Determination of Biological Activity and Biochemical Content of Ethanol Extract from Fruiting Body of Tricholoma bufonium (Pers.) Gillet.

The current study investigates the biochemical composition and biological activities of ethanol extract from the fruit body of Tricholoma bufonium, marking the first detailed examination of this species. The primary goal was to assess the antimicrobial, anti-biofilm, and antioxidant properties of ethanol extract from the fruit body of T. bufonium against a range of bacterial strains. Conventional microbiological and biochemical techniques were employed to assess the antimicrobial efficacy of the extract and to determine its minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. Furthermore, a GC-MS analysis identified bioactive compounds, such as palmitic acid and oleic acid, which are likely contributors to the observed antimicrobial activity. The anti-biofilm activity was tested using glucose monohydrate-modified environments for biofilm formation, while the antioxidant potential was measured using the DPPH radical scavenging assay, CUPRAC (cupric ion reducing antioxidant capacity) assay, and FRAP (ferric ion reducing antioxidant power) assay. The ethanol extract exhibited potent antimicrobial activity, particularly against Enterococcus faecium, Bacillus subtilis, and Staphylococcus aureus MRSA, with MIC values as low as 0.0338 mg/mL for several pathogens. Additionally, the extract exhibited significant anti-biofilm activity against Bacillus cereus and antioxidant activity with an EC50 value of 11.745 mg/mL. These results suggest that ethanol extract from the fruit body of T. bufonium may be a potent candidate for developing novel antimicrobial agents, particularly against resistant strains such as MRSA, while also providing antioxidant benefits.

Antioxidative and Antimicrobial Activity of 2-Amino Substituted Halochalcone N-Glycoside Derivative Compounds

Introduction: The aim of this study was to specify the antioxidant and antimicrobial activity synthesized 2-amino substituted halochalcone N-glycoside derivative compounds. Methods: The antioxidant capacity of synthesized compounds (1-4) was determined by cupric ion reducing antioxidant capacity (CUPRAC), and ferric reducing antioxidant power (FRAP). Antimicrobial activity was determined on 9 microorganism by agar well diffusion method.Results: According to the study results, compound 3 showed the highest antioxidant activity. The compound 3 showed antimicrobial activity against Yersinia pseudotuberculosis, Pseudomonas aeruginosa, Candida albicans while compound 4 showed antimicrobial activity against Escherichia coli, Yersinia pseudotuberculosis, Pseudomonas aeruginosa, Enterococcus faecalis, Bacillus cereus, Staphylococcus aureus.Conclusion: 2-amino substituted halochalcone N-glycoside derivative compounds could be evaluated in the pharmaceutical and cosmetic fields due to their antioxidant and antimicrobial potential.

Design, synthesis, and quantum chemical calculations of triazole-based aroylhydrazone molecules: Dual assessment of antioxidant properties via in vitro and in silico approaches

In this study, five novel Triazole-Based Aroylhydrazone compounds (6a-e) were synthesized in high yields (87–79 %), starting from deferasirox which has known iron chelator, and characterized with NMR, FTIR, Mass, and UV–vis spectroscopic methods. Quantum chemical calculations of all synthesized molecules were performed at the DFT/B3LYP/6-311G(d,p) level. It was observed that both theoretical and experimental spectral values were compatible. Optimized molecular geometries were acquired from DFT calculations and used in molecular docking studies. Tyrosinase (PDB ID: 3NM8) protein was utilized in molecular docking studies. 6d-3NM8 complex had the highest docking score with the -10.1 kcal/mol. Stabilization of the 6d-3NM8 complex during 100 ns was determined via molecular dynamics simulation. Intramolecular interactions of all molecules were also examined by NCI-RDG analysis. The frequently used cupric ion reducing antioxidant capacity method (CUPRAC) was used to determine the antioxidant activity of synthesized compounds and Trolox which is unknown in the literature on its activity in DMSO. The highest TEAC value was 1.76 (6d) while the lowest one was 1.34 (6e). It is possible to say that the synthesized molecule 6d has the potential to be an antioxidant reagent based on the results of in vitro and in silico antioxidant activity.

