Evaluation of the carbonylation of filamentous fungi proteins by dry immune dot blotting

The development of mycological gerontology requires effective methods for assessing the biological age of fungal cells. This assessment is based on the analysis of a complex of aging and oxidative stress markers. One of the most powerful such markers is the protein carbonylation. In this study, the already known method of dry immune dot blotting is adapted for mycological studies of the content of protein carbonyl groups. After testing the method on a number of filamentous fungi species, some features of the accumulation of carbonylated proteins in mycelium were established. Among these features: (i) a weak effect of exogenous oxidative stress on the accumulation of carbonyls in a number of fungi, (ii) reversibility of the carbonyl accumulation, (iii) possibility of arbitrary regulation of carbonyl content by fungus itself and (iv) the influence of hormesis. In addition, two polar strategies for the accumulation of carbonyl modification were revealed, named Id-strategy (Indifferent) and Cn-strategy (Concern). Thus, even the analysis of one marker allows making some preliminary general assumptions and conclusions. For example, the idea that fungi can freely regulate their biological age is confirmed. This feature makes fungi very flexible in terms of responding to environmental influences and promising objects for gerontology.

Effects of oxidative modification by malondialdehyde on the in vitro digestion properties of rice bran protein

As a secondary oxidation product derived during rice bran oxidative rancidity, malondialdehyde (MDA) was selected to study the impact of oxidative rancidity on the digestion properties of rice bran protein. MDA modification promoted the content of protein carbonyl groups and disulfide bonds from 2.0 to 10.34 nmol/mg, 12.04–16.51 nmol/mg, respectively. The results of FT-IR, intrinsic fluorescence spectra, surface hydrophobicity, molecular weight distribution and protein electrophoresis implied that MDA changed the secondary structure of rice bran protein, and promoted protein aggregation and cross-link through non-disulfide covalent bonds. Besides, increasing concentration of MDA caused the loss of degradation potential of protein, which accompanied by the loss of pepsin and trypsin digestibility from 33.86 to 24.29%, 70.10 to 47.05%, respectively. Furthermore, MDA treatment led to the decrease in antioxidant activities of digestion products, which might be attributed to the less formation of digestion products with molecular weight of 500–1500 Da. Correlation analysis between parameters of rice bran protein oxidation and protein digestion revealed that rice bran protein oxidation was significantly related to the reduction of digestion parameters. Overall, incubation with MDA altered the structure of rice bran protein, which had a significant correlation with the deterioration of digestion properties.

Effects of Excess Manganese on the Oxidative Status, and the Expression of Inflammatory Factors and Heat Shock Proteins in Cock Kidneys

Manganese (Mn) can have adverse effects on organisms as a result of heavy or chronic exposure, including neurological damage. This study examined the effects of chronic exposure to manganese chloride (MnCl2) on various biochemical indices of inflammatory cytokines, antioxidant enzymes, and heat shock proteins (HSPs) in the kidneys of Hy-line cocks. The exposures were carried out using 600, 900, or 1800mg/kg doses of MnCl2 administered for periods of 30, 60, and 90days. The exposure experiments indicated that Mn concentration in the kidneys increased over time and that Mn exposure potentially caused ultrastructural changes to the cells. Treatment with Mn was seen to increase the levels of various biomarkers, including protein carbonyl group content; DNA-protein cross-links (DPCs) and the mRNA expression of inflammatory factors such as tumor necrosis factor-α (TNF-α), nuclear factor-κB p50 (NF-κB p50), cyclooxygenase-2 (COX-2), and prostaglandin E synthase (PGES). The levels of other biomarkers were found to decrease as a result of Mn exposure, including the mRNA expression of oxidation indexes such as copper-zinc superoxide dismutase (CuZn-SOD), manganese superoxide dismutase (Mn-SOD), glutathione peroxidase (GSH-Px), and catalase (CAT). Accompanying the above changes, Mn exposure was seen to result in the relative mRNA and protein expression of HSPs 90, 70, 60, 40, and 27 increasing significantly. Thus, in cock kidneys, HSPs attenuated the biological changes caused by toxic exposure to Mn. This mechanism needs further exploration.

Biochemical and biomolecular effects induced by a static magnetic field in Saccharomyces cerevisiae: Evidence for oxidative stress.

