Purified Chick Embryo Cell Culture Rabies Vaccine: interchangeability with Human Diploid Cell Culture Rabies Vaccine and comparison of one versus two-dose post-exposure booster regimen for previously immunized persons

The equivalence and interchangeability of Purified Chick Embryo Cell Culture Rabies Vaccine (PCECV) to Human Diploid Cell Culture Rabies Vaccine (HDCV) and the immunogenicity of a reduced post-exposure regimen with PCECV was investigated. Statistical analyses revealed no difference ( P≤0.05) between the geometric mean titers (GMT) on day 49 of subjects that received PCECV or HDCV. In Year 2, subjects were boosted with one or two dose(s) of PCECV. No significant difference ( P≤0.05) was detected between the GMT of the two groups on days 7 and 365 post-booster. Subjects that received HDCV initially developed an adequate anamnestic response to PCECV. On day 21 post-booster, the GMT of subjects that received two boosters was higher (151.6 IU/ml) than those that received one booster (120.9 IU/ml). However, this difference may not be clinically significant.

A Novel Pharmacological Approach to Treating Cardiac Ischemia: BINARY CONJUGATES OF A1 AND A3 ADENOSINE RECEPTOR AGONISTS

Adenosine released during cardiac ischemia exerts a potent, protective effect in the heart via activation of A(1) or A(3) receptors. However, the interaction between the two cardioprotective adenosine receptors and the question of which receptor is the more important anti-ischemic receptor remain largely unexplored. The objective of this study was to test the hypothesis that activation of both receptors exerted a cardioprotective effect that was significantly greater than activation of either receptor individually. This was accomplished by using a novel design in which new binary conjugates of adenosine A(1) and A(3) receptor agonists were synthesized and tested in a novel cardiac myocyte model of adenosine-elicited cardioprotection. Binary drugs having mixed selectivity for both A(1) and A(3) receptors were created through the covalent linking of functionalized congeners of adenosine agonists, each being selective for either the A(1) or A(3) receptor subtype. MRS 1740 and MRS 1741, thiourea-linked, regioisomers of a binary conjugate, were highly potent and selective in radioligand binding assays for A(1) and A(3) receptors (K(i) values of 0.7-3.5 nm) versus A(2A) receptors. The myocyte models utilized cultured chick embryo cells, either ventricular cells expressing native adenosine A(1) and A(3) receptors, or engineered atrial cells, in which either human A(3) receptors alone or both human A(1) and A(3) receptors were expressed. The binary agonist MRS 1741 coactivated A(1) and A(3) receptors simultaneously, with full cardioprotection (EC(50) approximately 0.1 nm) dependent on expression of both receptors. Thus, co-activation of both adenosine A(1) and A(3) receptors by the binary A(1)/A(3) agonists represents a novel general cardioprotective approach for the treatment of myocardial ischemia.

Skin Reaction to Yellow Fever Vaccine After Immunization with Rabies Vaccine of Chick Embryo Cell Culture Origin

Skin reaction to yellow fever vaccine was examined after immunization with rabies vaccine. The two vaccines contained substrates from chick embryo cells (rabies vaccine) and chick whole embryo (yellow fever attenuated vaccine), as well as gelatin. A prick test with gelatin showed negative results in all vaccinees examined. An intradermal skin test revealed that the yellow fever vaccine had reacted with an anti-egg protein antibody-like substance in a case with a history of egg allergy before rabies vaccination. A case inoculated two times with the rabies vaccine revealed a positive reaction to egg-white protein as well as the yellow fever vaccine. This case had no anamnesis of egg allergy. Thus, an antibody reactive to the egg-white protein and/or the yellow fever vaccine was inducible by the rabies vaccine. The reaction of this antibody was not systemic but local at the skin test by the yellow fever vaccine. The period of the rabies vaccine sensitization reactive to the yellow fever vaccine could be estimated as longer than 14.3 +/- 9.6 days (mean +/- SD), based on a follow-up examination of the positive skin reaction in 41 of 84 cases examined. We therefore conclude that the yellow fever vaccine can be safely administered at an interval of at least four weeks after a second rabies vaccination.

