Relationship between Contingent Negative Variation and afterimage duration in migraine patients.

Abnormalities in electrocortical parameters and persistence of afterimage after visual stimulation are known to occur in migraine patients. The results of studies on Contingent Negative Variation (CNV) and afterimage persistence in migraine patients suggest a link between these two phenomena and a connection to the pathomechanism of migraine. To date, no studies have investigated both afterimage duration and CNV parameters in the same subjects. The aim of this study was to investigate the relationship between the early component of CNV (iCNV) and the duration of the afterimage in migraine patients. Sixty seven migraine patients from the headache center of the University of Rostock Medical Center were examined for iCNV amplitude, iCNV habituation and afterimage duration. The subjects also completed questionnaires developed for this study and the MIDAS (Migraine Disability Assessment) questionnaire. Associations were found between iCNV amplitude and afterimage duration and between habituation capacity and afterimage duration. A deficit in habituation capacity correlated with a significantly prolonged afterimage duration. Increased iCNV amplitude and prolonged afterimage duration were also significantly correlated. Conclusions about the pathophysiology of migraine can be drawn from the results of this study. The results support the hypothesis of cortical hyperexcitability as a consequence of a low pre-activation level, which may be a possible contributory cause of migraine. Furthermore, they allow assessment of whether the afterimage examination, which is easier and quicker to perform than the CNV examination, can be used as a diagnostic tool or as a parameter to monitor the course of therapy in people with migraine.

Early and late contingent negative variation (CNV) reflect different aspects of deficits in schizophrenia.

Abnormal reward processing and psychomotor slowing are well-known in schizophrenia (SZ). As a slow frontocentral potential, contingent negative variation (CNV) is associated with anticipatory attention, motivation and motor planning. The present study aims to evaluate the early and late amplitude and latencies of CNV in patients with SZ compared to healthy controls during a reward processing task and to show its association with clinical symptoms. We recruited 21 patients with SZ and 22 healthy controls to compare early and late CNV amplitude and latency values during a Monetary Incentive Delay (MID) Task between groups. Patients' symptom severity, levels of negative symptoms and depressive symptoms were assessed. Clinical features of the patients were further examined for their relation with CNV components. In conclusion, we found decreased early CNV amplitudes in SZ during the reward condition. They also displayed diminished and shortened late CNV responses for incentive cues, specifically at the central location. Furthermore, early CNV amplitudes exhibited a significant correlation with positive symptoms. Both CNV latencies were linked with medication dosage and the behavioural outcomes of the MID task. We revealed that early and late CNV exhibit different functions in neurophysiology and correspond to various facets of the deficits observed in patients. Our findings also emphasized that slow cortical potentials are indicative of deficient motivational processes as well as impaired reaction preparation in SZ. To gain a deeper understanding of the cognitive and motor impairments associated with psychosis, future studies must compare the effects of CNV in the early and late phases.

Exploring the cortical habituation in migraine patients based on contingent negative variation.

Cognitive dysfunction has frequently been found in patients with migraine. The so-called contingent negative variation (CNV) and EEG power spectral densities may be the best choices to explore the underlining pathophysiology, such as cortical inhibition and habituation. Thirty migraine patients without aura and healthy controls matched for sex, age, and education were recruited separately for CNV recording. The amplitudes, latencies, and squares of different CNV components, such as oCNV, iCNV, tCNV, and PINV, were selected and analyzed. Behavioral data, such as manual reaction time (RT), were analyzed. We used the Person correlation coefficient R to analyze different ERP components in relation to clinical characteristics. A multiple regression analysis was conducted for the migraine group. Spectral analysis of EEG data from all channels using the fast Fourier transform (FFT). The migraine group had longer A-latency, C-latency, and iCNV-latency than the control group. The migraine group had higher iCNV-amplitude, oCNV-amplitude, and tCNV-amplitude than the control group, especially those located in the occipital area. The iCNV-square, oCNV-square, tCNV-square, or PINV-square in the migraine group was significantly larger than the control group. Different correlations were found between clinical characteristics and ERP components. The delta or theta activity in the migraine group was statistically lower than in the control group. Our study has revealed that migraine attacks may influence responsivity, pre-activation, habituation, and cortical inhibition not only on the behavioral level but also on the electrophysiological level. Abnormal changes in cortical habituation and inhibition can be interpreted as CNV components. Additionally, analyses have revealed correlations between CNV components and various factors, including age, the clinical course of the condition, attack frequency, pain intensity, and duration. Thus, repetitive migraine attacks can lead to a reduction in cortical inhibition and subsequent impairment in executive function.

