Effects of dietary manganese supplementation on the growth performance, tissue manganese content and antioxidant capacity of coho salmon (Oncorhynchus kisutch) post-smolts

A 10-week growth trial was carried out to study the impact of dietary manganese (Mn) level on growth, tissue Mn content and anti-oxidative capacity of coho salmon (Oncorhynchus kisutch) post-smolts (186.51 ± 1.43 g). Six iso-nitrogenous (42.40% Protein) and iso-lipidic (10.27% Lipid) purified diets based on casein-gelatin were formulated to contain varied graded (MnSO4·H2O as a Mn source) levels of 0.40, 9.50, 16.80, 25.80, 33.40 and 41.20 Mn mg/kg. Fish were reared in a freshwater flow-through rearing system and fed to apparent satiation 3 times daily. No significant effects were found on the survival rate, morphological indices, whole-body proximate composition (p > .05) among the groups. However, higher dietary Mn supplementation level than 25.80 mg/kg improved the growth performance, increased the deposition of Mn in whole-body and vertebral, improved hepatic digestive enzyme activity, enhanced anti-oxidative enzyme activities, and decreased the malondialdehyde content in liver and serum (p < .05). Hepatic and serum total antioxidant capacity, glutathione peroxidase, glutathione reductase, total superoxide dismutase, Mn-superoxide dismutase and catalase activities were enhanced with increasing level of dietary Mn, and then stabilized when dietary Mn level up to 25.80 mg/kg. Using the cubic analysis (based on specific growth rate and feed conversion ratio), the optimal dietary Mn levels were 33.25 and 29.55 mg/kg, respectively. Using the broken-line analysis (based on the deposition of Mn, Mn-superoxide dismutase in liver and serum), the dietary Mn requirements of coho salmon post-smolts were estimated to be 24.52, 26.34, 24.74 and 26.05 mg/kg, respectively.

Influences of oxidative stress and energy balance on pregnancy rates after the transfer of embryos to repeat-breeder Japanese Black cattle.

The purpose of this study is to reassess our previously reported findings on the diminished pregnancy rate of embryo transfer (ET) in summer for repeat-breeder (RB) cows compared with other seasons, with a focus on oxidative stress and energy balance. The study involved Japanese Black cattle, including 224 heifers (aged <3years) and 278 (aged <9years) cows with one or more deliveries, defined as RB animals. Evaluation of the effects of season on pregnancy rates revealed significantly lower rates in RB cows during summer compared with spring and autumn. Moreover, serum malondialdehyde (MDA) concentration in RB cows during summer was significantly higher than in winter, with no difference in RB heifers. Seasonal effects on serum nonesterified fatty acid (NEFA) concentration in both RB heifers and RB cows showed no significant differences. However, median MDA and NEFA concentrations in RB cows were significantly elevated compared with RB heifers, suggesting that factors contributing to low fertility should consider parity. Furthermore, our study indicated that RB cows were under oxidative stress, as demonstrated by significantly elevated MDA concentrations compared with normally reproducing cows in summer. Further investigation is necessary to gain a better understanding of these observations in the future.

Changes in Biochemical Components of Obesity in Wistar Rats Fed Mentha Piperita-Supplemented High-Fat Diet

The research is designed to assess changes in biochemical components of obesity in Wistar rats fed peppermint-supplemented high-fat diet. Twenty (20) female Wistar rats of about 117.88±10.82g were divided into four (4) groups of five (5) rats each and randomly assigned to the formulated diets. The groups had similar average weights. Groups A and B were fed standard diet and High-fat diet respectively, while groups C and D were respectively fed 4% peppermint-supplemented standard diet and 4% peppermint-supplemented high-fat diet. The changes in weight and blood glucose level were monitored on weekly basis. At the end of six (6) weeks of the experiment, organ to body weight ratio, serum insulin level and HOMA-IR were determined. Markers of hepatic functions (transaminases, total bilirubin, albumin, globulin, conjugated bilirubin and total protein), markers of renal functions (electrolyte concentrations, urea and creatinine), serum activities of antioxidant enzymes (catalase, glutathione peroxidase, reduced glutathione and superoxide dismutase) and serum concentrations of malondialdehyde (MDA) were also determined. No significant (p≤0.05) differences were observed among the groups in weight gain, organ to body weight ratio, blood glucose levels, activities of antioxidant enzymes and serum MDA concentrations. The group fed 4% peppermint-supplemented high-fat diet had significantly (p≤0.05) higher activities of ALT, AST, total cholesterol and low-density lipoprotein cholesterol but significantly (p≤0.05) lower high-density lipoprotein cholesterol when compared with the group fed standard diet or high-fat diet. Serum creatinine and bicarbonate were significantly (p≤0.05) lower in the group fed 4% peppermint-supplemented high-fat diet compared with the group fed the standard diet. It is therefore concluded that including peppermint leaf powder in the diet for a period of 6 weeks may not have a significant impact on the body weight, organ development and glucose utilisation. However, it is important to note that consumption of high-fat diet supplemented with peppermint might lead to dyslipidemia, liver and kidney dysfunctions without altering oxidative balance in Wistar rats.