Exposure to static magnetic fields (SMF) can cause changes in microorganism metabolism altering key subcellular functions. The purpose of this study was to investigate whether an applied SMF could induce biological effects on growth of Saccharomyces cerevisiae, and then to probe biochemical and bio-molecular responses. We found a decrease in growth and viability under SMF (250mT) after 6h with a significant decrease in colony forming units followed by an increase between 6 h and 9 h. Moreover, measurements of antioxidant enzyme activities (catalase, superoxide dismutase, glutathione peroxidase) demonstrated a particular profile suggesting oxidative stress. For instance, SOD and catalase activities increased in magnetized cultures after 9 h compared with unexposed samples. However, SMF exposure caused a decrease in glutathione peroxidase activity. Finally, SMF caused an increase in MDA levels as well as the content of protein carbonyl groups after 6 and 9 h of exposure.

Ethyl-acetate fraction of Trichilia catigua restores long-term retrograde memory and reduces oxidative stress and inflammation after global cerebral ischemia in rats

We originally reported that an ethyl-acetate fraction (EAF) of Trichilia catigua prevented the impairment of water maze learning and hippocampal neurodegeneration after transient global cerebral (TGCI) in mice. We extended that previous study by evaluating whether T. catigua (i) prevents the loss of long-term retrograde memory assessed in the aversive radial maze (AvRM), (ii) confers hippocampal and cortical neuroprotection, and (iii) mitigates oxidative stress and neuroinflammation in rats that are subjected to the four vessel occlusion (4-VO) model of TGCI. In the first experiment, naive rats were trained in the AvRM and then subjected to TGCI. The EAF was administered orally 30min before and 1h after TGCI, and administration continued once per day for 7days post-ischemia. In the second experiment, the EAF was administered 30min before and 1h after TGCI, and protein carbonylation and myeloperoxidase (MPO) activity were assayed 24h and 5days later, respectively. Retrograde memory performance was assessed 8, 15, and 21days post-ischemia. Ischemia caused persistent retrograde amnesia, and this effect was prevented by T. catigua. This memory protection (or preservation) persisted even after the treatment was discontinued, despite the absence of histological neuroprotection. Protein carbonyl group content and MPO activity increased around 43% and 100%, respectively, after TGCI, which were abolished by the EAF of T. catigua. The administration of EAF did not coincide with the days of memory testing. The data indicate that antioxidant and/or antiinflammatory actions in the early phase of ischemia/reperfusion contribute to the long-term antiamnesic effect of T. catigua.

Effects of Melaxen and Valdoxan on Free-Radical Oxidation in Rat Heart with Experimental Hyperthyroidism

The rate of free-radical oxidation in rat heart with experimental hyperthyroidism (EH) was estimated after Melaxen and Valdoxan administration. It was found that the content of protein carbonyl groups increased if the pathology developed in cardiomyocytes. This indicated that they were oxidized. Increased concentrations of primary lipid peroxidation products (diene conjugates), inhibition of aconitate hydratase activity, and accumulation of citrate were noted. Injection of Melaxen and Valdoxan to rats with EH normalized these parameters. The melatonin-correcting capability of the used drugs and the antioxidant activity of melatonin probably facilitated normalization of free-radical homeostasis in rat cardiomyocytes with the pathology.

Влияние мелаксена и вальдоксана на свободнорадикальное окисление в сердце крыс при экспериментальном гипертиреозе

Проведена оценка интенсивности свободнорадикального окисления в сердце крыс при введении мелаксена и вальдоксана при экспериментальном гипертиреозе. Установлено, что при развитии патологии в кардиомиоцитах происходило увеличение содержания карбонильных групп белков, что свидетельствует об их окислительной модификации. При этом отмечено возрастание концентрации первичных продуктов перекисного окисления липидов (диеновых конъюгатов), ингибирование активности аконитатгидратазы и накопление цитрата. Введение мелаксена и вальдоксана крысам при экспериментальном гипертиреозе приводило к изменению исследуемых параметров в сторону нормы. Вероятно, мелатонин-корригирующая способность используемых препаратов и антиоксидантная активность мелатонина способствовали нормализации свободнорадикального гомеостаза в кардиомиоцитах крыс с патологией.