Comparing monetary gain and loss in the monetary incentive delay task: EEG evidence.

What is more effective to guide behavior: The desire to gain or the fear to lose? Electroencephalography (EEG) studies have yielded inconsistent answers. In a systematic exploration of the valence and magnitude parameters in monetary gain and loss processing, we used time-domain and time-frequency-domain analyses to uncover the underlying neural processes. A group of 24 participants performed a monetary incentive delay (MID) task in which cue-induced anticipation of a high or low magnitude of gain or loss was manipulated trial-wise. Behaviorally, the anticipation of both gain and loss expedited responses, with gain anticipation producing greater facilitation than loss anticipation. Analyses of cue-locked P2 and P3 components revealed the significant valence main effect and valence × magnitude interaction: amplitude differences between high and low incentive magnitudes were larger with gain vs. loss cues. However, the contingent negative variation component was sensitive to incentive magnitude but did not vary with incentive valence. In the feedback phase, the RewP component exhibited reversed patterns for gain and loss trials. Time-frequency analyses revealed a large increase in delta/theta-ERS oscillatory activity in high- vs. low-magnitude conditions and a large decrease of alpha-ERD oscillatory activity in gain vs. loss conditions in the anticipation stage. In the consumption stage, delta/theta-ERS turned out stronger for negative than positive feedback, especially in the gain condition. Overall, our study provides new evidence for the neural oscillatory features of monetary gain and loss processing in the MID task, suggesting that participants invested more attention under gain and high-magnitude conditions vs. loss and low-magnitude conditions.

Exploration of cortical inhibition and habituation in insomnia: Based on CNV and EEG.

Cognitive dysfunction with abnormal cortical inhibition and habituation has frequently been found in patients with insomnia. And the so-called contingent negative variation (CNV) and EEG power spectral density (FFT) may be the best choice to explore the underlining pathophysiology. We used polysomnography (PSG) to record such objective PSG parameters. The amplitudes, latencies, areas of different CNV components such as oCNV, iCNV and tCNV, PINV have been selected and analyzed. Behavioral data such as manual reaction time (RT) has been analyzed. Spectral analysis was performed with fast Fourier transformation (FFT) on all channels to make a spectral analyses of EEG datas. The A-latency located in CZ or PZ were statistically longer in insomnia group than control group, the iCNV-latency located in insomnia group were statistically shorter than control group. The iCNV-amplitude located in insomnia group was lower than control group. The oCNV-amplitude or the tCNV-amplitude located in insomnia group was higher than control group. The oCNV-square, tCNV-square, or PINV-square located in insomnia group were significant larger than control group. β1 or β2 activity distributed in bilateral hemisphere were significantly increased in insomnia group than control group with different distributions. Our study revealed varied attentional and information processing in insomnia patients. Above all, we made a hypothesis with ceiling theory: Frontal lobe play an important role in maintaining cognitive processing, which needs much more energy consumption and leads to decreased fast EEG activity in frontal cortex, which contributes to reduced cortical inhibition, represented as abnormal CNV.