Resveratrol Administration Ameliorates Hepatotoxicity in Mercuric Chloride-‎Induced Liver Injury in Rats

Mercuric chloride (HgCl2) pollution and poisoning has been a worldwide health ‎concern for decades, especially after the industrial revolutions. The aim of this study ‎was to investigate the role of resveratrol in reversing the deleterious effects of ‎HgCl2 exposure to resume the normal functions of hepatocyte. To achieve the study, ‎mature Sprague Dawley rats were assigned to five groups. Negative control group ‎‎(C) kept without any treatment; vehicle-treated group (D) received dimethyl ‎sulfoxide (DMSO); resveratrol-treated group (R), received 100 mg/kg of resveratrol; ‎HgCl2-intoxicated group (HD), received i.p. injection of HgCl2 at a dose of 1 mg/kg ‎for 30 consecutive days along to oral gavage of DMSO; and finally HgCl2-‎intoxicated group treated with resveratrol (HR) as same treatment strategy of R-‎group. At the endpoint of the experiment, blood samples were collected for ‎biochemical liver function tests along with serum concentrations of ‎malondialdehyde (MDA), glutathione (GSH), body weight, as well as ‎histopathological investigation was done too. Study results revealed a significant ‎‎(P&lt;0.05) elevation in serum AST, ALP, GGT, and MDA in HD group in comparison ‎with HR group. However, resveratrol treatment has led to a significant (P&lt;0.05) ‎increase in serum levels of GSH in HR group in comparison with the HD group. ‎Histopathological sections showed vacuolar degeneration in HD hepatocytes while ‎resveratrol treatment protected the hepatocytes against the chemical injury. ‎Altogether, It is concluded that resveratrol administration has the ability to increase ‎the resistance of liver against the HgCl2-induced hepatotoxicity via increase the ‎antioxidant yields such as GSH resulted in reduction of hepatocellular texture ‎damage.

ANTIOXIDANT EFFECTIVENESS OF THE PROPHYLACTIC COMPLEX WITH QUERCETIN IN OVARIECTOMIZED RATS

Introduction. Premature ovarian failure is characterized by the onset of menopause before age 40 and is accompanied by a decrease in estrogen levels in the body. This condition has a negative impact on almost all physiological systems of the body. Recently, there has been an increase in the number of patients with this pathology, which is associated with the spread of medical intervention in the reproductive sphere, environmental pollution, and stress. Aim. The purpose of this study was to experimentally clarify the state of such indicators of the antioxidant-prooxidant system as catalase, malondialdehyde, antioxidant-prooxidant index in blood serum, mucous membranes of the digestive tract, liver and alveolar process of the lower jaw of female rats after surgical menopause (ovariectomy) in in combination with a diet deficient in calcium and protein composition, as well as establishing the antioxidant efficiency of a complex based on quercetin, vitamins and macro and microelements. Methods. The research was conducted on 24 female Wistar rats aged 2.5 months, which were divided into 3 groups (8 in each): 1 – intact animals; 2 – animals in which surgical menopause was modeled by ovariectomy and kept on a diet with inferior composition; 3 – animals that, against the background of ovariectomy, were prophylactically administered a complex of quercetin, vitamins, macro- and microelements at a dose of 500 mg/kg. After 4 months, the animals were removed from the experiment. Malondialdehyde (MDA) content, catalase activity, and antioxidant-prooxidant index (API) were determined in blood serum, homogenates of mucous membranes of the digestive tract, liver, and alveolar process. Results. The analysis of the data obtained by us showed that in the group of rats with which the pathology was modeled, there was a drop in the activity of the catalase enzyme in the mucous membranes of the digestive tract and liver from 5.61% to 38.49%, an increase in the MDA content from 25.78% to 50.00% and a decrease in the API index from 25.08 to 56.24% relative to the indicators of the group of intact rats with a high level of reliability (p &lt; 0.05–0.001). The content of MDA in the blood serum increased by 31,91%, and in the bone tissue of the jaws – by 23,28%, API was lower in the blood by 16,45% and by 10,39% in the bone tissue (p &lt; 0,05–0,001). The obtained data allow us to assert about the increased formation of reactive oxygen species and inhibition of their inactivation processes. It should be noted that the activity of catalase in the blood serum and alveolar process of the jaws of animals with pathology, on the contrary, increased by 10.19% and 10.78%, respectively, which indicates compensatory antioxidant capabilities in these tissues. When using the biologically active complex, there was an increase in catalase activity in the mucous membranes of the digestive tract by 5,63–40,8%, a decrease in the activity of MDA by 17.84–23.03%, and an increase in the API index by 27,93–80,84% (p&lt;0,05–0,001). In the bone tissue of the jaws of animals under the influence of the complex, the activity of catalase increased by 7,52%, and API – by 33,3%, while the content of MDA decreased by 19,43%. In the blood of animals with pathology, the complex of drugs contributed to a decrease in the MDA level by 17,38%, an increase in the activity of catalase by 6,16% and the API index by 28,44%. Based on the above, it can be stated that the complex developed by us has a powerful antioxidant effect in conditions of estrogen, dietary protein and calcium deficiency in female rats. Conclusions. Menopause of surgical genesis in combination with nutritional deficiency of protein and calcium contributed to suppression of antioxidant protection against the background of activation of pro-oxidant processes. The use of a biologically active complex with quercetin, vitamins, calcium citrate, macro- and microelements increased the activity of the antioxidant defense system directed against free radical processes.