The influence of 20 and 1% glucose and fructose, which were components of larval diet, on the level of oxidized proteins and lipids, low molecular mass antioxidant content as well as activities of antioxidant and associated enzymes in adult fruit fly Drosophila melanogaster were investigated. The restriction of carbohydrates in larval diet leads to oxidative stress in adult insects. It is supported by 40-50% increased content of protein carbonyl groups and by 60-70% decreased level of protein thiol groups as well as by a 4-fold increase of lipid peroxide content in 2-day-old flies of both sexes, developed on the diet with 1% carbohydrates. Oxidative stress, induced by carbohydrate restriction of the larval diet, caused the activation of antioxidant defence, differently exhibited in male and female fruit flies. Caloric restriction increased activity of superoxide dismutase and thioredoxin reductase associating only in males with 2-fold higher activity of NADPH-producing enzymes--glucose-6-phosphate dehydrogenase and isocitrate dehydrogenase. Carbohydrate restriction in the larval diet caused the increase of uric acid content, but the decrease in catalase activity in males. In females the values of these parameters were changed in opposite direction compared with males. The obtained results let us conclude the different involvement of low molecular mass antioxidants, glutathione and uric acid, and antioxidant enzyme catalase in the protection of male and female fruit fly macromolecules against oxidative damages, caused by calorie restriction of larval diet.

Dormancy removal of apple seeds by cold stratification is associated with fluctuation in H2O2, NO production and protein carbonylation level

Reactive oxygen (ROS) and nitrogen (RNS) species play a signaling role in seed dormancy alleviation and germination. Their action may be described by the oxidative/nitrosative “window/door”. ROS accumulation in embryos could lead to oxidative modification of protein through carbonylation. Mature apple (Malus domestica Borkh.) seeds are dormant and do not germinate. Their dormancy may be overcome by 70–90 days long cold stratification. The aim of this work was to analyze the relationship between germinability of embryos isolated from cold (5°C) or warm (25°C) stratified apple seeds and ROS or nitric oxide (NO) production and accumulation of protein carbonyl groups. A biphasic pattern of variation in H2O2 concentration in the embryos during cold stratification was detected. H2O2 content increased markedly after 7 days of seeds imbibition at 5°C. After an additional two months of cold stratification, the H2O2 concentration in embryos reached the maximum. NO production by the embryos was low during entire period of stratification, but increased significantly in germination sensu stricto (i.e. phase II of the germination process). The highest content of protein carbonyl groups was detected after 6 weeks of cold stratification treatment. Fluctuation of H2O2 and protein carbonylation seems to play a pivotal role in seed dormancy alleviation by cold stratification, while NO appears to be necessary for seed germination.

Protective Effect of Myricetin on Proteins and Lipids of Erythrocytes Membranes

Myricetin (3, 5, 7-Trihydroxy -2- (3, 4, 5-trihydroxyphenyl) - 4-chromenone), a naturally occurring flavonol, is a potent scavenger of reactive oxygen species (ROS) and effectively prevent erythrocyte oxidation. The protective effect of myricetin on proteins and lipids of erythrocytes membranes was investigated. Erythrocytes membranes were subjected to oxidative stress by incubating them with 10 −5 M tert-butylhydroperoxide; this caused a significant increase in membrane protein carbonyl group content and membrane lipid peroxides while caused a significant decrease of membrane total thiol group and Na + , K + -ATPase activity. The presence of myricetin in micromolar concentration in the incubation medium decreased significantly protein carbonylation, lipid peroxidation and caused an increase in total thiol group and Na + , K + ATP-ase activity.

Oxidative stress responses in blood and gills of Carassius auratus exposed to the mancozeb-containing carbamate fungicide Tattoo

Intensive use of pesticides, particularly dithiocarbamates, in agriculture often leads to contamination of freshwater ecosystems. To our knowledge, the mechanisms of toxicity to fish by the carbamate fungicide Tattoo that contains mancozeb [ethylenebis(dithiocarbamate)] have not been studied. The present study aimed to evaluate the effects of Tattoo on goldfish gills and blood, tissues that would have close early contact with the pollutant. Exposure of goldfish Carassius auratus to 3, 5 or 10mgL−1 of Tattoo for 96h resulted in moderate lymphopenia (by 8 percent) with a concomitant increase in both stab (by 66–88 percent) and segmented (by 166 percent) neutrophils. An increase in the content of protein carbonyl groups in blood (by 137–184 percent) together with decreased levels of protein thiols (by 23 percent) and an enhancement of lipid peroxide concentrations (by 29 percent) in gills after exposure to 10mgL−1 of Tattoo demonstrated the induction of mild oxidative stress in response to Tattoo exposure. At the same time, the activities of selected antioxidant enzymes were enhanced in gills: superoxide dismutase by 18–25 percent and catalase by 27 percent. A 34 percent increment in low molecular mass thiol concentrations (mainly represented by glutathione) also occurred in gills and could be related to increased activity (by 13–30 percent) of glucose-6-phosphate dehydrogenase. The results indicate that Tattoo exposure perturbs free radical processes, i.e. induces mild oxidative stress and enhances the activity of certain antioxidant and associated enzymes in goldfish gills. It is clear that goldfish respond to the presence of waterborne pesticide by adjusting antioxidant defenses through upregulation of activities of antioxidant and associated enzymes.