Spatial selectivity of the visual duration aftereffect in the sub-second range: An event-related potentials study

Prolonged adaptation to one duration biases the perception of subsequent durations in a phenomenon known as duration adaptation. There is controversy regarding the neural substrates of the visual duration aftereffect: some researchers have argued that duration adaptation occurs in early visual cortical areas, whereas others have argued for high-level visual areas. Investigation of spatial selectivity of the duration aftereffect could help reveal the neural mechanisms underlying duration adaptation. In the present study, we used event-related potentials (ERPs) to demonstrate spatial selectivity of the visual duration aftereffect in the sub-second range. We measured the duration aftereffect at a 20° adapt-test distance that spanned 10° on either side of fixation. Our results revealed that duration adaptation first affected the N1 ERP component and then influenced the contingent negative variation (CNV) component. Moreover, the CNV component showed position effects in the temporal encoding stage, and the post-stimulus positive component was subject to a duration effect but not a position effect. These findings indicate that sub-second duration coding is closely linked to the early processing of the visual cortex. Moreover, the adaptation not only affects the earlier temporal coding stage, but also the later temporal processing stage.

Pre-motor deficits in left spatial neglect: An EEG study on Contingent Negative Variation (CNV) and response-related beta oscillatory activity

Right Brain Damaged patients with left spatial neglect (N+), are characterised by poor allocation of attention in the contralesional left side of space. In a recent study (Lasaponara et al., 2018) we showed during orienting of spatial attention with endogenous central cues, both the EEG markers reflecting the early phases of orienting (Early Directing Attention Negativity) and those reflecting the late setting-up of sensory facilitation in the visual cortex (Late Directing Attention Positivity) are disturbed in N+ when these patients attend the left side of space. In the healthy brain, endogenous cues also elicit EEG activity related to the preparation of manual responses to upcoming spatial targets. Here, we wished to expand on our previous findings and investigate the EEG correlates of cue-related response preparation in N+ patients. To this aim we investigated the Contingent Negative Variation (CNV) response and the pre-motor Beta-oscillatory activity evoked by spatially informative central cues during the performance of a Posner task. Due to concomitant contralesional motor impairments, N+ an N- patients performed the task only with the ipsilesional right-hand. Compared to healthy controls and patients without neglect, N+ displayed a pathological suppression of CNV component that was independent of cue direction. In addition, the amplitude of the CNV in response to right-pointing cues was positively correlated with neglect severity in line bisection. N+ also displayed a pathological enhancement of pre-motor Beta oscillations over the left hemisphere during the time period that preceded manual responses to targets in the left side of space, particularly to invalidly cued ones. Synchronization in the Beta-band (ERS) was also correlated with lower detection rate and slower RTs to Invalid targets in the left side of space. These results provide new insights on the premotor components of the spatial orienting deficits suffered by patients with left spatial neglect and can help improving its diagnosis and rehabilitation.

Recruitment dynamics of cognitive control in insomnia.

Individuals with insomnia disorder (ID) commonly report complaints of cognitive control functioning. Conversely, both behavioral and neurological evidence supporting subjective cognitive control impairments in insomnia remain remarkably scarce and inconclusive. To investigate this discrepancy, the present study used next to behavioral measures, event-related potentials (ERPs) to assess proactive control and reactive control in insomnia. Individuals with insomnia disorder (n = 18) and good sleeper controls (GSC; n = 15) completed the AX-continuous performance task, while electroencephalography (EEG) was recorded. This task required participants to maintain specific cue-information active to prepare an adequate response to a subsequent probe, which allowed us to measure participants' reliance on both proactive and reactive control. The results indicate that, although ID show a comparable level of performance as GSC, they show a reduced proactive engagement of cue-induced maintenance and response preparation processes (as reflected by the P3b and the contingent negative variation components). Moreover, in contrast to GSC, ID fail to engage reactive control (as indexed by the P3a component) to overcome invalid response tendencies. This study provides neurological evidence for impairments in cognitive control functioning in insomnia. As such, our study contributes to a better understanding of the discrepancy between the commonly reported cognitive impairments in insomnia and the scarce objective evidence supporting these cognitive complaints.

The reinforcing value of delay escape in attention deficit/hyperactivity disorder: An electrophysiological study.