Hydrogen attenuates endothelial glycocalyx damage associated with partial cardiopulmonary bypass in rats.

Cardiopulmonary bypass (CPB) causes systemic inflammation and endothelial glycocalyx damage. Hydrogen has anti-oxidant and anti-inflammatory properties; therefore, we hypothesized that hydrogen would alleviate endothelial glycocalyx damage caused by CPB. Twenty-eight male Sprague-Dawley rats were randomly divided into four groups (n = 7 per group), as follows: sham, control, 2% hydrogen, and 4% hydrogen. The rats were subjected to 90 minutes of partial CPB followed by 120 minutes of observation. In the hydrogen groups, hydrogen was administered via the ventilator and artificial lung during CPB, and via the ventilator for 60 minutes after CPB. After observation, blood collection, lung extraction, and perfusion fixation were performed, and the heart, lung, and brain endothelial glycocalyx thickness was measured by electron microscopy. The serum syndecan-1 concentration, a glycocalyx component, in the 4% hydrogen group (5.7 ± 4.4 pg/mL) was lower than in the control (19.5 ± 6.6 pg/mL) and 2% hydrogen (19.8 ± 5.0 pg/mL) groups (P < 0.001 for each), but it was not significantly different from the sham group (6.2 ± 4.0 pg/mL, P = 0.999). The endothelial glycocalyces of the heart and lung in the 4% hydrogen group were thicker than in the control group. The 4% hydrogen group had lower inflammatory cytokine concentrations (interleukin-1β and tumor necrosis factor-α) in serum and lung tissue, as well as a lower serum malondialdehyde concentration, than the control group. The 2% hydrogen group showed no significant difference in the serum syndecan-1 concentration compared with the control group. However, non-significant decreases in serum and lung tissue inflammatory cytokine concentrations, as well as in serum malondialdehyde concentration, were observed. Administration of 4% hydrogen via artificial and autologous lungs attenuated endothelial glycocalyx damage caused by partial CPB in rats, which might be mediated by the anti-inflammatory and anti-oxidant properties of hydrogen.

Effects of exposing Japanese quail eggs to a low dose of gamma radiation and in ovo feeding by two sources of trace elements on embryonic development activities

The present study investigated the influence of exposing quail eggs to low-dose gamma radiation (GR) and in ovo feeding with 2 sources of a mixture of trace elements (Zn, Fe, and Cu), including sulfate (TES) and loaded with montmorillonite (TEM), on embryonic development activities and prehatch quality. A total of 960 eggs on the seventh day of incubation were randomly divided into 6 groups (160 eggs/group) with 4 replicate of 40 eggs in each. A 3 × 2 factorial arrangement experiment was performed and included 3 sources in ovo feeding with a mixture of trace elements (Zn, Fe, and Cu), including 0 mg/egg, 50 mg TES/egg, and 50 mg TEM/egg with egg irradiation using 0 and 0.2 Gy from GR. Eggs injected with 50 mg TEM/egg and exposed to 0.2 Gy from GR (TEM/GR) was significantly (P ≤ 0.05 and 0.01) higher in hatchability, hatch body weight, and relative organ weight (liver, gizzard, proventriculus, heart, and intestine). The obtained results indicated significant (P ≤ 0.05) decreased in the serum concentration of malondialdehyde (MDA) in TEM/GR group. There was significant (P ≤ 0.05) increased of catalase (CAT) activity and the concentrations of growth hormone (GH) and insulin-like growth factor-1 (IGF-1) in TEM/GR group; however; total antioxidant capacity (T-AOC) was significant (P ≤ 0.05) increased in CT/GR group. Serum concentrations of immunoglobulin M (IgM) (P ≤ 0.05) and tumor necrosis factor-alpha (TNF-α) were increased in the TEM/CR group; the concentration of transforming growth factor beta (TGF-β) significant (P ≤ 0.05) increased in the TEM/GR group; and interleukins (IL6 and IL10) showed no significant differences among the groups. Our results showed increase in thyroxine and myostatin concentrations with TES/CR and CT/GR of our study groups, respectively. The relative mRNA expression levels of the GH, IGF-1, and Fas cell surface death receptor (FAS) genes were significantly (P ≤ 0.05 and 0.01) upregulated in the liver tissue of the TEM/GR group compared with the other groups. In conclusion, TEM/GR was the best treatment for improving prehatch quality, increasing serum antioxidant enzyme activities, and promoting the expression of growth and immune genes in fertilized quail eggs.

Probiotic-Fermented Distillers Grain Alters the Rumen Microbiome, Metabolome, and Enzyme Activity, Enhancing the Immune Status of Finishing Cattle.