Effect of the Phenological Stage on the Phenolic Composition, and Antioxidant and Antimicrobial Properties of Cirsium vulgare (Savi) Ten. Extracts.

Cirsium vulgare (Savi) Ten. is a plant from the Asteraceae family that is commonly used in traditional medicine. The purpose of this work was to investigate the antioxidant and antimicrobial characteristics of phenolic compounds found in ethanol and dry extracts of C. vulgare leaves, inflorescence, and roots during various phenological stages. Apigenin-7-O-glucoside and chlorogenic acid were identified in practically all C. vulgare extracts. Extracts from leaves collected at the end of the phenological dormancy period and in the first growing year had the highest antioxidant (cupric ion-reducing antioxidant capacity of 12,938 Trolox equivalents/g dry weight) and antimicrobial activity (against Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Proteus vulgaris, and Candida albicans) with MIC values of ethanol extract from 16.7 mg/mL to 8.35 mg/mL. These extracts included a high concentration of chlorogenic acid and apigenin-7-O-glucoside. Also, dry extracts from C. vulgare roots and inflorescences showed a higher antimicrobial effect compared to ethanolic extracts with MIC values from 5.57 mg/mL to 3 mg/mL. The study emphasizes the critical role of phenological stages and raw material composition in the accumulation of phenolic compounds and their biological activity in C. vulgare. The findings suggest that extracts from C. vulgare leaves, especially those collected at the end of the phonological dormancy period, are promising candidates for further research into bioactive compounds with potential medicinal applications. The strong antioxidant and antibacterial properties of these extracts highlight their potential for development into natural pharmaceutical products.

Antioxidant capacity and phenolic profile of defatted seed powder derived from organically grown vine crops

Grape seeds derived from winemaking pomace represent a significant waste stream with potential for valorization due to their rich phenolic content. The present study aimed to assess the phenolic composition and antioxidant potency of defatted grape seeds powder (DP) resulting after oil extraction through three different methods (i.e., cold stirring (ST), ultrasound-assisted extraction (US), and microwave-assisted extraction (MW)) from two grape varieties (i.e., Fetească Regală and Merlot) grown under organic farming conditions. The phenolic profile was determined using High-performance liquid chromatography with diode array detection and mass spectrometry operating in electrospray ionization positive mode, while the total phenolic content was determined using an adapted Folin-Ciocalteu technique, The samples' antioxidant properties were determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging ability assessments, as well as Cupric ion reducing antioxidant capacity and Ferric reducing antioxidant power assays. The phenolic qualitative profile differed between the two varieties and was unaffected by the defatting method. Instead, at the level of each variety, significant variations (p<0.05) were observed in the quantity of phenolic compounds, depending on the defatting method. Moreover, the total phenolic content was significantly higher in the MW-DP compared to ST-DP and US-DP (p<0.05), particularly from Fetească Regală grape seeds. The DPPH assay demonstrated the highest antioxidant activity in MW-DP from Fetească Regală. The study underscores the importance of the extraction methods and grape variety on the phenolic profile and antioxidant potency of DP. These findings suggest potential applications in pharmaceuticals, functional foods, and cosmetics, offering sustainable solutions for grape pomace utilization.

Exploring the In Vitro and In Vivo Therapeutic Efficacy of Juniperus oxycedrus Cade Oil: Antioxidant, Anti-inflammatory, and Anti-asthmatic Effects in an Allergic Asthma Model.

