Bamboo Charcoal as Adsorbent for SPE Coupled with Monolithic Column-HPLC for Rapid Determination of 16 Polycyclic Aromatic Hydrocarbons in Water Samples

The coupling of solid-phase extraction (SPE) using bamboo charcoal (BC) as an adsorbent with a monolithic column-high performance liquid chromatography (MC-HPLC) method was developed for the high-efficiency enrichment and rapid determination of 16 polycyclic aromatic hydrocarbons (PAHs) in water. Key influence factors, such as the type and the volume of the elution solvent, and the flow rate and the volume of the sample loading, were optimized to obtain a high SPE recovery and extraction efficiency. BC as an SPE adsorbent presented a high extraction efficiency due to its large specific surface area and high adsorption capacity; MC as an HPLC column accelerated the separation within 8 min because of its high porosity, fast mass transfer, and low-pressure resistance. The calibration curves for the PAHs extracted were linear in the range of 0.2-15 µg/L, with the correlation coefficients (r(2)) between 0.9970-0.9999. This method attained good precisions (relative standard deviation, RSD) from 3.5 to 10.9% for the standard PAHs I aqueous solutions at 5 µg/L; the method recoveries ranged in 52.6-121.6% for real spiked river water samples with 0.4 and 4 µg/L. The limits of detection (LODs, S/N = 3) of the method were determined from 11 and 87 ng/L. The developed method was demonstrated to be applicable for the rapid and sensitive determination of 16 PAHs in real environmental water samples.

LC/MS analysis of stratum corneum lipids: ceramide profiling and discovery

Ceramides (CERs) in the upper layer of the skin, the stratum corneum (SC), play a key role in the skin barrier function. In human SC, the literature currently reports 11 CER subclasses that have been identified. In this paper, a novel quick and robust LC/MS method is presented that allows the separation and analysis of all known human SC CER subclasses using only limited sample preparation. Besides all 11 known and identified subclasses, a 3D multi-mass chromatogram shows the presence of other lipid subclasses. Using LC/MS/MS with an ion trap (IT) system, a Fourier transform-ion cyclotron resonance system, and a triple quadrupole system, we were able to identify one of these lipid subclasses as a new CER subclass: the ester-linked ω-hydroxy fatty acid with a dihydrosphingosine base (CER [EOdS]). Besides the identification of a new CER subclass, this paper also describes the applicability and robustness of the developed LC/MS method by analyzing three (biological) SC samples: SC from human dermatomed skin, human SC obtained by tape stripping, and SC from full-thickness skin explants. All three biological samples showed all known CER subclasses and slight differences were observed in CER profile.

Analytical platform for verification and quantitation of target peptides in human serum: Application to cathelicidin

A selective and sensitive, fully automated platform for verification and quantitative determination of target peptides in biofluids is proposed and then validated by development of a method for analysis of cathelicidin in human serum. The method is based on the on-line coupling of solid-phase extraction (SPE) and tandem mass spectrometry with direct infusion. Mass spectrometry analysis was carried out by multiple reaction monitoring using three transitions (one for quantitative analysis and two for qualitative analysis), all them confirmed by in silico fragmentation of the target peptide. Samples were prepared in the SPE workstation on a polymeric divinylbenzene resin by preconcentration, deproteinization, and cleanup, removing salts and interferences after direct injection of human serum. The analytical process required 12 min. The limits of detection and quantitation were 2.5 and 8.25 μg/L, respectively (0.20 and 0.66 pg on column). Repeatability and within-laboratory reproducibility were 2.4% and 2.7%, respectively. A dual-cartridge configuration was used to test recovery of cathelicidin in serum, resulting in 80%. Because quantitative retention in the cartridge was assessed, determination of cathelicidin was validated without using synthetic peptides labeled with stable isotopes. The hyphenated system allows full automation, thereby improving reproducibility and accuracy, as demanded by clinical analysis.

