Spore Germination of Diplazium simplicivenium Holtt. (Athyriaceae) in Peninsular Malaysia

Diplazium simplicivenium Holtt. a species of fern in Peninsular Malaysia. This study reports an efficient method for D. simplicivenium spore sterilisation and the effect of plant growth regulators (PGRs) via green globular bodies (GGB). Sterilisation with 0.1% mercuric chloride effectively allowed spore germination. The result showed that the culture media supplemented with 0.5 mg/L gibberellin positively affects the weight of GGB and the number of shoots with significant differences (ANOVA, p < 0.05). The soaking technique established in this study for spore sterilisation is an efficient approach, and the optimal plant and concentration of plant hormones were identified. This procedure can be applied to other indigenous ferns and closely similar species.

Metabolic regulation mechanism of melatonin for reducing cadmium accumulation and improving quality in rice

Melatonin acts as a potential regulator of cadmium (Cd) tolerance in rice. However, its practical value in rice production remains unclear. In order to support the hypothesis that melatonin affects Cd accumulation and rice quality, we undertook the experiments. The results showed that exogenous melatonin application was associated with reduced Cd accumulation (23–43%) in brown rice. Fourier transform infrared spectroscopy (FTIR) analysis showed that exogenous melatonin affected the rice protein secondary structure and starch short-range structure. Metabolomics based on LC–MS/MS revealed that exogenous melatonin altered the brown rice metabolic profile, decreased fatty acid metabolite content, but increased amino acid metabolite, citric acid, melatonin biosynthetic metabolite, and plant hormone contents. These findings indicate that exogenous melatonin can effectively reduced Cd accumulation and improve rice quality through metabolic network regulation, serving as an effective treatment for rice cultivated in Cd-contaminated soil.

Comparative transcriptome revealed the molecular responses of Aconitum carmichaelii Debx. to downy mildew at different stages of disease development

BackgroundAconitum carmichaelii Debx. has been widely used as a traditional medicinal herb for a long history in China. It is highly susceptible to various dangerous diseases during the cultivation process. Downy mildew is the most serious leaf disease of A. carmichaelii, affecting plant growth and ultimately leading to a reduction in yield. To better understand the response mechanism of A. carmichaelii leaves subjected to downy mildew, the contents of endogenous plant hormones as well as transcriptome sequencing were analyzed at five different infected stages.ResultsThe content of 3-indoleacetic acid, abscisic acid, salicylic acid and jasmonic acid has changed significantly in A. carmichaelii leaves with the development of downy mildew, and related synthetic genes such as 9-cis-epoxycarotenoid dioxygenase and phenylalanine ammonia lyase were also significant for disease responses. The transcriptomic data indicated that the differentially expressed genes were primarily associated with plant hormone signal transduction, plant-pathogen interaction, the mitogen-activated protein kinase signaling pathway in plants, and phenylpropanoid biosynthesis. Many of these genes also showed potential functions for resisting downy mildew. Through weighted gene co-expression network analysis, the hub genes and genes that have high connectivity to them were identified, which could participate in plant immune responses.ConclusionsIn this study, we elucidated the response and potential genes of A. carmichaelii to downy mildew, and observed the changes of endogenous hormones content at different infection stages, so as to contribute to the further screening and identification of genes involved in the defense of downy mildew.

Plant hormone profiling of scion and rootstock incision sites and intra- and inter-family graft junctions in Nicotiana benthamiana

