Productivity Scenedesmus sp. in Various Cultivation Media

Scenedesmus sp. is one of the microalgae that is used as natural food for fish seeds or larvae. Scenedesmus sp. contains 8-56% protein, 10-52% carbohydrates, 2-40% fat, and 3-6% nucleic acid. Scenedesmus sp. also contains fatty acids in the form of lauric acid (0.22%), myrsiric acid (0.34%), linolenic acid (16.16%), glycerol trilaurate (3.73%), and vanil laurate (35.52%). Cultivation of Scenedesmus sp. can be done using inorganic fertilizer or waste water media. The types of inorganic fertilizer commonly used for cultivation are urea, KCL, TSP, Intermediate, Raja Bandeng brand. Meanwhile, waste water media can be used for cultivating Scenedesmus sp. namely tofu liquid waste water, milk waste water, bean sprout extract media, textile waste water. The results of cultivation in this media have an influence on the productivity of Scenedesmus sp.

Scenedesmus obliquusmicroalga‐based biorefinery – from brewery effluent to bioactive compounds, biofuels and biofertilizers – aiming at a circular bioeconomy

AbstractThe circular bioeconomy concept relies on the exploitation of wastes as a feedstock of different biotechnological processes to obtain, as much as possible, a huge spectrum of biochemical components through a biorefinery platform. This work deals with the treatment of brewery effluent through the cultivation ofScenedesmus obliquusmicroalga and the use of the biomass in a complex biorefinery. The treatment proved efficient in the removal of nutrients (N, P and COD removals of 88, 30 and 71% respectively). Several compounds and products were obtained from the biomass, such as: (a) phenols (0.249–1.016 mg GAE mL−1) and flavonoids (0.05–0.167 mg CE mL−1) by subcritical water extraction (SWE) at 120 °C, the extraction efficiency being five times higher at 200 °C; (b) biohydrogen by dark fermentation (67.1 mL H2gVS−1); (c) bio‐oil (64%), biochar (30%) and biogas (6%) by pyrolysis; and (d) enhanced capacity of germination/growth of wheat and barley seeds byS. obliquusculture and biomass (pellet, after centrifugation); better results were obtained with the biomass cultivated in brewery effluent (when compared with synthetic medium), and the biomass pellet was better than the whole culture; barley seeds treated with the pellet from the brewery effluent had the highest germination index (GI) of 85 compared with the control (tap water) GI of 35. Cultivation of scenedesmus sp. using optimized minimal nutrients and flocculants – a potential platform for mass cultivation

ABSTRACTA major constraint in the microalgal technology is the economics involved in cultivation and harvesting. This work is focussed on the optimization of nutrients for cultivation and harvesting using ‘Scenedesmus sp’. Response surface methodology (RSM) using ‘Face centered central composite design’ (FCCD) available in Design expert 10.0.4 was used to develop the regression model for optimization of nutrients and flocculation conditions. The optimum nutrient conditions were 500 ppm of urea, 250 ppm of potassium dihydrogen phosphate and 1000 ppm of potassium hydrogen carbonate under artificial light conditions and 500 ppm of urea and 2000 ppm of potassium hydrogen carbonate under sunlight conditions. The optimum conditions were predicted using the model and compared with experimental data. The model has an R2 value of 0.9769 and 0.9798 for artificial light and sunlight conditions, respectively. In the case of harvesting studies, 98% flocculation efficiency was obtained for a combination of pH 10.4, temperature 45°C, 200 mg/l of leaf powder of Cassia auriculata. The model has an R2 value of 0.9989. The present studies indicated that cultivation of Scenedesmus sp. with the optimized nutrients and harvesting conditions facilitate a platform for energy efficient mass cultivation.

Comparison between coagulation-flocculation and ozone-flotation for Scenedesmus microalgal biomolecule recovery and nutrient removal from wastewater in a high-rate algal pond

The removal of nutrients by Scenedesmus sp. in a high-rate algal pond, and subsequent algal separation by coagulation-flocculation or flotation with ozone to recover biomolecules, were evaluated. Cultivation of Scenedesmus sp. in wastewater resulted in complete NH3-H removal, plus 93% total nitrogen and 61% orthophosphate removals. Ozone-flotation obtained better water quality results than coagulation-flocculation for most parameters (NH3-N, NTK, nitrate and nitrite) except orthophosphate. Ozone-flotation, also produced the highest recovery of lipids, carbohydrates and proteins which were 0.32 ± 0.03, 0.33 ± 0.025 and 0.58 ± 0.014 mg/mg of biomass, respectively. In contrast, there was a low lipid extraction of 0.21 mg of lipids/mg of biomass and 0.12–0.23 mg of protein/mg of biomass in the coagulation-flocculation process. In terms of biomolecule recovery and water quality, ozone showed better results than coagulation-flocculation.

Influence of light intensity and tannery wastewater concentration on biomass production and nutrient removal by microalgae Scenedesmus sp.