A protein from Naegleria amoebae causes apoptosis in chick embryo and cho cells after they become confluent

Exposure for less than an hour to a protein isolated from Naegleria amoebae initiates a process that has no apparent effect on the appearance or growth of chick embryo or CHO cell cultures for 4 to 9 days; after the development of confluency, at some unknown signal, all of the cells undergo an apoptotic death within a 12- to 24-hour period. Abnormalities detected among the last mitotic cells include chromosomal breakage and early reversal in metaphase to telo/interphase daughter nuclei with irregular shapes. Additional events in the dying cultures include the development of a cytoplasmic amoebic-related immunogen, gross DNA fragmentation, cell blebbing, shrinkage, and apoptotic body formation. Culture death included all cells, those present in confluent cultures when the protein was added, and in other cultures, those formed during a more than 30-fold increase in cells as the cultures became confluent. The increase in the number of cells followed by the uniformity and synchrony of their death pattern indicates that the signal to kill has increased and spread throughout the culture; upon an unknown condition related to confluency, events are initiated that lead to the unusual apoptotic death of the culture.

Human diploid cell culture rabies vaccine (HDCV) and purified chick embryo cell culture rabies vaccine (PCECV) both confer protective immunity against infection with the silver-haired bat rabies virus strain (SHBRV)

The demonstration of extensive differences in the antigenic makeups of the silver-haired bat rabies virus (SHBRV) and canine rabies virus (COSRV) strains raised concerns as to whether current licensed rabies vaccines are sufficiently protective against SHBRV. NIH mouse protection test results show that both the human diploid cell culture rabies vaccine (HDCV) and the purified chicken embryo cell rabies vaccine (PCECV) protected against lethal infection with SHBRV as well as the canine rabies strain COSRV. However, in this investigation, the potencies of both vaccines in mice were found to be significantly higher for COSRV than for SHBRV. The in vivo protection data are confirmed by in vitro virus neutralizing antibody (VNA) test results which demonstrate that mice immunized with HDCV or PCECV develop significantly higher VNA titres against COSRV than against SHBRV. In contrast, VNA tests of sera from individuals immunized with HDCV or PCECV showed that humans, as opposed to mice, develop significantly higher VNA titres against SHBRV than against COSRV. These data suggest that HDCV and PCECV will protect humans against infection with the silver-haired bat rabies virus strain in addition to canine rabies virus strains.

Sodium ion-dependent (N-methylamino)-alpha-isobutyric [correction of isobutryic] acid uptake by embryonic chick cells exposed to ethanol in ovo:response to the stimulation/downregulation of protein kinases.

Amino acid uptake, critical for embryonic development, was studied in cultured chick cells isolated from 5-day-old chick embryos that had received treatment in ovo with either vehicle (sterile chick Ringer's solution) or vehicle plus ethanol (1.5 g/kg). Upon being placed in culture, the cells were grown in the absence of ethanol per se. The uptake of (N-methylamino)-alpha-isobutyric [correction of isobutryic] acid (AIB) was used to estimate amino acid uptake via system A, a Na+-dependent system that transports short-chain amino acids. The uptake of AIB in cultured chick embryo cells in response to the stimulation/downregulation of three protein kinases [tyrosine kinase, cyclic AMP-dependent kinase A (PKA) and protein kinase C (PKC)] was determined. Acute activation of PKC by treatment of the cells with a phorbol ester (TPA) inhibited Na+-dependent AIB uptake. Conversely, treatment of the cells with TPA for 24 h, to downregulate PKC activity, significantly increased AIB uptake. The data suggest that in these cells, PKC plays an important role in the regulation of amino acid uptake via system A. Activation of PKA via treatment of the cells with forskolin, an adenylyl cyclase activator, had little effect on AIB uptake in cells from vehicle-treated embryos and only a slight depressing effect on uptake by cells from ethanol-treated embryos. Insulin and insulin-like growth factor-1 (IGF-1) both stimulated AIB uptake, but the higher concentrations of insulin necessary to increase uptake to a level comparable to that of IGF-1 stimulation suggested that insulin could be acting through the IGF-1 receptor. Thus, while AIB uptake by cells of in ovo ethanol-treated embryos was significantly increased by treatment with either insulin or IGF-1, IGF-1 appeared to be the more physiologically important compound in the chick embryo. Overall, there was a consistent trend for cells isolated from in ovo ethanol-treated embryos to have higher levels of AIB uptake relative to cells isolated from vehicle-treated embryos, regardless of the in vitro treatment.

The reverse transcriptase activity in cell-free medium of chicken embryo fibroblast cultures is not associated with a replication-competent retrovirus.

