Immunohistochemical expression of Ki-67 in gall bladder carcinoma

Introduction: Gall bladder carcinoma accounts for 0.3-0.7% of all cancers and has a peak incidence among women aged 70 and 79 years. Recently many molecular markers like Ki-67, cyclin D1, MUC1 and p53 with prognostic impact on GBC have been described. Ki-67 is a proliferative marker which is expressed in all active phases of the cell cycle and it can be detected in the nucleus of tumour cells indicating its proliferative activity. The aim of this study is to evaluate the expression of Ki-67 in carcinoma of gall bladder by immunohistochemistry and to correlate the mean Ki-67 labelling index with the various histological grades of gallbladder carcinoma. Materials and Methods: All the histopathologically proven cases of gall bladder malignancy from July 2014 to June 2017 (n=44) were examined for Ki-67 expression immunohistochemically.Ki-67 labelling index was calculated. ANOVA test and independent ‘t’ test were applied to see correlation of Ki-67 expression with various grades of gall bladder carcinoma. Results: The Ki-67 labelling index was minimum in well-differentiated adenocarcinoma of gall bladder and maximum in poorly differentiated adenocarcinoma of gall bladder. The p value amongst these groups was found to be statistically significantly different. Conclusion: 88.64% of Gall bladder carcinoma was positive for Ki-67 and the Ki-67 labelling index increased significantly as the histological grade increased. Adenoma pleomórfico de glándula lagrimal: localización atípica de un tumor salival

Introduction: In daily ophthalmological practice, lacrimal gland tumors are rare. They represent 5 to 7.5% of all intraorbital tumors. The most common epithelial tumor of this gland is the pleomorphic adenoma, with a percentage of 25 to 50% of its tumor lesions. Objective: In this communication we presented a case of lacrimal gland pleomorphic adenoma and study the expression of Ki67 and the location and expression of MUC-1 and its correlation with tumor prognosis. We also carried out a retrospective descriptive study of the literature on the subject published between 1951 and 2020, using the MEDLINE database. Material and methods: The surgical piece examined, was processed according to the paraffin embedding technique, was performed with a histopathological diagnosis of pleomorphic lacrimal gland adenoma. Histological sections were stained with Hematoxylin / Eosin. Immunostaining with Ki67 and MUC-1 was performed with the DAKO LSAB + kit. Results: The diagnosis of hypercellular pleomorphic adenoma with areas of the remaining lacrimal gland was made. Ki67 labeling was low (≤15%). MUC-1 expression was intense, situated to the apical cell membrane of approximately 10% epitheliocytes from pseudoductal and cystic structures.Conclusions: Through histopathological evaluation, the correlation of Ki67 expression and the location and expression of MUC1, we verified that it is a non-recurrent pleomorphic adenoma without malignant transformation.

Correlation of Ki-67 Expression as Tumor cell Proliferation Activity Marker with Cutaneous Squamous Cell Carcinoma Grading.

Cutaneous Squamous Cell Carcinoma (cSCC) is a malignant keratinocyte tumour that develops through the suprabasal epidermis. This malignant tumour is the second most common skin malignancy after Basal Cell Carcinoma (BCC). The increased incidence of cSCC is directly proportional to increasing age. Generally, the predisposing factor of cSCC is exposure to recurrent sunlight for a long time, so localisation of cSCC is a part of the body that often exposed to direct sunlight, such as the forehead, face, ears, scalp, neck, and back of the hand. The carcinogenesis process of cSCC is a cumulation of a series of events, one of which plays an important role is the proliferation index assessed by Ki-67. Forty-eight tissue paraffin blocks were diagnosed histopathologically as cutaneous squamous cell carcinoma from the Anatomical Pathology Laboratory of the Faculty of Medicine, Universitas Sumatera Utara and the Anatomical Pathology Unit of Haji Adam Malik General Hospital Medan, as the research sample. The results of protein expression from Ki-67 were assessed based on area. There was no significant correlation between cSCC grading and Ki-67 expression (p > 0.05). Ki-67 antigen tumour marker, widely used to determine the level of tumour cell proliferation.