A total of 30 Simmental crossbred cattle (6.50 months old, 265.0 ± 22.48 kg) were randomly divided into three groups, with 10 heads per group, and fed for 45 days. The diet treatments consisted of the Control group without PFDG supplementation, the PFDG-15% group with 15% PFDG substituting for 15% concentrate, and PFDG-30% group with 30% PFDG substituting for 30% concentrate. The results showed that compared with the Control group, the average daily gain (ADG) of the cattle in the PFDG-30% group decreased significantly (0.890 vs. 0.768 kg/d, p = 0.005). The serum malondialdehyde content of cattle in the PFDG-15% and PFDG-30% groups decreased significantly (p = 0.047) compared to that of the Control group. However, the serum superoxide dismutase activity of cattle in the PFDG-30% group was significantly higher than that of the Control group (p = 0.047). Meanwhile, both the PFDG-15% and PFDG-30% groups (1758.47 vs. 2061.30 μg/mL) showed higher serum levels of immunoglobulin G, while the interleukin-10 concentration was lower in the PFDG-30% group (p = 0.027). In addition, the PFDG-15% and PFDG-30% groups shifted the rumen microbiota by improving the abundances of F082 (related to propionic acid production) and fiber-degrading bacteria (Lachnospiraceae_UGG-009 and Prevotellaceae_UCG-001) and reducing the abundance of the disease-associated bacteria Selenomonas. A Kyoto encyclopedia of genes and genomes (KEGG) analysis illustrated that three key metabolic pathways, including phenylalanine metabolism, pyrimidine metabolism, and tryptophan metabolism, were enriched in the PFDG-15% group, but eight key metabolic pathways, including arachidonic acid metabolism, were enriched in the PFDG-30% group. Importantly, both the PFDG-15% and PFDG-30% groups increased (p < 0.01) the activities of cellulase, lipase, and protease in the rumen. Finally, the different bacterial abundance in the rumen was associated with changes in the ADG, serum antioxidant capacity, immune status, rumen enzyme activity, and metabolites. These results suggest that PFDG alters rumen microbiome abundance, metabolome, and enzyme activity for enhancing serum antioxidant capacity and the immune status, but when the supplemental level reaches 30%, it has a negative effect on ADG and the anti-inflammatory factors in finishing cattle.

Pde5 Inhibition Reduced Blood Pressure and Alleviated Target Organ Damage in Chronic Intermittent Hypoxia.

The role of phosphodiesterase 5 (Pde5) in obstructive sleep apnea (OSA) induced damage remains unclear. Our study aimed to investigate the role of Pde5 in chronic intermittent hypoxia (CIH) model. C57BL/6J wild-type (WT) mice (n=48) and Pde5 knockout (Pde5-/-) mice (n=24) were randomly assigned to CIH group and room air (RA) group. After 6 weeks, some WT mice (n=24) in CIH group were given sildenafil or saline gavage for another 4 weeks. Blood pressure was regularly measured during the experiment. Echocardiography was used to estimate cardiac function. We collected organs from each group of mice and measured their physical indicators. Histochemical staining was used to explore the size of cardiomyocyte and fibrosis area of various organs. Cyclic guanosine monophosphate (cGMP) and Malondialdehyde (MDA) concentrations in serum were measured by ELISA assay. Compared to the RA-treated group, the 6-week CIH resulted in a significant increase in blood pressure, altered heart structure and reduced serum cGMP in WT mice. Pde5-/- mice and sildenafil intragastric administration significantly reduced systolic blood pressure in CIH condition and attenuated the damage of target organs. In CIH model, we found that the cardiomyocyte size and fibrosis area of heart and kidney significantly reduced in Pde5-/- groups. Besides, endogenous and exogenous inhibition of Pde5 reduced MDA level and inflammatory and oxidative stress markers expression in CIH condition. In the present study, we found that Pde5 inhibition could reduce blood pressure and alleviate target organ damage in the CIH model, which may be mediated through the oxidative stress pathway.

Exploring fibrous ingredients for fish: The case of feeding sugar beet pulp to tambaquí (Colossoma macropomum)

For a long time, co-products of food processing have been used in animal feed, but far less in fish because of their assumed inability to cope with high-fiber diets. Research on feeding co-products to fish species that naturally consume fibrous diets are yet lacking. We here evaluated the impact of sugar beet pulp in the diet of tambaquí on nutrient metabolism, oxidative stress, inflammation, and intestinal histomorphometry. A total of 18 tambaquí fish (1616 ± 107 g; 2 years old) were randomly divided over 6 similar tanks with 3 fish per tank and randomly attributed to one of the six dietary treatments 0, 5, 10, 15, 20 and 25 % beet pulp addition and reared for 8 weeks. Water quality parameters (pH, NH3–N, EC, TDS, DO, and temperature) were assessed and recorded twice a week for each tank. A quadratic increase in intestinal villus length, paravilli and absorptive surface were observed with beet pulp addition. Ammonia and pH as quality indicators were significantly changing with beet pulp addition. A higher supply of glucogenic substrate to the citric acid cycle was noticed with beet pulp addition due to the positive correlation with blood propionylcarnitine: acetylcarnitine ratio while there was no effect on ketone body synthesis as measured through the 3-hydroxybutyrylcarnitine: acetylcarnitine ratio. No pronounced change of serum and whole fish histamine and lowered concentrations of serum malondialdehyde were observed with beet pulp addition. In conclusion, beet pulp induced a marked increase in intestinal villus architecture without signs of inflammation or oxidative stress. Large-scale studies need to clarify if these features lead to improved growth performance but this work opens options for further study. The non-linear pattern of some blood components with increasing beet pulp may call for future optimal dosing and feed form of beet pulp together.