Electrochemical study of bovine serum albumin damage induced by Fenton reaction using tris (2,2′-bipyridyl) cobalt (III) perchlorate as the electroactive indicator

Oxidative protein damage is the most critical factor implicated in carcinogenesis and other disorders. However, the electrochemical detection of oxidative protein damage and the protections of protein from damage by antioxidants have been reported rarely. In this work, a novel, sensitive and inexpensive method for direct electrochemical detection of bovine serum albumin (BSA) damage in aqueous solutions is described. The simple electrochemical procedure has been constructed on the glassy carbon electrode (GCE) surface by BSA direct adsorption technique. Co(bpy)33+ was used as a redox indicator to monitor BSA damage induced by hydroxyl radical (OH), which was produced from Fenton reaction. The simplest fabrication procedure and the most classic method of free radical production were used to research the protein damage, demonstrating obvious virtues of brachylogy, rigor and precise. The electrochemical behaviors of the underlying electrodes were characterized by differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). Ultraviolet–visible (UV–vis) spectroscopy was used to verify the BSA damage with the universal index of the protein carbonyl group content. The optimizations of the Fe2+/H2O2 ratio and incubation time on BSA damage were explored. Moreover, the protections of BSA from damage by antioxidants were investigated. These conclusions demonstrated that the proposed method could be used to detect protein damage induced by Fenton reaction through a simple electrochemical approach.

An electrochemical biosensor for analysis of Fenton-mediated oxidative damage to BSA using poly-o-phenylenediamine as electroactive probe

A sensitive electrochemical procedure based on bovine serum albumin (BSA)/poly-o-phenylenediamine (PoPD)/carbon-coated nickel (C-Ni) nanobiocomposite film modified glassy carbon electrode (BSA/PoPD/C-Ni/GCE) has been developed to explore the electrochemical detection of BSA damage induced by hydroxyl radical. It is the first time that the electrochemical method has been applied for the analysis of Fenton-mediated oxidative damage to proteins. The hydroxyl radical was generated by Fenton reaction (Fe2+/H2O2), which was also validated by ultraviolet–visible (UV–vis) spectroscopy. The decrease in intensity of the PoPD oxidation signals was used as an indicator for the detection of BSA damage. Damage to BSA was also validated by horizontal Attenuation Total Reflection Fourier Transform Infra-red (ATR-FTIR) spectroscopy and the change of protein carbonyl group content achieved by UV–vis spectroscopy. Effects of H2O2 concentration, the ratio of Fe2+ and H2O2 and incubation time on BSA damage were examined. Protections of BSA from damage by antioxidants were also investigated. These conclusions demonstrated that the proposed electrochemical method is expected to the further application for protein damage studies.

Oxidative stress induced by the Fe/ascorbic acid system or model ischemia in vitro: effect of carvedilol and pyridoindole antioxidant SMe1EC2 in young and adult rat brain tissue.