Pesticide dissipation curves in peach, pear and tomato crops in Uruguay*

Dissipation curves of azoxystrobin and of the neonicotinoids acetamiprid and thiacloprid in peach; azinphos-methyl and carbaryl in pear and azoxystrobin, chlorfenapyr and chlorpyrifos in high-tunnel tomato crops were studied in the Southern region of Uruguay. An analytical methodology based on solid phase extraction (SPE) and detection by High Performance Liquid Chromatography with Diode Array Detector (HPLC/DAD) was used for acetamiprid and thiacloprid. Coupled SPE and detection by Gas Chromatography with Mass Selective Detector (GC/MSD) was used for the detection of azinphos-methyl, azoxystrobin, carbaryl, chlorfenapyr and chlorpyrifos residues. Curves were modeled mathematically with Solver program of Microsoft Excel®. The best fit for acetamiprid and thiacloprid in peach was achieved with the exponential model (r2=0.961 and 0.944, respectively). In the case of peach fruits there is not a Maximum Residue Limit (MRL) for acetamiprid in the Codex Alimentarius, while 0.5 mg/kg is the value rated for thiacloprid. The MRLs accepted by the European Union (EU) are 0.1 mg/kg for acetamiprid and 0.3 mg/kg for thiacloprid. According to the curves determined in these experiments, thiacloprid residues 10 to 12 days after application (daa) were below the MRLs established by both sources. In the case of acetamiprid, 25 daa would be required, according to the exponential mathematical model, to get residues levels below the MRL values established by the EU. For azinphos methyl in pear, the residues detected were mathematically fitted to an exponential model (r2=0.999). According to it, residue levels under the MRL established by the EU (0.05 mg/kg) are gotten in our conditions in 20 daa. In plastic tunnel tomato chlorfenapyr residues were not detected from 16 daa, having the dissipation curve an exponential trend. In the same condition, there was not a decay of the azoxystrobin concentration during a 24-day trial, being it around 0.40 ± 0.05 mg/kg.

Room temperature ionic liquid‐mediated molecularly imprinted polymer monolith for the selective recognition of quinolones in pork samples

A novel molecularly imprinted polymer monolith was prepared by the room temperature ionic liquid-mediated in situ molecular imprinting technique, using norfloxacin (NOR) as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker. The optimal synthesis conditions and recognition properties of NOR-imprinted monolithic column were investigated. The results indicated that the imprinted monoliths exhibited good ability of selective recognition against the template and its structural analog. Using the fabricated material as solid-phase extraction sorbent, a sample pre-treatment procedure of molecularly imprinted solid-phase extraction coupling with HPLC was developed for determination of trace quinolone residues in animal tissues samples. The recoveries ranging from 78.16 to 93.50% for eight quinolones antibiotics such as marbofloxacin, NOR, ciprofloxacin, danofloxacin, difloxacin, oxolinic acid, flumequine and enrofloxacin were obtained.

On-line Coupling of Solid-Phase Extraction to Liquid Chromatography--A Review

Solid-phase extraction (SPE) is an effective sample preparation method for removal of interfering compound and enrichment of analyte. Liquid chromatography (LC) with various detectors is a main separation and detection technique used in the analytical field. This article reviews the literatures about the on-line coupling of SPE with LC. The advantages of on-line coupling are reduction of analysis time, sample contamination, and analyte losses, as well as improvement of precision and accuracy. The SPE sorbents including traditional materials, such as chemically bonded silica, ion-exchange and carbon-based materials, and some novel sorbents, such as restricted access material, molecularly imprinted polymer, immunosorbent, and monolithic material, used in the on-line analysis are discussed in detail. The on-line coupling of SPE-LC to other sample preparation techniques, such as supercritical fluid extraction, subcritical water extraction, microwave-assisted extraction, ultrasound-assisted extraction, and derivatization technique are also reviewed.

Detection of cocaine and its metabolites in urine using solid phase extraction-ion mobility spectrometry with alternating least squares

A reliable, alternative screening method for detection of cocaine and its metabolites, benzoylecgonine and cocaethylene in urine is demonstrated using solid phase extraction (SPE) coupled with ion mobility spectrometry (IMS). Data analysis with alternating least squares (ALS) is used to model IMS spectral datasets and separate the reactant ion peak from the product ion peaks. IMS has been used as a screening device for drug and explosive detection for many years. It has the advantages of atmospheric pressure operation, simple sample preparation, portability, fast analysis, and high sensitivity when compared to similar methods. Coupling SPE with IMS decreases the detection limits of drug metabolites in urine while removing salts and other polar compounds that suppress ionization during the measurement. The IMS analysis time in this experiment is 20 s, much shorter than traditional chromatographic analysis. The application of ALS further increases the sensitivity and selectivity of this method. The detection limits of benzoylecgonine and cocaethylene are 10 ng/mL and 4 ng/mL, respectively. Commercial adulteration of urine specimens does not influence the ability to detect cocaine metabolites after sampling the urine with SPE. This method provides forensic chemists a viable approach for fast and simple drug screening.