The delay aversion hypothesis argues that the tendency for impulsive choice (preference for smaller sooner over larger later rewards) is motivated by the escape of negative affective states associated with delay. This model predicts that individuals with ADHD find the imposition of delay before an outcome or event especially aversive and its escape reinforcing. Consistent with this, fMRI studies show that ADHD is associated with amygdala hyper-sensitivity to cues of delay. However, evidence that delay escape is reinforcing is lacking. Here we extend fMRI research by using electrophysiological methods to study the reinforcing properties of delay-escape in ADHD. Thirty controls and 25 adolescents with ADHD aged 10–15 years performed the Escape Delay Incentive (EDI) task- in which pre-target cues indicated three conditions: i) CERTAIN DELAY: delay would follow a response irrespective of response speed ii) CONDITIONAL DELAY: delay would only follow if the response was too slow and iii) NO DELAY: delay would follow the response whatever the speed. We focused on the Contingent Negative Variation (CNV), a cue-evoked marker of motivated response preparation, across two time windows (CNV1 and CNV2). We took measures of parent, teacher and self-rated ADHD symptoms, task performance (RT) and self-rated delay aversion. We isolated CNV components and compared these between ADHD and controls. Adolescents with ADHD displayed a larger CNV2 to the CONDITIONAL DELAY than the CERTAIN DELAY cues compared to controls. However, this effect was not mirrored at the performance level and was unrelated to self-reported delay aversion. Our study provides the first ERP evidence that delay escape differentially reinforcers neural activation of attention preparation in ADHD cases. Future studies should examine the impact of varying cognitive load on task EDI performance.

Contingent negative variation in acute trauma patients: A prospective exploratory study

It is still unclear why some individuals completely recover after an acute trauma and others develop a long-lasting post-traumatic stress disorder (PTSD). This study investigates whether the contingent negative variation (CNV) – a slow negative event-related potential – may be associated with the occurrence of PTSD after acute trauma. CNV (auditory 2-stimulus reaction time paradigm) was recorded within one month as well as 6 months after an acute trauma (dangerous or grave physical injuries, witnessing of attempted suicide or murder, robbery, extortion, accidents, heavy illness, death or loss of an important person, hostage-taking) in 39 otherwise healthy adults and compared with CNV recordings in 38 healthy control subjects without potentially traumatizing experience in their history. According to their subsequent clinical course, these subjects were divided into two groups: participants who recovered completely 1 month after the trauma (PTSD- group, n = 31), and those who began to experience PTSD (PTSD+ group, n = 8). Patients from both trauma groups were characterized by a significantly longer reaction time immediately after the trauma. The PTSD+ group demonstrated lower amplitudes of the late CNV component immediately and six months after the trauma compared with the PTSD- and the control group. Whether the lower CNV amplitudes in patients who develop PTSD after the acute trauma, which is already present in the first days after the trauma, may be related to a higher risk for development of PTSD in these subjects, this has to be clarified in further prospective studies.

The role of the P3 and CNV components in voluntary and automatic temporal orienting: A high spatial-resolution ERP study

A main distinction has been proposed between voluntary and automatic mechanisms underlying temporal orienting (TO) of selective attention. Voluntary TO implies the endogenous directing of attention induced by symbolic cues. Conversely, automatic TO is exogenously instantiated by the physical properties of stimuli. A well-known example of automatic TO is sequential effects (SEs), which refer to the adjustments in participants’ behavioral performance as a function of the trial-by-trial sequential distribution of the foreperiod between two stimuli. In this study a group of healthy adults underwent a cued reaction time task purposely designed to assess both voluntary and automatic TO. During the task, both post-cue and post-target event-related potentials (ERPs) were recorded by means of a high spatial resolution EEG system. In the results of the post-cue analysis, the P3a and P3b were identified as two distinct ERP markers showing distinguishable spatiotemporal features and reflecting automatic and voluntary a priori expectancy generation, respectively. The brain source reconstruction further revealed that distinct cortical circuits supported these two temporally dissociable components. Namely, the voluntary P3b was supported by a left sensorimotor network, while the automatic P3a was generated by a more distributed frontoparietal circuit. Additionally, post-cue contingent negative variation (CNV) and post-target P3 modulations were observed as common markers of voluntary and automatic expectancy implementation and response selection, although partially dissociable neural networks subserved these two mechanisms. Overall, these results provide new electrophysiological evidence suggesting that distinct neural substrates can be recruited depending on the voluntary or automatic cognitive nature of the cognitive mechanisms subserving TO.