This investigation aimed to explore the antioxidant, anti-inflammatory effects of Cade oil and its efficacy within a Wistar allergic asthma model. The antioxidant activity was assessed through various in vitro tests using chain-breaking antioxidant effects (radical scavenging and reducing abilities assays). In vivo experiments involved Wistar rats categorized into four groups: negative control group, Ovalbumin-sensitised/challenged group, Cade oil-treated group, and Ovalbumin-sensitised/challenged Cade oil-treated group. These experiments aimed to evaluate oxidative stress parameters in the lungs and erythrocytes. The results indicated that the Cade oil exhibited significant antioxidant capabilities, evidenced by its radical scavenging activity against DPPH, ABTS, and Galvinoxyl radicals, with IC50 values ranging from 21.92 to 24.44 µg/mL. Besides, the reducing abilities methods showed A0,5 value ranging from 11.51 to 30.40 µg/mL for reducing power, Cupric ion reducing antioxidant capacity, and O-phenanthroline assays. Additionally, the IC50 value for β-carotene scavenging was found to be (8.2 ± 0.25 µg/ml). Analysis revealed high levels of polyphenols and flavonoids in Cade oil, indicating rich polyphenol (275.21 ± 3.14 mg GAE/g DW) and flavonoid (28.23 ± 1.91 µg QE/mg) content. In vivo findings highlighted Cade oil's efficacy in reducing inflammatory cell recruitment, enhancing antioxidant status, reducing lipid peroxidation, and improving histopathological alterations within the allergic asthma model. These results demonstrated that Cade oil has a potent antioxidant, anti-inflammatory, and anti-asthmatic properties, suggesting its potential therapeutic application in asthma treatment.

LC−MS/MS−based metabolite profiling and antioxidant evaluation of three Indonesian orange varieties

The orange peel contains bioactive components with health−promoting effects, a rich source of antioxidants, and could be used for functional food. However, due to many factors, the composition and concentration of bioactive compounds from orange peels differ because of their variety. This study aims to identify the metabolite and evaluate the antioxidant activity of peels from Indonesian orange varieties, i.e., Rimau Gerga Lebong (RGL, Bengkulu), Brastagi (JBM, North Sumatra), and Garut (JSG, West Java). The metabolite profile was analyzed using LC−MS/MS and discriminated based on orange variety using principal component analysis (PCA) and hierarchical clustering analysis (HCA). Antioxidant activity was measured using the 2,2 −diphenyl− 1 −picrylhydrazyl (DPPH), cupric ion−reducing antioxidant capacity (CUPRAC), and ferric−reducing antioxidant power (FRAP) methods. In this study, the metabolite profile of orange peel proved to be rich in flavonoids like hesperidin, rutin, nobiletin, polyphenol−organic acids like p − coumaric acid, citric acid, quinic acid, and other metabolites from the class terpenoids, amino acids, and sugars. PCA separated each variety and HCA heatmaps showed variations in the distribution of identified metabolites. In the RGL variety, 47 metabolites were identified, dominated by flavonoids and terpenoids. As many as 44 metabolites in the JBM variety contain many amino acids and flavonoids. In JBM, 44 metabolites were identified with carboxylic and amino acids as the main compounds. RGL orange peel extract had the highest antioxidant activity using the DPPH and CUPRAC methods, whereas the FRAP method did not show significant differences. Regarding metabolites and antioxidant activity, RGL, JBM, and JSG orange peels can be developed as alternative functional foods.

The Effect of Various Extraction Methods and Solvents on the Phytochemical Contents and Antioxidant Capacities of Safflower Florets (Carthamus Tinctorius L.) from South Sulawesi

Safflower is a traditional medicine commonly applied for the treatment of varicella in South Sulawesi. The aim of this study is to study the effect of extraction methods and solvents on phytochemical content and antioxidant capacity in the extraction of safflower. The study began with sample extraction with various solvents, i.e., water, ethanol 70, methanol, ethyl acetate and hexane and various methods, i.e., reflux, Soxhlet, hot maceration, cold maceration and room temperature maceration. Each extract was standardized for its phytochemical content, i.e., phenolics, flavonoids and carotenoids and the antioxidant capacities were also determined by the radical scavenging 2,2-diphenyl-1-picrylhydrazyl, total antioxidant capacity, cupric ion reducing antioxidant capacity and Ferric reducing antioxidant power. The results indicated that the optimum total phenolic content was obtained when the florets were refluxed with methanol, flavonoids were refluxed with 70 % ethanol and carotenoids were Soxhleted with methanol. The research data also showed that 70 % ethanol and methanol have maximal antioxidant capacity. The results conclude that 70 % ethanol and methanol as solvents have the potential to extract the highest levels of phytochemical content and antioxidant substances. HIGHLIGHTS The phytochemical content and antioxidant capacity of safflower floret extract are influenced by the selection of solvent and extraction method. Polar solvents (70 % ethanol or methanol) can efficiently extract higher levels of phytochemical components and antioxidant substances from safflower florets. The hot extraction approach exhibits a higher potential for extracting phytochemical content compared to the cold extraction method, but its antioxidant capacity exhibits the opposite. The application of cold maceration, combined with methanol or 70 % ethanol as solvents, has been identified as a promising method for the extraction of phytochemical compounds and antioxidant substances. Characterization of polyphenols and carbohydrates exuded by Phaeodactylum tricornutum diatom grown under Cu stress