On-line coupling of solid-phase extraction to liquid chromatography–tandem mass spectrometry for the determination of macrolide antibiotics in environmental water

An automated on-line solid-phase extraction–liquid chromatography–tandem mass spectrometry (SPE–LC–MS/MS) system was developed for the determination of macrolide antibiotics including erythromycin (ETM), roxithromycin (RTM), tylosin (TLS) and tilmicosin (TMC) in environmental water samples. A Capcell Pak MF Ph-1 column packed with restricted access material (RAM) was used as SPE column for the concentration of the analytes and clean-up of the sample. One milliliter water sample was injected into the conditioned SPE column and the matrix was washed out with 3 mL high purity water. By rotation of the switching valve, macrolides (MLs) were eluted in the back-flush mode and transferred to the analytical column by the chromatographic mobile phase. The matrix effect was evaluated by the directly injection LC–MS and on-line SPE–LC–MS methods. The limits of detection (LODs) and limits of quantification (LOQs) obtained are in the range of 2–6 and 7–20 ng L −1, respectively, which means that the proposed method is suitable for trace analysis of MLs at low level concentration. The intra- and inter-day precisions are in the range of 2.9–7.2% and 3.3–8.9%, respectively. In the three fortified levels (20, 200 and 2000 ng L −1), recoveries of MLs ranging from 86.5% to 98.3% are obtained.

Evaluation of a molecularly imprinted polymer as in‐line concentrator in capillary electrophoresis

Molecularly imprinted polymers (MIPs) have been evaluated as sorbent for the construction of an in-line solid phase extraction concentrator in capillary electrophoresis to be applied in the monitoring of triazine herbicides: atrazine and its three metabolites, desethylatrazine, desisopropylatrazine and desethyldesisopropylatrazine. Initially, the electrophoretic separation of these compounds was optimized. The electrolyte consists of an aqueous solution of 75 mM phosphoric acid (H(3)PO(4)) adjusted to pH 2.1 and containing 0.7 mM cetyltrimethylammonium bromide. After the fabrication and assembly of the concentrator into the capillary, these optimal CE conditions were applied to evaluate the performance of this device. Efficiencies of 40 000-55 000 plates could be achieved and the separation time was around one hour. Different parameters affecting the in-line molecularly imprinted solid-phase extraction in capillary electrophoresis such as composition and volume of the elution plug were optimized. The method was evaluated in terms of linearity, precision and limits of detection and quantification. MIPs were compared with Oasis hydrophilic-lipophilic-balance (HLB) particles for the in-line coupling of solid-phase extraction and capillary electrophoresis. The superior selectivity of MIPs is demonstrated through direct injection of a urine sample spiked with 10 microg/mL atrazine, desethylatrazine, desisopropylatrazine and desethyldesisopropylatrazine. Recoveries were between 92 and 102% compared with an aqueous solution.

On-line solid phase extraction coupled to flame atomic absorption spectrometry for the determination of trace copper and zinc in environmental and biological samples

In this work, bamboo charcoal (BC) was used as a sorbent for on-line solid phase extraction (SPE) coupling with flame atomic absorption spectrometry (AAS) for trace copper and zinc determination in environmental and biological samples. Under the optimum pH of 5.5 (for Zn) and 7.0 (for Cu), trace copper and zinc were effectively adsorbed on the microcolumn and the retained cations were efficiently eluted with HCl or HNO3 with an appropriate concentration and flow rate for on-line AAS determination. With a sample loading time of 60 s at a sample flow rate of 7.6 mL min−1, the enhancement factors of 39 (for Cu) and 30 (for Zn) and detection limits (3σ) of 0.60 µg L−1 (for Cu) and 0.36 µg L−1 (for Zn), respectively, were achieved. The sample throughput was 45 h−1. At the level of 20 µg L−1of Cu(II) and Zn(II), the precision (RSD, n = 11) were found to be 0.26% and 1.6%, respectively. The proposed method has been successfully applied to the determination of copper and zinc in environmental and biological samples.

