Investigation of controlled salmeterol xinafoate and fluticasone propionate release from double molecular imprinted nanoparticles

AbstractSalmeterol xinafoate (SAM) and fluticasone propionate (FLU) are one of the drug combinations used together in the treatment of lung diseases such as asthma and chronic obstructive pulmonary disease (COPD). The aim of this study is to investigate the usability of novel dual molecular imprinted nanoparticles (poly(2-hydroxyethyl methacrylate-N-methacryloyl-(L)-alanine-N-methacryloyl-(L)-histidine) [p(HEMA-MAAL-MAH)], abbr. DMIPNPs) as a controlled drug release systems. In this study, SAM and FLU drugs were chosen as model drugs because they are used in the treatment of these diseases. DMIPNPs were prepared by surfactant-free emulsion polymerization method and characterized by scanning electron microscopy (SEM) and fourier transform infrared spectrometer (FTIR). In in vitro drug release experiments, drug release conditions were optimized. SAM and FLU release from DMIPNPs experiments were also performed in the simulated lung fluid (SLF). The amount of released SAM and FLU were found as 4.79 and 5.68 mg/g in the SLF medium at the end of 48 h, respectively. The release kinetics of SAM and FLU from DMIPNPs were calculated in the SLF medium. The release of SAM and FLU was determined to be compatible with the Higuchi release models. According to these results, these DMIPNPs, dual-template molecular imprinted nanoparticles with dual monomers, are promising materials that can be used in the controlled release of two different drugs. Chitosan-coated liposome with lysozyme-responsive properties for on-demand release of levofloxacin

As an anti-infection antibiotic delivery route, a drug-controlled release system based on a specific condition stimulus response can enhance drug stability and bioavailability, reduce antibiotic resistance, achieve on-demand release and improve targeting and utilization efficiency. In this study, chitosan-coated liposomes containing levofloxacin (Lef@Lip@CS) were prepared with lysozyme in body fluids serving as an intelligent “switch” to enable accurate delivery of antibiotics through the catalytic degradation ability of chitosan. Good liposome encapsulation efficacy (64.89 ± 1.86 %) and loading capacity (5.28 ± 0.18 %) were achieved. The controlled-release behavior and morphological characterization before and after enzymatic hydrolysis confirmed that the levofloxacin release rate depended on the lysozyme concentration and the degrees of deacetylation of chitosan. In vitro bacteriostatic experiments showed significant differences in the effects of Lef@Lip@CS before and after enzyme addition, with 6-h inhibition rate of 72.46 % and 100 %, and biofilm removal rates of 51 % and 71 %, respectively. These findings show that chitosan-coated liposomes are a feasible drug delivery system responsive to lysozyme stimulation.

Application and prospect of novel nanomedicine in cancer therapy

With the rapid development of nanotechnology, the application of new nanodrugs in cancer treatment has gradually become a research hotspot. In this paper, we explore the design principles, preparation methods and their anticancer effects in vitro and in vivo, and explore potential clinical applications. The results show that nanomedicine improve the therapeutic effect and reduce the side effects by achieving accurate delivery and controlled release of drugs, providing a new solution for cancer treatment. In terms of design principles, nanomdrugs mainly make use of unique properties such as small size effects, surface effects and quantum size effects of nanomaterials to combine drug molecules with nanocarriers to achieve precise delivery and controlled release of drugs. In terms of preparation methods, physical, chemical and biological methods are used to prepare nanoscale drugs with different morphology and structures. In vitro and in vivo experiments showed that nanomedicines can significantly increase the concentration of the drug in the tumor cells, enhance the therapeutic efficacy, and reduce the toxicity to normal tissues. However, nanomedicine still face many challenges in the field of cancer therapy. Looking ahead, with the continuous advances in nanotechnology, it is reasonable to believe that nanomedicine will play a greater role in the field of cancer therapy. Through in-depth study of the mechanism of action of nanomedicine, optimizing the preparation process, improving the precision of the controlled drug release system, and focusing on the biological safety issues, we are expected to bring safer and more effective treatment methods for cancer patients. At the same time, the combined application of nanomedicine and other therapeutic means such as immunotherapy and gene therapy will also become a research hotspot in the future, providing more comprehensive and personalized treatment options for cancer treatment.