ABSTRACT Many previous studies have suggested that various plant hormones play essential roles in the grafting process. In this study, to understand the plant hormones that accumulate in the graft junctions, whether these are supplied from the scion or rootstock, and how these hormones play a role in the grafting process, we performed a hormonome analysis that accumulated in the incision site of the upper plants from the incision as “ungrafted scion” and lower plants from the incision as “ungrafted rootstock” in Nicotiana benthamiana. The results revealed that indole-3-acetic acid (IAA) and gibberellic acid (GA), which regulate cell division; abscisic acid (ABA) and jasmonic acid (JA), which regulate xylem formation; cytokinin (CK), which regulates callus formation, show different accumulation patterns in the incision sites of the ungrafted scion and rootstock. In addition, to try discussing the differences in the degree and speed of each event during the grafting process between intra- and inter-family grafting by determining the concentration and accumulation timing of plant hormones in the graft junctions, we performed hormonome analysis of graft junctions of intra-family grafted plants with N. benthamiana as scion and Solanum lycopersicum as rootstock (Nb/Sl) and inter-family grafted plants with N. benthamiana as scion and Arabidopsis thaliana as rootstock (Nb/At), using the ability of Nicotiana species to graft with many plant species. The results revealed that ABA and CK showed different accumulation timings; IAA, JA, and salicylic acid (SA) showed similar accumulation timings, while different accumulated concentrations in the graft junctions of Nb/Sl and Nb/At. This information is important for understanding the molecular mechanisms of plant hormones in the grafting process and the differences in molecular mechanisms between intra- and inter-family grafting.

Occurrence of plant hormones in composts made from organic fraction of agri-food industry waste

Utilizing the organic fraction of agri-food industry waste for fertilization represents one approach to waste management, with composting emerging as a popular method. Composts derived from this waste may contain plant hormones alongside primary macronutrients. This study aimed to evaluate the content of plant hormones in composts crafted from the organic fraction of agri-food industry waste. The presence of these substances was ascertained using liquid chromatography–mass spectrometry (LC-MS) analysis, applied to extracted samples from three composts produced in a bioreactor and three obtained from companies. The results indicate the presence of 35 compounds, which belong to six types of plant hormones: auxins, cytokinins, gibberellins, brassinosteroids, abscisic acid, and salicylic acid, in composts for the first time. The highest amount of plant hormones was noted in buckwheat husk and biohumus extract (35 compounds), and the lowest in hemp chaff and apple pomace (14 compounds). Brassinosteroids (e.g., brassinolide, 28-homobrassinolide, 24-epicastasterone, 24-epibrassinolide, and 28-norbrassinolide) and auxins (e.g., indolilo-3-acetic acid) are dominant. The highest concentration of total phytohormones was reported in biohumus extract (2026.42 ng g−1 dry weight), and the lowest in organic compost (0.18 ng g−1 dry weight).

Silencing of early auxin responsive genes MdGH3-2/12 reduces the resistance to Fusarium solani in apple1

Apple replant disease (ARD) has led to severe yield and quality reduction in the apple industry. Fusarium solani (F. solani) has been identified as one of the main microbial pathogens responsible for ARD. Auxin (indole-3-acetic acid, IAA), as an endogenous hormone in plants, is involved in almost all plant growth and development processes and plays a role in plant immunity against pathogens. Gretchen Hagen3 (GH3) is one of the early/primary auxin response genes. The aim of this study was to evaluate the function of MdGH3-2 and MdGH3-12 in the defense response of F. solani by treating MdGH3-2/12 RNAi plants with F. solani. These results show that under F. solani infection, RNAi of MdGH3-2/12 inhibited plant biomass accumulation and aggravated root damage. After inoculation of F. solani, MdGH3-2/12 RNAi inhibited the biosynthesis of acid-amido synthetase. This led to the inhibition of free IAA combining with amino acid, resulting in excessive free IAA accumulation. Excessive free IAA alters plant tissue structure, accelerates fungal hyphal invasion, reduced the activity of antioxidant enzymes (SOD, POD and CAT) and increased reactive oxygen species (ROS) level, reduced total chlorophyll content and photosynthetic ability, while regulating the expression of PR-related genes including PR1, PR4, PR5 and PR8. It also changed the content of plant hormones and amino acids, and ultimately decreased the resistance to F. solani. In conclusion, these results demonstrate that MdGH3-2 and MdGH3-12 play an important role in F. solani tolerance to ARD.