Reverse transcriptase (RT) activity has previously been reported in concentrated medium of primary chicken embryo cell cultures using the traditional RT assay. Recently, using the newly-developed and highly-sensitive product-enhanced reverse transcriptase (PERT) assay, RT activity has been detected in live, attenuated vaccines grown in chicken cell substrates. Furthermore, this activity has been associated with particles that contain RNA related to an ancient, endogenous avian retrovirus family designated as EAV-0. To investigate whether the RT activity present in vaccines produced in specific pathogen-free chicken cell substrates is associated with an infectious retrovirus that can replicate in human cells. The kinetics of RT activity produced by 10-day-old chicken embryo fibroblast (CEF) cultures was determined by analyzing cell-free medium in a PCR-based RT (PBRT) assay. Material containing the peak PBRT activity was used as the inoculum to infect various human cell lines and peripheral blood mononuclear cells. Filtered supernatants from control and test cultures were analyzed for the presence of replication-competent retroviruses by the PBRT assay. The cells were monitored for other adventitious agents by routine observation for cytopathic effect (CPE) and by transmission electron microscopy (TEM) at culture termination. The PBRT activity did not increase above the background level in the human target cells through at least five cell passages, thus indicating the absence of a replicating retrovirus. No other adventitious agents were detected based upon TEM analysis and the absence of CPE. The RT activity produced by chicken primary cell cultures is not associated with a retrovirus that can replicate in human cells.

Chicken interferon types I and II enhance synergistically the antiviral state and nitric oxide secretion.

This report shows that chicken type I and type II interferons (IFNs), like their mammalian counterparts, act synergistically such that a mixture of the two has much greater activity than that expected from the separate contribution of each type. The degree of antiviral synergy was measured by virus plaque reduction and cytopathic effect (CPE) inhibition in both primary and secondary chicken embryo cell cultures. Mixtures of the two Ch-IFNs produced antiviral effects 3-10 times greater than that expected from strict additivity of each IFN acting alone. At high concentrations of IFN mixtures there was a qualitative shift to an exponential IFN action does-response curve that revealed synergistic effects greater than 100-fold. Synergy resulted even with mixtures containing less than 1 U/ml of either type of Ch-IFN. The antiviral effects developed more rapidly with mixtures than when type I or II Ch-IFN was used alone. Mixtures of the two types of Ch-IFN synergistically potentiated nitric oxide secretion in cells of the HD11 chicken macrophage line. Molecular mechanisms are cited that may account for synergy between type I and type II IFNs, and speculation is offered on the epidemiologic and therapeutic implications of synergy in vivo.

Availability of New Rabies Vaccine for Human Use

On October 20, 1997, the Food and Drug Administration licensed a new rabies vaccine for both pre-exposure and postexposure prophylactic use in humans. This purified chick embryo cell culture (PCEC) vaccine (RabAvert) is manufactured by Chiron Behring GmbH and Company. The addition of PCEC to the current products available for pre-exposure and postexposure prophylactic use in humans allows for greater flexibility in treatment choices for the vaccination candidate who develops a sensitivity to one of the other available vaccines. Although derived from chick embryo cells, antibodies to chick cell proteins were not detected in recipients of the vaccine.

CELL–CELL CONTACTS ACCELERATE CELL COLONY GROWTH IN SPARSE CULTURES OF CHICK EMBRYO FIBROBLASTS

There is no cell proliferation in very sparcely plated chick embryo cell cultures. Substituting conditioned medium or adding of ethanol-fixed homologous cells to the cultures accelerates cell colony growth. The mechanism for the mitogenic action of fixed cells is considered to be the contact stimulation of cell proliferation, and addition of extra cells to sparse culture is believed to mimic the cell micro-environment existing in subconfluent cultures. The role of diverse cell–cell contacts in cultured cell growth regulation is discussed. The procedure used (addition of ethanol-fixed cells) may improve normal cell cloning techniques.