Natural and Synthetic Polymers for Biomedical and Environmental Applications.

Natural and synthetic polymers are a versatile platform for developing biomaterials in the biomedical and environmental fields. Natural polymers are organic compounds that are found in nature. The most common natural polymers include polysaccharides, such as alginate, hyaluronic acid, and starch, proteins, e.g., collagen, silk, and fibrin, and bacterial polyesters. Natural polymers have already been applied in numerous sectors, such as carriers for drug delivery, tissue engineering, stem cell morphogenesis, wound healing, regenerative medicine, food packaging, etc. Various synthetic polymers, including poly(lactic acid), poly(acrylic acid), poly(vinyl alcohol), polyethylene glycol, etc., are biocompatible and biodegradable; therefore, they are studied and applied in controlled drug release systems, nano-carriers, tissue engineering, dispersion of bacterial biofilms, gene delivery systems, bio-ink in 3D-printing, textiles in medicine, agriculture, heavy metals removal, and food packaging. In the following review, recent advancements in polymer chemistry, which enable the imparting of specific biomedical functions of polymers, will be discussed in detail, including antiviral, anticancer, and antimicrobial activities. This work contains the authors' experimental contributions to biomedical and environmental polymer applications. This review is a vast overview of natural and synthetic polymers used in biomedical and environmental fields, polymer synthesis, and isolation methods, critically assessessing their advantages, limitations, and prospects.

In vitro inhibition of uterine contractions using electrospun nanofibers loaded with nifedipine and ML7.

To formulate a nanofiber-based controlled drug delivery system that could be effective in preventing uterine contractions and can be used for the treatment of preterm labor. We utilized uterine tissue samples obtained from ten pregnant women who underwent cesarean section at term to investigate the effect of nanofibers on spontaneous and induced myometrial contractions. We prepared nifedipine and ML7-loaded nanofibers using the electrospinning method with Poly(D,L-lactide-co-glycolide) (PLGA) polymer, resulted in seven groups of nanofibers, including a control group. Group I served as the control, Group II was non-drug loaded nanofiber, Group III was nifedipine (10-5 M) loaded nanofiber, Group IV was ML7 (3x10-5 M) loaded nanofiber, Group V was ML7 (3x10-5 M) and nifedipine (10-5 M) nanofiber, Group VI was ML7 (3x10-5 M) and nifedipine (3x10-5 M) nanofiber, and Group VII was ML7 (3x10-5 M) and nifedipine (10-4 M) nanofiber. To evaluate the contractile response, the nanofibers loaded with different doses of ML7 and nifedipine were applied onto the tissue strips, and in vitro organ bath experiments were performed. Full-thickness uterine samples were cleared of the serosa and surrounding tissues, and eight strips (3x10 mm) were prepared from each sample. The seven different nanofiber formulations were gently placed and sutured onto the strips, with one strip always kept as the time control. We recorded spontaneous, KCl-induced, and stimulated cumulative oxytocin-induced contractions from all samples in all groups. After completing all experiments, the viability of the strips was checked, and weight measurement was recorded. The administration of drug-loaded polymers resulted in a significant decrease in both the frequency and intensity of spontaneous and induced contractions in all groups (p<0.01). No significant difference was observed between the control group and the non-drug-loaded nanofiber group in post hoc analysis (p=0.704). In terms of amplitude and frequency of contractions, the most significant decrease was observed in group VII at cumulative oxytocin doses compared to the control and non-drug-loaded nanofiber groups (p<0.05). Moreover, group VI also showed a significant decrease in contraction intensity and frequency compared to the control and non-drug-loaded nanofiber groups (p<0.05). While the use of nifedipine and/or ML7-loaded nanofibers decreased both intensity and frequency of contraction, this attenuation was not significant compared to the control and empty polymer groups. However, a more significant inhibition was observed when ML7 was used with nifedipine at doses of 3x10-5 M and 10-4 M. The results indicate that human uterine contractions can be inhibited using calcium channel blocker (nifedipine) and myosin light chain kinase inhibitor (ML7) loaded nanofibers in uterine tissue strips. These results strongly suggested the potential for the development of locally effective and safe controlled drug release systems to prevent premature birth.

Ibuprofen-Loaded Silver Nanoparticle-Doped PVA Gels: Green Synthesis, In Vitro Cytotoxicity, and Antibacterial Analyses.