Measuring expectation of pain: Contingent negative variation in placebo and nocebo effects.

Expectation is an important mechanism underlying placebo response. Here, we analysed expectation of placebo hypoalgesia and nocebo hyperalgesia by using, for the first time, the contingent negative variation (CNV), also known as expectancy wave. Subjects were presented a green or red cue followed by a train of either non painful or painful electrical stimuli, and expected hypoalgesia after the green and hyperalgesia after the red cue. In experiment 1, expectation was reinforced using a conditioning procedure whereby the green and red cues were paired with non painful and painful stimuli, respectively (acquisition). In a second session (test) the intensity of the stimuli was kept constant, regardless of cue. In experiment 2 no conditioning was performed and participants expected an altered pain perception indicated by the visual cues. CNV mean amplitude, time necessary to stop the train of stimuli (reaction time) and pain ratings were measured. A difference in pain perception occurred when electrical stimuli followed the presentation of the green cue compared to the red in the test session, whereas reaction times showed no changes. The same difference occurred in the early CNV component, related to cognitive stimulus anticipation, whereas the late CNV component, related to motor preparation, did not change. Moreover, these differences in pain perception and CNV amplitude were less robust in the experiment 2. Placebo hypoalgesia and nocebo hyperalgesia differently affect sensory (pain perception) and motor components (pain avoidance) of pain. Furthermore, CNV is an electrophysiological objective measure capable of dissecting these components. Dissection of placebo hypoalgesia, differentiating the sensory component (pain perception) from the motor component (pain avoidance). Study of these components using the contingent negative variation (CNV) as an electrophysiological objective measure.

Cue validity probability influences neural processing of targets

The neural bases of the so-called Spatial Cueing Effect in a visuo-auditory version of the Central Cue Posneŕs Paradigm (CCPP) are analyzed by means of behavioral patterns (Reaction Times and Errors) and Event-Related Potentials (ERPs), namely the Contingent Negative Variation (CNV), N1, P2a, P2p, P3a, P3b and Negative Slow Wave (NSW). The present version consisted of three types of trial blocks with different validity/invalidity proportions: 50% valid – 50% invalid trials, 68% valid – 32% invalid trials and 86% valid − 14% invalid trials. Thus, ERPs can be analyzed as the proportion of valid trials per block increases. Behavioral (Reaction Times and Incorrect responses) and ERP (lateralized component of CNV, P2a, P3b and NSW) results showed a spatial cueing effect as the proportion of valid trials per block increased. Results suggest a brain activity modulation related to sensory-motor attention and working memory updating, in order to adapt to external unpredictable contingencies.

Modulation of ERP components by task instructions in a cued go/no-go task.

The present study investigated how components of ERPs are modulated when participants optimize speed versus accuracy in a cued go/no-go task. Using a crossover design, 35 participants received instructions to complete the task prioritizing response speed in half of the task, and accurate responding in the other half of the task. Analysis was performed on the contingent negative variation (CNV), P3go, and P3no-go and the corresponding independent components (IC), as identified by group independent component analysis. After speed instructions, the IC CNV(late), P3go(anterior), P3no-go(early), and P3no-go(late) all had larger amplitudes than after accuracy instructions. Furthermore, both the IC P3go(posterior) and IC P3go(anterior) had shorter latencies after speed than after accuracy instructions. The results demonstrate that components derived from the CNV and P3 components are facilitated when participants optimize response speed. These findings indicate that these ERP components reflect executive processes enabling adjustment of behavior to changing demands.