Abstract Due to the absorption capacity of nutrients by microalgae, their uses for industrial wastewater treatment have shown increasing interest in research. Tannery wastewaters have high organic content and are rich in nitrogenous compounds which are essential for microalgae cultivation. Thus, the aim of this study was to use tannery wastewater as alternative cultivation medium for microalgae biomass production aiming to treat this effluent. Biomass production and removal of ammoniacal nitrogen, phosphorus and chemical oxygen demand by the microalgae Scenedesmus sp., cultivated in tannery wastewater, was studied under different wastewater concentrations (between 20% and 100%), light intensity (between 80 and 200 μmol photons m−2 s−1) at temperature of 25 °C and constant aeration. The results showed that the adaptation of microalgae for this nutrient source was effective. The cultivation of Scenedesmus sp. showed maximum biomass concentration (0.90 g L−1) and maximum removal of ammoniacal nitrogen (85.63%), phosphorus (96.78%) and COD (80.33%) at a tannery wastewater of 88.4% and light intensity of 182.5 μmol photons m−2 s−1.

Identification of fatty aids through cultivation of Scenedesmus sp. from the Caspian Sea

ABSTRACTBiomass concentrations of Scenedesmus sp. with variation of light intensities and nitrogen sources were investigated. At the end of the cultivation period, maximum biomass concentration was recorded under nitrogen deficiency and low illumination (1,000 lux), which was about 3-fold higher than other conditions. However, applying high illumination (6,500 lux) did not significantly boost the biomass concentration. At desired illumination, the fact was revealed that the excessive amount of nitrogen source may waste the chemical and financial resources. The presence of C16:0, C18:1, C18:2, and C18:3 as the major constituents (81.84% of the algal oil) makes Scenedesmus sp. biomass a suitable feedstock for biodiesel production.

Digestion of settleable solids from recirculating fish tank as nutrients source for the microalga Scenedesmus sp. cultivation

The high concentration of nitrogen and phosphorus in wastewater incorporated with the ability to use carbon dioxide as the carbon source make the microalgae become more attractive in wastewater treatment process. This study evaluates the optimal conditions for the digestion of settelable solids from the recirculating aquaculture system to produce the biomass of the green microalga Scenedesmus sp. After solids separation, aerobic digestion of settleable solids under disperse condition produced nitrate as the final product of consequently ammonification and nitrification processes. With the optimal digestion procedure, nitrate concentration during aerobic digestion in 2000 mL vessel increased from 9.63±0.65 mg N/L to 58.66±0.06 mg N/L in 10 days. Thereafter, cultivation of Scenedesmus sp. was performed in 1000 mL Duran bottle with air bubbling. The highest Scenedesmus sp. specific growth rate of 0.321±0.01/d was obtained in treatment using liquid fraction after aerobic digestion as the whole culture medium for Scenedesmus sp. cultivation. With this study, digestion of 8,800±128.12 mg dry weight/L of settleable solids from fish pond finally produced 1,235±21 mg dry weight/L of Scenedesmus sp. biomass.

Wastewater from Instant Noodle Factory as the Whole Nutrients Source for the Microalga Scenedesmus sp. Cultivation

Cultivation of microalgae using wastewater exhibits several advantages such as nutrient removal and the production of high valuable products such as lipid and pigments. With this study, two types of wastewater from instant noodle factory; mixed liquor suspended solids (MLSS) and effluents after sedimentation tank were investigated for green microalga Scenedesmus sp. cultivation under laboratory condition. Optimal wastewater dilution percentage was evaluated in 24 wells microplate. MLSS and effluent without dilution showed the highest specific growth rate (μ) of 1.63±0.11 day－1 and 1.57±0.16 day－1, respectively, in which they were significantly (p < 0.05) higher than Scenedesmus sp. grown in BG11 medium (1.08±0.14 day－1). Ten days experiment was also conducted using 2000 ml Duran bottle as culture vessel under continuous light at approximately 5000 lux intensity and continuous aeration. It was found that maximum biomass density of microalgae cultivated in MLSS and effluent were 344.16 ± 105.60 mg/L and 512.89 ± 86.93 mg/L respectively and there was no significant (p < 0.05) difference on growth to control (BG11 medium). Moreover, cultivation microalgae in wastewater could reduce COD in wastewater by 39.89%-73.37%. Therefore, cultivation of Scenedesmus sp. in wastewater from instant noodle factory can yield microalgae biomass production and wastewater reclamation using photobioreactor simultaneously.

Changes in the biological activity of agropyrene on combined application with humus substances in the cultivation ofScenedesmus obliquus (TURP.) krüger

The work is concerned with the question of the effect of humic acids on the biological action of agropyrene, the effective substance from the rootstock of Agropyron repens (L.) P. BEAUV. The test organism was the alga Scenedesmus obliquus (TURP.) KRÜGER, cultivated in mineral solution. The basic biological test was the determination of the number and size of the cells of this alga. It was found that the way in which agropyrene acts on combined application with humus acids depends on which humus fraction is used. It can display a synergic action if humic acid is used, since this substance probably facilitates the penetration of agropyrene into the cells. On the other hand, fulvic acid counteracts the activity of agropyrene and this most probably by mutual binding and blocking of active groups and links.
