Printed Electrochemical Biosensors: Opportunities and Metrological Challenges.

Printed electrochemical biosensors have recently gained increasing relevance in fields ranging from basic research to home-based point-of-care. Thus, they represent a unique opportunity to enable low-cost, fast, non-invasive and/or continuous monitoring of cells and biomolecules, exploiting their electrical properties. Printing technologies represent powerful tools to combine simpler and more customizable fabrication of biosensors with high resolution, miniaturization and integration with more complex microfluidic and electronics systems. The metrological aspects of those biosensors, such as sensitivity, repeatability and stability, represent very challenging aspects that are required for the assessment of the sensor itself. This review provides an overview of the opportunities of printed electrochemical biosensors in terms of transducing principles, metrological characteristics and the enlargement of the application field. A critical discussion on metrological challenges is then provided, deepening our understanding of the most promising trends in order to overcome them: printed nanostructures to improve the limit of detection, sensitivity and repeatability; printing strategies to improve organic biosensor integration in biological environments; emerging printing methods for non-conventional substrates; microfluidic dispensing to improve repeatability. Finally, an up-to-date analysis of the most recent examples of printed electrochemical biosensors for the main classes of target analytes (live cells, nucleic acids, proteins, metabolites and electrolytes) is reported.

Impedance-based drug-resistance characterization of colon cancer cells through real-time cell culture monitoring

Interest in impedance-based cellular assays is rising due to their remarkable advantages, including label-free, low cost, non-invasive, non-destructive, quantitative and real-time monitoring. In order to test their potential in cancer treatment decision and early detection of chemoresistance, we devised a new custom-made impedance measuring system based on electric cell-substrate impedance sensing (ECIS), optimized for long term impedance measurements. This device was employed in a proof of concept cell culture impedance analysis for the characterization of chemo-resistant colon cancer cells. Doxorubicin-resistant HT-29 cells were used for this purpose and monitored for 140 h. Analysis of impedance-based curves reveal different trends from chemo-sensitive and chemo-resistant cells. An impedance-based cytoxicity assay with different concentrations of doxorubicin was also performed using ECIS. The obtained results confirm the feasibility of ECIS in the study of drug resistance and show promises for studies of time-dependent factors related to physiological and behavioral changes in cells during resistance acquisition. The methodology presented herein, allows the continuous monitoring of cells under normal culture conditions as well as upon drug exposure. The ECIS device used, sets the basis for high-throughput early detection of resistance to drugs, administered in the clinical practice to cancer patients, and for the screening of new drugs in vitro, on patient-derived cells.

Functional miRNA Screening Identifies Wide-ranging Antitumor Properties of miR-3622b-5p and Reveals a New Therapeutic Combination Strategy in Ovarian Tumor Organoids.

Novel therapeutic strategies are urgently required for the clinical management of chemoresistant ovarian carcinoma, which is the most lethal of the gynecologic malignancies. miRNAs hold promise because they play a critical role in determining the cell phenotype by regulating several hundreds of targets, which could constitute vulnerabilities of cancer cells. A combination of gain-of-function miRNA screening and real-time continuous cell monitoring allows the identification of miRNAs with robust cytotoxic effects in chemoresistant ovarian cancer cells. Focusing on miR-3622b-5p, we show that it induces apoptosis in several ovarian cancer cell lines by both directly targeting Bcl-xL and EGFR-mediating BIM upregulation. miR-3622b-5p also sensitizes cells to cisplatin by inhibiting Bcl-xL in ovarian cancer cell lines escaping BIM induction. miR-3622b-5p also exerts antimigratory capacities by targeting both LIMK1 and NOTCH1. These wide-ranging antitumor properties of miR-3622b-5p in ovarian cancer cells are mimicked by the associations of pharmacologic inhibitors targeting these proteins. The combination of an EGFR inhibitor together with a BH3-mimetic molecule induced a large decrease in cell viability in a panel of ovarian cancer cell lines and several ovarian patient-derived tumor organoids, suggesting the value of pursuing such a combination therapy in ovarian carcinoma. Altogether, our work highlights the potential of phenotype-based miRNA screening approaches to identify lethal interactions which might lead to new drug combinations and clinically applicable strategies.