This study is focused on analysing polyphenols and carbohydrates released by Phaeodactylum tricornutum (P. tricornutum) diatoms cultured in natural seawater enriched with sublethal and lethal Cu doses. Cu concentrations of 0.31, 0.79 and 1.57 µM reduced cell densities by 37, 82 and 91%, respectively, compared to the control. The total sum of all identified polyphenols and total carbohydrates released by cells grown under lethal Cu levels increased up to 18.8 and 107.4 times, respectively, compared to data from a control experiment. Four different in vitro assays were used to estimate the antioxidant activities of the extracellular compounds: 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical inhibition, cupric ion reducing antioxidant capacity (CUPRAC), ferric reducing antioxidant power and Cu complexing ability (CCA). The highest antioxidant activities were observed in the Cu lethal treatments, where the CCA assay exhibited a greater increase (up to 32.2 times higher than that found in the control experiment) to reduce the concentration of free Cu in the medium and its toxicity. The presence of Cu stimulated the release of polyphenols and carbohydrates to the medium as a detoxification mechanism to survive under lethal levels of Cu regulating its speciation.

Characterization and antioxidant activity of peel extracts from three varieties of citrus sinensis

High volume of postharvest materials including peels from citrus fruits is periodically generated, which contributes to environmental pollution. Investigating the chemical composition cum antioxidant property of these ‘wastes’ would be instructive in achieving value addition in the food and pharmaceutical value chain. On this premise, this study carried out phytochemical screening and antioxidant activity of three (3) commonly cultivated citrus varieties namely Citrus sinensis ‘valencia’, Citrus sinensis ‘washinton’ and Citrus sinensis ‘thompson navel’. The peels were extracted using ethanol and hexane in a Soxhlet extractor and thereafter subjected to phytochemical and Gas Chromatography/Mass Spectrometry (GC/MS) analyses, ferric ion reducing antioxidant power (FRAP), hydrogen peroxide scavenging and cupric ion reducing antioxidant capacity (CUPRAC) assays to evaluate their antioxidant potentials. Results show that Citrus sinensis peel extracts contain alkaloids, flavonoids, phenols, phytosterols, diterpenes, tannins and glycosides. GC/MS analysis identified about 48 compounds in each extract; with the predominant bioactive compounds being limonene (16.5%), ascorbic acid (17.7%), stearic acid (26.3%), linalool (4.7%), linoleic acid (16.18%), palmitic acid (15.23%), pentadecyclic acid (1.1%). Ethanol and hexane extracts of Valencia exhibited higher FRAP (9.09 ± 0.13) and CUPRAC (2.04 ± 0.06) values while the ethanol extract of Ibadan sweet demonstrated greater hydrogen peroxide scavenging activity (1.39 ± 0.00). Citrus peels are rich in bioactive compounds with excellent antioxidant activity and may serve as potential sources of natural antioxidants for food products or pharmaceutical formulations.

CYTOTOXICITY SCREENING AND ANTIOXIDANT CAPACITY ASSESSMENT OF THE INNER PERIANTH SEGMENTS OF 14 RUMEX SPECIES GROWN IN TÜRKİYE

Objective: Breast cancer is one of the most prevalent cancer types worldwide. Antioxidant sources may prevent the occurrence of cancer. Natural sources rich in phenolics, thus, may provide alternate agents in the management of breast cancer. Rumex species are widely distributed in Turkish flora. Emerging evidence has pointed out the antitumoral property of Rumex species on a variety of cancer cells. In the present study, we propose to test the ethanolic extracts of the inner perianth segments of 14 Rumex species on four breast cancer cells with different origins. We also demonstrated their toxicity on healthy cells. Material and Method: We performed the resazurin reduction assay to examine the cytotoxicity and toxicity. Furthermore, we determined the phenolic contents of the extracts as an indicator of their antioxidant profile and ascertained their antioxidant activities by DPPH radical, ABTS radical cation scavenging activity and cupric ion-reducing antioxidant capacity assays. Result and Discussion: The ethanolic extracts of the inner perianth segments of Rumex species exhibited remarkable cytotoxicity profiles neither on breast cancer cells nor on healthy H9c2 rat myoblastoma cells. However, they usually displayed strong antioxidant activities due to possessing high phenolic content.