Effect of guanidinoacetic acid on performance, egg quality, yolk fatty acid composition, and nutrient digestibility of aged laying hens fed diets with varying substitution levels of corn with low-tannin sorghum

A study was conducted to evaluate the efficiency of guanidinoacetic acid (GAA) in diets containing varying levels of corn replacement with low-tannin sorghum (LTS) for laying hens in the later stage of production. In a 12-wk study, a total of 288 laying hens at 52 wk of age were divided into 6 treatment groups. Each treatment group had 8 replicates, each of which consisted of 6 hens. A 2 × 3 factorial design was used to investigate the impact of substituting corn with LTS at 3 levels (100% corn, 50% LTS, and 100% LTS) with 2 doses of GAA supplementation (0 and 0.6 g/kg). The results indicate that there were interaction effects (P < 0.05) between diet type and GAA supplementation on protein digestibility and AMEn, with the GAA supplement being more effective in the 100% LTS group. Replacing corn with LTS at both levels had no negative effects on performance and metabolic profile. In contrast, the 100% LTS diet increased monounsaturated fatty acids in the yolk (P < 0.05), but decreased the yolk color index, the ratio of polyunsaturated fatty acids (PUFA) to saturated fatty acids (SFA) in the yolk, ileal digestibility of energy, and AMEn when compared to the 100% corn diet (P < 0.05). Regardless of the diet, dietary supplementation with GAA resulted in increases (P < 0.05) in shell-breaking strength, the PUFA to SFA ratio in egg yolk, and concentrations of creatine and nitric oxide in serum. There was also a decrease (P < 0.05) in serum malondialdehyde concentration with GAA supplementation. In conclusion, the positive effects of GAA on protein digestibility and AMEn were found to be more pronounced when corn was completely replaced with LTS. However, the positive effects of GAA on egg-laying performance, eggshell quality, antioxidant status, and yolk fatty acid composition remained consistent regardless of the extent to which corn was substituted with LTS.

Effects of a mixture of glycerol monolaurate and cinnamaldehyde supplementation on laying performance, egg quality, and antioxidant status in laying hens.

This study aimed to determine the effects of a mixture of glycerol monolaurate and cinnamaldehyde (GCM) supplementation on the laying performance, egg quality, antioxidant capacity, and serum parameters of laying hens. A total of 1120 14-week-old Jingfen-1 strain laying hens with similar performance were randomly allocated to four dietary treatments: control, and GCM groups supplemented with 250, 500, or 1000 mg kg-1 for 12 weeks. Compared with the control group, GCM-supplemented groups significantly reduced (P < 0.05) the rate of unqualified eggs of laying hens aged 17-24 weeks. Supplementation of GCM significantly increased (P < 0.05) yolk color and serum glutathione peroxidase (GSH-Px) activity but decreased (P < 0.05) the hydrogen peroxide (H2 O2 ) content in the serum of laying hens at the age of 20 weeks. Furthermore, groups supplemented with GCM showed a significant increase (P < 0.05) in Haugh unit, yolk color, activities of total superoxide dismutase and GSH-Px, and the glucose content in serum, and a decrease (P < 0.05) in the content of urea nitrogen and H2 O2 and malondialdehyde in serum of laying hens at the age of 24 weeks. 500 mg kg-1 GCM supplementation significantly increased (P < 0.05) the number of large white follicles and 1000 mg kg-1 GCM supplementation decreased the number of large yellow follicles in 28-week-old laying hens. These results indicated that GCM supplementation has positive effects on reducing egg loss and improving egg quality in the early laying period of laying hens. © 2023 Society of Chemical Industry.

Glutathione-rich yeast hydrolysate makes the contributions to growth performance, healthy of Pacific white shrimp (Litopenaeus vannamei), and helps shrimp resist nitrite stress

Seven diets were formulated to investigate the effects of glutathione-rich yeast hydrolysate (GYH) on growth performance, antioxidant capacity, and innate immunity of Litopenaeus vannamei. Based on the control diet (28% fishmeal content, CON), six diets supplement with GYH 0% (G0), 0.3% (G0.3), 0.5% (G0.5), 0.7% (G0.7), 0.9% (G0.9) and 1.1% (G1.1) were prepared after cottonseed protein concentrate replacing fishmeal 28.5% of the CON. After the 8 weeks feeding experiment, the shrimp fed diet containing 0.5%− 0.9% GYH obtained better growth performance compared to the CON and G0 groups, as well as the significantly lower feed conversion rate (P < 0.05). The intestinal villus height of shrimp fed diet with 0.3–0.9% GYH was significantly higher than the G0 group (P < 0.05). Decreased serum malondialdehyde content and significantly increased total antioxidant capacity, reduced glutathione content, activity of glutathione peroxidase and phenoloxidase in the hepatopancreas were found in shrimp fed diet with 0.7–0.9% GYH (P < 0.05). In addition, supplement 0.5–0.9% GYH significantly upregulated the expression levels of genes igf-1, pi3k, akt, tor, s6k1, and 4ebp1 in muscle and increased the crude protein content of the whole shrimp (P < 0.05). The expression levels of genes lgbp, proPO and lzm in the hepatopancreas were significantly upregulated in shrimp fed 0.7% GYH supplement (P < 0.05). Serum CAT activity in the G0.9 group was significantly higher than that of the G0 group before and after nitrite stress (P < 0.05). The shrimp fed diet supplemented with 0.5–1.1% GYH exhibited better intestinal tissue structure compared to groups CON and G0 after nitrite stress. In conclusion, the optimum level of dietary GYH was 0.67% based on the analysis of the quadratic term fitting results of weight gain rate. The diets supplemented with appropriate GYH could contribute to the growth and health of shrimp.