Continuous cell culture monitoring using a compact microplate reader with a silicone optical technology-based spatial filter.

Continuous cell monitoring is very important for the maintenance and control of cell multiplication and differentiation. This paper presents a compact microplate reader that is able to continuously measure a 24-well microplate (6 × 4 wells) using the optical absorption measurement method. The 24-channel plate reader consisted of a spatial filter, light emitting diode light source, and color sensors and was similarly sized with the cell culture microwell plates. A spatial filter was previously fabricated by our group using silicone optical technology (SOT). This SOT-based spatial filter has an excellent noise reduction effect. Light reflection at the optical path interface can be absorbed and only forward light can be transmitted; accordingly, a larger S/N ratio than that of conventional optical systems is expected. The fabricated 24-channel plate reader permits real-time cell monitoring during cultivation on the clean bench and in cell culture conditions by incorporating the SOT spatial filter. Using the device, it was possible to continuously evaluate the concentration and pH of reagents in the 24 wells in real time. Moreover, cell activity and protein production were detectable using the device. These results suggest that the newly fabricated device is a promising tool for the evaluation of cell behaviors for cell management.

Modeling Temporal Information of Mitotic for Mitotic Event Detection

Due to the enormous potential and influence that stem cells may have in regenerative medicine, there has been a rapidly growing interest in developing tools to analyze and characterize the behaviors of these cells in vitro. Among these behaviors, mitosis, or cell division, is very important because stem cells proliferate and renew themselves through mitosis. However, current automated systems for mitosis detection often require traditional computer vision technology and machine learning methods; automated mitosis detection and recognition are difficult to achieve and mainly rely on manual annotation. In this paper, we proposed an effective method to capture video-wide temporal information for automated mitosis detection and recognition, which is a nondestructive imaging modality, thereby allowing continuous monitoring of cells in culture. In this approach, we postulate that a function capable of ordering the frames of a video temporally well captures the evolution of the appearance within the video. We learn such ranking functions per video via a ranking machine and use the parameters of these functions as a new video representation. Here, we utilized the CNN model (VGG-16) and some classic low-level feature extraction methods (HOG, SIFT, and GIST) to extract low-level features for each frame. The proposed method is easy to interpret and implement, fast to compute and effective in recognizing mitosis events. In a comparison experiment, our approach significantly outperformed previous approaches in terms of both detection accuracy and computational efficiency. The data that we validate the proposed method with includes C3H10 mesenchymal and C2C12 myoblastic stem cell populations. Our approach achieves an F-score of 95.8 percent on the C2C12 dataset and an F-score of 95.3 percent on the C3H10 dataset. The results on both datasets outperform traditional mitosis recognition methods based on probability models. These experiments all demonstrate the significance of our approach.

Sigma-1 and Sigma-2 receptor ligands induce apoptosis and autophagy but have opposite effect on cell proliferation in uveal melanoma.

Uveal melanoma is the most common primary intraocular tumor in adults, with about 1200–1500 new cases occurring per year in the United States. Metastasis is a frequent occurrence in uveal melanoma, and outcomes are poor once distant spread occurs and no clinically significant chemotherapeutic protocol is so far available. The aim of the present study was to test the effect of various σ1 and σ2 receptor ligands as a possible pharmacological strategy for this rare tumor. Human uveal melanoma cells (92.1) were treated with various concentrations of different σ2 ligands (haloperidol and haloperidol metabolite II) and σ1 ligand ((+)-pentazocine) at various concentrations (1, 10 and 25 μM) and time points (0, 4 h, 8 h, 24 h and 48 h). Cell proliferation and migration were evaluated respectively by continuous cell monitoring by xCELLigence analysis, clonogenic assay and wound healing. Apoptosis and autophagy were also measured by cytofluorimetric and microscopy analysis. Our results showed that σ2 receptor ligands significantly reduced cell proliferation whereas (+)-pentazocine exhibited opposite results. All tested ligands showed significant decrease in cell migration. Interestingly, both σ1 and σ2 receptor ligands showed significant increase of autophagy and apoptosis at all concentrations. Taken all together these results suggest that sigma receptors mediates opposite biological effects but they also share common pharmacological effect on apoptosis and autophagy in uveal melanoma. In conclusion, these data provide the first evidence that sigma receptors may represent a “druggable” target to develop new chemotherapic agent for uveal melanoma.