Synthesis, antioxidant, DNA interaction, electrochemical, and spectroscopic properties of chromene-based Schiff bases: Experimental and theoretical approach

Two chromene-based Schiff base ligands were synthesized by the condensation of 5-hydroxy-2,2-dimethyl-2H-chromene-6-carbaldehyde with 2-aminodiphenylamine and 1,8-diaminonaphthalene: (E)-2,2-dimethyl-6-(((2-(phenylamino)phenyl)imino)methyl)-2H-chromen-5-ol (2) and (E)-6-(((8-aminonaphthalen-1-yl)imino)methyl)-2,2-dimethyl-2H-chromen-5-ol (3). The compounds were characterized by Fourier transform infrared (FT-IR), absorption (UV-Vis) and nuclear magnetic resonance (NMR) spectroscopies, in addition to mass spectrometry (MS) analyses; the results of which were rationalized using density functional theoretical (DFT) calculations. Antioxidant activity of the Schiff bases was determined using the DPPH free radical scavenging and CUPric Ion Reducing Antioxidant Capacity (CUPRAC) assays. Compound 3 (IC50 = 14 μM) showed greater free radical scavenging ability than compound 2 (35 μM), and was also more effective than ascorbic acid and Trolox. DFT studies indicate that the HAT mechanism is favored over SET for DPPH scavenging. Both compounds exhibited stronger Cu(II) ion reducing capacity than Trolox and ascorbic acid with Trolox equivalent activity coefficient (TEAC) values of 2.0 and 1.6 for compounds 2 and 3, respectively. Furthermore, DNA cleavage and binding properties were determined using gel electrophoresis, UV-Vis spectroscopy, and cyclic voltammetry. The compounds showed copper-mediated cleavage of plasmid DNA and strong binding to DNA, with binding constants on the order of 104 M−1. The strength of DNA binding was verified by molecular docking analysis using two DNA models: a B-DNA dodecamer (-8.4 to -9.3 kcal/mol) and a hexamer (-7.8 to -7.9 kcal/mol). Overall, the chromene Schiff bases show promise as antioxidant and DNA binding agents.

Phytochemical analyses by LC-HRMS, FTIR spectral analysis, antioxidant, antidiabetic and antityrosinase activity of Crataegus orientalis Pall. ex M. Bieb fruit extracted with various solvents.

Crataegus orientalis Pall. ex M. Bieb fruit (COPMB) is extensively used as a source of various products in the medicinal-aromatic field and holds the potential for erosion control, ornamental purposes, food source, and economic benefits for forest villagers from its fruits. This study aims to determine the chemical components and biological activities of extracts prepared from COPMB using different solvents. The present work was designed to define the antioxidant activity [phosphomolybdenum (total antioxidant capacity), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP), cupric ion-reducing antioxidant capacity (CUPRAC) and metal chelating activity (MCA)], phytochemical screening analysis, enzyme inhibitor (α-amylase, α-glucosidase and tyrosinase) potential, and liquid chromatography-high resolution mass spectrometry (LC-HRMS) secondary metabolite profiling in different extracts of COPMB. The results of LC-HRMS revealed that fumaric acid was the main phenolic compound in all extracts. Among the extracts, ethyl acetate extract has the highest phytochemical and antioxidant properties [total phenolic content (TPC): 32.5 mg GAE/g, total flavonoid content (TFC): 12.2 mg QE/g, ABTS: 213.0 mg TE/g; CUPRAC: 126.0 mg TE/g, MCA: 145.0 mg EDTA/g; FRAP: 122.8 mg TE/g; TAC: 2.8 mmol TE/g]. Ethyl acetate and methanol extracts are more effective in α-amylase (0.27 ± 0.01 mg/mL; 0.12 ± 0.00 mg/mL), α-glucosidase (0.63 ± 0.02 mg/mL; 0.77 ± 0.02 mg/mL) and tyrosinase (0.03 ± 0.00 mg/mL; 0.03 ± 0.00 mg/mL) enzyme inhibition potentials compared to standard acarbose (0.75 ± 0.02 mg/mL for α-amylase; 1.11 ± 0.03 mg/mL for α-glucosidase) and kojic acid (0.04 ± 0.00 mg/mL). The findings from this study suggest that COPMB could serve as a valuable source of natural agents for the food and pharmaceutical industry. © 2024 Society of Chemical Industry.