Effects of Fermented Soybean Meal Substituting Plant Protein and Fish Meal on Growth, Flesh Quality, and Intestinal Microbiota of Largemouth Bass (Micropterus salmoides)

This study investigated the effects of replacing soybean meal (SM), corn gluten meal (CGM), and fish meal (FM) with fermented soybean meal (FSM) on growth performance, flesh quality, and intestinal microbiota of largemouth bass. In a diet with 400 g/kg FM inclusion (FSM-0), FSM was used to substitute 120 g/kg SM, 120 g/kg SM + 80 g/kg CGM, and 65 g/kg FM at the inclusion of 100 g/kg (FSM-10-SM), 200 g/kg (FSM-20), and 100 g/kg (FSM-10-FM), respectively. Then, largemouth bass weighing 225.7 ± 2.6 g was fed the four diets for 60 days. The WG of FSM-20 group was increased by 11.8% ( P &lt; 0.05 ), while FSM-10-SM and FSM-10-FM groups did not differ significantly from FSM-0 group in growth performance. Compared with the FSM-0 group, the flesh hardness, shear force, total collagen, hot soluble collagen, and total free amino acid contents of FSM-20 group were significantly increased, and FSM-10-SM group also presented higher flesh hardness and shear force. The flesh superoxide dismutase (SOD) and total antioxidant capacity (TAOC) activities in FSM-20 and FSM-10-FM groups and glutathione peroxidase (GSH-Px) activity in FSM-10-FM group were increased ( P &lt; 0.05 ), while the lactic acid content in FSM-20 group was decreased, when compared to FSM-0 group ( P &lt; 0.05 ). In serum biochemical indexes, the glucose, total protein, cholesterol and malondialdehyde contents in the three FSM groups, and the triglyceride content in FSM-10-SM and FSM-20 groups were significantly lower than those of FSM-0 group ( P &lt; 0.05 ). In intestinal histology, the intestinal villus width, muscle thickness in FSM-10-SM group and intestinal villus width in FSM-20 group were significantly higher than those of FSM-0 group. In intestinal microbiota, the three FSM groups showed higher abundance of Firmicutes and lower abundance of Proteobacteria than FSM-0 group. In summary, the replacement of 120 g/kg SM or 65 g/kg FM with 100 g/kg FSM (FSM-10-SM and FSM-10-FM groups) did not exhibit significant effect on growth performance, but partially improved the flesh quality of largemouth bass. The substitution of 120 g/kg SM + 80 g/kg CGM with 200 g/kg FSM (FSM-20 group) significantly improved weight gain and flesh quality of largemouth bass.

148 Evaluation of Multicarbohydrase Supplementation in Soluble and Insoluble Fiber-Based sow Diets on Markers of Immune Function, Inflammation, and Oxidative Status at mid and Late Gestation

Abstract Carbohydrase supplementation in soluble and insoluble fiber-based diets can improve the health, gastrointestinal function, and livability of growing pigs. However, its efficacy in sow diets is incompletely understood. The experimental objective was to investigate the effects of soluble (20% sugar beet pulp) versus insoluble (20% reduce oil corn DDGS) dietary fiber with or without multicarbohydrase supplementation on systemic markers of gut barrier integrity, inflammation, and oxidative stress of sows at mid and late gestation. On d 28 of gestation, 36 confirmed gestating sows (186 ± 4.6 kg BW) were blocked by parity and randomly assigned to 1 of 4 diets (n = 8) in a 2×2 factorial arrangement. Factors included fiber type of either insoluble fiber (IF; 16.91 IDF%) or soluble fiber (SF; 5.26 SDF%) and with (+) or without (-) enzyme supplementation (0.05%, Rovabio Advance P10; Adisseo, Antony, France). Diets were fed from d 28 to d 109 of gestation at a feeding level of 2.4 kg/d. On d 50 and d 99 of gestation, before an 8-d metabolism collection period, serum and plasma were collected via jugular venipuncture and analyzed for malondialdehyde, total antioxidant capacity (TAC), lipopolysaccharide binding protein (LBP), and a 13-plex cytokine array. Data were analyzed as repeated records using a linear mixed model (PROC Mixed, SAS 9.4) with block as a random effect, and fiber type, enzyme supplementation, and collection period and their interaction as fixed effects. Relative to mid gestation, serum malondialdehyde concentrations were greater in late gestation (5.53 vs. 7.52 μmol/mL; Period P = 0.015), and a similar effect was observed for plasma TAC (5.81 vs. 6.59 mmol/L; Period P = 0.008). Sows fed SF tended to have increased circulating Interleukin (IL)-10 (4.97 vs. 8.59 ng/mL; Fiber P = 0.096) and had increased serum IL-6 (0.65 vs. 1.40 ng/mL; Fiber P = 0.032). Multicarbohydrase supplementation decreased serum IL-4 (17.97 vs. 9.67 ng/mL; Enzyme P = 0.012). Irrespective of collection period, multicarbohydrase supplementation increased Granulocyte-Macrophage Colony-stimulating factor in sows fed IF but reduced it in those fed SF (220, 430, 550, 260 pg/mL for IF-, IF+, SF-, and SF+, respectively; Fiber×Enzyme P = 0.042). Sows fed SF+ had increased serum IL-1 receptor antagonist (2.12, 2.20, 2.69, 4.18 ng/ml for IF-, IF+, SF-, and SF+, respectively; Fiber×Enzyme P = 0.035), and IL-2 (1.53, 1.67, 1.64, 2.89 ng/mL for IF-, IF+, SF-, and SF+; Fiber×Enzyme P = 0.042). In the presence of IF, multicarbohydrases increased serum LBP, but not when supplemented with SF (9.68, 10.90, 10.02, 9.68 ng/mL for IF-, IF+, SF-, and SF+, respectively; Fiber×Enzyme P = 0.028). Circulating IL-8 (0.24 vs. 0.10 ng/mL) and Tumor Necrosis Factor-a (1.26 vs. 0.60 ng/ml) were reduced in sows fed multicarbohydrases (Enzyme P &amp;lt; 0.05). Collectively, supplementing multicarbohydrases may reduce systemic endotoxin and inflammatory pressures throughout gestation, but mechanisms are unique to dietary fiber type.