On-line coupling of solid-phase extraction to high-performance liquid chromatography for determination of estrogens in environment

A method based on on-line solid-phase extraction (SPE) coupling to high-performance liquid chromatography (HPLC) for the determination of estrogens has been developed. This method can continuously perform extraction of estrone, estradiol, estriol and diethylstilbestrol from aqueous samples without any other pretreatment, which can then be analyzed by HPLC with a UV detector at 230 nm. A pre-concentration column was adapted with methanol/water for chromatographic separation and two kinds of sorbents were involved, which are octadecyl-bonded silica and cigarette filter. The condition of pH of samples, sample loading flow rate and desorption time were all optimized, and the performances of both two sorbents were satisfactory. The on-line SPE system requires very low maintenance and just involved a switching-valve-filter system and a flow-inject pump, and the operation of the whole SPE-HPLC instrumentation is quite simple. The detection limits for pre-concentrating 50 mL of standard solution using cigarette filter as sorbent ranged from 0.98 to 78.1 ng L −1. The enhancement factors were in the range of 197–326. The recoveries of estrogens spiked in real water samples ranged from 85 to 112%. The precisions for nine replicate measurements of a standard mixture (5.0 μg L −1) were in the range of 1.0–3.4%.

On-line coupling of solid-phase extraction, derivatization reaction and spectrophotometry by sequential injection analysis: Application to trifluoperazine assay in human urine

Introduction: This study deals with a new methodology of on-line solid-phase extraction (SPE), derivatization reaction and spectrophotometric measurement utilizing sequential injection analysis (SIA) technique. The method was applied for the assay of trifluoperazine in human urine. Methods: An SPE procedure was automated to perform sample clean-up, extraction and preconcentration into a homemade microcolumn connected to a SIA manifold. Unlike previous SIA methods, a simple SIA manifold included one syringe pump and one multi-position valve was constructed. This offered simplicity and rapidity to the proposed method (sampling frequency 11 sample h − 1 ). Spectrophotometric measurement was based on a fast oxidation of trifluoperazine by Ce(IV) in sulfuric acidic media resulting in a spectrophotometrically detectable chromophore measured at wavelength 500 nm. The SPE procedure was optimized by the univariate method while the derivatization reaction and spectrophotometric measurement were optimized by multivariate methods. Results: The method was linear in a range of 70–200 ng ml − 1 and accurate with a recovery of 92.7%. Relative standard deviation of repeatability ( n = 10 in a day) and intermediate precision ( n = 5 over a week) did not exceed 4.3% indicating satisfactory precision. The limits of detection and quantification were 18.2 and 55.2 ng ml − 1 , respectively. The sensitivity of the method was improved by the preconcentration, the use of extended pathlength in the detection device and the optimization of absorbance measurement. The method is selective in the presence of chlordiazepoxide, which is sometimes taken in combination with trifluoperazine. Discussion: The method is suitable for the application of overdose and therapeutic drug monitoring in human urine.

Analysis of steroids in songbird plasma and brain by coupling solid phase extraction to radioimmunoassay

It is a common practice to extract steroids from plasma, serum, or tissue samples prior to steroid measurement by radioimmunoassay (RIA) or enzyme immunoassay (EIA). Steroid extraction is critical because it can remove substances that interfere with the RIA or EIA. Steroid extraction is commonly achieved using organic solvents, such as diethyl ether or dichloromethane. However, organic solvent extractions can suffer from low recovery, imprecise recovery, or incomplete removal of assay interference. Here, we describe validations of a simple protocol to extract steroids (e.g., dehydroepiandrosterone, corticosterone, and estradiol) from avian plasma, serum, and brain tissue using solid phase extraction (SPE) with commercially available C 18 columns. We compare various methods for (1) eluting steroids from columns, (2) drying eluates, and (3) resuspending dried eluates prior to RIA. The SPE method yields high and consistent recoveries. The SPE method also effectively separates steroids from interfering substances, even when extracting steroids from lipid-rich plasma and brain tissue. These data indicate that SPE is superior to organic solvent extraction on several measures. SPE should be broadly useful for extracting steroids from plasma or tissue samples.

Recent advances in coupling solid-phase extraction and capillary electrophoresis (SPE–CE)

Solid-phase extraction (SPE) is a good strategy for overcoming the low sensitivity of capillary electrophoresis (CE). Here, we present a critical review of applications of coupling between SPE and CE reported between 2000 and March 2007. We highlight selected developments and advances and focus on various combinations of these two techniques, which include at-line, in-line and on-line modes of SPE–CE. We discuss the advantages and the drawbacks of the different set-ups and methodologies. We also outline novel trends in SPE–CE coupling.