Abnormal proactive and reactive cognitive control during conflict processing in major depression.

According to the Dual Mechanisms of Control framework, cognitive control consists of two complementary components: proactive control refers to anticipatory maintenance of goal-relevant information, whereas reactive control acts as a correction mechanism that is activated when a conflict occurs. Possibly, the well-known diminished inhibitory control in response to negative stimuli in Major Depressive Disorder (MDD) patients stems from a breakdown in proactive control, and/or anomalies in reactive cognitive control. In our study, MDD patients specifically showed increased response latencies when actively inhibiting a dominant response to a sad compared with a happy face. This condition was associated with a longer duration of a dominant ERP topography (800-900 ms poststimulus onset) and a stronger activity in the bilateral dorsal anterior cingulate cortex, reflecting abnormal reactive control when inhibiting attention to a negative stimulus. Moreover, MDD patients showed abnormalities in proactive cognitive control when preparing for the upcoming imperative stimulus (abnormal modulation of the contingent negative variation component), accompanied by more activity in brain regions belonging to the default mode network. All together, deficits to inhibit attention to negative information in MDD might originate from an abnormal use of both proactive resources and reactive control processes.

The learning-oddball paradigm: Data of 24 separate individuals illustrate its potential usefulness as a new clinical tool

ObjectiveIn a previous article reporting group data, we presented event-related potentials (ERPs), which were evoked by randomly presented target stimuli in a ‘learning-oddball’ task. These ERPs contained a large N2-P3 complex that decreased and a Contingent Negative Variation (CNV) that increased when the targets were presented in a regular fashion. Using the learning-oddball paradigm, the aim of the present paper was to determine ERP effects of introducing regularity in individual participants. MethodsThe data from the previous study were re-analyzed at the level of the individual participant, extracting individual sigmoid curves by means of wavelet-denoising and focusing on RTs, and CNV, N2, and P3 ERP components. ResultsMost participants displayed significant sigmoid curves with respect to the P3 component (22 of the 24 participants – 22/24), the N2 component (20/24), and/or the CNV (19/24) component. In contrast, reaction times (RTs) appeared less sensitive to incidental learning (15/24). Modest correlations were observed between RTs and N2 component amplitudes. ConclusionsIt is possible to extract significant ERP changes to introducing regularity in individual participants. SignificanceTracking ERP changes within the learning-oddball paradigm might be a useful tool to assess pattern detection capacities in individual patients.

Attention-Related Potentials Allow for a Highly Accurate Discrimination of Mild Cognitive Impairment Subtypes

The three most frequent forms of mild cognitive impairment (MCI) are single-domain amnestic MCI (sd-aMCI), single-domain dysexecutive MCI (sd-dMCI) and multiple-domain amnestic MCI (md-aMCI). Brain imaging differences among single domain subgroups of MCI were recently reported supporting the idea that electroencephalography (EEG) functional hallmarks can be used to differentiate these subgroups. We performed event-related potential (ERP) measures and independent component analysis in 18 sd-aMCI, 13 sd-dMCI and 35 md-aMCI cases during the successful performance of the Attentional Network Test. Sensitivity and specificity analyses of ERP for the discrimination of MCI subgroups were also made. In center-cue and spatial-cue warning stimuli, contingent negative variation (CNV) was elicited in all MCI subgroups. Two independent components (ICA1 and 2) were superimposed in the time range on the CNV. The ICA2 was strongly reduced in sd-dMCI compared to sd-aMCI and md-aMCI (4.3 vs. 7.5% and 10.9% of the CNV component). The parietal P300 ERP latency increased significantly in sd-dMCI compared to md-aMCI and sd-aMCI for both congruent and incongruent conditions. This latency for incongruent targets allowed for a highly accurate separation of sd-dMCI from both sd-aMCI and md-aMCI with correct classification rates of 90 and 81%, respectively. This EEG parameter alone performed much better than neuropsychological testing in distinguishing sd-dMCI from md-aMCI. Our data reveal qualitative changes in the composition of the neural generators of CNV in sd-dMCI. In addition, they document an increased latency of the executive P300 component that may represent a highly accurate hallmark for the discrimination of this MCI subgroup in routine clinical settings.