Tamyb10-D1 restores red grain color and increases grain dormancy via suppressing expression of TaLTP2.128, non-specific lipid transfer protein in wheat

Grain dormancy of wheat is closely associated with grain color: red-grained lines show higher dormancy than white-grained lines. The production of red pigments is regulated by R-1, Tamyb10 gene. However, the relation between grain color and dormancy remains unknown. For this study, we generated transgenic lines which were introduced a DNA fragment containing Tamyb10-D1 gene and its a 2 kb promoter including the 5′ untranslated region into white-grained wheat. Transgenic lines showed red-grained and higher dormant traits. Contents of plant hormones and gene expression of embryos at 30 days after pollination were examined in a wild type and a transgenic line. No differences were observed in the contents of plant hormones, but several genes are differentially expressed between these lines. One differentially expressed gene, TaLTP2.128, is a member of non-specific lipid transfer proteins. It was expressed higher in white grains than in red grains. A putative amino acid sequence showed similarity to that of OsHyPRP5, which is identified as QTL controlling low-temperature germinability in rice. Expression of TaLTP2.128 was increased by grain imbibition. The increasing levels were higher not only in other white-grained lines, but also in non-dormant red-grained lines. TaLTP2.128 was expressed at a quite early stage of germination. These study findings indicate that Tamyb10 regulates dormancy release by the modification of TaLTP2.128 acting as trigger of germination.

Attenuated total reflection Fourier-transform infrared spectroscopy for the prediction of hormone concentrations in plants.

Plant hormones are important in the control of physiological and developmental processes including seed germination, senescence, flowering, stomatal aperture, and ultimately the overall growth and yield of plants. Many currently available methods to quantify such growth regulators quickly and accurately require extensive sample purification using complex analytic techniques. Herein we used ultra-performance liquid chromatography-high-resolution mass spectrometry (UHPLC-HRMS) to create and validate the prediction of hormone concentrations made using attenuated total reflection Fourier-transform infrared (ATR-FTIR) spectral profiles of both freeze-dried ground leaf tissue and extracted xylem sap of Japanese knotweed (Reynoutria japonica) plants grown under different environmental conditions. In addition to these predictions made with partial least squares regression, further analysis of spectral data was performed using chemometric techniques, including principal component analysis, linear discriminant analysis, and support vector machines (SVM). Plants grown in different environments had sufficiently different biochemical profiles, including plant hormonal compounds, to allow successful differentiation by ATR-FTIR spectroscopy coupled with SVM. ATR-FTIR spectral biomarkers highlighted a range of biomolecules responsible for the differing spectral signatures between growth environments, such as triacylglycerol, proteins and amino acids, tannins, pectin, polysaccharides such as starch and cellulose, DNA and RNA. Using partial least squares regression, we show the potential for accurate prediction of plant hormone concentrations from ATR-FTIR spectral profiles, calibrated with hormonal data quantified by UHPLC-HRMS. The application of ATR-FTIR spectroscopy and chemometrics offers accurate prediction of hormone concentrations in plant samples, with advantages over existing approaches.

Transcriptomics reveals a core transcriptional network of K-type cytoplasmic male sterility microspore abortion in wheat (Triticum aestivum L.)