On-line coupling of solid-phase extraction and capillary electrophoresis for the determination of cefoperazone and ceftiofur in plasma

We present a method for determining two cephalosporins (cefoperazone and ceftiofur) in plasma by on-line solid-phase extraction (SPE)—capillary zone electrophoresis (CZE) with a T-split interface. Using this interface, a part of the SPE elution plug containing the cephalosporins is injected while the rest of the sample is flushed to waste. SPE was carried out using a C 18 micro-precolumn and the cephalosporins presented good retention properties with breakthrough volumes above 1 ml. Using a desorption volume of 426 nl of acetonitrile, recoveries were 75 and 90%, for cefoperazone and ceftiofur, respectively. The resulting elution volume was about 1.8 μl. A deproteinization step was included prior to SPE for the analysis of plasma samples with recoveries of 90 and 57% for cefoperazone and ceftiofur, respectively. With UV detection at 254 nm, linear relationships between the injected concentration and peak area was measured between 10 and 500 ng ml −1 for standards, and 200 and 1500 ng ml −1 for plasma samples. Intra-day ( n = 5) and inter-day ( n = 5) peak area repeatability were lower than 12% RSD. The detection limits obtained for spiked plasma (100 ng ml −1 cefoperazone and ceftiofur) are sufficient for applying the method to pharmacokinetic studies.

Flow-injection interface for on-line coupling solid-phase extraction and X-ray fluorescence measurements

A flow-injection (FI) manifold including a solid-phase extraction (SPE) mini-column has been coupled on-line to an energy-dispersive X-ray fluorescence detector (EDXRFD) by locating a lab-modified 18-μL flow-cell, 10-mm path length, connected to the SPE column by PTFE tubing of 0.5 mm i.d., in the X-ray spectrometer chamber. The optical window of the flow-cell was adjusted and fixed to the X-ray irradiation zone of the spectrometer. Two PTFE tubes connected the flow-cell to the FI device and were introduced into the spectrometer chamber by a small orifice without distortion nor modification of the instrument. The SPE–EDXRFD coupling was tested for Pb and Cd aqueous solutions using Dowex 50 cation-exchange resin as sorbent, and flushing the eluate through the flow-cell for monitoring. The LODs and LOQs thus obtained were 1 and 3.2 μg for Pb and 1.8 and 4.8 μg for Cd, respectively; values which allow using the approach for the analysis of waste water by injecting 20 mL of sample into the FI manifold. The linear dynamic ranges are a function of the sample volume circulated through the mini-column. For a sample volume of 20 mL the ranges are between 1 and 4000 μg for Pb and between 1.8 and 2000 μg for Cd. The method was validated by the standard addition method using ground-water samples. The SPE–EDXRFD coupling enables to carry out the study of those variables influencing the SPE process – namely, the effect of the sample volume flushed through the column, concentration of analytes in the sample, amount of resin packed, breakthrough volume of the resin, elution profiles, sample pH and retention and elution flow-rates – in an automatic, cheap, fast and precise way.

Integrated multilayer microfluidic device with a nanoporous membrane interconnect for online coupling of solid-phase extraction to microchip electrophoresis

An integrated microfluidic device was developed for online coupling of solid-phase extraction to microchip electrophoresis (chip SPE-CE). With a nanoporous membrane sandwiched between two PDMS substrates, SPE preconcentration and electrophoretic separation can be carried out in upper and lower fluidic layers, separately and sequentially. During the SPE process, the thin membrane can act as a fluid isolator to prevent intermixing between two fluidic channels. However, when a pulse voltage is applied, the membrane becomes a gateable interconnect so that a small plug of concentrated analytes can be online injected into the lower channel for subsequent separations. This multilayer design provides a universal solution to online SPE-CE hyphenation. Both electroosmotic flow and hydrodynamic pumps have been adopted for SPE operation. SPE was performed on a 2.5 mm long microcolumn, with two weirs on both sides to retain the C(18)-coated silica beads. Rhodamine 123 and FITC-labelled ephedrine were used to test the operational performance of the hyphenation system. High separation efficiency and thousand-fold signal enhancement were achieved.