New effective strategies and new highly effective neuroprotective agents are being searched for the therapy of human stroke and cerebral ischemia. The compound SMe1EC2 is a new derivative of stobadine, with enhanced antioxidant properties compared to the maternal drug. Carvedilol, a non-selective beta-blocker, possesses besides its cardioprotective and vasculoprotective properties also an antioxidant effect. We compared the effect of carvedilol and SMe1EC2, antioxidants with a similar chemical structure, in two experimental models of oxidative stress in young and adult rat brain tissue. SMe1EC2 was found to improve the resistance of hippocampal neurons to ischemia in vitro in young and even in 18-month-old rats and inhibited formation of protein carbonyl groups induced by the Fe2+/ascorbic acid pro-oxidative system in brain cortex homogenates of young rats. Carvedilol exerted a protective effect only in the hippocampus of 2-month-old rats and that at the concentration 10-times higher than did SMe1EC2. The inhibitory effect of carvedilol on protein carbonyl formation induced by the pro-oxidative system was not proved in the cortex of either young or adult rats. An increased baseline level of the content of protein carbonyl groups in the adult versus young rat brain cortex confirmed age-related changes in neuronal tissue and may be due to increased production of reactive oxygen species and low antioxidant defense mechanisms in the adult rat brain. The results revealed the new pyridoindole SMe1EC2 to be more effective than carvedilol in neuroprotection of rat brain tissue in both experimental models involving oxidative stress.

Implications of the Up-regulation of Genes Encoding Protein Degradation Enzymes and Heat Shock Protein 90 for Intertidal Green Macroalga Ulva fasciata Against Hypersalinity-Induced Protein Oxidation

The molecular acclimation of intertidal green macroalga Ulva fasciata Delile to high salinity stress were examined by the construction of a forward cDNA library via the suppressive subtractive hybridization between 30‰ and 90‰ (24 h) and by the time course dynamics of several abundantly expressed genes. Among the genes with known sequences, the expressed sequence tags are abundant in the function of protein synthesis (ribosomal protein) and destination. The cDNAs of ATP-dependent Clp protease (UfClpC), 20S proteasome β-subunit type 1 domain (UfPbf1), ubiquitin-conjugating enzyme E2 I (UfUbc9), and heat shock protein 90A (UfHsp90A) were cloned. UfClpC transcript increased 3 h after 90‰ treatment, followed by a decrease, while UfPbf1 and UfUbc9 transcripts increased after 12 h and decreased at 48 h. The transcripts of UfHsp90A increased 1 h after 90‰ treatment, followed by a drop and to the control level at 48 h. Protease activity increased 3 h after 90‰ treatment and decreased to the control level at 48 h. H₂O₂ contents increased 1 h after 90‰ treatment and then remained unchanged, but protein carbonyl group contents increased after 48 h. The treatments of reactive oxygen species scavengers partially alleviated 90‰ damage (partial growth rescue) and suppressed the increases in H₂O₂ content, protein carbonyl group content, protease activity, and UfClpC, UfPbf1, UfUbc9, and UfHsp90A transcripts by 90‰. The induction of specific chaperones and proteases at the molecular level for protein quality control can be considered as one of the molecular mechanisms of hypersalinity acclimation in U. fasciata.

Cytotoxicity of chromium ions may be connected with induction of oxidative stress

Chromium ions are frequently found in aquatic ecosystems and are known to be inducers of oxidative stress in fish solid tissues. The present study was designed to determine whether fish blood samples can be used to allow nonlethal diagnostic testing for chromium intoxication. First, we confirmed that 96 h exposures to water containing 10.0 mg L −1 chromium ions, either Cr 3+ or Cr 6+, induced oxidative stress in brain of goldfish ( Carassius auratus). Multiple blood parameters were then evaluated. Cr 6+ exposure triggered a 579% increase in the number of erythrocytes containing micronuclei, a frequently used marker of cellular toxicity. Leucocyte numbers were also perturbed by exposure to either Cr 3+ or Cr 6+ indicating that chromium ions could impair the immune system as well. The content of protein carbonyl groups, a marker of oxidative damage to proteins, was enhanced in fish plasma by exposure to either chromium ion and activities of catalase and lactate dehydrogenase also were affected. The data demonstrate that chromium ions induced oxidative stress in goldfish blood and were cytotoxic for erythrocytes. This indicates that analysis of plasma can be used as a good early nonlethal diagnostic marker of fish intoxication by transition metal ions.