BackgroundCytoplasmic male sterility (CMS) plays a crucial role in hybrid production. K-type CMS, a cytoplasmic male sterile line of wheat with the cytoplasms of Aegilops kotschyi, is widely used due to its excellent characteristics of agronomic performance, easy maintenance and easy restoration. However, the mechanism of its pollen abortion is not yet clear.ResultsIn this study, wheat K-type CMS MS(KOTS)-90-110 (MS line) and it’s fertile near-isogenic line MR (KOTS)-90-110 (MR line) were investigated. Cytological analysis indicated that the anthers of MS line microspore nucleus failed to divide normally into two sperm nucleus and lacked starch in mature pollen grains, and the key abortive period was the uninucleate stage to dinuclear stage. Then, we compared the transcriptome of MS line and MR line anthers at these two stages. 11,360 and 5182 differentially expressed genes (DEGs) were identified between the MS and MR lines in the early uninucleate and binucleate stages, respectively. Based on GO enrichment and KEGG pathways analysis, it was evident that significant transcriptomic differences were “plant hormone signal transduction”, “MAPK signaling pathway” and “spliceosome”. We identified 17 and 10 DEGs associated with the IAA and ABA signal transduction pathways, respectively. DEGs related to IAA signal transduction pathway were downregulated in the early uninucleate stage of MS line. The expression level of DEGs related to ABA pathway was significantly upregulated in MS line at the binucleate stage compared to MR line. The determination of plant hormone content and qRT-PCR further confirmed that hormone imbalance in MS lines. Meanwhile, 1 and 2 DEGs involved in ABA and Ethylene metabolism were also identified in the MAPK cascade pathway, respectively; the significant up regulation of spliceosome related genes in MS line may be another important factor leading to pollen abortion.ConclusionsWe proposed a transcriptome-mediated pollen abortion network for K-type CMS in wheat. The main idea is hormone imbalance may be the primary factor, MAPK cascade pathway and alternative splicing (AS) may also play important regulatory roles in this process. These findings provided intriguing insights for the molecular mechanism of microspore abortion in K-type CMS, and also give useful clues to identify the crucial genes of CMS in wheat.

Silicon application enhances wheat defence against Sitobion avenae F. by regulating plant physiological-biochemical responses

Sitobion avenae F. is a highly prevalent and devastating pest in wheat crops, leading to significant yield losses. Silicon (Si) has been widely recognized as an effective inducer of plant resistance against aphids. Nevertheless, the underlying mechanisms governing the physiological and biochemical responses of plants induced by Si defense against S. avenae F. remain incompletely understood. In this study, we conducted experiments by treating wheat leaves with varying concentrations of Tetraethyl orthosilicate (TEOS) spray under aphid infestation. We meticulously observed and recorded the life cycle of S. avenae F. and measured the content of plant hormones, secondary metabolites, and the activity of defense enzymes in wheat leaves. Furthermore, we utilized structural equation modeling to discern the causal correlation between aphid performance and the physiological-biochemical responses of wheat under TEOS sprays. Our findings revealed that a concentration of 3 mmol/L TEOS significantly shortened the net reproductive rate, intrinsic rate and finite rate of increase, and mean generation time of S. avenae F., while simultaneously prolonging the population doubling time. Additionally, the content of lignin, total phenolics, flavonoids, alkaloids, tannin, jasmonic acid (JA), and salicylic acid in wheat leaves exhibited a substantial increase. Furthermore, the activity of phenylalanine ammonia-lyase (PAL), polyphenol oxidase (PPO), catalase, and lipoxygenase in wheat leaves was significantly enhanced. Our results suggest that TEOS spray reduced the survival and population growth of S. avenae F. while enhancing the defense response of wheat against aphids by activating the activity of PAL and PPO in wheat, and increasing the content of total phenolic and JA. This work provides valuable insights for the development of appropriate Si fertilizers for effective pest management and offers robust theoretical support for wheat aphid control through agricultural fertilization strategies.

Serendipita indica Promotes the Growth of Tartary Buckwheat by Stimulating Hormone Synthesis, Metabolite Production, and Increasing Systemic Resistance.