Parietal Influence on Temporal Encoding Indexed by Simultaneous Transcranial Magnetic Stimulation and Electroencephalography

Previous studies have suggested that contingent negative variation (CNV), as recorded by electroencaphalography (EEG), may serve as an index of temporal encoding. The interpretation of these studies is complicated by the fact that, in a majority of studies, the CNV signal was obtained at a time when subjects were not only registering stimulus duration but also making decisions and preparing to act. Previously, we demonstrated that repetitive transcranial magnetic stimulation (rTMS) of the right supramarginal gyrus (rSMG) in humans lengthened the perceived duration of a visual stimulus (Wiener et al., 2010a), suggesting the rSMG is involved in basic encoding processes. Here, we report a replication of this effect with simultaneous EEG recordings during the encoding of stimulus duration. Stimulation of the rSMG led to an increase in perceived duration and the amplitude of N1 and CNV components recorded from frontocentral sites. Furthermore, the size of the CNV amplitude, but not N1, positively correlated with the size of the rTMS effect but negatively correlated with bias (the baseline tendency to report a comparison stimulus as shorter), suggesting that the CNV indexes stimulus duration. These results suggest that a feedforward mechanism from parietal to prefrontal regions mediates temporal encoding and demonstrate a dissociation between early and late phases of encoding processes.

Time-Resolved Influences of Functional DAT1 and COMT Variants on Visual Perception and Post-Processing

Thus, we examined the influences of functional variants related to dopamine inactivation in the dopamine transporter (DAT1) and catechol-O-methyltransferase genes (COMT) on the time-course of visual processing in a contingent negative variation (CNV) task.Methods64-channel EEG recordings were obtained from 195 healthy adolescents of a community-based sample during a continuous performance task (A-X version). Early and late CNV as well as preceding visual evoked potential components were assessed.ResultsSignificant additive main effects of DAT1 and COMT on the occipito-temporal early CNV were observed. In addition, there was a trend towards an interaction between the two polymorphisms. Source analysis showed early CNV generators in the ventral visual stream and in frontal regions. There was a strong negative correlation between occipito-temporal visual post-processing and the frontal early CNV component. The early CNV time interval 500–1000 ms after the visual cue was specifically affected while the preceding visual perception stages were not influenced.ConclusionsLate visual potentials allow the genomic imaging of dopamine inactivation effects on visual post-processing. The same specific time-interval has been found to be affected by DAT1 and COMT during motor post-processing but not motor preparation. We propose the hypothesis that similar dopaminergic mechanisms modulate working memory encoding in both the visual and motor and perhaps other systems.

Time-dependent post-imperative negative variation indicates adaptation and problem solving in migraine patients

According to the Seligman theory of learned helplessness, depression is caused by a repetitive experience of loss of control resulting in internal, stable and global attributional styles for negative events. In depressed patients and healthy controls experiencing such events, an increased amplitude of the post-imperative negative variation (PINV) has been described. The aim of the study was to investigate a possible correlation between migraine, depression, learned helplessness and PINV. 24 patients suffering from migraine without aura and 24 healthy controls were exposed to a situation of loss of control whilst the contingent negative variation (CNV) from C3, C4 and Cz were recorded. Before conducting the experiment, the subjects were asked to answer the Beck Depression Inventory (BDI) and the German attributional style questionnaire (GASQ). Amplitudes of total CNV, early and late component and PINV were calculated in eight blocks of four recordings each. The results confirm findings of a pronounced PINV in situations of loss of control, though high amplitudes were not correlated with low values in the GASQ and therefore with learned helplessness. High PINV in migraine patients correlated with high scores in the BDI and the list of the complaints questionnaire. However, this was not the case in healthy controls. In this experimental situation, PINV in migraine patients can be interpreted as an expectancy potential in order to avoid failure and helplessness.