Expression of Ki-67 in Adult and Children Acute Lymphoblastic Leukemia and Its Clinical Significance

To investigate the relationship of Ki-67 level with clinical features, immunophenotype, gene mutation, curative efficacy and prognosis in patients with acute lymphoblastic leukemia(ALL). Flow cytometry gated at CD45/SSC was used to detect the expression of Ki-67, and the correlation of Ki-67 expression with clinical manifestation, laboratorial indexes, curative efficacy and prognosis was analysed. Ki-67 expression level increased in ALL patients, the median expression rate was 29.22%, there was significant difference as compared with the healthy control (P<0.01). In adult ALL, the median expression rate of Ki-67 in the high-risk group was 31.49%, and the difference was statistically significant as compared with the low-risk group (P<0.05). In children ALL, the median expression rate of Ki-67 in high-risk group was 42.28%, and the difference was statistically significant (P<0.05). The results of unvariate analysis showed that the age, WBC count at newly diagnosed and extramedullary invasion were adverse factors affecting OS and DFS; the results of multivariate analysis showed that age and extramedullary invasion were independent risk factors for OS and DFS in patients. Age≥14 years old, intramedullary invasion are the poor factors for prognosis; the Ki-67 level is not an independent factor for the prognosis of patients.

Ki67 expression in triple negative breast cancer: Correlation of Ki67 expression with other prognostic factors in breast cancer in Indian patients

Introduction: Triple negative breast cancer ( TNBC) cases are not sensitive to hormonal therapy and till date no specific targeted medication has been found for TNBC. The present study aims to know the expression of Ki67 in TNBC tissues and in non-TNBC tissues using immunohistochemistry and to do correlation analysis of ki 67 expression with other clinicopathological parameters. Material and Methods: The present study was conducted on 160 cases of breast cancer received as specimens from January to august 2016 in the Pathology Department, Sri Guru Ram Das institute of medical sciences, Amritsar. Histopathological typing and grading was done followed by immunohistochemistry for ER, PR, Her2neu and ki67. Results: Ki67-positive expression were identified in 86.6% TNBC cases (including 14 cases of strong Ki67 expression). By contrast, among 130 cases of non-TNBC, 98 cases were Ki67-positive, accounting for 75.3%,(only 2 cases showed strong positivity) indicating a significant difference compared with that in the TNBC group that Ki67 proliferation index was higher in the cases with size >2cm also cases with lymph node metastasis showed high Ki67 proliferation index. Conclusion: Ki-67 index is relatively higher in TNBC than in non-TNBC cases. Ki67 expression correlated with tumor size and lymph node metastasis in breast cancer in the present study. Thus the increased expression of Ki67 may predict the increased proliferation of breast cancer cells, increased invasiveness, faste r rate of growth and the high incidence of lymph node metastasis.

Correlation of Ki67 Expression with Hormone Receptors, Human Epidermal Growth Factor Receptor-2 (HER-2) Status, P53 Mutation and Clinicopathological Characteristics in Pathologic Specimens of Breast Cancer Patients

Background: Breast cancer is the main cause of cancer in women and the second cause of ma­lignancy deaths. Ki-67 is one of the molecular markers used to evaluate cancer prognosis along with other factors such as age, tumor size, lymph node involvement, estrogen receptor (ER), progesterone receptor (PR), P53, human epidermal growth factor receptor-2 (HER-2), histolog­ical and nuclear grades. This study was aimed to evaluate the correlation of KI-67 expression with some biomarkers and clinico-pathological characteristics in breast cancer patients. Mate­rials and Methods: A total of 513 cases (all female) aged 40- 80 years, were randomly selected from patients who were admitted in two centers affiliated with Tehran University of Medical Sciences (Buo-alli and Kasra hospitals) over a 7-year period (2010-2015). Assessment of tumors for HER-2, P53, ER PR, pathological type and histologic grade was performed. Ki-67 labelling index (Ki-67LI) was defined as the percentage of MIB1-positive cells among a total number of 1,000 malignant cells at high-power magnification (×400). Results: Our study showed that age, ER and PR status were negatively correlated with Ki-67LI (P&lt;0.05). Moreover, number of lymph nodes involved, HER-2, P53 and nuclear grades had a positive correlation with Ki-67LI (P&lt;0.05), whereas, tumor size and histological grade showed no significant correlation with Ki-67LI (P =0.195 and P=0.721, respectively). Conclusion: Results of our study and other studies confirm that the expression of Ki-67 is significantly associated with ER, PR, HER-2 and P53 status. On the other hand, Ki-67 relationship with clinical characteristics such as age, tumor size and lymph node metastasis is not completely established and needs further research.[GMJ.2016;5(2):90-97]

Radically resected pituitary adenomas: prognostic role of Ki 67 labeling index in a monocentric retrospective series and literature review