The main objective of this study was to investigate the influence of Serendipita indica on the growth of Tartary buckwheat plants. This study highlighted that the roots of Tartary buckwheat can be colonized by S. indica and that this fungal endophyte improved plants height, fresh weight, dry weight, and grain yield. In the meantime, the colonization of S. indica in Tartary buckwheat leaves resulted in elevated levels of photosynthesis, plant hormone content, antioxidant enzyme activity, proline content, chlorophyll content, soluble sugars, and protein content. Additionally, the introduction of S. indica to Tartary buckwheat roots led to a substantial rise in the levels of flavonoids and phenols found in the leaves and seeds of Tartary buckwheat. In addition, S. indica colonization reduced the content of malondialdehyde and hydrogen peroxide when compared to non-colonized plants. Importantly, the drought tolerance of Tartary buckwheat plants is increased, which benefits from physiology and bio-chemical changes in plants after S. indica colonized. In conclusion, we have shown that S. indica can improve systematic resistance and promote the growth of Tartary buckwheat by enhancing the photosynthetic capacity of Tartary buckwheat, inducing the production of IAA, increasing the content of secondary metabolites such as total phenols and total flavonoids, and improving the antioxidant enzyme activity of the plant.

The Technical System of Rapid Propagation with Photinia frasery Leaves Inducing Calli, First Generation Culture, Increasing Reproduction, Rooting and Training Seedlings to Transplant

The explants of Photinia frasery with leaves were first generation cultured. And then, the calli which were induced by explants were cultured and proliferated, followed by the effective seedlings which were proliferation-targeted cultured, rooting-targeted cultured. Finally, the seedlings are trained and transplanted. The study results showed: (1) Primary culture of Photinia frasery leaves, the best plant hormone combination in medium is: MS+0.4 mg · L−1BA+1.6 mg · L−1NAA; (2) The result of orthogonal test, proliferation culture of Photinia frasery, the best plant hormone combination in medium is: MS+2.0 mg·L−1BA+0.1 mg·L−1IBA+2.0 mg·L−1GA+40 g sucrose; (3) Rooting culture of Photinia frasery, the best plant hormone combination in medium is: 1/2MS+0.4 mg·L−1IBA. The results showed that different impacts of plant hormone type and concentration combination on the primary culture, reproducing culture and rooting culture of Photinia frasery leaves were obvious.

Comparative protein profiling of two soybean genotypes with different stress tolerance reveals major components in drought tolerance

IntroductionDrought stress has gradually become a limiting factor for plant growth. Soybean, a crop with a higher water demand than other plants, is particularly sensitive to water deficits.MethodTo explore the effect of drought stress on soybean protein expression, we used drought-tolerant soybean genotype HeiNong44 (HN44) and sensitive soybean genotype HeiNong65 (HN65) as experimental materials, PEG-6000 as an osmotic regulator, and tandem mass tag (TMT) technology to identify the differential expression of soybean proteins.ResultsWe identified that 48 upregulated and 38 downregulated proteins in HN44 and 55 upregulated and 13 downregulated proteins in HN65.DiscussionKEGG analysis showed that these differentially expressed proteins were involved in carbohydrate metabolism, signal transduction, amino acid metabolism, lipid metabolism, and programmed cell death. Drought stress usually caused an increase in the activity of plant antioxidant enzymes, the obstruction of photosynthetic synthesis, and a significant change in the content of plant hormones. We found that HN44 responds to drought stress mainly through sugar decomposition, increased antioxidant enzyme activity and lipid metabolism. HN65 responds to drought stress mainly through ABA synthesis, increased antioxidant enzyme activity and amino acid metabolism. Our study also found that differential proteins in the starch and sucrose metabolic pathway play a key role in supplying energy and regulating osmotic potential.

Relationship between seasonal variation in isoprene emission and plant hormone profiles in the tropical plant Ficus septica.

