Ligand design for dual enantioselective control in Cu-catalyzed asymmetric conjugate addition of R2Zn to cyclic enone

A new chiral N-heterocyclic carbene (NHC) ligand derived from a natural α-aminoester has been designed and synthesized. The coupling of N-methylbenzimidazole with an α-chloroacetamide derivative, which was prepared from chloroacetyl chloride and (S)-serine methyl ester, gave the corresponding ester/amide-functionalized azolium compound 20. The reaction of 2-cyclohexen-1-one (17) with Et2Zn in the presence of catalytic amounts of Cu(OTf)2 and 20 produced (R)-3-ethylcyclohexanone (18) as a major product. In contrast, the enantioselective conjugate addition (ECA) reaction catalyzed by Cu(OTf)2 under the influence of a hydroxy-amide-functionalized azolium compound 15, which was derived from (S)-tert-leucinol, produced (S)-18 in preference to (R)-18. A series of azolium salts were synthesized from (S)-serine esters, and the reaction conditions for the ECA reaction were optimized to produce (R)-18 with 69% ee. The best results were obtained in the case of the reaction of 4,4-dimethyl-2-cyclohexen-1-one (34) with Et2Zn catalyzed by Cu(OTf)2 in combination with azolium compounds. When the reaction of 34 with Et2Zn was carried out in the presence of catalytic amounts of Cu(OTf)2 and 20, (S)-3-ethyl-4,4-dimethylcyclohexanone (35) was obtained with 97% ee, whereas the ECA reaction under the influence of hydroxy-amide-functionalized azolium 15 afforded (R)-35 with >99% ee. In this manner, the reversal of enantioselectivity was achieved by controlling the structure of chiral ligands.

A new C2-symmetric azolium compound for Cu-catalyzed asymmetric conjugate addition of R 2Zn to cyclic enone

A new chiral N-heterocyclic carbene (NHC) ligand was designed. Thus, an efficient synthetic route to C 2-symmetric bis(hydroxyamide)-functionalized benzimidazolium salts from chiral β-amino alcohols was developed. The combination of Cu(OTf) 2 and the chiral azolium compound efficiently promoted the conjugate addition reaction of cyclic enone with dialkylzinc to give the corresponding adduct in good yield. Among a series of chiral NHC proligands, the functionalized benzimidazolium chloride possessing a tert-butyl group as a stereodirecting group was found to be the best choice of ligand. Under optimized reaction conditions, an excellent enantioselectivity (96% ee) was realized by allowing 2-cyclohepten-1-one to react with Bu 2Zn at room temperature.

Reversal of Stereoselectivity in the Cu-Catalyzed Conjugate Addition Reaction of Dialkylzinc to Cyclic Enone in the Presence of a Chiral Azolium Compound

Reversal of enantioselectivity in a Cu-catalyzed asymmetric conjugate addition reaction of dialkylzinc to cyclic enone with use of the same chiral ligand was successfully achieved. The reaction of 2-cyclohexen-1-one (30) with Et(2)Zn catalyzed by Cu(OTf)(2) in the presence of an azolium salt derived from a chiral beta-amino alcohol gave (S)-3-ethylcyclohexanone (31) in good enantioselectivity. Among a series of chiral azolium compounds examined, the benzimidazolium salt (10) having both a tert-butyl group at the stereogenic center and a benzyl substituent at the azolium ring was found to be the best choice of ligand in the Cu(OTf)(2)-catalyzed reaction. Good enantioselectivity was observed when the reaction was conducted by employing a benzimidazolium derivative rather than an imidazolium derivative. The influence of the substituent at the azolium ring on the stereoselectivity of the reaction was also examined. In addition, from the results of the reaction catalyzed by Cu(OTf)(2) combined with an azolium compound derived from (S)-leucine methyl ester, it was found that the hydroxy side chain in the chiral ligand is probably crucial for the enantiocontrol of the conjugate addition reaction. On the other hand, it was discovered from a screening test of copper species that the reversal of enantioselectivity was realized by allowing 30 to react with Et(2)Zn in the presence of Cu(acac)(2) combined with the same ligand precursor to afford (R)-31 as a major product. The influence of the stereodirecting group at the chiral ligand on the stereoselectivity in the Cu(acac)(2)-catalyzed reaction differed completely from that observed in the Cu(OTf)(2)-catalyzed reaction. Reaction with a cyclic enone consisting of a seven-membered ring such as 2-cyclohepten-1-one (40) resulted in increasing the enantioselectivity of the reaction. Thus, treatment of 40 with Et(2)Zn catalyzed by Cu(OTf)(2) combined with a benzimidazolium salt produced the corresponding (S)-conjugate adduct in a 92:8 enantiomer ratio (er), while the Cu(acac)(2)-catalyzed reaction with the same ligand afforded (R)-product in a 9:91 er.