In contrast to conventional drug delivery systems, controlled drug release systems employ distinct methodologies. These systems facilitate the release of active substances in predetermined quantities and for specified durations. Polymer hydrogels have gained prominence in controlled drug delivery because of their unique swelling-shrinkage behavior and ability to regulate drug release. In this investigation, films with a hydrogel structure were crafted using polyvinyl alcohol, a biocompatible polymer, and silver nanoparticles. Following characterization, ibuprofen was loaded into the hydrogels to evaluate their drug release capacity. The particle sizes of silver nanoparticles synthesized using a green approach were determined. This study comprehensively examined the structural properties, morphological features, mechanical strength, and cumulative release patterns of the prepared films. In vitro cytotoxicity analysis was employed to assess the cell viability of drug-loaded hydrogel films, and their antibacterial effects were examined. The results indicated that hydrogel films containing 5% and 10% polyvinyl alcohol released 89% and 97% of the loaded drug, respectively, by day 14. The release kinetics fits the Korsmeyer-Peppas model. This study, which describes nanoparticle-enhanced polyvinyl alcohol hydrogel systems prepared through a cost-effective and environmentally friendly approach, is anticipated to contribute to the existing literature and serve as a foundational study for future research.

Posaconazole Loading and Release Behavior in Surface-Modified Mesoporous Silica Nanoparticular System

In conventional drug treatments, high toxic effects, low solubility, and low bioavailability of the active substance cause insufficient drug release in the target tissue and undesirable side effects in healthy tissue. Various drug delivery systems are utilized to eliminate these undesirable effects. Mesoporous silica nanoparticles (MSN) are biocompatible biomaterials that have a large surface area, high pore volume, and enhanced adsorption capacity. With MSN-mediated controlled drug release, the active substance concentration in the blood is kept within the desired therapeutic range. Posaconazole (PCZ) is an antifungal agent. Absorption of PCZ is difficult due to its low solubility in aqueous and acidic environments, low therapeutic effect and low bioavailability. The use of controlled drug release systems avoids these problems and facilitates the absorption and release of PCZ. In this study, it is aimed to enhance the PCZ adsorption and release by using a drug delivery system. MSNs were synthesized by sol-gel method, and surface modification of nanoparticles was achieved using (3-Aminopropyl) triethoxysilane (APTES). PCZ was loaded on APTES-modified MSN successfully. MSN, APTES-modified MSN and PCZ loaded APTES-modified MSN were characterized. Diffusion controlled release of PCZ was observed in drug release studies.

Hydrogels are used in the treatment of soft tissue damage, controlled drug release systems, tissue/organ production with 3D bioprinters, smart material production, and many other tissue engineering studies. Although polymers obtained from natural polymers or synthetically produced polymers are used in hydrogel production, they may have various biocompatibility problems. In this study, Pericardial fluid structure (PFS) was used to increase the biocompatibility of the alginate and was used in the production of bioink for use in 3D bioprinters. PFS is a structure isolated from pericardial fluid (PF) and consists of complex components that are very similar to natural Extracellular Matrix (ECM) both morphologically and in content. According to the results of SEM images, the collagen-elastin fiber network was clearly observed in the groups with PFS added, since PFS contains high levels of collagen and elastin proteins. It was concluded that the biocompatibility of the material was also increased thanks to the structure similar to the natural ECM in the alginate hydrogels with PFS added.

Preparation of a novel alginate hydrogel microspheres covered by hollow silica for controlled-release application

Sodium alginate hydrogel has been widely used in the field of controlled-release. In this paper, in order to prevent the negative effect of fast degradation and explosive release of pure sodium alginate hydrogel in the drug delivery process, we developed a novel alginate hydrogel supported by hollow silica microsphere for drug delivery. Briefly, the drugs were fused with sodium alginate and the hollow silica microspheres followed by ion crosslinking to form a composite hydrogel microsphere with a core–shell structure (SA@SiO2). The structure and morphology of the composite hydrogel were characterized by Fourier infrared spectroscopy (FT-IR), thermogravimetric analysis (TG), scanning electron microscope (SEM), transmission electron microscope (TEM) along with fluorescence microscope. On this basis, using three different types of organic compounds (rhodamine B, Indocyanine green and salicylic acid) as model molecules, the controlled-release performance of the SA@SiO2 microspheres were investigated in detail. The research results show that the drug delivery system has broad-spectrum controlled-release performance for different types of molecules, and the controlled-release time can exceed 40 h. At the same time, the pH- responsive property of the sodium alginate hydrogel endows the delivery system with a certain targeting function. This study provides a new strategy for constructing an efficient targeted drug controlled-release system.