Effects of dietary supplementation with microencapsulated Galla chinensis tannins on growth performance, antioxidant capacity, and lipid metabolism of young broiler chickens.

This study aimed to investigate the impacts of dietary supplementation with Galla chinensis tannins (GCT) on the growth performance, antioxidant capacity, and lipid metabolism of young broilers. Overall, a total of 216 healthy 1 day-old broilers were randomly allocated to CON group and GCT group, and provided with a basal diet or a basal diet added with 300 mg/kg microencapsulated GCT, respectively, in a 21 days trial. Our findings indicated that dietary GCT addition had no significant effects (p > 0.05) on growth performance. However, GCT supplementation led to a significant reduction in the total cholesterol (TC) concentration in the serum and liver (p < 0.05). Furthermore, GCT supplementation significantly increased the ratios of high-density lipoprotein (HDL) to low-density lipoprotein (LDL) and HDL to TC in the serum, in addition to elevating the activities of enzymes related to lipid metabolism in the liver (p < 0.05). Dietary GCT addition also improved the antioxidant capacity of the broilers, as evidenced by a significant decrease in the concentration of malondialdehyde in serum and liver (p < 0.05). Additionally, the GCT group exhibited significantly increased expressions of hepatic genes associated with antioxidant enzymes (HO-1, GPX1, SOD2, SIRT1, CPT-1, and PPARα) (p < 0.05), while the mRNA expression of SREBP-1 was significantly decreased (p < 0.05) compared with the CON group. In conclusion, dietary addition of 300 mg/kg microencapsulated GCT improved the antioxidant status and lipid metabolism of broilers without affecting their growth performance.

Effect of mineral-vitamin premix supplementation on behavioral, performance, hormonal, oxidative stress, and serum biochemical profiles on rutting male Camelus dromedarius in Egypt

IntroductionThe rutting period imposes a stressful condition on male camels, which results in elevated serum cortisol levels and alterations in their sexual behavior. Therefore, the current work was carried out to investigate the effect of mineral-vitamin premix supplementation on behavior, reproductive performance, hormones, serum oxidative stress profile, and other serum biochemical parameters of Camelus dromedarius during the breeding season.MethodsFourteen mature, fertile male Camelus dromedarius were divided into two groups, a control group (n = 7) and a mineral-vitamin premix group (n = 7). The present study lasted for 95 days during the rutting period (1st February to 5th May). Each camel in the premix group received a daily diet of 50 g of mineral-vitamin premix throughout the whole rutting period, during which the frequencies and durations of the following behaviors: maintenance, posture, aggressiveness, and sexual activity were collected every 20 min. At the end of the study, blood samples were collected.ResultsResults revealed that the premix group showed higher (P &amp;lt; 0.05) maintenance (feeding and rumination), standing, and overall sexual desire-related behavior frequency, besides more times (P &amp;lt; 0.001) for rumination, standing, walking, and lying while showing lower (P &amp;lt; 0.001) frequencies of overall aggressive behaviors than the control group. The serum concentration of malondialdehyde, nitric oxide, cortisol, blood glucose, and urea evidenced a significant decrease in the premix group compared with the control one, while significantly elevated levels of reduced glutathione, testosterone, total antioxidant capacity, triiodothyronine, and thyroxin, total protein, albumin, globulin, calcium, phosphorus, potassium, and magnesium were recorded in the premix group in comparison with the control.ConclusionIt could be concluded that daily dietary supplementation of 50 g of mineral-vitamin premix to male camels during the breeding season is necessary to overcome the oxidative stress and serum cortisol concentration with a subsequent decrease in aggressive behavior and improvement to testosterone level in blood, body condition score and body weight gain.