An immunogenicity and efficacy study of purified chick embryo cell culture rabies vaccine manufactured in Japan

Purified chick embryo cell rabies vaccine manufactured by the Chemo-Sero-Therapeutic Institute(Kaketsuken) at Kumamoto, Japan (Kaketsuken) was submitted to an immunogenicity and efficacy study. 52 severely rabies exposed patients were treated with the conventional five doses intramuscular WHO approved (‘Essen’) postexposure schedule. This included the administration of 40 IU kg −1 of equine rabies immune globulin on Day 0. A control group of equally severely exposed subjects were treated with human diploid cell rabies vaccine manufactured by the Swiss Serum and Vaccine Institute as well as human rabies immune globulin. There were no deaths in either group in the more than 2 years follow-up period. Subjects treated with the chick embryo vaccine showed greater suppression of the neutralizing antibody response by the equine rabies immune globulin than those given the human diploid cell vaccine and human rabies immune globulin. A group of 20 less severely rabies exposed patients who received only the chick embryo vaccine without immune globulin all had antibody titers greater than the WHO minimal acceptable level on Day 14, 30, 90 and 180. Fourteen subjects among the severely exposed vaccine and immune globulin study group were given vaccine boosters on Day 180 because of low antibody titers. It is concluded that chick embryo rabies vaccine manufactured by Kaketsuken is an immunogenic and effective rabies vaccine, but that the potency of future batches must be increased to provide a greater safety margin.

Modulation of tensin and vimentin expression in chick embryo developing cartilage and cultured differentiating chondrocytes.

It has been proposed that tensin, in association with several other proteins, mediates the micro-filament-integrin link. Here we describe the isolation of clones spanning about 5 kb from the 3' end of tensin mRNA from cultured chick embryo chondrocyte and embryonic heart cDNA libraries. Tensin expression was investigated in cultured chick embryo cells. It was observed that tensin expression is dependent upon substrate adhesion and it is turned off after 7 days of suspension culture. This process is reversible. Tensin expression is also regulated during cartilage cell differentiation in vivo; at Hamburger and Hamilton stage 39-40, non-hypertrophic tibial chondrocytes express both RNA and protein while hypertrophic chondrocytes do not. In the culture system the expression of vimentin, a major component of intermediate filaments, showed an opposite behaviour since the suspension culture enhances the accumulation of both vimentin and its mRNAs. Therefore in chick embryo cultured chondrocytes and in vivo, during cartilage development, cell shape changes and/or integrin-extracellular matrix protein interactions may be involved in the regulation of these two genes coding for cytoskeletal proteins.

Scanning electron microscopy studies on the morphology and release of vesicular stomatitis virus

In spite of convincing evidence to the contrary, the overwhelming majority of virologists still refer to vesicular stomatitis virus (VSV) as “bullet-shaped”. The term bacilliform, which refers to a cylinder with 2 rounded ends, is used infrequently and conditionally to describe VSV.We have used scanning electron microscopy (SEM) to study the morphology of the Indiana strain of VSV attached to, being, or having been released from 3 types of cultured cells, namely, a human melanoma line known as MEWO, human foreskin cells, and primary cultures of chick embryo cells. We have used crude virus suspensions and virus concentrated ten-fold by highspeed centrifugation and resuspension, as well as fresh virus preparations and virus suspensions that have been kept frozen at -70°C and thawed before use.Virus suspended in medium containing 10% fetal bovine serum was used for transmission electron microscopy (TEM). The suspension were negative stained with 1% phosphotungstic acid containing 0.1% bacitracin and viewed in an Hitachi H-7000 TEM. For scanning electron microscopy, virus was adsorbed for 1.5 hours at 37°C unto confluent layers of cells grown in 35 mm plastic petri dishes.

Toxicological studies with primary cultures of chick embryo cells: DNA fragmentation under the influence of DNase I-inhibitors.

Chicken embryo brain and liver cells in vitro exhibited spontaneous DNA fragmentation as determined by viscometry of alkaline cell lysates. Ca2+ and Mg2+ enhanced, while Zn2+, the Ca2+ chelator ethylenglycol-bis(beta-aminoethyl-ether)-N,N,N'-tetraacetic acid (EGTA), spermine and--to a lesser extent--spermidine and Hoechst 33,258 inhibited spontaneous DNA fragmentation. Under the same conditions chromatin condensation, as assessed by nucleoid sedimentation, increased. Exposure of chicken embryo cells to various genotoxic agents, i.e. doxorubicin, bleomycin, methyl methanesulfonate, thiyl radicals, H2O2, UV light, and X-rays, increased DNA fragmentation in a dose dependent manner. Zn2+ or EGTA diminished DNA fragmentation in cells exposed to bleomycin, thiyl radicals, H2O2 and UV light. An apparent sensitisation to X-irradiation has been observed in Zn2+ or EGTA-pretreated cells. It is suggested by the present investigations that, with agent specific peculiarities, apoptotic phenomena are implicated when nucleotoxicity is assessed in chicken embryo cells by physico-chemical short-term tests in vitro.