Self‐Powered Electrically Controlled Drug Release Systems Based on Nanogenerator

AbstractPrecision medicine requires precise regulation of drugs in terms of time, space, and dosage. Exogenous control systems, such as electrical responsiveness, have made great progress. However, wearable or implantable controlled drug release devices still face major challenges due to limitations including limited battery life, large size, and fixed power supply. To overcome these limitations, the fabrication of autonomous devices is available to endure extended periods without reliance on external power sources. As a promising strategy, nanogenerators (NGs) turn body mechanical energy into electricity, powering long‐term drug release. In this review, the current status of drug delivery systems (DDS) is briefly outlined and the importance of self‐driven controlled drug release systems is emphasized. The main types and operational mechanisms of various nanogenerators are introduced. This review also focuses on summarizing the latest progress of self‐powered controlled drug release systems based on nanogenerators (NG‐based CDRs). Additionally, their applications in the field of drug release are introduced in detail. Finally, the existing challenges and future trends of self‐powered NG‐based CDRs are discussed from the perspectives of clinical needs and practical translation.

Biomedical Approach of Nanotechnology and Biological Risks: A Mini-Review

Nanotechnology has played a prominent role in biomedical engineering, offering innovative approaches to numerous treatments. Notable advances have been observed in the development of medical devices, contributing to the advancement of modern medicine. This article briefly discusses key applications of nanotechnology in tissue engineering, controlled drug release systems, biosensors and monitoring, and imaging and diagnosis. The particular emphasis on this theme will result in a better understanding, selection, and technical approach to nanomaterials for biomedical purposes, including biological risks, security, and biocompatibility criteria.

Cellulose-Silk Fibroin Composite Particles for Hydrophilic Drug-Controlled Release System: Preparation and Characterization

The objective of this study was to investigate the dissolution of cellulose (C) powder as a potential material for particle formation when combined with a silk fibroin (SF) solution. Each polymer solution as well as a mixture of cellulose and silk fibroin in different ratios were prepared into particles using the solvent emulsion-diffusion method. The particle morphology was characterized by scanning electron microscopy (SEM) and by attenuated reflection Fourier-transformed infrared spectroscopy (ATR-FTIR) and thermogravimetric analyzer (TGA) for the C-SF molecular interaction and thermal properties, respectively. The results showed that the cellulose particles appeared as separate fibers and loosely bound together, while silk fibroin particles were spherical in shape with dense texture and smooth surface. The C/SF mixed particles with different ratios could be formed into spherical particles and a ratio of 1:3 has the most spherical and solid. The ATR-FTIR results indicated the absorption spectra of the main functional groups both cellulose and silk fibroin. Blue dextran helped to enhance the temperature of the maximum decomposition rate (Td,max) of the particles via hydrogen bond interaction between the blue dextran and polymers. A burst release of blue dextran was observed in the first 12 h due to the swelling of particle surfaces and the slowest release found in the silk fibroin particles. The C/SF composite showed different blue dextran- eleased profiles and the particle with a high content of silk fibroin-released blue dextran in slowly rate. The results of this work can be used as a basis for applying C/SF composite particles in a hydrophilic drug release-controlled system.

Surface Engineering of Bioactive Coatings for Improved Stent Hemocompatibility: A Comprehensive Review.