In Ficus septica, the short-term control of isoprene production and, therefore, isoprene emission has been linked to the hormone balance between auxin (IAA) and jasmonic acid (JA). However, the relationship between long-term changes in isoprene emission and that of plant hormones remains unknown. This study tracked isoprene emissions from F. septica leaves, plant hormone concentrations and signalling gene expression, MEP pathway metabolite concentrations, and related enzyme gene expression for 1 year in the field to better understand the role of plant hormones and their long-term control. Seasonality of isoprenes was mainly driven by temperature- and light-dependent variations in substrate availability through the MEP route, as well as transcriptional and post-transcriptional control of isoprene synthase (IspS). Isoprene emissions are seasonally correlated with plant hormone levels. This was especially evident in the cytokinin profiles, which decreased in summer and increased in winter. Only 4-hydroxy-3-methylbut-2-butenyl-4-diphosphate (HMBDP) exhibited a positive connection with cytokinins among the MEP metabolites examined, suggesting that HMBDP and its biosynthetic enzyme, HMBDP synthase (HDS), play a role in channelling of MEP pathway metabolites to cytokinin production. Thus, it is probable that cytokinins have potential feed-forward regulation of isoprene production. Under long-term natural conditions, the hormonal balance of IAA/JA-Ile was not associated with IspS transcripts or isoprene emissions. This study builds on prior work by revealing differences between short- and long-term hormonal modulation of isoprene emissions in the tropical tree F. septica.

Transcriptome analysis reveals that auxin promotes strigolactone-induced adventitious root growth in the hypocotyl of melon seedlings.

Strigolactone (SL) and auxin are two important phytohormones involved in plant root development, but whether they show synergistic or mutual promotion effects during adventitious root (AR) formation has not been adequately explored. In this study, we investigated the mechanisms of GR24 (synthetic SL) and indole-3-acetic acid (IAA; a type of auxin) in the formation of ARs using melon as the study material. Morphological measurements showed that the AR number, length, superficial area, and volume under the GR24 treatment were 1.60-3.27, 1.58-3.99, 2.06-3.42, and 3.00-6.11 times greater than those of the control group, respectively, at 6-10 days; the GR24+IAA treatment further promoted AR formation in melon seedlings, and the AR number, length, superficial area, and volume under the GR24+IAA treatment were 1.44-1.51, 1.28-1.73, 1.19-1.83, and 1.31-1.87 times greater than those obtained with the GR24 treatment, respectively. Transcriptome analysis revealed 2,742, 3,352, and 2,321 differentially expressed genes (DEGs) identified from the GR24 vs. control, GR24+IAA vs. control, and GR24+IAA vs. GR24 comparisons, respectively. The GR24 treatment and GR24+IAA treatment affected auxin and SL synthesis as well as components of the phytohormone signal transduction pathway, such as auxin, brassinosteroid (BR), ethylene (ETH), cytokinin (CK), gibberellin (GA), and abscisic acid (ABA). The concentrations of auxin, GA, zeatin (ZT), and ABA were evaluated using high-performance liquid chromatography (HPLC). From 6 to 10 days, the auxin, GA, and ZT contents in the GR24 treatment group were increased by 11.48%-15.34%, 11.83%-19.50%, and 22.52%-66.17%, respectively, compared to the control group, and these features were increased by 22.00%-31.20%, 21.29%-25.75%, 51.76%-98.96%, respectively, in the GR24+IAA treatment group compared with the control group. Compared to that in the control, the ABA content decreased by 10.30%-11.83% in the GR24 treatment group and decreased by 18.78%-24.00% in the GR24+IAA treatment group at 6-10 days. Our study revealed an interaction between strigolactone and auxin in the induction of AR formation in melon seedlings by affecting the expression of genes related to plant hormone pathways and contents.

Increasing Rice Seedling Growth under Induced Water-Deficient Conditions Using Indole-3-Acetic Acid Produced from <em>Enterobacter</em> sp. Bacteria