Lens-free shadow image based high-throughput continuous cell monitoring technique

A high-throughput continuous cell monitoring technique which does not require any labeling reagents or destruction of the specimen is demonstrated. More than 6000 human alveolar epithelial A549 cells are monitored for up to 72h simultaneously and continuously with a single digital image within a cost and space effective lens-free shadow imaging platform. In an experiment performed within a custom built incubator integrated with the lens-free shadow imaging platform, the cell nucleus division process could be successfully characterized by calculating the signal-to-noise ratios (SNRs) and the shadow diameters (SDs) of the cell shadow patterns. The versatile nature of this platform also enabled a single cell viability test followed by live cell counting. This study firstly shows that the lens-free shadow imaging technique can provide a continuous cell monitoring without any staining/labeling reagent and destruction of the specimen. This high-throughput continuous cell monitoring technique based on lens-free shadow imaging may be widely utilized as a compact, low-cost, and high-throughput cell monitoring tool in the fields of drug and food screening or cell proliferation and viability testing.

Electric impedance sensing in cell-substrates for rapid and selective multipotential differentiation capacity monitoring of human mesenchymal stem cells

Biosensor systems which enable impedance measurements on adherent cell layers under label-free conditions are considered powerful tools for monitoring specific biological characteristics. A radio frequency identification-based sensor platform was adopted to characterize cultivation and differentiation of human bone marrow-derived multipotent stem cells (bmMSC) over periods of up to several days and weeks. Electric cell-substrate impedance sensing was achieved through fabrication of sensitive elements onto glass substrates which comprised two comb-shaped interdigitated gold electrodes covering an area of 1.8mm×2mm. The sensing systems were placed into the wells of a 6-well tissue culture plate, stacked onto a reader unit and could thus be handled and operated under sterile conditions. Continuous measurements were carried out with a sinusoidal voltage of 35mV at a frequency of 10kHz.After seeding of human bmMSC, this sensor was able to trace significant impedance changes contingent upon cell spreading and adhesion. The re-usable system was further proven suitable for live examination of cell-substrate attachment or continuous cell monitoring up to several weeks. Induction of either osteogenic or adipogenic differentiation could be validated in bmMSC cultures within a few days, in contrast to state-of-the-art protocols, which require several weeks of cultivation time. In the context of medical cell production in a GMP-compliant process, the here presented interdigitated electric microsensor technology allows the documentation of MSC quality in a fast, efficient and reliable fashion.

Automated Mitosis Detection of Stem Cell Populations in Phase-Contrast Microscopy Images

Due to the enormous potential and impact that stem cells may have on regenerative medicine, there has been a rapidly growing interest for tools to analyze and characterize the behaviors of these cells in vitro in an automated and high throughput fashion. Among these behaviors, mitosis, or cell division, is important since stem cells proliferate and renew themselves through mitosis. However, current automated systems for measuring cell proliferation often require destructive or sacrificial methods of cell manipulation such as cell lysis or in vitro staining. In this paper, we propose an effective approach for automated mitosis detection using phase-contrast time-lapse microscopy, which is a nondestructive imaging modality, thereby allowing continuous monitoring of cells in culture. In our approach, we present a probabilistic model for event detection, which can simultaneously 1) identify spatio-temporal patch sequences that contain a mitotic event and 2) localize a birth event, defined as the time and location at which cell division is completed and two daughter cells are born. Our approach significantly outperforms previous approaches in terms of both detection accuracy and computational efficiency, when applied to multipotent C3H10T1/2 mesenchymal and C2C12 myoblastic stem cell populations.