Establishing the feasibility of coupled solid-phase extraction–solid-phase derivatization for acidic herbicides

The significance of this research is that it improves analytical methodology used for organic chemicals in aqueous solutions by establishing the feasibility of heterogeneous chemical derivatization at the liquid–solid interface (i.e., solid-phase reaction or solid-phase derivatization). A solid-phase derivatization method for determining chlorinated herbicide acids was developed. Solid-phase extraction was used to concentrate and retain analytes on sorbents for subsequent solid-phase derivatization. Background interferences were removed from the chromatograms by electronically subtracting the responses of blank, nonfortified analyses from spiked samples. Two extraction sorbents (octadecyl bonded silica and polystyrene-divinylbenzene) and two derivatizing reagents (BF3–MeOH and trimethylsilyldiazomethane) were investigated. Recovery of 13 chlorinated herbicide acids—including pentachlorophenol, dinoseb, and bentazon (having a derivatizable functional group, OH or NH, bonded directly to a phenyl group); dicamba, picloram, acifluorfen, 3,5-dichlorobenzoic acid, and dacthal (having a derivatizable functional group, –COOH, bonded directly to a phenyl group); and 2,4-D, 2,4,5-T, dichlorprop, 2,4,5-TP, and 2,4-DB (having a derivatizable functional group, COOH, bonded directly to a sp3 carbon atom)—was tested. The analytical method developed was proven successful for determining acidic herbicides, except for the dacthal diacid metabolite, in aqueous samples.

PEPPeR, a Platform for Experimental Proteomic Pattern Recognition

Quantitative proteomics holds considerable promise for elucidation of basic biology and for clinical biomarker discovery. However, it has been difficult to fulfill this promise due to over-reliance on identification-based quantitative methods and problems associated with chromatographic separation reproducibility. Here we describe new algorithms termed "Landmark Matching" and "Peak Matching" that greatly reduce these problems. Landmark Matching performs time base-independent propagation of peptide identities onto accurate mass LC-MS features in a way that leverages historical data derived from disparate data acquisition strategies. Peak Matching builds upon Landmark Matching by recognizing identical molecular species across multiple LC-MS experiments in an identity-independent fashion by clustering. We have bundled these algorithms together with other algorithms, data acquisition strategies, and experimental designs to create a Platform for Experimental Proteomic Pattern Recognition (PEPPeR). These developments enable use of established statistical tools previously limited to microarray analysis for treatment of proteomics data. We demonstrate that the proposed platform can be calibrated across 2.5 orders of magnitude and can perform robust quantification of ratios in both simple and complex mixtures with good precision and error characteristics across multiple sample preparations. We also demonstrate de novo marker discovery based on statistical significance of unidentified accurate mass components that changed between two mixtures. These markers were subsequently identified by accurate mass-driven MS/MS acquisition and demonstrated to be contaminant proteins associated with known proteins whose concentrations were designed to change between the two mixtures. These results have provided a real world validation of the platform for marker discovery.

Analysis of hormonal steroids in fish plasma and bile by coupling solid-phase extraction to GC/MS

An analytical procedure for the simultaneous determination of twelve endogenous steroids (testosterone, androstenedione, 17beta-estradiol, estrone, pregnenolone, progesterone, dihydroandrostenedione, dihydrotestosterone, 11alpha-ketotestosterone, 17alpha-hydroxyprogesterone, 17alpha-hydroxypregnenolone, 17alpha,20beta-dihydroxy-4-pregnen-3-one) in plasma and bile samples by solid-phase extraction (SPE) and gas chromatography/mass spectrometry (GC-MS) has been developed. After enzymatic hydrolysis for bile samples only, samples were concentrated and purified using two successive SPE (C(18) and NH(2)) cartridges. Analytes were derivatized with a mixture of N-methyl-N-(trimethylsilyl)trifluoroacetamide (MSTFA) / mercaptoethanol / ammonium iodide (NH(4)I) and determined by GC-MS in selective ion monitoring mode. For most of the steroids monitored, recoveries were in the range 90-120% in plasma and in the range 60-70% in bile, and the reproducibility was below 10% for the complete procedure. Limits of detection obtained ranged from 0.1 to 0.4 ng/g in fish plasma and from 1.6 to 14 ng/g in fish bile. The developed method was successfully applied to the determination of plasma steroids in flounders (Platichthys flesus) collected from two French estuaries.