Purpose: Water deficiency stress affects rice growth and productivity around the world while reducing indole-3-acetic acid (IAA) levels within plants. This study aims to determine the effects of IAA, produced from the endophytic bacteria Enterobacter sp. (RD4-1-1), on the germination and seedling growth of two lowland rice varieties, namely, RD31 and RD41, under water deficiency stress. Research Method: Water deficiency was simulated in culture medium [using the percentage of polyethylene glycol 6000 (PEG)] and soil cultivation (using different watering frequencies). The effects of RD4-1-1 IAA at various concentrations on rice under water deficiency stress were assessed by measuring a variety of characteristics related to germination and seedling growth. Findings: The effects of RD4-1-1 IAA supplementation in rice depended on the availability of water and the rice variety. RD4-1-1 IAA promoted the germination and growth of rice seedlings according to most measured characteristics, but the effective concentration levels differed with water deficiency in RD31 and RD41 varieties. In particular, the appropriate concentration of bacterial IAA was lower and was in a narrower range when applied to plants in soil compared with that required when IAA was used in plants cultured on medium. Originality/ Value: This study is one of few studies to report the effects of appropriate concentrations of microorganism-produced plant hormones on rice growth under water-deficient conditions.

Transcriptomic and metabolomic analyses unveil the growth advantage mechanism conferred by heterosis of Michelia 'Zhongshanhanxiao'.

Michelia compressa (Maxim.) Sarg. is one of the important timber trees in Taiwan province, P. R. China. Michelia 'Zhongshanhanxiao' is a group of variants found among the progeny of M. compressa that exhibit higher growth rates compared with normal individuals, with a significantly increased stem diameter and height, as well as enlarged leaves and flowers. However, the molecular mechanisms fostering the growth advantage and morphological variations are unknown and deserve further study. Through analysing the transcriptome, metabolome and physiological processes of leaves, we identified remarkable differences in gene expression and metabolic profiles between Michelia 'Zhongshanhanxiao' and both the maternal M. compressa and its normal progeny. These differences were widely associated with a plant-pathogen interaction, phenylpropanoid biosynthesis, cyanoamino acid metabolism, carbon fixation in photosynthetic organisms and plant hormone signal transduction. Additionally, physiological measurements showed that Michelia 'Zhongshanhanxiao' possesses stronger photosynthetic capacity and higher plant hormone content. These results suggest that the heterosis of Michelia 'Zhongshanhanxiao' is regulated by candidates related to cell division, resistance to pathogens and the accumulation of organic compounds. The findings of this study provide crucial information on the molecular mechanisms underlying the growth advantages conferred by heterosis in trees.

Effects of plant age on antioxidant activity and endogenous hormones in Alpine Elymus sibiricus of the Tibetan Plateau.

Elymus sibiricus L. is a perennial forage species that has potential to serve as a forage source in livestock grazing systems. However, E. sibiricus has been shown to have a rapid and substantial reduction of aboveground biomass and seed yield after 3 or 4 years and an accelerated aging process. To determine possible aging mechanisms, we planted E. sibiricus seeds in triplicate blocks in 2012, 2015, and 2016, respectively, and harvested samples of leaves and roots at the jointing and heading stages in 2018 and 2019 to determine oxidative indices and endogenous hormones. The fresh aboveground biomass of 4- and 5-year old plants declined by 34.2% and 52.4% respectively compared with 3-year old plants, and the seed yield declined by 12.7% and 34.1%, respectively. The water content in leaves was 51.7%, 43.3%, and 35.6%, and net photosynthesis was 7.73, 6.35, and 2.08 µmol/m2·s in 3-, 4-, and 5-year old plants, respectively. The superoxide anion radical generation rate in leaves and roots did not show any aging pattern. There was a non-significant increase in malondialdehyde concentration with plant age, particularly in leaves and roots at the heading stage in 2019. The superoxide dismutase activity showed a declining trend with age of plant roots at the jointing stage in both 2018 and 2019. The peroxidase activity declined with plant age in both leaves and roots, for example, and the catalase activity in roots 4- and 7-year old plants declined by 13.8% and 0.85%, respectively, compared to 3-year old plants at the heading stage in 2018. Therefore, the reduced capacity of the antioxidant system may lead to oxidative stress during plant aging process. Overall, the concentrations of plant hormones, auxin (IAA), gibberellin (GA), zeatin (ZT), and abscisic acid (ABA) were significantly lower in roots than in leaves. The IAA concentration in leaves and roots exhibited different patterns with plant age. The ZT concentrations in leaves of 3-year old plants was 2.39- and 2.62-fold of those in 4- and 7-year old plants, respectively at the jointing stage, and in roots, the concentration declined with plant age. The changes in the GA concentration with plant age varied between the physiological stages and between years. The ABA concentrations appeared to increase with plant age, particularly in leaves. In conclusion, the aging process of E. sibiricus was apparently associated with an increase in oxidative stress, a decrease of ZT and an increase of ABA, particularly in roots. These findings highlight the effects of plant age on the antioxidant and endogenous hormone activity of E. sibiricus. However, these plant age-related trends showed variations between plant physiological stages and between harvest years that needs to be researched in the future to develop strategies to manage this forage species.