Regulation of chicken Hsp70 and Hsp90 family gene expression by transforming growth factor-beta 1.

Transforming growth factor-beta 1 (TGF beta) is a regulator of protein synthesis in cultured chicken embryo cells (CEC). Preceding a gradual increase in overall protein synthesis, members of the Hsp70 family (Hsp70, Hsc70, and Grp78) and the Hsp90 family (90-2 and 90-3) of molecular chaperones are induced rapidly and represent a new class of TGF beta-inducible proteins (I.M. Takenaka and L.E. Hightower, J. Cell. Physiol., 152:568-577, 1992). Herein, 32P-labeled cDNA probes encoding Hsc70 and Hsp90 were used to show that levels of the corresponding mRNAs increased as a fraction of total RNA and in polysomes within five hours of treatment of CEC with TGF beta. This cytokine did not increase rates of hsc70 and hsp90 gene transcription as measured by run-on transcription assays of isolated nuclei. However, the Hsp RNA inductions were inhibited by dactinomycin, indicating a requirement for newly synthesized RNA. Both Hsc70 and Hsp90 mRNAs had relatively short half-lives, measured by Northern blot analyses of dactinomycin chases, which were not altered substantially in TGF beta-treated cells. In contrast, Hsp mRNA half-lives increased in heat shocked CEC exposed to dactinomycin during recovery, revealing a difference in regulation of these genes in stressed cells compared with TGF beta-treated cells. Our results support the conclusion that hsc70 and hsp90 gene expression is regulated posttranscriptionally in TGF beta-treated CEC, and the mechanism likely involves a nuclear event such as increasing the half-lives of nuclear RNA transcripts, processing, or transport into the cytoplasm.

Transforming growth factor-beta 1 rapidly induces Hsp70 and Hsp90 molecular chaperones in cultured chicken embryo cells.

In this report we show that: (1) molecular chaperones in the heat shock protein (hsp) family are a new class of cellular proteins induced by Transforming Growth Factor-beta 1 (TGF beta), a cytokine present in serum, (2) rapid induction of Hsc70 precedes a general increase in protein synthesis and may be a preparatory event, (3) TGF beta is a potent regulator of overall protein synthesis in chicken embryo cells (CEC), and (4) isoforms of Hsp90 with different biochemical properties exist, raising the possibility that they may have different functions. TGF beta can substitute for serum in stimulating synthesis of members of the Hsp90 and Hsp70 families of stress proteins, whereas other cytokines, including PDGF, FGF, and EGF, were not effective nor did they enhance the stimulatory effect of TGF beta on the hsp's. Analysis of the induction of hsp's using one- and two-dimensional polyacrylamide gel electrophoresis indicated that members of the Hsp70 family of molecular chaperones were induced rapidly by TGF beta, reaching maximum rates of accumulation by 5 hours of treatment. Total protein synthesis increased more slowly, undergoing an approximately twofold increase in 24 hours. Using a modified protocol for two-dimensional gel electrophoresis, the Hsp90 protein family was separated into four isoelectric forms, two of which were phosphorylated (Hsp90-2 and -4). These phosphorylated isoforms turned over faster than the unphosphorylated forms of Hsp90. All four isoforms were heat inducible, but only Hsp90-2 and -3 were induced rapidly by TGF beta, again within 5 hours of treatment. The effects of serum on these protein families were similar to those of TGF beta, suggesting that this cytokine may be the serum component primarily responsible for up-regulating members of the Hsp90 and Hsp70 families. We hypothesize that cells rapidly increase their chaperoning capacity for newly synthesized polypeptides in preparation for an increase in the rate of synthesis of proteins up-regulated by TGF beta.

Maintenance of motility of fowl spermatozoa in vitro is prolonged by a low molecular weight factor derived from cultured chick embryo cells.

Gel filtration of a conditioned medium composed of the supernatant fluid removed from a 5-day culture of skeletal muscle cells from 9-day-old chick embryos with Bio-Gel P-2 revealed one peak of motility-prolonging activity (about 0.3 kDa), which was not present in fresh medium. Spermatozoa incubated in this fraction of the conditioned medium maintained their motility for at least 36 h at 37 degrees C. Both the formation of lipid peroxide and the leakage of lactic dehydrogenase of spermatozoa incubated in the conditioned medium fraction were lower than those incubated in the corresponding fresh medium. Initial rate of oxygen consumption of the spermatozoa incubated in the conditioned medium fraction increased compared with that of the fresh medium fraction. These results suggest that a low molecular weight factor(s) supplied by cultured cells effectively prolongs the motility of fowl spermatozoa, and that the effect could result from inhibition of the structural damage to the sperm membrane.