In vitro research to evaluate the antioxidant effects, inhibiting enzymes, methicillin-resistant Staphylococcus aureus strains of Terminalia catappa extracts

AimContribute to the well-being of the Ivorian population through scientific proof of the use of Terminalia cattapa leaves extracts in traditional medicine. MethodologyThe antioxidative effect of aqueous, water-alcoholic, ethyl acetate and hexane derivatives of Terminalia catappa leaves was measured by six tests: DPPH (2-diphenyl-1-picrylhydrazyl), ABTS (2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)), FRAP (ferric ion reducing antioxidant power), PPBD (phosphomolybdenum), CUPRAC (cupric ion reducing antioxidant capacity) and MCA (metal chelating activity). The inhibition of the activity of five enzymes (acetylcholinesterase, butyrylcholinesterase, tyrosinase, amylase and glucosidase) was also determined. In addition, the broadcast method was used to evaluate impact of the samples on methicillin-resistant Staphylococcus aureus (MRSA) strains. ResultsHighest antioxidant capacity was obtained in the ethanolic sample for DPPH, ABTS, FRAP and PPBD tests. Ethyl acetate sample produced the best CUPRAC and MCA tests. Among inhibitory effect of samples on the five enzymes, aqueous sample showed the strongest anti-acetylCholynesterase and anti-butyrylCholynesterase activity. Best anti-tyrosinase and anti-amylase activity were obtained with the ethanolic sample. Hexane sample obtained the best anti-glucosidase activity while ethanolic and aqueous samples obtained none. All extracts showed no activity on the MRSA strains tested. ConclusionThe chemical compounds present in Terminalia cattapa leaves extracts would not be responsible for the use made in traditional medicine against infections.

Antioxidant Activities of Stem, Leaves and Fruits Extracts of Pepino (Solanum muricatum Aiton).

<b>Background and Objective:</b> Pepino (<i>Solanum muricatum</i> Aiton), rich with vitamin C and flavonoids, constitutes an abundant source of potent antioxidants. This research was conducted to determine antioxidant activity from three different parts of pepino based on equivalence with ascorbic acid, to analyze the relationship between total phenolic content (TPC) and total flavonoid content (TFC) on antioxidant activities and to determine flavonoid compounds. <b>Materials and Methods:</b> Antioxidant activities were determined using 2,2-Diphenyl-1-Picrylhydrazyl (DPPH) and Cupric Ion Reducing Antioxidant Capacity (CUPRAC) methods. The TPC and TFC were determined by UV-visible spectrophotometry. The correlation between TPC, TFC and antioxidant activity was analyzed using Pearson's method. Flavonoid compound content was performed by HPLC. <b>Results:</b> The ethyl acetate pepino fruit extract expressed the highest antioxidant activity by DPPH and CUPRAC assays. The highest TPC was obtained from the ethyl acetate extract of pepino stem (18.493 g GAE/(100 g)), while the highest TFC was obtained from the hexane extract of pepino leaves (9.541 g QE/(100 g)). <b>Conclusion:</b> The DPPH and CUPRAC assays demonstrated that pepino exhibits potential as a source of natural antioxidants, especially in its fruit part.