Spectral light quality regulates the morphogenesis, architecture, and flowering in pepper (Capsicum annuum L.).

Transparent plastic films with poor light transmittance seriously affect the mass composition of visible light in many greenhouses, which leads to the reduction of photosynthesis in vegetable crops. Understanding the regulatory mechanisms of monochromatic light in the vegetative and reproductive growth of vegetable crops is of great importance for the application of light-emitting diodes (LEDs) in the greenhouse. In this study, three monochromatic light treatments (red-, green- and blue-light) were simulated by using LEDs to explore light quality-dependent regulation from the stage of seedling to flowering in pepper (Capsicum annuum L.). The results showed that light quality-dependent regulation guides the growth and morphogenesis in pepper plants. Red- and blue-light played opposite roles in determining the plant height, stomatal density, axillary bud growth, photosynthetic characteristics, flowering time and hormone metabolism, while green light treatment resulted in taller plants and fewer branches, which was similar to the red-light treatment. The weighted correlation network analysis (WGCNA) based on mRNA-seq results revealed that the two modules named "MEred" and "MEmidnightblue" were positively correlated with red- and blue-light treatment, respectively, exhibiting high correlations with the traits such as plant hormone content, branching and flowering. Moreover, our results suggest that the light response factor ELONGATED HYPOCOTYL 5 (HY5) is essential for blue light-induced plant growth and development by regulating photosynthesis in pepper plants. Hence, this study uncovers crucial molecular mechanisms of how light quality determines the morphogenesis, architecture, and flowering in pepper plants, thus providing a basic concept of manipulating light quality to regulate pepper plant growth and flowering under greenhouse conditions.

Systemic enantioselectivity study of penthiopyrad: enantioselective bioactivity, acute toxicity, degradation and influence on tomato.

In order to promote the green development of agriculture, it is important to study the enantioselective effect of chiral pesticides. The bioactivity of the chiral fungicide penthiopyrad (PEN) racemate and enantiomers against phytopathogens, toxicity to non-target organisms, effect on tomato fruit growth and maturation, and environmental fate in tomato cultivation were evaluated at an enantioselective level in this study. The results indicated that at the same efficacy, the optically pure S-(+)-PEN could lower the dosage of racemate by 20-96%. The S-enantiomer had low toxicity to earthworms. Besides, the S-(+)-PEN did not cause significant abiotic stress to the tomato and increased fruit fresh weight and size via modulating the contents of plant hormones. However, the content of hydrogen peroxide (H2 O2 ), superoxide (O2 - ) and malondialdehyde in the R-enantiomer treatment group was significantly higher than the control group. The effect of the racemate on tomato fruit was between the enantiomers. Furthermore, compared to R-(-)-PEN and racemate, the S-enantiomer degraded more quickly in tomato fruit, leaves, and soil, reducing the danger of human exposure. The S-enantiomer is highly effective and less toxic. The development of enantiomer pure S-(+)-PEN products might be an efficient and low-risk strategy. The results lay the foundation for comprehensive evaluation and proper application of PEN. © 2023 Society of Chemical Industry.