Ki-67 Labeling Index is an immunocytochemical marker of cell proliferation. The correlation of Ki-67 expression with pituitary adenomas recurrence has been investigated and is highly debated. Aim of this study was to evaluate whether Ki-67 correlates with recurrence even in patients with an apparently completely removed pituitary adenoma. We retrospectively reviewed the database of the Hypothalamic-Pituitary Disease Unit at the Catholic University of Rome, collected between 2003 and 2011. Inclusion criteria were: patients who underwent surgery at the Department of Neurosurgery with an apparently complete removal of a pituitary adenoma; Ki-67 histological evaluation by the same operator and values of <3%. All patients underwent endocrine evaluation of the hypothalamic-pituitary function, ophthalmologic and neuro-radiological examinations, during the preoperative period and follow-up. Out of 490 patients recorded on the database of the Hypothalamic-Pituitary Disease Unit at the Catholic University of Rome, 191 cases met the inclusion criteria. Recurrence was observed in 49 cases (25.7% of the patients who had undergone radical excision). Optional cut-off value was identified at Ki-67 values of 1.50%. This was associated with worse disease-free survival time, even after correction for age at treatment, gender, positivity to p53, functional classification and Knosp grading. Ki-67 labeling index may be useful in postoperative management, even in patients who underwent radical PA removal. We suggest a Ki-67 cut-off value of 1.5% to plan an adequate clinical follow-up.

P5-11-12: Correlation of Ki67 Expression between Initial Biopsy and Surgical Specimen in Untreated Breast Cancer Patients. Does Menstrual Cycle Matter?

Abstract Background: The measure of proliferative index by Ki67 expression is being increasingly used to identify breast cancer subtype and to guide treatment decisions. Findings on initial tumor biopsy are in some cases, such as for primary systemic treatment, the only data available prior to treatment. In addition, results from window studies to develop novel anti-tumor agents depends on initial breast cancer biopsy findings. This study aims to compare the Ki67 expression on initial biopsy with matched primary breast cancer surgical specimens. In addtion, we aimed to explore the possible effect of hormonal levels — as reflected by menstrual cycle phase — on discordant Ki67 cases. Material and Methods: A total of 50 random cases were analyzed by 2 blinded pathologists. Ki67 expression was collected as a continuous variable. Tumors were then classified as high — or low-proliferative according to a Ki67 value of &amp;gt; or = 15% or &amp;lt; 15%, respectively. Sperman Rank test was used to study the correlation between initial and surgical biopsies. Data on menstrual cycle phase at the time of biopsy and surgery were retrived from medical history of all premenopausal patients. Results: Baseline patients characteristics are as follows: median age 48 (35-56 years), premenopausal 27 (54%), postmenopausal 23 (46%), invasive ductal carcinoma 38 (76%). A differential expression of Ki67 was found in 36 patients (72%), 20 pre- and 16 post-menopausal. Spearman's Rank Order correlation did not show any significant difference between biopsy and matched surgical specimen in terms of Ki67 expression. Indeed, there was a strong positive correlation between initial and surgical biopsy, which was statistically significant across all patients (rs = .556, P &amp;lt; 0.001), and regardless the menopausal status. However, 15% of pre-menopausal patients transformed from Ki67 low to high. Therefore, data on pre-menopausal patients were analyzed according to the phase of menstrual cycle at the timing of initial and surgical biopsies. The Spearman's Rank Order gave no correlation among 14 premenopausal patients with differential menstrual cycle phase at the timing of initial biopsy and surgery (rs = .411, P = 0.145). Conclusions: Initial breast cancer biopsy can be used with high confidence for Ki67 determination. However, it should be noted that menstrual cycle phase could affect Ki67 expression as women with distinct menstrual cycle phase at the time of initial biopsy and final surgery do not show concordant results. Correlation of Ki-67 expression in cutaneous primary melanoma with prognosis in a prospective study: Different correlation according to thickness

Background: The proliferative activity of melanoma cells, evaluated as MIB-1 immunoreactivity against a Ki-67 epitope, has been considered an indicator of poor prognosis in thick primary melanomas. Objective: The aim of the study was to assess the correlation between the growth fraction of primary melanomas, assessed by means of Ki-67 immunoreactivity, and metastatic relapse. Methods: Ki-67 reactivity in primary melanomas was evaluated as at least 5% of positive neoplastic cells and was assessed on fresh specimens of 55 primary lesions at the time of excision. Such reactivity was correlated with metastatic relapse of patients in a prospective study, by means of multivariate Cox regression models (follow-up, 3-120 months). Results: Ki-67 immunoreactivity was associated with increasing thickness (P = .003). Positive correlation was found between Ki-67 reactivity and metastatic dissemination in primary melanomas less than 1.5 mm thick (n = 23; mean thickness, 0.75 ± 0.3 mm; P = .002). On the contrary, a negative correlation was found between Ki-67 reactivity and metastatic activity in primary melanomas thicker than 1.5 mm (n = 32; mean thickness, 4.0 ± 1.6 mm; P = .019). Conclusion: Ki-67 proliferative activity appears to be a possible predictor of metastasis in primary melanomas, in particular, an indicator of poor prognosis in lesions less than 1.5 mm thick.