Synthesis of Chiral Bulky Monophosphite Ligands and Their Application in Cu-Catalyzed Asymmetric Conjugate Addition of Diethylzinc to Cyclic Enones

A series of new bulky monophosphite ligands were synthesized from axially chiral BINOL and highly sterically hindered adamantanecarbonyl chloride. These ligands were applied to the Cu-catalyzed asymmetric 1,4-conjugate addition of diethylzinc to cyclic enones and were found to have moderate enantioselectivities (up to 61% ee). The stereochemically matched combination of (S)-BINOL in ligand (S)-(2-(adamantane-1-carboxylic acid)-1,1′-binapthalen-2′-yl)-((S)-1,1′-binaphthalen-2,2′-yl)phosphite induces a moderate enantioselectivity. The sense of enantioselectivity is mainly controlled by the configuration of the 2,2′-o, o-(1,1′-binaphthyl)-dioxo-phosphite moiety of the ligands.

Enantioselective Total Synthesis of Clavirolide C. Applications of Cu-Catalyzed Asymmetric Conjugate Additions and Ru-Catalyzed Ring-Closing Metathesis

The first enantioselective total synthesis of clavirolide C, a member of the dolabellane family of diterpenes isolated from Pacific soft coral Clavularia viridis, is disclosed. The total synthesis features the application of chiral amino acid based ligands in Cu-catalyzed asymmetric conjugate addition (ACA) reactions and a relatively rare application of catalytic ring-closing metathesis to access an 11-membered ring structure. The total synthesis effort has spawned the development of a new protocol for NHC.Cu-catalyzed ACA of alkylaluminum reagents to beta-substituted cycloalkenones. The enantioselective clavirolide C synthesis requires 17 steps (longest linear sequence), affords the target molecule in 3.5% overall yield, and confirms the stereochemical assignment for the natural product.

Cu-Catalyzed Asymmetric Conjugate Additions of Dialkyl- and Diarylzinc Reagents to Acyclic β-Silyl-α,β-unsaturated Ketones. Synthesis of Allylsilanes in High Diastereo- and Enantiomeric Purity

A readily available and simple (MW = 444.5 g/mol) valine-based chiral phosphine is used to promote highly efficient catalytic asymmetric conjugate additions of dialkyl- and diarylzinc reagents to acyclic beta-silyl-alpha,beta-unsaturated ketones. The catalytic asymmetric protocol allows access to versatile allylsilanes that bear a trisubstituted olefin in high diastereo- and enantiomeric purity.

Catalytic Enantioselective Alkylations of Tetrasubstituted Olefins. Synthesis of All-Carbon Quaternary Stereogenic Centers through Cu-Catalyzed Asymmetric Conjugate Additions of Alkylzinc Reagents to Enones

A method for Cu-catalyzed asymmetric conjugate addition (ACA) of dialkylzinc reagents to tetrasubstituted five- and six-membered cyclic enones that afford quaternary all-carbon stereogenic centers in up to 95% ee is reported. Catalytic ACAs are practical and efficient. Reactions proceed to >98% conversion in undistilled commercial grade toluene in the presence of 2 mol % of an air-stable Cu salt (CuCN) and a readily available chiral ligand. Enantioselective ACA reactions deliver products that can be readily functionalized to afford a variety of synthetically versatile compounds in high optical purity.