Real-time impedance analysis of host cell response to meningococcal infection

Measuring cell proliferation and cell death during bacterial infection involves performing end-point assays that represent the response at a single time point. A new technology from Roche Applied Science and ACEA Biosciences allows continuous monitoring of cells in real-time using specialized cell culture microplates containing micro-electrodes. The xCELLigence system enables continuous measurement and quantification of cell adhesion, proliferation, spreading, cell death and detachment, thus creating a picture of cell function during bacterial infection. Furthermore, lag and log phases can be determined to estimate optimal times to infect cells. In this study we used this system to provide valuable insights into cell function in response to several virulence factors of the meningitis causing pathogen Neisseria meningitidis, including the lipopolysaccharide (LPS), the polysaccharide capsule and the outer membrane protein Opc. We observed that prolonged time of infection with pathogenic Neisseria strains led to morphological changes including cell rounding and loss of cell–cell contact, thus resulting in changed electrical impedance as monitored in real-time. Furthermore, cell function in response to 14 strains of apathogenic Neisseria spp. ( N. lactamica and N. mucosa) was analyzed. In contrast, infection with apathogenic N. lactamica isolates did not change electrical impedance monitored for 48 h. Together our data show that this system can be used as a rapid monitoring tool for cellular function in response to bacterial infection and combines high data acquisition rates with ease of handling.

New technology for investigating trophoblast function

Measuring trophoblast function involves performing end-point assays that represent the response at a single time point. New technology from Roche Applied Science enables continuous monitoring of cells in real-time using specialized culture dishes containing micro-electrodes. The xCELLigence System allows continuous measurement and quantification of cell adhesion, proliferation, migration and invasion, thus creating a true picture of trophoblast function. Lag and log growth phases can be determined thus pinpointing optimal times to treat and harvest cells. Use of this system will provide valuable insights into trophoblast functions as well as the behaviour of other cell types found at the maternal–fetal interface.

Mid-IR Contact Probe for Examination of Live Cell Cultures

Continuous monitoring of live cell cultures and in-vitro tissues is a major challenge in the study of model systems for cancer research. Specifically, monitoring chemical changes often requires the cells to be stained with specific fluorophores (limited chemical content) or harvested from culture (precluding longitudinal study). Recently, mid-infrared spectroscopy is being increasingly applied to measure chemical content of cells and tissues. There are no reports, however, of in-vitro monitoring of cell cultures. The major reason is the high absorption of water in this spectral region and lack of instrumentation to address this need. This project seeks to apply mid-IR in a non-destructive manner to live cell cultures and tissues while maintaining rich information content. We accomplish this by the design of a contact-method probe. We have designed a fiber optic-base beam guidance geometry and coupled it to a total internal reflection sensing element. The entire sensor is coupled to a commercial spectrometer, thus allowing for rapid translation to other laboratories. It is believed it can be a cost effective solution using current technologies with adequate SNR for down-stream chemometric analysis. One of the major design challenges was to optimally guide and utilize light from the spectrometer within the constraints of leveraging as many commercially available components, processes, and methods to most quickly translate this idea into a working device. Our major task was optical modeling and subsequent fabrication of the device. The optical models show 6 percent throughput is possible using currently available parts; 1 percent is required. The design was implemented with the optical system placed wholly within a single chassis. While a single chassis design allows for miniaturization, it presents substantial alignment challenges due to the lack of degrees of freedom for movement. These challenges are the current work focus. Proof of principle studies are on-going. We anticipate that the goal of continuous cell monitoring and in-vitro cell spectroscopy will be attained upon final integration of the device with our experimental setup.
