 2 μg/L is associated with chlorophyll a concentration thresholds of 23, 68, 84, and 104 μg/L, respectively. When managing for these various microcystin levels, exceeding these reported chlorophyll a concentrations should be a trigger for further testing and possible management action.

Anthropogenic and climate-induced change favors toxic cyanobacteria blooms: Evidence from monitoring a highly eutrophic, urban Mediterranean lake

Cyanobacteria Harmful Algal Blooms (or CyanoHABs) represent one of the most conspicuous waterborne microbial hazards in aquatic environments mostly due to the production of harmful secondary metabolites, known as cyanotoxins. In this paper we assessed cyanobacteria blooms in a shallow, Mediterranean, highly eutrophic lake (Lake Pamvotis, NW Greece) in relation to critical environmental parameters using a multi-approach methodology. Microscopic (identification of cyanobacteria), immunological (ELISA), and molecular techniques (PCR) combined with physico-chemical parameters were used to monitor cyanobacteria blooms and the associated cyanotoxin production for 14 months on a monthly basis. Cyanobacteria were the main phytoplankton component, representing more than 75% of the total phytoplankton abundance throughout the study period; dominant species belonged to Microcystis, Anabaena and Aphanizomenon. Microcystins (MCs) were detected throughout the year in all sampling stations in concentrations ranging from 0.16 to 27μgL−1, indicating that toxic Anabaena and Microcystis strains are persistent and dominant. Saxitoxins (STXs) found in two samples (concentrations 1.3 and 2.1μgL−1) in the warm period, are reported here for the first time. The total MC concentration was positively correlated with temperature, nitrate nitrogen, ammonia nitrogen, and soluble reactive phosphorus, suggesting that high-temperatures and eutrophic conditions promote the growth of MC-producing genotypes. We also investigated a Microcystis strain specific growth rate, which reached its maximum at 30°C. Our data, combined with long-term data comparison of key limnological features, spanning a 25-year period, suggest that water temperature and nutrient loads may act synergistically to promote cyanobacterial dominance and persistence in Lake Pamvotis.

Cyanotoxin occurrence and potentially toxin producing cyanobacteria in freshwaters of Greece: A multi-disciplinary approach

Cyanobacteria harmful algal blooms (or CyanoHABs) represent one of the most conspicuous waterborne microbial hazards in aquatic environments mostly due to the production of harmful secondary metabolites, known as cyanotoxins. In freshwaters of Greece only the presence of microcystins (MCs) has been reported despite the increasing occurrence of species able to produce other cyanotoxins too. In this paper, we studied the occurrence of potentially toxic cyanobacteria in water samples collected from six lakes and reservoirs in Greece. A multi-technique approach was applied by the use of microscopy, molecular, and immunological methods. Cyanobacteria were found in all the sites ranging from 4.7 × 103 to 5.3 × 108 individuals L−1, representing >70% of the total phytoplankton abundance. Microcystins (MCs), cylindrospermopsins (CYNs), and saxitoxins (STXs) were detected using ELISA, in concentrations ranging from 3.9 to 108 μg L−1, from 0.3 to 2.8 μg L−1 and from 0.4 to 1.2 μg L−1, respectively. In half of the samples examined more than one cyanotoxins were detected. Our results document the first report on the occurrence of CYN and STX in freshwaters of Greece and show that potential STX producers are Cylindrospermopsis raciborskii and Aphanizomenon flos-aquae. Further studies are needed to assess potential CYN producers. This study provides further data on the distribution and toxicity of C. raciborskii and Aph. flos-aquae and documents a C. raciborskii dominated bloom producing STX in Europe.