Abstract Background: Retinopathy is one of almost insidious advanced neuron-vascular complications of diabetes, oneof the most common complications that lead to permanent visual impairment in the middle-aged population. Objectives: The study investigated the association between poor oxidative stress index and DNA methylation. Materials and Methods: The study included 120 participants, of whom 40 were healthy, 40 patients with retinopathy, and 40 non-retinopathy diabetes patients (type 2 diabetes mellitus (T2DM)). The total antioxidant, capacity (TAC) evaluated using Cupric ion reducing antioxidant capacity method. Reactive oxygen species (ROS) was measured using Erel method. Global DNA methylation has measured employing 5 mC% estimating using MethylFlash™ Methylated DNA Quantification Kit. Results: Current investigation indicated significant (P ≤ 0.05) accrue in TAC, ROSs, and oxidative stress index (OSI) in diabetic compared with controls. In addition, a significant (P ≤ 0.05) increase of ROSs and OSI were recorded in retinopathy compared with T2DM patients was reported. The analysis of global DNA methylation revealed that diabetic patients have a significantly (P ≤ 0.05) more 5 mC% as compared with healthy subjects. Also, the present study showed a significant rise in methylation in retinopathy compared as with T2DM patients. Regarding the correlation analysis, the current study conducted significantly (P < 0.05) positive association between DNA methylation and each of the ROSs and OSI. Also, a significant positive correlation was reported between DNA methylation and ROSs and OSI in retinopathy and T2DM. Conclusion: Global DNA methylation has an increased level in patients and could be engaged with emergence and progress of retinopathy in diabetic with poor control of oxidative stress and increased ROSs.

Cytotoxicity of Sambucus nigra L. on Cancer Cell Line and In Vitro Antioxidant Properties

Objective: Essential oils, free fatty acids, flavonoids, glycosides, phenolic acids, carotenoids, vitamins, and minerals are found in significant quantities in the characteristic chemical composition of Sambucus nigra L. This study aimed to evaluate the antioxidant potential of Sambucus nigra L. Fructus and evaluate the cytotoxicity on the cancer cell line.
 Methods: The Sambucus nigra L. fruits were collected from Yalova Atatürk Horticultural Central Research Institute in September 2021. The ethanol extract was prepared. Antioxidant property of Sambucus nigra L. fruit extracts was evaluated with 2,2-diphenyl-1-picryl-hydrazyl- hydrate (DPPH) radical scavenging activity, Cupric Ion Reducing Antioxidant Capacity (CUPRAC). Also, total phenolic content, total flavonoid content, and total anthocyanin content were calculated. Liver hepatocellular carcinoma cell line (HepG2) was used for cytotoxicity assay and an 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay was applied.
 Results: The total phenolic, total flavonoid contents and total monomeric anthocyanins were 9.75±0.92 mg GAE/ mg fruit extract, 0.07437±0.004 mg quercetin/ mg fruit extract, respectively. 0.45±0.0014 mg catechin/gram of fruit extract and 2.08±0.025 mg Cyanidin-3 glucosides/g fruit extract. CUPRAC and DPPH results showed that Sambucus nigra L. extract has strong antioxidant activity. The results of the cytotoxicity assay indicated that while concentrations of the extract increased, the viability of HepG2 decreased.
 Conclusion: Our findings suggest that the Sambucus fruit extract is particularly rich in antioxidant components that are possibly modulating their beneficial use for hepatocellular malignancies, significantly reducing the number of viable cancer cells and inducing cell death.

Liposomes as a carrier for 4–hydroxycoumarin: characterisation, stability and antioxidant potential