Reduction of lipid and protein damage in patients with disorders of propionate metabolism under treatment: a possible protective role of l-carnitine supplementation

Disorders of propionate metabolism are autosomal recessive diseases clinically characterized by acute metabolic crises in the neonatal period and long-term neurological deficits whose pathophysiology is not completely established. There are increasing evidences demonstrating antioxidant properties for l-carnitine, which is used in the treatment of propionic and methylmalonic acidemias to increase the excretion of organic acids accumulated in tissues and biological fluids of the affected patients. In this work we aimed to evaluate lipid (malondialdehyde content) and protein (carbonyl formation and sulfhydryl oxidation) oxidative damage in plasma from patients with propionic and methylmalonic acidemias at the moment of diagnosis and during treatment with l-carnitine. We also correlated the parameters of oxidative damage with plasma total, free and esterified l-carnitine levels. We found a significant increase of malondialdehyde and carbonyl groups, as well as a reduction of sulfhydryl groups in plasma of these patients at diagnosis compared to controls. Furthermore, patients under treatment presented a marked reduction of the content of protein carbonyl groups, similar to controls, and malondialdehyde content in relation to patients at diagnosis. In addition, plasma total and free l-carnitine concentrations were negatively correlated with malondialdehyde levels. Taken together, the present data indicate that treatment significantly reduces oxidative damage in patients affected by disorders of propionate metabolism and that l-carnitine supplementation may be involved in this protection.

Protective effect of resveratrol on formation of membrane protein carbonyls and lipid peroxidation in erythrocytes subjected to oxidative stress

Many of the biological actions of resveratrol have been attributed to its antioxidant properties. In this work, we subjected human erythrocytes to in vitro oxidative stress by incubating them with tert-butylhydroperoxide (t-BHP). This caused a significant increase in the malondialdehyde (MDA) level and the protein carbonyl group content above the basal values. The presence of trans-resveratrol at micromolar concentrations in the incubation medium protected the erythrocytes from t-BHP-induced oxidative stress, as evidenced by the decrease in the MDA level and the protein carbonyl group content. The effect of resveratrol was concentration and time-dependent. Our findings help to explain some of the beneficial effects of resveratrol.

Buffer modulation of menadione-induced oxidative stress in Saccharomyces cerevisiae

The objective of this study was to compare, in vivo, the effects of bicarbonate and phosphate buffers on survival and menadione-induced oxidative stress in yeast cells. The latter were treated with different concentrations of menadione in the presence of these two buffers. At 25 mM concentration of buffers, menadione only slightly reduced yeast surviving; at 50 mM concentration, cell killing by menadione was much more pronounced in bicarbonate than in phosphate buffer. Although the content of protein carbonyl groups did not show development of oxidative stress under menadione-induced stress, inactivation of aconitase and decrease in glutathione level mirrored its induction. However, cellular glutathione and aconitase activity decrease did not correlate with yeast survival. In vitro, aconitase was more quickly inactivated in 50 mM carbonate, than in 50 mM phosphate buffer. The possible involvement of the carbonate radical in these processes is discussed.

Hypersalinity and hydrogen peroxide upregulation of gene expression of antioxidant enzymes in Ulva fasciata against oxidative stress.

The modulation of manganese superoxide dismutase (MnSOD), FeSOD, ascorbate peroxidase (APX), glutathione reductase (GR), and catalase (CAT) gene expression and activities and antioxidants in Ulva fasciata against hypersalinity (90 per thousand)-induced oxidative stress was studied. Increases in H(2)O(2) contents but no changes in lipid peroxidation and protein carbonyl group contents suggest oxidative damage did not occur in 90 per thousand condition. Antioxidants were consumed for reactive oxygen species (ROS) scavenging indicated by decreased ascorbate and glutathione contents by 90 per thousand. Antioxidant enzymes were differently expressed by 90 per thousand for ROS removal. MnSOD activity and transcript increased 1 h after 90 per thousand treatment with a peak at hour 3, while FeSOD activity increased fast to the plateau after 1 h and its transcript increased after 3 h. APX activity increased 1 h after 90 per thousand but its transcript rose till 3 h, and GR activity increased after 1 h with a peak at hour 3 but its transcript increased till 3 h. CAT activity and transcript increased after 12 h. Enzyme activity is transcriptionally regulated by 90 per thousand except a fast increase in FeSOD, APX, and GR activities during 1 h. APX is responsible for early H(2)O(2) decomposition while CAT scavenges H(2)O(2) in the later period. The inhibition of 90 per thousand induced increase of H(2)O(2) content and FeSOD activity and transcript by treatment of a H(2)O(2) scavenger, dimethylthiourea, and the increase of FeSOD transcript of 30 per thousand grown thalli by H(2)O(2) treatment suggest that H(2)O(2) mediates the upregulation of FeSOD by hypersalinity while other enzymes is modulated by factors other than H(2)O(2).