Efficacy of acyclovir against herpesvirus infection in Quaker parakeets

SUMMARY We evaluated the efficacy of acyclovir against experimentally induced herpesvirus infection (Pacheco's parrot disease) in Quaker parakeets. Thirty-two of 40 birds were challenge-exposed with 0.1 ml of a suspension of herpesvirus (104 median cell culture infective doses [ccid50]) given im. Treatment with acyclovir was started 24 hours later and was continued for 7 days. The birds were allotted to 5 groups of 8 birds each. There was a considerable difference in mortality between groups 1–5. Of 8 birds in each group, 6 died in group 1 (control), 1 died in group 2 (gavage), 3 died in group 3 (low dose, im), 4 died in group 4 (high dose, im), and none died in group 5 (contact controls). There was a significant (P = 0.023) difference in mortality between groups 1 and 2, thus the oral form of acyclovir administered by gavage was the most efficacious therapeutic regimen. Clinical signs and death occurred after discontinuation of acyclovir in groups 2 and 3, whereas the mean time of death for the control group was 6 days after challenge exposure. Herpesvirus was recovered by inoculation of chick embryo cell culture with pooled tissue suspensions from all birds that died. Histologic evidence of herpesvirus infection was found in most birds that died, with the control group having the most severe lesions. Surviving Quaker parakeets were transferred to cages with seronegative Quaker parakeets with no known exposure to herpesvirus. There have been no deaths attributable to herpesvirus infection in a period exceeding 2 years.

The method of isolation of primary cells and their subculture influences the expression of LDL receptors on pigeon and chicken embryo cells in culture.

Previous studies from this laboratory have shown that pigeon embryo fibroblasts in culture, derived from embryo explants, do not express LDL receptors. In contrast, pigeon peritoneal macrophages and pigeon hepatocytes in culture express LDL receptors. The presence of an active LDL receptor pathway in pigeons has been further confirmed by in vivo studies which showed that receptor-mediated mechanisms were responsible for greater than 80 percent of whole body LDL clearance. The purpose of the present study was to determine the factors responsible for the failure of explant-derived pigeon embryo cells to express LDL receptors. To address this problem, both chicken and pigeon embryo cells were studied. Primary pigeon and chicken embryo cells isolated by collagenase digestion expressed high levels of LDL receptor activity using 125I-labelled pigeon LDL as the ligand. Cells isolated from chicken embryo by enzyme digestion expressed an approximately 10-fold greater LDL receptor activity than cells derived from explants. Pigeon LDL and beta-VLDL, but not methyl-LDL, HDL or mammalian LDL, bound to a limited number of receptors on pigeon embryo cells with specificity and high affinity. Other properties of the pigeon LDL receptor were similar to the mammalian LDL receptor with the exception that the pigeon LDL receptor was resistant to the proteolytic enzyme, pronase. In both pigeon and chicken embryo cells LDL receptor activity was lost with subsequent passage in culture. As a result, the failure of explant derived pigeon cells to express LDL receptors appears to be the result of two factors, the use of explant rather than enzyme-isolated cells and the loss of any remaining LDL receptor activity as a result of passage in culture. Whether this difference between pigeon and mammalian cells with respect to LDL receptor activity is the result of differences in the molecular structure of the pigeon LDL receptor remains to be determined.

Isolation and characterization of attenuated plaque variants of infectious bursal disease virus

Attenuated plaque variants were obtained from infectious bursal disease virus adapted to chick embryo cell cultures. The large plaque (Lp) clone and the small plaque (Sp) clone formed homogeneous plaques about 5 and 1 mm in diameter, respectively. Neutralization tests indicated that these clones differed little from their parent strain in antigenicity. Sp clones showed a retarded growth rate in chick embryo cell cultures as compared with Lp clones. The clones were significantly less pathogenic for chick embryos than the parent strain, although Lp clones were more pathogenic than Sp clones, and they were much less pathogenic for 1-day-old chicks and 28-day-old chickens. Both clones had immunizing potency in 28-day-old chickens, although the Lp clone had a somewhat higher potency than the Sp clone. These findings suggest the Lp and Sp clones, in particular the Lp clones, to be useful as live virus vaccine strains.