Cardiovascular diseases continue to be a major contributor to illness and death on a global scale, and the implementation of stents has given rise to a revolutionary transformation in the field of interventional cardiology. The thrombotic and restenosis complications associated with stent implantation pose ongoing challenges. In recent years, bioactive coatings have emerged as a promising strategy to enhance stent hemocompatibility and reduce thrombogenicity. This review article provides an overview of the surface engineering techniques employed to improve the hemocompatibility of stents and reduce thrombus formation. It explores the mechanisms underlying thrombosis and discusses the factors influencing platelet activation and fibrin formation on stent surfaces. Various bioactive coatings, including anticoagulant agents, antiplatelet agents, and surface modifications, are discussed in detail, highlighting their potential in reducing thrombogenicity. This article also highlights a multitude of surface modification techniques which can be harnessed to enhance stent hemocompatibility including plasma treatment, physical vapor deposition (PVD), chemical vapor deposition (CVD), and electrodeposition. These techniques offer precise control over surface properties such as roughness, charge, and composition. The ultimate goal is to reduce platelet adhesion, tailor wettability, or facilitate the controlled release of bioactive agents. Evaluation methods for assessing hemocompatibility and thrombogenicity are also reviewed, ranging from in vitro assays to animal models. Recent advances in the field, such as nanotechnology-based coatings and bioactive coatings with controlled drug release systems, are highlighted. Surface engineering of bioactive coatings holds great promise for enhancing the long-term outcomes of stent implantation by enhancing hemocompatibility and reducing thrombogenicity. Future research directions and potential clinical applications are discussed, underscoring the need for continued advancements in this field.

Prolonged Anesthesia Effects of Locally Administered Ropivacaine via Electrospun Poly(caprolactone) Fibrous Membranes.

Prolonged analgesia is important to safeguard the patient's comfort and safety during and after surgery in clinical practice. To meet the demand for prolonged analgesia, medical professionals often resort to increasing drug frequency, which may lead to poor patient compliance and serious complications due to drug overdose. Therefore, it is of great interest to develop controlled-release drug delivery systems for local anesthetics, enabling slow and controlled drug release to prolong the analgesic effect and minimize systemic toxicity. In this study, we utilized an electrospinning technique to fabricate nonwoven poly(caprolactone) (PCL) fibrous membranes loaded with Ropivacaine and performed proof-of-principle experiments on both in vitro drug release tests and in vivo animal tests, to further prolong the analgesic effect of Ropivacaine and improve postoperative local pain management and chronic pain treatment. Material characterization and in vitro drug release studies confirmed the feasibility of the Ropivacaine-loaded PCL fibrous membranes for sustained release. The drug loading content and drug loading efficiency of Ropivacaine-loaded fibrous membrane are 8.7 ± 0.3 wt% and 96 ± 3 wt%, respectively. Evaluation in an animal model demonstrated prolonged anesthesia effects along with excellent biocompatibility and stability. At 72 h, the cumulative release accounted for approximately 50% of the drug loading content. This study offers novel approaches and strategies for clinical postoperative pain management and chronic pain treatment, while providing new insights and directions for the design of local anesthetic controlled-release delivery systems.

A Comprehensive Review on Role of Polymers in Transdermal Drug Delivery System

Transdermal drug delivery system has been emerged as one of the most effective controlled drug release system in topical formulation as it doesn’t undergo first pass metabolism in liver and can be directly absorbed into the systemic circulation. Polymers are used in TDDS as to increase its effectiveness in the control release of drug from the transdermal patch. Use of polymers in the TDDS has decreased the control release rate of drug from the patch. Use of polymers in combination has given more effectiveness of the drug release. This article gives a quality of information about the use of polymers in the Transdermal drug delivery system. Polymers also help in the adhesive nature of the drug to the skin and promote more extent of drug release from the dosage form. An advance in the use and amount of polymers used in the TDDS has been increased recently. This article provides the information about the use of Natural, semisynthetic and synthetic polymers in the TDDS. Use of natural polymers has been showed more impact in the TDDS and their use has showed fewer side effects like irritation, allergy, and this has been increased patient compliance and more effectiveness in drug release. Semisynthetic and synthetic polymers has been developed and used excessively in the TDDS for effective control release of the drug from the patches. A transdermal patch is defined as medicated adhesive patch which is placed above the skin to deliver a pre-determined dose of medication via skin with a predetermined rate of release to reach into the bloodstream. Future Opportunities in the Field of Drug Delivery Research