P-Chiral o-Phosphinophenol as a P/O Hybrid Ligand: Preparation and Use in Cu-Catalyzed Asymmetric Conjugate Addition of Diethylzinc to Acyclic Enones

[reaction: see text] (S)-2-(tert-Butylmethylphosphino)phenol and its methyl ether were synthesized from tert-butyldichlorophosphine via optically active phosphine-boranes as the intermediates. The former compound was used as a P/O hybrid ligand in the Cu-catalyzed asymmetric conjugate addition of diethylzinc to acyclic enones to achieve high enantioselectivity of up to 96%.

Enantioselective Synthesis of Nitroalkanes Bearing All-Carbon Quaternary Stereogenic Centers through Cu-Catalyzed Asymmetric Conjugate Additions

The first examples of catalytic asymmetric conjugate addition (ACA) of alkylzinc reagents to trisubstituted nitroalkenes, leading to the formation of nitroalkanes bearing a quaternary carbon stereogenic center, are reported. Reactions are promoted in the presence of 4 mol % of a readily available amino acid-based phosphine and 2 mol % (CuOTf).C6H6. Cu-catalyzed reactions proceed efficiently in up to 98% ee and can be carried out with a variety of dialkylzinc reagents and trisubstituted nitroolefins. We highlight the synthetic utility of the products obtained by efficient conversion of optically enriched nitroalkanes to the corresponding carboxylic acids.

Efficient Cu-catalyzed asymmetric conjugate additions of alkylzinc reagents to aromatic and aliphatic acyclic nitroalkenes.

An efficient and highly enantioselective (up to 95% ee) Cu-catalyzed method for asymmetric conjugate addition (ACA) of alkylzinc reagents to acyclic disubstituted nitroalkenes is presented. Reactions are typically effected at ambient temperature in the presence of 2 mol % chiral dipeptide phosphine and 1 mol % (CuOTf)(2).C(6)H(6). Biphenol-based ligands for Cu-catalyzed asymmetric conjugate addition

The synthesis and characterization of seven new phosphoramidite ligands are described, as well as their use in copper-catalyzed 1,4-addition of diethylzinc to a wide range of substrates. Enantioselectivities of up to >99.5% are obtained.

New chiral amino-phosphoramidite and bisphosphoramidite ligands derived from ( R, R)-1,2-diaminocyclohexane: application in Cu-catalyzed asymmetric conjugate addition of diethylzinc to 2-cyclohexenone

New bidentate amino-phosphoroamidite and diphosphoroamidite ligands derived from inexpensive ( R, R)-1,2-diaminocyclohexane have been synthesized and screened in the Cu-catalyzed asymmetric conjugate addition of Et 2Zn to 2-cyclohexenone. The highest 74% ee value was reached with the N, N′-dimethyl substituted P, N-ligand and Cu(OAc) 2·H 2O.

Efficient cu-catalyzed asymmetric conjugate additions of alkylzincs to trisubstituted cyclic enones.

The first examples of efficient catalytic asymmetric conjugate addition (ACA) of alkylzincs to trisubstituted cyclic enones is disclosed. These Cu-catalyzed reactions proceed efficiently with five- and seven-membered ring substrates to afford the desired products in >/=95% ee. Intermediate enolates can be trapped with alkyl halides to generate a quaternary stereogenic center. The requisite chiral ligand is prepared from commercially available materials and can be used in situ without further purification in the presence of commercial grade (CuOTf)2.PhMe.

Cu-catalyzed asymmetric conjugate additions of alkylzinc reagents to acyclic aliphatic enones.

Cu-catalyzed enantioselective conjugate additions to acyclic aliphatic enones are reported. The resulting enolates may be functionalized intra- and intermolecularly, leading to the formation of an additional C-C bond. The utility of the present method is not limited to reactions involving Et2Zn; a variety of alkylzincs may be used. Moreover, many of the requisite substrates can be easily accessed through catalytic olefin cross metathesis.