Gallic Acid Protects Against Methotrexate-Induced Intestinal Mucositis; Oxidative Stress, Histopathology and Inflammatory Status

Background: Gastrointestinal (GI) mucositis is one of the serious side effects of methotrexate (MTX) treatment. It is known that oxidative stress plays an important role in drug-induced side effects. Objectives: The present study aimed to assess the effect of gallic acid (GA) against MTX-induced intestinal mucositis in male Wistar rats. Methods: Twenty-eight adult male Wistar rats were randomly divided into 4 groups (n = 7), including (1) control group; (2) GA group (gallic acid: 30 mg/kg/day, orally); (3) MTX group [20 mg/kg, intra peritoneal (IP)]; and (4) (MTX + GA) group (MTX: 20 mg/kg, IP and gallic acid: 30 mg/kg/day, orally). Then amounts of malondialdehyde (MDA), nitric oxide (NO), glutathione peroxidase (GPx), glutathione (GSH), superoxide dismutase (SOD), interleukin 2 (IL-2) and interleukin 6 (IL-6) were analyzed in serum samples and then the histopathological examinations of the duodenum and jejunum of animals groups. Results: The results showed that treatment with GA significantly reduced the MTX-induced elevation of serum MDA (P &lt; 0.001), NO (P &lt; 0.001), IL-2 (P &lt; 0.001) and IL-6 (P &lt; 0.001) contents and increased MTX-induced reduction in GSH (P &lt; 0.001) content, GPx (P &lt; 0.001) and SOD (P &lt; 0.001) activity. In addition, the histopathological results showed that MTX leads to intestinal tissue damage, and gallic acid can remarkably improve the pathological changes. Conclusions: Our results indicate that gallic acid can mitigate oxidative stress and pro-inflammatory parameters and also moderately prevent histopathological damage of the small intestine of rats exposed to MTX.

Dietary L-theanine supplementation improves antioxidant capacity and lipid metabolism in finishing pigs

The objective of this study was to investigate the effects of dietary L-theanine (LTH) supplementation on finishing pigs' antioxidant capacity and lipid metabolism. Results showed that dietary LTH supplementation increased the activity of antioxidant enzymes and decreased the content of malondialdehyde in serum, longissimus dorsi muscle (LD muscle), and liver of finishing pigs (P < 0.05). Dietary LTH supplementation decreased serum low-density lipoprotein cholesterol content and increased high-density lipoprotein cholesterol content (P < 0.05). Moreover, LTH-containing diet decreased the contents of triglyceride and total cholesterol in serum, LD muscle, and liver (P < 0.05). In liver and LD muscle, LTH activated Nrf2-Keap1 signaling pathway, increased mRNA expression levels of genes related to lipolysis and fatty acid oxidation and reduced the expression levels of genes related to adipogenesis (P < 0.05). According to our study, nutritional supplementation with LTH enhanced finishing pigs' antioxidant capacity and lipid metabolism.

Early supplementation with zinc proteinate does not change rectal microbiota but increases growth performance by improving antioxidant capacity and plasma zinc concentration in preweaned dairy calves.

The present study evaluated the effects of early supplementation with zinc proteinate (ZnP) or zinc oxide (ZnO) for 2 weeks on the growth performance, redox status, plasma trace element concentrations, and rectal microbiota of preweaned dairy calves. A total of 60 newborn healthy female Holstein dairy calves, with initial body weight (BW): 41.33 ± 0.62 kg, were randomly allocated to 5 groups of 12 each: a control group (CON); three groups supplemented with 261 (L-ZnP), 523 (M-ZnP), and 784 (H-ZnP) mg/day ZnP, equivalent to 40, 80, and 120 mg/day zinc, respectively; and one group supplemented with 232 mg/day ZnO, equivalent to 180 mg/day zinc (ZnO). Zinc supplements were administered on days 1-14, and the calves were followed up until day 70. Zinc supplementation increased total dry matter intake (DMI) and starter DMI compared with the CON group (p < 0.01). The final BW, average daily gain, and feed efficiency were higher in the M-ZnP, H-ZnP, and ZnO groups (p < 0.05). The incidence of diarrhea on days 1-28 was reduced by zinc administration (p < 0.01), whereas the incidence on days 29-56 was lower in the M-ZnP and ZnO groups (p < 0.05). Serum glutathione peroxidase activity, total antioxidant capacity, immunoglobulin G and plasma zinc concentrations were increased linearly (p < 0.05), while the serum concentration of malondialdehyde was decreased linearly (p < 0.01), as the dose of ZnP increased. ZnP yielding 80 mg/day zinc had similar effects as ZnO yielding 180 mg/day zinc, except that final BW was higher in the ZnO group (p < 0.05). At the phylum level, ZnO decreased the relative abundance of Firmicutes while increasing the abundance of Bacteroidetes (p < 0.05). At the genus level, ZnO increased the relative abundances of Prevotella, Subdoligranulum, and Odoribacter (p < 0.05). These findings indicated that early supplementation with ZnP did not affect the rectal microbiota of preweaned dairy calves but increased their growth performance, antioxidant capacity, and plasma zinc concentration. In summary, ZnP is an organic zinc source with greater bioavailability than ZnO for preweaned dairy calves. Early dietary supplementation with ZnP yielding 80 mg/day zinc is recommended.