Much of the priorities in drug delivery research are focused on targeted drug delivery for cancer therapies and a wide range of controlled drug release systems for commonly used active pharmaceutical ingredients (APIs). In this “thousand words article” we highlight some of the emerging health threats and future opportunities for drug delivery research. Important emerging health threats include viral pandemics beyond COVID, antibiotic-resistant pathogens, the need for new antifungal therapies, and emerging diseases caused by increasing pollution and climate change. Fundamentally new drugs may be needed. For example, one little known research effort focuses on the development of new antibiotics based on metal-organic frameworks. Finally, new delivery approaches will be needed. This is illustrated by the development of a topical peptide delivery system as a wound dressing for burn patients, combining biotechnology (a new peptide) with polymer science (a new topical delivery system) to address a medical need (burn injury) for which there is currently no effective treatment. Another important trend is the shift in our collective understanding of impact, moving away from “counting papers” to considering the societal benefit of the research including its potential for commercialization. To remain relevant in the coming decade, we need to anticipate and embrace future challenges. This is particularly important for younger scientists.

Dual-Targeted Nanoparticle-in-Microparticle System for Ulcerative Colitis Therapy.

Conventional oral therapy for ulcerative colitis (UC) is associated with premature release or degradation of drugs in the harsh gastrointestinal environment, resulting in reduced therapeutic effectiveness. Consequently, the present study aims to develop a dual-targeted delivery system with a nanoparticle-in-microparticle (nano-in-micro) structure. The prepared Asiatic Acid-loaded delivery system (AA/CDM-BT-ALG) has pH-sensitive properties. Cellular uptake evaluation confirms that nanoparticles exhibit targeted absorption by macrophages and Caco-2 cells through mannose (Man) receptor and biotin-mediated endocytosis, respectively. Therefore, this mechanism effectively enhances intracellular drug concentration. Additionally, the biodistribution study conducted on the gastrointestinal tract of mice indicates that the colon of the microspheres group shows higher fluorescence intensity with longer duration than the other groups. This finding indicates that the microspheres exhibit selective accumulation in areas of colon inflammation. In vivo experiments in colitis mice showed that AA/CDM-BT-ALG significantly alleviates the histopathological characteristics of the colon, reduced neutrophil, and macrophage infiltration, and decreases pro-inflammatory cytokine expression. Furthermore, the effect of AA/CDM-BT-ALG on colitis is validated to be closely related to the TLR4/MyD88/NF-κB signaling pathway. The present findings suggest that the development of a dual-targeted delivery system is accomplished effectively, with the potential to serve as a drug-controlled release system for treatingUC.

Intravitreal implants manufactured by supercritical foaming for treating retinal diseases

Chronic retinal diseases, such as age-related macular degeneration (AMD), are a major cause of global visual impairment. However, current treatment methods involving repetitive intravitreal injections pose financial and health burdens for patients. The development of controlled drug release systems, particularly for biological drugs, is still an unmet need in prolonging drug release within the vitreous chamber. To address this, green supercritical carbon dioxide (scCO2) foaming technology was employed to manufacture porous poly(lactic-co-glycolic acid) (PLGA)-based intravitreal implants loaded with dexamethasone. The desired implant dimensions were achieved through 3D printing of customised moulds. By varying the depressurisation rates during the foaming process, implants with different porosities and dexamethasone release rates were successfully obtained. These implants demonstrated controlled drug release for up to four months, surpassing the performance of previously developed implants. In view of the positive results obtained, a pilot study was conducted using the monoclonal antibody bevacizumab to explore the feasibility of this technology for preparing intraocular implants loaded with biologic drug molecules. Overall, this study presents a greener and more sustainable alternative to conventional implant manufacturing techniques, particularly suited for drugs that are susceptible to degradation under harsh conditions.

Application of Calcium Carbonate as a Controlled Release Carrier for Therapeutic Drugs

A drug carrier usually refers to a tool that can carry the effective ingredients of drugs into the human body. The drug-controlled release system prepared by a new drug carrier can allow the gradual release of the drug in the human body at a stable rate, thus decreasing the frequency of administration and reducing the toxicity and side effects thereof; however, existing drug carriers generally have problems such as low drug loading, poor biocompatibility, stability, and specificity, each of which could be improved. Calcium carbonate can be used as a sustained-release carrier of active substances, with good biocompatibility, biodegradability, low cost, easy preparation, and broad application prospects. This paper reviews the synthesis and structural characteristics of calcium carbonate carrier materials and the related research progress of calcium carbonate as a controlled release carrier for therapeutic drugs, providing a reference for promoting the research and application of calcium carbonate as a drug carrier.