<p>The aims of the present research were to develop and characterize 4-hydroxycoumarin-loaded liposomes <em>via </em>the determination of encapsulation efficiency, antioxidant capacity, vesicle size, polydispersity index (PDI), zeta potential, conductivity, mobility, density, surface tension, viscosity, and 60-day storage stability. The encapsulation efficiency of 4-hydroxycoumarin in liposomal particles was 96.7 ± 1.2%. The ABTS and DPPH radical scavenging capacity of the prepared liposomes was 89.76 ± 0.56% and 93.18 ± 0.23%, respectively, whereas cupric ion-reducing antioxidant capacity amounted to 0.367 ± 0.003 mmol Trolox equivalent (TE)/L. The density of liposomes with 4-hydroxycoumarin was 1.007 ± 0.002 g/cm<sup>3</sup>, surface tension was 22.7 ± 0.2 mN/m, and viscosity was 14.3 ± 0.2 mPa·s. Vesicle size and PDI of 4-hydroxycoumarin-loaded liposomes were changed from 1286.3 ± 73.8 nm to 2077.3 ± 63.2 nm and from 0.409 ± 0.050 to 0.676 ± 0.064, respectively, during the 60-day stability study. The zeta potential of the obtained liposomes was changed from -16.73 ± 0.47 mV to -10.31 ± 0.42 mV, while mobility varied from -1.311 ± 0.036 µmcm/Vs to -0.806 ± 0.031 µmcm/Vs. The conductivity did not change during 60 days and amounted to approximately 0.020 mS/cm. Overall, due to the high encapsulation efficiency and antioxidant capacity, the obtained results qualify liposomes to be used as 4-hydroxycoumarin carriers for future examination of its biological activities and potential application in medicine and pharmaceutical products. However, future experiments should include the optimization of the liposomal composition with the aim of improving 4-hydroxycoumarin-loaded liposome stability.</p>

Genistein Co-Amorphous Systems with Amino Acids: An Investigation into Enhanced Solubility and Biological Activity.

Genistein, an isoflavone known for its antioxidant and antidiabetic effects, suffers from the drawback of low solubility. To overcome this limitation, co-amorphous systems were synthesized by incorporating amino acids that were chosen through computational methods. The confirmation of the amorphous state of lysine and arginine-containing systems was ascertained by X-ray powder diffraction. Subsequently, the characterization of these systems was extended by employing thermo-gravimetry, differential scanning calorimetry, Fourier-transform infrared spectroscopy, and scanning electron microscopy. The investigation also included an assessment of the physical stability of the samples during storage. The apparent solubility of the systems was studied in an aqueous medium. To evaluate the in vitro permeability through the gastrointestinal tract, the parallel artificial membrane permeability assay was employed. The biological properties of the systems were assessed with regard to their antioxidant activity using 2,2-diphenyl-1-picrylhydrazyl and cupric ion-reducing antioxidant capacity assays, as well as their ability to inhibit α-glucosidase. The systems' glass transition temperatures were determined, and their homogeneity confirmed via differential scanning calorimetry analysis, while Fourier-transform infrared spectroscopy analysis provided data on molecular interactions. Stability was maintained for the entire 6-month storage duration. The co-amorphous system containing lysine displayed the most pronounced apparent solubility improvement, as well as a significant enhancement in antioxidant activity. Notably, both systems demonstrated superior α-glucosidase inhibition relative to acarbose, a standard drug for managing type 2 diabetes. The results indicate that co-amorphous systems with lysine and arginine have the potential to significantly enhance the solubility and biological activity of genistein.

Catechin Composition, Phenolic Content, and Antioxidant Properties of Commercially-Available Bagged, Gunpowder, and Matcha Green Teas

The leaves of the Camellia sinensis plant are used to produce many different types of teas, and green tea is particularly noted for its health promoting properties which are attributed to high concentrations of phenolic compounds known as catechins. Green tea is available in forms such as bagged, gunpowder (rolled leaf), and matcha, a fine powder. This study evaluated the phenolic content, catechin composition, caffeine concentration, and antioxidant properties of fifteen commercially-available green teas with an emphasis on identifying differences between ceremonial and culinary matcha. Gunpowder and bagged green teas had higher total phenolic contents and greater CUPRAC (cupric ion reducing antioxidant capacity) and ORAC (oxygen radical absorbance capacity) antioxidant capacities than matcha teas. Among matcha samples, less expensive culinary teas had greater total phenolic contents and higher antioxidant capacities than ceremonial products. Hierarchical cluster analysis was used to determine similarities and differences in catechin composition and revealed that bagged and gunpowder green teas had the highest levels of the stereoisomers epi/catechin gallate, which correlated with their strong antioxidant properties. Bagged and gunpowder teas were the lowest cost products in this study, indicating that they are an economical choice for consumers who want a green tea beverage with high catechin content, although they generally had lower caffeine levels than matcha.
